Acute kidney injury (AKI) is a highly morbid and lethal syndrome that is often clinically “silent” in early stages. Multimodal imaging, which integrates complementary structural and functional insights from various techniques, holds great potential to enhance diagnostic sensitivity and accuracy. However, developing a single-agent probe that effectively combines multiple imaging modalities for AKI remains a significant challenge. Herein, we show neodymium-enriched carbon dots (Nd-CDs) as a novel bimodal imaging probe for the early diagnosis of AKI by using computed tomography (CT) and the second near-infrared window (NIR-II) fluorescence. Nd-CDs with high metal doping level (17.9%) can be achieved on a gram-scale (2.1 g) through a “bottom-up” strategy. The ultra-small hydrodynamic size (<2 nm) endows Nd-CDs with rapid renal clearance. Benefiting from narrow-band NIR-II fluorescence emission at 1060 nm and strong X-ray attenuation, Nd-CDs provide high-spatial-resolution CT and highly sensitive fluorescence imaging in vivo . In a cisplatin-induced AKI rat model, Nd-CDs-based bimodal imaging reveals a pronounced delay in renal metabolism caused by tubular injury, which precedes abnormalities in blood biochemical parameters, enabling early diagnosis and progression monitoring of AKI. The proposed strategy provides new insights into the development of single-agent probes integrating multiple imaging modalities and opens up a new way for early diagnosis of AKI.

The multifunctional theranostic nanoplatforms, which can realize changing the contrasts of medical images and enhance cancer therapies simultaneously, have attracted tremendous attention from chemists and medicine in past decades. Herein, a nanoscale metal–organic framework-based material was first synthesized and then decorated with platinum (NMOF545@Pt) successfully for multimodal imaging-guided synergistic cancer therapy. The obtained NMOF545@Pt is advantageous in shortening the longitudinal relaxation time (T1), enhancing photoacoustic effects, and elevating X-ray absorption efficiently. Thus, the enchantments of tripe imaging modalities, computed tomography (CT)/magnetic resonance imaging (MRI)/photoacoustic imaging (PAI), were realized with NMOF545@Pt administration simultaneously and can be cleared from the mice. Meanwhile, in vitro and in vivo experiments demonstrate that the synthesized NMOF545@Pt can dramatically increase photothermal therapy (PTT) and radiotherapy (RT) efficacy. Convincing evidence proves that tumor growth can be wholly inhibited without noticeable side effects or organ damage. The results demonstrated the promise of multifunctional nanocomposites NMOF545@Pt to improve biomedical imaging and synergistic tumor treatments.

In this work, a novel bi-modal imaging probe with enhanced CT contrast efficiency and FL brightness was constructed, in which the combination of a binary CT contrast agent BaHoF 5 and Cu-doped QDs served as a vehicle; hyaluronic acid (HA) was employed as a tumor-targeting ligand. With its CT contrast efficiency about 2.1- and 3.9-fold higher than PEG-BaHoF 5 and Iohexol, the CT contrast efficiency and the fluorescent brightness of the bi-modal probe were both enhanced. Likewise, its fluorescent brightness is almost 6-fold brighter after Cu-doped QDs loading. The most important contribution of this work lies on the proposed strategy. The inherent contradiction of the imaging sensitivity of CT and FL imaging is well balanced and a great CT/FL bi-modal imaging performance is simultaneously obtained even at low concentration (400 μg/mL) of the probe, which was superior to the previous CT/FL bi-modal probes. Moreover, since BaHoF 5 as a binary CT contrast agent was introduced instead of conventional Au and Bi 2 S 3 , the CT/FL bi-modal probe would be more suitable for different patients under different operation voltages. In addition, the in vitro tumor cell imaging also demonstrated a good photo-stability, FL brightness, and tumor-targeting capability of the probe, indicating its great potential in practical bi-modal imaging for further tumor diagnosis and therapy.
A novel bi-modal imaging probe with enhanced CT contrast efficiency and FL brightness was fabricated, in which its CT contrast efficiency was about 2.1- and 3.9-fold higher than PEG-BaHoF 5 and Iohexol, respectively, and its fluorescent brightness almost 6-fold brighter after Cu-doped QDs loading.

Theranostics of platinum (Pt)-based chemotherapy are able to self-track the biodistribution and pharmacokinetics while performing therapeutic effects. Pt-based CT imaging is expected to visualize and monitor the tumor throughout the entire tumor inhibition stage. However, a sufficient Pt concentration is necessary for CT imaging, which may bring about severe nephrotoxicity. A Bio-Pt-I compound is designed and synthesized by conjugation of iodine and biotin to the structure of Pt and further self-assembles into nanoparticles. The introduction of iodine not only enhances the CT imaging signal with a much lower dose of Pt but also overcomes the resistance of tumor cells to Pt-containing nanomedicine by inhibiting the expression of Bcl-2. Furthermore, biotin-mediated tumor targeting increases drug accumulation in tumors. This work combines CT imaging based self-track with efficient cisplatin-resistance reversion ability, which may promote the clinical transformation of Pt-containing nanomedicine.

Multifunctional nanoprobes that can be applied for real-time monitoring or precision treatment of tumors have received wide interest among researchers. However, most of these nanoprobes are obtained through chemical synthesis, and thereby may contain toxic residues or harmful reagents. In this article, a nano-bismuth-sphere-cluster (Bi) is synthesized via a one-step method (after an irradiation with ultra-violet) and is then applied in dual-mode computed tomography/photoacoustic imaging. Bismuth potassium citrate granules, which is a common gastric drug that is highly safe and has a low price (<1 China Yuan/g), is used as the only raw material. The results show that the Bi cluster has good stability with sizes of about 25–55 nm, and a photothermal conversion efficiency as high as 39.67%. After being adsorbed onto doxorubicin, the Bi cluster can be used directly in animal experiments. Due to the effect of enhanced permeability and retention, the probe can easily enter tumor cells. Drug release can be controlled by a near-infrared laser and the acidic environment of tumor cells, which indicates that the combined chemo-photothermal therapy is achieved. This work presents a new dual-mode bio-imaging and combined chemo-photothermal therapeutic nanoprobe that can be applied in theragnostics for tumors.

Objective To fabricate a novel nano-hydroxyapatite mineralized silk fibroin (MSF) scaffold in order to diminish the resorption of alveolar ridge and accelerate new bone formation within tooth sockets. Also, to investigate the biocompatibility and osteogenic ability of the MSF in vitro , and the effect of site preservation of the MSF graft in post-extractive sockets in vivo . Methods SEM, EDX, FTIR and XRD were used to analyze the mineral crystals deposited on the silk fibroin (SF) surface. Pre-osteoblasts (MC3T3-E1) were seeded on SF and MSF scaffolds. Cell viability, distribution and differentiation were examined using a live-dead assay, histological analysis and Alizarin Red S staining. Furthermore, prepared grafts (SF or MSF scaffold) were implanted into the maxillary right first molar sockets of Sprague Dawley rats for 6 weeks and newly formed bone tissue was analyzed by micro-CT and histological examination. Results The SEM, EDX, FTIR and XRD analysis demonstrated that granulate nano-hydroxyapatite (nHA) crystals were uniformly distributed on the SF scaffold. In addition, the MSF hydrophilicity measured by water contact angle and swelling ratio was superior to plain SF scaffold. The effect of nHA inorganic crystals on osteogenic differentiation of MC3T3-E1 cells indicated the MSF scaffolds improved osteogenesis. Furthermore, MSF grafts induced more bone formation and reduced the height of alveolar bone resorption after tooth extraction. Significance The MSF scaffold partially simulated the structure and composition of natural bone matrix. It induced osteogenic differentiation of MC3T3-E1 cells in vitro , and also promoted new bone regeneration in tooth extraction sockets in vivo , indicating it is a biomaterial with great potential for tooth extraction site preservation.

Purpose MicroRNA-34c-5p (miR-34c-5p) plays a pivotal role in bone remodeling, yet its therapeutic potential is hindered by challenges such as instability, limited cellular internalization, and immune responses. This study was aimed at developing innovative scaffolds capable of efficiently delivering microRNAs (miRNAs), specifically miR-34c-5p.Methods Chitosan (CS)/sodium tripolyphosphate (STPP)/sodium alginate (SA) scaffolds, referred to as CTS scaffolds, were synthesized at a specific ratio and characterized using dynamic light scattering and scanning electron microscopy (SEM). Cytotoxicity assessments were conducted through cell activity staining. The loading capacity and releasing performance of miRNAs were quantified using spectrophotometry. Subsequently, the in vivo efficacy of miR-34c-5p Agomir/Antagomir in regulating bone repair was evaluated in the rabbit cranial bone defect model, with micro-CT scanning and histological analysis conducted at 4, 8, and 12 weeks. Results CTS scaffolds with a composition ratio of 1:0.2:0.1 were successfully synthesized, exhibiting a mean particle size of 360.1 nm. SEM revealed scaffolds had the porous spongy structure. Cell activity staining confirmed the excellent biocompatibility of the CTS scaffolds. Spectrophotometry demonstrated miR-34c-5p Antagomir were continually released, reaching 91.41% within 30 days. Differential new bone formation was observed between the miR-34c-5p Agomir and Antagomir groups. Micro-CT imaging and histological staining revealed varying degrees of bone regeneration, with notable improvements in the miR-34c-5p Antagomir group. Conclusion CTS scaffolds with a composition ratio of 1:0.2:0.1 demonstrate favorable biocompatibility and enable efficient loading and sustained release of miR-34c-5p Antagomir. The study suggests potential applications of miR-34c-5p Antagomir in promoting bone repair and highlights the promise of innovative scaffolds for therapeutic miRNAs administration in bone regeneration.

This study followed the one-step and two-step technologies to produce rice straw biochar (BC), chitosan-modified rice straw biochar (CT-BC), and thiol grafted chitosan-modified biochar (TH@CT-BC) for cadmium (Cd) removal from contaminated water . Here, chitosan and thiol were introduced by two-step combination to enhance the adsorption ability of biochar. Our results revealed the maximum Cd adsorption at pH 5.5 with TH@CT-BC (261.47 mg g −1 ) followed by CT-BC (103.14 mg g −1 ) and BC (29.64 mg g −1 ). Obtained data was best fitted by the Langmuir and pseudo-second-order kinetic models (with R 2 values of 0.997 for TH@CT-BC) as compared to Freundlich and Temkin models (0.949 and 0.925, respectively). TH@CT-BC retained its efficiency in spiked river water system, removing up to 89% of Cd in river water spiked with 30 mg −1 L Cd. The experimental investigations and data calculations demonstrated the surface complexation and electrostatic interaction as dominant underlying mechanisms for Cd removal by TH@CT-BC. Moreover, it is essential to utilize the large amount of produced rice straw to develop new materials which would help in converting the waste to useable product for environmental remediation. Thus, this study demonstrates the production of TH@CT-BC from rice straw as an effective adsorbent of Cd from aqueous systems which can be studied as a potential candidate for practical wastewater treatment applications.

The CO 2 -WAG injection method proposed for unconventional tight oil reservoirs has been successfully applied in field. However, for water-sensitive tight oil reservoirs, there is a lack of research focusing on the displacement mechanisms and effects of WAG injection at the pore scale. And the gap between nuclear magnetic resonance (NMR) and computerized tomography (CT) scanning technologies is not well explored. This study integrated deuterium oxide-shielded NMR and contrast agent (n-butane iodide and lead iso-octanoate mixture) enhanced micro-CT to study the immiscible CO 2 -WAG mechanisms for water-sensitive tight oil. The aim is to investigate the microscopic distribution and displacement mechanisms of residual oil after CO 2 flooding and WAG injection in water-sensitive cores. Experimental results showed that NMR experiments and CT experiments identified the microscopic distribution of oil at different injection stages successfully. The contrast agent, despite its similar viscosity ratio, had poor representativeness in other properties, leading to significant differences in the microscopic fluid distribution results compared to NMR experiments. The primary WAG flood mechanism observed was that CO 2 dispersed oil droplets, with most being displaced out of the pores, while water played a mobility control role by blocking large pores. However, due to water sensitivity, medium and small pores also experienced blockage, making the effect of WAG injection after CO 2 flooding not significant. In the NMR experiment, the final recovery of core 6–1 after WAG injection was 64.71%. In the CT scan experiment, due to the smaller volume of the microcore, the water sensitivity effect was relatively weaker, and under the stronger influence of the capillary end effect, the final recovery reached 76.91%. In the five sets of long-core experiments, by controlling the gas–water ratio and slug size, the recovery ranged from 56.51 to 64.04%. The core flood experiments validated the representativeness of the microscopic experiments. And for water-sensitive tight oil reservoirs, controlling the volumes of water and CO 2 during the WAG injection process is crucial.

Multifunctional nanotheranostic agents have been highly commended due to the application to image-guided cancer therapy. Herein, based on the chemically disordered face centered cubic (fcc) FePt nanoparticles (NPs) and graphene oxide (GO), we develop a pH-responsive FePt-based multifunctional theranostic agent for potential in vivo and in vitro dual modal MRI/CT imaging and in situ cancer inhibition. The fcc-FePt will release highly active Fe ions due to the low pH in tumor cells, which would catalyze H2O2 decomposition into reactive oxygen species (ROS) within the cells and further induce cancer cell apoptosis. Conjugated with folic acid (FA), the iron platinum-dimercaptosuccinnic acid/PEGylated graphene oxide-folic acid (FePt-DMSA/GO-PEG-FA) composite nanoassemblies (FePt/GO CNs) could effectively target and show significant toxicity to FA receptor-positive tumor cells, but no obvious toxicity to FA receptor-negative normal cells, which was evaluated by WST-1 assay. The FePt-based multifunctional nanoparticles allow real-time monitoring of Fe release by T2-weighted MRI, and the selective contrast enhancement in CT could be estimated in vivo after injection. The results showed that FePt-based NPs displayed excellent biocompatibility and favorable MRI/CT imaging ability in vivo and in vitro. Meanwhile, the decomposition of FePt will dramatically decrease the T2-weighted MRI signal and increase the ROS signal, which enables real-time and in situ visualized monitoring of Fe release in tumor cells. In addition, the self-sacrificial decomposition of fcc-FePt will be propitious to the self-clearance of the as-prepared FePt-based nanocomposite in vivo. Therefore, the FePt/GO CNs could serve as a potential multifunctional theranostic nanoplatform of MRI/CT imaging guided cancer diagnosis and therapy in the clinic.

As promising luminescence nanoparticles, near-infrared (NIR) persistent luminescence nanoparticles (PLNPs) have received extensive attention in the field of high-sensitivity bioimaging in recent years. However, NIR PLNPs face problems such as short excitation wavelengths and single imaging modes, which limit their applications in vivo reactivated imaging and multimodal imaging. Here, we report for the first time novel Gd2GaTaO7:Cr3+, Yb3+ (GGTO) NIR PLNPs that integrate X-ray activated NIR persistent luminescence (PersL), high X-ray attenuation and excellent magnetic properties into a single nanoparticle (NP). In this case, Cr3+ is used as the luminescence center. The co-doped Yb3+ and coating effectively enhance the X-ray activated NIR PersL. At the same time, the presence of the high-Z element Ta also makes the GGTO NPs exhibit high X-ray attenuation performance, which can be used as a CT contrast agent to achieve in vivo CT imaging. In addition, since the matrix contain a large amount of Gd, the GGTO NPs show remarkable magnetic properties, which can realize in vivo MR imaging. GGTO NPs combine the trimodal benefits of X-ray reactivated PersL, CT and MR imaging and are suitable for single or combined applications that require high sensitivity and spatial resolution imaging

Background Circular RNAs (circRNAs) are involved in the progression of osteoporosis; however, their impact on osteogenic differentiation has yet to be fully elucidated. In this study, we identified a novel circRNA known as circZfp644-205 and investigated its effect on osteogenic differentiation and apoptosis in osteoporosis. Methods CircZfp644-205, miR-445-3p, and SMAD2 levels were measured using quantitative real-time polymerase chain reaction (qRT-PCR). MC3T3-E1 cells were subjected to microgravity (MG) to establish a cell model. Osteogenic differentiation was assessed using qRT-PCR, Alizarin Red S staining, alkaline phosphatase staining, and western blot. The apoptosis was evaluated using flow cytometry. The relationship between miR-445-3p and circZfp644-205 or SMAD2 was determined using bioinformatics, RNA pull-down, and luciferase reporter assay. Moreover, a hindlimb unloading mouse model was generated to investigate the role of circZfp644-205 in vivo using Micro-CT. Results CircZfp644-205 expression was up-regulated significantly in HG-treated MC3T3-E1 cells. Further in vitro studies confirmed that circZfp644-205 knockdown inhibited the osteogenic differentiation and induced apoptosis of pre-osteoblasts. CircZfp644-205 acted as a sponge for miR-455-3p, which reversed the effects of circZfp644-205 on pre-osteoblasts. Moreover, miR-455-3p directly targeted SMAD2, thus inhibiting the expression of SMAD2 to regulate cellular behaviors. Moreover, circZfp644-205 alleviated the progression of osteoporosis in mice. Conclusions This study provides a novel circRNA that may serve as a potential therapeutic target for osteoporosis and expands our understanding of the molecular mechanism underlying the progression of osteoporosis.
