Semi-rational engineering of Issatchenkia terricola acetaldehyde dehydrogenase for enhanced thermostability and catalytic efficiency in detoxification of aldehydes

Jing Lu,Lina Wu,Jingwen Zhao,Huilin Shen,Bingxin Shi,Yuchen Gao,Shuai Zhao,Lei Wang,Yawei Shi

Acetaldehyde dehydrogenase is crucial for detoxifying aldehydes in pharmaceutical, food, and biofuel industries. We enhanced Issatchenkia terricola ALDH (ist-ALDH) thermostability and catalytic activity toward aliphatic and aromatic aldehydes via molecular engineering. The combinatorial mutant H62G/D240P/F429Vshifted the optimal temperature from 35 °C to 55 °C. It had better thermal stability with a half-life (t 1/2 ) of 315.07 ± 4.24 min at 40 °C, 14.41-fold that of the wild-type (WT). Kinetic studies showed higher catalytic efficiency for both aliphatic and aromatic aldehydes. H62G/D240P/F429V had 11.96-fold higher catalytic activity for acetaldehyde than WT, with K m , V max , k cat , and k cat / K m improved by 3.44-, 10.99-, 10.98-, and 39.30-fold, respectively. For aromatic substrates like furfural and 5-hydroxymethylfurfural, catalytic efficiencies were 19.41- and 46.77-fold of the WT levels. Molecular dynamics and structural analysis indicated that enhanced hydrophobic packing and optimized electrostatic potential stabilized H62G/D240P/F429V rigid conformation. Substrate binding studies showed the hydrogen bond distance between Cys327 and acetaldehyde in the mutant decreased from 2.18 Å to 2.09 Å compared to the WT. Additionally, an additional hydrogen bond was formed between Cys328 and the aldehyde group, with a bond distance of 1.94 Å. These findings establish a basis for the property engineering of tailoring ALDH properties, with implications for developing aldehyde detoxification systems in industrial biocatalysis.

How microorganisms respond to single and binary antibiotic exposure: Focusing on detoxification, nitrogen, phosphorus, and cell metabolism mechanisms

Taotao Tang,Bin Yan

The coexistence of multiple antibiotics poses a significant threat to biological treatment systems. However, few studies have systematically explored their comprehensive impact on the metabolic performance of microorganisms. This study examined the effects of sulfadiazine (SDZ) and norfloxacin (NOR) on microbial enzyme activity, electron transport system, oxidative stress, detoxification, and metabolic capabilities after long-term exposure to environmental concentrations (0.01–0.5 mg/L). Our results demonstrated that single and coexposure to SDZ and NOR significantly enhanced the activities of ammonia monooxygenase and hydroxylamine oxidase, induced oxidative damage, and inhibited electron transfer and nitrate reductase activity. Metatranscriptomics analysis revealed that SDZ and NOR, either alone or in combination, shortcut nitrification and denitrification, replacing the traditional nitrification-denitrification pathway as the dominant nitrogen removal pathway. Compared to single exposure, microorganisms under coexposure conditions exhibited enhanced detoxification capacity by upregulating genes associated with toxic efflux and carbon metabolism and promoting polyphosphate accumulation and transformation. Coexposure particularly worsens the ability of cell mobility but can mainly improve toxicity resistance between microorganisms by enhancing putrescine and glutamate transport. This research enhances our understanding of the impact of multi-pollutant interactions on wastewater treatment performance and biological metabolic mechanisms.

A double-lyophilization method for the preparation of CS/GO-COOH scaffold and its application in blood detoxification

Gang Zhou,Lei Wang,Jianchao Li,Jun Tai,Haisheng Su,Jing Zhang,Yuan Xi,Yubo Fan

The accumulation of uremic toxins in blood might induce chronic renal failure (CRF). The incidence of CRF was as high as 10%. The traditional therapy for CRF was hemodialysis, which was more effective to remove small molecules, such as urea and creatinine. However, this detoxification method ignored the tissue functional adaption due to the retention of macromolecule uremic toxins. To solve this problem, this paper developed a new kind of chitosan/carboxyl graphite oxide (CS/GO-COOH) scaffold via a double-lyophilization method. Then, this synthetic scaffold was characterized by Fourier transform infrared spectroscopy, scanning electron microscope, hydrophilic test, mechanical property, and in vitro detoxification test. Covalent bonding and hydrogen bonding were formed, indicating the strong interactions between CS and GO-COOH. There were interconnected networks in the synthesized scaffold. The mechanical test suggested that the GO-2500 scaffold had excellent mechanical strength, which was 7.41 ± 0.82 MPa with 25% shrink. What is more, GO-2500 could totally rebound within 1s, after compressed to 90% shrink. The rates of GO-2500 were 1587 ± 60 and 246 ± 10% according to the water uptake and retention data, respectively. Furthermore, the detoxification of GO-2500 to urea, creatinine, VB12, and β2-m were 67.59 ± 2.31, 39.67 ± 2.95, 31.51 ± 2.62, and 83.82 ± 7.76 mg/g, respectively. The resulting CS/GO-COOH scaffold held great potential for the detoxification of uremic toxins.

Maize root exudates promote Bacillus sp. Za detoxification of diphenyl ether herbicides by enhancing colonization and biofilm formation

Yanning Tian,Fangya Zhong,Na Shang,Houyu Yu,Dongmei Mao,Xing Huang

Diphenyl ether herbicides are extensively utilized in agricultural systems, but their residues threaten the health of sensitive rotation crops. Functional microbial strains can degrade diphenyl ether herbicides in the rhizosphere of crops, facilitating the restoration of a healthy agricultural environment. However, the interplay between microorganisms and plants in diphenyl ether herbicides degradation remains unclear. Thus, the herbicide-degrading strain Bacillus sp. Za and the sensitive crop, maize, were employed to uncover the interaction mechanism. The degradation of diphenyl ether herbicides by strain Bacillus sp. Za was promoted by root exudates. The strain induced root exudates re-secretion in diphenyl ether herbicide-polluted maize. We further showed that root exudates enhanced the rhizosphere colonization and the biofilm biomass of strain Za, augmenting its capacity to degrade diphenyl ether herbicide. Root exudates regulated gene fliZ, pivotal in biofilm formation. Wild-type strain Za significantly reduced herbicide toxicity to maize compared to the ZaΔfliZ mutant. Moreover, root exudates promoted strain Za growth and chemotaxis, which was related to biofilm formation. This mutualistic relationship between the microorganisms and the plants demonstrates the significance of plant-microbe interactions in shaping diphenyl ether herbicide degradation in rhizosphere soils.

The bonding and intercalation efficacy of polyethyleneimine enhanced BiOBr semiconductor-organic framework degradation and detoxification of bisphenol AF

Runren Jiang,Guanghua Lu,Tianjian Dang,Min Wang,Jianchao Liu,Zhenhua Yan

Photocatalysis is a promising method for water pollution treatment, but it is challenged due to its low efficiency and insufficient understanding of the toxicity after degradation. In this work, we construct a polyethyleneimine-enhanced BiOBr semiconductor organic framework (PEI/BiOBr SOF) by utilizing PEI's bonding and intercalation effects. PEI/BiOBr SOF enhances the photocatalytic degradation of bisphenol AF (BPAF) at a reaction rate 10 times that of BiOBr. Under the dual effects of intercalation and bonding, PEI/BiOBr SOF can realize extended the light absorption, increase the specific surface area and surface-active sites, inhibit carrier complexation, and promote the production of reactive oxygen species. The toxicity changes of the BPAF were evaluated by combining chemical product analysis and biological toxicity experiments. The mutual validation of the toxicity prediction results of the ECOASR product and the zebrafish toxicity experiment confirmed that the toxicity of BPAF decreased after degradation. Furthermore, the mechanism of photocatalytic degradation and detoxification of BPAF with PEI as the circuit of charge transmission of the BiOBr SOF was proposed.

Glutathione S-Transferases Mediate In Vitro and In Vivo Inactivation of Genipin: Implications for an Underlying Detoxification Mechanism

Yulin Zhao,Haoyan Huang,Ning Lv,Chunyan Huang,Huili Chen,Han Xing,Chaorui Guo,Ning Li,Di Zhao,Xijing Chen,Yongjie Zhang

Genipin (GP), the reactive metabolite of geniposide (GE), is responsible for GE-induced hepatotoxicity. As a potential detoxification pathway, the inactivation of GP by glutathione S-transferases (GSTs) has not yet been characterized. In this study, the thiol-GSH conjugates of GP, M532-1 and M532-2 were first identified and the catalytic activities of GSTs were investigated both in vitro and in vivo. GSTA1-1 and GSTA4-4 showed high activity in the formation of both thiol-GSH conjugates, whereas GSTA4-4 specifically catalyzed M532-2 formation in vitro. The active GST isoforms protect against alkylation of N-acetylcysteine (NAC), a classic model nucleophile. GST inhibition attenuated M532-1 formation in rat bile, confirming the in vivo catalytic role of GSTs. In conclusion, this study demonstrated the inactivation of GP by GSTs and implied that interindividual variability of GSTs may be a risk factor for susceptibility to GE-induced hepatotoxicity.

Hydrogen-bond based charge bridge in a heterojunction system for the synergistic degradation and detoxification of two PPCPs

Runren Jiang,Guanghua Lu,Tianjian Dang,Min Wang,Jianchao Liu,Zhenhua Yan,Haijiao Xie

The use of photocatalysis to remove pharmaceuticals and personal care products (PPCPs) is emerging as a promising method, but few studies have investigated the simultaneous degradation of multiple PPCPs. Here, we constructed a hydrogen-bonded bridged MIL-101(Cr)–NH 2 /BiOCl-OH (MCN/BOCH) system to degrade sulfamethoxazole and ethyl p-aminobenzoate alone or in combination. The MCN/BOCH system effectively removed two PPCPs under visible light and exerted a synergistic effect on degrading the combined pollutants. Meanwhile, the degradation products of the mixture changed significantly compared with that of a single pollutant, and the toxicity of the composite pollutants was significantly reduced through the combined effect of synergistic degradation. The bridging of hydrogen bonds effectively accelerated charge transfer at the heterojunction interface under visible light. Moreover, the separated redox side of the MCN/BOCH heterojunction system and the electron donor and acceptor effect of the two PPCPs significantly enhanced the synergistic photocatalytic degradation performance. This work might be useful for controllable designs of hydrogen-bonded photocatalysts for the treatment of complex PPCP pollution.

Efficient removal and detoxification of Cr(VI) by PEI-modified Juncus effuses with a natural 3D network structure

Shengli Zhang,Wei Li,Menglin Li,Tong Lin,Kai Su,Hongwei Yang,Junmin Chen

In the present study, a novel PEI-modified Juncus effuses (PEI-JE) adsorbent was prepared based on the natural 3D network structure of JE via a simple and environmental-friendly method with the help of the ethanol/aqueous solution. SEM, EDS, ATR-FTIR and XPS were performed to reveal the physical and chemical structure of the PEI-JE. The Cr(VI) removal by the PEI-JE was investigated by bath adsorption experiments. Results showed that the developed PEI-JE exhibited the same 3D network structure as the JE, which not only was easy to separate from aqueous solution but also can expose more active sites and facilitate mass transfer. The optimal pH for Cr(VI) adsorption on the PEI-JE was 3, and adsorption equilibrium time was related to initial Cr(VI) concentrations. The adsorption kinetic obeyed pseudo-second-order kinetic model, and the adsorption isotherm showed the best fit with Freundlich model. The calculated maximum adsorption capacity was 268.93 mg/g at 25℃. Coexisting ions including Na + , K + , Al 3+ , H 2 PO 4 − , Cl − , NO 3 – had no remarkable effect on Cr(VI) adsorption, but the presence of SO 4 2− , HCO 3 – and CO 3 2– caused the efficiency losses of 10.9%, 41% and 66%, respectively. After 10 cycles of adsorption/desorption, Cr(VI) removal rate on the PEI-JE almost kept unchanged, demonstrating that the PEI-JE possessed good reusability. Mechanism study suggested that Cr(VI) removal was involved in a three-step reaction, namely electrostatic adsorption, redox and complexation. Film diffusion was the rate-limiting step and chemical adsorption was the main mechanism. The Cr(VI) adsorbed on the PEI-JE surface was completely reduced to Cr(III), demonstrating that PEI-JE is useful for Cr (VI) removal and detoxification.

Effects of azadirachtin on detoxification-related gene expression in the fat bodies of the fall armyworm, Spodoptera frugiperda

Yu Haikuo,Yang Xianmei,Dai Jinghua,Li Yuning,Veeran Sethuraman,Lin Jintian,Shu Benshui

The fall armyworm, Spodoptera frugiperda, has become a worldwide pest and threatens world food production. A previous study indicated that azadirachtin, the most effective botanical insecticide for S. frugiperda , inhibits larval growth of the insect. The effect of azadirachtin on the tissues of the larvae, however, remains to be determined. In this study, the effects of azadirachtin on the structure of fat bodies were analyzed. Comparative transcriptomic analysis was conducted between controls and samples treated with 0.1 μg/g azadirachtin for 7 days to explore potential relevant mechanisms. The expression of 5356 genes was significantly affected after azadirachtin treatment, with 3020 up-regulated and 2336 down-regulated. Among them, 137 encode detoxification enzymes, including 53 P450s , 20 GSTs , 27 CarEs , 16 UGTs , and 12 ABC transporters . Our results indicated that azadirachtin could destroy fat body structure and change the mRNA levels of detoxification-related genes. The up-regulated genes encoding detoxification enzymes might be related to detoxifying azadirachtin. Our results elucidate a preliminary mechanism of azadirachtin detoxification in the fat bodies of S. frugiperda larvae.

A mix & act liposomes of phospholipase A2-phosphatidylserine for acute brain detoxification by blood‒brain barrier selective-opening

Zinan Zhang,Wenbin Cao,Huanchun Xing,Shuai Guo,Lijuan Huang,Lin Wang,Xin Sui,Kui Lu,Yuan Luo,Yongan Wang,Jun Yang

In the treatment of central nervous system disease, the blood–brain barrier (BBB) is a major obstruction to drug delivery that must be overcome. In this study, we propose a brain-targeted delivery strategy based on selective opening of the BBB. This strategy allows some simple bare nanoparticles to enter the brain when mixed with special opening material; however, the BBB still maintains the ability to completely block molecules from passing through. Based on the screening of BBB opening and matrix delivery materials, we determined that phospholipase A2-catalyzed 1-palmitoyl-2-oleoyl- sn -glycero-3-phosphoserine liposomes can efficiently carry drugs into the brain immediately. At an effective dose, this delivery system is safe, especially with its effect on the BBB being reversible. This mix & act delivery system has a simple structure and rapid preparation, making it a strong potential candidate for drug delivery across the BBB.

Triclosan exposure induces immunotoxic impacts by disrupting the immunometabolism, detoxification, and cellular homeostasis in blood clam (Tegillarca granosa)

Yu Han,Weixia Zhang,Yu Tang,Wei Shi,Zhiquan Liu,Imane Lamine,Hangjun Zhang,Jing Liu,Guangxu Liu

Omnipresent presence of triclosan (TCS) in aqueous environment puts a potential threat to organisms. However, it's poorly understood about its immunometabolic impacts of marine invertebrates. In present study, we use a representative bivalve blood clam ( Tegillarca granosa ) as a model, investigating the effects of TCS exposure at 20 and 200 μg/L for 28 days on immunometabolism, detoxification, and cellular homeostasis to explore feasible toxicity mechanisms. Results demonstrated that the clams exposed to TCS resulting in evident immunotoxic impacts on both cellular and humoral immune responses, through shifting metabolic pathways and substances, as well as suppressing the expressions of genes from the immune– and metabolism–related pathways. In addition, significant alterations in contents (or activity) of detoxification enzymes and the expression of key detoxification genes were detected in TCS-exposed clams. Moreover, exposure to TCS also disrupted cellular homeostasis of clams through increasing MDA contents and caspase activities, and promoting activation of the apoptosis–related genes. These findings suggested that TCS might induce immunotoxic impacts by disrupting the immunometabolism, detoxification, and cellular homeostasis.

Mesoporous cobalt-manganese layered double hydroxides promote the activation of calcium sulfite for degradation and detoxification of metronidazole

Akbar Mamatali,Dedong Wu,Haijiao Xie,Pengfei Xiao

Metronidazole (MNZ), a commonly used antibiotic, poses risks to water bodies and human health due to its potential carcinogenic, mutagenic, and genotoxic effects. In this study, mesoporous cobalt-manganese layered double hydroxides (Co x Mn y -LDH) with abundant oxygen vacancies (O v ) were successfully synthesized using the co-precipitation method and used to activate calcium sulfite (CaSO 3 ) with slight soluble in water for MNZ degradation. The characterization results revealed that Co 2 Mn-LDH had higher specific areas and exhibited good crystallinity. Co 2 Mn-LDH/CaSO 3 exhibited the best catalytic performance under optimal conditions, achieving a remarkable MNZ degradation efficiency of up to 98.1 % in only 8 min. Quenching experiments and electron paramagnetic resonance (EPR) tests showed that SO 4 •− and 1 O 2 played pivotal roles in the MNZ degradation process by activated CaSO 3 , while the redox cycles of Co 2+ /Co 3+ and Mn 3+ /Mn 4+ on the catalyst surface accelerated electron transfer, promoting radical generation. Three MNZ degradation routes were put forward based on the density functional theory (DFT) and liquid chromatography-mass spectrometer (LC-MS) analysis. Meanwhile, the toxicity analysis result demonstrated that the toxicity of intermediates post-catalytic reaction was decreased. Furthermore, the Co 2 Mn-LDH/CaSO 3 system displayed excellent stability, reusability, and anti-interference capability, and achieved a comparably high removal efficiency across various organic pollutant water bodies. This study provides valuable insights into the development and optimization of effective heterogeneous catalysts for treating antibiotic-contaminated wastewater.
