Enrichment, Antioxidant and Enzyme Inhibition Activities of Flavonoids from Artemisia Selengensis Turcz.

Ting Wang,Weiming Wang,Zhichun Shi,Dan Wang,Jun Li,Liqiu Sun,Ming Zhao

Macroporous resin was used to enrich flavonoids in the ethyl acetate extract of Artemisia Selengensis Turcz. Based on a single factor experiment, the enrichment process was optimized using the response surface method. The optimal parameters of the enrichment process were a sample concentration of 0.3 mg/mL, a loading rate of 1 mL/min, an elution flow rate of 2 mL/min, and a total flavonoid content of 155.38±0.97 mg/g. The flavonoids enriched by AB-8 macroporous resin demonstrated significant scavenging activities against DPPH, ABTS + , and hydroxyl free radicals, and also exhibited certain inhibitory effects on α -amylase and α -glucosidase. Among them, the scavenging ability of the flavonoids enriched by AB-8 macroporous resin on hydroxyl free radical (IC 50 =30.31±1.92 μg/mL) was the closest to Vc, and the inhibitory effect on α -glucosidase (IC 50 =16.19±1.35 μg/mL) was the best. These findings confirmed the potential of Artemisia Selengensis Turcz. was a natural antioxidant and hypoglycemic drug. Graphical

Differential Diagnosis and Precision Therapy of Two Typical Malignant Cutaneous Tumors Leveraging Their Tumor Microenvironment: A Photomedicine Strategy

Peiru Wang,Weitao Yang,Shuzhan Shen,Chao Wu,Long Wen,Qian Cheng,Bingbo Zhang,Xiuli Wang

Elevated hydrogen peroxide (H2O2) in biological tissues is generally recognized to be relevant to the carcinogenesis process that regulates the proliferative activity of cancer cells and the transformation of malignant features. Inspired by this observation, it can be hypothesized that imaging H2O2 in the tumor microenvironment (TME) could help diagnose tumor types and malignancy, and even guide precise therapy. Thus, in this study, a noninvasive photomedicine strategy is demonstrated that leverages the different levels of H2O2 in the TME, and two representative skin cancers, malignant melanoma (MM, clinically higher incidence of metastasis and recurrence) and cutaneous squamous cell carcinoma (cSCC, relatively less dangerous), are differentially diagnosed. The working probe used here is one we previously developed, namely, intelligent H2O2 responsive ABTS-loaded HRP@Gd nanoprobes (iHRANPs). In this study, iHRANPs have advantages over ratiometric imaging due to their bimodal imaging elements, in which the inherent magnetic resonance imaging (MR) mode can be used as the internal imaging reference and the H2O2 responsive photoacoustic (PA) imaging modality can be used for differential diagnosis. Results showed that after intravenous injection of iHRANPs, the tumor signals on both MM and cSCC are obviously enhanced without significant difference under the MR modality. However, under the PA modality, MM and cSCC can be easily distinguished with obvious variations in signal enhancement. Particularly, guided by PA imaging, photothermal therapy (PTT) can be precisely applied on MM, and a strong antitumor effect was achieved owing to the excessive H2O2 in the TME of MM. Furthermore, exogenous H2O2 was injected into cSCC to remedy H2O2 deficiency in the TME of cSCC, and an evident therapeutic efficacy on cSCC can also be realized. This study demonstrated that MM can be differentially diagnosed from cSCC by noninvasive imaging of H2O2 in the TME with iHRANPs; meanwhile, it further enabled imaging-guided precision PTT ablation, even for those unsatisfactory tumor types (cSCC) through exogenously delivering H2O2.

An autocatalytically-activatable hydrogen peroxide photoacoustic sensor for in situ visualization precise diagnosis and drug intervention tracing in diabetes syndrome

Shulong Wang,Yanni Luo,Changchun Wen,Shulin Zhao,Liangliang Zhang

In situ visualization for the diagnosis of diabetic syndrome and visual monitoring the response to drug treatment is a challenge. Herein, we designed and prepared an autocatalytically-activatable hydrogen peroxide photoacoustic (PA) sensor. We first prepared the FeMoO x nanoparticle with catalase activity, then combined it to 2,2′-azino-bis(3-ethylbenzothi-azoline-6-sulfonic acid) (ABTS) and distearoylphos-phoethanola-mine-polyethylene-glycol (DSPE-PEG) to construct a autocatalytically-activatable PA sensor (FeMoO x @ABTS@DSPE-PEG). In its presence, ABTS can be converted into oxidized ABTS · + by H 2 O 2 . ABTS · + exhibits strong light absorption in the near-infrared region, and can serve as an ideal contrast agent for PA imaging. H 2 O 2 as a biomarker of oxidative stress response is closely related to the occurrence and development of diabetes mellitus and its complications. Therefore, FeMoO x @ABTS@DSPE-PEG was used as a PA sensor of H 2 O 2 for visual monitoring of the progression of diabetes-induced liver injury and metformin-mediated treatment of diabetes. The autocatalytically-activatable PA sensor developed in this study provides a promising platform for in situ visual diagnosis of diabetes and its syndrome and monitoring the response to therapy.

Comparison of microwave, ultrasound and ultrasound-microwave assisted solvent extraction methods on phenolic profile and antioxidant activity of extracts from jackfruit (Artocarpus heterophyllus Lam.) pulp

Ming Cheng,Jiali He,Huaijie wang,Chuan Li,Gang Wu,Kexue Zhu,Xiaoai Chen,Yanjun Zhang,Lehe Tan

This study aimed to investigate the impact of different solvents (water, 60% methanol, 60% ethanol and 60% acetone) and methods (conventional solvent extraction, ultrasound-assisted extraction, microwave-assisted extraction and ultrasound-microwave assisted extraction) on the phenolic profile and antioxidant activity of jackfruit ( Artocarpus heterophyllus Lam.) pulp. The Folin-Ciocalteu assay was used to determine the total phenolic content (TPC), and antioxidant activity was measured by DPPH and ABTS assays. The phenolic profiles in different extracts were identified using the UPLC-ESI-Q-TOF-MS/MS technique. The results showed that significant differences ( p  < 0.05) in TPC were found among different extraction methods, the microwave-assisted extraction with 60% ethanol had the highest TPC (2.40 mg GAE/g) compared with the extracts obtained from other methods. Meanwhile, the highest DPPH radical and ABTS radical cation was found in the 60% ethanol-MAE extracts. Correlation analysis further showed a significant positive correlation between TPC and antioxidant activity (R 2  > 0.8, p  < 0.001). Seventy-eight phenolic compounds were identified by UPLC-ESI-Q-TOF-MS/MS analysis. The most abundant phenolic compounds in jackfruit pulp were identified in negative mode, with 60% ethanol-MAE being the most effective. The quantity of phenolic compounds extracted varied with different extraction methods indicated that the extraction methods had effects on the phytochemical characteristics.

Kochiae Fructus: Evaluation on the antioxidant properties and oral safety of its water decoction

Xue-Lin Chen,Bing Li,Xia Zhao,Li-hong Li,Han-Lei Wang,Kun Zhang,Mei Han,Yu-Mei Zhang

Kochiae Fructus (KF) was listed as ‘top grade’ medicinal material by the ‘Shennong's Herbal Classic of Materia Medica’ and has been used in traditional Chinese medicine to delay aging and treat inflammation, such as rubella, eczema, cutaneous pruritus, etc. Our research focused on the antioxidant capability of water decoction and fractions from KF based on 2,2-iphenyl-1-picrylhydrazyl (DPPH) free radical and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radical scavenging assay, the ferric reducing antioxidant power assay, and inhibitory effects on DNA and protein oxidative damage. The results of total phenolics and flavonoids contents showed that ethyl acetate fraction (EAF) possessed the highest phenolics and flavonoids with values of 112.90 ± 9.58 mg gallic acid equivalents/g and 329.60 ± 20.93 mg rutin equivalents/g, respectively. At the same time, the results of antioxidant capacities showed that EAF possessed best antioxidant abilities. In addition, in this work, we evaluated the oral safety of the water decoction of KF (KFWD) via the 14-day acute and 28-day subacute toxicity tests. The results of in vivo toxicity assessment showed that KFWD did not cause significant changes in the general clinical symptoms, hematology and biochemical parameters, organ weights, or histopathological appearances in mice or rats. In summary, the reason why KF has the traditional effect on delaying aging may be related to the fact that its rich in flavonoids and phenolics. Simultaneously, no toxicity was detected after acute or subacute treatment of KFWD, providing valuable evidence for the traditional safe use of KF.

Purpose The early stage of this study verified that a turmeric extract (TUR) including 59% curcumin (CU), 22% demethoxycurcumin (DMC), and 18% bisdemethoxycurcumin (BDMC), could enhance the stability of CU and had greater antidepressant potential in vitro. The objective of the study was to develop a nano-delivery system containing TUR (TUR-NE) to improve the pharmacokinetic behavior of TUR and enhance its antidepressant effect. Methods The antidepressant potential of TUR was explored using ABTS, oxidative stress-induced cell injury, and a high-throughput screening model. TUR-NE was fabricated, optimized and characterized. The pharmacokinetic behaviors of TUR-NE were evaluated following oral administration to normal rats. The antidepressant effect of TUR-NE was assessed within chronic unpredictable mild stress model (CUMS) mice. The behavioral and biochemical indexes of mice were conducted. Results The results depicted that TUR had 3.18 and 1.62 times higher antioxidant capacity than ascorbic acid and CU, respectively. The inhibition effect of TUR on ASP+ transport was significantly enhanced compared with fluoxetine and CU. TUR-NE displayed a particle size of 116.0 ± 0.31 nm, polydispersity index value of 0.121 ± 0.007, an encapsulation rate of 98.45%, and good release and stability in cold storage. The results of pharmacokinetics indicated the AUC(0-t) of TUR-NE was 8.436 and 4.495 times higher than that of CU and TUR, while the Cmax was 9.012 and 5.452 times higher than that of CU and TUR, respectively. The pharmacodynamic study confirmed that the superior antidepressant effect of TUR-NE by significantly improving the depressant-like behaviors and elevating the content of 5-hydroxytryptamine in plasma and brain in CUMS mice. TUR-NE showed good safety with repeated administration. Conclusion TUR-NE, which had small and uniform particle size, enhanced the bioavailability and antidepressant effect of TUR. It could be a promising novel oral preparation against depression.

Optimization of ultrasound-assisted deep eutectic solvent extraction, characterization, and biological studies of polysaccharide from seeds of Toona sinensis

Wen Yao,Chen Geping,Zhang Xiaobing

The present study aimed to evaluate the polysaccharides extracted from Toona sinensis seeds using ultrasound-assisted deep eutectic solvent (DES). The extracted polysaccharides (TSRP) were purified and studied for their hypoglycemic and antioxidant activities. The purified polysaccharides (TSRPs-1) were hydrolyzed and derivatized to study their monosaccharide composition. The optimum extraction conditions were DES (choline chloride to oxalic acid) at a molar ratio of 1.473:1, ultrasound time of 50.643 min, and a material-to-liquid ratio of 1:31.838 g/mL. The ultrasound-assisted DES extraction was more effective (56.1%) as compared to the water extraction (35.89%). The inhibition of α-glucosidase by TSRPs-1 was 42.77% at a concentration of 0.8 mg/mL. At 3.2 mg/mL, TSRPs-1 inhibited α-amylase by 34.38%. The clearance of DPPH by TSRPs-1 was 24.36% at 3.2 mg/mL. The IC 50 for ABTS· + clearance by TSRPs-1 was 0.233 mg/mL. It was shown that the monosaccharides of TSRPs-1 were composed of Man, Rha, GlcA, GalA, Glc, Gal, Xyl, and Ara in a molar ratio of 1:563.13:41.81:51.20:21.81:2244.58:3.73:1130.48. In this paper, the process of extracting TSRP with ultrasound-assisted DES was investigated and optimized. TSRPs-1 possesses certain hypoglycemic and antioxidant activities. Therefore, this paper can provide an experimental basis for the establishment of an industrialized production process of polysaccharides and the study of their biological activities.

In Vitro DNA-Binding, Anti-Oxidant and Anticancer Activity of Indole-2-Carboxylic Acid Dinuclear Copper(II) Complexes

Xiangcong Wang,Maocai Yan,Qibao Wang,Huannan Wang,Zhengyang Wang,Jiayi Zhao,Jing Li,Zhen Zhang

Indole-2-carboxylic acid copper complex (ICA-Cu) was successfully prepared and characterized through elemental analysis, IR, UV-Vis,1H-NMR, TG analysis, and molar conductance, and its molecular formula was [Cu2(C9H6O2N)4(H2O)2]·2H2O. The binding ability of ICA-Cu to calf thymus DNA (CT-DNA) was examined by fluorescence spectrometry and the viscosity method. The results indicated that, upon the addition of increasing amounts of CT-DNA, the excitation and emission intensity of ICA-Cu decreased obviously and the excitation spectra shifted towards a long wavelength. ICA-Cu could displace ethidium bromide (EB) from the EB-DNA system, making the fluorescence intensity of the EB-DNA system decrease sharply; the quenching constantKSVvalue was 3.99 × 104M−1. The emission intensity of the ICA-Cu-DNA system was nearly constant, along with the addition of Na+in a series of concentrations. The fluorescence of the complex could be protected after the complex interacted with DNA. A viscosity measurement further supported the result that the ICA-Cu complex may interact with DNA in an intercalative binding mode. The antioxidant activities of ICA-Cu were evaluated by a 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, a hydroxyl radical (OH) scavenging assay, and a 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay. The ICA-Cu exhibited the highest inhibitory effects on the ABTS radical (94% inhibition at 60 µM), followed by OH and DPPH radicals (the degrees of inhibition being 71% and 56%, respectively). The in vitro cytotoxicity activity of ICA-Cu against two human breast cancer cell lines, MDA-MB-231 and MCF-7, was investigated by 3-[4,5-dimethyltiazol2-yl]-2.5-diphenyl-tetrazolium bromide (MTT) assay and cellular morphological analysis. The results showed that, upon increasing the concentration of ICA-Cu, an increase was observed in growth-inhibitory activity and the inhibition percentage were greater than 90% at 20 µM in both cell lines. Also, cellular morphological changes in the two cell lines agreed with the cytotoxicity results.Keywords:DNA-binding;antioxidant;anticancer;indole-2-carboxylicacid;copper(II) complexes

Improved flavonoid content in mulberry leaves by solid-state fermentation: Metabolic profile, activity, and mechanism

Qiannan Zhao,Xiaohai Yan,Yuan Yue,Tianli Yue,Yahong Yuan

This study was aimed at investigating the mechanisms of release and transformation of total flavonoids (TFs) from mulberry leaves (MLs) by solid-state fermentation (SSF) via Aspergillus cristatus . An intelligent model based on a back-propagation artificial neural network (BP–ANN) was established to predict the TFs in fungi co-fermented MLs. After prediction and experimental validation, the TFs released by SSF (72.55 mg rutin equivalents/g dry weight) were significantly higher than that of unfermented (24.42 mg rutin equivalents/g dry weight). In addition, the biotransformation mechanism of flavonoids during SSF was proposed by analyzing the untargeted metabolome , including the flavonoid and phenylpropanoid biosynthetic pathways . Pearson's correlation coefficient analysis indicated that the 29 up-regulated catabolites (especially loureirin D and arachidoside) after SSF contributed to increased ABTS (2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) radical scavenging activities and α-glucosidase inhibitory activities. Furthermore, structural characteristics (by scanning electron microscopy, thermogravimetric analysis , and FTIR spectroscopy) and analyses of dynamic changes in carbohydrate-hydrolyzing enzymes revealed that the destruction of hemicellulose was essential to releasing TFs by SFF. Industrial relevance In this study, a back-propagation artificial neural network (BP–ANN) was successfully used to establish a co-fermentation with Aspergillus cristatus for releasing and transforming flavonoids of mulberry leaves (MLs) through solid-state fermentation (SSF). The present study has immeasurable importance toward making maximum use of MLs. Moreover, the findings of this study also provide new insights into understanding the release and biotransformation mechanism of flavonoids from MLs by SSF.

Comparative Study on the Antioxidative Effects and α-Glucosidase Inhibitory Potential In Vitro among Ellagic Acid and Its Metabolites Urolithins

Zhao-Rong Li,Rui-Bo Jia,Yurong Mo,Haozheng Wang,Donghui Luo,Chunxia Zhou,Mouming Zhao

The current study compared the radical scavenging and α-glucosidase inhibition potentials of ellagic acid (EA) and its metabolites, urolithins (Uros), and further explored the structure–activity relationship. The outcomes indicated that urolithin M5 (Uro-M5), EA, and urolithin M6 (Uro-M6) exhibited superior 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity; EA and urolithin D (Uro-D) expressed better ABTS scavenging ability, and EA and Uro-M5 showed preferable α-glucosidase inhibition activity. The results of CD spectra and fluorescence spectral analysis explained the interaction between Uros and α-glucosidase. Correlation analysis indicated that hydroxyl groups were crucial for the antioxidative effect, while C-8 OH contributed greatly to the α-glucosidase inhibition activity. Quantum mechanical analysis showed that both EA and Uros exhibited strong electrophilic properties. These comparative results showed a biological discrepancy between Uros and provided essential information for exploring the bioactive application of EA as a functional ingredient or dietary supplement.

Antioxidant and anti-inflammatory effects of different ratios and preparations of Angelica sinensis and chuanxiong rhizoma extracts

Mengtian Zhang,Wei Geng,Xiqin Guan,Shijie Gao,Jinlong Mao

Ethnopharmacological relevance Angelica sinensis (AS) and Chuanxiong rhizoma (CR) are frequently prescribed in clinical settings for their ability to enrich blood, regulate menstrual cycles, and alleviate pain. Despite their widespread use, there is a relative dearth of studies exploring their anti-inflammatory properties. Aim of the study To evaluate the antioxidant and anti-inflammatory effects of Angelica sinensis - Chuanxiong rhizoma (ASCR) extracts and investigate its anti-inflammatory mechanisms. Materials and methods AS and CR were combined in six ratios and extracted using five solvents. The quality of the resulting ASCR extracts was assessed by determining the content of ferulic acid (FA) using HPLC. The antioxidant effects of the ASCR extracts were evaluated in vitro using the DPPH and ABTS assays, as well as in HUVECs exposed to H 2 O 2 -induced oxidative damage. Additionally, the anti-inflammatory effects of the extracts were investigated in vivo through the assays of ear edema in mice and paw edema in rats. Biochemical markers including NO , MDA , and SOD in paw tissues, as well as PGE2 , TNF-α, and COX-2 in rat serum, were measured to further elucidate the anti-inflammatory mechanisms of ASCR extracts. Results The WA-2-1 was obtained by combining AS and CR in a 2:1 ratio through first water then ethanol extraction, and showed favorable antioxidant and anti-inflammatory activities. The extract demonstrated effective scavenging abilities against DPPH• and ABTS + • radicals while also protecting against H 2 O 2 -induced oxidative damage. Furthermore, in vivo studies revealed that WA-2-1 had significant inhibitory effects on ear and paw edema as well as the ability to decrease NO and MDA levels, enhance SOD activity, and downregulate the expression of COX-2, PGE2, and TNF-α. Conclusions The combination of AS and CR exhibits favorable anti-inflammatory effects, attributed to its dual actions of mitigating oxidative stress and suppressing the production of inflammatory mediators in serum or tissues during the inflammatory process.

Preparation of antibacterial composite film based on arginine-modified chitosan and its application in the preservation of ready-to-eat sea cucumber

Jinghe Sun,Yimeng Li,Tingting Yan,Jingfeng Yang

An edible composite film was developed and applied for ready-to-eat sea cucumber storage to improve the product quality. The PAC film base is first prepared by mixing 0.5 % glycerin (GL) with 4 % polyvinyl alcohol (PVA) and 1 % arginine-modified chitosan (Arg-CTS) in the same volume. After the addition of nano-ZnO (ZnO) and thymol (Thy) to the PAC film base, the mechanical properties and functions were tested. Compared to the PAC film, the PAC-ZnO-ThyH composite film showed a 1.34-fold increase in the DPPH scavenging rate and a 2.19-fold increase in the ABTS scavenging rate. Contrary to the PAC film, the inhibition zone diameter of Escherichia coli and Staphylococcus aureus significantly increased by 2.35 and 4.08 folds in the PAC-Zno-ThyH film, respectively. After applying the PAC-ZnO-ThyH film to store ready-to-eat sea cucumber for 10 days, there was a significant reduction in weight loss, total volatile basic nitrogen (TVB-N), and lipid oxidation levels to 1.47 and 1.26 folds to the Ctrl group. After preservation, the hardness and chewiness of ready-to-eat sea cucumber were maintained at 1079.62 ± 138.86 N and 913.73 ± 175.79 N, respectively. The novel PAC-ZnO-ThyH composite film can be used as an active food packaging for promising seafood applications.
