A biocompatible tea polyphenol nanoplatform for efficient cytosolic delivery of protein therapeutics

Hui Wang,Cui Tang,Hanfeng Zhang,Lina Guo,Chan Zou,Shengting Zhang,Jinlong Liu,Qianping Huang,Yujun Liu,Wenhu Zhou,Guoping Yang,Xiang Gao

Intracellular protein therapeutics offer great promise for treating diseases involving intracellular targets, yet their clinical translation remains limited by inefficient delivery systems. Here, we report the development of a simple, versatile, and biocompatible protein delivery platform based on tea polyphenol nanoparticles (TPNs) formed via the oxidative self-polymerization of epigallocatechin gallate (EGCG). Through noncovalent interactions between EGCG and proteins, TPNs enable efficient encapsulation of proteins with diverse molecular weights and isoelectric points under mild conditions. Using urate oxidase (UOx) as a model therapeutic protein, we demonstrate that TPNs protect UOx from enzymatic degradation, preserve its native structure and enzymatic activity, and achieve effective cytosolic delivery via glutathione-triggered release. In disease models of hyperuricemia and gouty arthritis, TPNs/UOx significantly prolonged UOx circulation, lowered serum uric acid levels, reduced joint inflammation, and improved liver and kidney function, with favorable biocompatibility. This work establishes TPNs as a versatile and stimuli-responsive platform for intracellular protein delivery, offering new strategies for the clinical application of protein therapeutics.

Tea stain-inspired solar energy harvesting polyphenolic nanocoatings with tunable absorption spectra

Yang Lu,Zou Yuan,Xia Wei,Li Haotian,He Xinyu,Zhou Yi,Liu Xianhu,Zhang Chaoqun,Li Yiwen

Discovery and development of new sustainable photothermal materials with tunable light absorption spectra play a key role in solar energy harvesting and conversion. One possible solution to this quest is to check nature as a source of matters or inspiration. Inspired by the formation of tea stains, a unique class of dark stain materials generated by the interfacial reaction between tea polyphenols and metal substance, we reported the facile preparation and screening of a series of photothermal nanocoating layers via the metal ion (i.e. Cu(II),Fe(III), Ni(II), Zn(II)) promoted in situ polymerization of typical phenolic moieties of tea polyphenols (i.e., catechol and pyrogallol). It was found that those resulting metal-polyphenolic nanocoatings showed various promising features, such as high blackness and strong adhesion, excellent and tunable light absorption properties, good hydrophilicity and long-term stability. We further fabricated the photothermal composite devices by in situ formation of metal-polyphenolic nanocoatings on pristine silks for solar desalination, which demonstrated promising durable evaporation behaviors with excellent evaporation rates and steam generation efficiencies. We believe that this work could provide more opportunities towards new types of bio-inspired and sustainable photothermal nanomaterials for solar energy harvesting applications such as water desalination.

Green synthesis of nano silver by tea extract with high antimicrobial activity

Yun Fang,Ci-Qing Hong,Fang-Rong Chen,Fang-Ze Gui,Yun-Xin You,Xiong Guan,Xiao-hong Pan

In this study, a facile synthetic pathway was proposed to fabricate silver nanoparticles by using tea extract as reducing agent and stabilizer to reduce silver nitrate, which was further evaluated for the antimicrobial activity against bacterium ( Escherichia coli ) and pathogenic fungus of tea ( Pseudopestalotiopsis theae ). The average particle size of silver nanoparticles is 15.41 nm by X-ray diffraction (XRD) and Scherrer equation, the morphology is relatively regular and well dispersed by scanning electron microscope (SEM) and transmission electron microscope (TEM), the zeta potential is −13.91 ± 0.89 mV and the aggregate particle size distribution in water is around 773.49 nm. In addition, the synthesized nano silver had good antimicrobial effect on Pseudopestalotiopsis theae ( P. theae ) and Escherichia coli ( E. coli ), and the inhibition ratio was 96.05% and 61.87%, respectively. And the reaction of silver nanoparticles with bacteria and fungi could not be driven by the electrostatic interactions, but might by intermolecular forces. This study provides a facile and effective method for the preparation of nano silver, and broadens the application of nano silver in antimicrobial agent.

Rapid Authentication of White Tea Vintages Using SERS Fingerprints and Machine Learning

Lin Hui,Chen Zhenglong,Ren Chunfeng,You Ruiyun,Lu Yudong

The aging process has a significant impact on the quality and market value of white tea (WT), as described in a traditional proverb: “One year’s tea, three years’ medicinal value, and seven years becomes a treasure.” However, the lack of a rapid and reliable method for age identification poses challenges for market regulation. This study proposes a novel approach that combines surface-enhanced Raman spectroscopy (SERS) with machine learning (ML) to accurately identify the age of white tea. SERS is used to obtain molecular fingerprint spectra from Fujian white tea samples covering seven different years (2011−2023). Seven machine learning models, including logistic regression (LR), support vector machine (SVM), and random forest (RF), were systematically evaluated. The LR model, after being preprocessed through principal component analysis—logistic discriminant analysis, demonstrated relatively excellent performance. This SERS-ML method can perform rapid analysis and requires very little samples preparation. Our work establishes a robust, efficient, and field-deployable strategy for identifying the age of white tea, which is of great significance for combating fraud and protecting consumers. Graphical

Carboxymethyl cellulose/tea polyphenol/polypyrrole-derived carbon aerogel for electromagnetic wave absorption

Qian Liang,Haoqing Cheng,Fuzhi Yan,Xiaomin Hu,Fen Zhu,Huabo Huang,Xianghua Yu,Yulan Liu,Jiayou Ji,Liang Li

A green route is developed for Fe₃O₄/FeN/N co-doped porous carbon aerogels from sodium carboxymethyl cellulose, tea polyphenols and polypyrrole. • Tea polyphenol−Fe 3+ coordination mitigates pyrolysis-induced framework shrinkage and enables uniform polypyrrole dispersion. • The CTP2-700 aerogel exhibits a minimum reflection loss of −58.4 dB (3.1 mm) and effective bandwidth of 5.3 GHz at 2.1 mm (20 wt% loading). • RCS simulations demonstrate good radar stealth; this work guides the design of lightweight biomass carbon-based electromagnetic absorbers. In recent years, the fabrication of lightweight porous carbon aerogels from biomass precursors has attracted considerable attention in the field of electromagnetic wave absorption. Herein, novel Fe 3 O 4 /FeN/N porous carbon aerogels were successfully synthesized using sodium carboxymethyl cellulose as the carbon source, with metal-phenolic networks constructed through the coordination of tea polyphenols and Fe 3+ . These networks not only enable homogeneous dispersion of polypyrrole but also mitigate the shrinkage of carbon frameworks during high-temperature pyrolysis, thereby maintaining the structural integrity of the aerogel products. The as-prepared carbon aerogels possess a hierarchical porous structure and well-developed conductive networks. Electromagnetic parameter tests revealed that the materials achieved a minimum reflection loss of −58.4 dB at a matching thickness of 3.1 mm, together with a broad effective absorption bandwidth of 5.3 GHz at a thickness of 2.1 mm. This work offers an innovative strategy for the green and efficient utilization of cellulose-based materials and demonstrates their potential for developing lightweight, high-performance electromagnetic wave absorbers.

Multi-omics insights into aroma formation in congou black tea during fermentation

Jingtao Zhou,Muxue Qin,Jiasi Li,Junyu Zhu,Yazhong Chen,De Zhang,Bernard Ntezimana,Zhi Yu,Yuqiong Chen,Dejiang Ni

Phenylalanine degradation dominates aroma formation via transaminase pathway‌. • Fatty acid autoxidation surpasses enzymatic pathways in aldehyde production‌. • Glycoside hydrolysis activates terpenoid release during early fermentation‌. • Ribosomal proteins sustain enzyme synthesis despite cellular disruption‌. • Protein phosphatases modulate enzyme activities through modifications. This study used multi-omics technologies to analyze aroma formation during Congou black tea fermentation. Volatile compounds were analyzed by headspace solid phase microextraction coupled with gas chromatography mass spectrometry using two columns of different polarity. Fermentation increased total volatile normalized peak area fivefold, with alcohols, aldehydes, and acids increasing over sevenfold. 29 differential metabolites were screened, including amino acid derived phenylacetaldehyde, phenylethanol, and 2-methylbutanal; fatty acid derived ( E,E )-2,4-heptadienal, hexanal, and 1-hexanol; and isoprenoid derived linalool, geraniol, and beta ionone. Transcriptomic, proteomic, and enzyme analyses revealed that biosynthesis contributed to early accumulation of amino acid and isoprenoid derived aromas, whereas ortho quinone mediated Strecker degradation and free radical induced fatty acid auto oxidation dominated generation of amino and fatty acid derived aromas during middle and late fermentation. In conclusion, aroma formation during fermentation results from biosynthesis and non-enzymatic oxidation, with the latter possibly dominating amino and fatty acid derived aromas. Tea Polyphenols Enhance Embryonic Development and Mitigate DMSO-Induced Toxicity in Drosophila

Quanyong Wu,Caohuizi Xiao,Boya Ouyang,Jesus Simal-Gandara,Yaping Qi,Zi Chen

Tea polyphenols (TPs), potent antioxidants found in tea, offer cellular protection and health benefits. Dimethyl sulfoxide (DMSO), a widely used organic solvent in medicine, remains controversial due to its potential embryonic toxicity. This study explored the effects of TPs from oolong and black tea, DMSO exposure, and their combined treatment on embryonic development using a fruit fly model. TPs from oolong tea (rich in catechin and EGCG) and black tea (mainly catechin) promoted embryonic development, enhanced larval vitality, and improved oxidative stress resistance in adults. Catechin intervention mirrored the effects of TPs from oolong and black tea, while the effect of EGCG was sex-dependent. DMSO showed dose-dependent toxicity, with 1% concentrations disrupting organ development and preventing emergence. Notably, EGCG and oolong tea TPs supported normal organ formation. Furthermore, catechin and tea TPs alleviated 0.5% DMSO-induced damage, boosting larval vitality, adult locomotion, and body weight in males, while reducing oxidative stress.

Effects of Three Different Withering Treatments on the Aroma of White Tea

Huiting Wu,Yuyu Chen,Wanzhen Feng,Shanshan Shen,Yuming Wei,Huiyan Jia,Yujie Wang,Weiwei Deng,Jingming Ning

White tea (WT) is a slightly fermented tea, and withering is a critical step in its processing. The withering treatment can affect white tea’s aroma; different treatments’ effects were investigated in this study. White tea was withered indoors (IWT), in a withering-tank (WWT), or under sunlight (SWT). Quantitative descriptive analysis (QDA) results showed that SWT had a more obvious flower aroma, and WWT had a more pronounced grassy aroma. Volatile compounds were extracted and subsequently detected with solvent-assisted flavor evaporation (SAFE) and headspace solid-phase microextraction (HS-SPME) combined in addition to gas chromatography–mass spectrometry (GC-MS). A total of 202 volatile compounds were detected; 35 of these aroma-active compounds met flavor dilution (FD) factor ≥ 4 or odor activity value (OAV) ≥ 1. The nine key potent odorants for which both conditions were met weredimethyl sulfide,2-methyl-butanal,1-penten-3-one,hexanal,(Z)-4-heptenal,β-Myrcene, linalool, geraniol, and trans-β-ionone. These results were used with QDA to reveal that SWT had a stronger floral aroma mainly due to an increase of geraniol and linalool. Moreover, WWT had a stronger grassy aroma mainly due to increasedhexanal. The results could be used to select processing methods for producing white tea with a superior aroma. Keywords:white tea;withering treatment;volatile compounds;aroma extract dilution analysis;odor activity value

Deciphering the Aroma-Driven Mechanism of Sweet Flavor in Keemun Black Tea

Wenjing Huang,Zhibin Ye,Shengmei Xie,Zihao Qi,Mengying Zhang,Shimao Fang,Jingming Ning

The sweetness of Keemun black tea (KBT) is a critical determinant of its overall sensory quality. This study used sensory analysis to quantitate the sweet odor and flavor of KBT. The results indicated that volatile compounds, rather than soluble sugars or free amino acids, are the main contributors to KBT’s sweet flavor. Ten odorants with an odor activity value > 1 were identified by the Sensomics approach and screened by applying the odor-induced taste enhancement (OITE) method, i.e. β-damascenone, (E)-β-ionone, linalool, methional, dimethyl sulfide, geraniol, β-myrcene, coumarin, citral, and 2-phenylacetaldehyde, which were identified as the key drivers of KBT’s sweet flavor. Omission and addition experiments confirmed that the intensity of sweet odor was correlated with sweet flavor (r = 0.71). These findings demonstrate that the sweet flavor of KBT mainly originates from its odorants, which synergistically enhance sweetness through OITE, resulting in the high overall flavor quality of KBT.

Monitoring Antioxidant Preservation in Microwave-Dried Tea Using H2O2-Responsive Electrochemical Sensor

Jiaoling Wang,Hao Li,Xinxin Wu,Xindong Wang,Xinai Zhang

Considering the demand for nutritional assessment and product quality control in the tea industry, this work develops an effective electrochemical sensor based on gold nanoparticles electrodeposited onto a zeolitic imidazolate framework (Au/MOF(Zn)) for evaluating the antioxidant activity of tea subjected to microwave-assisted drying (MAD) through hydrogen peroxide (H2O2) scavenging. The MOF(Zn) enables uniform deposition of AuNPs, which significantly enhances the electrocatalytic oxidation of H2O2. The fabricated sensor exhibits a wide linear detection range from 400 μM to 1.8 mM for H2O2with a correlation coefficient of 0.9983. The experimental results demonstrate acceptable selectivity, with signal interference <5% from common tea compounds like inorganic ions, sugars, and organic acids. Electrochemical methods, including cyclic voltammetry (CV) and differential pulse voltammetry (DPV) analysis, were employed to quantify H2O2by measuring oxidation currents in phosphate-buffered saline (PBS, pH 7.0). The relative standard deviation (RSD) for repeatability and reproducibility was 5.1% and 6.8%, respectively, confirming high reliability. This sensor was successfully applied to assess antioxidant capacity in tea extracts obtained from fresh leaves subjected to microwave-assisted drying under varying power and duration. Results indicate that increasing microwave power enhances antioxidant activity, while prolonged drying at low power initially increases activity (peaking at 120 s) but reduces it upon extended exposure. Optimal antioxidant preservation was achieved at 120 s. This real-time, reliable sensing strategy offers theoretical foundations for optimizing tea processing parameters to preserve bioactive compounds, particularly polyphenols like catechins, thereby improving tea quality and health benefits.

Injectable carboxymethyl chitosan-genipin hydrogels encapsulating tea tree oil for wound healing

Kepeng Hu,Erna Jia,Qimeng Zhang,Wei Zheng,Rongjiao Sun,Mengao Qian,Ying Tan,Wanle Hu

Injectable hydrogel is of interesting for wound healing due to it can be used as carriers of bioactive molecules for the reparation of tissues with minimal invasiveness. However, the integration of lipid-soluble substances into hydrogel network is difficult because of the polarity differences. Here, the tea tree oil (TTO) is encapsulated into the hydrogel network via a previous emulsification process, and a tough and antibacterial injectable hydrogel is synthesized by the Schiff base reaction between carboxymethyl chitosan (CMCS) and genipin (GP). CMCS is served as both an emulsifier and a gel-forming material to construct the heterogeneous hydrogel. The obtained hydrogels present high adhesive strength (∼162.75 kPa), great antibacterial properties (over 90 %) and excellent biocompatibility. Moreover, an anal fistula-like wound healing experiment concluded that the heterogeneous hydrogel has good slow-release properties of TTO for an accelerate healing process, this hydrogel shows great potential for the treatment of complex anal fistula wounds.

Aroma formation and transformation during sealed yellowing process of Pingyang yellow tea

Rong-Jian Dong,Xie-Huang Sheng,Qian-Tu Xie,Xiaolin Huang,Fujie Yan,Songbai Liu

Yellow tea, a unique type of tea in China which is characterized with yellow color, has gained increasing popularity due to its pleasant taste. However, transformation of aroma compounds during sealed yellowing has been poorly understood. Results of sensory evaluation exhibited that yellowing time was the key factor for flavor and fragrance formation. A total of 52 volatile components during sealed yellowing process of Pingyang yellow soup were further collected and analyzed. The results demonstrated that the sealed yellowing process significantly increased the ratio of alcohol and aldehyde compounds in the aroma volatiles of yellow tea, which were primarily composed of geraniol, linalool, phenylacetaldehyde, linalool oxide and cis -3-hexenol, and their proportion increased with the prolongation of sealed yellowing. Mechanistic speculation revealed that the sealed yellowing process promoted release of alcoholic aroma compounds from their glycoside precursors and enhanced Strecker and oxidative degradation. This study revealed the transformation mechanism of aroma profile during the sealed yellowing process, which would facilitate processing of yellow tea.
