Aspirin eugenol ester inhibits agonist-induced platelet aggregation in vitro by regulating PI3K/Akt, MAPK and Sirt 1/CD40L pathways

Dong-Shuai Shen,Ya-Jun Yang,Xiao-Jun Kong,Ning Ma,Xi-Wang Liu,Shi-Hong Li,Zeng-Hua Jiao,Zhe Qin,Mei-Zhou Huang,Jian-Yong Li

Aspirin eugenol ester (AEE) was a promising drug candidate for treating inflammation, pain and fever and preventing cardiovascular diseases with fewer side effects than its precursors. Previous researches indicated that AEE could markedly inhibit agonist-induced platelet aggregation in vitro and ex vivo , however, the anti-platelet aggregation mechanisms of AEE remain to be defined. Here, AEE in vitro effects on agonist-induced granule-secretion, intercellular Ca 2+ mobilization and thromboxane A 2 (TXA 2 ) generation were examined. Vasodilator-stimulated phosphoprotein (VASP), mitogen-activated protein kinase (MAPK), Akt, Sirt 1 and CD40L expressions were also studied. In agonist-activated platelets in vitro , AEE markedly attenuated granule secretion markers (P-selectin expression and ATP release), intercellular Ca 2+ mobilization and thromboxane B 2 (TXB 2 ) formation. AEE also attenuated CD40L activation, suppressed extracellular-signal-regulated protein kinase 2 (ERK2), c-Jun N-terminal kinase 1 (JNK1) and Akt phosphorylation, and recovered Sirt1 expression, but the activation of p38, VASP Ser157 and VASP Ser239 , and the levels of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) were not affected by AEE. Overall, this study demonstrates that AEE inhibits agonist-induced platelet aggregation in vitro by regulating PI3K/Akt, MAPK and Sirt 1/CD40L pathways.

Preparation of eugenol/1−methylcyclopropene composite microsphere by S/O/W method and study on the postharvest quality of loquat

Jingyi Su,Zhaoxin Cao,Yueyang Mao,Wen Shen,Xinyu Wang,Siyu Zhang,Yanqi Chen,Xueme Ge

The S/O/W method of solvent evaporation was utilized in this work for manufacturing poly ( ε −caprolactone) (PCL) microspheres loaded with 1−methylcyclopropene (1−MCP−α−CD) and eugenol (EUG) for temperature- and humidity-regulated EUG release. The composite microspheres with higher encapsulation efficiency (71.39 % ± 2.71 %) were attained by optimizing the rotation speed and mass ratio of PCL to composite powder . The microspheres had good spherical shape without large pores. The rate of release could be managed by different temperature and relative humidity conditions with the release of microspheres (96.82 % ± 0.81 % in 12 days) at the temperature of 60 °C (90 % relative humidity) and (86.50 % ± 2.71 % in 12 days) at the 90 % relative humidity (60 °C). Microsphere encapsulation of EUG and 1−MCP−α−CD was demonstrated by Fourier transform infrared spectroscopy and X-ray diffractometer. Thermal gravity analysis demonstrated that the degradation of EUG@β−CD and 1−MCP−α−CD was delayed after encapsulation. The deterioration of loquat (Baiyu, white-fleshed) can be postponed after treatment of EUG/1-MCP composite microspheres. Relative conductivity, total soluble solids, hardness, color, titratable acid , and weight loss of the loquat were also improved. It could significantly inhibit polyphenol oxidase (PPO) and peroxidase (POD) activities and inhibited browning of loquat tissue. EUG/1−MCP composite microspheres we formulated in this work might be employed to increase the shelf life of loquat.

Cutoff Ostwald ripening stability of eugenol-loaded Pickering emulsion stabilized by chitosan particles and long-term antibacterial coating activity evaluation

Mengjiao Wang,Huiran Qi,Wenyu Du,Junhua Lu,Liying Han,Shupeng Su

Plant essential oils such as eugenol exhibit broad-spectrum antimicrobial and antioxidant properties but suffer from high volatility, poor water solubility, and rapid release, limiting their application in food preservation. To address these challenges, we developed chitosan-based Pickering emulsions with effective suppression of Ostwald ripening and long-term coating antimicrobial activity. The Ostwald ripening inhibition, long-term stability, release behavior, and coating antimicrobial performance of eugenol-loaded chitosan Pickering emulsions were systematically investigated. Chitosan Pickering emulsions encapsulating pure eugenol exhibited optimal stability at pH 6.5; however, complete suppression of Ostwald ripening was not achieved. At pH 6.5, the incorporation of 2.5% corn oil effectively inhibited Ostwald ripening of the eugenol emulsions, conferring long-term stability over a 150-day storage period. Chitosan Pickering emulsions exhibited superior sustained release of eugenol compared to Tween emulsions, attributed to their inherent film-forming capability. The 95% eugenol-loaded chitosan Pickering emulsion exhibited the slowest release rate and the most prolonged coating antimicrobial activity, achieving a maximum coating duration of 48 days under simulated conditions. These findings highlight the potential of this chitosan Pickering emulsion system as a promising and adaptable platform for developing high-performance, long-lasting antimicrobial edible coatings to extend food shelf life. β-Cyclodextrin/tea saponin complex-stabilized eugenol emulsion functionalized composite films with antibacterial and sustained-release properties for fruit preservation

Longbin Li,Chi Feng,Fuhou Lei,Jiufang Duan,Kun Wang,Jianxin Jiang

β-Cyclodextrin/tea saponin (CT) complexes were prepared. • CT-stabilized eugenol emulsion shows superior storage and environmental stability. • CT complex retards the release rate of eugenol in the composite film. • Cationic guar gum maintained emulsion structural integrity during film formation. • The film integrates antibacterial, UV-shielding, and sustained-release properties. Addressing the severe postharvest losses of perishable fruits and environmental concerns caused by plastic packaging, this study developed a multifunctional composite film. Specifically, a composite of β-cyclodextrin and tea saponin—a byproduct of camellia oil processing—was synthesized and served as a stabilizer for the eugenol emulsion. This composite exhibited favorable surface activity, endowing the emulsion with excellent storage stability. Further, incorporating the emulsion into a κ-carrageenan/cationic guar gum film-forming matrix, the obtained film exhibited favorable mechanical properties (tensile strength: 22.62 MPa, elongation at break: 33.16%), outstanding antioxidant activity (scavenging rates of 60.77% for DPPH and 94.03% for ABTS radicals), and excellent sustained-release effect. Strawberry preservation experiment further confirmed that the composite film can inhibit microbial growth and reduce water loss, thereby extending the shelf life of fruits. This study provides valuable insights for advancing green packaging technologies and promoting the sustainable utilization of food processing byproducts. Bio-based thiol-ene photo-cured resins derived from resveratrol and eugenol with excellent mechanical, flame retardant and dielectric properties

Yunran Zhang,Guangjian Dai,Aoyun Kang,Wenjie Yang,Heng Zhang,Chunxiang Wei,San-E Zhu,Hongdian Lu,Bin Yu,Wei Yang

Thiol-ene photocurable resins have demonstrated significant potential for applications in flexible electronic packaging owing to their tunable mechanical properties, low polymerization energy requirements, and excellent dielectric characteristics. However, most of these resins are flammable with lots of fire hazards. To address this issue, we employed resveratrol, rich in aromatic ring structures and hydroxyl functional groups, to synthesize a biomass-derived monomer with carbon-carbon double bonds, namely RST. In addition, eugenol was used to react with hexachlorocyclotriphosphazene (HCCTP), yielding a reactive bio-based flame retardant additive (HECTP). It was cross-linked with pentaerythritol tetrakis(3-mercaptopropionate) (PETMP) containing four thiol groups to prepare RST/4SH/HECTP resins. The results showed that the addition of HECTP remarkably enhanced the char formation ability and self-extinguishing performance. In particular, the total heat release (THR) declined from 19.2 kJ/g for RST/4SH to 13.6 kJ/g for RST/4SH/HECTP10 % with a reduction of 29.2 %. The loading of HECTP also enhanced the tensile strength and storage modulus of RST/4SH due to the improved crosslinking density of RST/4SH/HECTP resins. Moreover, the dielectric constant of RST/4SH was significantly reduced with the addition of HECTP. For RST/4SH/HECTP10 %, there was a dielectric constant of 1.78 that was lower than most reported results, highlighting its potential for electronic packaging materials.

Synergistic Antibacterial and Pro-Healing Effects of a Novel Eugenol/Nano-Haliotidis Concha Electrospun Membrane for Vibrio vulnificus-Infected Wound

Fuyu Zhao,Xianjun Fu,Wuyi Zhou,Xia Ren

Wounds caused byVibrio vulnificus(V. vulnificus) infection often exhibit delayed healing and are prone to complications, making them a significant challenge in clinical treatment. Current conventional treatments, such as antibiotics and gauze dressings, have limited effectiveness. To address this, this study developed a multifunctional fiber membrane using electrospinning technology. Micron- or nano-sizedHaliotidis Concha(HC) and eugenol (Eu) were loaded onto the membrane to promote healing inV. vulnificus-infected wounds. The prepared fiber membranes exhibited diameters of approximately 0.35 ± 0.01 μm. Membranes loaded with nano-HC demonstrated significant antibacterial efficacy, achieving a 96.2% inhibition rate againstV. vulnificus, which was markedly superior to the micron-HC group (p< 0.05). Notably, the nano-HC/Eu membranes exhibited exceptionally high flexibility with an elongation at break of 878.1 ± 35.3%, while maintaining a tensile strength of approximately 2.2 MPa. Furthermore, these membranes exhibited excellent biocompatibility, with cell viability exceeding 85% for fibroblasts, and demonstrated good hemocompatibility. They also effectively promoted cell migration, indicating their potential as wound scaffold materials. In aV. vulnificus-infected skin wound model, the nano-HC/Eu fiber membrane accelerated collagen deposition and promoted wound healing, achieving a wound closure rate of 94.7 ± 1.1% on day 15. In summary, this study developed a multifunctional fiber membrane with antibacterial, antioxidant, and wound healing properties, offering a novel dressing for treatingV. vulnificusinfections.

Qianqian Zhou,Peiyun Li,Shiyuan Fang,Wenru Liu,Jun Mei,Jing Xie

This research was to evaluate the effects of gelatin (G) active coating containing eugenol/β-cyclodextrin (βCD) emulsions combined with superchilling (−0.9 °C) on physicochemical, microbiological, and organoleptic properties of Chinese seabass samples during 30 days of storage. Results showed that seabass samples dipped in G-βCD coatings containing 0.15% or 0.3% eugenol combined with superchilling could significantly lower the total volatile basic nitrogen, K value, total viable count, H2S-producing bacteria,Pseudomonasspp. and Psychrophilic counts, and free fatty acids. Further, G-βCD coatings containing eugenol with superchilling (−0.9 °C) were more effective in retarding the water migration by low field NMR and MRI results, maintaining quality of seabass during storage according to organoleptic evaluation results. Keywords:Chinese seabass;superchilling;active coating;eugenol;shelf life

Electrochemical determination of eugenol using a three-dimensional molecularly imprinted poly (p-aminothiophenol-co-p-aminobenzoic acids) film modified electrode

Lite Yang,Faqiong Zhao,Baizhao Zeng

A novel electrochemical sensor for eugenol determination was presented, which was composed of molecularly imprinted poly ( p -aminobenzenethiol-co- p -aminobenzoic acid) and ionic liquid (IL, i.e. 1-aminopropyl-3-methylimidazolium bromide) functionalized graphene-carbon nanotubes composite (GN-CNTs-IL). The porous molecularly imprinted copolymer film was prepared on the GN-CNTs-IL by cyclic voltammetry. The template molecule was eugenol, the supporting electrolyte was 25   mM tetrabutylammonium perchlorate ethanol solution, and the concentration ratio of p -aminothiophenol and p -aminobenzoic acid was 1:3. The 3D porous GN-CNTs-IL composite was synthesized through a simple one-step hydrothermal method. After optimizing the preparation and measurement conditions, the obtained sensor showed a linear response to eugenol in the concentration range of 5.0   ×   10 −7 to 2.0   ×   10 −5   M, and the limit of detection was 1.0   ×   10 −7   M (S/N   =   3). The sensor had good selectivity, 100-fold glucose, lactose and sucrose did not affect the detection of 5.0   ×   10 −6   M eugenol, while 10-fold analogues (i. e. vanillin, methyl eugenol, paeonol and methyl isoeugenol) had no interference. In addition, it also displayed good reproducibility and practicability, and was successfully applied to detect eugenol in real samples.

UPLC-Q-TOF/MS-Based Plasma Metabolomics to Evaluate the Effects of Aspirin Eugenol Ester on Blood Stasis in Rats

Dongshuai Shen,Ning Ma,Yajun Yang,Xiwang Liu,Zhe Qin,Shihong Li,Zenghua Jiao,Xiaojun Kong,Jianyong Li

Aspirin eugenol ester (AEE) is a novel compound that is formed from the esterification of aspirin (acetylsalicylic acid (ASA)) and eugenol. This study aimed to investigate the effects of AEE on blood stasis in rats and to characterize the underlying mechanisms using a plasma metabolomic study. The results indicate that AEE and ASA could modulate whole blood viscosity (WBV), plasma viscosity (PV), blood coagulation parameters, platelet count, platelet aggregation, lactate dehydrogenase (LDH), creatinine (CR) and the levels of thromboxane A2 (TXA2) and 6-keto prostaglandin F1α (6-keto-PGF1α). The metabolic profiles of the plasma samples from all groups were clearly separated in the score plots. Nineteen potential metabolites were selected and identified, and disordered levels of these metabolites could be regulated by AEE and ASA. Pathway analysis showed that the mechanism of action of AEE on blood stasis might be principally related to the metabolism of amino acid, fatty acid, energy and glycerophospholipid. The above results indicate that AEE protected the rats against blood stasis, and that this effect might have been caused by the anticoagulation activity of AEE and its abilities to maintain a balance between TXA2and PGI2, reduce blood viscosity, inhibit platelet aggregation and normalize the plasma metabolic profile. Keywords:blood stasis;AEE;aspirin;metabolomics;UPLC-Q-TOF/MS

A Promising Plant-Based Eugenol-Loaded Nano Delivery System (EUG@CMC-PGMA-CS) for Enhanced Antibacterial and Insect Repellent Behavior

Jihao Zuo,Ruopeng Lan,Ningning Lv,Yitong Lin,Li Hao,Xinhua Zhou,Hongjun Zhou

Pathogens and pests pose significant threats to global crop productivity and plant immunity, necessitating urgent measures from researchers to prevent pathogen contamination and pest damage to crops. A natural plant-based antibacterial agent, eugenol (EUG), has demonstrated excellent antimicrobial and insect repellent capabilities, but the characteristics of volatilization and poor dissolution limit the practical application. The nanoization of pesticide formulations holds promise in the development of highly effective pesticides for antibacterial and insecticidal purposes. Herein, a eugenol-loaded nano delivery system (EUG@CMC-PGMA-CS) was synthesized using glycidyl methacrylate (GMA) as a functional monomer to connect carrier core structure carboxymethyl cellulose (CMC) with shell structure chitosan (CS), and EUG was encapsulated within the carrier. EUG@CMC-PGMA-CS demonstrated excellent leaf affinity, with minimum contact angles (CAs) of 37.83 and 70.52° on hydrophilic and hydrophobic vegetable leaf surfaces, respectively. Moreover, the maximum liquid holding capacity (LHC) of EUG@CMC-PGMA-CS on both hydrophilic and hydrophobic vegetable leaf surfaces demonstrates a noteworthy 55.24% enhancement compared to the LHC of pure EUG. The in vitro release curve of EUG@CMC-PGMA-CS exhibited an initial burst followed by stable sustained release. It is with satisfaction that the nano delivery system demonstrated exceptional antibacterial properties against S. aureus and satisfactory insecticidal efficacy against Spodoptera litura. The development of this eugenol-loaded nano delivery system holds significant potential for enhanced antibacterial and insect repellents in agriculture, paving the way for the application of volatile bioactive substances.

Sustainable and Biodegradable Copolymers from SO2 and Renewable Eugenol: A Novel Urea Fertilizer Coating Material with Superio Slow Release Performance

Limin Liu,Yonghao Ni,Yunfei Zhi,Wenbo Zhao,Manoj Pudukudy,Qingming Jia,Shaoyun Shan,Kaifu Zhang,Xuefen Li

To enhance SO2 utilization, improve fertilizer use efficiency, and minimize the negative environmental impact, the novel slow release sulfur-containing urea fertilizers with good biodegradation performance were developed by coating with the sustainable poly(eugenol sulfone) derived from renewable eugenol and SO2. The poly(eugenol sulfone) was synthesized by a simple free radical polymerization under mild conditions, and structural features of the synthesized copolymers were studied by various characterization techniques. Characterization results revealed that the copolymers exhibited the strict alternating copolymerization structures containing O═S═O. A set of systematically designed experiments were carried out to determine the influences of the amount of initiator, reaction time, and reaction temperature on the molecular structure, release, and biodegradation behavior of the coated fertilizers. The obtained results proved that the coated fertilizers showed excellent release and biodegradation features. Moreover, the release and biodegradation rate of the coated fertilizers can be adjusted by changing the molecular weight of poly(eugenol sulfone). In addition, the kinetic study on the slow release characteristics of poly(eugenol sulfone)-coated fertilizers showed that the best fitting effect was obtained by the Ritger–Peppas equation. This work offers a simple and useful strategy for designing sulfur-containing urea fertilizers with excellent slow release and biodegradation performance and provides a new route for sulfur recycling. In the future, the fertilizer will be deeply tested to evaluate their impact on plant growth, chemical, and biological soil properties.

Reusable magnetic molecular imprinted polymers based on magnetic graphene oxide for selective identification and detection of eugenol in environmental water samples

Sha Ou,Wu Yunfan,Dai Xincheng,Li Huiwen,Xu Yuan,Wang Ziwen,Zhang Pu

In this study, a reliable method for determining eugenol content in environmental water samples was established by combining magnetic solid-phase extraction with high-performance liquid chromatography. Magnetic molecular imprinted polymers MGO@MIPs were prepared through surface molecular imprinting technique with eugenol as the template molecule. The material displayed good superparamagnetic properties and magnetic responsiveness in favor of rapid separation. The adsorption properties of MGO@MIPs for eugenol were evaluated through adsorption kinetics and selectivity experiments. MGO@MIPs were found to have favorable reusability and obvious selectivity for eugenol. In addition, adsorption and elution conditions were investigated. Under optimal conditions, a linear relationship was obtained between the concentration of eugenol and its peak area in the range of 0.02–5 mg/L ( R 2  = 0.9998) and the limit of detection was 4.0 × 10 –6  mg/mL. The performance of the established method was assessed with the average recovery of 96.59–102.20% and the relative standard deviation (RSD) below 3.5%. The application of this method provides a new perspective for the separation, enrichment and detection of eugenol in water environment.
