A disease-targeting nanoplatform that integrates imaging with therapeutic activity would facilitate early diagnosis, treatment, and therapeutic monitoring. To this end, a macrophage membrane-coated Cu–WO3–x-Hydro820 (CWHM) nanoreactor was prepared. This reactor was shown to target inflammatory tissues. The reactive oxygen species (ROS) such as H2O2 and ·OH in inflammatory tissues can react with Hydro820 in the reactor to form the NIR fluorophore IR820. This process allowed photoacoustic/fluorescence dual-mode imaging of H2O2 and ·OH, and it is expected to permit visual diagnosis of inflammatory diseases. The Cu–WO3–x nanoparticles within the nanoreactor shown catalase and superoxide enzyme mimetic activity, allowing the nanoreactor to catalyze the decomposition of H2O2 and ·O2– in inflammatory cells of hepatic tissues in a mouse model of liver injury, thus alleviating the oxidative stress of damaged liver tissue. This nanoreactor illustrates a new strategy for the diagnosis and treatment of hepatitis and inflammatory liver injury.

A multidimensional colorimetric sensor array was produced based on a catalytic – regulated gold nanorod (Au NR) etching process as a receptor for protein detection. With the peroxidase-like property of gold nanosphere (Au NP) and H 2 O 2 , 3,3′,5,5′-tetramethylbenzidine (TMB) (colorless) is oxidized into TMB 2+ (yellow). Different concentrations of TMB 2+ promote different degrees of Au NR etching, and the localized surface plasmon resonance band of the Au NR is blueshifted. The blueshift results in color displays (gray, purple, blue and pink), which can be clearly distinguished by the naked eye. When the surfaces of the Au NP are covered by proteins, the catalytic activity is influenced. On this basis, the dynamically tunable system obtains triple channel information including the plasma resonance effect, the catalytic activity of Au NP and the etching of Au NR by TMB 2+ (catalytic products of TMB in the Au NP-H 2 O 2 reaction system), which can be adjusted using different proteins. The array can correctly identify native and thermally denatured proteins. This may provide a new method for studying conformational changes in biological macromolecules. Furthermore, pure protein (different concentrations) and binary protein mixtures can be efficiently differentiated in the urine sample.

Background As the major active component of turmeric (Curcuma longa), curcumin is widely used as a spice and food coloring agent, and also possesses multiple biological activities and therapeutic potential for neurodegenerative diseases. To answer the paradox between curcumin’s biological activities and poor systemic bioavailability, we proposed that degradation products of curcumin may make important contributions to its biological activities, which needs to be verified. In addition, curcumin is usually heated or boiled used as a spice, it is necessary to explore whether boiled curcumin, which degrades readily, is still biologically active. Methods Thus, in the present study we investigated the protective effects of curcumin and boiled curcumin mixture on H2O2-induced oxidative damage in PC12 cells, a widely used model for neurons. Results Results showed that in spite of high degradation rates, boiled curcumin mixture still possessed similar protective activities like parent curcumin, and could effectively rescue PC12 cells against H2O2-induced damage, via decreasing production of reactive oxygen species and malondialdehyde, reducing caspase-3 and caspase-9 activities. Moreover, curcumin’s degradation products including ferulic acid, vanillin and vanillic acid could also improve PC12 cells survival rate. Conclusion Our findings indicated that boiled curcumin mixtures still possessed protective activity for PC12 cells, and supported the contribution of degradation products to biological activities of curcumin.

Accurate and effective detection of single-stranded nucleic acids is vital in both disease diagnosis and pathological studies. Hence, we develop a PAMmer-assisted CRISPR/Cas9 system mediated G4-EXPAR (Cas-G4EX) strategy for site-specific detection of ssRNA and ssDNA . PAMmer-assisted CRISPR/Cas9 executes the site-specific cleavage of target ssRNA or ssDNA and released product fragment with the desired sequence at the 3′-terminal. This fragment serves as a primer to activate subsequent sequence-dependent exponential amplification reaction (EXPAR). The G-rich EXPAR products assembles with hemin to form a G-Quadruplex (G4/hemin). G4/hemin catalyzes ABTS-H 2 O 2 system with the appearance of vivid green color, realizing naked-eye analysis. Cas-G4EX integrates the superiority of CRISPR/Cas9 and EXPAR, presenting outstanding site-specific recognition and high-performance amplification efficiency. Meanwhile, the programmability of CRISPR/Cas9 system makes the proposed method become a universal detection paradigm for any ssRNA or ssDNA. Cas-G4EX assay shows the linear relationship from 250 aM to 2.5 nM for ssRNA detection with the actual LOD of 250 aM, and that ranges from 100 aM to 1 nM for ssDNA detection with the actual LOD of 100 aM. Additionally, the acceptable recoveries of 101.48%–109.61% for ssRNA and 93.25%–111.98% for ssDNA in real detection of human serum are obtained for detection of single-strand nucleic acid in real samples. Cas-G4EX also exhibits the excellent discrimination for single-base mutation of single-stranded nucleic acids. Therefore, Cas-G4EX assay provides a promising platform in the applications of molecular diagnosis and pathological analysis.

A series of Fe–Ba mixed oxides, including a pure Fe-containing sample as a reference, have been synthesized via a sol–gel process using Fe3+or Fe2+salts and BaSO4as raw materials, with Pluronic P123 serving as a template. These oxides have been thoroughly characterized and subsequently utilized as catalysts for the chlorination of various organic molecules. Commercial hydrochloric acid, known for its relative safety, and environmentally friendly aqueous hydrogen peroxide were employed as the chlorine source and oxidant, respectively. The pure Fe-containing catalyst displays excellent thermal stability between 600 and 800 °C and exhibited moderate to high conversions in the chlorination of toluene, benzene, andtert-butyl hydroperoxide, with remarkableortho-selectivity in chlorination of toluene. The combination of Fe3+salt with BaSO4in the sol–gel process results in a Fe–Ba mixed oxide catalyst composed of BaO2, BaFe4O7, and Fe2O3, significantly enhancing the chlorination activity compared to that displayed by the pure Fe catalyst. Notably, the chlorination oftert-butyl hydroperoxide (TBHP) does not require additional oxidants such as H2O2, and involves both electrophilic substitution and nucleophilic addition. Notably, the chlorination of bromobenzene yields chlorobenzene as the sole product, a transformation that has not been previously reported. Overall, this catalytic chlorination system holds promise for advancing the chlorination industry and enhancing pharmaceutical production.

Similar geometries caused difficulty in (MoO 4 ) 2− and (CO 3 ) 2− separation. • A target-reshaping separation strategy based on CDFT descriptors was proposed. • Aliquat 336 selectively recovers 99.81% Mo as [Mo(O 2 ) 4 ] 2− without acidification. • Alkaline raffinate is directly converted to SPC achieving acid free valorization. Fly ash from petroleum refining waste alkali liquor incineration represents a dual-component secondary resource rich in sodium carbonate and molybdates. Conventional solvent extraction or ion-exchange resin for Mo recovery were usually applied in acidic aqueous environment, and selective separation of Mo from this highly alkaline (pH > 10) solution of fly ash was heavily hindered by similar ionic radii and charge densities of MoO 4 2− and CO 3 2− . Herein, we report an acid-free Mo extraction strategy based on peroxo complexation induced ionic reshaping. By introducing H 2 O 2 into fly ash solution, MoO 4 2− is quantitatively converted into the bulky [Mo(O 2 ) 4 ] 2− complex, expanding the molecular volume to 140.20 Å 3 and elevating ionic softness to 0.31 eV −1 . Approximately 99.81% of Mo in solution was selectively extracted using extractant Aliquat 336, and yielded H 2 MoO 4 product with the purity of 99.65%. Moreover, Aliquat 336 exhibits the highest softness among the tested extractants, which results in a stronger affinity for the softened peroxo-reshaped Mo species. Conceptual density functional theory (CDFT) calculations and hard-soft acid-base analysis reveal that these geometric and electronic alterations dramatically enhance Mo orbital compatibility and binding energy (−5.725 eV) with extractant Aliquat 336. Subsequently, Mo depleted raffinate was subjected into the production of sodium percarbonate, and around 83.68% of sodium carbonate was recycled as sodium percarbonate (SPC) with total metallic impurities below 0.02%. Overall, the introduction of H 2 O 2 contributes to selective Mo recovery and simultaneous value enhancement of Na 2 CO 3 , and this method provides a sustainable strategy for the valorization of complex hazardous wastes. Developing a novel EPCS combining GOase with Au/TiO 2 . • Achieving ∼92.3% DFF yield with ∼97.0% selectivity within 2 h in pure water. • The by-product H 2 O 2 is in-situ decomposed into O 2 by Au/TiO 2 . • •OH, •O 2 – and O 2 precisely required for GOase activation generated by Au/TiO 2 . 2,5-Diformylfuran (DFF), as the oxidation product of the biomass platform molecule 5-hydroxymethylfurfural (HMF), has attracted considerable attention because of its broad utility as a precursor for pharmaceuticals, renewable polymers, and other functional products. Efficient oxidation of HMF into DFF through biocatalysis in pure water is extremely desirable but still faces significant challenges: complex multi-enzyme systems and low reaction efficiency. Herein, we develop a novel enzyme-photo-coupled catalytic system (EPCS) combining GOase with Au/TiO 2 photocatalyst, achieving “waste-to-wealth”. The mechanism study reveals that the Au/TiO 2 acts as the role of “killing two birds with one stone”: in-situ decomposing inhibitory H 2 O 2 by-product into O 2 and H 2 O, thereby mitigating enzyme inhibition; and ingeniously generating reactive oxygen species (ROS) (hydroxyl radicals (·OH) and superoxide radicals ( )) and regenerative O 2 that precisely are required for GOase activation. Under visible light conditions, the EPCS achieves (92.3±1.4)% DFF yield with (97.0±0.8)% selectivity within 2 h, significantly outperforming the single enzyme system (DFF yield: (40.9±2.4)%). The “green chemistry” concept is implemented in the EPCS, which provides a generalizable strategy for developing efficient, green, and concise oxidation systems by turning a toxic by-product into catalytic promoters. In conventional Electro-Fenton systems, the oxidation process dominated by the Fe 2+ /Fe 3+ cycle and · OH radicals, although effective in degrading organic pollutants, is still limited by a narrow pH applicability range (2–4). To address this challenge, a coral-like ZnO nanospheres modified Cu(OH) 2 @CF composite electrode was synthesized in this study, which significantly broadened the applicable pH range (2–10). The ZnO/Cu(OH) 2 @CF composite electrode enhanced the selective generation of single-linear oxygen ( 1 O 2 ) and overcame the inherent defects, such as the extremely short lifetime of · OH (<1 μs), while enabling the efficient in situ generation of H 2 O 2 (272.51 mg/L, 30 min). And the complete degradation of tigecycline (TGC) was realized within 30 min with an operating cost of only 0.044 ¥·m −3 (energy consumption 0.076 kWh·m −3 ). and a low Tafel slope of only 24.80 mV·dec -1 is required for an efficient Electro-Fenton reaction. Notably, the ZnO/Cu(OH) 2 @CF composite electrode exhibited high degradation rates for a variety of typical antibiotics and could be adapted to different water quality conditions. After 10 cycles of use, its degradation rate of TGC was still as high as 92.67 %. The continuous flow reactor not only maintained more than 90 % degradation efficiency for TGC after 300 min of operation, but also maintained a high degradation rate of 80 % for mixed antibiotic wastewater. The dominant mechanism of action of 1 O 2 was deeply analyzed by bursting experiments and electron paramagnetic resonance (EPR). The two-dimensional correlation spectroscopy (2DCOS), three-dimensional fluorescence spectroscopy (3DEEMs) and liquid chromatography-mass spectrometry (LC-MS) techniques were used to reveal the degradation pathways of the pollutants and the toxicity evolution of the intermediates.

For the mono-metal Pd based catalysts, the O–O bond in O2 and H2O2 is easily dissociated due to the higher energy sites of Pd, which decreases the selectivity and productivity of H2O2. To address this issue, non-noble metals M (M = Sn, Fe, Co, Ni) were doped into palladium precursors to create a series of yolk–shell structure bimetallic catalysts Pd–M@HCS (H = hollow; C = carbon; S = sphere) with Pd–M as the core and porous carbon as the shell. The results indicated that the catalyst Pd–M@HCS doped with Sn, Fe, Co, and Ni has a weak dissociation ability for O2, and both the adsorption and dissociation ability for H2 are improved. Of these catalysts, the catalyst Pd–Sn@HCS gives the best performance, which is benefited by the suitable Pd–Sn nanoparticle size and the best Pd2+/Pd0 ratio. The influence of the nPd/nSn ratio in the Pd–Sn@HCS system on catalyst performance was further explored, and the findings revealed that Pd–Sn@HCS had the best catalytic activity and stability when the nPd/nSn ratio was 2. The catalyst's H2 conversion, H2O2 selectivity, and productivity were 35%, 97%, and 3961 mmol gPd−1 h−1, respectively. After 10 cycles, the H2O2 productivity of this catalyst remained at 92.1%, which was greater than the H2O2 productivity of the typical Pd-based catalyst Pd/C, which stayed at 37% under the same conditions.

This study investigated the reparative effects of Brassica rapa L. ethanolic extract (BREE) on H 2 O 2 -induced oxidative injury in PC-12 cells. Brassica rapa L. , a cruciferous plant rich in bioactive flavonoids, exhibits potent antioxidant and neuroprotective properties against oxidative stress, a key pathological driver in neurodegenerative diseases. This study investigated the reparative effects of Brassica rapa L. ethanolic extract (BREE) on H 2 O 2 -induced oxidative injury in PC-12 cells, employing a multi-omics approach integrating LC-MS/MS metabolite profiling, molecular docking, and transcriptomic validation. BREE demonstrated robust radical-scavenging activity. Molecular docking identified direct interactions among flavonoids within BREE, suggesting a role in redox modulation. Subsequent experiments confirmed the activation of the PI3K/Akt, Keap1-Nrf2, and cell cycle pathways. Functionally, BREE significantly restored cell viability to 162.3% ± 6.9% of that in the control group. These findings validate BREE's dual action in scavenging ROS and reprogramming redox signaling networks, positioning it as a candidate for combating oxidative stress-associated neurodegeneration and as a potential functional food for age-related cognitive decline.

The photo-Fenton method presents a promising approach for wastewater treatment, yet it faces several significant challenges, including the continuous supply of H 2 O 2 , slow mass transfer rates and a limited operational pH range. In this context, we describe the successful preparation of a novel catalyst: n-p type graphitic carbon nitride nanosheets engineered with nitrogen vacancies and decorated with Cu 2 O-T, which is characterized by controllable crystal planes. This photo-self-Fenton-like catalyst is designed for the in-situ production and activation of H 2 O 2 . The optimized catalyst, CNC-20, demonstrates remarkable efficiency in degrading various organic pollutants, showcasing high cyclic stability and wide applicability. Notably, CNC-20 achieves a degradation rate of 98.8% for tetracycline hydrochloride within 60 min, with a reaction rate constant 4.2 times greater than that of pristine carbon nitride. Characterization studies illustrate that the construction of ultrathin carbon nitride nanosheets featuring nitrogen vacancies, combined with the co-exposure of Cu 2 O-T crystalline planes (111) and (100), significantly enhances carrier separation efficiency, thereby facilitating effective H 2 O 2 generation. Furthermore, Cu 2 O-T plays a crucial role in the direct capture of in-situ generated H 2 O 2 , rapidly activating it to generate abundant ·OH, which effectively mitigates the issue of slow mass transfer associated with H 2 O 2 . This study presents a novel approach integrating in-situ H 2 O 2 generation and activation, effectively overcoming the inherent limitations of traditional photo-Fenton reactions and providing a promising strategy for pollutant degradation in wastewater treatment systems.

Oral medication for ulcerative colitis (UC) is often hindered by challenges such as inadequate accumulation, limited penetration of mucus barriers, and the intricate task of mitigating excessive ROS and inflammatory cytokines. Here, we present a strategy involving sodium alginate microspheres (SAMs) incorporating M2 macrophage membrane (M2M)–coated Janus nanomotors (denominated as Motor@M2M) for targeted treatment of UC. SAM provides a protective barrier, ensuring that Motor@M2M withstands the harsh gastric milieu and exhibits controlled release. M2M enhances the targeting precision of nanomotors to inflammatory tissues and acts as a decoy for the neutralization of inflammatory cytokines. Catalytic decomposition of H2O2 by MnO2 in the oxidative microenvironment generates O2 bubbles, propelling Motor@M2M across the mucus barrier into inflamed colon tissues. Upon oral administration, Motor@M2M@SAM notably ameliorated UC severity, including inflammation mitigation, ROS scavenging, macrophage reprogramming, and restoration of the intestinal barrier and microbiota. Consequently, our investigation introduces a promising oral microsphere formulation of macrophage-biomimetic nanorobots, providing a promising approach for UC treatment.
