A rapid, simple, and stable detection method for streptomycin has been established based on the coloration reaction of 3,3′,5,5′-tetramethylbenzidine (TMB) catalyzed by peroxidase-mimicking DNAzymes (PMDs). It is discovered that the catalytic activity of PMDs constructed with G-quadruplex and hemin is weakened in the presence of streptomycin. In the presence of streptomycin, the OD 450nm value of the TMB colorimetric sample catalyzed by PMDs is significantly lower than that of the control sample (without streptomycin). Hence, the detection of streptomycin can be accomplished by detecting the change of OD 450nm (△OD 450nm ). Furthermore, when the concentration of the substrate TMB is lower, the catalytic activity of PMDs is inhibited more significantly by streptomycin. Under optimized conditions, this method has good detection specificity, could effectively distinguish streptomycin from other antibiotics (ampicillin, chloramphenicol, geneticin, and kanamycin), has a limit of quantitation (LOQ) of 5 mg/L, excellent linear performance, good repeatability, and a total detection time of less than 2 h, and can be employed for the detection of actual samples (laver and egg drop soup). This method circumvents the complex preparation process of experimental materials, is simple to operate, does not require fluorescent labeling, and is of low cost, providing a novel method for food safety supervision.

Facile synthesis of Hemin@PMo 12 via manual grinding, no complex equipment and harsh conditions required. • Solid-phase Hemin@PMo 12 enables easy recovery, good reusability and high stability. • Hemin@PMo 12 shows excellent peroxidase-mimicking activity, outperforming natural HRP in kinetic parameters. • The sensor achieves accurate AA detection in apple samples via UV–vis and smartphone analysis. Polyoxometalate-based nanocomposites, as promising peroxidase mimetics for biosensing, have drawn extensive attention. In this study, a facile manual grinding method was developed for the synthesis of Hemin@PMo 12 nanocomposites from hemin and phosphomolybdic acid (H 3 PMo 12 ), without complicated equipment or harsh reaction conditions. Hemin@PMo 12 exhibited excellent peroxidase-mimicking activity, capable of catalyzing the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H 2 O 2 ) to form a blue oxidized product. Ascorbic acid (AA) could specifically reduce the oxidized TMB, leading to a noticeable color fading and a significant decrease in absorbance, which enabled the colorimetric detection of AA. The proposed method based on Hemin@PMo 12 demonstrated a wide linear range of 3–200 μM and a low limit of detection (LOD) of 1.1 μM for AA detection. Furthermore, the developed sensor was successfully applied to the quantitative determination of AA in fresh fruit samples (e.g., apple), yielding satisfactory recoveries ranging from 98.4% to 102.7%. Notably, the solid-phase nature of Hemin@PMo 12 endowed it with prominent advantages of easy recovery from the reaction system and good reusability. These advantages well made up for the inherent drawbacks of traditional nanozymes, namely the difficulty in separation and the restriction to single-use applications. With the merits of simple synthesis, easy recovery, reusability, high sensitivity, and good selectivity, Hemin@PMo 12 provided a promising candidate for the development of practical, low-cost colorimetric sensors for AA detection in food analysis and related fields. p53 gene is one of the most important anticancer genes and has been recognized as a typical biomarker for the early diagnosis of cancers. In this paper, a portable point-of-care testing (POCT) platform was developed for the colorimetric and photothermal dual-modal analysis of the p53 gene. Long concatemer G4 structures with high catalytic activity were formed by a target-driven hybridization chain reaction, and their catalytic properties can further be enhanced with the help of iodide ions. This dual-modal method realized the sensitive multicolor visual point-of-care detection of the p53 gene, where color variations from colourless to blue were easily distinguished by the naked eye. The temperature output was achieved by effective photothermal conversion and regulation guided by the accumulated concatemer G4 complexes in the TMB–H2O2 system, avoiding the need for complex instruments. The proposed dual-modal assay for p53 gene detection is cost-effective, simple, highly sensitive (3.34 pM), stable, and remarkably selective. It was also applied to determine the concentration of the p53 gene in spiked serum samples.

Malathion is one of the most commonly used organophosphorus pesticides that can cause serious harm to the ecological environment and human health. Herein, we demonstrated a label-free chemiluminescent aptasensor for the sensitive detection of malathion based on exonuclease-assisted dual signal amplification and G-quadruplex/hemin DNAzyme. Upon the addition of malathion, the aptamer probe specifically bound to the target to form a complex malathion-S3, leaving a duplex S1-S2. The complex malathion-S3 was digested by exonuclease I and the target was released. The released target was recycled to perform exonuclease I-assisted signal amplification. Furthermore, after treatment with exonuclease III, the duplex S1-S2 was converted into the secondary target ST. The secondary target ST interacted with the hairpin H1 to form a complex H1-ST, which was further digested by exonuclease III and released the secondary target. The released secondary target was recycled to perform exonuclease III-assisted signal amplification. After complete amplification, large numbers of G-quadruplex/hemin DNAzymes were generated. Under the optimal experimental conditions, the prepared aptasensor showed an excellent linear response to malathion with a detection limit of 0.47 pM. The relative standard deviations were in the range of 4.2–6.9%. Moreover, the aptasensor was successfully applied to detect malathion in spiked food and traditional Chinese medicine samples.

A novel polymer-based monolithic column was prepared via redox initiation system within the confines of a stainless steel column with 4.6 mm i.d. In the processes, hemin and lauryl methacrylate were used as co-monomers; ethylene dimethacrylate as crosslinking agent; n-butyl alcohol, ethanediol, and N , N -dimethylformamide as tri-porogens; benzoyl peroxide and N , N -dimethyl aniline as redox initiation system. The resulting polymer-based monolithic columns were characterized by scanning electron microscopy, nitrogen adsorption-desorption instrument, and mercury intrusion porosimeter, respectively. The results illustrated that the improved monolith had relative uniform porous structure, good permeability, and low back pressure. Aromatic compounds were used to test the chromatographic behavior of the monolith, resulting in highest column efficiency of 19 880 plates per meter with reversed-phase mechanism. Furthermore, the homemade monolith was used as the stationary phase of high performance liquid chromatography to separate proteins from complex bio-matrix, including human plasma, egg white, and snailase. The results showed that the monolithic column occupied good separation ability with these complex bio-samples. Excellent specific character of the homemade hemin-based monolith was that it could simultaneously remove high-abundance proteins (including human serum albumin, immunoglobulin G, and human fibrinogen) from human plasma and separate other proteins to different fractions.

A novel colorimetric sensing platform based on the peroxidase activity of hemin regulated by oligonucleotide and pesticide was reported for the ultrasensitive and selective detection of isocarbophos. Oligonucleotides can accumulate on the surface of hemin in acid condition and temporarily inhibit its catalytic activity, which results in the loss of one electron of TMB molecule and produce the blue products. With the addition of isocarbophos, the pesticide molecules can interact with oligonucleotides to form some complexes, which relieve the inhibition of ssDNA to hemin and further enhance its catalytic activity. Thus, the TMB molecules are further oxidized to lose another electron and produce the yellow product in a few minutes, which has the characteristic absorption peak at 450 nm. The color change of the sensing system is related to the amount of isocarbophos, so this method can quickly discriminate whether the target pesticide exceeds the maximal residue limit just by naked eyes. To improve the performance of sensing platform, some important parameters like buffer condition and ssDNA have been investigated, and the peroxidase activity of hemin was further studied to verify the catalytic mechanism. The proposed sensing platform has a detection limit as low as 0.6 μg/L and displays good selectivity against other competitive pesticides. Moreover, the developed sensing platform also exhibits favorable accuracy and stability, indicating that it has potential applications in the detection of pesticide residues in agricultural products.

Recently, mimic enzymes have obtained particular interest by their high activity, stability, and biocompatibility. In this work, by coupling copper hydroxide nanozyme and G-quadruplex/hemin DNAzyme to form a double-integrated mimic enzyme, a visual, sensitive and selective immunosensor was established to detect microcystin-LR (MC-LR). In this immunoassay , the microplates were modified with core-shell silica/nickel silicate as the substrate to capture MC-LR antigens. Then, Cu(OH) 2 nanocages with fine regulation were used as the label to capture the secondary antibody for immunoreaction and the DNA primer for propagation, followed by using hybridization chain reaction to amplify the DNA primer, thus numerous DNAzymes (G-quadruplex/hemin) can be formed on the surface of Cu(OH) 2 nanocages with the aid of hemin . Such double-integrated mimic enzyme including Cu(OH) 2 nanozymes and DNAzymes showed excellent peroxidase activity for the chromogenic reaction of 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), which realized the visual detection of MC-LR in the range from 0.007 to 75 μg/L with the detection limit of 6 ng/mL, and thus provided the probability for the portable assessment of MC-LR in real sample.

Telomerase, a widely accepted cancer biomarker for early cancer diagnostics, is considered as an important therapeutic target. To now, it is still a challenging subject to develop a simple and sensitive strategy for telomerase activity detection. Herein, we reported a simple colorimetric strategy for label-free quantification of human telomerase activity in urine by using hemin-graphene nanomaterial (H-GNs). H-GNs possessed tailored dispersibility in the high salt concentration and highly active biomimetic oxidation catalyst property. In this strategy, H-GNs were adjusted to coagulate to appropriate degree by carefully selecting the contained NaCl amount in the presence of original TS primer. The supernatant of the solution contained few H-GNs and showed light blue color. Under the action of telomerase, TS primer was elongated with repeating sequences of (TTAGGG) n . These negatively charged DNA enhanced individual H-GNs electrostatic repulsion and resisted salt-induced H-GNs coagulation. As a result, the supernate of the corresponding solution containing more dispersed H-GNs and showed dark blue color after chromogenic reaction. Thus, telomerase activity could be quasi-quantified by naked eye and precise quantified by UV spectrometer. The proposed method has the linear range from 100 to 2300 HeLa cells/mL and the detection limit was 60 cells/mL. It has been successfully applied to detect telomerase activity in real urine samples. Obtained results were in good agreement with the clinical diagnosis. Therefore, this colorimetric approach affords simplicity, sensitivity and reliability in telomerase activity detection.

A nanocomposite nanozyme has been fabricated through mineralizing gold-silver bimetals into Hemin (Hem)-coupled melamine (MA) polymer matrix for visual colorimetric analysis of H 2 O 2 and glucose. Catalytic Hem was cross-linked onto MA scaffold for the mineralization of Au–Ag bimetals yielding the rod-like nanocomposite of MA-Hem/Au–Ag. It was discovered that the resulting nanocomposite could present high aqueous stability and especially improved catalysis, which was more than four-fold higher than that of native Hem. Catalytic kinetics studies indicate that the prepared nanocomposite nanozyme could present much higher affinities to the substrates than those of native Hem or even horseradish peroxidase . Herein, the so mineralized Au–Ag bimetals with the “silver effect” would act as “nanowires” for promoting the electron transferring of nanocomposite nanozyme. Moreover, the Hem-coupled MA polymer matrix with high specific surface area could ensure the high adsorption capacity for the reactant substrates and targeting analytes. The application feasibility of the developed nanocomposite nanozyme was demonstrated subsequently by the colorimetric assays for H 2 O 2 and glucose separately in milk and blood samples, with the linear ranges of 0.010–2.50 mM and 0.0050–2.0 mM, respectively. Such a bimetal mineralization-based fabrication route may open a new door toward the design of diverse nanocomposites nanozymes with improved catalysis and adsorption performances.

A highly sensitive, low-cost colorimetric aptasensor was developed for the determination of 8-hydroxy-2′-deoxyguanosine (8-OHdG) in human urine. The method is based on a conformational switching of the 8-OHdG aptamer to form a G-quadruplex structure in the presence of 8-OHdG. The resulting G-quadruplex assembles into a peroxidase-like DNAzyme with hemin , which effectively catalyzes the oxidation of 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS 2− ) by H 2 O 2 to ABTS + , resulting in an increase in the absorption signal at 416 nm along with a color change of the solution. The response signals linearly correlated with the concentration of 8-OHdG, ranging from 466 pM to 247 nM with a detection limit of 141 pM. The relative standard deviation and the recovery were 1.97–3.47% ( n = 11) and 98.8–100.2%, respectively. The proposed method avoids the label and derivatization steps in common methods and allows direct analysis of the samples by the naked eye without costly instruments, which is reliable, inexpensive, and sensitive.

Specific and cost-effective methodologies for human papillomavirus (HPV) gene detection are significant for clinical diagnosis and cancer control. Herein, a label-free and fluorimetric/colorimetric dual-mode sensing strategy was developed for the quantitative determination of HPV DNA based on the integration of fluorescent DNA-silver nanoclusters (DNA/AgNCs) and G-quadruplex/hemin DNAzyme. The fluorimetric sensing strategy was based on the phenomena that the fluorescence enhancement of DNA/AgNCs obtained in proximity of guanine-rich DNA sequences and the photoinduced electron transfer (PET) effect between the electron donor (DNA/AgNCs) and electron receptor (G-quadruplex/hemin). The colorimetric sensing strategy was relied on the peroxidase-like activity of G-quadruplex/hemin DNAzyme. By integrating DNA/AgNCs and DNAzyme, this dual-mode strategy could produce two independent signals to improve the analytical diversity and accuracy. Under optimized conditions, the fluorimetry and colorimetry of the strategy displayed a linear range of 0.01–4 and 0.02–4 nM, with the low detection limit of 2.3 and 5.2 pM, respectively. Additionally, this dual-mode strategy has been successfully applied to HPV DNA analysis in different real samples with excellent results. Moreover, the sensing platform could be developed for different HPVs DNA assay by only adjusting the recognition sequence, which provided a universal strategy for various kinds of virus analysis.

The stability, repeatability and sensitivity of an electrochemical biosensor material are closely connected with the dispersibility of metal organic frameworks (MOFs) in aqueous media. Herein, a nanocomposite based on Cu–MOF/hemin, which is not only highly water-soluble but also simple and efficient in synthesis, was used for the construction of a non-enzymatic sensor to detect hydrogen peroxide (H2O2). The Cu–MOF/hemin was characterized via scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDS)-mapping, X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), X-ray photoelectron spectroscopy (XPS) and thermal gravimetric analysis (TGA), which indicate that hemin and the Cu–MOF were successfully combined. As a H2O2 electrochemical biomimetic enzyme, the Cu–MOF/hemin exhibited excellent electrocatalytic performance, which was confirmed by the electrochemical experiments and chromogenic reactions, and the possible mechanism of the reactions has been deduced. The electrochemical sensor based on the biomimetic enzyme exhibited an extended linear detection range from 0.01–5.0 mM (R = 0.998), low detection limit of 4.14 μM, and high selectivity and stability under the optimized conditions. More importantly, the practical application ability of the sensor was verified by the test of H2O2 in human serum samples and it could be used for the real-time detection of H2O2 released from living cells with satisfactory results. Therefore, this novel nanocomposite has certain potential in preparing electrochemical sensing platforms for nonenzymatic biosensing and provides a new method for clinical diagnosis and real-time monitoring.
