Bimetallic Fe/Mn metal-organic-frameworks and Au nanoparticles anchored carbon nanotubes as a peroxidase-like detection platform with increased active sites and enhanced electron transfer

Xueming Dang,Huimin Zhao

A hybrid of metal-organic frameworks (Fe, Mn) and Au nanoparticles anchored carbon nanotubes (Au/MOFs(Fe, Mn)/CNTs) was fabricated by a facile one-step hydrothermal process. The Au/MOFs(Fe, Mn)/CNTs exhibit highly enhanced peroxidase-like activity, because of the increased active sites, the enhanced partial charge density and electron transfer among the Fermi level of MOFs, Au and CNTs, as well as the synergistic action of Fe and Mn. The hybrid nanomaterials with high catalytic velocity (V max  = 17.65 × 10 −7  M s −1 ) and affinity (K m  = 0.33 mM) for substrates of H 2 O 2 display a great detection performance in the range of 0.34–53.05 nM with a limit of detection (LOD) of 0.18 nM (S/N = 3). Based on the cascade reaction of the artificial peroxidase and glucose oxidase (GOx), the glucose was further better detected in a linear range of 0.005–0.3 μM with a LOD of 0.002 μM (S/N = 3). Notably, considering the regulated peroxidase-like activity by sulfadimethoxine (SDM) aptamer , the concentration of SDM ranging from 0.54 to 41.58 μg L −1 was also detected with a LOD of 0.35 μg L −1  (S/N = 3). The as-synthesized hybrid nanozymes hold promising applications as the multifunctional sensing platform in environmental and biologic target detection with the combination of corresponding enzyme or specific aptamer.

Enhancing the peroxidase-like activity of ficin by rational blocking thiol groups for colorimetric detection of biothiols

Wen Zheng,Dongjun Shen,Yadi Pan,Danyang Yi,Yijuan Long,Huzhi Zheng

The peroxidase-like activity of ficin is relatively low, which limits its application. It was found that thiol groups of ficin could inhibit its peroxidase-like activity. So, two procedures, i.e., direct blocking with N -ethylmaleimide (NEM), or using tris (2-carboxyethyl) phosphine hydrochloride (TCEP) to interrupt disulfide bonds then blocking thiol groups with NEM, were applied to block thiol groups of ficin, ficin-NEM (ficin-N) and ficin-TCEP-NEM (ficin-TN) were produced, respectively. The blocking of thiol groups accelerated the peroxidase activity dramatically. The peroxidase catalytic activity of ficin-N and ficin-TN toward the peroxidase substrate 3,3′,5,5′-tetramethylbenzidine (TMB) oxidation by H 2 O 2 was about 2.5-fold and 5-fold increase compared with ficin, respectively, which accompanied a color change from colorless to blue and followed classic Michaelis-Menten model. The kinetic parameters indicated that higher affinity of ficin-N ( K m  = 0.31) and ficin-TN ( K m  = 0.39) to H 2 O 2 compared with ficin ( K m  = 0.58), and ficin-TN had the highest K cat which increased by 6.5 times and 4.5 times for TMB and H 2 O 2 , respectively. According to these findings, a colorimetric method with high sensitivity for the detection of biothiols was developed due to sulfhydryl compounds inhibited the peroxidase activity of ficin. Comparing with ficin and ficin-N, ficin-TN had the widest detection range (0.01–16 μM) and the lowest detection limit (3 nM). The practical applications of ficin-TN for biothiol determination in human serum samples have been demonstrated with satisfactory results. Ficin-N and ficin-TN are promising to apply to the bioanalysis.

A Novel Luminescent “Nanochip” as a Tandem Catalytic System for Chemiluminescent Detection of Sweat Glucose

Ya Gao,Yu Huang,Jianmei Chen,Yuhang Liu,Yurui Xu,Xinghai Ning

Accurate sweat glucose detection is a promising alternative to invasive finger-prick blood tests, allowing for self-monitoring of blood glucose with good patient compliance. In this study, we have developed a tandem catalytic system, termed as a luminescent “nanochip” (LAON), which was composed of gold nanoparticles (AuNPs) and N-(aminobutyl)-N-(ethylisoluminol) (ABEI)-engineered oxygen-doped carbon nitride (O-g-C3N4), for chemiluminescent detection of sweat glucose. The LAON exhibits dual catalytic activity of glucose oxidase and peroxidase and can not only oxidize glucose to generate H2O2 but catalyze H2O2-mediated luminol chemiluminescence, resulting in sensitive detection of glucose. We identify that the LAON can precisely detect glucose with a detection limit of 0.1 μM, enabling us to measure glucose levels in different biological samples. Particularly, the LAON is capable of sensitively and accurately monitoring dynamic changes in sweat glucose during exercise. Therefore, the LAON provides an alternative approach to supersede invasive blood tests and may improve the management of diabetes mellitus.

Efficient bionic nanozyme based on AuPt NPs@ZIF-90 used for cyclic catalysis multimodal tumor therapy

Wan Huang,Song Zhang,Li Luo,Yalong Pan,Lijun Han,Yao Yu

Multimodal therapy based on nanozyme is expected to become a novel option for tumor treatment. However, the catalytic efficiency of nanozyme and the hypoxia microenvironment of tumors limit the therapeutic effect of nanozyme. Here in this article, we screened a small molecule midazole-2-carboxaldehyde (ICA) to preparation ZIF-90 and embedded gold and platinum nanoparticles to obtain ZAAP. ZAAP possessed a multi-enzymatic cascade of catalytic process including greatly enhanced peroxidase activity via "bionic" catalytic microenvironment (enhanced 23-fold), catalase and glucose oxidase activities, resulting in glucose decomposition to continuously supply H2O2, peroxidases for the catabolism of H2O2 to generate ROS and peroxidase-induced oxygen generation for continuous oxidation of glucose. All the above process built a catalysis cycle which greatly promotes the generation of ROS and oxygen as well as the consumption of glucose, leading to the chemical dynamic therapy function and alleviating the tumor hypoxia. In addition to the photothermal effect of ZAAP, a synergistic treatment of chemical dynamic/ photothermal/ starvation therapy can be achieved and the tumor inhibition rate reached 96.4% within 2 weeks, indicating that ZAAP show great potential in nanozyme based synergistic multimodal tumor treatment.

Coordination Self-Assembled AuTPyP-Cu Metal–Organic Framework Nanosheets with pH/Ultrasound Dual-Responsiveness for Synergistically Triggering Cuproptosis-Augmented Chemotherapy

Jianshuai Bao,Jiefei Wang,Sudi Chen,Shiqi Liu,Zhen Wang,Weiwei Zhang,Chenhui Zhao,Yuling Sha,Xiaoyan Yang,Yusen Li,Yong Zhong,Feng Bai

Reactive oxygen species (ROS) mediated tumor cell death is a powerful anticancer strategy. Cuproptosis is a copper-dependent and ROS-mediated prospective tumor therapy strategy. However, the complex tumor microenvironment (TME), low tumor specificity, poor therapy efficiency, and lack of imaging capability impair the therapy output of current cuproptosis drugs. Herein, we designed a dual-responsive two-dimensional metal–organic framework (2D MOF) nanotheranostic via a coordination self-assembly strategy using Au(III) tetra-(4-pyridyl) porphine (AuTPyP) as the ligand and copper ions (Cu2+) as nodes. The dual-stimulus combined with the protonation of the pyridyl group in AuTPyP and deep-penetration ultrasound (US) together triggered the controlled release in an acidic TME. The ultrathin structure (3.0 nm) of nanotheranostics promoted the release process. The released Cu2+ was reduced to Cu+ by depleting the overexpressed glutathione (GSH) in the tumor, which not only activated the Ferredoxin 1 (FDX1)-mediated cuproptosis but also catalyzed the overexpressed hydrogen peroxide (H2O2) in the tumor into reactive oxygen species via Fenton-like reaction. Simultaneously, the released AuTPyP could specifically bind with thioredoxin reductase and activate the redox imbalance of tumor cells. These together selectively induced significant mitochondrial vacuoles and prominent tumor cell death but did not damage the normal cells. The fluorescence and magnetic resonance imaging (MRI) results verified this nanotheranostic could target the HeLa tumor to greatly promote the self-enhanced effect of chemotherapy/cuproptosis and tumor inhibition efficiency. The work helped to elucidate the controlled assembly of multiresponsive nanotheranostics and the high-specificity ROS regulation for application in anticancer therapy.

Bacteria-Mediated Tumor-Targeting Delivery of Multienzyme-Mimicking Covalent Organic Frameworks Promoting Pyroptosis for Combinatorial Sono-Catalytic Immunotherapy

Yunyun Liu,Lihua Xiang,Yitong Li,Shen Zhang,Ying Zhang,Hui Shi,Hui Liu,Dou Du,Bangguo Zhou,Beibei Ye,Shaoyue Li,Haohao Yin,Huixiong Xu,Yifeng Zhang

Pyroptosis, an inflammatory cell death, has attracted great attention for potentiating a strong immune response against tumor cells. However, developing powerful pyroptosis inducers and then activating specific pyroptosis still remains challenging. Herein, a PEG-CuP-COF@∆St nanosystem is rationally designed, consisting of PEG-CuP-COF nanozyme pyroptosis inducers and tumor-targeting bacteria of the Salmonella Typhimurium strain VNP20009 (ΔSt), with an affinity for the tumor hypoxic microenvironment. The PEG-CuP-COF nanozymes possessed excellent sonodynamic performance and multienzyme-mimicking activities to generate reactive oxygen species (ROS) and then induce potent pyroptosis. The superoxide dismutase- and peroxidase-mimicking activities of PEG-CuP-COF catalytically produced hydrogen peroxide (H 2 O 2 ) and hydroxyl radicals (•OH) which have important value in triggering acute inflammatory responses and pyroptosis. Moreover, PEG-CuP-COF showed outstanding glutathione peroxidase-mimicking activities, impairing the antioxidant defense in tumor cells and enhancing sonodynamic efficiency by making them more vulnerable to ROS-induced damage. During in vivo studies, PEG-CuP-COF@∆St nanosystem with its self-driven property exhibited impressive tumor-targeting capability and activated Caspase-3/gasdermin E-dependent pyroptosis to inhibit tumor growth. More importantly, it induced a powerful immune memory effect to prevent bone metastasis. In summary, this study introduces an innovative approach for combinatorial sono-catalytic immunotherapy using bacteria-mediated tumor-targeting delivery of nanozymes as specific pyroptosis inducers.

Supplemented collagen peptide composition improves skin aging, elasticity, and tissue regeneration in zebrafish

Juan Huang,Haiyu Ouyang,Wenjing Zhou,Difan Zhu,Haimei Zhi,Wei Li

A supplemented collagen peptide composition (SCPC) containing microbe-fermented collagen peptide, collagen tripeptide, elastin peptide, and sodium hyaluronate was investigated to determine its effects on skin aging, skin elasticity, and tissue regeneration in zebrafish. β-Galactosidase activity was inhibited, while telomerase activity was elevated, by exposure to 0.977 μL/mL of SCPC in an H 2 O 2 -induced aging zebrafish model, indicating that SCPC can delay cell senescence. Expression of the collagen I gene col1a1a was up-regulated by exposure to 62.5 μL/mL of SCPC, indicating that SCPC can improve skin elasticity. Regrowth of the injured caudal fin was enhanced by exposure to only 0.122 μL/mL of SCPC. These improvements to skin aging, elasticity, and tissue regeneration in zebrafish suggest that this SCPC has potential as a functional food with beneficial effects on skin health and aging resistance in humans.

Hydrogen sulfide-generating semiconducting polymer nanoparticles for amplified radiodynamic-ferroptosis therapy of orthotopic glioblastoma

Anni Zhu,Shuai Shao,Jinyuan Hu,Wenzhi Tu,Zheming Song,Yue Liu,Jiansheng Liu,Qin Zhang,Jingchao Li

A variety of therapeutic strategies are available to treat glioblastoma (GBM), but the tumor remains one of the deadliest due to its aggressive invasiveness, restrictive blood-brain barrier (BBB), and exceptional resistance to drugs. In this study, we present a hydrogen sulfide (H2S)-generating semiconducting polymer nanoparticle (PFeD@Ang) for amplified radiodynamic-ferroptosis therapy of orthotopic glioblastoma. Our results showed that in acidic tumor microenvironment (TME), H₂S donors produce large amounts of H2S, which inhibits mitochondrial respiration and alleviates cellular hypoxia, thus enhancing the radiodynamic effect during X-ray irradiation; meanwhile, Fe3⁺ is reduced to Fe²⁺ by tannic acid in acidic TME, which promotes iron-dependent cell death process in tumors. H2S facilitates the ferroptosis process by increasing local H2O2 concentration via inhibiting catalase activity. Such an amplified radiodynamic-ferroptosis therapeutic strategy could remarkably inhibit glioma progression in an orthotopic GBM mouse model. Our study demonstrates the potential of PFeD@Ang for GBM treatment via targeted delivery and combinational therapeutic actions of RDT and ferroptosis therapy.

Rapid and robust modification of PVDF ultrafiltration membranes with enhanced permselectivity, antifouling and antibacterial performance

Kai Cheng,Na Zhang,Na Yang,Shuai Hou,Jiahui Ma,Luhong Zhang,Yongli Sun,Bin Jiang

Zwitterionic materials are well known to possess extraordinary water affinity. However, micromolecular zwitterions have rarely been exploited for membrane hydrophilic modification. Herein, novel antifouling and antibacterial polyvinylidene fluoride (PVDF) ultrafiltration membranes with enhanced permselectivity were facilely fabricated through one-step co-deposition of dopamine and a newly synthesized micromolecular zwitterion (DMAPAPS). With the assistance of CuSO 4 /H 2 O 2 -triggered oxidation, a uniform and hydrophilic coating (water contact angle = 33°) was constructed on membrane surface in only 40 min. The impacts of the DMAPAPS concentration and coating time on membrane performance were investigated. Results showed that our strategy endowed the optimized membrane with high pure water flux of 364 L/m 2 ∙ h and BSA rejection of 98.6%. The flux recovery ratios of membrane were as high as 96.3% for bovine serum albumin, 98.1% for humic acid and 95.1% for sodium alginate after 3-cycle filtration, respectively. Moreover, the membrane displayed superior antibacterial activity towards E. coli and S. aureus as well as excellent chemical and mechanical stability. This work provides a versatile platform for the robust, time-saving and cost-effective fabrication of antifouling and antibacterial membranes.

Determination of the binding constant of specific interactions and binding target concentration simultaneously using a general chemiluminescence method

Lingfeng Gao,Li Ju,Hua Cui

The measurement of the binding constant of specific interactions and concentration of a target is of considerable importance in clinical diagnosis, therapy, bioassays and drug design. The development of methods that combine high sensitivity with generalization and simplicity for the measurement of both binding constant and target concentration is highly desirable. Previously, we developed a label-free chemiluminescence (CL) strategy for the measurement of the target concentration and binding constants between DNA aptamers and target simultaneously based on the fact that the target could enhance the CL produced by the reaction of the N-(4-aminobutyl)-N-ethylisoluminol (ABEI) functionalized gold colloid with H2O2. In this study, the enhancement and inhibition effect of various targets on the CL reaction is studied. The generalization of the proposed CL strategy for various targets is also explored. The results demonstrate that the proposed CL strategy is suitable for targets that can cause a change in CL intensity, which includes enhancement and inhibition. It could be applied for the measurement of the dissociation constants of aptamer-binding targets, antibody–antigen complexes, protein-binding small molecules and double-strand DNA hybrids from the millimole to picomole level. It could also be used for the sensitive determination of target concentration, including 2,4,6-trinitrotoluene (TNT), dopamine, tetracycline, human IgG (hIgG), tuberculosis (TB) DNA and mannose, with the detection limit of 0.93 nM–4.1 fM. This strategy is of great potential in fundamental research as well as in applications in life sciences.

An integrated microwave-ultraviolet catalysis process of four peroxides for wastewater treatment: Free radical generation rate and mechanism

Shiyu Zuo,Dongya Li,Haiming Xu,Dongsheng Xia

The Advanced Oxidation Processes (AOPs) still require further research in response to ever-increasing standard of reclaimed water reuse and operating environment requirements. In this study, an efficient microwave-ultraviolet catalyzed oxidation system (MW-UV) was successfully developed, and its degradation effect was studied by using activated H 2 O 2 , sodium percarbonate (SPC), persulfate (PS), and peroxymonosulfate (PMS) to degrade toxic organic contaminants in wastewater and actual industrial wastewater. It was shown that MW-UV had an exceptionally high capability to react with these four common peroxides to produce reactive radicals ( OH and SO 4 − ). This can destroy multiple toxic organic contaminants through nonselective oxidation with a wide range of pH applications, has strong adaptability to anions in bodies of water, and can effectively treat wastewater. The sequence of predominant free radical generation rate by MW-UV was SPC > H 2 O 2  > PS > PMS, with higher generation rate constant than other Fenton processes. The free radical evolution mechanisms for H 2 O 2 , SPC, PS, and PMS activation were also investigated. The thermal and non-thermal effects of microwave irradiation have a significant enhancement for UV reactions, and thus the synergy between the MW and UV can effectively activate peroxide. This treatment method could provide scientific basis and reference for degrading toxic organic contaminants in small-scale and limited space wastewater treatment.

Dual signal amplification photoelectrochemical biosensor for highly sensitive human epidermal growth factor receptor-2 detection

Xiaoxi Guo,Shuping Liu,Minghui Yang,Huitong Du,Fengli Qu

Photoelectrochemical (PEC) biosensor for highly sensitive detection of breast cancer biomarker human epidermal growth factor receptor-2 (HER2) is reported utilizing a dual signal amplification strategy. The biosensor was prepared based on tungsten sulfide nanowire array on Ti mesh (WS 2 NW/TM). Such WS 2 NW/TM electrode can generate photoelectric signal under visible light excitation . The HER2 aptamer was wrapped onto the nanowire array surface for specific binding with HER2 molecules, and gold nanoparticles (Au NPs) that modified with glucose oxidase (GOx) and HER2 binding peptide was utilized for signal amplification. The H 2 O 2 that generated by GOx catalyzed glucose reaction and localized surface plasmon resonance of Au NPs can both enhance the PEC current intensity of the biosensor, leading to dual signal amplification. The PEC current intensity is enhanced linearly with HER2 concentration in the 0.5–10 ng/mL range with limit of detection of 0.36 ng/mL. Such biosensor was applied for the detection of HER2 in breast cancer serum samples with detection results in good agreement with commercial ELISA results.
