Folic acid-functionalized AIE Pdots based on amphiphilic PCL-b-PEG for targeted cell imaging

Yan Zhang,Yujue Chen,Xing Li,Jibo Zhang,Jinlong Chen,Bin Xu,Xueqi Fu,Wenjing Tian

Fluorescent block amphiphilic copolymers are one of the most important bioimaging materials which are highly desirable for early stage cancer diagnosis and treatment. However, sometimes the application of the fluorescent block copolymers is severely limited because of the aggregation-caused quenching (ACQ) effect. In this study, new aggregation induced emission polymer dots (AIE Pdots) were prepared through a self-assembly process by using an AIE-conjugated block copolymer containing an AIE fluorophore, 9,10-bis(4-hydroxystyryl)anthracene, hydrophobic poly(ε-caprolactone) segments, hydrophilic poly(ethylene glycol) segments and folate groups. The AIE Pdots are monodispersed in H2O with an average diameter of 15 nm, and possess strong emission and high solid state fluorescence quantum efficiency (Φ = 27.0%). Furthermore, the AIE Pdots show good stability and little toxicity to living cells and thus can be utilized for targeted HeLa cell imaging. Biological imaging investigation indicated that the folic acid functionalized AIE Pdots can be applied for targeted HeLa intracellular imaging.

Dual-Functional AIE Fluorescent Probe for Visualization of Lipid Droplets and Photodynamic Therapy of Cancer

Shizeng Pei,Haoyang Li,Linfeng Chen,Gang Nie,Huiling Wang,Chunrong Liu,Caihong Zhang

Abnormal lipid droplets (LDs) are known to be intimately bound with the occurrence and development of cancer, allowing LDs to be critical biomarkers for cancers. Aggregation-induced emission luminogens (AIEgens), with efficient reactive oxygen species (ROS) production performance, are prime photosensitizers (PSs) for photodynamic therapy (PDT) with imaging. Therefore, the development of dual-functional fluorescent probes with aggregation-induced emission (AIE) characteristics that enable both simultaneous LD monitoring and imaging-guided PDT is essential for concurrent cancer diagnosis and treatment. Herein, we reported the development of a novel LD-targeting fluorescent probe (TDTI) with AIE performance, which was expected to realize the integration of cancer diagnosis through LD visualization and cancer treatment via PDT. We demonstrated that TDTI, with typical AIE characteristics and excellent photostability, could target LDs with high specificity, which enables the dynamic tracking of LDs in living cells, specific imaging of LDs in zebrafish, and the differentiation of cancer cells from normal cells for cancer diagnosis. Meanwhile, TDTI exhibited fast ROS generation ability (achieving equilibrium within 60 s) under white light irradiation (10 mW/cm2). The cell apoptosis assay revealed that TDTI effectively induced growth inhibition and apoptosis of HeLa cells. Further, the results of PDT in vivo indicated that TDTI had a good antitumor effect on the tumor-bearing mice model. Collectively, these results highlight the potential utility of the dual-functional fluorescent probe TDTI in the integrated diagnosis and treatment of cancer.

A cooperative effect of copper-induction and AIE leading to bright luminescence of gold nanoclusters

Yongjie Zhang,Luyao Feng,Jingyan Luan,Guomei Zhang,Ning Sheng,Jinglin Shen

Emission enhancement of gold nanoclusters protected with 6-propyl-2-thiouracil (PRT-AuNCs) is achieved through incorporating a suitable amount of Cu2+. Sophisticated mechanisms are involved in the modulation of emission properties of the PRT-AuNCs/Cu2+ system, instead of merely a metal ion induced aggregation. X-ray photoelectron spectroscopy, IR spectroscopy and mass spectroscopy reveal that the Cu2+ is reduced by 6-propyl-2-thiouracil to form a Cu(I)–thiolate complex, which then co-assembles with the AuNCs to form a PRT-Au/Cu composite at an appropriate Cu2+ concentration. Systematic microscopic characterization studies including scanning electron microscopy, X-ray diffraction etc. demonstrate the crucial effect of compact aggregation driven by metallophilic interactions on the emission enhancement of PRT-Au/Cu. Detailed analysis of the photophysical parameters validates that the presence of Cu(I) changes the radiative transition mechanisms of PRT-Au/Cu with respect to primitive PRT-AuNCs. As a result, a cooperative effect of copper-induction and aggregation induced emission (AIE) leads to a strongly emissive composite with bathochromic-shifted emission energy. Moreover, the high molar ratio between Cu and Au in PRT-Au/Cu can effectively reduce the cost of the material in production, and we have developed a WLED to exemplify the potential application of this composite material.

Biotin receptor and mitochondria dual targeted AIE photosensitizer for fluorescence imaging guided photodynamic anticancer therapy

Liying Pan,Shihua Wang,Mintao Xie,Manshan Liang,Lichao Yu,Bing Du,Xiaoxia Ye,Yanshu Luo,Yun-Yun Quan,Zu-Sheng Huang

Rational design of organelle targeted therapeutic platform in where the drug can be targeted to destroy tumor cells as much as possible, is one of the effective ways to improve the therapeutic outcomes and minimize the cytotoxicity to normal cells. Here, photosensitizer TSPy-B with aggregation-induced emission (AIE) feature, biotin receptor and mitochondria dual-targeted capability, and high reactive oxygen species (ROS) generation efficiency was designed for fluorescence imaging guided photodynamic therapy (PDT) by linking biotin group to D-π-A type AIE dyes via ester bonds. Notably, cell imaging experiments showed that TSPy-B could selectively target tumor cells with biotin receptor overexpression and further exhibit mitochondria targeting ability. The in vitro anticancer mechanism confirmed that after cell internalization, TSPy-B localized in the mitochondrial site and produced ROS resulting in mitochondrial dysfunction and cell death. ROS generation experiments showed that TSPy-B could simultaneously produce 1 O 2 and highly toxic free radicals which not depended on the oxygen level of solid tumor. The cell killing effect of TSPy-B was also confirmed in hypoxic culture model. In vivo photodynamic therapy experiments demonstrated that TSPy-B could effectively inhibit tumor growth and had good biological safety. This work provides a choice for development of highly targeted photosensitizers for clinical fluorescence imaging guided PDT.

Color-Shifting Fluorescent System Based on an AIE Polymer with an Upper Critical Solution Temperature

Shunfeng Yu,Kang Liu,Yueyi Tian,Xiaoxiao Chen,Jie Zhang,Mei Gao,Jinhui Wang,Mingming Zhang,Chuanzhuang Zhao

The design of thermoresponsive luminescent polymers with both a tunable upper critical solution temperature (UCST) and a tunable color is of great challenge. Herein, a series of thermoresponsive copolymers with UCST and aggregation-induced emission (AIE) behaviors are prepared, and a color-shifting fluorescent system based on the energy conversion between the AIE polymers and fluorescent dyes is constructed. The copolymers are prepared from acrylamide and 6-(4-vinylphenyl)-2,4-diamino-1,3,5-triazine (VPhDT). Benefitting from VPhDT, an AIE-active monomer that can provide multiple hydrogen bonds and enhance association of the polymer chains, the obtained copolymers exhibit both UCST and AIE behaviors in aqueous solutions. Utilizing the AIE copolymers as the donor and fluorescent dyes as the acceptors, color-shifting fluorescent systems that can emit blue, green, and red lights are established. The color-shifting fluorescent systems also exhibit tunable luminescent intensities and colors with the stimuli of temperature and pH. Such a color-shifting fluorescent system may have promising applications in the fields of temperature sensors and information displays.

Cellulose nanofiber encapsulated polyethylene glycol phase change composites containing AIE-gen for monitoring leak process

Yabi Yang,Shuang Liu,Guoliang Zhang,Ziqing Hu,Liang Jin,Bingqing Quan,Xiaoyun Hao,Hao Wu,Xiaofan Ji,Xiang Lu

The design of cellulose nanofiber (CNF)-encapsulated polyethylene glycol (PEG)-based phase change composites (PCCs) has shown great promise toward overcoming the phase transition process’s leakage. Whereas, the leakage is inescapable for intricate industrial occasions such as extrusion and collision. Thereby it’s requisite to monitor the products’ leakage. Herein, a fluorophore with aggregation-induced emission (AIE) effect was employed to prepare fluorescent PCCs. In detail, Pmix2 was picked to dip into the skeleton CNF6 to construct CNF6/Pmix2 with dim green. What’s more, the free fall of 500 g weight, 983 times of CNF6/Pmix2′s weight, was applied to the materials to simulate the possible force that occurred in real industrial processes. Obviously, CNF6/Pmix2′s leakage is seen on dark occasions, whereas CNF6/PEG’s leakage trace is hardly observed, evidencing CNF6/Pmix2 possesses the capacity of monitoring leakage. Additionally, CNF6/Pmix2 displays excellent thermal energy storage and reusability, illustrating promising energy conversion and storage applications.

Mechanochromic, thermoresponsive and triboluminescence behaviors of one divinyl thioxanthene based AIE luminogen with variable conformations

Nengni Xu,Wenhua Xu,Meng Sun,Yi Yuan,Xinjun Luan,Ying Wang,Hui Wang

Strategic structure designs to develop organic multi-stimuli-responsive fluorescent materials are highly desired, but hard to achieve. We report here a novel multi-stimuli responsive luminescent material TXENE (1,2-di(9H-thioxanthen-9-ylidene)ethane) composed of double thioxanthene chromophores and a diene liker that integrate polymorphism, aggregation-induced emission (AIE), mechanoluminescence (MCL), thermoresponsive luminescence (TCL), and triboluminescent (TL) properties into one single molecule. Three crystalline polymorphs including TXENE-G and TXENE-Y with a terminal up–down conformer and TXENE-R with an up–up form are obtained. The X-ray structure analysis revealed that not only the intermolecular interactions but also the terminal chromophore-thioxanthene conformation (up–down vs. up–up) dominate polymorphism, thus leading to distinct luminescent properties. It is shown that MCL, TCL and TL properties can be systematically tuned via its aggregation state regulation; typically, the metastable TXENE-G and TXENE-R are MCL and TCL active, while TXENE-Y is TL active. Lastly, we illustrate the possible application of color-changeable indicators for information anti-counterfeiting and thermal sensing by using TXENE. This study demonstrates that multiple unique functions can be integrated into one single compound by fine controlling its aggregation states, which render a new strategy for the investigation of multifunctional organic materials.

An AIE fungal vacuole membrane probe toward species differentiation, vacuole formation visualization, and targeted photodynamic therapy

Bingnan Wang,Siyuan Wang,Chunyang Li,Jianqing Li,Meixi Yi,Jing-Wen Lyu,Bing Gu,Ryan T.K. Kwok,Jacky W.Y. Lam,Anjun Qin,Ben Zhong Tang

Vacuoles are unique organelles of fungi. The development of probes targeting the vacuoles membrane will enable visualization of physiological processes and precise diagnosis and therapy. Herein, a zwitterionic molecule, MXF-R, comprising of an aggregation-induced emission (AIE) photosensitizing unit and an antibiotic moxifloxacin, was found capable of specifically imaging vacuole membrane and using for targeted antifungal therapy. MXF-R demonstrated a higher signal-to-noise ratio, stronger targeting capability, and better biocompatibility than the commercial probe FM4-64. By using MXF-R, real-time visualization of vacuole formation during Candida albicans ( C. albicans ) proliferation was achieved. More importantly, owing to its varying staining ability towards different fungus, MXF-R could be used to quickly identify C. albicans in mixed strains by fluorescence imaging. Moreover, MXF-R exhibits a remarkable ability to generate reactive oxygen species under white light, effectively eradicating C. albicans by disrupting membrane structure. This antifungal therapy of membrane damage is more effective than clinical drug fluconazole. Therefore, this work not only presents the initial discovery of a probe targeting vacuolar membrane, but also provides a way to develop novel materials to realize integrated diagnosis and therapy.

Highly Efficient AIE-Active palmatine photosensitizer for photodynamic inactivation of Listeria monocytogenes in apple juice preservation

Shiyu Peng,Qianying Li,Hui Wei,Ping Li,Shuyuan Liu,Lijuan Nie,Yuankui Leng,Xiaolin Huang

Photodynamic sterilization is a promising alternative to conventional antibacterial approaches with merits of high efficiency and safety. The exploration of aggregation-induced emission (AIE)-type photosensitizers from natural sources is highly valued by researchers and food industry. Herein, the use of palmatine (PA), an aggregation-induced emission-active natural product from traditional Chinese medicine, as photosensitizers for photodynamic inactivation of a tenacious foodborne pathogen of Listeria monocytogenes is investigated. Antibacterial and antibiofilm activity against L. monocytogenes of PA-mediated photodynamic process were first assessed. The results showed that PA-mediated photodynamic process could inhibit the growth of L. monocytogenes , and the minimum bactericidal concentration was determined as 80 μM. At this PA dosing level, well-established biofilms of L. monocytogenes can be effectively destroyed under light irritation. Afterward, cell- and gene-level investigations were conducted to explore the antibacterial mechanism. It is concluded that the exceptional photodynamic antibacterial activity of PA against L. monocytogenes is attributed to the disruption of the cell wall and membrane, increased cell permeability, leakage of functional proteins, and damage to DNA structure. Subsequently, PA-mediated photodynamic process was applied to reduce bacterial activity in apple juice while preserving its quality. Overall, this work highlights the significant potential of PA-mediated photodynamic strategy for controlling foodborne pathogens in food systems.

An AIE-active “turn-off” fluorescent sensor for highly selective and sensitive detection of Cu2+ions

Xiaoling Niu,Hong Zhang,Xiaojing Wu,Shengbo Zhu,Yongliang Liu,Min Tian

Developing highly selective and sensitive sensor for Cu 2+ ions is very important in the fields of chemical, biological and environmental. Herein, a novel Cu 2+ ions sensor based on tetraphenylethylene (TPE) derivative with aggregation-induced emission (AIE) property was designed and synthesized. It was prepared by introducing four Cu 2+ ion chelating centers onto TPE core, which exhibited a sensitive fluorescent “turn-off” behaviour and highly selective responses to Cu 2+ ions in the present of various interferential cations. It also showed a visible colour change from bright yellow to colourless at the addition moment of Cu 2+ ions. The responses mechanism was revealed by 1 H NMR spectrum, dynamic light scattering (DLS) analysis and density functional theory (DFT) calculation. The results showed that the coordination of the sensor's N, O atoms with Cu 2+ ions induced the TPE-Schiff base's structure planarization, and formed a very tight Cu 2+ complex in nano-size aggregation state, which led to the fluorescent emission “ turn-off ”. This study provides a valuable approach to design and prepare AIE-active fluorescent sensor for the detection of Cu 2+ ions.

A novel AIE fluorescent probe for the monitoring of aluminum ions in living cells and zebrafish

Yabing Gan,Guoxing Yin,Jianhua Wang,Peng Yin

A novel fluorescent probe BTD with aggregation induced emission (AIE) characteristics for the monitoring of Al3+ was developed. This fluorescent probe could be used to detect Al3+ in aqueous solution under mild conditions, along with high sensitivity and high selectivity. The detection limit of the probe BTD for Al3+ is as low as 3.25 nM, which is below the WHO recommendation concentration (7.41 μM) for drinking water. Furthermore, this probe was successfully applied to the sensing of Al3+ in living cells and zebrafish.

Detecting the insoluble protein aggregates in live cells using an AIE derivative of fluorescent protein chromophore

Lihua Liu,Wenhan Jin,Yanan Huang,Jianan Dai,Xuwei Zheng,Yu Liu,Minzi Ju,Baoxing Shen

Many incurable or unmanageable human protein conformational diseases are associated with the misfolding or aggregation of the aberrantly processed or mutant proteins . In this work, we report an aggregation-induced emission (AIE) derivative of fluorescent protein chromophore used to detect the insoluble protein aggregates in live cells. Based on the 4-hydroxybenzylidene-imidazolinone (HBI), we designed and synthesized a series of AIEgens that span a wide range of viscosity coefficients (χ), thus mimicking the viscous microenvironment of a wide variety of amorphous protein aggregates. The mechanism of these AIEgens were systematically investigated using a combination of photophysical studies, computational analyses and structure-function studies. With the aid of the AggTag method, the optimized probe was used to realize the imaging of the aggregated proteome under the control of the proteostasis network. Besides, we also described the formation and decomposition of protein aggregates under the control of small molecule proteostasis regulators. Briefly, we developed a series of AIEgens that explore varying viscosity sensitivities to visualize protein aggregation in live cells as well as study other biological processes that associated with local viscosity changes.
