Oviductus Ranae alleviates D-galactose-induced ovarian aging by inhibiting ferroptosis and regulating the GPX4/ACSL4 pathway

Ling Xiaomei,Xie Cong,Li Ming,Li Jie,Liu Shun,Sun Liyue,Zeng Xinxin,Zhang Zhen,Zhang Xuhui,Liang Lei

Background Ovarian dysfunction caused by aging restricts female reproductive capacity and causes age-related health problems. Ferroptosis is an important mode of cell death during accelerated aging. Oviductus Ranae (OR), a traditional Chinese medicine, has been used to treat ovarian age-related diseases in women. However, the mechanisms through which OR mitigates ovarian aging, especially ferroptosis regulation, remain unclear. This study investigated the pharmacological effects and mechanisms of OR for ovarian aging in rats. Methods Sprague–Dawley female rats were divided into six groups: aging group induced with D-galactose; experiment groups treated with OR at low, middle, and high concentrations; positive control treated with estradiol valerate; and control group with no treatment. After 42 d, ovarian tissue and serum were collected for biochemical determination, hematoxylin and eosin staining, immunohistochemistry, enzyme-linked immunosorbent assay, reverse transcription-quantitative polymerase chain reaction, western blotting, and RNA sequencing analysis. To elucidate the mechanism underlying the effects of OR on ferroptosis in ovarian granulosa cells (OGCs), in vitro experiments were conducted. Results Treatment with OR improved ovarian follicular development and serum hormone levels, reduced iron deposition, enhanced cell proliferation, and inhibited apoptosis in D-galactose-induced ovarian aging rats. Ferroptosis was alleviated in the ovaries and serum by elevated glutathione and decreased lipid peroxidation production. RNA-seq showed that OR induced changes in 2,768 genes that were involved in ovarian steroidogenesis, glutathione metabolism, lipid metabolism, and ferroptosis pathways. Notably, OR downregulated ACSL4, LPCAT3, and ALOX12 mRNA and protein expressions, while upregulating FTH1, FTL, SLC7A11, GSS, and GPX4, which serve as critical regulators in ferroptosis. OGCs pretreated with OR-containing serum before erastin exposure exhibited enhanced viability, reduced ferrous iron, total iron, and lipid peroxidation levels, improved antioxidant activity, and stabilized mitochondrial function, indicating effective ferroptosis inhibition. Furthermore, OR-containing serum pretreatment inhibited ferroptosis in OGCs by regulating the GPX4/ACSL4 axis. Conclusions OR resists ovarian damage and prevents aging through antiferroptosis mechanisms by regulating the GPX4/ACSL4 axis in OGCs and aging rats. These results highlight the potential of OR as a therapeutic agent for the prevention and treatment of ovarian aging.

A Long-Wavelength Fluorescent Probe for Saccharides Based on Boronic-Acid Receptor

Hongyan Bai,Qian Sun,Haiyu Tian,Junhong Qian,Lingyi Zhang,Weibing Zhang

A single boronic acid-based fluorescent probe (compound CSP) for saccharides was designed and synthesized. The probe, with an α , β -unsaturated ketone conjugated into the coumarin fluorophore, was synthesized by 4 steps from the commercial material 4-diethylamino salicylaldehyde. The electron push-pull effect is enhanced with the N , N -diethyl amino as the electron donor and the carbonyl as the electron acceptor. Both the absorption (463 nm) and emission (616 nm) maxima of CSP are in the visible wavelength region with a Stokes shift of about 150 nm, which ensures CSP a potential probe for biological application. Under near physiological conditions, significant fluorescence enhancement of CSP was observed upon the addition of some saccharides, namely, D -sorbitol, D -fructose, D -glucose, D -mannose and D -galactose. The probe showed relatively high sensitivity towards D -fructose and D -sorbitol, and their detection limits were 0.05 mmol/L and 0.1 mmol/L, respectively.

Boosted high-throughput D⁺ transfer from D₂O to unsaturated bonds via Pdδ+ cathode for solvent-free deuteration

Zhang Xiu-Feng,Zhang Shi-Nan,Zhang Zhao,Leng Bing-Liang,Lu Kai-Yuan,Chen Jie-Sheng,Li Xin-Hao

Deuterated organic compounds have gained significant attention due to their diverse applications, including reaction mechanism studies, probes for metabolism and pharmacokinetics, and raw materials for labeled compounds and polymers. Conventional reductive deuteration methods are limited by the high cost of deuterium sources (e.g., D₂ gas) and challenges in product separation and D₂O recycling. Electrochemical deuteration using D₂O is promising, but existing methods still suffer from low Faradaic efficiency (FE) and high separation costs. Herein, we report a deuterium ion diffusion-based all-solid electrolyser, featuring a RuO₂ anode for D + generation from pure D 2 O and a Pd/nitrogen-doped carbon-based liquid diffusion cathode (Pd δ+ /NC LDC) with tunable electron deficiencies Pd δ+ /NC to enhance selective deuteration. This system achieves over 99% selectivity for deuterated benzyl alcohol with a FE of 72%, and demonstrates broad applicability for the deuteration of aldehydes, ketones, imines, and alkenes with high FE and selectivity. Moreover, the Pd δ+ /NC-based electrolyser can achieve ten-gram-scale production of deuterated benzyl alcohol over 500 hours, showcasing its potential for high-throughput, solvent-free deuteration reactions in practical applications.

Effects of Saikosaponin D on CYP1A2 and CYP2D6 in HepaRG Cells

Hongfang Li,Yunyan Tang,Yang Wang,Weipeng Wei,Chengchen Yin,Fushang Tang

Background Bupleurum is one of the most important traditional Chinese medicines and an ingredient in many compound preparations. It is widely used together with other drugs in clinical practice, and thus there is great potential for drug–drug interactions. Saikosaponin D (SsD) is a major bioactive triterpenoid saponin extracted from Bupleurum with anti-inflammatory, anticancer, antioxidative, and antihepatic fibrosis effects. Effects of the main components of Bupleurum on cytochromes P450 (CYPs) need to be clarified in the clinical application of combination therapies of formulations containing SsD or Bupleurum . Purpose This study aimed to investigate the effects of SsD on the CYP1A2 and CYP2D6 mRNAs, protein expression, and relative enzyme activities in HepaRG cells. Methods HepaRG cells were cultured with SsD at concentrations of 0.5, 1, 5 and 10 μM for 72 hours. mRNA and protein expression of CYP1A2 and CYP2D6 were analyzed with real-time PCR and Western blot analysis. Relative enzyme activities were analyzed with HPLC based on consumption of the specific probe substrate. Results SsD significantly induced expression of mRNA and increased relative activity of CYP1A2 in HepaRG cells after the cells had been treated with SsD at concentrations of 1, 5 and 10 μM. SsD also induced protein expression of CYP1A2 at concentrations of 5 and 10 μM. SsD exhibited an inductive effect on CYP2D6 mRNA and protein expression, while increasing the relative activity of CYP2D6 at concentrations of 5 and 10 μM. Conclusion This study is the first to investigate the effect of SsD on CYP1A2 and CYP2D6 in HepaRG cells, and the results may provide some useful information on potential drug–drug interactions related to clinical preparations containing SsD or Bupleurum .

Acteoside palliates d-galactose induced cognitive impairment by regulating intestinal homeostasis

Maiquan Li,Mingzhi Zhu,Wei Quan,Weisu Huang,Xia Liu,Can Zhang,Baiyi Lu,Xing Xiao,Zhonghua Liu

Acteoside, an important phenylethanol glycoside, is the main active component in Osmanthus fragrans flower. Our previous study found that acteoside showed high antiaging effect but its absorption rate was low. We speculated acteoside palliated aging-related cognitive impairment before being absorbed, that was intestinal homeostasis underlie the antiaging effect of acteoside. In this study, acteoside was confirmed to palliate cognitive impairment in d -galactose induced aging mice. Acteoside treatment dramatically reduced oxidative stress, alleviated intestinal inflammation, restored intestinal mucosal barrier, rebuilt gut microbiome structure and upregulated gut microbiome metabolites short-chain fatty acids (SCFAs) and amino acids (AAs). Furthermore, antibiotic treatment revealed that the antiaging ability of acteoside was abolished in microbiota depleted mice, which offered direct evidence for the essential role of gut microbiota in the attenuation of cognitive impairment of acteoside. Together, our study indicated that acteoside palliated cognitive impairment by regulating intestinal homeostasisand acteoside intake might be a promising nutritional intervention in prevention of neurodegenerative diseases.

Oxyresveratrol prevents lipopolysaccharide/d-galactosamine-induced acute liver injury in mice

Ya-Nan Jia,Hai-Peng Lu,Ya-Lin Peng,Bao-Shun Zhang,Xiao-Bao Gong,Jun Su,You Zhou,Min-Hui Pan,Li Xu

Oxyresveratrol (Oxy) is a natural polyhydroxystilbene abundant in mulberry that has anti-inflammation and anti-oxidant activities. We evaluated the protective effect of Oxy in the context of the lipopolysaccharide and d -galactosamine (LPS/ d -GalN) induced acute liver injury. Oxy restricted the development of histopathological changes, markedly reduced the activity of alanine transaminase (ALT) and aspartate transaminase (AST), which are indicators of impaired liver function. Oxy significantly regulated the contents of oxidative stress related enzymes and products, and inhibited expressions of inflammatory mediators and cytokines. Oxy treatment diminished the Toll-like receptor 4/nuclear factor-kappa B (TLR4/NF-κB) signaling pathway in liver, activated the Kelch-like ECH-associated protein 1(Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) pathway, and increased expressions of heme oxygenase 1 (HO-1) and quinine oxidoreductase 1(NQO1). Pretreatment with Oxy decreased LPS/ d -GalN stimulated hepatocyte apoptosis by efficaciously raising the B-cell lymphoma 2 (Bcl-2)/Bcl-2 associated X (Bax) ratio, inhibiting the expression and activation of caspases , and activating the phosphoinoside-3-kinase (PI3K)-Akt pathway. Our results demonstrate the hepatoprotective efficacy of Oxy. The protection is mainly due to the prevention of TLR4/NF-κB pathway activation, induced activation of Keap1-Nrf2 signaling pathway, and decreased hepatocyte apoptosis. Oxy warrants further study as a potential therapeutic agent candidate for the management of acute liver injury.

A 2-D photonic crystal hydrogel for selective sensing of glucose

Fei Xue,Zihui Meng,Fengyan Wang,Qiuhong Wang,Min Xue,Zhibin Xu

A novel 2-D photonic crystal (PC) sensing material for the visual detection of glucose with high selectivity at a physiological ionic strength (150 mM) is developed. A monolayer polystyrene crystalline colloidal array (CCA) is embedded in a phenylboronic acid (PBA) functionalized hydrogel film to diffract light to sensitively report on the hydrogel surface area. This 2-D PC sensor is superior to the previously reported 3-D PC sensors due to its fast preparation and simple detection. The binding of glucose would increase the cross-linking of the hydrogel that could shrink the hydrogel to increase the Debye diffraction ring diameter. At a physiological ionic strength of 150 mM, the 2-D PC sensor exhibits significant sensitivity for glucose across the entire human physiological glucose range. Additionally, the 2-D PC sensor shows high selectivity for glucose rather than other sugars (fructose and galactose). The diffraction color change of the 2-D PC sensor can be observed. For validation, the prepared 2-D PC hydrogel sensor is applied for the sensing of glucose in the artificial tear fluid.

Separation of d-glucose and d-fructose by porous organic cage

Zhihui Liu,Jian Liang,Yan Qiao,Zongxi Zhang,Yu Liu,Yingxiong Wang

Porous organic cages (POCs) are discrete, three-dimensional molecular species constructed from organic building blocks via covalent bonds. In this work, we found that RCC3, a tetrahedral [4 + 6] porous organic cage possessing 6 vicinal diamine pairs, can selectively separate d -glucose and d -fructose. RCC3 exhibited high binding capacity for d -glucose (0.4 mg/mg of RCC3). This binding process can be reversibly controlled. After a capture-dissociation cycle, the recovery yields of d -glucose and RCC3 were 75% and 83%, respectively. In addition, three binding/dissociation experiments on the binary system of d -glucose and RCC3 demonstrated its reusability. The binding mechanism of RCC3 to d -glucose was investigated via nuclear magnetic resonance spectroscopy (NMR), matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS), and control experiments with various POCs and monosaccharides, revealing the aminal formation between diamines on RCC3 and the carbonyl group of d -glucose. This work demonstrated a novel strategy for the molecular recognition and isolation of d -glucose utilizing porous organic cages.

Weihua Yu,Jianhua Kuang,Qingqing Hu,Zhonghua Wang,Yunwen Liao,Zhengjun Cheng

Graphical A new ratio fluorescent probe (RF-probe) based on the mixing of red-emitting D-penicillamine (DPA) stabilized copper nanoclusters (DPA-CuNCs) with pyridoxal 5’-phosphate (PLP) was designed for sensitive and selective determination of Al 3+ . The excess accumulation of nonessential Al 3+ in the human body can cause serious health problems, thus there is significant interest in the detection of Al 3+ . A new ratio fluorescent probe (RF-probe) based on the mixing of red-emitting D-penicillamine (DPA) stabilized copper nanoclusters (DPA-CuNCs) with pyridoxal 5’-phosphate (PLP) was designed for sensitive and selective determination of Al 3+ . The −CHO group of PLP is expected to form an imine linkage with the free −NH 2 group available of the DPA on the surface of DPA-CuNCs. The content of Al 3+ could be mensurated by the as-prepared RF-probe with a linear response in the range of 1.33–46.67 μmol ⋅ L −1 , and corresponding limit of detection (LOD) was calculated to be 0.33 μmol ⋅ L −1 . Moreover, the RF-probe was successfully extended to the assay of Al 3+ in aluminum carbonate chewing tablets, baking powder, pond water and drink water with satisfactory results compared to the standard atomic absorption spectroscopy (AAS).

Dihydromyricetin Improves Cognitive Impairments in d-Galactose-Induced Aging Mice through Regulating Oxidative Stress and Inhibition of Acetylcholinesterase

Cui-Cui Sun,Zhong-Ping Yin,Ji-Guang Chen,Wen-Jun Wang,Guo-Dong Zheng,Jing-En Li,Ling-Li Chen,Qing-Feng Zhang

Scope Alzheimer's disease (AD) is a neurodegenerative disease with phenomena of cognitive impairments. Oxidative stress and cholinergic system dysfunction are two widely studied pathogenesis of AD. Dihydromyricetin (DMY) is a natural dihydroflavonol with many bioactivities. In this study, it is aimed to investigate the effects of DMY on cognitive impairment in d -galactose ( d -gal) induced aging mice. Methods and Results Mice are intraperitoneally injected with d -gal for 16 weeks, and DMY is supplemented in drinking water. The results show that DMY significantly improves d -gal-induced cognitive impairments in novel object recognition and Y-maze studies. H&E and TUNEL staining show that DMY could improve histopathological changes and cell apoptosis in mice brain. DMY effectively induces the activities of catalase, superoxide dismutase and glutathione peroxidase, and reduces malondialdehyde level in mice brain and liver. Furthermore, DMY reduces cholinergic injury by inhibiting the activity of Acetylcholinesterase (AChE) in mice brain. In vitro studies show that DMY is a non-competitive inhibitor of AChE with IC50 value of 161.2 µg mL −1 . Conclusion DMY alleviates the cognitive impairments in d -gal-induced aging mice partly through regulating oxidative stress and inhibition of acetylcholinesterase.

Gold nanoparticles in situ generated on carbon dots grafted paper: application in enantioselective fluorescence sensing of D-alanine

Siqi Chen,Tingting Li,Donglian Deng,Xiaoyue Zhang,Yibing Ji,Ruijun Li

The integration of paper-based devices with fluorescent nanomaterials as recognition elements has offered novel opportunities for biosensing with enhanced sensitivity and selectivity, as well as stable signals. This work innovatively designed a fluorescent paper-based device for enantioselective sensing of D-alanine (D-Ala), where carbon dots (CDs) grafted cellulose paper (PCD) was used as the fluorophore and the in situ generated gold nanoparticles (AuNPs) were used as quenchers. D-α-Amino acid oxidase (DAAO) stereo-selectively catalyzed D-Ala to produce hydrogen peroxide (H2O2), which mediated the in situ formation of PCD/AuNPs composites and quenched the fluorescence of PCD based on fluorescence resonance energy transfer (FRET). Under optimal conditions, the designed PCD/AuNPs demonstrated a reliable quenching response to D-Ala in the range of 1–100 μM and the limit of detection was 0.55 μM. Moreover, the platform was applied to detect D-Ala in serum, simulated gastric fluid samples, and was further employed to determine D-Ala in BGC-823 gastric cancer cells with satisfactory results. Ultimately, the PCD/AuNPs platform provided a novel strategy for the development of reliable and sensitive assays based on paper-based devices. It has great potential to be utilized in enantioselective sensing and clinical early diagnosis.
