Allium mongolicum Regel is prone to rapid senescence and quality deterioration during postharvest storage. Herein, polyvinyl alcohol/pullulan/ZnO nanoparticles (PVA/PUL/ZnO-Nps) composite films were prepared via solution casting and studied to analyze the effects of ZnO-Nps on the PVA/PUL film matrix. Results revealed that the incorporation of suitable ZnO-Nps effectively reduced the light transmittance, improved water contact angle, water vapor permeability, and mechanical properties of the composite films, as well as enhanced their antimicrobial activity. The composite films were used for the postharvest preservation of A. mongolicum Regel. Results revealed that the PVA/PUL/ZnO-Nps film effectively reduced malondialdehyde accumulation content, superoxide radical generation rate, hydrogen peroxide content, improve the activity of related enzymes, and extend the storage time compared with that of polyethylene films. Therefore, the PVA/PUL/ZnO-Nps film can be used as a novel packaging material for the postharvest preservation of A. mongolicum Regel.

This study fabricated novel smart colorimetric film from pullulan (P) and sodium alginate (SA) incorporated with anthocyanin (ACNs) loaded casein carboxymethylcellulose (CAS-CMC-ACNs) nanocomplex for the freshness monitoring of the fish and shrimp . The ACNs loaded nanocomplex exhibited large particle size (176.902 ± 4.641 nm) than CAS-CMC (155.454 ± 4.152 nm) nanocomplex without ACNs, and can be loaded in P/SA film with uniform distribution. The P/SA film incorporated with CAS-CMC-ACNs nanocomplex had a more compact structure, improved barrier properties, mechanical strength, and thermal stability than P/SA film incorporated with free ACNs, which could be attributed to hydrogen bonding among the components in the film matrix, as confirmed by SEM micrographs and FT-IR. Intriguingly, the composite film exhibited antibacterial effect against gram-negative bacteria E. coli ( Escherichia coli ) and gram-positive bacteria S. aureus (Staphylococcus aureus) with inhibition zone of 7 ± 0.283 mm and 8.6 ± 0.141 mm, and antioxidant effect. The composite film demonstrated a visible response to CO 2 and pH buffer. Furthermore, the composite smart film demonstrated the potential to monitor the freshness of fish and shrimp stored at 25 °C for 24 h via a visual color change from pink to dark grey. The findings revealed that the fabricated film is an effective tool for evaluating the freshness of fish and shrimp.

Loquat leaf extract (LLE) was added to guar gum and pullulan as an environmentally friendly packaging film (GPE) to preserve Chinese water chestnuts (CWCs). The effect of the amount of LLE on the guar gum/pullulan composite film was investigated. The optimal amount of LLE was 4% (GPE4), with lower water vapor permeability (WVP) and greater mechanical strength, antioxidant, and comparable antibacterial performance than many pullulan-based films. Upon packing the CWCs for 4 days, the weight loss rate of GPE4 was only 1.80 ± 0.05%. For GPE4, the POD activity, the soluble solid content, and the vitamin C (Vc) content of the CWCs were 21.61%, 36.16%, and 26.22% higher than those of the control sample, respectively. More importantly, GPE4 was effective in preserving the quality of CWCs after 4 days of storage, better or at least comparable to non-biodegradable plastic wrapping (PE). Therefore, it can be concluded that GPE films hold significant promise as a sustainable alternative packaging material for preserving fruit-based foods like CWCs, potentially replacing PE in the future.

Antimicrobial packaging is important for preventing food spoilage and extending shelf life. Herein, antimicrobial core-shell nanofiber films were fabricated via coaxial electrospinning with pullulan/nanochitin-nisin complexes as the core and gelatin-zein as the shell. Particle size and zeta potential analyses confirmed the successful formation of nanochitin-nisin complexes. Fourier-transform infrared spectrometry (FTIR) revealed binding via electrostatic interactions and hydrogen bonding. The suitable viscosity, conductivity, and surface tension of the spinning solutions ensured good spinnability, yielding uniform nanofibers, as observed using scanning electron microscopy. FTIR and X-ray diffraction analyses indicated that hydrogen bonding stabilized the core-shell structure without altering the crystallinity of biopolymers. The resulting films exhibited enhanced thermal stability, mechanical strength, reduced water vapor permeability (by ~40 %), and improved hydrophobicity. The antibacterial activities against Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922 were comparable to those of uniaxial nanofiber films. After 15 days of refrigerated storage at 4 °C, sea bass fillets wrapped in the films showed reductions in total volatile basic nitrogen, total viable counts, and thiobarbituric acid reactive substances by approximately 50 %, 90 %, and 75 %, respectively, compared to those wrapped in uniaxial nanofiber films. These findings indicate the potential of the core-shell nanofiber films for antimicrobial food packaging applications.

In this work, electrospun short nanofiber aerogel-templated oleogels (ESNF-ATO) were fabricated by electrospun pullulan nanofibers. Spinning solutions with different concentrations were utilized to maintain fiber diameter, and short nanofibers were homogenized to prepare aerogel templates for oil adsorption. The shape of the templates could be easily controlled, and the fabrication process unchanged the properties of pullulan and the diameter of short nanofibers. Increasing the fiber diameter resulted in more flexible aerogel templates, which exhibited lower compressive strengths but higher recovery rates in 100-cycle fatigue tests. Simple linear fitting further confirmed the above results. Oleogels could be formed by rapid oil adsorption (normally within 100 s), and capillarity mainly induced this procedure. The thinner nanofibers provided the templates a high oil adsorption capacity (up to 69.05 g/g) but a relatively weakened oil holding capacity. ESNF-ATO exhibited good rheological properties, and could effectively realize the sustained-release of free fatty acids or lipo-soluble compounds. The significance of this work lied in the innovativeness of the combination of electrospun short nanofibers and aerogel templates, and the characteristics of fiber-based templates and oleogels were adjustable by modulating the fiber diameter.

Combined treatment of drugs and therapeutic genes has emerged as a new modality of anticancer therapy. In this study, a new amphiphilic bifunctional pullulan derivative (named as PSP) containing stearic acid and low-molecular weight (1 kDa) branched polyethylenimine was prepared and evaluated as a nano-carrier for the co-delivery of a drug and gene for potential cancer therapy. The amphiphilic PSP could self-assemble into cationic core–shell nano-micelles in water, with a critical micelle concentration of around 58.9 mg L−1. PSP nanomicelles had an average size of 188.75 ± 3.18 nm, and a positive zeta potential of 17.83 ± 0.75 mV. The drug loading content and encapsulation efficiency of the PSP nanomicelles for doxorubicin (DOX), an anti-tumor drug, were about 5.10% and 56.07%, respectively, and DOX in PSP nanomicelles showed sustained release. The flow cytometry and confocal laser scanning microscopy showed that PSP/DOX nanomicelles could be successfully internalized by MCF-7 cells. The in vitro IC50 of PSP/DOX nanomicelles was slightly lower than that of free DOX against MCF-7 cells. Additionally, PSP nanomicelles condensed DNA efficiently to form compact structures, and induced comparable GFP gene expression level to Lipo2000 at N/P = 10 in gene transfection studies. In comparison with single DOX or p53 delivery, the co-delivery of DOX and therapy gene p53 using PSP micelles displayed higher cytotoxicity and induced a higher apoptosis rate of tumor cells in vitro. Moreover, PSP exhibited good blood compatibility and low cytotoxicity in the hemolysis and MTT assays, respectively. Altogether, PSP nanomicelles have a great potential in delivering hydrophobic anticancer drugs and therapeutic genes simultaneously for improved cancer therapy.

In this study, composite hydrogels were prepared by a simple synthetic technique to adsorb methylene blue from water. The hydrogel was composed of polyacrylamide (PAM) as the carrier, grafted with pullulan polysaccharide, and modified with graphene oxide. The morphology and properties of PUL/PAM/GO composites were characterized by Thermogravimetric Analysis (TGA), Fourier Transform Infrared (FTIR) spectroscopy, and Scanning Electron Microscopy (SEM). Then, the adsorption performance of PUL/PAM/GO hydrogel for MB was researched, including the dose, temperature, contact time, pH, adsorption isotherm, adsorption kinetics, and adsorption thermodynamics and swelling properties. The effects of various salts in real wastewater on adsorption were simulated. The removal rate of MB by PUL/PAM/GO was high-speed, and the sample can remove 83.2% of the dye in 140 min. The adsorption kinetics was more consistent with pseudo-second-order reaction, and the Langmuir model was suitable for describing the adsorption process. The maximum adsorption capacity was 438.7 mg g−1. Thermodynamic parameters covered the enthalpy changes (ΔH0), free energy (ΔG0), and entropy (ΔS0), which stated that adsorption by the PUL/PAM/GO hydrogels was exothermic and spontaneous.Graphical Abstract

The development of green, edible functional coating materials is crucial for addressing food safety issues. Traditional petroleum-based packaging materials, however, pose significant environmental pollution and resource waste due to their poor biodegradability. In this study, carvacrol (CA) encapsulated in β-cyclodextrin (β-CD) to form microcapsules (CMP) was integrated with a polysaccharide base (CTS/PUL) to create CP-CMP edible coatings. The physicochemical properties and microstructure of these films were evaluated using scanning electron microscopy, and fourier transform infrared spectroscopy. Successful incorporation of CMP into the CP coating via hydrogen bonding was indicated by changes in functional group vibrations, enhancing the barrier and thermal stability of the CP-CMP coating. The antioxidant capabilities of the coating, demonstrated by high DPPH (80.49 %) and ABTS (89.40 %) radical scavenging activities, were remarkable. Furthermore, it significantly inhibited Staphylococcus aureus and Escherichia coli . Applying the developed CP-CMP coating to fresh-cut green peppers extended shelf life from 8 days (control) to 12 days through dual actions: membrane-disrupting antimicrobial activity and integrated antioxidant functions that preserved color and retarded VC and chlorophyll degradation. Thus, the CP-CMP coating exhibits potent antioxidant and antibacterial properties, emerging as a promising alternative to traditional packaging materials. This innovative approach holds significant potential for reducing the environmental impact of white pollution.

Reducing the dosage of chemotherapeutic drugs via enhancing the delivery efficiency using novel nanoparticles has great potential for cancer treatment. Here, we focused on improving mitoxantrone delivery by using cholesterol-substituted pullulan polymers (CHPs) and selected a suitable nano-drug size to inhibit the growth of bladder cancer cells. We synthesized three kinds of CHPs, named CHP-1, CHP-2, CHP-3. Their chemical structures were identified by NMR, and the degree of cholesterol substitution was 6.82%, 5.78%, and 2.74%, respectively. Their diameters were 86.4, 162.30, and 222.28 nm. We tested the release rate of mitoxantrone in phosphate-buffered saline for 48 h: the release rate was 38.73%, 42.35%, and 58.89% for the three CHPs. The hydrophobic substitution degree in the polymer was associated with the self-assembly process of the nanoparticles, which affected their size and therefore drug release rate. The release of the three drug-loaded nanoparticles was significantly accelerated in acid release media. The larger the nanoparticle, the greater the drug release velocity. At 24 h, the IC 50 value was 0.25 M, for the best inhibition of mitoxantrone on bladder cancer cells. 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) experiments demonstrated that drug-loaded CHP-3 nanoparticles with the largest size were the most toxic to bladder cancer cells. Immunofluorescence and flow cytometry revealed that drug-loaded CHP-3 nanoparticles with the largest size had the strongest effect on promoting apoptosis of bladder cancer cells. Also, the three drug-loaded nanoparticles could all inhibit the migration of MB49 cells, with large-size CHP-3 nanoparticles having the most powerful inhibition.

Clove essential oil (CEO) exhibited potent antibacterial efficacy and are obtained from Eugenia caryophyllata tree flower buds. Herein, CEO nanoemulsions were prepared using various concentrations of casein protein treated with ultrasound for different time interval. The study demonstrated that CEO nanoemulsions with 5% casein protein subjected to ultrasound for 10 min displayed the most minimal particle size. The pullulan‑sodium alginate film incorporated with nanoemulsions treated with ultrasound exhibited enhanced physico-mechanical characteristics. Based on the structural analysis, the application of ultrasonic treatment improved intermolecular compatibility and organized molecular structure by strengthening hydrogen bonds . Furthermore, the composite film displayed remarkable efficacy against E. coli and S. aureus as well as longer retention of essential oils. The use of the developed films to protect cherry fruits and mushrooms produced promising results, emphasizing their potential in food packaging applications.

The escalating environmental burden of conventional plastic food packaging has intensified the search for sustainable, biodegradable alternatives. However, single-component polysaccharide edible films often exhibit suboptimal mechanical and barrier properties, which limits their practical applicability. In this study, pullulan (PUL), konjac glucomannan (KGM) and low-acyl gellan gum (LGG) were blended as film-forming substrates with glycerol as a plasticiser. Single-factor experiments combined with Box–Behnken response surface methodology (RSM) were employed to optimise the formulation of ternary edible composite films. The effects of PUL, KGM and LGG concentrations on mechanical properties and water vapour permeability (WVP) were systematically investigated. The optimised film was comprehensively characterised for microstructure, physical barrier performance, antioxidant activity, soil disintegration, bacteriostatic capacity and plum preservation efficiency. The optimal formulation comprised 1.43 g/100 mL PUL, 0.24 g/100 mL KGM and 0.48 g/100 mL LGG. Under these conditions, the composite film exhibited a transparent and smooth appearance, with a continuous and dense internal microstructure. The comprehensive performance score (K) reached 0.805 ± 0.013, with a WVP of 2.75 ± 0.17 g·μm/(m 2 ·h·kPa), light transmittance of 89.93% ± 1.26%, tensile strength (TS) of 3.05 ± 0.14 N/cm 2 , elongation at break (EB) of 64.57% ± 0.58% and DPPH radical scavenging activity of 34.12% ± 0.96%. Storage trials on two plum varieties demonstrated that the composite coating effectively mitigated fruit weight loss, retarded peel browning and pulp softening, and maintained superior sensory quality throughout storage. Overall, the PUL/KGM/LGG ternary edible composite film exhibited favourable mechanical strength, promising water vapour barrier properties and moderate antioxidant activity, suggesting its potential as a useful reference for the further development and industrial adoption of biodegradable edible films in fruit and vegetable preservation.

To address the limitations of high moisture sensitivity and lack of bioactivity in polysaccharide-based packaging, pullulan-cellulose nanocrystal (CNC) composite films were functionalized with a thymol-decanoic acid based hydrophobic deep eutectic solvent (TD-HDES). Two distinct fabrication strategies, mixing-hot pressing (MHP) and immersion-hot pressing (IHP), were developed and compared. At equivalent TD-HDES loadings, IHP-prepared films exhibited significantly superior water vapor barrier properties, surface hydrophobicity, and antioxidant and antibacterial properties compared to their MHP counterparts. Infrared spectroscopy and scanning electron microscopy revealed a surface accumulation of TD-HDES in IHP films, forming a distinct HDES-enriched surface layer driven by pressure- and temperature-induced migration. While TD-HDES incorporation reduced thermal stability without altering the crystalline structure of the pullulan-CNC matrix, the MHP method yielded higher mechanical strength and toughness due to better bulk integration. A shelf-life study demonstrated that IHP-fabricated films effectively preserved fresh rice noodles. After 4 days at 4°C, samples exhibited lower total plate counts, inhibited hardness and chewiness, and reduced cooking loss compared to polyethylene or pure pullulan-CNC films. This study highlights the potential of IHP-induced surface engineering for developing high-performance bioactive packaging for starch-based foods.
