p53 gene is one of the most important anticancer genes and has been recognized as a typical biomarker for the early diagnosis of cancers. In this paper, a portable point-of-care testing (POCT) platform was developed for the colorimetric and photothermal dual-modal analysis of the p53 gene. Long concatemer G4 structures with high catalytic activity were formed by a target-driven hybridization chain reaction, and their catalytic properties can further be enhanced with the help of iodide ions. This dual-modal method realized the sensitive multicolor visual point-of-care detection of the p53 gene, where color variations from colourless to blue were easily distinguished by the naked eye. The temperature output was achieved by effective photothermal conversion and regulation guided by the accumulated concatemer G4 complexes in the TMB–H2O2 system, avoiding the need for complex instruments. The proposed dual-modal assay for p53 gene detection is cost-effective, simple, highly sensitive (3.34 pM), stable, and remarkably selective. It was also applied to determine the concentration of the p53 gene in spiked serum samples.

A novel polymer-based monolithic column was prepared via redox initiation system within the confines of a stainless steel column with 4.6 mm i.d. In the processes, hemin and lauryl methacrylate were used as co-monomers; ethylene dimethacrylate as crosslinking agent; n-butyl alcohol, ethanediol, and N , N -dimethylformamide as tri-porogens; benzoyl peroxide and N , N -dimethyl aniline as redox initiation system. The resulting polymer-based monolithic columns were characterized by scanning electron microscopy, nitrogen adsorption-desorption instrument, and mercury intrusion porosimeter, respectively. The results illustrated that the improved monolith had relative uniform porous structure, good permeability, and low back pressure. Aromatic compounds were used to test the chromatographic behavior of the monolith, resulting in highest column efficiency of 19 880 plates per meter with reversed-phase mechanism. Furthermore, the homemade monolith was used as the stationary phase of high performance liquid chromatography to separate proteins from complex bio-matrix, including human plasma, egg white, and snailase. The results showed that the monolithic column occupied good separation ability with these complex bio-samples. Excellent specific character of the homemade hemin-based monolith was that it could simultaneously remove high-abundance proteins (including human serum albumin, immunoglobulin G, and human fibrinogen) from human plasma and separate other proteins to different fractions.

Telomerase, a widely accepted cancer biomarker for early cancer diagnostics, is considered as an important therapeutic target. To now, it is still a challenging subject to develop a simple and sensitive strategy for telomerase activity detection. Herein, we reported a simple colorimetric strategy for label-free quantification of human telomerase activity in urine by using hemin-graphene nanomaterial (H-GNs). H-GNs possessed tailored dispersibility in the high salt concentration and highly active biomimetic oxidation catalyst property. In this strategy, H-GNs were adjusted to coagulate to appropriate degree by carefully selecting the contained NaCl amount in the presence of original TS primer. The supernatant of the solution contained few H-GNs and showed light blue color. Under the action of telomerase, TS primer was elongated with repeating sequences of (TTAGGG) n . These negatively charged DNA enhanced individual H-GNs electrostatic repulsion and resisted salt-induced H-GNs coagulation. As a result, the supernate of the corresponding solution containing more dispersed H-GNs and showed dark blue color after chromogenic reaction. Thus, telomerase activity could be quasi-quantified by naked eye and precise quantified by UV spectrometer. The proposed method has the linear range from 100 to 2300 HeLa cells/mL and the detection limit was 60 cells/mL. It has been successfully applied to detect telomerase activity in real urine samples. Obtained results were in good agreement with the clinical diagnosis. Therefore, this colorimetric approach affords simplicity, sensitivity and reliability in telomerase activity detection.

A highly sensitive, low-cost colorimetric aptasensor was developed for the determination of 8-hydroxy-2′-deoxyguanosine (8-OHdG) in human urine. The method is based on a conformational switching of the 8-OHdG aptamer to form a G-quadruplex structure in the presence of 8-OHdG. The resulting G-quadruplex assembles into a peroxidase-like DNAzyme with hemin , which effectively catalyzes the oxidation of 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS 2− ) by H 2 O 2 to ABTS + , resulting in an increase in the absorption signal at 416 nm along with a color change of the solution. The response signals linearly correlated with the concentration of 8-OHdG, ranging from 466 pM to 247 nM with a detection limit of 141 pM. The relative standard deviation and the recovery were 1.97–3.47% ( n = 11) and 98.8–100.2%, respectively. The proposed method avoids the label and derivatization steps in common methods and allows direct analysis of the samples by the naked eye without costly instruments, which is reliable, inexpensive, and sensitive.

Inspired by natural enzymes, artificial enzymes have garnered significant interest due to their simplicity of production, robustness under harsh conditions, and enhanced stability. This study introduces, for the first time, a novel Z/Ce@hemin composite enzyme, constructed by anchoring hemin onto zeolitic imidazolate framework-8 (ZIF-8)-encapsulated ceria (CeO 2 ) nanoparticles. This innovative design overcomes the challenges of hemin dimerization, enhances substrate affinity, and accelerates mass transport, leading to significantly improved peroxidase-like activity. The enzyme also demonstrates remarkable stability and resistance to environmental interference. Utilizing this Z/Ce@hemin composite, a sensitive and selective colorimetric detection system was developed for rapid and accurate quantification of hydrogen peroxide (H 2 O 2 ), a key oxidative stress marker. This approach was successfully applied in cellular models and Caenorhabditis elegans models of Parkinson’s disease, enabling precise monitoring of H 2 O 2 . The study provides a groundbreaking platform for oxidative stress modeling and expands possibilities in neurodegenerative disease research and therapeutic screening.

A hemin (Hm)-engineered cerium-based metal–organic framework (MOF), UiO-66(Ce)@Hm, was prepared and used as a nanozyme for electrochemical sensing of hydrogen peroxide (H2O2) and dissolved hydrogen (H2) in beverages. UiO-66(Ce)@Hm showed enhanced electrocatalytic activity toward H2O2 because of the synergistic catalytic effects of Hm and UiO-66(Ce). The resulting sensor enabled H2O2 detection from 0.5 to 250 μM with a detection limit of 0.32 μM, and was successfully applied to milk analysis. In addition, dissolved H2 in hydrogen-rich water (HRW) promoted the catalytic conversion of o-phenylenediamine/H2O2 to electroactive 2,3-diaminophenazine, allowing H2 determination from 45 to 900 μM with a detection limit of 26.2 μM. These results demonstrate the potential of UiO-66(Ce)@Hm for food safety monitoring and quality assessment of HRW beverages.

Atherosclerotic Cardiovascular Disease (ASCVD) is one of the major fatal diseases for humans and the fatality rate is increasing rapidly. Low density lipoprotein (LDL) is a serum key marker in the diagnosis of ASCVD because LDL level is associated with the prevalence of ASCVD. Herein, a light addressable potentiometric sensor (LAPS) was constructed for rapid detection of LDL using reduced graphene oxide-polyaniline-hemin nanocomposites (RGO-PANI-HNCs) and low density lipoprotein aptamer (LDLapt) as sensitive membrane of LAPS. Under the effect of bias voltage, the specific binding between LDL and LDLapt, and forming the LDL-LDLapt complex will cause the voltage change on the sensitive membrane surface of the LAPS system, which will cause the change of original current–voltage (I-V) balance in the circuit, resulting in the shift of I-V curve. The correlation between the potential shift of LAPS and LDL concentration was linear from 1.0 to 100.0 μg/mL under optimal conditions. The sensitivity is 4.7671 mV/μg/mL, and the detection limit is 0.79 μg/mL. Also, the LDL biosensor showed satisfactory specificity, selectivity, reproducibility, and showed a good precision in real human serum samples, which provides a simple and effective way for the early diagnosis of ASCVD.

Graphical A new type of nanocomposite enzymes has been developed, composed of Pd−Pt nanoparticles, hemin-rGO and CNTs-COOH. The nanocomposites show a high peroxidase-like activity and a linear response toward H 2 O 2 in the range of 0.05–500 μM with a determination limit of 0.01 μM. Based on this, a paper sensor was constructed for H 2 O 2 detection in real samples. Nanocomposite enzymes are attracting intensive attention worldwide. In the present study, a novel colorimetric H 2 O 2 paper sensor was developed based on a new type of nanocomposites composed of Pd−Pt nanoparticles, hemin functionalized reduced graphene oxide (hemin-rGO) and carboxylated carbon nanotubes (CNTs-COOH). The proposed nanocomposites show a high peroxidase-like activity. It provides a sensitive platform showing a linear response toward H 2 O 2 in the range of 0.05–500 μM with a determination limit of 0.01 μM. Based on this, the composites were applied to construct a paper sensor for H 2 O 2 detection in real samples. Herein, an ultrasensitive and stereo-selective electrochemiluminescent (ECL) biosensor based on ECL signal amplification of luminol by the synergetic catalysis of a hemin-functionalized composite and gold–platinum nanowires (Au-PtNWs) has been designed for the detection of D-alanine (D-Ala). In the sensor design, D-amino acid oxidase (DAAO) was used to generate the ECL co-reactant (H2O2) for luminol in situ. The reduced graphene oxide-hemin-cysteine (rGO-H-Cys) composite and Au-PtNWs acted not only as amplification labels to amplify signals via synergetic catalysis, but also as an ideal nanocarrier to accelerate electron transfer. A great difference in ECL intensities toward D-Ala and L-Ala was obtained, and an obviously higher intensity was seen for D-Ala. Under optimal conditions, the ECL intensity had a linear relationship with the logarithm of the D-Ala concentration in the range from 1.0 × 10−9 to 5.0 × 10−3 M with a detection limit of 3.3 × 10−10 M (S/N = 3). This study provides a new method for the determination of D-Ala in food with high sensitivity, excellent selectivity, and good stability and reproducibility.

Glypican-3 (GPC3) is a highly specific tumor marker for hepatocellular carcinoma (HCC), and plays an important role in reflecting the existence, therapeutic evaluation, monitoring and prognosis of HCC. Herein, an electrochemical aptasensor was designed for GPC3 detection with the reduced graphene oxide-hemin nanocomposites (RGO-Hemin) modified on the screen-printed electrode surface as the sensing platform and GPC3 aptamer as recognize molecule. In the existence of GPC3, the aptamer can specifically bind with the target GPC3 and form GPC3-aptamer conjugations on the sensing surface, which would increase the resistance of the electron transfer on the electrode and make the decrease of electrochemical signals of Hemin in RGO-Hemin nanocomposites. The electrochemical current change was recorded by differential pulse voltammetry (DPV). Scanning electron microscopy (SEM), Raman microscope (RM), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to characterize the GPC3 electrochemical aptasensor. Under the optimum conditions, the current response of the electrochemical aptasensor is linearly correlated with the concentration of GPC3 (0.001–10.0 μg/mL) with the detection limit of 2.86 ng/mL (S/N = 3) and the sensitivity of 0.134 μA/μM/cm 2 . In addition, the aptasensor was applied to the determination of GPC3 in spiked human plasma and the recoveries fluctuated from 102.68% to 117.29%. All these results show that the aptasensor has good specificity, sensitivity, stability and reproducibility for GPC3 detection.

A novel hemin/phytic acid doped polyaniline (PA-PANI) hydrogel composite was prepared through a simple chemical and self-assembly method, which was modified onto electrode for electrochemical detection of H 2 O 2 released from living cells. It showed good analytical performance with high sensitivity, selectivity and a rapid response for the analysis of H 2 O 2 in the range of 2 to 102 μM, with the detection limit of 1.2 μM. The favorable results mainly originated from both the high conductivity of PA-PANI hydrogel and its network structure preventing hemin from self-dimerization to provide active catalytic species. Furthermore, PA-PANI with good biocompatibility allowed living cells to adhere and resulted in a short diffusion distance between H 2 O 2 released from cells and electrode.

Lysozyme aptamer-functionalized magnetic alginate hydrogel was prepared for separation and enrichment of lysozyme. Luminol-labeled aptamer was used as a signal tag, and the signal tag was adsorbed on magnetic carboxylated carbon nanotubes based on the π-interaction. When lysozyme was added, the aptamer specifically binds to the lysozyme, causing the signal tag to detach from the magnetic carboxylated carbon nanotubes. When the aptamer/lysozyme complex bound to the complementary single strand of aptamer on the hemin@HKUST-1, lysozyme was released. The released lysozyme can be recombined with the signal tag adsorbed on the magnetic carboxylated carbon nanotube, allowing more signal tag to be dispersed into the solution. Determination of lysozyme was achieved by releasing the luminol-labeled aptamer to generate a chemiluminescence signal at a wavelength of 425 nm. It was proved by experiments that the synthesized hemin@HKUST-1 had a strong catalytic effect on the luminol-NaOH-H 2 O 2 system. The chemiluminescence signal was increased nearly 100 times. The complementary pairing allowed the luminol to be immobilized on the surface of hemin@HKUST-1. The generation and consumption of short-lived reactive oxygen species were concentrated on the surface of the MOFs, which improves the chemiluminescence efficiency. The introduction of hemin@HKUST-1 and DNA solved the defects of chemiluminescence analysis. The chemiluminescence assay was able to detect lysozyme with linear range of 1.05 × 10 −6 U∙mg −1 (6.00 × 10 −13 mol∙L −1 )–1.25 × 10 −2 U∙mg −1 (7.14 × 10 −9 mol∙L −1 ); the detection limit was 3.50 × 10 −7 U∙mg −1 (2.00 × 10 −13 mol∙L −1 ) ( R 2 = 0.99). The recovery of lysozyme in spiked saliva samples was 97.4–102.8%.
Schematic presentation of chemiluminescence assay. Lysozyme (Lys) was captured by aptamer-modified magnetic sodium alginate (M-Alg-Apt); Glycine (pH = 2) as eluent for Lys. Luminol-modified Apt (Apt-luminol) as signal tag; magnetic carbon nanotubes (MCNTs) as adsorption matrix; cDNA was complementary to Apt; hemin@HKUST-1 as catalyst.
