Injectable microcarriers enhance stem cell delivery and promote corneal regeneration for alkali burn

Jinming Li,Qiaoyu Li,Zhaoxiang Lu,Wenyu Wu,Xiaojun He,Zhou Yang,Yun Feng

Corneal alkali burns represent severe ocular injuries characterized by acute inflammation and long-term stromal fibrosis, potentially causing irreversible vision loss. Stem cell therapy has emerged as a promising treatment due to its regenerative and immunomodulatory properties. However, conventional cell injection methods are limited by poor cell viability, low retention rates, and injection-induced mechanical trauma, significantly impairing therapeutic outcomes. Here, we present collagen-modified chitosan porous microspheres (ColMS) as a cell-laden porous microcarrier system to enhance adipose-derived stem cells (ADSCs) delivery efficiency for corneal repair following alkali burns. The ColMS possess tunable pore sizes (~6–18 μm) and particle diameters (~163–313 μm), providing a three-dimensional scaffold to support ADSC adhesion, proliferation, and enhanced cell viability during injection. In vitro experiments confirmed that ADSC-loaded ColMS (ColMS-ADSCs) exhibits robust immunomodulatory capacity and epithelial regenerative capabilities. Using a rat corneal alkali burn model, subconjunctival injection of ColMS-ADSCs suppressed acute inflammation in the early stage and effectively reduced corneal fibrosis in the late stage, leading to substantial restoration of corneal transparency. The treatment promoted epithelial regeneration, with epithelial thickness restored to near-normal levels and decreased myofibroblast activation, as evidenced by a 1.5-fold reduction in α-SMA fluorescence intensity compared to the alkali burn group. This work demonstrate that ColMS effectively enhance stem cell delivery efficiency, offering a promising clinical strategy for the treatment of corneal alkali burns, potentially opening new avenues toward improved vision restoration.

Curcumin-Loaded Mesenchymal Stem Cell-Derived Exosomes: A Potential Effective Strategy for NAFLD

Ruixi Luo,Jing Zhang,Peng Chen,La Wang,Wenjia Wang,Ping Wang,Xing Zhao,Weiyi Tian

The aim of this study was to construct curcumin-loaded mesenchymal stem cell-derived exosomes (Cur-exos) and explore their effects on nonalcoholic fatty liver disease (NAFLD) models both in vitro and in vivo. Cur-exos were prepared using three common drug-loading techniques—incubation, sonication, and freeze–thaw cycles. Among these, Cur-exos prepared via repeated freeze–thawing demonstrated higher encapsulation efficiency and drug-loading rates than those produced by other methods. In vitro, Cur-exos exhibited enhanced stability, were efficiently taken up by hepatocytes, and mitigated palmitic acid (PA)–induced lipotoxicity in HepG2 cells. Moreover, Cur-exos significantly ameliorated liver damage in NAFLD model mice, reduced inflammation, decreased reactive oxygen species (ROS) levels, and mitigated endoplasmic reticulum (ER) stress. Additionally, Cur-exos effectively regulated lipid metabolism disorders and improved impaired glucose tolerance. Overall, we demonstrate that, compared with free curcumin, Cur-exos significantly improve curcumin stability and offer superior therapeutic effects in NAFLD mouse models. This study provides a novel approach for developing treatments for NAFLD.

Hydroxybutyl chitin combined with bone marrow mesenchymal stem cells promotes diabetic wound healing

Shuo Wang,Tianjiao Xu,Shourui Liu,Ruibao Ju,Jinhua Chi,Zhiwen Jiang,Lixin Xia,Wanshun Liu,Baoqin Han

Diabetic wounds are a common complication of diabetes mellitus, characterized by low healing rates and high recurrence rates, which severely impact patients’ quality of life. Although studies suggest that bone marrow-derived mesenchymal stem cells (BMSCs) possess the potential to promote chronic wound healing, their direct application is limited by low survival rates and impaired functionality. To address this issue, we developed a thermosensitive hydrogel (HBCT hydrogel) for encapsulating/delivering BMSCs by modifying chitin via hydroxybutylation. HBCT can form a hydrogel at physiological temperature and exhibits excellent biocompatibility as well as the ability to enhance cell migration. More importantly, the HBCT hydrogel maintains/sustains the viability of BMSCs. In a rat model of diabetic full-thickness skin defects, the combined application of BMSCs and the HBCT hydrogel improved wound healing efficiency by promoting collagen deposition, enhancing the expression of epidermal growth factor, and increasing markers of neovascularization. This study combines the HBCT thermosensitive hydrogel with BMSCs, offering/providing a promising strategy for tissue regeneration and repair in diabetic wounds.

Preparation of fruit stem preservation column and its application in postharvest grape storage

Youwei Yu,Xinyu Yang,Silong Jia,Qiyuan Feng,Tian Li,Hesheng Wang,Linna Wang,Limin Guo,Baoguo Ji,Shaoying Zhang

Postharvest fruits and vegetables are susceptible to spoilage, which may result in food safety hazards and economic losses. In this study, a stem-insertable preservation column was developed and applied for the preservation of post-harvest grapes. The preservation column incorporates orange essential oil Pickering emulsion as the active component, with natural antimicrobial substances being effectively stabilized through physical emulsification technology. Experimental results showed that the inserted preservation columns enable targeted release of active compounds through the vascular system and achieved significant inhibition against pathogenic microorganisms including Staphylococcus aureus at grape stem junctions. The preservation column treatment reduced grape decay incidence by over 60 % compared to untreated controls. Furthermore, this technology effectively mitigates fruit water loss, decreasing the mass reduction rate by 45 %, while improving nutrient retention - notably reducing vitamin C loss from 70.22 % in controls to 52.20 % in the P-CZ treatment group. These effects collectively maintain optimal sensory characteristics and marketability of grape berries. The developed method exhibits multiple advantages including operational simplicity, absence of chemical residues, and consistent preservation efficacy. These findings confirm that the essential oil-based preservation column technology represents a safe and effective innovation for grape postharvest management.

Amino acid catabolism regulates hematopoietic stem cell proteostasis via a GCN2-eIF2α axis

Changzheng Li,Binghuo Wu,Yishan Li,Jie Chen,Zhitao Ye,Xiaobin Tian,Jin Wang,Xi Xu,Shuai Pan,Yucan Zheng,Xiongwei Cai,Linjia Jiang,Meng Zhao

Summary Hematopoietic stem cells (HSCs) adapt their metabolism to maintenance and proliferation; however, the mechanism remains incompletely understood. Here, we demonstrated that homeostatic HSCs exhibited high amino acid (AA) catabolism to reduce cellular AA levels, which activated the GCN2-eIF2α axis, a protein synthesis inhibitory checkpoint to restrain protein synthesis for maintenance. Furthermore, upon proliferation conditions, HSCs enhanced mitochondrial oxidative phosphorylation (OXPHOS) for higher energy production but decreased AA catabolism to accumulate cellular AAs, which inactivated the GCN2-eIF2α axis to increase protein synthesis and coupled with proteotoxic stress. Importantly, GCN2 deletion impaired HSC function in repopulation and regeneration. Mechanistically, GCN2 maintained proteostasis and inhibited Src-mediated AKT activation to repress mitochondrial OXPHOS in HSCs. Moreover, the glycolytic metabolite, NAD + precursor nicotinamide riboside (NR), accelerated AA catabolism to activate GCN2 and sustain the long-term function of HSCs. Overall, our study uncovered direct links between metabolic alterations and translation control in HSCs during homeostasis and proliferation.

Mechanical Signal-Tailored Hydrogel Microspheres Recruit and Train Stem Cells for Precise Differentiation

Zehao Chen,Zhendong Lv,Yaping Zhuang,Qimanguli Saiding,Wu Yang,Wei Xiong,Zhen Zhang,Hao Chen,Wenguo Cui,Yuhui Zhang

The aberrant mechanical microenvironment in degenerated tissues induces misdirection of cell fate, making it challenging to achieve efficient endogenous regeneration. Herein, a hydrogel microsphere-based synthetic niche with integrated cell recruitment and targeted cell differentiation properties via mechanotransduction was constructed for enhanced endogenous regeneration. Through the incorporation of microfluidics and photo-polymerization strategies, we prepared fibronectin (Fn) modified methacrylated gelatin (GelMA) microspheres with the independently tunable elastic modulus(1-10Kpa) and ECM ligand density (2 and 10 μg/ml), allowing a wide range of cytoskeleton modulation to trigger the corresponding mechanobiological signaling to direct cell fate. The combination of the soft matrix (2Kpa) and low ligand density (2 μg/ml) could support the nucleus pulposus (NP)-like differentiation of intervertebral disc (IVD) progenitor/stem cells by translocating Yes-associated protein (YAP), without the addition of inducible biochemical factors. Meanwhile, platelet-derived growth factor-BB was loaded onto Fn-GelMA microspheres (PDGF@Fn-GelMA) via the heparin-binding domain of Fn and exhibited continuous release for over 28 days to initiate endogenous cell recruitment. In in vivo experiments, hydrogel microsphere-niche maintained the IVD structure and stimulated ECM synthesis, thereby enhancing the endogenous regeneration of NP. Overall, this synthetic niche with cell recruiting and mechanical training capabilities offered a promising novel strategy for endogenous tissue regeneration. This article is protected by copyright. Uniformly sized iron oxide nanoparticles for efficient gene delivery to mesenchymal stem cells

Qianhao Xu,Tianyuan Zhang,Qiyue Wang,Xinchi Jiang,Ai Li,Ying Li,Ting Huang,Fangyuan Li,Ying Hu,Daishun Ling,Jianqing Gao

Recent advances in nanomaterials have made iron oxide nanoparticles (IONPs) as an innovative approach for the delivery of genes. However, the effectiveness of IONPs-assisted gene delivery is currently suffering from their poor uniformity, which not only exhibits detrimental effect on the magnetic property, but also leads to the poor reproducibility to maintain the optimal gene delivery. To this end, the present study developed extremely uniform 15 nm-sized IONPs with a good monodispersity in water phase. These ultra-uniform IONPs exhibit notable potentials for an efficient gene delivery to human mesenchymal stem cells (hMSCs) with the assistance of magnetic force, which partly owes to their abilities to facilitate the cellular uptake, as well as to induce the rapid DNA release and the following nuclear transport. Besides, no significant detrimental effects of these IONPs are observed either on the proliferation or the multi-lineage differentiation potential of hMSCs. The current study highlights the potential advantages of designing extremely uniform magnetic nanomaterials for an efficient and safe delivery of genes to stem cells.

Enhanced Mesenchymal Stem Cells-derived Exosomes Secretion by Electrical Stimulation of Triboelectric Nanogenerator

Shanguo Zhang,Tianyi Jiang,Haoxiu Sun,Ming Li,Depeng Yang,Wenlong Wu,Qi Gu,Aitong Xu,Yu Li,Hongyuan Jiang

Exosomes derived from mesenchymal stem cells (MSCs) exhibit immunomodulatory, tissue repair, anti-inflammatory, and anti-aging properties, making them valuable tools in cell therapy and regenerative medicine. Electrical stimulation shows promise for enhancing exosome production; however, current methods rely on large, expensive power supplies to generate electrical signals. These systems are prone to electrode electrolysis and bubble formation in the buffer solution. In this study, we propose a novel self-powered method based on triboelectric nanogenerators to enhance exosome production from MSCs. This approach utilizes the triboelectric effect to generate an electrical output with an open-circuit voltage of 1800V and a short-circuit current of 88   µA, effectively avoiding harmful electrochemical reactions. By optimizing parameters such as current strength, electrical stimulation duration, and treatment cycles, exosome production is increased by up to 3.2 times. Further investigations reveal that the enhanced exosome production is closely linked to calcium ion influx. Moreover, exosomes produced under electrical stimulation retain their biological activity and significantly promote wound healing. Thus, this self-powered, portable, and efficient electrical stimulation method holds significant potential for advancing exosome production.

Dynamic adaptive coassembled sericin protein orchestrating stem cell development for nucleus pulposus regeneration

Xing Li,Yize Zhao,Peiyang Gu,Chengkun Zhao,Yujiang Fan,Xingdong Zhang,Chao Zhao,Peter X. Ma,Ganjun Feng,Yong Sun

The zonal structure of natural extracellular matrix (ECM) creates a dynamic microenvironment that regulates mechanical and biochemical signals to support stem cell development. Here, we engineered a dynamic adaptive protein gel by coassembling bioactive sericin with an amphiphilic tripeptide, forming a nucleus pulposus (NP)–like viscoelastic network with rapid stress relaxation. This coassembled protein gel sequentially activated integrin β3–mediated mechanotransduction, promoted cytoskeletal remodeling and Yes-associated protein nuclear translocation, and subsequently up-regulated bioactive factors and ECM synthesis to orchestrate stem cell development. Incorporation of kartogenin provided sustained chondrogenic cues, thereby promoting stem cell differentiation and preserving NP tissue integrity in vivo. Together, these findings establish a phase separation–driven, adaptive protein matrix that orchestrates both mechanical and biochemical signaling for robust intervertebral disc regeneration.

Periodontal ligament stem cells-loaded photocrosslinking GelMA/PEGDA scaffolds for periodontal bone regeneration

Bei Gao,Jie Yao,Yijie Wang,Chen Chen,Zhen Chai,Wanting Wan,Shiyang Wu,Zhongbo Liu,Rui Zou

Background Matrix stiffness is a key feature of the mechanical microenvironment. Periodontal tissue is a multicomponent structure composed of cementum, periodontal ligament, and alveolar bone, and is essential for the physiological reconstruction and stress of teeth. Simulating the stiffness of periodontal tissue with a biomimetic extracellular matrix is of great significance for regulating cell behavior and tissue repair. In this study, the application of GelMA-PEGDA hybrid hydrogels with different matrix stiffnesses for periodontal bone regeneration was explored. Results In this work, a GelMA‒PEGDA hybrid hydrogel ink was prepared, and the proportion of PEGDA significantly increased the matrix stiffness of the hybrid hydrogel, which had good biocompatibility. In addition, matrix stiffness promoted the osteogenic differentiation and mineralization capacity of periodontal ligament stem cells on the surface of the gels. A high-precision hydrogel scaffold was obtained by introducing a photoinhibitor. The porosity and biocompatibility of the scaffold were good. Conclusions This 3D-printed hydrogel provides a promising strategy for periodontal bone tissue regeneration.

Carbon dots for tracking and promoting the osteogenic differentiation of mesenchymal stem cells

Dan Shao,Mengmeng Lu,Duo Xu,Xiao Zheng,Yue Pan,Yubin Song,Jinying Xu,Mingqiang Li,Ming Zhang,Jing Li,Guangfan Chi,Li Chen,Bai Yang

Mesenchymal stem cells (MSCs) hold great potential for tissue engineering and regeneration medicine. However, for clinical use, MSCs may be detrimental due to their uncertain fate during the transplantation. It is therefore highly desirable to develop biocompatible nanomaterials to integrate cell fate regulation with monitoring for MSC-based therapy. Herein, we employ recently developed citric acid-based carbon dots (CDs) and their derivatives (Et-IPCA) for labeling and tracking of rat bone marrow mesenchymal stem cells (rBMSCs). We further investigate their biocompatibility and effects on the osteogenic differentiation of rBMSCs. These highly fluorescent probes provide labeling of rBMSCs by internalization without affecting cell viability or inducing apoptosis when the concentration is lower than 50 μg mL−1. Importantly, the presence of the CDs and Et-IPCA facilitates high-efficiency osteogenic differentiation of rBMSCs by promoting osteogenic transcription and enhancing matrix mineralization. Compared to Et-IPCA, CDs considerably provide long-term tracking and promote the differentiation of rBMSCs toward osteoblasts through the ROS-mediated MAPK pathway. Taken together, our results consistently demonstrate that carbon dots are capable of both tracking and enhancing the osteogenic differentiation of MSCs. This study sheds new light on the potential of carbon dots as a bifunctional tool in the thriving field of MSC-based therapy.

Single-cell analysis defines the lineage plasticity of stem cells in cervix epithelium

Zhao Zixian,Wang Yujia,Wu Yingchuan,Li Dandan,Zhang Ting,Ma Yu,Teng Xiaoming,Zuo Wei

Information about the dynamic change and post-injury regeneration of cervical epithelium is relatively rare, even though it is tightly related to gynecologic malignancy. Here, using a feeder cell-based culturing system, we stably cloned mouse and human P63 and KRT5 expressing cells from the adult cervix as putative cervical stem/progenitor cells (CVSCs). When subjected to differentiation, the cultured cells gave rise to mature cervical epithelium by differentiating into squamous or glandular cells. The ability of endogenous mouse CVSCs to reconstitute cervical epithelium after injury was also evident from the genetic lineage tracing experiments. Single-cell transcriptomic analysis further classified the CVSCs into three subtypes and delineated their bi-lineage differentiation roadmap by pseudo-time analysis. We also tracked the real-time differentiation routes of two representing single CVSC lines in vitro and found that they recapitulated the predicted roadmap in pseudo-time analysis. Signaling pathways including Wnt, TGF-beta, Notch and EGFR were found to regulate the cervical epithelial hierarchy and implicated the different roles of distinct types of cells in tissue homeostasis and tumorigenesis. Collectively, the above data provide a cloning system to achieve stable in vitro culture of a bi-lineage stem/progenitor cell population in the cervix, which has profound implications for our understanding of the cervix stem/progenitor cell function in homeostasis, regeneration, and disease and could be helpful for developing stem cell-based therapies in future.
