Carbon Dots-Modified Nanoporous Membrane and Fe3O4@Au Magnet Nanocomposites-Based FRET Assay for Ultrasensitive Histamine Detection

Yijie Mao,Yu Zhang,Wei Hu,Weiwei Ye

Histamine can be formed by enzymatic decarbonylation of histidine, which is an important indicator of seafood quality. A rapid and sensitive assay method is necessary for histamine monitoring. A fluorescence resonance energy transfer (FRET) assay system based on a carbon dot (CD)-modified nanoporous alumina membrane and Fe3O4@Au magnet nanocomposites has been developed for histamine detection in mackerel fish. CDs immobilized on nanoporous alumina membranes were used as donors, which provided a fluorescence sensing substrate for histamine detection. Fe3O4@Au magnet nanocomposites can not only act as acceptors, but also concentrate histamine from fish samples to increase detection sensitivity. Histamine was detected by the fluorescence signal changes of CDs capturing histamine by an immune reaction. The fluorescence signals of CDs were quenched by Fe3O4@Au magnet nanocomposites via the FRET mechanism. With an increase of histamine, the fluorescence intensity decreased. By recording fluorescence spectra and calculating intensity change, histamine concentration can be determined with a limit of detection (LOD) of 70 pM. This assay system can be successfully applied for histamine determination in mackerel fish to monitor the fish spoilage process in different storage conditions. It shows the potential applications of CDs-modified nanoporous alumina membranes and Fe3O4@Au magnet nanocomposites-based biosensors in the food safety area. Keywords:fluorescence resonance energy transfer;CDs;nanoporous alumina membrane;food safety

Exactly Matching the FRET Energy of the Functionalized Upconversion Nanoprobe to Boost the Sensing Performance toward Perchlorate

Zhongfei Shang,Meimei Wang,Jiguang Li,Zhenzhen Cai,Luyan Yang,Xincun Dou,Baiyi Zu

The design and development of ultra-accurate probe is of great significance, but there is still a big challenge for chemical sensing in complex practical detection scenes. Here, a unique optical nanoprobe with high sensitivity and anti-interference capability toward perchlorate is designed by exactly matching the FRET energy of the functionalized ligand with the green emission of the UCNPs. Benefiting from the design of specific recognition sites and the intrinsic superiority of the FRET mechanism, the nanoprobe demonstrates an ultralow detection limit of 1.6 nM, rapid response within 4 s, and excellent anti-interference ability even in the presence of 22 potential interferents. The reliable detection performance in complicated real cases is further verified in recognizing trace perchlorate from interferents with various fluorescent emissions and colors existing simultaneously. The proposed design strategy of nanoprobe is expected to open up a brand new methodology for ultra-accurate sensing.

Smartphone-assisted dual-mode turn-off-on red-emission AuNCs/WS2 FRET sensor for detection of carbendazim

Sheng Gong,Fan Nie,Lingxuan Ma,Jiali Wan,Yu Yang,Wensheng Zhang,Jinchao Wei,Peng Li

The overuse of pesticides poses a significant threat to human health, driving the need for accurate and rapid strategies to identify various pesticides. A fluorescent nano-sensor (R-AuNCs@Aptamer) was developed by integrating an amino-modified carbendazim aptamer with red-colored gold nanoclusters (R-AuNCs). We subsequently engineered an aptamer-triggered turn-off-on Förster resonance energy transfer (FRET) platform, employing R-AuNCs@Aptamer in conjunction with tungsten disulfide nanosheets (WS 2 NS). In this setup, the carbendazim aptamer-stabilized red-emitting R-AuNCs (∼655 nm) served as the energy donor, while WS 2 NS acted as a fluorescence quencher. Upon the introduction of carbendazim, alterations in fluorescence intensity were monitored at 655 nm, facilitating the recognition of carbendazim. The resulting fluorescent nano-sensor displayed outstanding selectivity towards carbendazim, exhibiting a broad detection range of 0.1 to 1000 μg/L and remarkable sensitivity with a low limit of detection. It demonstrated reliability and practicality when applied to Notoginseng Radix et Rhizoma and Ginseng Radix et Rhizoma samples, with recoveries ranging from 90 % to 110 %. By changing the aptamer sequence, this sensor can be customized to detect different pollutants, exhibiting excellent tailorability and broad application potential.

Signal “Off-On” Biosensor Based on Fluorescence Resonance Energy Transfer (FRET) for Detection of Sorghum Mosaic Virus

Zhenlong Han,Pengfei Du,Ronghui Wen,Baoshan Chen,Xipu He

A fluorescence off-on method for the detection of Sorghum mosaic virus (SrMV) based on fluorescence resonance energy transfer (FRET) was developed in this study. The biosensor was realized by the adsorption of SrMV coat protein (CP) on gold nanoparticle (AuNP) and conjugation of anti-SrMV antibody (Ab) on cadmium telluride quantum dot (CdTe QD). The optimum quenching efficiency was about 50% obtained at a CdTe QD-Ab/AuNP-CP ratio of 1:7. Moreover, the feasibility of the developed biosensor was verified by the detection of purified CP and the limit of detection was estimated as 0.02 μg/mL. This strategy was also successfully applied to monitor SrMV CP in plant sap with a recovery rate between 97.7 and 107.4%. The developed biosensor is a simple, rapid, and efficient technique which does not need excessive washing and separation steps. Keywords:Sorghum mosaic virus;quantum dot;AuNP;FRET;immune complex

In situ monitoring of the structural change of microemulsions in simulated gastrointestinal conditions by SAXS and FRET

Xia Lv,Shuguang Zhang,Huipeng Ma,Peipei Dong,Xiaodong Ma,Ming Xu,Yan Tian,Zeyao Tang,Jinyong Peng,Haibo Chen,Jianbin Zhang

Microemulsions are promising drug delivery systems for the oral administration of poorly water-soluble drugs. However, the evolution of microemulsions in the gastrointestinal tract is still poorly characterized, especially the structural change of microemulsions under the effect of lipase and mucus. To better understand the fate of microemulsions in the gastrointestinal tract, we applied small-angle X-ray scattering (SAXS) and fluorescence resonance energy transfer (FRET) to monitor the structural change of microemulsions under the effect of lipolysis and mucus. First, the effect of lipolysis on microemulsions was studied by SAXS, which found the generation of liquid crystalline phases. Meanwhile, FRET spectra indicated micelles with smaller particle sizes were generated during lipolysis, which could be affected by CaCl 2 , bile salts and lecithin. Then, the effect of mucus on the structural change of lipolysed microemulsions was studied. The results of SAXS and FRET indicated that the liquid crystalline phases disappeared, and more micelles were generated. In summary, we studied the structural change of microemulsions in simulated gastrointestinal conditions by SAXS and FRET, and successfully monitored the appearance and disappearance of the liquid crystalline phases and micelles.

Fluorescence turn-on sensing of L-cysteine based on FRET between Au-Ag nanoclusters and Au nanorods

Jian-Jun Li,Dan Qiao,Jing Zhao,Guo-Jun Weng,Jian Zhu,Jun-Wu Zhao

The applications of metallic nanoclusters and nanoparticles in biological sensing have attracted special attention owing to their optical interaction based on fluorescence and surface plasmon resonance (SPR). In this work, we designed a fluorescent nanoprobe for the determination of L -cysteine ( L -Cys) based on fluorescence resonance energy transfer (FRET) from gold‑silver bimetallic nanoclusters (Au-Ag NCs) to gold nanorods (AuNRs). Firstly, the negatively charged Au-Ag NCs protected by bovine serum albumin (BSA) are directly adsorbed on the surface of the positively charged AuNRs through electrostatic interaction , and the FRET effect leads to distinct fluorescence quenching of Au-Ag NCs at 615   nm. The SPR wavelength of AuNRs is dependent on the aspect ratio, so the SPR of AuNRs could be tuned to have a better spectral overlap with fluorescence of Au-Ag NCs, which enhances the fluorescence quenching effect. Because the SH group of L -Cys has an affinity with gold, the addition of L -Cys can result in the release of Au-Ag NCs from the surface of AuNRs via forming Au S bonds. Thus, the introduction of L -Cys could effectively restore the fluorescence emission of the AuNRs/Au-Ag NCs system because of the restraint of FRET effect. Under the optimized conditions, the fluorescence recovery of AuNRs/Au-Ag NCs probe exhibits a linear response to L -Cys concentration ranging from 5 to 100   μM, and the corresponding theoretical detection limit (LOD) is 1.73   μM. Meanwhile, this method displays excellent sensitivity and selectivity for L -Cys over other amino acids , and it has been successfully applied to detect L -Cys in real samples.

A fluorometric aptamer nanoprobe for alpha-fetoprotein by exploiting the FRET between 5-carboxyfluorescein and palladium nanoparticles

Li Guiyin,Zeng Junxiang,Liu Huiling,Ding Ping,Liang Jintao,Nie Xinmin,Zhou Zhide

Alpha-fetoprotein (AFP) is a reliable clinical marker of hepatocellular carcinoma (HCC). A highly sensitive fluorometric aptamer nanoprobe is described for AFP detection. It is based on fluorescence resonance energy transfer (FRET) between AFP aptamer labelled with 5-carboxyfluorescein (FAM) and palladium nanoparticles (PdNPs). The PdNPs quench the green fluorescence of the FAM-AFP aptamer via interactions between nitrogen functional groups of the AFP aptamer and PdNPs. When AFP was introduced into the FAM-AFP aptamer-PdNPs FRET system, the AFP aptamer preferentially combines with AFP. This results in a conformational change and weakens the interaction between the aptamer and the PdNPs. Thus, the fluorescence of FAM recovers. The fluorescence recovery of FAM increases linearly in the 5.0–150 ng·mL −1 AFP concentration range and has a 1.4 ng·mL −1 detection limit. The assay was applied to the analysis of spiked diluted human serum. The recovery values ranged from 98.3 to 112.9%, with relative standard deviations of <1.1%. This biosensing strategy provides a reliable and ultrasensitive protocol for the quantification of biomarkers with relevant antigens and aptamers. Schematic presentation of a fluorometric aptamer nanoprobe for AFP assay based on fluorescence resonance energy transfer (FRET) between AFP aptamer labelled with 5-carboxyfluorescein (FAM) and palladium nanoparticles (PdNPs).

Cyclodextrin Functionalized Carbon Dots Using IFE and FRET Dual Mechanism for Sequential Detection of Morin and Al3+

Jinxia Xu,Yueyan Zang,Fanyong Yan,Jingru Sun,Yuyang Zhang,Chunhui Yi

Carbon dots (CDs) are emerging photoluminescent materials with excellent optical properties. However, the lack of active sites in primitive CDs has limited their development applications. Herein, functionalized carbon dots (Z-CDs) are successfully prepared by surface modification of CDs with mono (6-amino-6-deoxy) cyclodextrin (β-CD). The introduction of β-CD increases the spatial potential resistance between CDs, which effectively reduces the self-quenching effect. Moreover, the conjugated domains of Z-CDs are expanded, which improves the optical properties with a quantum yield of 48.74%. Z-CDs are able to be used in the sequential detection of morin and Al 3+ , and the fluorescence mechanisms are confirmed to be internal filtration effect and fluorescence resonance energy transfer, respectively. The limits of detection are 0.817 and 0.231 × 10 −6 m . This study not only provides an idea to solve the problem of self-quenching of CDs but also enriches the detection means of flavonoids and ions, which is expected to be applied to biosensing and environmental monitoring.

Photoactivated multifunctional nanoplatform based on lysozyme-Au nanoclusters-curcumin conjugates with FRET effect and multiamplified antimicrobial activity

Ying Zhang,Hongjun Yan,Rixiang Su,Peiyuan Li,Fangzhou Wen,Yingbin Lv,Jinyun Cai,Wei Su

Photodynamic therapy is a promising alternative to antibiotics in the treatment of bacterial infection, which has the advantages of less side effects, no drug resistance and accurate treatment. However, most photosensitizers (PSs) have some disadvantages, such as poor water solubility, serious self-quenching, poor stability and so on. Therefore, solving the limitations of traditional photosensitizers has become a hot issue in photodynamic therapy. On this basis, the lysozyme (Lys)-gold nanocluster (AuNCs)-curcumin (Cur) conjugate was used as a new type of water-soluble photosensitizer and combined with the bacterial surface through electrostatic interaction to form a bacterial surface enriched in PSs. At the same time, real-time bacterial imaging can be achieved by using the fluorescence properties of Lys-AuNCs-Cur. Under 405  nm light and dark conditions, the inactivation effect of Lys-AuNCs-Cur on Staphylococcus aureus and Escherichia coli was higher than that of free Cur. Moreover, Lys-AuNCs-Cur can also produce enough ROS to kill methicillin-resistant Staphylococcus aureus . The excellent antibacterial properties of nanoparticles may be attributed to the synergism of antibacterial protein lysozyme and AuNCs-Cur nanocomplexes and the increase of 1 O 2 production. The results of scanning electron microscope and fluorescence microscopic imaging showed that the cell membrane of E. coli and S. aureus was seriously damaged, and facilitated penetration of Lys-AuNCs-Cur nanocomplexes into the bacteria through the damaged cell membrane, and improve the photoinactivation efficiency of bacteria. More importantly, nanoparticles exhibited almost negligible dark cytotoxicity. The coupling of Cur and gold nanoclusters can significantly improve the stability of free curcumin molecules and the ability to generate ROS. Therefore, this work is of great significance for the development of new antimicrobial agents against bacterial infection.

An off–on fluorescence aptasensor for trace thrombin detection based on FRET between CdS QDs and AuNPs

Pu Li,Chen Luo,Xiaoxiao Chen,Chaobiao Huang

An off–on fluorescence aptasensor was developed for trace thrombin detection based on fluorescence resonance energy transfer (FRET) between CdS QDs and gold nanoparticles (AuNPs). Using DNA pairwise hybridization of the aptamer to the complementary DNA (cDNA), the CdS QDs (energy donor) were tightly coupled to the AuNPs (energy acceptor), resulting in the occurrence of FRET and there was a dramatic fluorescence quenching of CdS QDs (turn off). When the thrombin was added to the fluorescence aptasensor, the specific binding of the aptamer to the target formed a G-quadruplex that caused the AuNPs receptor to detach and the DNA duplex to be disassembled. The process would inhibit the FRET which contribute to the recovery of fluorescence (turn on) and an “off–on” fluorescence aptasensor for thrombin detection was constructed accordingly. Under optimal conditions, the fluorescence recovery showed good linearity with the concentration of thrombin in the range of 1.35–54.0 nmol L−1, and the detection limit was 0.38 nmol L−1 (S/N = 3, n = 9). Importantly, the fluorescence aptasensor presented excellent specificity for thrombin, and was successfully applied to the quantitative determination of thrombin in real serum with satisfactory recoveries of 98.60–102.2%.

A robust photoswitchable dual-color fluorescent poly (vinyl alcohol) composite hydrogel constructed by photo-responsive FRET effect

Xin Zhang,Beibei Wang,Zhiyu Zheng,Guiqiang Yang,Chao Zhang,Liqiong Liao

Stimuli responsive fluorescence characteristics are significant in some fields, such as sensors, information encryption , and bionic camouflage. In this work, a robust poly(vinyl alcohol) (PVA) hydrogel with photoswitchable dynamic fluorescence behaviors was constructed by incorporating dual-color fluorescent nanoparticles (DCFNs). The DCFNs were composed of the spiropyran-modified β-cyclodextrin (β-CD-SP) and nitrobenzoxadiazolyl (NBD) derivatives, which could exist in the PVA network through non-covalent interaction. The SP moiety functioned as an acceptor and realized the photoisomerization to further quench or recover the fluorescence of the NBD moiety based on the fluorescence resonance energy transfer (FRET) effect. Therefore, under the irradiation of ultraviolet light (UV) or visible light (Vis), the hydrogel showed a fluorescence emission peak of merocyanine (MC) at 660 nm (red) or NBD at 550 nm (green), respectively. Additionally, the pH responsive characteristic of the MC chromophore also endowed the hydrogel with pH responsive fluorescence behaviors. Utilizing these advantages, specific patterns can be displayed on the hydrogel in dual mode through a hollowed-out photomask. The hydrogel could also be utilized as dual-color “building blocks” to construct various 2D or 3D structures. This composite hydrogel has potential application in various fields, such as sensors, optical switch , information storage, and biomimetic components.

A biosensor using semi-DNA walker and CHA -FRET loop for ultrasensitive detection of single nucleotide polymorphism

Yunshan Zhang,Shijie Xu,Jian Chen,Lanyue Wang,Lina Bian,Jing ye,Lin Weng,Xiaoyu Zhao,Cheng-Te Lin,Shuang Li,Diming Zhang

Single nucleotide polymorphisms (SNPs) are emerging as important biomarkers for disease diagnosis, prognostics and disease pathogenesis. However, it is still challenging to make highly specific and sensitive SNPs detection to distinguish target DNA with single-base difference, so that flap endonuclease 1 (FEN 1) manipulated semi-DNA walker reaction and catalytic hairpin assembly (CHA)-FRET loop for ultrasensitive SNP detection has been proposed. Herein, FEN 1 was employed to specifically recognize and cut the 5′ flap of three-base overlapping structure formed by only hybridizing with mutant target (MT). Simultaneously, FEN 1 drove MT hybridization to autonomously walk along downstream probe (DP) and the cut sequence (CS) from semi-DNA walker reaction was collected by magnetic bead-based separation to avoid false-positive signal. Next, the cleaved CS triggered CHA and generated fluorescence resonance energy transfer (FRET) via Cy3 and Cy5. FEN 1 manipulated semi-DNA walker reaction to specifically recognize the target DNA and combined CHA-FRET loop to dually amplify signal. Thus, the SNP biosensor demonstrated superior performance for ultrasensitive detection of mutant KRAS gene with a low detection limit as low as 80 aM and was successfully applied to monitor the expression of mutant KRAS gene in human cancer cell lysates. This strategy provides an ultrasensitive way for the detection of biomolecules and reveals an effective avenue for diseases diagnosis.
