Precision medicine urges the development of theranostics which can efficiently integrate precise diagnosis and effective therapy. In this study, a facile synthesis of Ir/Gd bimetallic oxide nanotheranostics (termed BSA@Gd2O3/IrO2 NPs) with good biocompatibility was demonstrated using a biomineralization method where bovine serum albumin (BSA) served as a versatile template. BSA@Gd2O3/IrO2 NPs exhibited high longitudinal relaxivity (5.2 mM−1 s−1) and X-ray absorption capability (14.5 Hu mM−1), illustrating them to be a good contrast agent for magnetic resonance (MR) and computed tomography (CT) dual-modal imaging. Moreover, BSA@Gd2O3/IrO2 NPs can act as not only a photothermal conversion agent with ultrahigh efficiency (66.7%) as well as a good photosensitizer, but also an effective catalase to decompose endogenous H2O2 to produce O2, thus relieving hypoxia and enhancing the phototherapeutic effect. Both in vitro and in vivo experiments demonstrated the high effectiveness of BSA@Gd2O3/IrO2 NPs in MR/CT dual-modal imaging and photothermal and photodynamic synergistic tumor treatments. This work sheds new light on the development of versatile nanotheranostic systems using mild and robust biomineralization methods.

Carbon quantum dots (CQDs) with good biosafety can be doped with metals to construct multimodal imaging probes with a wide range of applications in fluorescence and molecular imaging. However, developing CQDs with both long-wavelength emission and a variety of imaging modes remains a challenge. In this article, novel bismuth- and gadolinium-codoped carbon quantum dots (Bi,Gd-CQDs) were developed using a hydrothermal method for green and red fluorescence imaging, X-ray computed tomography (CT) imaging, and T1-weighted magnetic resonance imaging (T1-weighted MRI). In addition to exceptional water solubility, strong steady fluorescence characteristics, and great biocompatibility, the obtained Bi,Gd-CQDs have a high X-ray attenuation coefficient and a short magnetic resonance T1 relaxation time. Additionally, cell imaging involved red fluorescence imaging with deep penetration. Therefore, the prepared Bi,Gd-CQDs with green and red dual-emissions can be used as nanoprobes for fluorescence, CT, and T1-weighted MR imaging, expanding their application prospects in the field of biomedicine.

Integrating multimodal bioimaging and different therapies into one nanoplatform is a promising strategy for biomedical applications, but remains a great challenge. Herein, we have synthesized a biocompatible folic acid (FA) functionalized gold nanorod@polypyrrole@Fe3O4 (GNR@PPy@Fe3O4-FA) nanocomposite through a facile method. The conjugated FA has endowed the nanocomposite with the ability to recognize targeted cancer cells. Importantly, the nanocomposite has been successfully utilized for magnetic resonance (MR), computed tomography (CT) and photoacoustic (PA) multimodal imaging. Moreover, the GNR@PPy@Fe3O4-DOX nanocomposite shows pH-responsive chemotherapy and enables the integration of photothermal therapy and chemotherapy to achieve superior antitumor efficacy. The GNR@PPy@Fe3O4-DOX nanocomposites have a drug release of 23.64%, and the photothermal efficiency of the GNR@PPy@Fe3O4 nanocomposites reaches 51.46%. Cell viability decreases to 15.83% and 16.47% because of the combination of chemo-photothermal therapy effects. Moreover, the GNR@PPy@Fe3O4-DOX-FA nanocomposite could target cancer cells via folic acid and under a magnetic field. The in vivo multimodal imaging and chemo-photothermal therapy effects showed that the GNR@PPy@Fe3O4-DOX-FA nanocomposites are a good contrast and theranostic agent. Thus, this multifunctional nanocomposite could be a promising theranostic platform for cancer diagnosis and therapy.

As an excellent target for cancer theranostics, fibroblast activation protein (FAP) has become an attractive focus in cancer research. A class of FAP inhibitors (FAPIs) with a N-(4-quinolinoyl)-Gly-(2-cyanopyrrolidine) scaffold were developed, which displayed nanomolar affinity and high selectivity. Compared with 90Y, 177Lu, 225Ac, and 188Re, 211At seems to be more favored as a therapeutic candidate for FAPI tracers which have fast washout and short retention in tumor sites. Thus, the current study reported the synthesis of two FAPI precursors for 211At and 131I labeling and the preliminary evaluation of 131I-labeled FAPI analogues for cancer theranostics. FAPI variants with stannyl precursors were successfully synthesized and labeled with 131I using a radioiododestannylation reaction. Two radioactive tracers were obtained with high radiochemical purity over 99% and good radiochemical yields of 58.2 ± 1.78 and 59.5 ± 4.44% for 131I-FAPI-02 and 131I-FAPI-04, respectively. Both tracers showed high specific binding to U87MG cells in comparison with little binding to MCF-7 cells. Compared to 131I-FAPI-02, 131I-FAPI-04 exhibited higher affinity, more intracellular uptake, and longer retention time in vitro. Biodistribution studies revealed that both tracers were mainly excreted through the kidneys as well as the hepatobiliary pathway due to their high lipophilicity. In addition, higher accumulation, longer dwell time, and increased tumor-to-organ ratios were achieved by 131I-FAPI-04, which was clearly demonstrated by SPECT/CT imaging. Furthermore, intratumor injection of 131I-FAPI-04 significantly suppressed the tumor growth in U87MG xenograft mice without significant toxicity observed. The above results implied that FAP-targeted alpha endoradiotherapy (specific to 211At) should be used to treat tumors in the near future, considering the chemical similarity between iodine and astatine can ensure the labeling of the latter onto the designed FAPIs.

Background/Objectives: The purpose of this study was to develop the gemcitabine-loaded drug-eluting beads (G-DEBs) for transarterial chemoembolization (TACE) in rabbit renal tumors and to evaluate their antitumor effect using 2-deoxy-2-[(18)F]fluoro-D-glucose positron emission tomography/X-ray computed tomography (18F-FDG PET/CT). Methods: DEBs were prepared by polyvinyl alcohol-based macromer crosslinked withN-acryl tyrosine andN,N′-methylenebis(acrylamide). Gemcitabine was loaded through ion change to obtain G-DEBs. Their particle size and drug release profile were characterized. VX2 tumors were implanted in the right kidney of rabbits to establish the renal tumor model. The tumor-bearing rabbits received pre-scan by18F-FDG PET/CT, followed by targeted transarterial injection of G-DEBs under digital subtraction angiography (DSA) guidance. The rabbits received another18F-FDG PET/CT scan 10 or 14 days after the treatment. The therapeutic effect was further validated by histopathological analysis of the dissected tumors. Results: The average particle size of the microspheres was 58.06 ± 0.50 µm, and the polydisperse index was 0.26 ± 0.002. The maximum loading rate of G-DEBs was 18.09 ± 0.35%, with almost 100% encapsulation efficiency. Within 24 h, GEM was eluted from G-DEBs rapidly and completely, and more than 20% was released in different media. DSA illustrated that G-DEBs were delivered to rabbit renal tumors. Compared with the untreated control group with increased tumor volume and intense18F -FDG uptake, the G-DEBs group showed significant reductions in tumor volume and maximum standard uptake value (SUVmax) 10 or 14 days after the treatment. Histopathological analysis confirmed that the proliferating area of tumor cells was significantly reduced in the G-DEBs group. Conclusions: Our results demonstrated that G-DEBs are effective in TACE treatment of rabbit VX2 renal tumors, and18F-FDG PET/CT provides a non-invasive imaging modality to monitor the antitumor effects of TACE in renal tumors.

Nowadays, in order to improve the diagnostic accuracy of the disease, more and more contrast agents have been applied in the clinical imaging modalities. A combination of nanotechnology with optical, computed X-ray tomography (CT), and magnetic resonance imaging (MRI) has great potential to improve disease diagnosis and therapy. Herein, we developed a novel multimodal contrast agent for deep-tissue bioimaging based on PEGylated Mn 2+ doped NaLuF 4 :Yb/Er nanoparticles (PEG-UCNPs). The multimodal nanoprobes have revealed the intensely red upconversion luminescence emission for deep-tissue upconversion luminescence (UCL) imaging. Moreover, Owing to the high longitudinal relaxivity, the PEG-UCNPs can be used as T 1 -weighted MRI contrast agents . Additionally, with the high X-ray mass absorption coefficient of Lu 3+ , the novel nanoprobes are appropriate for CT imaging. With integration the high paramagnetic property, superior X-ray mass absorption coefficient and excellent upconversion luminescence in one system, the multimodal nanoprobes can provide a unique opportunity for MRI, CT and UCL imaging. More importantly, modification with PEG endows the novel nanoprobes with high biocompatibility, which would bring more opportunities for the biomedical applications in clinic.

Self-assembled DNA nanostructures, as a new type of nanocarriers, have shown great potential in molecular imaging probes. DNA bipyramid nanostructures (DBNs), as a classic kind of DNA nanostructure, can be precisely constructed through the unparalleled base-pairing precision of oligonucleotide strands. DBNs were prepared by self-assembly of six oligonucleotides with equal molar amounts in a single annealing step, and purified by high performance liquid chromatography (HPLC). DBNs were stable in 10% FBS as well as 80% mouse serum for at least 8 h. To prepare 99mTc-labeled DBNs, N-hydroxysuccinimidyl S-acetylmercaptoacetyl triglycinate (Sacetyl-MAG3-NHS ester) tagged single-stranded DNA (ssDNA) was first radiolabeled with Tc-99m, and DBNs with an overhang were assembled and then hybridized with 99mTc-ssDNA to prepare 99mTc-labeled DNA bipyramid nanostructures (99mTc-MAG3-DBNs). DBNs were radiolabeled, with the radiochemical purity being over 90%. The plasma half-life of 99mTc-MAG3-DBNs in normal KM mice was about 6 min. The biodistribution and SPECT/CT imaging were conducted with 99mTc-MAG3-DBNs in KM mice, and both showed that 99mTc-MAG3-DBNs mainly concentrated in the intestine, liver, and kidneys, and there was also prominent uptake in the gallbladder and bladder. We successfully obtained a new class of SPECT molecular probes based on this DNA polyhedron structure.

Black phosphorus (BP) nanostructures have been receiving enormous attention in diverse bioapplications owing to their unique physicochemical properties. However, it is still challenging to investigate their blood circulation, biodistribution, and pharmacokinetics after administration due to the difficulty in quantifying them. Here, we report the quantification of blood circulation, biodistribution, and pharmacokinetics of dextran-modified BP nanoparticles (i.e., BP-DEX NPs) in balb/c mice through the highly sensitive noninvasive photoacoustic (PA) imaging and single-emission computed tomography/computed tomography (SPECT/CT) imaging. We first prepare water-soluble and biocompatible small BP-DEX NPs and label them with radioisotopes 99mTc. We then quantify their half-lives of distribution and elimination phases to be 0.2 and 9.5 h by counting the γ-emissions in the blood of mice administrated with BP-DEX NPs. We also show the dynamic variation of BP-DEX NPs in the tumor, liver, spleen, and kidney through in vivo PA imaging and illustrate the accumulation of nanoparticles in major organs by SPECT/CT imaging, which follows an order of spleen > liver > lung > kidney. Our work demonstrates the renal clearance and hepatobiliary excretion of BP-DEX NPs, which could be potentially translated for clinical in vivo imaging and therapy of cancer.

In this study, a hollow Ag 2 S/Ag nanocomposite shell comprised of elemental Ag and compound Ag 2 S was synthesized, after loading the sonosensitizer and CT contrast agent, a multifunctional HASAIC nanoprobe was obtained for the first time by wrapping it with a thermosensitively supported lipid bilayer. The elemental Ag in the probe can catalyze H 2 O 2 to O 2 to relieve the hypoxic environment at the tumor site and improve the effect of sonodynamic therapy. Moreover, the hollow Ag 2 S/Ag nanocomposite shell avoids the key limitation of pure Ag shell during the process of catalyzing H 2 O 2 to O 2 , the pure Ag shell will dissolve and could not be used for photothermal therapy (PTT) and photoacoustic imaging (PAI), and enables the probe to use stable Ag 2 S to employ PTT and PAI though solving the problem of hypoxia. The synergy combination of the nano size (70 nm) HASAIC probe, low cytotoxicity, good safety, and high biocompatibility boost up the tumor killing capability. Because of the high efficiency, the HASAIC nanoprobe has been employed for real-time ( in vitro and in vivo) CT and PAI imaging in a 4T1 tumor-bearing mouse. Laser and ultrasound irradiation demonstrated a higher survival rate of HASAIC-injected mice compared to a probe without elemental silver, thereby indicating the high efficiency of our diagnosis and treatment probe.

CT (i.e., hypochlorite concentration C × exposure
time T)
value, borrowed from water disinfection that assumes the equivalent
contribution of C and T, is widely used to indicate membrane chlorination
intensity. Although intentional chlorine treatment is commonly applied
to polyamide nanofiltration membranes for surface modification or
fouling control, whether C and T play equivalent roles in membrane
rejection of diverse contaminants, such as per- and polyfluoroalkyl
substances (PFASs), antibiotics, and endocrine disrupting compounds
(EDCs), remains unclear. Herein, we investigated the impacts of C
and T on contaminant rejection across CT values of 1000–20,000
ppm × h. Results reveal near-equivalent effects on the rejection
of PFASs and antibiotics at low chlorination intensities (e.g., ≤
2000 ppm × h), but significantly inequivalent impacts on the
rejection of all contaminants at higher intensities (e.g., CT ≥
10,000 ppm × h). High-C chlorination induced more severe polyamide
degradation than long T chlorination, drastically compromising contaminant
rejection (e.g., sulfadiazine rejection of 98.2% vs 59.3% for membranes
chlorinated at 2000 ppm × 10 h and 20,000 ppm × 1 h, respectively).
Based on these results, a conceptual framework was established to
decouple the traditional CT paradigm, revealing the fundamental mechanisms
of chlorination-induced changes in membrane rejection.

Titanium-based materials hold great biomedical potential owing to excellent biocompatibility. But titanium diboride (TiB 2 ), an emerging metal boride, is rarely explored for antitumor use, and developing multifunctional TiB 2 -based nanoplatforms with enhanced catalytic activity is urgently needed. Herein, we constructed a nanoplatform (TiB 2 -PEG-DOX) by modifying TiB 2 nanosheets (TiB 2 NSs) with amino polyethylene glycol (NH 2 -PEG-NH 2 ) and loading doxorubicin (DOX) via electrostatic interaction. We first clarified the boron (B)-mediated Ti valence cycle: B regulates Ti electron transfer, promotes Ti 4+ reduction to Ti 2+ , enhancing H 2 O 2 -rich tumor microenvironment Fenton-like reaction to generate cytotoxic •OH for chemodynamic therapy (CDT). Meanwhile, TiB 2 -PEG-DOX shows 53.53% photothermal conversion efficiency under 808 nm near-infrared (NIR) irradiation for efficient photothermal therapy (PTT), and NIR further triggers on-demand DOX release. DOX accumulates at tumor sites via the enhanced permeability and retention (EPR) effect to exert chemotherapy (CT). In vitro, it eliminates 86.32% of HCT-116 colorectal cancer cells via CDT/PTT/CT synergy; in vivo, it achieves nearly complete tumor ablation in mice with excellent biocompatibility. This work provides a high-performance synergistic antitumor platform and establishes a B-modulated metal valence cycling paradigm, paving the way for titanium-based nanomedicines.

In this work, multifunctional polystyrene@chitosan@gold nanoparticles–Fe3O4 nanoparticles–folic acid/indocyanine green (PS@CS@Au–Fe3O4–FA/ICG) nanocomposites that can integrate diagnosis and therapy into one nanoplatform was fabricated. The constructed PS@CS@Au–Fe3O4–FA/ICG nanocomposites possessed excellent computed tomography, magnetic resonance and fluorescence imaging performances which could be used to diagnose tumor and monitor the nanocomposites’ localization in vivo. Moreover, PS@CS@Au–Fe3O4–FA/ICG nanocomposites can be delivered in a targeted manner to cancer cells by folic acid in a receptor-mediated way, and tumor growth could be efficiently inhibited, even completely ablated, under NIR irradiation. That is, the constructed PS@CS@Au–Fe3O4–FA/ICG nanocomposites had great potential for computed tomography, magnetic resonance and fluorescence imaging performances, which could be used to diagnose tumors and to monitor local therapy and fluorescence imaging-guided targeted therapy of cancer cells using nanocomposites.
