BSA-carbon dots a promising “off–on” fluorescence probe for detecting glyphosate residues in agricultural products

Wang Qian,Wang Miao,Zheng Lufei,She Yongxin,Wang Jing,Jia Minghong,Abd El-Aty A. M.

The widespread and extensive use of glyphosate in agriculture has raised concerns about its potential impact on the quality and safety of agricultural products. Conventional detection methods require long analysis times, making them impractical for the rapid detection of large quantities of samples. Therefore, developing a fast and simple detection system for glyphosate pesticide residues is urgent. In this study, the development of a facile fluorescence probe synthesized using a simple one-pot hydrothermal method for the determination of glyphosate is an important step toward addressing the need for a fast and simple detection system. The present sensor was created using bovine serum albumin (BSA) as a precursor, and the sensor operates by producing an “off–on” fluorescent signal. The bovine albumin-derived BSA-CDs emitted light yellow fluorescence, but this fluorescence was quenched (or suppressed) by the presence of Cu 2+ ions. However, the fluorescence can be restored by the presence of glyphosate, which interacts with the Cu 2+ ions to form a complex and release the BSA-CDs from suppression. The functional groups in glyphosate can capture Cu 2+ and break the BSA-CDs/Cu 2+ combinatorial system. The BSA-CDs/Cu 2+ fluorescence quenching system had good selectivity for glyphosate. The detection limit of the BSA-CD/Cu 2+ fluorescence sensor was 0.05 µg/mL. This developed method was utilized to successfully detect glyphosate in Chinese wheat. The average recoveries ranged from 98.9 to 100.7%, with a relative standard deviation < 3.0%, showing good prospects for practical applicability.

Coating carboxymethylpachymaran on bovine serum albumin (BSA) nanoparticles for the encapsulation and oral delivery of curcumin

Lan Wang,Jin Mao,Qi Zhou,Qianchun Deng,Lei Zheng,Jie Shi

Curcumin is a hydrophobic substance with various beneficial biological activities. Its weak solubility and instability, however, now restrict its usage as a functional food. Herein, the purpose of this work is to fabricate curcumin-loaded bovine serum albumin/carboxymethylpachymaran nanoparticles (BMCNPs) to overcome these challenges. Under optimal conditions, the BMCNPs were spherically shaped with a surface charge of −31.4 mV, polydispersity index of 0.16 and particle size of 158.3 nm were characterized by electron microscopy and dynamic light scattering. The interactions between curcumin, bovine serum albumin (BSA), and carboxymethylpachymaran (CMP) were investigated. BMCNPs had a much better curcumin encapsulation efficiency (74.1%) as well as strong antioxidant activity, storage stability, and ionic strength tolerance. The in vitro release revealed that curcumin was more successfully sustained in the small intestine stage (68.7%) and that BMCNPs were able to prevent it from releasing too early in the stomach (21.4%). Not to mention, the results of the cytotoxicity test performed on the HT-29 and IEC-6 cell lines demonstrated the acceptable biocompatibility of BMCNPs.

Water-soluble gold nanoclusters prepared by protein-ligand interaction as fluorescent probe for real-time assay of pyrophosphatase activity

Hao-Hua Deng,Fei-Fei Wang,Xiao-Qiong Shi,Hua-Ping Peng,Ai-Lin Liu,Xing-Hua Xia,Wei Chen

This paper reports a new and facile method for the synthesis of water-soluble thiolate-protected AuNCs via protein-ligand interaction. Using 3-mercaptopropionic acid (MPA) as a model ligand and bovine serum albumin (BSA) as a model protein, water-soluble AuNCs (BSA/MPA-AuNCs) with intense orange-yellow fluorescent emission (quantum yield=16%) are obtained. Results show that AuNCs produced with this method have hydrophobic interactions with BSA. The synthetic strategy is then successfully extended to produce water-soluble AuNCs protected by other thiolates. Moreover, a sensitive and eco-friendly sensing system is established for detection of the activity of inorganic pyrophosphatase (PPase), which relies on the selective coordination of Fe 3+ with BSA/MPA-AuNCs, the higher affinity between pyrophosphate (PPi) and Fe 3+ , and the hydrolysis of PPi by PPase . A good linearity between the fluorescence intensity and PPase activity within the range from 0.1 to 3   U/L is found, with a detection limit down to 0.07   U/L. Additionally, the fluorescent assay developed here is utilized to assay the PPase activity in real biological samples and as well as to evaluate PPase inhibitor, illustrating the great potential for biological analysis.

Preparation of Boronic Acid-Functionalized Silica Nanocomposites for Selective Enrichment of Glycoproteins

Peng Yang,Zeng-Lian Tian,Li-Ping Xie,Lin Su,Jun-Yi He,Yan-Ping Wu,Wei-Hua Jia

A facile method was developed for synthesis of boronic acid-functionalized silica nanocomposites (SiO 2 -BA) by ‘thiol-ene’ click reaction, where silica nanoparticles were synthesized by using tetraethoxysilane (TEOS) and γ -mercaptopropyl trimethoxysilane ( γ -MPTS) as precursors. The morphology and structure properties of the resultant SiO 2 -BA were characterized by transmission electronic microscopy (TEM), Fourier transform infrared spectroscopy (FT-IR), and Brunner-Emmet-Teller measurements (BET). The adsorption behavior of the SiO 2 -BA for glycoproteins was evaluated. Under the optimized conditions, the SiO 2 -BA exhibited higher adsorption capacity towards glycoproteins (ovalbumin, OVA, 7.64 μmol/g) than non-glycoproteins (bovine serum albumin, BSA, 0.83 μmol/g). In addition, the practicality of the SiO 2 -BA was further assessed by selective enrichment of glycoproteins from egg white samples. Graphical

BSA-chitosan nanocarrier shields dihydromyricetin from digestive degradation and reshapes gut microbiota and their metabolites in vitro

Jingran Liu,Hao Yang,Shuning Yi,Yingyuan Zhao,Xianhui Chang,Qingyun Lyu,Lei Chen,Qian Shen,Wenping Ding,Xi Chen

This study preliminarily confirmed that the primary cause for the low bioavailability of Dihydromyricetin (DHM) was its rapid structural alteration induced by pH fluctuation during digestion, using the in vitro digestion model with HPLC-ESI-MS/MS analysis. Thus, for stable gastrointestinal delivery, DHM was encapsulated using bovine serum albumin (BSA) - chitosan nanocarrier. Physicochemical characterization revealed spherical DHM loaded BSA-chitosan nanoparticles (DBC-NPs, 217.3 ± 3.2 nm) with high encapsulation efficiency (86.66%). In vitro stability tests confirmed their resistance to upper gastrointestinal pH variations and indicated the potential to reach the colon intact. Untargeted metabolomics and 16S rRNA sequencing were performed using in vitro colonic fermentation (three replicates in each group) of gut bacteria from the pooled stools of 6 donors, applying a statistical threshold of p-value <0.05. Furthermore, DBC-NPs modulated gut microbiota (increasing Dorea / Bifidobacterium / Akkermansia , reducing Escherichia-Shigella / Clostridium ) and their corresponding metabolites (such as promoting beneficial phenyllactic acid while inhibiting harmful metabolites such as 3-Oxocholic acid). This study revealed the capability of BSA-chitosan in protecting DHM integrity during digestion and regulative effect on the gut microbiome and their metabolites, offering new strategies for functional foods and precision nutrition interventions.

Three-core fiber sensor based on antibody-functionalized asymmetric C-shaped microcavities for BSA and temperature sensing

Yuxi Ma,Mao-qing Chen,Riqing Lv,Lu Cai,Yong Zhao

An asymmetric dual C‑shaped microcavity sensor based on three‑core fiber (TCF) is proposed and fabricated for label‑free bovine serum albumin (BSA) detection and temperature sensing. Two spatially isolated microchannels of different lengths are directly constructed in the side cores of TCF. One microchannel is modified with BSA‑specific antibodies for specific biomolecular recognition, and the other is filled with thermo‑responsive material to realize temperature sensing. Benefiting from this structural design, high‑sensitivity specific BSA detection and real‑time temperature monitoring are successfully integrated into a single optical fiber. Experimental results verify prominent BSA sensing performance and reliable temperature response of the proposed sensor. After antibody functionalization, the sensor delivers 4.3‑fold higher BSA detection sensitivity than the sensor without antibody treatment, and accurate local temperature measurement is realized in the low‑temperature region. With merits of high integration, good mechanical stability, and low fabrication cost, the presented sensor shows application potential in biological protein analysis, precision medical diagnosis, and low‑temperature reagent preservation.

Development and characterization of BSA-pullulan gel matrices incorporating orange peel flavonoids: Structural interactions and antimicrobial functionality

Mahmoud Salah,Mabrouk Sobhy,Yajing Fang,Salwa M. El-Sayed,Ibrahim Khalifa,Ali Korin,Mohammed Mansour,Syed Asim shah Bacha,Sajid Maqsood,Yun Wang

This study investigates the molecular interactions and functionality of bovine serum albumin-pullulan (BSA-Pn) hydrogels integrated with citrus flavonoid extracts (Fs). Structural characterization via vibrational and fluorescence spectroscopies revealed noncovalent interactions, primarily H-bonding, between BSA, pullulan, and flavonoids, as confirmed by molecular docking modelling. Surface chemistry analysis revealed that increased C1s-peak intensity in BSA-Pn-Fs-9 composite, thereby indicating a structural reorganization. The hydrogel demonstrated potent an antimicrobial activity against Gram-positive ( Bacillus cereus and Staphylococcus aureus ) and Gram-negative ( Escherichia coli ) bacteria, with inhibition zones of 1.34–1.69 cm, and suppressed fungal growth ( Penicillium expansum and Aspergillus westerdijkiae ) by 40.72–43.24 %. Thermal stability of flavonoids within matrix was enhanced, as evidenced by thermal structure analysis. Antioxidant assays showed significant radical scavenging with values of 33.49–93.47 % versus free radicals scavenging, confirming preserved flavonoid bioactivity. These findings underscore the structure-function synergy of BSA-Pn-Fs composites, driven by polysaccharide-protein-flavonoid interactions, positioning them as promising candidates for bioactive and functional biomaterials/additives.

BSA/PEI/GOD Modified Cellulose Nanocrystals for Construction of Hydrogel-Based Flexible Glucose Sensors for Sweat Detection

Tianjun Zhou,Pan Li,Yujie Sun,Wenxiang Wang,Liangjiu Bai,Hou Chen,Huawei Yang,Lixia Yang,Donglei Wei

With the miniaturization, integration and intelligence of sweat electrochemical sensor technology, hydrogel flexible sensors have demonstrated immense potential in the field of real-time and non-invasive personal health monitoring. However, it remains a challenge to integrate excellent mechanical properties, self-healing properties, and electrochemical sensing capabilities into the preparation of hydrogel-based flexible sensors. The utilization of CBPG (cellulose nanocrystals (CNCs)@bovine serum albumin (BSA)@polyethyleneimine (PEI) glucose oxidase (GOD) nanomaterial) as both an enhancing phase and sensor probe within a hydrogel matrix, with poly(vinyl alcohol) (PVA) serving as the primary network constituent, has been proposed as a non-invasive technique for monitoring trace glucose levels in sweat. In this study, BSA was initially attached to CNCs through electrostatic interactions. To further boost the surface active sites of the nanomaterial (CNCs@BSA), PEI was grafted onto the nanomaterial surface. The resulting CNC@BSA@PEI nanomaterials were then used as carriers for GOD. The prepared hydrogel exhibited good self-healing properties (87.5%) and excellent breaking strength (0.8 MPa), effectively converting glucose stimulation in human sweat into electrical output. The sensor had a detection range of 1.0 ~ 100.0 μM and a detection limit as low as 0.9 μM. Due to its ability to specifically recognize trace glucose levels in sweat, the CBPG-PVA sensor can perform highly selective, flexible, and reliable real-time monitoring of human sweat, offering significant potential for wearable biofluid monitoring in personalized health applications

Thiol-Disulfide Exchange Engineered BSA-Derived Nanoprodrug for Precise Delivery of Hydropersulfides Against Hepatic Ischemia/Reperfusion Injury

Qiuyue Wang,Xue Zhang,Baomei Zhang,Zerui Li,Xinlei Meng,Xia Ren,Wenchu Li,Xutao Ma,Hui Xin,Zhiyu He

Hydropersulfides, noted for their potent reducing and nucleophilic characteristics, protect cells against severe oxidative damage. Nevertheless, the free hydropersulfides groups exhibit instability in blood circulation, potentially eliciting disproportionation reactions among themselves. To address this issue, a ROS-responsive bovine serum albumin-derived hydropersulfide nanomedicine (BSA-SS-PBAP NPs) was engineered by a thiol-disulfide exchange strategy that employed BSA-SH with multiple exposed thiol groups and phenylboronic acid pinacol ester-SS-pyridine (PBAP-SS-Py). Upon intravenous administration in ischemia/reperfusion-induced acute liver injury (ALI) mice at 0 h post-reperfusion, BSA-SS-PBAP NPs exhibited excellent colloidal stability in the blood circulation, rapidly targeted the damaged liver, and were internalized by hepatocytes. Under pathological conditions with elevated ROS levels, the responsively released hydropersulfides reshaped the redox equilibrium by eliminating free radicals, replenishing the thiol pool, and restoring the GSH/GSSG cycle, therefore reducing lipid peroxide accumulation. This process subsequently suppressed the Bcl-2/Bax/Cyt-C/cleaved caspase-3/9/12 apoptotic pathways (apoptosis reduced by over 90%), thereby rescuing dying hepatocytes, alleviating the inflammatory cascade, and ultimately improving liver function (AST and ALT normalized within 12 h). The therapeutic administration, administered 3 h post-reperfusion, emulated a clinical regimen and displayed comparable hepatoprotective benefits. This BSA-derived hydropersulfides prodrug platform holds tremendous translational potential for ALI and other oxidative stress-related disease therapies.

Near-infrared fluorescence probe: BSA-protected gold nanoclusters for the detection of metronidazole and related nitroimidazole derivatives

Lei Meng,Jian-hang Yin,Yaqing Yuan,Na Xu

In this work, a novel near-infrared fluorescence probe based on BSA-protected gold nanoclusters (AuNCs@BSA) is designed for the detection of metronidazole (MTZ) and nitroimidazole derivatives. Compared with other wavelengths for fluorescence probes, near-infrared light for MTZ detection has many advantages including effective tissue depth penetration, imaging sensitivity and non-invasivity, and good signal to background noise ratio. The water soluble AuNCs@BSA reported are synthesized through a simple stirring method using BSA and aqueous HAuCl4 as sources, and there is no further sophisticated modification before the detection. The effects of a series of ions (anions and cations), glucose, amino acids and MTZ on the fluorescence intensity of the prepared AuNCs@BSA are studied, and only the MTZ quenches the fluorescence significantly. The mechanism – that electrons transfer from the NCs to the nitro group of MTZ – has been provided, which means that this probe has the potential to determine other nitroimidazole derivatives, including dimetridazole, ronidazole, tinidazole and ornidazole. For MTZ detection, it has a much lower detection limit of 0.01 μM and a wider range between 0.1 to 10 000 μM than other fluorescence probes. Furthermore, a real sample test in human saliva has been successfully performed.

Evaluating the therapeutic efficacy of radiolabeled BSA@CuS nanoparticle-induced radio-photothermal therapy against anaplastic thyroid cancer

Chunmei Zhang,Jinyan Chai,Qiang Jia,Jian Tan,Zhaowei Meng,Ning Li,Menghui Yuan

Bovine serum albumin (BSA) has been employed as a mild biological template in nanoscale particles. Copper sulfide (CuS) has been used for photothermal therapy (PTT) in several studies. In this study, we aimed to synthesize the 131 I-labeled BSA-modified CuS nanoparticles ( 131 I-BSA@CuS), with attributes of both radiotherapy and PTT, as a therapeutic agent against anaplastic thyroid carcinoma (ATC). BSA@CuS nanoparticles were prepared using the solvothermal reaction and then labeled with Na 131 I by the chloramine-T method. The products were characterized and their cytotoxicity was investigated in vitro and in vivo. The therapeutic efficacy of 131 I-BSA@CuS was evaluated in ARO cell (an ATC cell line) subcutaneous tumors. The nanoparticles showed good biocompatibility and low toxicity in vitro and in vivo. BSA@CuS rapidly and effectively converted the light energy from an 808 nm laser into thermal energy with a conversion efficiency of 28.07%. SPECT/CT imaging demonstrated that the accumulation of radioactivity peaked within 24 hr and resided in the tumors for 5 days post intratumoral injection. In vivo assays indicated that, compared to monotherapy, the synthesized nanoparticles employing both PTT and radiotherapy possess better therapeutic efficacy against tumors. The synthesized nanomaterial showed uniform dispersion, good stability and aqueous solubility, excellent photothermal properties, and long-term retention in ATC. Hence, combined radiotherapy and PTT can significantly inhibit tumor growth compared to monotherapy, and can be applied in clinical settings.

Exploring binding interaction of baricitinib with bovine serum albumin (BSA): multi-spectroscopic approaches combined with theoretical calculation

Rong-Juan Zhang,Song-Bo Kou,Lu Hu,Li Li,Jie-Hua Shi,Shao-Liang Jiang

Baricitinib (BRT) is an orally administered small molecule JAK inhibitor, which mainly inhibits JAK-1 and JAK-2. And it is used to treat autoimmune diseases such as rheumatoid arthritis (RA), atopic dermatitis and systemic lupus erythematosus. In this paper, the interaction between BRT and bovine serum albumin (BSA) was studied by fluorescence spectroscopy, UV spectrophotometry, fourier transform infrared spectroscopy (FT-IR) and molecular docking approaches. It has been proved that the fluorescence quenching mechanism of BSA in the mutual complexation process of BRT and BSA was a static quenching. The number of binding sites (n) of BRT-BSA complex was about 1. The binding site was located at site II of BSA and its binding constant was about 5.01 × 10 3 M −1 at 298 k. It is indicated that there was a weak binding between BRT and BSA. By analyzing the sign and magnitude of the free energy change (ΔG 0 ), enthalpy change (ΔH 0 ) and entropy change (ΔS 0 ), it can be found that the complexation process of BRT and BSA is spontaneous and enthalpy-driven. Van der Waals forces and hydrogen bonds play a critical part in the complexation process of BRT with BSA. Based on synchronous fluorescence and three-dimensional fluorescence, it was found that the microenvironment around the residues of Tyr and Trp of BSA had no significant change after binding to BRT. FT-IR and molecular docking studies confirmed that the secondary structure of protein changed during the complexation of BRT with BSA. Experimental results showed that the addition of some metal ions (Ca 2+ , Zn 2+ , Cu 2+ and Co 2+ ) significantly increased the binding interaction of BRT with BSA. This study is helpful to further evaluate the pharmacological characteristics of BRT and provides an important reference for the design of new drugs.
