Dynamic culturing of large cell-loaded PCL/gelatin methacryloyl scaffolds for bone critical size defect repair

Guangxin Shen,Botao Gao,Jiayi Guo,Weikang Xu,Guangfu Chen,Shuai Huang,Zhiwen Zeng,Xiaodong Zhao

Due to the limited ability to self-repair, the regeneration of bone critical-sized defects (CSD) is a significant challenge. Bone tissue engineering scaffolds are considered promising candidates for CSD repair, but low cell infiltration efficiency and a lack of nutrients greatly restrict bone regeneration abilities. Herein, we developed a dynamic culturing of large biomimetic bone scaffolds, PCL/GelMA@cells that combining 3D printed polycaprolactone (PCL) multi-channel cylinder with gelatin methacryloyl (GelMA) encapsulated with bone marrow mesenchymal stem cells (BMSCs) and rat aortic endothelial cells (RAECs). A cell dynamic culture system was fabricated to simulate the dynamic microenvironment. Compared to static culturing, dynamic culturing proved to enhance the nutrient exchange within the large scaffold to promote the cells infiltration, growth, proliferation and induce osteogenic and angiogenic differentiation. Furthermore, a rat cranial CSD (D = 10 mm) repair model verified the accelerated vascular ingrowth and new bone formation with the implantation of dynamic culturing of PCL/GelMA@cells scaffold (∼10 times higher than Blank group), indicating the great potential of dynamical culturing of scaffolds for bone repair. In summary, the results highlight the significant advantages of the dynamical culturing of cell-loaded scaffolds for bone regeneration, offering a promising strategy for addressing critical size bone defects.

Fracture toughness and bonding strengths of epoxy composites containing POSS-PCLn: An impact of phase separation from macro-to nano-scales

Gaoming Li,Yeping Wu,Guohua Hang,Xibing Shen,Zhongtao Chen,Mao Chen,Xiuli Zhao,Yinyu Zhang,Sixun Zheng

Epoxy thermosets have been widely used in engineering applications due to their excellent thermal stability and chemical resistance. However, their inherent brittleness causes low bonding strength. Therefore, improvement of toughness is required. By using a variety of modifiers, epoxy thermosets can be toughened via the generation of fine phase-separated morphologies. In this work, we reported the toughening of epoxy thermosets with polyhedral oligomeric silsesquioxane (POSS)-capped poly(ε-caprolactone)s (PCL) . By taking control of types of R groups ( i.e., isobutyl and/or isooctyl) of POSS cages and the lengths of PCL chains, the epoxy thermosets were regulated with the morphologies from nano-to macro-scales. It was found that the improvement of toughness was quite dependent on types of R groups of POSS cages. Under the identical condition, the thermosets contain isooctyl POSS-PCL n had the fracture toughened larger than those containing isobutyl POSS groups. For the thermoset containing 15 wt% of isooctyl POSS-PCL 1.00 , the critical stress intensity factor ( K IC ) was enhanced from 0.70 to 1.69 MPa m 1/2  at room temperature and from 1.11 to 2.51 MPa m 1/2  at low temperature and the thermal stability remained invariant. Furthermore, the bonding strength on stainless steel and anodized aluminum substrates were significantly improved with the incorporation of 15 wt% of isooctyl POSS-PCL 1.00 . These findings provide a novel approach to regulate phase structure in epoxy thermosets, offering the insights into structure-performance relationships, thus opening up a new way for designing epoxy resins with high toughness and adhesive strength.

High-voltage resistance of PCL/PEO blending polymer-based solid electrolyte for high-temperature lithium metal batteries

Xiong Zhen-Yu,Wang Guan-Hua,Wang Hong-Yu,Li Peng-Yuan,Sun Yang,Ma Yu-Han,Guo Tao,Shi Fa-Nian,Zhang Yu-Hang

Polyethylene oxide (PEO) is regarded as the most promising candidate for the next generation solid polymer lithium metal batteries (LMBs). However, it suffers from low mechanical strength and limited electrochemical stability window (ESW), which restricts its application in high energy density LMBs. In this work, polycaprolactone (PCL) is chosen as the main matrix to fabricate a blending polymer-based electrolyte with PEO, which shows an improved electrochemical performance with a high lithium ion transference number of 0.65. The improvement can be attributed to the competition between ester and ether groups of different polymer chains, which can both interact with lithium ions to release more free cations. Moreover, the high-voltage resistance of PCL can broaden the electrochemical stability window to 4.8 V of PCL/PEO solid electrolyte to match LiNi 0.8 Co 0.1 Mn 0.1 O 2 (NCM811) cathode, and its high-temperature resistance ensure a satisfying dimensional stability for the assembled NCM811||Li cell to operate at 55 ℃, with a high initial specific capacity of 210 mAh/g and a retention of 95% within 100 cycles. Compared to other blending systems, the PCL/PEO blend exhibits superior compatibility, mechanical properties, and electrochemical stability, offering a promising strategy for the commercialization of high-voltage, safe, and high-energy-density solid-state lithium metal batteries operable at elevated temperatures.

Schiff base-grafted bacterial cellulose-PCL composites: Humidity/temperature-driven hydrogen bond reconfiguration for mechanically adaptive nanofiber

Chenchen Qin,Bianjing Sun,Xinyu Chen,Du Ping,Wuxin Bai,Lin Shi,Ahsan Nazir,Chuntao Chen,Dongping Sun

A novel graft copolymer BC-g-PCL was synthesized and characterized by integrating bacterial cellulose (BC) with polycaprolactone (PCL) to improve mechanical performance and sustainability. Molecular dynamics simulations reveal that hydrogen bonding is crucial for improving the strength and toughness of the material, with temperature fluctuations affecting the interactions of the polymer chains. The BC-g-PCL exhibits notable water plasticity and solvent responsiveness, enabling reversible transitions between dry and wet states due to the high surface wettability from exposed hydroxyl groups. Mechanical testing demonstrates that BC-g-PCL retains excellent ductility and strength across varying humidity conditions due to the dynamic formation of hydrogen bonds. Furthermore, recyclability and biodegradability assessments indicate substantial stability against organic solvents and successful regeneration via N , N -dimethylacetamide (DMAC)/lithium chloride (LiCl) dissolution, while maintaining robust mechanical properties even after multiple recycling cycles. These results make BC-g-PCL a promising eco-friendly alternative to traditional plastics, highlighting its potential for advanced functional materials and sustainable engineering applications derived from biological resources.

Developing topological engineering PCL-based comb polyurethanes: investigation of the aggregation structure, deformation behavior and optimized properties

Hengyu Yan,Mingjie Zhang,Qing Tang,Chao Long,Hecheng Wu,Ao Wang,Nan He,Wenkai Liu,Zhen Li,Feng Luo,Jiehua Li,Hong Tan

Recent advances reported polyurethanes with hard-segment main chains and soft-segment side chains. However, fundamental understanding of their aggregation structures, deformation mechanisms, and potential applications remained limited. Besides, existing designs were confined to polyether or zwitterion side chains, with polyester-based variants unexplored. Herein, we synthesized a kind of novel comb polyurethanes with polycaprolactone (PCL) sidechains attached to hard backbone (SPPU). Comprehensive characterizations revealed distinct aggregation structures in SPPU compared to linear PCL-based polyurethanes (PPUs) from different aspects. Strain-dependent analysis was first applied to uncover the distinctive deformation mechanism of SPPUs in this study. While PPUs’ deformation was mainly driven by soft-segment elongation, low hard-segment SPPUs underwent whole-chain slippage-dominated deformation (enhancing chain mobility) and high hard-segment SPPUs demonstrated hard-segment elongation-based deformation (reinforcing rigidity). SPPUs exhibited enhanced room-temperature loss modulus and absolute damping capacity compared to PPU. Low hard-segment SPPU exhibited significantly higher loss factor and displayed superior impact protection performance. High hard-segment SPPUs exhibited substantially increased modulus than linear PUs. SPPUs also exhibited improved acid/alkali corrosion-resistance, extending the service life in corrosive environments. Superior melt flowability of SPPUs at high temperature highlighted the topological design in improving processability. The synergy of tunable stiffness, optimized damping and durability positioned SPPUs as transformative materials for optimization of conventional linear PCL-based PUs. This work afforded the first polyester-based comb PU system and crucial molecular design insights for advanced high-performance polyurethane materials.

Acetal-Linked Paclitaxel Polymeric Prodrug Based on Functionalized mPEG-PCL Diblock Polymer for pH-Triggered Drug Delivery

Yinglei Zhai,Xing Zhou,Lina Jia,Chao Ma,Ronghua Song,Yanhao Deng,Xueyao Hu,Wei Sun

The differences in micro-environment between cancer cells and the normal ones offer the possibility to develop stimuli-responsive drug-delivery systems for overcoming the drawbacks in the clinical use of anticancer drugs, such as paclitaxel, doxorubicin, and etc. Hence, we developed a novel endosomal pH-sensitive paclitaxel (PTX) prodrug micelles based on functionalized poly(ethylene glycol)-poly(ε-caprolactone) (mPEG-PCL) diblock polymer with an acid-cleavable acetal (Ace) linkage (mPEG-PCL-Ace-PTX). The mPEG-PCL-Ace-PTX5with a high drug content of 23.5 wt % was self-assembled in phosphate buffer (pH 7.4, 10 mM) into nanosized micelles with an average diameter of 68.5 nm. The in vitro release studies demonstrated that mPEG-PCL-Ace-PTX5micelles was highly pH-sensitive, in which 16.8%, 32.8%, and 48.2% of parent free PTX was released from mPEG-PCL-Ace-PTX5micelles in 48 h at pH 7.4, 6.0, and 5.0, respectively. Thiazolyl Blue Tetrazolium Bromide (MTT) assays suggested that the pH-sensitive PTX prodrug micelles displayed higher therapeutic efficacy against MCF-7 cells compared with free PTX. Therefore, the PTX prodrug micelles with acetal bond may offer a promising strategy for cancer therapy. Keywords:prodrug;polymer micelles;pH-sensitive;acetal;paclitaxel

Rational design of antibacterial and biodegradable PCL/oxidized cassava starch/Nisin ternary films based on molecular docking

Xiangrui Ren,Li Niu,Liping Zhao,Wenqing Zhao,Qian Lin,Shuangfeng Guo,Yayun Hu,Wenhao Li

This study focuses on the rational design of biodegradable active packaging films by combining oxidized cassava starch (OCS), polycaprolactone (PCL), and Nisin via hot-pressing. Systematic characterization demonstrated that OCS incorporation reduced PCL crystallinity through hydrogen bonding, markedly altering film microstructure and functionality, and enhancing biodegradability in synergy with Nisin. Key experimental data revealed that the ternary composite films exhibited superior comprehensive performance: the soil degradation rate reached 15.98% after 72 days, a 316% increase compared to pure PCL. Regarding antimicrobial efficacy, the inhibition rate against Staphylococcus aureus reached 85.79%. Additionally, all composite films maintained low water absorption (<2%), and some formulations exhibited excellent flexibility with a peak elongation at break of 117.98%. Molecular docking further revealed that hydrogen bonds and salt bridges stabilized the structure, with the strongest binding affinity reaching −5.0 kcal/mol, influenced by the oxidation pattern of OCS. This research presents a novel, eco-friendly ternary composite film with tunable degradation and strong antibacterial activity. This work provides a strategic framework for designing next-generation carbohydrate-based active packaging and offers a practical engineering strategy to tailor bio-based films for food preservation applications.

The preparation of PCL/MSO/SiO2 hierarchical superhydrophobic mats for oil-water separation by one-step method

Guohui Zhang,Panpan Wang,Xiaoxiao Zhang,Chunhui Xiang,Lili Li

Hydrophobic polycaprolactone (PCL) membranes with hierarchical structure were fabricated via one-step electrospinning technique. Meanwhile, the addition of the liquid methyl silicone oil (MSO) and hydrophobic silica (SiO 2 ) nanoparticles in the PCL membrane were expected to improve the hydrophobicity of PCL membranes. FE-SEM images showed the microsphere-fiber interlinked hierarchical morphology of PCL/MSO/SiO 2 composite fiber. FT-IR confirmed the formation of PCL/MSO/SiO 2 composite fibers. XPS approved its hierarchical structure of the composition. By comparing with pure PCL membrane which the water contact angle (WCA) was 132.2 ± 1.2°, the WCAs of PCL/MSO and PCL/SiO 2 membrane increased to 143.3 ± 1.2° and 141.7 ± 1.9°, respectively. It was worth noting the WCA of PCL/MSO/SiO 2 composite mat could reach 151.6 ± 1.7° and the sliding angle was 7.4 ± 1.2°. The excellent surface roughness and air-pocket capacity on the hierarchical membranes were believed to contribute to the superhydrophobicity of PCL/MSO/SiO 2 membrane. The unique microsphere-fiber interlinked hierarchical structure and superhydrophobicity of membrane could provide adequate support for oil adsorption and oil-water separation. The maximum n-hexane adsorption capacity of PCL/MSO/SiO 2 membrane was 41.37 g g −1 . Separation efficiency for n-hexane-water mixture of PCL/MSO/SiO 2 membrane was as high as 99.92%. The PCL/MSO/SiO 2 hierarchical superhydrophobic mats indicate the potential applications of environmental-friendly biopolymers as separator.

An asymmetric Janus PCL/chitosan bilayer membrane harmonizing mechanical integrity and Osteo-immunomodulatory bioactivity for periodontal regeneration

Lin Huang,Fengxiao He,Tao Wu,Jinghao Fan,Lei Liu,Xiang Zhang,Jiangchen Liu,Shiqian Huang,Decheng Wu,Waruna Lakmal Dissanayaka

This study introduces a new bioactive Janus membrane for guided bone regeneration (GBR) to treat periodontal bone defects. • Unlike conventional barrier membranes that lack long-term stability or biological function, our bilayer membrane, composed of poly(ε-caprolactone) (PCL), chitosan (CS), and the natural polyphenol proanthocyanidin (PA), combines mechanical strength with antioxidant, antibacterial, and anti-inflammatory properties. • Its asymmetric design, with in situ biomineralized hydroxyapatite particles (HAp) on a chitosan layer, supports bone growth on one side while preventing bacterial invasion on the PA-incorporated side. • This innovative approach addresses major challenges in periodontal tissue engineering and offers a promising platform for more effective and biologically intelligent therapies in clinical bone regeneration. The achievement of periodontal bone regeneration using a barrier membrane in guided bone regeneration (GBR) technology is often limited by either its inferior mechanical strength or bioactivity. To address this limitation, we developed a novel Janus bi-layer membrane by sequentially electrospinning poly(ε-caprolactone) (PCL) mixed with proanthocyanidin (PCL-PA) and chitosan (CS) in acetic acid, then biomineralized it in simulated body fluid (BSF) to form hydroxyapatite particles (HAp) in situ on a CS nanofiber layer (CS-HAp): PCL-PA/CS-HAp. This bi-layer membrane exhibited superior mechanical performance compared to single-layer membranes, with a peeling strength of 58 J/m 2 and tensile strength exceeding 5.0 MPa under hydrated conditions. In vitro antioxidant assays demonstrated the strong free-radical-scavenging ability of PCL-PA/CS-HAp. In addition, the PA-incorporated membranes exhibited greater than 95% bacterial inhibition against both Gram-negative E. coli and Gram-positive S. aureus . The bilayer membrane also alleviated oxidative stress induced by hydrogen peroxide (H 2 O 2 ), thereby promoting the proliferation of MC3T3-E1 cells, as monitored by cytoskeletal staining. Flow cytometry analysis showed that the PA-incorporated membrane can stabilize immunomodulatory effects by about 10% in lipopolysaccharides (LPS)-stimulated RAW264.7 cells. When used in a rat periodontal bone defect model, significant bone regeneration was observed in the PCL-PA/CS-HAp group, with a bone volume to total volume ratio of 36.2% and a significant reduction in bone defect size after 8 weeks, outperforming other treatment groups ( p  < 0.001). The PCL-PA/CS-HAp bi-layer membrane represents a promising strategy for guided bone regeneration, effectively combining mechanical stability and bioactive properties. A bi-layer barrier membrane, PCL-PA/CS-HAp, prepared via electrospinning, was effective for periodontal bone regeneration in a rat model. 3D-printed BaTiO3-modified PCL/β-TCP scaffolds with multimodal optimization for enhanced critical-size bone regeneration

Liu Xuanhao,Zhang Jing,Xu Biying,Xu Yang,Wang Yuxin,Huang Zhi,Liu Bin,Fuxiang Song

Multimodal optimization improved scaffold structure, mechanics, and bioactivity. • Low-dose BT generated a detectable but mild local piezoelectric response. • PTB4 achieved the best overall balance of printability and biological performance. • PTB4 enhanced osteogenic differentiation and VEGF-related endothelial responses. • PTB4 promoted osteogenesis–angiogenesis coupling and calvarial bone regeneration. Critical-size bone defects (CSDs) remain a major clinical challenge. Although three-dimensional (3D) printing enables precise structural control and defect-specific scaffold fabrication, conventional polymer-based scaffolds often exhibit insufficient mechanical strength and limited osteogenic activity. In this study, poly(ε-caprolactone)/β-tricalcium phosphate (PCL/β-TCP) scaffolds containing 0, 1, 2, 4, or 8 wt% barium titanate (BaTiO 3 , BT) were fabricated by direct ink writing. Their rheology, microstructure, mechanical properties, local electromechanical response, cytocompatibility, angiogenesis-related activity, osteogenic differentiation, and bone regeneration in vivo were evaluated. All formulations exhibited suitable printability and regular porous structures. Moderate BT incorporation increased surface roughness and mechanical strength, with compressive strength remaining within the range of human cancellous bone. Piezoresponse force microscopy confirmed a detectable but mild local piezoelectric response. However, its direct osteogenic contribution was not isolated from other BT-related material effects and requires further verification. All scaffolds exhibited good cytocompatibility. PTB4 showed the most favorable biological performance, including enhanced MC3T3-E1 cell proliferation, increased ALP-positive area, greater mineralized deposition, and more pronounced VEGF-related fluorescence in HUVECs. After 12 weeks in a rat calvarial critical-size defect model, PTB4 exhibited significantly higher bone volume fraction and bone mineral density than the blank and PT groups. Histological and immunohistochemical analyses showed more extensive new bone formation, collagen deposition, and stronger osteogenesis–angiogenesis coupling. Overall, PTB4 achieved the best balance among printability, mechanical compatibility, cytocompatibility, angiogenesis-related activity, and osteogenic performance, supporting low-dose BT modification as a promising strategy for safe, printable scaffolds for critical-size craniofacial and oral bone defect repair. ROS-Responsive mPEG–PCL Nanoparticles for Co-Delivery of Paclitaxel Prodrug and R848 in Synergistic Chemo-Immunotherapy

Qi Zhang,Luyao Wang,Zhaofan Yang,Hao Meng,Shixian Lv,Linlin Liu

Chemo-immunotherapy aims to integrate chemotherapy-induced antigen release with innate immune activation to achieve enhanced antitumor efficacy. However, the pharmacokinetic mismatch and systemic toxicity of free small molecules often compromise the synergistic effects. Herein, we propose a reactive oxygen species (ROS)-responsive nanoplatform based on mPEG–PCL for the co-delivery of a paclitaxel (PTX) prodrug and the TLR7/8 agonist R848. The nanoparticles remain stable under physiological conditions while releasing drug in oxidative tumor microenvironments, thereby achieving synchronous intratumoral delivery and activation. In vitro, the system exhibited efficient intracellular uptake and potent cytotoxicity against tumor cells while maintaining low toxicity toward normal cells due to prodrug design. Moreover, the platform effectively induced immunogenic cell death and promoted dendritic cell maturation, highlighting its capacity for chemo-immunotherapeutic synergy. This co-delivery strategy provides a versatile approach to realize controllable and safe chemo-immunotherapy through stimuli-responsive nanomedicine design.

Synthesis of Poly(methyl/amino)silsesquioxane Microsphere as an Efficient Compatibilizer for Enhanced PHB/PCL Blend Properties

Haiying Chen,Cheng Chen,Min Zhou,Xiang Wu,Tao Wang,Lingjun Wei

To enhance the compatibility of polyhydroxybutyrate/polycaprolactone (PHB/PCL) binary blend system, a novel covalent compatibilizer, poly(methyl/amino)silsesquioxane (PMASQ) was synthesized via the base-catalyzed sol-gel method using Methyltrimethoxysilane (MTMS) and γ-Aminopropyl trimethoxysilane (APTMS). The films were prepared through the solution casting method with the blend of PHB/PCL (25:75, w/w) incorporating with various content of PMASQ (1   wt%, 3   wt%, 5   wt%). As a result, ATR-FTIR analysis confirmed the successful synthesis of PMASQ and the subsequent grafting of PHB and PCL segments onto the PMASQ. At 3   wt% PMASQ, the blend exhibited optimal mechanical enhancement, with tensile strength and elongation at break increasing by 1.23-fold and 6.17-fold, respectively, compared to uncompatibilized blends. Furthermore, the incorporation of PMASQ resulted in a reduction in crystallinity while enhancing thermal stability within the PHB/PCL blend system. An improvement in continuity was also observed in the PHB/PCL/PMASQ blends, characterized by a reduction in the size of the dispersed phase and an emergence of a homogeneous morphology. These results demonstrate PMASQ’s dual role as a reactive compatibilizer and thermal stabilizer, mitigating inherent brittleness and phase incompatibility in PHB/PCL systems. The improved physicochemical properties and processability of the blend highlight PMASQ’s potential to expand the utility of biodegradable PHB/PCL composites in packaging, biomedical, and environmentally sustainable industrial applications.
