Conditioned Media from LPS-Activated Human Adipose-Derived Mesenchymal Stem Cells Enhances Bone Regeneration in Type 2 Diabetic Rats

Li Yu,Zhang Zhanzhao

Type 2 diabetes mellitus disrupts bone metabolism and impairs bone regeneration, making the bone repair in patients with type 2 diabetes mellitus become a great challenge. The aim of this study is to investigate the effect of conditioned media from LPS-activated human adipose-derived mesenchymal stem cells (LPS-CM) on bone regeneration in type 2 diabetic rats. Type 2 diabetic rats were successfully induced via a high-fat diet and low-dose streptozotocin intraperitoneal injection. After treatment with LPS-CM, the proliferation and migration of bone marrow-derived mesenchymal stem cells from type 2 diabetic rats (BMSCs) were significantly enhanced. In vitro osteogenic induction experiment showed that BMSCs treated with LPS-CM displayed increased ALP activity and calcium deposition. The beneficial effects of LPS-CM on osteogenic differentiation were also confirmed by elevated expression of osteogenesis-related genes including OCN, OPN, RUNX2, and Osx. Furthermore, in vivo study using a rat calvarial bone defect model revealed that LPS-CM had a better osteogenic capacity than conditioned media from unstimulated mesenchymal stem cells. These findings demonstrated that LPS-CM can enhance bone regeneration in type 2 diabetic rats by promoting the proliferation, migration and osteogenic differentiation of BMSCs, suggesting that LPS-CM may provide a promising route for bone repair in patients with type 2 diabetes mellitus.

Eriocitrin in combination with resveratrol ameliorates LPS-induced inflammation in RAW264.7 cells and relieves TPA-induced mouse ear edema

Junlei Liu,Huarong Huang,Zebin Huang,Yuran Ma,Lanyue Zhang,Yan He,Dongli Li,Wenfeng Liu,Susan Goodin,Kun Zhang,Xi Zheng

Eriocitrin is a flavonoid that is isolated from orange peel. Resveratrol is a polyphenol compound, which is present in various fruits, such as grapes. The aim of the present study was to investigate both in vitro and in vivo anti-inflammatory effects of eriocitrin combined with resveratrol using lipopolysaccharide (LPS)-induced RAW264.7 cells and a mouse model of ear edema. The results showed that eriocitrin combined with resveratrol strongly inhibited LPS-induced secretion of nitric oxide (NO), tumor necrosis factor-α (TNF-α), and Interleukin-1β (IL-1β). Moreover, Eriocitrin combined with resveratrol potently inhibited nuclear factor-κB (NF-κB), phosphor-STAT3, and phosphor-AKT, which was accompanied by inhibition of phosphorylation in mitogen-activated protein kinase (MAPK) signaling pathways . Treatment with eriocitrin combined with resveratrol alleviated edema and subcutaneous tissue inflammation caused by 12-O-tetradecanoylphorbol-13-acetate (TPA) in vivo . This study also demonstrated that treatment with eriocitrin and resveratrol decreased the levels of the pro-inflammatory cytokines TNF-α and IL-1β. The results of the present study indicate that eriocitrin combined with resveratrol effectively inhibits inflammatory responses both in vitro and in vivo .

CLG from Hemp Seed Inhibits LPS-Stimulated Neuroinflammation in BV2 Microglia by Regulating NF-κB and Nrf-2 Pathways

Shanshan Wang,Qian Luo,Yuefang Zhou,Peihong Fan

The healthy benefits of hemp (Cannabis sativa L.) seed have often been attributed to its oils and proteins. Recent studies reveal that hemp seed phenylpropionamides could also show various bioactivities. Continuation of our study on hemp seed provided a phenylpropionamide, coumaroylaminobutanol glucopyranoside (CLG). This work investigated the neuroprotective effect of CLG and its underlying mechanism using lipopolysaccharide-induced BV2 microglia. Our study demonstrated that CLG increased adenosine monophosphate-activated protein kinase (AMPK) expression, suppressed the nuclear factor-kappa B (NF-κB) signaling pathway by inhibiting the phosphorylation of IκBα and NF-κB p65 and decreased proinflammatory cytokine levels in a concentration-dependent manner. Furthermore, CLG reduced the production of cellular reactive oxygen species and stimulated the nuclear factor erythroid 2-related factor 2 (Nrf-2) signaling pathway. Collectively, these results suggested that CLG effectively and simultaneously inhibited inflammatory responses and oxidative stress through the NF-κB and Nrf-2 signaling pathways. AMPK was also involved in the anti-inflammatory effect of CLG. This study provides new insights into the diverse bioactive constituents of hemp seed.

Design, synthesis and bioactivity evaluation of fisetin derivatives as potential anti-inflammatory agents against LPS-induced acute lung injury

Xiemin Wang,Jun Yang,Bingyu Ding,Pan Chen,Zhengwei Xu,Yunxi Zhao,Pengqin Chen,Nipon Chattipakorn,Guang Liang,Di Wu,Qidong Tang

Acute lung injury (ALI) refers to a common and life-threatening disease attributed to inflammation. However, effective drug treatments have been rare for ALI. Natural products have been considered as a vital source of drug discovery which indicates that it’s a workable method to find new anti-inflammatory drugs in natural products. Inspired by the various biological activities of fisetin , we reported the design and synthesis of a series of fisetin derivatives which were also evaluated for their anti-inflammatory activities in J774A.1 macrophages. Most of the obtain derivatives could effectively inhibit the release of IL-6 and TNF-α in vitro experiments without cytotoxicity. The most promising compound 5b exhibited significant in vivo anti-inflammatory activity in the model of LPS-induced ALI in mice. On the whole, this study could provide novel candidates for the treatment of ALI.

A high precision apparatus for intracellular thermal response at single-cell level

Wenjuan Tian,Cangling Wang,Jianqing Wang,Qiuhua Chen,Jianfei Sun,Can Li,Xing Wang,Ning Gu

In this work, a nanoprobe that is highly thermo-sensitive to tiny temperature changes was prepared based on a thermocouple metal junction. A series of electro-element apparatuses were integrated to accomplish single-cell temperature measurement. The temperature measurement probe (TMP) was constructed by tungsten (W), polyurethane (PU), and platinum (Pt). The tip size of TMP was characterized at less than 500 nm, and the tip angle was between 10 and 20° with the resistance in the range of 500 to 1500 Ω. The single-cell temperature measurement probes were calibrated and calculated with a Seebeck coefficient ranging from 6 to 8 μV °C−1 at a precision of 0.1 °C. Monitoring the temperature at a single-cell level by inserting the TMP in marine lung epithelia (MLE)-12 cells displayed that the stimulation of lipopolysaccharide (LPS) and cobalt chloride induced different single-cell temperature fluctuation. This investigation could help reveal complex cellular functions and develop novel diagnoses.

Prenylated benzenepropanoic acid analogues from the Citrus grandis (L.) Osbeck and their anti-neuroinflammatory activity

Jingwen Li,Menglong Duan,Xinsheng Yao,Danmei Tian,Jinshan Tang

Phytochemical studies of the air-dried pericarp of Citrus grandis led to the isolation of four new compounds including three prenylated benzenepropanoic acids ( 2 , 3 and 5 ) and one alkamidic glycoside ( 6 ), together with ten known compounds ( 1 , 4 and 7 – 14 ). The structures of these compounds were determined by the NMR spectroscopy, optical rotation data and modified Mosher's method. Meanwhile, the anti-neuroinflammatory activities of all isolated compounds were evaluated by detecting the production of nitric oxide (NO) in LPS-stimulated BV2 cells. The results showed that compounds 1 , 2 , 5 and 13 exhibited strong inhibition effects on NO production in LPS-stimulated BV2 cells. Mechanistically, compounds 1 , 2 and 5 could suppress the expressions of iNOS. In addition, compounds 1 , 2 and 5 also showed obvious inhibition effects on COX-2 expression, another vital enzyme in the inflammation process, in LPS-treated BV2 cells. These findings shed light on the potent anti-neuroinflammatory effects of Citrus grandis .

Effect of RUNX1/FOXP3 axis on apoptosis of T and B lymphocytes and immunosuppression in sepsis

Yangfa Chao,Wenting Huang,Zhiheng Xu,Ping Li,Shaodong Gu

Lymphocyte apoptosis is a latent factor for immunosuppression in sepsis. Forkhead box protein P3 (FOXP3) can interact with RUNX family transcription factor 1 (RUNX1) in regulatory T cells. Our research was to probe whether RUNX1/FOXP3 axis affects immunosuppression in the process of sepsis by modulating T and B lymphocyte apoptosis. We constructed sepsis model in mice and mouse CD4 + T and CD19 + B lymphocytes. RUNX1 and FOXP3 expressions and apoptosis in cells were assessed by western blot, quantitative real-time PCR, and flow cytometer. Inflammation of serum and pathological damage was assessed by ELISA and H&E staining. Relationship between RUNX1 and FOXP3 was assessed by co-immunoprecipitation. The findings showed that RUNX1 ameliorated the survival rate, pathological damage, and decreased inflammation-related factors, and inhibited apoptosis of CD4 + T and CD19 + B cells in cecal ligation and puncture mice. Furthermore, RUNX1 up-regulated the viability and down-regulated apoptotic rate with the changed expressions of apoptosis-related molecules in lipopolysaccharide (LPS)-mediated CD4 + T and CD19 + B cells. Additionally, FOXP3 interacted with RUNX1, and its silencing decreased RUNX1 expression and reversed the inhibitory effect of RUNX1 on apoptosis of LPS-mediated CD4 + T and CD19 + B cells. In summary, the RUNX1/FOXP3 axis alleviated immunosuppression in sepsis progression by weakening T and B lymphocyte apoptosis.

Echinacea polysaccharide alleviates LPS-induced lung injury via inhibiting inflammation, apoptosis and activation of the TLR4/NF-κB signal pathway

Haihua Zhang,Wuying Lang,Shiyong Wang,Binru Li,Guangyu Li,Qiumei Shi

Lung injury is a common critical life-threatening syndrome. Inflammation is a key factor in the pathogenesis of lung injury. It is reported that Echinacea Polysaccharides (EP) has anti-inflammatory activity. However, the effect of EP on lung injury remains unclear. In our study, murine model of lung injury was induced with 2.5 mg/kg LPS before administration of 5 mg/kg or 10 mg/kg EP. EP ameliorated LPS-induced lung pathological damage, along with reduction in lung wet/dry weight ratio and myeloperoxidase activity. EP decreased the number of leukocytes, eosinophils, neutrophils, lymphocytes and macrophages in bronchoalveolar lavage fluid, and the release of tumour necrosis factor-α (TNF-α), interleukin-6 (IL-6) and interleukin-1β (IL-1β) in LPS-treated lung. EP suppressed LPS-induced apoptosis along with down-regulation of Bcl2-associated X (Bax) and cleaved caspase-3 (CC3), and elevated B-cell lymphoma-2 (Bcl-2). Besides, RAW 264.7 cells were treated with EP 100 μg/ml for 1 h and then incubated with 1 μg/ml LPS for 24 h. TNF-α, IL-6 and IL-1β levels were lowered by treatment of EP in LPS-treated RAW 264.7 cells. Moreover, EP down-regulated the expression of toll-like receptor 4 (TLR4), myeloid differentiating factor 88 (MyD88), p-IκBα, nuclear factor kappa-B (NF-κB), p-NF-κB, and up-regulated the inhibitor of NF-κB (IκBα) in vivo and in vitro following LPS induction, which is consistent with the effect of TAK-242. In conclusion, EP may alleviate LPS-induced lung injury via inhibiting inflammation, apoptosis and activation of TLR4/NF-κB signal pathway.

Digestion characteristics of yeast protein and identification of bioactive peptides with cytoprotective effects against LPS-induced injury in C2C12 myoblasts

Xin Cao,Hongjuan Liu,Miao Yang,Zhixian Chen,Qian Cheng,Jialin Sun,Xudong Wu,Yan Zhang,Liping Hao

Yeast protein (YP) is a sustainable alternative protein, offering a favorable amino acid profile. However, its digestion kinetics and potential as a source of bioactive peptides (BAPs) remain neglected. This study applied the INFOGEST digestion model to compare in vitro digestion kinetics of YP, whey and soy protein. YP exhibited delayed amino acid release and distinct proteomic features. Analysis of the final intestinal chyme of YP identified 312 unique peptides. In silico screening identified several low-molecular-weight peptides (<1000 Da) with high bioactivity potential and favorable pharmacokinetics. Crucially, cell-based assays revealed that two high-abundance peptides, GDWPLPT and PLPPAPAPK, significantly alleviated lipopolysaccharide (LPS)-induced injury in C2C12 myotubes. These peptides reduced reactive oxygen species (ROS) levels and interleukin-6 (IL-6) secretion, while restoring mitochondrial membrane potential and adenosine triphosphate (ATP) synthesis. Overall, this study provides a comprehensive characterization of the digestion kinetics and peptide profiles of YP. Beyond these findings, the cytoprotective potential of the identified peptides was validated in a muscle injury model, thereby proving YP as a promising source of functional food ingredients for future applications.

Formononetin attenuates LPS-induced neuroinflammation and depressive-like behaviors by regulating the Keap1/Nrf2/HO-1 pathway in the hippocampus

Siqi Quan,Mengyuan Li,Zhouli Yue,Hui Liu,Shuaijun Peng,Roujia Guo,Xiaoyan Fang,Yucheng Li

Neuroinflammation and oxidative stress are major pathogenic drivers of depression, with the Keap1/Nrf2/HO-1 pathway serving as a key regulator of both processes. Formononetin (FMN) is a naturally occurring isoflavone with anti-inflammatory and antioxidant activities; however, its potential to exert antidepressant effects through activation of this pathway has not been investigated. In this study, lipopolysaccharide (LPS, 1 mg/kg, i.p.) was used to induce neuroinflammation and depressive-like behaviors in mice, and FMN (20 or 40 mg/kg) was administered for seven days prior to LPS challenge. FMN dose-dependently alleviated LPS-induced behavioral deficits, including reduced sucrose preference, decreased locomotor activity, and prolonged immobility in the tail suspension test. Hematoxylin-eosin (HE) and Nissl staining revealed that FMN markedly attenuated LPS-induced hippocampal neuronal damage. FMN also inhibited microglial activation, as evidenced by reduced Iba-1 expression, decreased NF-κB phosphorylation, and downregulation of both mRNA and protein levels of pro-inflammatory cytokines, including IL-1β, TNF-α, and IL-6. Furthermore, LPS-induced lipid peroxidation was significantly attenuated, as indicated by lower levels of 4-hydroxynonenal (4-HNE). Molecular docking suggested a potential direct interaction between FMN and Keap1, while Western blot analysis confirmed that FMN downregulated Keap1 and upregulated Nrf2 and HO-1 protein expression. Collectively, these findings demonstrate that FMN alleviates LPS-induced neuroinflammation and depressive-like behaviors, likely through activating the Keap1/Nrf2/HO-1 pathway, supporting its potential as a preventive candidate for inflammation-associated depression.

Sophoricoside ameliorates LPS/D-GalN-induced acute liver failure by inhibiting ferroptosis via activation of the Nrf2/GPX4 signaling pathway

Shaowei Zheng,Ya Gao,Houkang Cao,Kai Guo,Wenxiu Wang,Guiying Lu,Shasha Wang,Mingli Zhong,Bo Li,Kefeng Zhang,Riming Wei,Jianzhao Wu

Background Acute liver failure (ALF) is associated with ferroptosis and currently lacks specific drugs and therapies. Previous studies have shown the hepatoprotective activity of Sophoricoside (SOP) in diverse models of hepatic injury. However, the role of SOP in ALF caused by lipopolysaccharide (LPS)/D-galactosamine (D-GalN) is still unknown. Purpose To investigate the therapeutic effects of SOP on ALF and its underlying mechanisms of action. Methods In vivo , an ALF mouse model was established using LPS and D-GalN. RNA sequencing was used to identify differentially expressed genes, which were subsequently confirmed by Western blot and qRT-PCR. In vitro , erastin was used to induce ferroptosis in HepG2 cells. Subsequently, the cells were treated with SOP, the nuclear factor erythroid 2-related factor 2 (Nrf2) inhibitor ML385, and the glutathione peroxidase 4 (GPX4) inhibitor RSL3. Additionally, siRNA was utilized for Nrf2 knockdown. Measurements of biochemical markers, reactive oxygen species (ROS) levels, alterations in mitochondrial membrane potential, qRT-PCR, and Western blot analysis were used to confirm the effectiveness and mechanism of SOP. Finally, the interaction between SOP and Nrf2 was verified through molecular docking, surface plasmon resonance (SPR), and cellular thermal shift assay (CETSA). Results In vivo , experimental analysis revealed that SOP ameliorated liver injury, hepatocyte Fe 2+ accumulation, and oxidative stress. RNA sequencing analysis indicated that SOP reversed the abnormal expression of genes related to Nrf2 and the ferroptosis pathway. In vitro , SOP ameliorated erastin-induced oxidative stress, ROS accumulation, and mitochondrial membrane potential loss. After the application of pharmacological inhibitors and siRNA knockdown, the SOP’s inhibitory effect on ferroptosis was antagonized. Conclusion SOP alleviated ALF by activating the Nrf2/GPX4 signaling pathway, thereby mitigating oxidative stress and ferroptosis.

A study on Glycyrrhiza uralensis polysaccharide: The structure elucidation, anti-inflammation effects and treatment mechanism of LPS-induced pneumonia mice

Yue Yu,Wenqing Jiang,Shouhai Chen,Wen Yin,Ping Hao,Ziye Zhu,Muhammad Shahid Mahmood,Rao Zahid Abbas,Khalid Mehmood,Qian Zhou,Yuecheng Liu,Weijie Qu,Ning Li,Xiaonan Yang,Yi Wu

Acute lung injury (ALI) is a life-threatening inflammatory pulmonary disorder characterised by diffuse alveolar damage, pulmonary oedema, and excessive immune responses. This study aimed to investigate the therapeutic potential of a purified polysaccharide, Glycyrrhiza polysaccharide fraction 2 (GCP-2), derived from Glycyrrhiza uralensis (liquorice), in a murine model of LPS-induced ALI. GCP-2 was obtained through a multi-step purification process involving aqueous extraction, ethanol precipitation, ion-exchange chromatography, and size-exclusion chromatography. ALI was induced in mice via intranasal administration of lipopolysaccharide (LPS), followed by GCP-2 treatment. Key outcomes were assessed using ELISA, qPCR, histopathology, and western blotting, while gut microbiota changes were evaluated by 16S rRNA sequencing. The results demonstrated that GCP-2 significantly alleviated lung injury, reduced pulmonary and systemic levels of pro-inflammatory cytokines, restored barrier-related proteins such as Zonula occludens-1 (ZO-1) and occludin, and improved histopathological features. Mechanistic studies revealed that these effects were mediated through the selective inhibition of the TLR4/NF-κB signalling pathway. Additionally, GCP-2 modulated gut microbiota composition by increasing beneficial taxa such as Bacteroidetes , Lactobacillus , and Bifidobacteria , while suppressing Firmicutes and Proteobacteria . These findings suggest that GCP-2 is a safe and effective plant-derived polysaccharide that mitigates LPS-induced ALI through anti-inflammatory and gut–lung axis regulatory mechanisms, supporting its potential as a therapeutic candidate for treating inflammatory lung diseases.
