Insights into the enhanced adsorption of glyphosate by dissolved organic matter in farmland Mollisol: effects and mechanisms of action

Jiao Yaqi,Jia Junxin,Gu Jiaying,Wang Sa,Zhou Qin,Li Hui,Li Li

Dissolved organic matter (DOM) is easy to combine with residual pesticides and affect their morphology and environmental behavior. Given that the binding mechanism between DOM and the typical herbicide glyphosate in soil is not yet clear, this study used adsorption experiments, multispectral techniques, density functional theory, and pot experiments to reveal the interaction mechanism between DOM and glyphosate on Mollisol in farmland and their impact on the environment. The results show that the adsorption of glyphosate by Mollisol is a multilayer heterogeneous chemical adsorption process. After adding DOM, due to the early formation of DOM and glyphosate complex, the adsorption process gradually became dominated by single-layer chemical adsorption, and the adsorption capacity increased by 1.06 times. Glyphosate can quench the endogenous fluorescence of humic substances through a static quenching process dominated by hydrogen bonds and van der Waals forces, and instead enhance the fluorescence intensity of protein substances by affecting the molecular environment of protein molecules. The binding of glyphosate to protein is earlier, of which affinity stronger than that of humic acid. In this process, two main functional groups (C–O in aromatic groups and C–O in alcohols, ethers and esters) exist at the binding sites of glyphosate and DOM. Moreover, the complexation of DOM and glyphosate can effectively alleviate the negative impact of glyphosate on the soil. This study has certain theoretical guidance significance for understanding the environmental behavior of glyphosate and improving the sustainable utilization of Mollisol. Graphical abstract

Dual beneficial activities of Paenibacillus ottowii NPDY10: glyphosate degradation, modulation of soil microbial communities, and antagonism against phytopathogenic fungi

Deng Yun,Niu Yuqing,Xu Qing,Xie DongRong,Su Yan,Liu DuanHua,Chen Yong-He,Han YongHe,Tian Dagang

Background The widespread use and environmental persistence of the herbicide glyphosate raise significant concerns for human and ecological health. Developing efficient and sustainable remediation strategies is therefore crucial. Microbial degradation and biocontrol are two promising approaches. This study aimed to isolate and characterize a bacterial strain capable of both degrading glyphosate and antagonizing soil-borne fungal pathogens, and to evaluate its impact on soil microbial communities. Results This study isolated and characterized a novel bacterial strain, NPDY10, identified as Paenibacillus ottowii , which exhibits dual beneficial functions: efficient glyphosate degradation and broad-spectrum antagonism against soil-borne fungal pathogens. Under optimal conditions (pH 7–7.5, 30–35 °C), strain NPDY10 completely degraded 400 mg L⁻¹ of glyphosate within four days, utilizing the herbicide as a carbon source. Metabolic profiling via UPLC-MS/MS confirmed glyphosate degradation via the aminomethylphosphonic acid (AMPA) pathway In a rice paddy field trial, the application of NPDY10 reduced glyphosate levels in paddy water, and a visible foam layer formed, serving as a visual indicator of active metabolism. Furthermore, strain NPDY10 demonstrated broad-spectrum antifungal activity in vitro, inhibiting 17 plant-pathogenic fungi with inhibition rates ranging from 60.00% to 92.00%. Analysis of soil microbial communities in a subsequent maize cultivation season revealed that NPDY10 treatment increased bacterial diversity and suppressed the relative abundance of pathogenic fungal genera such as Fusarium and Gibberella . Conclusion Paenibacillus ottowii NPDY10 represents a promising multi-functional agent for sustainable agriculture, potentially combining glyphosate bioremediation with direct and indirect biocontrol effects through modulation of soil microbial communities, thereby contributing to improved soil health.

An advanced ratiometric molecularly imprinted sensor based on metal ion reoxidation for indirect and ultrasensitive glyphosate detection in fruit

Xiaoyu Su,Kaiyi Zheng,Xiaoyu Tian,Xuan Zhou,Xiaobo Zou,Xuechao Xu,Zongbao Sun,Wen Zhang

An indirect and ultrasensitive ratiometric molecularly imprinted (MIP) sensor, based on metal ion reoxidation, is introduced for glyphosate (GLY) determination in fruit. As high-performance signal amplification substrates, carbon nanotubes (MWCNTs) and gold nanoparticles (AuNPs) are conveniently modified on GCE. The artificial antibody-MIP membrane, presents typical three-dimensional structure to GLY template. Built-in reference methylene blue (MB) is directly electropolymerized on MWCNTs-Au/GCE. Particularly, Cu 2+ and GLY interestingly form chelate complex, and the Cu 2+ ( I Cu ) in Cu(Ⅱ)-GLY-complex can be reoxidized, and indirectly quantizes GLY. The reference signal ( I MB ) presents noteworthy stability with different GLY levels, and the ratiometric readout ( I Cu / I MB ) is recognized as a more trustworthy indicator to quantize GLY. Proposed sensor presents broad range as 1.73 ∼ 400 ng/mL, and limit of detection is well found as 0.24 ng/mL (S/N = 3). Finally, as-fabricated method is verified with standard HPLC in real-fruit-sample, and the errors and recovery rates are calculated as 3.4% ∼ 6.7% and 94.4% ∼ 104.6%, respectively.

A Turn-On Fluorescence Sensor Based on Nitrogen-Doped Carbon Dots and Cu2+ for Sensitively and Selectively Sensing Glyphosate

Ziqiang Li,Shuang Liang,Li Zhou,Fengjian Luo,Zhengyun Lou,Zongmao Chen,Xinzhong Zhang,Mei Yang

Glyphosate has excellent herbicidal activity, and its extensive use may induce residue in the environment and enter into humans living through the food chain, causing negative impact. Here, water-soluble 1.55 nm size nitrogen-doped carbon quantum dots (NCDs) with strong blue fluorescence were synthesized using sodium citrate and adenine. The maximum excitation and emission wavelengths of NCDs were 380 nm and 440 nm, respectively. The above synthesized NCDs were first used for the construction of a fluorescence sensor for glyphosate detection. It was found that Cu2+could quench the fluorescence of NCDs effectively through the photoinduced electron transfer (PET) process, which was confirmed using fluorescence lifetime measurements. Additionally, the fluorescence was restored with the addition of glyphosate. Hence, a sensitive turn-on fluorescence sensor based on NCDs/Cu2+for glyphosate analysis was developed. The LODs of glyphosate for water and rice samples were recorded as 0.021 μg/mL and 0.049 μg/mL, respectively. The sensor was applied successfully for ultrasensitive and selective detection of glyphosate in environmental water and rice samples with satisfied recoveries from 82.1% to 113.0% using a simple sample pretreatment technique. The proposed strategy can provide a significant potential for monitoring glyphosate residue in water and agricultural product samples. Keywords:carbon quantum dots;fluorescence;glyphosate detection;water;rice

Sensitive and Label-Free Colorimetric Detection of Glyphosate Based on the Suppression Peroxidase-Mimicking Activity of Cu(II) Ions

Qing Li,Yumeng Guo,Xiangyi He,Guangli Li

The sensitive and accurate determination of glyphosate (Glyp) is urgently demanded because it is closely correlated with human health and environmental safety. In this work, we proposed a sensitive and convenient colorimetric assay by employing copper ion peroxidases for the detection of Glyp in the environment. Free Cu(II) ions displayed high peroxidase activity and can catalytically oxidize the colorless 3,3′,5,5′-tetramethylbenzidine (TMB) into blue oxTMB, resulting in an obviously visible discoloration reaction. Once the Glyp is added, the ability of copper ions to mimic peroxidase can be largely suppressed because of the generation of Glyp–Cu2+chelate. The favorable selectivity and sensitivity were demonstrated in the colorimetric analysis of Glyp. Furthermore, this rapid and sensitive method was successfully applied in the accurate and reliable determination of glyphosate in the real sample, holding promising applications in pesticide determination in the environment.Keywords:label-free;colorimetric analysis;copper ions;peroxidase-mimicking activity;glyphosate

A ratio fluorescence sensor based on rhodamine B embedded metal-organic framework for glyphosate detection in agri-food products

Chao-Qun Wan,Yue-Hong Pang,Yong-Wei Feng,Xiao-Fang Shen

Glyphosate, a broad-spectrum and high-efficiency herbicide, could accumulate in the human body through the consumption of agri-food products. Herein, a ratio fluorescence sensor based on rhodamine B-embedded amino-functionalized iron-based metal-organic framework (MOF, NH 2 -MIL-88(Fe)@RhB) bonded with Cu 2+ was developed for rapid detection of glyphosate. The synthesized NH 2 -MIL-88(Fe) was a biconical prism and had a cavity for the embedding of RhB as a reference compound. In the presence of Cu 2+ , Lewis interactions with NH 2 -MIL-88(Fe)@RhB cause the fluorescence signal to be turned off. When glyphosate was added, the signal was turned on due to chelation with Cu 2+ and hydrogen bonding interactions with NH 2 -MIL-88(Fe)@RhB. Under optimal conditions, the developed sensor exhibited a linear range of 0.60–45 μmol L −1 with a response time of less than 1 min. The sensor was applied in the analysis of agri-food products (tea, soybean, wheat, cucumber), with recoveries between 97.93% and 109.06%, indicating its promising application in agri-food safety.

A turn-on fluorescence sensor based on Cu2+ modulated DNA-templated silver nanoclusters for glyphosate detection and mechanism investigation

Yixia Yang,Behafarid Ghalandari,Liyun Lin,Xiao Sang,Wenqiong Su,Adeleh Divsalar,Xianting Ding

The abuse application of glyphosate can result in a potential hazard for environment and human, however its ultrasensitive detection remains challenging. Herein, a Cu 2+ modulated DNA-templated silver nanoclusters (DNA-AgNCs) sensor was constructed to sensitively determine glyphosate based on the turn-on fluorescence strategy. The fluorescence quenching of DNA-AgNCs occurred with the existence of Cu 2+ . Upon the presence of glyphosate, the functional groups on the surface of glyphosate could chelate with Cu 2+ , following the fluorescence recovery of DNA-AgNCs. Through the stoichiometric methods, we unveil that Cu 2+ -trigged fluorescence quenching mode is a combination of static and dynamic quenching with the static mode being predominant. In DNA-AgNCs/Cu 2+ system, the carboxylate, amine, and phosphonate groups of glyphosate interact with Cu 2+ through chelation, in which the carboxylate oxygen, the phosphonate oxygen atoms, and the monoprotonated secondary amine nitrogen atom and Cu 2+ form chelate rings. This fluorescence sensor showed a desired linearity of glyphosate analysis under the optimum conditions, ranging from 15 to 100 μg/L with a low detection down to 5 μg/L. Moreover, the proposed sensor was successfully utilized to measure glyphosate in real samples, indicating a promising application in pesticide residues detection.

A Rapid Detection Method for Atrazine, Diquat, and Glyphosate in Water Using Radar Fingerprint Spectra of Algal Photosynthetic Fluorescence Parameters

Ziqi Ye,Tingting Gan,Gaofang Yin,Nanjing Zhao,Xiaoxuan Tan,Letong Li,Xingchi Li,Yunfei Luo,Wenqing Liu

As typical pesticide pollutants in water environments, herbicides pose a serious threat to the safety of aquatic ecosystems. However, effective methods for rapid screening are still lacking at present. This study developed a stepwise prediction method for atrazine, diquat, and glyphosate using radar fingerprint spectra derived from algal photosynthetic fluorescence responses. Microcystis aeruginosa was selected as the indicator alga from four freshwater algae. Using a strategy of “clustering dimension reduction and comprehensive scoring”, 11 parameters were selected from 26 JIP test parameters, and the optimal exposure time was determined to be 60   min. Radar fingerprint spectra were constructed from the selected parameters, and radial, geometric, and template features were extracted. Four classification and four regression algorithms were evaluated across seven feature sets using nested cross validation. The selected combinations were integrated into a stepwise prediction method following an “identification first, then quantification” strategy. Nested cross validation yielded an identification accuracy of 91.11% and R² values of 0.979, 0.790, and 0.982 for atrazine, diquat, and glyphosate, respectively. The final method achieved an independent validation accuracy of 96.67%, and the corresponding LOQs were 1   μg∙L -1 , 1   mg∙L -1 , and 6   mg∙L -1 , respectively. In filtered actual water samples individually spiked with one target herbicide, mean recoveries ranged from 94.20% to 105.65%. This study provides a novel method for translating algal physiological stress responses into structured spectral features, thereby providing methodological support for rapid detection of herbicides and for emergency monitoring and early warning of water pollution.

Interfacial covalent assembly of CdTe QDs on Eu3+-modified NH2-UiO-66 for Ratiometric fluorescence detection and adsorption of glyphosate

Jun Chen,Kai Jiang,Tianxiang Hang,Ciyang Zhang,Ruijie Che,Mingzhu Xia,Wu Lei

A covalently coupled CdTe/NH₂-UiO-66(Eu) ratiometric probe was developed. • Adsorption experiments and IGM analysis indicated rapid glyphosate uptake mediated by multiple interfacial interactions. • Glyphosate enhanced blue emission and weakened green emission for ratiometric readout. • Lifetime, XPS, and TD-DFT analyses provided mechanistic insights into the fluorescence response. Rapid detection and effective adsorptive capture of glyphosate from water are important for aquatic environmental monitoring and agricultural product safety. To address the susceptibility of single-emission fluorescence signals to external fluctuations and the limited glyphosate-capture capability of conventional fluorescent probes, a dual-emission platform integrating glyphosate adsorption with ratiometric fluorescence detection was constructed through the covalent interfacial assembly of Eu 3+ -modified NH 2 -UiO-66 and mercaptopropionic acid-modified CdTe QDs. Adsorption experiments and independent gradient model (IGM) analysis indicated that the composite exhibited rapid glyphosate uptake mediated by multiple interfacial interactions. In the presence of glyphosate, CdTe/NH 2 -UiO-66(Eu) exhibits enhanced blue emission at 434 nm and weakened green emission at 524 nm, producing a pronounced I 434 /I 524 ratiometric response. The fluorescence intensity ratio shows a linear dependence on glyphosate concentration over the range of 0.5–80 μM, with a limit of detection of 0.217 μM. Based on fluorescence lifetime results and XPS analysis, the ratiometric fluorescence response mechanism of the probe was elucidated. TD-DFT calculations further suggested that glyphosate modifies the excited-state charge distribution and charge-transfer characteristics associated with the MOF component. This work provides an interfacial assembly strategy for constructing MOF-based platforms that combine adsorptive glyphosate capture with ratiometric fluorescence sensing. Tailoring diacid carbon chains of precursors to boost the peroxidase activity of carbon dot nanozymes for dual-mode glyphosate detection

Xiaomei Li,Yi Feng,Xue Huang,Wencai Jiang,Guilin Zou,Shiyu Long,Zhibo Hou,Yan Zhao,Guoqi Zhang

Background: carbon dot nanozymes (CDszymes) hold significant promise for sensing applications due to their excellent water solubility and intrinsic fluorescence. However, conventional approaches for synthesizing CDszymes with high peroxidase-like (POD) activity remain largely empirical, relying on trial-and-error precursor screening, which limits rational design and optimization. Results: carbon dot nanoenzymes (CDszymes) demonstrate potential applications in nanoenzymatic sensing due to their excellent solubility and intrinsic fluorescence. However, conventional methods for synthesizing CDszymes with high peroxidase (POD) activity rely on empirical precursor screening. In this study, CDszymes with optimal POD activity were rationally designed by regulating the carbon chain length of dicarboxylic acids. Among the six synthesized CDszymes, ethanedioic acid-derived CDszymes-2 exhibited the highest POD activity, which catalyzes the oxidation of TMB by H2O2 to produce blue oxidized TMB (oxTMB), alongside fluorescence quenching via an inner filter effect. Utilizing glyphosate's (GLY) inhibition of acetylcholinesterase (AChE), a dual-mode colorimetric/fluorescent sensing platform was established, attaining detection limits of 13 ng mL−1 (colorimetric) and 7 ng mL−1 (fluorescent). Dual-mode sensing was successfully applied to detect GLY in fruit and vegetable samples, demonstrating potential for ultrasensitive trace-level pesticide detection in agricultural products.

In-situ grown dual lanthanide metal-organic framework electrospinning nanofiber film for the ratiometric fluorescent sensing of glyphosate in foods

Licheng Yu,Yi Yang,Chenyang Wang,Cuiyu Huang,Xiwen He,Langxing Chen,Yukui Zhang

The extensive utilization of glyphosate (Gly) leads to food contamination that poses the potential threat to human health. Therefore, it is urgent need to develop high-sensitivity methods for Gly detection. Herein, a dual-lanthanide metal-organic framework (Ln-MOF) integrating electrospinning nanofiber membrane was fabricated via in-situ growth strategy for the ratiometric fluorescence sensing of Gly. This strategy equips the sensor with a porous structure, which is beneficial for the pre-concentration of low-concentration analytes. The binding of Gly and Tb 3+ weakened the ligand-to-metal energy transfer, leading to the luminescence decrease of Tb-MOF. Meanwhile, the luminescence of Eu-MOF remained unchanged due to the small channel, resulting in a ratiometric response. The sensor exhibited a good linearity in the concentration range of 0.1–20 μM with a detection limit of 32 nM, and achieved satisfactory performances in food detection. Collectively, the proposed sensor may provide a new inspiration and have great potential for practical applications.
