Approximately half of lung cancer patients experience bone metastasis, leading to bone loss, fracture, and other skeletal-related events. Although immunotherapies have significantly advanced the therapeutic landscape for lung cancer, bone metastases have an immunologically "cold" microenvironment, representing a challenging obstacle when treating lung cancer. The combination of immunotherapy and photothermal therapy (PTT) for treating tumor-induced bone defects holds promise for enhancing the efficacy of local tumor ablation and inhibiting tumor recurrence and metastasis through activating systemic immune responses. Herein, we developed an injectable hydrogel-based photothermal immunotherapy system (BP@Gel-CD[SA] hydrogel) incorporating STING agonists (SA) and black phosphorus nanosheets (BPNSs) for high-efficiency tumor elimination, immune activation, and bone regeneration. The photothermal and photodynamic activities of BPNSs induce hyperthermia and ROS-mediated apoptosis of tumor cells. Meanwhile, SA loaded into the nano-boxes in BP@Gel-CD[SA] hydrogel by host-guest interaction significantly activates the cGas-STING pathway. It stimulates immunogenic cell death (ICD), synergistically promoting immune cell infiltration. Single-cell RNA sequence analysis confirms the modulation of the tumor microenvironment (TME) through the PTT-mediated ICD effect and the transactivation of the cGAS-STING pathway in immune cells of the TME. More importantly, the system can significantly inhibit the growth of distant tumors via systemic immune activation and elicit long-term immune memory in addition to tumor eradication. In the long term, this hydrogel system can promote the formation of new bone at sites of tumor-induced bone destruction, improving bone strength in the affected area. Collectively, this strategy provides an effective and safe option for treating lung cancer bone metastases.

Osteoarthritis (OA) pathogenesis is profoundly influenced by dysregulated immune dynamics, where persistent interleukin-17 (IL-17)/T helper 17 (Th17) cell mediated inflammation coordinates with failed regenerative processes to perpetuate joint destruction. Here, we unveil the role of myeloid-derived suppressor cells (MDSCs) as dual-phase regulators that paradoxically orchestrate both inflammatory escalation and tissue repair in OA progression. Intra-articular administration of MDSCs in OA mice amplified IL-17 dependent inflammatory cascades and chemokine-driven leukocyte recruitment, revealing a context-dependent pro-inflammatory phenotype. Unexpectedly, MDSC depletion failed to attenuate joint damage, implying their indispensable yet multifaceted role in OA pathogenesis. Mechanistically, MDSCs exhibited functional plasticity by upregulating arginase-1 to polarize M2 macrophages, fostering a regenerative niche alongside their inflammatory activity. To resolve this duality, we developed a bio-responsive hydrogel-microsphere system integrating transforming growth factor β1 (TGF-β1) and interleukin-1 β1 antibody (anti–IL-1β) loaded mesoporous silica nanoparticles (MSNs). This spatiotemporally controlled platform selectively suppressed MDSC-mediated Th17 cell expansion while harnessing their intrinsic capacity to drive M2 macrophage polarization and chondrogenesis. The resultant shift from a pro-inflammatory to pro-regenerative microenvironment significantly attenuated cartilage erosion and restored joint integrity in OA models. Our findings redefine MDSCs as bifunctional immune orchestrators in OA and establish precision biomaterial guided immune decoding as a paradigm-shifting therapeutic strategy. By engineering MDSC plasticity through antagonistic cytokine delivery, this work provides a blueprint for microenvironment remodeling in degenerative joint diseases.

The simultaneous integration of high elasticity and lubricity-hallmarks of biological tissues-remains a fundamental challenge in synthetic hydrogels due to the intrinsic trade-off between “dehydration-induced” elasticity and “hydration-dependent” lubrication. Herein, inspired by the dynamic architecture of living systems, the construction of “living” biodegradable hydrogel microspheres is reported that reconcile this contradiction through internal nano-reinforcement and external molecular lubrication. Crystalline disc-like Laponite nanosheets are intercalated within GelMA networks, acting as dynamic, spatially confining crosslinkers that inhibit water infiltration and preserve network cohesion. Concurrently, zwitterionic brushes are grafted onto the microsphere surface, forming a robust hydration layer via dynamic charge–dipole interactions to enable long-lasting lubrication. This synergistic design endows the microspheres with tunable elasticity (14–4000 Pa) and adjustable friction coefficients (0.12–0.04), achieving a functional convergence of mechanical resilience and surface lubricity. Experimental evaluations confirm their efficacy in inhibiting excessive mechanical stress-induced calcium ion influx and downstream calcium signaling to prevent chondrocyte damage. This work offers a universal strategy to overcome the elasticity–lubrication paradox in hydrogels, unlocking their potential in biomedical engineering, drug delivery, and soft robotic interfaces.

Traditional fluorescent materials for information encryption have drawbacks such as low resolution and cumbersome process of information loading. We have developed an α-amylase-stimulated responsive fluorescent hydrogel system, which was used to store information with arbitrary architectures through combining 3D printing technology. By controlling the concentration of α-amylase to tune the fluorescence of the hydrogel, various types of information on the hydrogel at specific times can be obtained. Multiple dynamic encryptions of information can be easily performed, during which false information may be generated as a means to further improve the security of information storage. Moreover, displaying information from a two-dimensional to a three-dimensional plane of this responsive fluorescent hydrogel broadens the method of information storage and encryption. The introduction of a multicolor system and 3D printing of bacteria that secrete α-amylase in situ will provide a new design strategy for information encryption design.

Chronic wound management presents formidable clinical challenges due to compromised skin barrier function and pathological microenvironment characterized by persistent microbial infection, oxidative stress imbalance, and dysregulated inflammation. Traditional interventions inadequately address the need for real-time monitoring and multi-modal therapeutic integration. Here, a dual-layer multifunctional bioelectronic microneedle patch (DMB-MN) engineered with synergistic functionalities including localized photothermal antibacterial action is developed, intelligent thermoresponsive drug release, electrical stimulation, and pH-actuated closed-loop wound diagnostics. The photothermal platform achieves targeted hyperthermia (50.4 °C) under near-infrared irradiation, demonstrating exceptional antimicrobial performance with 99.88% Staphylococcus aureus eradication and accelerated wound closure (99.78% contraction in 14 days). Mechanistic studies reveal the therapeutic triad's profound healing effects through oxidative stress mitigation (72.66% DPPH radical scavenging), collagen matrix restructuring, and vascular regeneration alongside inflammatory pathway modulation (39.51% TNF-α reduction). Integrated biosensing components enable real-time wound pH tracking with 86.650 mV decade −1 sensitivity, while maintaining biosafety parameters. This bioelectronic-pharmacological DMB-MN platform establishes an advantageous “diagnose-actuate” therapeutic paradigm, synergistically combining critical chronic wound management modalities: antibacterial action, oxidative stress resolution, immunomodulation, and tissue regeneration guidance, which represents a transformative advance in precision chronic wound management through its closed-loop monitoring-treatment integration and bioelectronic control of healing microenvironments.

Vasovasostomy is a surgical procedure aimed at restoring male fertility, whereas low anastomotic efficiency, delayed healing, and postoperative stenosis are key problems encountered during vas deferens recanalization. Here, a directly coaxial-printable and robust PCB hydrogel stent is designed to enhance the effectiveness of vasovasostomy by photo-polymerizing host–guest complexations between poly(ethylene glycol) methyl ether acrylate and α-cyclodextrin acrylate, using N, N’-bis(acryloyl)cystamine as a degradable crosslinker. The PCB hydrogel exhibits continuous micro-stent fabrication capabilities and exceptional anti-deformation mechanical properties (elastic modulus, fracture strength, and toughness of 1.3 MPa, 2.5 MPa, and 480.4 kJ·m −3 , respectively) due to the formation of a polypseudorotaxane threading network integrated with nano-self-assembled structures. Rabbit microsurgical vasovasostomy experiments demonstrate that the time required for tissue anastomosis is reduced by more than half with the support of the PCB hydrogel. Moreover, the stent's excellent resistance to cell adhesion and complete degradability within 28 days in vivo effectively prevents stenosis and promotes healing of the vas deferens, achieving a 100% patency rate. This high-performance PCB stent presents a promising solution for effective vas deferens-related treatment and various other applications.

Current alveolar ridge preservation (ARP) materials face unresolved trade-offs between mechanical stability, bioactivity, and clinical operability. To address this, we developed a fish-derived methacrylated gelatin (FGelMA) hydrogel composited with magnesium silicate (MS) microparticles combining the low immunogenicity of FGelMA with the dual osteo-angiogenic potential of MS. To characterize the physical properties of this material, the composite hydrogels (MS/FGelMA) were tested using a mechanical tester and a rheometer, and then its biocompatibility and in vitro osteogenic properties were analyzed using bone marrow mesenchymal stem cells (BMSCs) in a three-dimensional environment. In vivo model was further established to evaluate the effect of MS/FGelMA on ARP in SD rats. The results indicated that MS/FGelMA hydrogels exhibited rapid crosslinking within 20 s (365 nm UV, 10 mW/cm2), excellent shear-thinning behavior enabled precise defect adaptation, enhanced mechanical robustness, improved osteogenesis and angiogenesis capacity, especially for the optimized 1%MS/15%FGelMA formulation. 1%MS/15%FGelMA had compressive strength of 231 ± 10.149 kPa (378.69% of pure 15%FGelMA), and 2.3–4.1 folds upregulation of osteogenic markers (RUNX2/ALP/OCN) and angiogenic marker (VEGF) in rat BMSCs cultured in 3D hydrogels compared with that in pristine FGelMA hydrogel. Micro-CT analysis revealed 1%MS/15%FGelMA had socket volume preservation of 61% (vs. 46% in controls) at 3 weeks and had bone density of 75% (vs. 62% in controls) at 6 weeks. In general, this species-independent, chairside-applicable platform demonstrates superior clinical translation potential for complex ARP scenarios.

Hydrogel-based cartilage tissue engineering (HCTE) offers a promising strategy for the regeneration of articular cartilage. However, the effectiveness of current HCTE is hindered by the low efficiency of stem cell transplantation and insufficient chondrogenic differentiation. Here, we develop a mechanically optimized and biochemically sustained biomaterial system that integrates a dynamic hydrazone/covalent dual-crosslinked hyaluronan (HA) hydrogel with kartogenin (KGN)-loaded poly(lactic- co -glycolic acid) microspheres to facilitate stem cell chondrogenesis and repair articular cartilage. This composite hydrogel features excellent injectability and self-healing behavior, allowing for minimally invasive implantation while protecting cell viability. The tunable viscoelasticity of the hydrogel enhances focal adhesion formation in bone-marrow mesenchymal stem cells (BMSCs), promoting their spreading, proliferation, and aggregation. Furthermore, the combined effects of hydrogel viscoelasticity and sustained KGN release markedly promote the expression of cartilage-specific genes and the secretion of extracellular matrix components through the integrin-mediated Hippo signaling pathway and KGN-mediated TGF-β signaling pathway. This dual-stimuli hydrogel embedded with BMSCs significantly promotes the regeneration of articular cartilage in rabbit models. Overall, the designed KGN composite dynamic HA-based hydrogel enables highly effective transplantation and promotes efficient chondrogenesis of stem cells, providing a promising approach for stem cell therapy and cartilage regeneration.

Dysregulation of extracellular matrix (ECM) homeostasis plays a pivotal role in the accelerated degradation of cartilage, presenting a notable challenge for effective osteoarthritis (OA) treatment and cartilage regeneration. In this study, we introduced an injectable hydrogel based on streamlined-zinc oxide (ZnO), which is responsive to matrix metallopeptidase (MMP), for the delivery of miR-17-5p. This approach aimed to address cartilage damage by regulating ECM homeostasis. The ZnO/miR-17-5p composite functions by releasing zinc ions to attract native bone marrow mesenchymal stem cells, thereby fostering ECM synthesis through the proliferation of new chondrocytes. Concurrently, sustained delivery of miR-17-5p targets enzymes responsible for matrix degradation, thereby mitigating the catabolic process. Notably, the unique structure of the streamlined ZnO nanoparticles is distinct from their conventional spherical counterparts, which not only optimizes the rheological and mechanical properties of the hydrogels, but also enhances the efficiency of miR-17-5p transfection. Our male rat model demonstrated that the combination of streamlined ZnO, MMP-responsive hydrogels, and miRNA-based therapy effectively managed the equilibrium between catabolism and anabolism within the ECM, presenting a fresh perspective in the realm of OA treatment.

Excessive intracellular iron accumulation can induce mitochondrial dysfunction, leading to chondrocyte ferroptosis, a key contributor to cartilage damage in osteoarthritis (OA). Here, micelle-microfluidic hydrogel microspheres, featuring keto-enol-thiol bridged nano-sized secondary structures that disintegrate within the intracellular peroxidative environment to reveal β-diketone groups with metal chelation capabilities, are utilized for the in situ removal of reactive iron, thereby facilitating cartilage repair through the restoration of mitochondrial homeostasis. The relevant experiments demonstrate that the microspheres reduce iron influx by downregulating transferrin receptor (TfR1) expression and decrease mitochondrial iron uptake by upregulating mitochondrial outer membrane iron-sulfur cluster protein (CISD1), thus restoring intracellular mitochondrial iron homeostasis. Furthermore, the antioxidant properties of the ketone-thioether segments synergistically mitigate chondrocyte phospholipid peroxidation via Nrf2/SLC7A11/GPX4 axis, inhibiting ferroptosis and slowing OA progression. In summary, this system that in situ sustainably chelates reactive iron via metal coordination exhibits great potential in the minimally invasive treatment of OA and other ferroptosis-mediated diseases.

Heart valve organoids hold promise for modeling human valve biology, yet existing platforms often lack human-relevant cell sources, preserved multicellular architecture, and physiologic mechanical cues, limiting their capacity to recapitulate extracellular matrix (ECM) formation and structural maturation. Here, a strategy is established that integrates human induced pluripotent stem cell-derived valve interstitial cells (hiPS-VICs), spheroid organization, bio-printable hydrogels, and controlled mechanical conditioning to engineer advanced valve organoids. 3D bio-printing of hiPS-VIC spheroids is conducted within GelMA/ChSMA constructs, systematically compares spheroid-based bio-printing (SBB) with single-cell bio-printing (SCB), and establishes mechanically-stimulated spheroid-based bio-printing (MSSB) to guide ECM remodeling. Spheroid bioprinting preserved cell–cell organization and improved post-print structural integrity, resulting in substantially enhanced ECM deposition and assembly over time when compared with SCB. Superimposing physiologic loading further promoted cellular spreading and ECM deposition, accompanied by transcriptional signatures associated with matrix production, matrix stabilization, and mechano-responsive pathways. Together, this spheroid-based, mechanically matured, human iPSC valve organoid platform provides a robust and physiologically relevant system that supports ECM development and structural organization beyond dissociated-cell approaches, offering a scalable foundation for human valve biology research, disease modeling, and translational applications.

Shape memory polymers (SMPs) are pivotal for applications in soft robotics, biomedical devices, and flexible electronics. However, their reliance on external stimuli for activation and the difficulty of precisely programming recovery dynamics remain significant limitations. Autonomous SMPs with delayed recovery offer a promising alternative, yet facile control of recovery onset and duration is still difficult to attain. Here, a semi-interpenetrating network (semi-IPN) hydrogel composed of poly(acrylic acid) and silk fibroin (PAA-SF) is presented, fabricated via a facile one-step UV polymerization. By modulating the SF content within the PAA network, the hydrogel exhibits tunable shape recovery kinetics across multiple programming environments, including organic solvents, air, and water, enabling spatiotemporal control over shape restoration. Moreover, the hydrogels show significantly enhanced mechanical properties and strong adhesion compared to neat PAA hydrogels. Benefiting from these combined properties and excellent cytocompatibility, demonstrating two proof-of-concept devices: a smart four-jaw gripper capable of retrieving objects in confined spaces, and a stiffness-switchable needle that reversibly transitions between soft and rigid states. This work not only introduces a robust and multifunctional shape memory hydrogel but also provides a versatile platform for designing stimulus-free, time-programmable soft actuators for biomedical and robotic applications.
