Characterization of Gas-Phase Charge-Site Isomerism in Flavonoids and Flavonoid Glycosides by Cyclic Ion Mobility-Mass Spectrometry with Computational Validation

Liping Xu,Shurui Gao,Yanqiu Wang,Lin Zhang,Hongli Li,Yu Wang,David Da Yong Chen

Flavonoids exhibit high structural complexity not only from their diverse cores and substitutions but also from glycosylation with various glycans, leading to numerous isomeric species. This study employed cyclic ion mobility mass spectrometry (cIM-MS) to achieve high-resolution separation of flavonoid and flavonoid glycoside isomers with subtle structural differences. A multipass cIM strategy featuring adjustable drift path lengths significantly enhanced both resolution and operational flexibility. Notably, multiple ion mobility peaks were frequently observed for individual pure compounds, which were identified as charge-site isomers arising primarily from sodium adduction at distinct molecular sites. Computational chemistry and collision cross section (CCS) analysis were integrated to elucidate the three-dimensional configurations corresponding to these different mobility peaks. The results reveal that sodium ions, by coordination with multiple electronegative oxygen atoms within the molecule, stabilize specific gas-phase conformations, leading to distinct CCS values resolvable by ion mobility spectrometry. This work advances the understanding of charge-site isomerism by providing atomic-level insights into metal ion–molecule interactions. It also establishes an integrated cIM-MS and computational modeling framework, which substantially enhances analytical depth for complex isomeric systems.

High-throughput sequencing analysis reveals effects of short-term low-temperature storage on miRNA-mediated flavonoid accumulation in postharvest toon buds

Hu Zhao,Xiaopu Shi,Cheng Shen,Caifeng Chen,Jiyuan Liu,Changqing Qu

Toon bud is rich in polyphenols and flavonoids, which not only have nutritional value but also have health benefits. However, the molecular mechanisms of flavonoid accumulation during storage remain unclear. In our report, small RNA libraries from toon buds were synthesized after three storage period (0, 3, and 5 days) at 4 °C. Overall, 303 homologous sequences with known microRNA (conserved miRNAs homology) and 19 putative novel miRNAs were obtained using Illumina sequencing technology. Furthermore, the potential target unigenes of these miRNAs were predicted. Of the predicted miRNAs and their target unigenes, 17 conserved miRNAs and their target unigenes were confirmed by quantitative real-time PCR (qRT-PCR). Exactly 11 miRNAs involved in the flavonoid pathway and 6 miRNAs targeting transcription factors displayed significant correlation with flavonoid accumulation in toon buds. The findings will be helpful for better understanding flavonoid accumulation in toon buds during storage.

Exploration of the metabolic benefits and mechanisms of the flavonoid naringin on lipid homeostasis in high-fat-diet-induced obese mice

Yunzhen Zhang,Guoping Zhao,Cheng Xue,Wenlu Li,Yanbo Wang

Natural flavonoids present safe and effective therapeutic potential for obesity prevention and management. This study elucidates naringin's anti-obesity mechanisms using biochemical, microbiome, and metabolomic analyses in high-fat diet (HFD)-fed mice. Naringin significantly reduced weight gain, adiposity, serum TC/TG levels, hepatic lipid deposition, and adipocyte hypertrophy. It also improved intestinal barrier damage and chronic inflammation, which are closely related to the gut microbiome and its metabolites. Specifically, naringin supplementation modulated the gut microbiota structure, significantly increasing the relative abundance of Lachnospiraceae_NK4A136_group and norank_f__Muribaculaceae , and decreased the relative abundance of norank_f__Desulfovibrionaceae . Simultaneously, naringin supplementation regulated the levels of gut microbiota metabolites, increasing SCFA levels and improving intestinal barrier damage by decreasing serum LPS and D-lactate levels. Mechanistically, naringin activated the PPARα/CPT-1α pathway in the liver, promoting fatty acid oxidation while inhibiting lipid synthesis (SREBP-1c/FASN). In adipose tissue, it activated the AMPK/PGC-1α pathway, inducing white adipose tissue browning (UCP1) and energy expenditure. Untargeted metabolomics further revealed naringin's regulation of cholesterol-bile acid and tryptophan metabolism pathways. Collectively, these findings highlight naringin's multi-targeted potential for treating obesity and metabolic disorders.

A Plant-Derived Flavonoid, Isobavachin, Promotes Osteogenesis and Alleviates Glucocorticoid-Induced Osteoporosis via Modulation of the ESR1-PI3K/Akt Signaling Pathway

Jingran Cui,Xuting Song,Heran Liu,Zhenhai Cui,Mengmeng Sun,Min He,Meiying Jin

Background: Glucocorticoid-induced osteoporosis (GIOP) is marked by impaired osteogenesis and reduced bone formation. Isobavachin (IBA), a flavonoid from Psoralea corylifolia, shows multiple potentials in anti-inflammatory and bone metabolism regulations, but its effects against GIOP remain unclear. This study investigated the osteoprotective effects and potential mechanism of IBA using zebrafish GIOP model. Methods: osteoprotective effects of IBA was assessed by fluorescence imaging in a prednisolone-induced zebrafish model, following with osteogenic gene expressions measured by RT-qPCR. Potential targets and pathways of IBA was filtered and predicted by network pharmacology, molecular docking, and molecular dynamics (MD) simulations, and finally validated with a pharmacological rescue experiment using a PI3K-specific inhibitor. Results: IBA improved bone mineralization and upregulated osteogenesis-related genes. Network pharmacology identified the PI3K-Akt pathway as a key pathway, with ESR1, GSK3B, MTOR, and CCND1 as core targets. PI3K inhibition attenuated the osteoprotective effects of IBA and suppressed downstream osteogenic gene expression. Conclusions: IBA alleviates GIOP by modulating the ESR1-associated PI3K-Akt signaling pathway and may serve as a multi-target therapeutic candidate for osteoporosis.

Integrated Transcriptomic and Metabolomic Analysis Identified Key Transcriptional Factors Involved in Flavonoid and Alkaloid Biosynthesis Among Different Tissues of Sophora flavescens

Ake Liu,Jingjing Dong,Mingyang Wang,Jin Li,Junjie Lu,Lintao Liu,Haiying Lei,Zhen Zeng,Huifang Song

Sophora flavescenshas anti-inflammatory, analgesic, antibacterial, antiviral, and anti-tumor properties due to its active compounds, including alkaloids and flavonoids. Exploring the biosynthetic regulation mechanism of these compounds is crucial. Here, we identified 227 flavonoids and 55 alkaloids among five tissues (roots, stems, leaves, flowers, and pods) using wide-targeted metabolomics. Flavonoids were predominantly in roots, whereas alkaloids were primarily in roots and flowers. RNA sequencing revealed 18,488 differentially expressed genes (DEGs) in non-root tissues compared to roots. These DEGs were significantly enriched in pathways such as plant hormone signal transduction, carbon metabolism, flavonoid biosynthesis, and isoflavonoid biosynthesis. Utilizing K-means clustering and WGCNA, we identified ten transcription factors (TFs) potentially regulating the biosynthesis and accumulation of flavonoids (e.g., SfWRKY50 and SfbHLH078) and seven TFs involved in alkaloid biosynthesis (e.g., SfbHLH109 and SfbHLH162) inS. flavescens. These TFs can serve as candidate genes for studying the biosynthetic mechanisms of flavonoids and alkaloids, providing insights into the production of key active compounds and supporting the rational use ofS. flavescenstissues.

Immobilized Fe3O4-Polydopamine-Thermomyces lanuginosus Lipase-Catalyzed Acylation of Flavonoid Glycosides and Their Analogs: An Improved Insight Into Enzymic Substrate Recognition

Wang Zhaoyu,Li Yang,Li Mingyi,Zhang Xiaohui,Ji Qingxia,Zhao Xiaojuan,Bi Yanhong,Luo Si

The conversion of flavonoid glycosides and their analogs to their lipophilic ester derivatives was developed by nanobiocatalysts from immobilizing Thermomyces lanuginosus lipase (TLL) on polydopamine-functionalized magnetic Fe3O4 nanoparticles (Fe3O4-PDA-TLL). The behavior investigation revealed that Fe3O4-PDA-TLL exhibits a preference for long chain length fatty acids (i.e., C10 to C14) with higher reaction rates of 12.6–13.9 mM/h. Regarding the substrate specificity, Fe3O4-PDA-TLL showed good substrate spectrum and favorably functionalized the primary OH groups, suggesting that the steric hindrances impeded the secondary or phenolic hydroxyl groups of substrates into the bonding site of the active region of TLL to afford the product.

Study on the stability of four flavonoid glycoside components in Myrica Rubra pomace and their mechanism of in vitro hypoglycaemic activity

Tian Siyi,Chang Guoli,Xiang Yannan,Cai Chenggang,Luo Xinyu,Zhu Ruiyu,Yang Hailong,Gao Haiyan

In order to investigate the hypoglycaemic mechanism and potential applications of four hypoglycaemic flavonoid glycosides, namely myricitrin, Cyanidin-3-O-glucoside (C3G), hyperoside and quercitrin in Myrica rubra pomace, the stability of these four flavonoid glycosides and their binding mechanisms were studied using molecular docking. The results demonstrated that pH value affects on the stability of these four components in M. rubra pomace. C3G exhibited the most significant inhibitory effect on α-glucosidase at pH 5, with myricitrin, hyperoside and quercitrin showing the highest inhibitory effect at pH 7. Moreover, an increase in temperature and storage time reduced the inhibitory effect of these four glycosidic components on α-glucosidase. Molecular docking analysis revealed that myricitrin formed hydrogen bonds with the active site residues of α-glucosidase, namely Phe550, Ile552, Asp555, Ser574 and Arg576, and also engaged in hydrophobic interactions with Lys551. Hyperoside formed hydrogen bonds with α-glucosidase, formed hydrophobic interactions with Lys50 and exhibited π-cation interaction with Lys53. Quercitrin formed hydrogen bonds with α-glucosidase, formed hydrophobic interactions with Lys500 and established salt bridges with Lys50. C3G formed hydrogen bonds and hydrophobic interactions with α-glucosidase and showed π-π interactions with Phe301. These findings will provide valuable insights for the application of these four chemicals.

Genome-wide analysis of UDP-glycosyltransferase gene family and identification of members involved in flavonoid glucosylation in Chinese bayberry (Morella rubra)

Ren Chuanhong,Cao Yunlin,Xing Mengyun,Guo Yan,Li Jiajia,Xue Lei,Sun Chongde,Xu Changjie,Chen Kunsong,Li Xian

Glycosylation was catalyzed by UDP-glycosyltransferase (UGT) and was important for enriching diversity of flavonoids. Chinese bayberry (Morella rubra) has significant nutritional and medical values because of diverse natural flavonoid glycosides. However, information of UGT gene family was quite limited in M. rubra. In the present study, a total of 152 MrUGT genes clustered into 13 groups were identified in M. rubra genome. Among them, 139 MrUGT genes were marked on eight chromosomes and 13 members located on unmapped scaffolds. Gene duplication analysis indicated that expansion of MrUGT gene family was mainly forced by tandem and proximal duplication events. Gene expression patterns in different tissues and under UV-B treatment were analyzed by transcriptome. Cyanidin 3-O-glucoside (C3Glc) and quercetin 3-O-glucoside (Q3Glc) were two main flavonoid glucosides accumulated in M. rubra. UV-B treatment significantly induced C3Glc and Q3Glc accumulation in fruit. Based on comprehensively analysis of transcriptomic data and phylogenetic homology together with flavonoid accumulation patterns, MrUFGT (MrUGT78A26) and MrUGT72B67 were identified as UDP-glucosyltransferases. MrUFGT was mainly involved in C3Glc and Q3Glc accumulation in fruit, while MrUGT72B67 was mainly involved in Q3Glc accumulation in leaves and flowers. Gln375 and Gln391 were identified as important amino acids for glucosyl transfer activity of MrUFGT and MrUGT72B67 by site-directed mutagenesis, respectively. Transient expression in Nicotiana benthamiana tested the function of MrUFGT and MrUGT72B67 as glucosyltransferases. The present study provided valuable source for identification of functional UGTs involved in secondary metabolites biosynthesis in M. rubra.

Alkaloids and flavonoids were the major bioactive constituents from S. flavescens , which had been widely used for liver disease treatment in China. However, the liver-protective components of flavonoids from S. flavescens and their mechanism of action were not clear. Aim of the study This work aimed to evaluate the in vitro hepatoprotective activities of 35 flavonoids from S. flavescens and screen active compounds. Furthermore, it was conducted to demonstrate the hepatoprotective effects of a new active compound (kurarinol A, 1 ) was isolated by authors and the ethyl acetate (EtOAc) extract form S. flavescens against carbon tetrachloride (CCl 4 )-induced hepatic injury in Kunming (KM) mice, meanwhile revealed the potential mechanism. Materials and methods The 35 flavonoids from S. flavescens were co-incubated with HepG2 cells and treated with 0.35% CCl 4 for 6 h cell viability was measured by (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt) (MTS) assay. Then, in vivo animal experiments, the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) in the serum were analyzed, the degree of hepatic injury was examined using hematoxylin-eosin (H&E) staining, the mRNA expression of Superoxide Dismutase 2 (SOD2), Nuclear factor E2-related factor 2 (Nrf2), heme oxygenase 1 (HO-1), Interleukin 6 (IL-6), Tumor Necrosis Factor-α (TNF-α), interleukin-1β (IL-1β), and the protein levels of nuclear factor-kappa B p65/p-p65 (NF-κB p65/p-p65), toll-like receptor 2 (TLR2), IL-1β and cyclooxygenase-2 (COX2) in hepatic tissues were detected. Results The lavandulyl flavonoid (kurarinol A, 1 ) and the EtOAc extract from S. flavescens showed protective effects on CCl 4 -injured HepG2 cells, increasing cell viability from 24.5% to 61.3% and 91.8%, respectively. What’s more, we found that treatment with kurarinol A ( 1 ) and the EtOAc extract lead to a significant reduction in hepatotoxicity in response to acute CCl 4 exposure. Compared with the model group, experimental results exhibited kurarinol A (10 mg/kg, i.p.) and the EtOAc extract (300 mg/kg, i.p.) could decrease the levels of AST, ALT, ALP and tissue damage. Further mechanistic investigations revealed that up-regulated the mRNA expression of SOD2, Nrf2, OH-1 and down-regulated the IL-1β in liver tissues, respectively. Additionally, Western blot analyses elucidated that inhibition of IL-1β, TLR2, COX-2, NF-κB (p65/p-p65) via TLR2/NF-κB signaling pathway by kurarinol A and the EtOAc extract contribute to its hepatoprotective activity. Conclusion These findings demonstrated that the novel compound (kurarinol A, 1 ) possessed notable hepatoprotective activity against CCl 4 . It was confirmed that kurarinol A had a certain effect on mice with liver damage induced by CCl 4 , and its mechanism could be include inhibiting inflammation and reducing of oxidative stress reaction by regulating expression of related genes and proteins. Thus, kurarinol A could as a novel active agent that contributes to the hepatoprotective activity of S. flavescens for the treatment of live injury.

Ultrasensitive Determination of Natural Flavonoid Rutin Using an Electrochemical Sensor Based on Metal-Organic Framework CAU−1/Acidified Carbon Nanotubes Composites

Yuhong Li,Jianxiong Tang,Yueli Lin,Jiejun Li,Yaqi Yang,Pengcheng Zhao,Junjie Fei,Yixi Xie

Rutin, a natural flavonol glycoside, is widely present in plants and foods, such as black tea and wheat tea. The antioxidant and anti-inflammatory effects of flavonoids are well known. In this study, a new electrochemical rutin sensor was developed using multiwalled carbon nanotubes/aluminum-based metal–organic frameworks (MWCNT/CAU-1) (CAU−1, a type of Al-MOF) as the electrode modification material. The suspension of multiwalled carbon tubes was dropped on the surface of the GCE electrode to make MWCNT/GCEs, and CAU−1 was then attached to the electrode surface by electrodeposition. MWCNTs and CAU−1 were characterized using scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS). Due to the synergistic effect of CAU−1 and MWCNT-COOH, the prepared sensor showed an ultrasensitive electrochemical response to rutin. Under optimized conditions, the sensor showed a linear relationship between 1.0 × 10−9~3.0 × 10−6M with a detection limit of 6.7 × 10−10M (S/N = 3). The sensor also showed satisfactory stability and accuracy in the detection of real samples. Keywords:electroanalysis;pyrolysis;carbon materials;drug analysis;modified electrode

Comparing the acceptor promiscuity of a Rosa hybrida glucosyltransferase RhGT1 and an engineered microbial glucosyltransferase OleDPSA toward a small flavonoid library

Lu Wang,Weiqing Han,Chenying Xie,Jingli Hou,Qinghong Fang,Jianchun Gu,Peng George Wang,Jiansong Cheng

Glycosylation is a widespread modification of plant secondary metabolites, and catalyzed by a superfamily of enzymes called UDP-glycosyltransferases (UGTs). UGTs are often involved in late biosynthetic steps and show broad substrate specificity or regioselectivity. In this study, the acceptor promiscuity of a Rosa hybrid UGT RhGT1 and an evolved microbial UGT OleD PSA toward a small flavonoid library was probed and compared. Interestingly, RhGT1 showed comparable acceptor promiscuity in comparison with OleD PSA , though the acceptor binding pocket of the latter is much more open and large. This clearly indicates that stabilization of the acceptor position by suitable hydrophobic interactions is important for the specificity or regioselectivity determination as well as overall fit of the acceptor into a ‘big enough’ binding pocket. This also poses a challenge for structure-based UGT engineering to alter the glucosylation pattern of flavonoids.

Integration of covalent organic frameworks and molecularly imprinted polymers for selective extraction of flavonoid naringenin from grapefruit (Citrus × paradisi Macf.) peels

Xiumei Chen,Yingying Sheng,Jinxin Che,Okwong Oketch Reymick,Nengguo Tao

Grapefruit ( Citrus × paradisi Macf.) peel, a by-product of the citrus-processing industry, possesses an important economic value due to the richness of bioactive compounds. In this study, boron-linked covalent organic frameworks integrated with molecularly imprinted polymers (CMIPs) were developed via a facile one-pot bulk polymerization approach for the selective extraction of naringenin from grapefruit peel extract. The obtained CMIPs possessed a three-dimensional network structure with uniform pore size distribution, large surface areas (476 m 2 /g), and high crystallinity. Benefiting from the hybrid functional monomer APTES-MAA, the acylamino group can coordinate with the boronate ligands of the boroxine-based framework to form B-N bands, facilitating the integration of imprinted cavities with the aromatic skeleton. The composite materials exhibited a high adsorption capacity of 153.65 mg/g, and a short adsorption equilibrium time of 30 min for naringenin, together with favorable selectivity towards other flavonoid analogues. Additionally, the CMIPs captured the template molecules through π–π* interaction and hydrogen bonding, as verified by FT-IR and XPS. Furthermore, they had good performance when employed to enrich naringenin in grapefruit peels extract compared with the common adsorbent materials including AB-8, D101, cationic exchange resin, and active carbon. This research highlights the potential of CMIPs composite materials as a promising alternative adsorbent for naringenin extraction from grapefruit peel.
