Colorimetric Biosensor Based on Magnetic Enzyme and Gold Nanorods for Visual Detection of Fish Freshness

Xia Xu,Xiaotian Wu,Shunqian Zhuang,Yucong Zhang,Yuting Ding,Xuxia Zhou

Histamine, an important safety index for aquatic products, can also be used as a freshness indicator for red-fleshed fish. In this work, magnetic graphene oxide (Fe3O4@GO, MGO) was applied to immobilize diamine oxidase (DAO) through a method of adsorption and covalent bonding. Under the optimized conditions, magnetic DAO prepared by adsorption immobilization had a higher enzyme activity than that of free enzyme, which was selected for the sensor construction. A colorimetric biosensor based on magnetic DAO induced etching of gold nanorods (AuNRs) was developed for the detection of histamine in fish. The developed biosensor showed an excellent response toward histamine with a low detection limit of 1.23 μM and had negligible interference from other diamines. With increasing the histamine concentration, the AuNRs after the reaction exhibited colors ranging from dark green to blue-green, blue, purple, red, and colorless. The etching induced multicolor change of AuNRs indicated the presence of different contents of histamine in mackerel during storage, and was consistent with the overall change in the content of the total volatile basic nitrogen (TVB-N). Thus, it was indicated that the proposed colorimetric biosensor with a naked-eye-detectable readout has a great potential to evaluate the freshness of red-fleshed fish high in histamine. Keywords:histamine;colorimetric biosensor;magnetic diamine oxidase;etching of gold nanorods;fish freshness

Comprehensive survey and health risk assessment of antibiotic residues in freshwater fish in southeast China

Yongyou Hua,Qinghua Yao,Jian Lin,Xi Li,Yan Yang

To assess the health risks from antibiotic residues in freshwater fish, this study analyzed residues of 65 antibiotics in 10 freshwater fish species sampled in southeast China. Eight antibiotics were detected by UPLC-MS/MS at an overall detection rate of 53.9 %, with 3.48 % of samples exceeding MRLs and 13.0 % suggesting the misuse of human antibiotics. Quinolones , particularly enrofloxacin , were the most frequently detected residues. Azithromycin , an antibiotic intended only for human use, was detected at a rate of 14.8 % at concentrations up to 453.1 µg/kg. Antibiotic residues varied in category and concentration across freshwater fish species. Residues were detected in benthic predatory fish at higher levels than in middle or upper omnivorous or herbivorous fish. Estimated daily intakes showed that the consumption of cultured fish from this region does not pose a serious risk to human health, the hazard index being less than one. However, these findings suggest that the abuse of antibiotics in aquaculture is still a problem, and more attention should be paid to their residues in fish to enhance food safety.

Anhydride structures affect the acylation modification and emulsion stabilization ability of mammalian and fish gelatins

Ting Zhang,Jiamin Xu,Shudan Huang,Ningping Tao,Xichang Wang,Jian Zhong

In this work, ring-like anhydrides (C4, C5, and C6) with different sizes and succinic anhydrides (C4, C10, C12, C14, and C16) with different side chain lengths were used to modify bovine bone gelatin (BBG) and cold-water fish skin gelatin (CFG), and the effect of acylated gelatins on fish oil-loaded emulsions stability was explored. The results showed that the degree of N -acylation decreased with increased ring sizes or side chain lengths, and the surface hydrophobicity of acylated gelatins increased with increased anhydride carbon numbers. Acylated CFGs had higher droplet stability and lower liquid-gel transition time than acylated BBGs. Only BBG-C12 had a slight increase on the creaming stability among these acylated gelatins. These results demonstrated that the gelatins could be modified by all the anhydrides and their emulsion stabilization ability was dependent on the gelatin type and anhydride structure. The results could be beneficial for protein-based emulsifier development and application.

Rheological properties and interactions of fish gelatin–κ-carrageenan polyelectrolyte hydrogels: The effects of salt

Lu Tong,Xinzi Kang,Qi Fang,Wenge Yang,Shijie Cen,Qiaoming Lou,Tao Huang

This study mainly explored the effects of low-concentration salts (0.1, 0.5 mM NaCl and Na 2 SO 4 ) on the gel, rheological and structural properties of fish gelatin (FG)–κ-carrageenan (κC) polyelectrolyte hydrogels. The results showed that κC could increase the gel strength, hardness, and chewiness of the FG–κC polyelectrolyte hydrogels, while the addition of salts had a negative effect. The rheological behaviors showed that the addition of salts reduced the apparent viscosity, gel, and melting points of the FG–κC polyelectrolyte hydrogels. Compared with NaCl, Na 2 SO 4 -treated FG–κC had lower gel strength, hardness, viscosity, gelation, and melting points, while the addition of salts increased the fluorescence intensity by unfolding FG molecules. The secondary structure analysis results showed that the addition of NaCl and Na 2 SO 4 decreased α-helix and β-sheet contents of FG–κC by destroying the hydrogen bond of FG–κC.

Dietary quenching enzyme AiiO-AIO6 promotes fish growth through intestinal microbiota: Zebrafish as a model

Yuan-Yuan Yao,Rui Xia,Ya-Lin Yang,Chen-Chen Gao,Feng-Li Zhang,Qiang Hao,Chao Ran,Zhen Zhang,Zhi-Gang Zhou

The growth performance of cultured aquatic animals is directly related to the economic benefit of the aquaculture industry. Quenching enzyme AiiO-AIO6 could degrade quorum sensing signal molecules N-acylhomoserine lactone (AHL), in this work, the effects of this AiiO-AIO6 on the growth of zebrafish was studied, the control group and AiiO-AIO6 additive group were set up with 120 fish in each group, and the feeding experiment was carried out for three weeks. The following results were obtained by observation of intestinal structure and morphology, quantitative PCR, gut microbes sequencing analysis and transfer, etc. The results showed that the final weight, weight gain rate and specific growth rate were significantly increased and feed conversion ratio and feeding rate of AiiO-AIO6 treated group were significantly decreased. Intestinal digestive enzyme assay showed that there was no significant difference in amylase, lipase and protease activities between the AiiO-AIO6 and control group. Hematoxylin and eosin (H&E) staining of intestinal morphology showed that the height of villus, the number of goblet cells and the height of columnar epithelial cells were significantly increased in the AiiO-AIO6 group. In addition, the relative expression levels of intestinal nutrient transporters Pept1a , GLUT2 and FATP1-a were also significantly increased. The analysis of intestinal microbiota structure and short-chain fatty acids showed that the addition of AiiO-AIO6 could change intestinal microbiota structure and increase the contents of acetic acid and isovalerate. The expression of growth-related genes in intestine, liver and muscle of two groups showed that growth hormone GH , GH receptor GHra , insulin-like growth factor IGF-I and myogenic factor Myf5 of AiiO-AIO6 group were significantly up-regulated. Results in the germ-free zebrafish model showed that the expression of growth-related genes was not directly regulated by AiiO-AIO6, but was mediated by the action of active intestinal microbiota. Overall, our data reflected that AiiO-AIO6 can promote the growth performance of zebrafish, and AiiO-AIO6 is a potential functional additive to make aquaculture efficient and high-yielding.

Biomimetic Mineralized Hydroxyapatite–Fish-Scale Collagen/Chitosan Nanofibrous Membranes Promote Osteogenesis for Periodontal Tissue Regeneration

Maoxue Li,Guoping Cheng,Shimeng Xiao,Bo Jiang,Shujuan Guo,Yi Ding

Commercial mammalian collagen-based membranes used for guided tissue regeneration (GTR) in periodontal defect repair still face significant challenges, including ethical concerns, cost-effectiveness, and limited capacity for periodontal bone regeneration. Herein, an enhanced biomimetic mineralized hydroxyapatite (HAp)–fish-scale collagen (FCOL)/chitosan (CS) nanofibrous membrane was developed. Specifically, eco-friendly and biocompatible collagen extracted from grass carp fish scales was co-electrospun with CS to produce a biomimetic extracellular matrix membrane. An enhanced biomimetic mineralized HAp coating provided abundant active calcium and phosphate sites, which promoted cell osteogenic differentiation, and showed greater in vivo absorption. In vitro experiments demonstrated that the HAp-FCOL/CS membranes exhibited desirable properties with no cytotoxicity, provided a mimetic microenvironment for stem cell recruitment, and induced periodontal ligament cell osteogenic differentiation. In rat periodontal defects, HAp-FCOL/CS membranes significantly promoted new periodontal bone formation and regeneration. The results of this study indicate that low-cost, eco-friendly, and biomimetic HAp-FCOL/CS membranes could be promising alternatives to GTR membranes for periodontal regeneration in the clinic.

Colorimetric aptasensor based on Fe3O4@MOF@Pt nanozymes for ultrasensitive detection of histamine in fish

Xia Xu,Xin Liu,Yixing Qiu,Yufan Song,Xuxia Zhou,Yuting Ding

The accurate determination of histamine in fish is imperative for food safety, given the toxic nature of this biogenic amine . This study presents an innovative colorimetric aptasensor for the ultrasensitive detection of histamine in fish, utilizing nanozymes based on magnetic metal-organic frameworks coated with Pt nanoparticles (Fe 3 O 4 @MOF@Pt), which exhibit high peroxidase-like catalytic activity, magnetism, and superior optical properties. The surface of Fe 3 O 4 @MOF@Pt nanozymes were modified with DNA strands complementary to the histamine aptamer , creating effective colorimetric probes. In the presence of histamine, these colorimetric probes compete with histamine molecules for aptamer binding on the enzyme plate. Color development is triggered by adding a TMB and H 2 O 2 substrate solution to the enzyme plate. This competitive binding-based aptasensor allows for rapid and accurate histamine quantification within a linear range of 10 pM to 10 μM, and a notable detection limit of 6.23 pM. Furthermore, the sensor demonstrated excellent specificity, distinguishing histamine from l -histidine and other biogenic amines. Tested on mackerel samples, the aptasensor showed spiked recoveries of 91.4%–109.6%, validating its practical applicability. This sensor offers an ultrasensitive, specific, and reusable tool for histamine detection, representing a significant advancement in ensuring the safety of fish products.

Proteomic study of the effect of different cooking methods on protein oxidation in fish fillets

Lyulin Hu,Sijie Ren,Qing Shen,Jianchu Chen,Xingqian Ye,Jiangang Ling

The effect of different cooking methods on protein and lipid oxidation of farmed sturgeon (Acipenser gueldenstaedtii) was investigated. In this study, the fish fillets were cooked using five different methods (boiling, steaming, microwaving, roasting and deep-frying), to an internal temperature of approximately 85 °C. General indexes for protein oxidation (carbonyls, free thiols and Schiff bases) and lipid oxidation were evaluated by spectrophotometric and fluorimetric methods. Side-chain modifications of amino acid residues were pinpointed by gel- and MS-based proteomic technology. The results are as follows: (1) cooking, especially roasting and frying, significantly increased the levels of carbonyls and Schiff bases and decreased the amounts of free thiols. (2) Oxidation of aromatic amino acids and extensive oxidation of lysine to carbonyl-containing compounds were characterized in all the cooked samples. (3) Indirect protein oxidation caused by interacting with lipid-derived aldehydes like malondialdehyde (MDA) and 4-hydroxynonenal (HNE) was also observed and much more pronounced in roasted and fried samples, giving a footprint of lipid–protein interaction during cooking. In summary, cooking treatment can induce protein oxidation in sturgeon fillets with a variety of modifications occurring at the protein primary structure. Molecular mapping of protein oxidation is conducive to understand the mechanism of cooking-induced protein oxidation and allows better control of the nutritional properties of protein-based products.

Application of magnetic molecularly imprinted polymers in the detection of malachite green in fish samples

Zheng-zhong Lin,Hong-yuan Zhang,Lu Li,Zhi-yong Huang

Magnetic molecularly imprinted polymers (MMIPs) synthesized through suspension polymerization were used as the adsorbent for the detection of malachite green (MG) in fish samples. The morphological, thermal and magnetic properties of the MMIPs were characterized. The influence of polymerization condition on the absorption capacity of the MMIPs for MG was explored. The results gave the optimum polymerization condition as follows: using MG (0.25   mmol) as template, methacrylic acid (1   mmol) as monomer , ethylene dimethacrylate (5   mmol) as cross linker, 2,2′-Azobis(2-methylpropionitrile) (100   mg) as initiator, Fe 3 O 4 (100   mg) as magnetic nuclei, and PVA (1%) as dispersant . Under the optimum condition the apparent maximum adsorption capacity of MMIPs is 5693   μg·g −   1 with dissociation constant being 23.5   mg·L −   1 . The MMIPs also have strong magnetic responses to an external magnet, which enable the rapid separation from the sample solution. The MMIPs were employed for the detection of MG residue in fish samples combined with HPLC . The recoveries of spiked MG in fish samples ranged from 82.7% to 111% with RSD in the range of 2.0%–12.8%. The MMIPs can retain 90% of the original adsorption capacity even if used for 5 times. Thus, the prepared MMIPs can be applied to the sample pretreatment for separation and enrichment of MG residue in fish samples.

Curcumin and its two analogues improve oxidative stability of fish oil under long-term storage

Qiaoxian Huang,Jian Chen,Chunjiang Liu,Cheng Wang,Chenghui Shen,Yiyi Chen,Qingyong Li

Adding antioxidant is one of the most effective ways to prevent oil systems from oxidation. Curcumin (Cur) and its derivatives have a significant antioxidant activity as potential antioxidants. In this work, the oxidative stability of fish oil treated with three different concentrations (0.1, 0.2 and 0.3 mg/g) of Cur, its two synthetic analogues (compounds 14 and 25 ) and α-tocopherol (positive control) are determined after 70 days at 4 and 25 °C. The antioxidants are evaluated by acid (AV), peroxide (PV), conjugated diene (CD) values, thiobarbituric acid-reactive substances (TBARS) as well as polyunsaturated fatty acid (PUFA) in fish oil. Results show that Cur and α-tocopherol could protect fish oil from oxidation more effectively than compound 14 and 25 at 4 °C, whereas compound 14 and 25 could protect fish oil from oxidation more effectively than Cur and α-tocopherol at 25 °C. Temperature is an important factor on lipid oxidation in the process of fish oil storage resulted from antioxidant activity of compounds determined by temperature. Curcumin shows the decreased ability to protect fish oil from oxidation with increasing temperature. Compounds 14 and 25 show increasing ability to protect fish oil from oxidation with increasing temperature. Practical applications: Compounds 14 and 25 are potential antioxidants to extend expiration date of the fish oil product. Temperature is a key variable for fish oil storage. Compounds 14 and 25 show higher biological stability than that of Curcumin. In long-term storage at 25 °C, compounds 14 and 25 perform better antioxidant activity in fish oil than Curcumin, and tocopherol.

Effect of wogonin, an active ingredient of Scutellariae Radix, on fish infected with Aeromonas hydrophila

Cui Jun,Zeng Dan,Guan Peipei,Jiang Ning

Exposure to pathogens induces hyperactivate inflammatory immune responses, oxidative stress and even death in fish. Therefore, suppressing excessive inflammatory responses is a means of treating fish diseases. Scutellariae Radix (SR), an herbal medicine, has anti-inflammatory and antioxidant properties. However, the active ingredients of SR that produce a marked effect and their mechanisms are poorly understood. In this study, 32 active ingredients of SR and their 124 target genes associated with anti-inflammatory effect were screened. These target genes in zebrafish were mainly enriched in biological processes associated with response to stress and stimulus. The KEGG enrichment analysis and pathway–pathway network analysis showed that the MAPK signalling pathway and nine other pathways were the key pathways associated with anti-inflammatory effect. A protein–protein interactions (PPI) network of target genes was constructed, and AKT2 and nine other genes were identified as the hub genes by cytoHubba. Combining the above results, we constructed an active ingredient–hub protein–key pathway network, showing that wogonin-AKT2 was the key regulated module. Molecular docking and SPR-based assays were used to investigate the possible interaction of wogonin with the AKT2 protein. The infected zebrafish and grass carp treated with wogonin displayed a higher survival rate and anti-inflammatory and antioxidant effects than the infected fish without wogonin treatment. Dietary wogonin also enhanced grass carp anti-inflammatory and antioxidant effects. These findings further reveal the molecular biological mechanism by which wogonin from SR exerts anti-inflammatory effects and provide a theoretical basis for the prevention of fish diseases.

Effects of Maillard reaction of different monosaccharide-modified on some functional properties of fish gelatin

Jinghong Xu,Jing Zhang,Peihan Wu,Feifei Wang,Zongcai Tu,Hui Wang,Debin Guo

In this work, the effects of Maillard reaction of different monosaccharide-modified fish gelatin were studied. The changes of gel properties, rheology and structure of fish gelatin before and after modification were compared and analyzed, and oil-in-woter emulsions were prepared. The results showed that the five-carbon monosaccharide had stronger modification ability than the six-carbon monosaccharide, which was mainly due to the different steric hindrance of the amino acids in the nuclear layer and the outer layer to the glycosylation reaction. With the progress of the Maillard reaction, the color of fish gelatin gradually became darker. The attachment of sugar chains inhibited the gelation process of fish gelatin, decreased the gelation rate, changed the secondary structure, increased the content of β-turn or α-helix, increased the degree of fluorescence quenching, and enhanced the emulsifying properties and emulsion stability. This study provides useful information for the preparation of different types of monosaccharide-modified proteins and emulsions.
