Modified chitosan-fabricated antifungal transdermal array for dermatophytosis therapy

Feng Feng,Xing Ge,Tianye Fang,Kaiyang Chen,Jiaying Zhu,Jun Yang,Lei Tao,Jianjun Dai,Yanmin Ju

Dermatophytosis is a complex and prevalent fungal skin infection that affects a significant portion of the global population. Conventional antifungal therapies are often compromised by their inability to effectively penetrate fungal biofilms and limited skin permeability, ultimately leading to unsatisfactory clinical results. To address these issues, we proposed a novel dissolvable microneedle (MN) patch (denoted as TB/QCS MNs) that encapsulated terbinafine (TB) within the quaternary ammonium chitosan (QCS) MN tips. The combination of QCS and TB exhibited a pronounced synergistic effect against Trichophyton mentagrophytes (T. mentagrophytes). Importantly, TB/QCS MNs effectively disrupted fungal biofilms and damaged extracellular polymeric substances. In comparison to TB alone, TB/QCS MNs effectively accelerated fungal clearance, shortened treatment time, and enhanced skin recovery in T. mentagrophytes-infected C57BL/6 mice. These findings demonstrated that TB/QCS MNs significantly improve therapeutic efficacy for dermatophytosis, offering a promising alternative for clinical treatment of skin fungal infections.

Xiangru Han,Liguang Tian,Qiuhong Chen,Linqing Wang,Yingqi Mi,Qing Li,Zhanyong Guo,Fang Dong

The aim of this study was to synthesize thiolated chitosan derivative and evaluated its antioxidant activity. Thiol groups were introduced into chitosan and hydroxypropyl trimethylammonium chloride chitosan quaternary ammonium salt (HACC) by microwave reaction, respectively. At the same time, a series of compounds containing thiol groups were introduced at the amino site. The structure of the chitosan derivatives was identified by FTIR and 1 H NMR. The thiol groups content, thermal stability, water solubility and cytotoxicity of the chitosan derivative were evaluated. The antioxidant activity of chitosan derivatives was evaluated using superoxide anion radical scavenging activity assay, DPPH radical scavenging activity assay, hydroxyl radical scavenging activity assay and reducing power assay. The results showed that the thiolated chitosan derivatives obtained in this study possessed excellent antioxidant activity, which provides a new idea for the further study of chitosan antioxidants.

Coordination of Mg2+ with Chitosan for Enhanced Triboelectric Performance

Jingjia He,Lili Wang,Kaiyuan Zheng,Shoukang Hu,Xueke Zhang,Ziyu Mu

In this work, Mg2+modified chitosan (Mg2+/CS) is proposed and successfully designed. By investigating the effects of the Mg2+and CS interaction on hydrogen bonding, dipoles, charge density, surface potential, and roughness, the coordination between Mg2+and CS is verified and the mechanism of coordination improving tribological properties is elucidated. The Mg2+/CS coordination structure enhances intermolecular interactions, promoting the formation of new hydrogen bonds and increasing the dipoles. Compared to CS, the relative dielectric constant of Mg2+/CS increased by 76%, the surface potential increased by 70 mV, and the root mean square roughness increased by 39.4 nm. The open-circuit voltage, short-circuit current, and charge density of the triboelectric nanogenerator (TENG) fabricated from Mg2+/CS were increased by 100%, 94%, and 75%, respectively, compared to the CS-TENG fabricated from pure CS. The coordination of Mg2+increased the charge density of the Mg2+/CS-TENG, significantly enhancing its charge transfer capability. The Mg2+/CS-TENG successfully provided power for photodetectors and LEDs. Mg2+/CS exhibited excellent flexibility and skin adhesion, and the Mg2+/CS-TENG successfully converted the mechanical energy generated by human joint motion into electrical signals. The coordination structure of Mg2+with CS enhances the triboelectric performance of Mg2+/CS-TENG, providing new light for the research of chitosan-based TENGs.

Genipin-enriched chitosan-Zein nanoparticles for improved curcumin encapsulation

Lingling Sha,Husnain Raza,Chengsheng Jia,Imran Mahmood Khan,Huqing Yang,Gang Chen

Polysaccharide-protein nanocomplexes are considered one of the desired systems to encapsulate bioactive compounds. The study prepared chitosan (CS)/genipin (GP)/zein nanoparticles with chemical cross-linkage for encapsulating and releasing curcumin. Fourier transform infrared spectra demonstrated that cross-linkage between molecules was attributed to electrostatic interactions and the formation of amido bonds and hydrogen bonding. Transmission electron microscopy showed that all prepared nanoparticles showed a spherical morphology. Particularly, cinnamaldehyde screening –NH 2 groups in CS significantly reduced the encapsulation efficiency of nanoparticles, demonstrating the significant role of the –NH 2 group in CS-coated zein nanoparticles. Additionally, the encapsulation efficiency was largely increased to 82.3 % at the ratio of CS/GP/zein ( w /w, 20:1:100), compared with 38.4 % using zein as a single encapsulation for curcumin. In vitro digestion, the addition of GP and CS decreased the release of curcumin from 84.8 % to 57.1 % after the initial digestion of 2 h. The GP/CS/zein particles retained 12 % of curcumin after 6- h of digestion, indicating the improvements in encapsulation efficiency and release properties.

A comparative study on Fe(III)-chitosan and Fe(III)-chitosan-CTAB composites for As(V) removal from water: preparation, characterization and reaction mechanism

Jiang Changjin,Zhang Ting,Li Shuhui,Yang Zhaoguang

Fe(III)-chitosan and Fe(III)-chitosan-CTAB composites were prepared using an ionotropic gelation method. Various techniques were used to analyze the morphology, structure, and property of the adsorbents, including SEM, EDS, FT-IR, XPS, and zeta potential. Compared with Fe(III)-chitosan, Fe(III)-chitosan-CTAB was more effective for As(V) adsorption at a wide range of pH (3.0–8.0). The adsorption of As(V) onto Fe(III)-chitosan and Fe(III)-chitosan-CTAB could reach equilibrium in 20 min, and their maximum adsorption capacities were 33.85 and 31.69 mg g ‒1 , respectively. The adsorption kinetics was best described by the pseudo-second-order model ( R 2  = 0.998 and 0.992), whereas the adsorption isotherm was fitted well by the Freundlich model ( R 2  = 0.963 and 0.987). The presence of H 2 PO 4 – significantly inhibited the adsorption of As(V) onto Fe(III)-chitosan and Fe(III)-chitosan-CTAB, and humic acid also led to a slight decrease in As(V) adsorption by Fe(III)-chitosan-CTAB. Over 94% of As(V) at the initial concentration of no more than 5 mg L –1 was removed from real water by the two adsorbents. 1% (w/v) NaOH solution was determined to be the most suitable desorption agent. Fe(III)-chitosan and Fe(III)-chitosan-CTAB still maintained their initial adsorption capacities after five adsorption–desorption cycles. Based on different characterization results, both electrostatic attraction and surface complexation mechanisms played important roles in As(V) adsorption on Fe(III)-chitosan and Fe(III)-chitosan-CTAB.

Novel dual-sensitization electrochemiluminescence immunosensor using photopermeable Ru(bpy)32+-doped chitosan/SiO2 nanoparticles as labels and chitosan-decorated Nafion/MWNTs composites as enhancer

Zheng Chang,Chao Zhang,Binghua Yao

A novel dual-sensitization electrochemiluminescence (ECL) immunosensor for the detection of tumour protein prostate specific antigen (PSA) at trace level using Ru(bpy) 3 2+ -doped chitosan/SiO 2 nanoparticles (Ru(bpy) 3 2+ /chitosan/SiO 2 NPs) as the first signal enhancers was fabricated. Due to chitosan with excellent pore forming capacity, these nanoparticles possess porous structures and better photopermeability, and therefore have higher luminescence efficiencies compared with Ru(bpy) 3 2+ /SiO 2 NPs reported in previous publications. Conversely, chitosan with good biocompatibility and high hydrophilicity was electrochemically decorated onto a Nafion/multiwall carbon nanotubes (Nafion/MWNTs) modified electrode surface and used as the second sensitizing matrix to seize large amounts of prostate specific capture antibody (Ab 1 ). The chitosan-decorated Nafion/MWNTs composites exhibited a 5.5-times higher ECL intensity than the unadorned Nafion/MWNTs films. Also, without additional reagents, such as (3-aminopropyl)triethoxysilane (APTS), the one-step functionalized Ru(bpy) 3 2+ /chitosan/SiO 2 NPs provided a large number of active arms to connect with PSA-detected antibodies (Ab 2 ) through the amino groups in chitosan. After a sandwich immunoreaction, the PSA antigen and Ru(bpy) 3 2+ /chitosan/SiO 2 NPs-labelled Ab 2 were sequentially captured onto the Ab 1 /chitosan/Nafion/MWNTs-modified electrode surface. The ECL signal increases were linearly related to the PSA antigen concentrations and ranged from 0.01 pg·mLl −1 to 10.0 pg·mLl −1 . Under the optimized experimental conditions, the immunosensor displayed excellent sensitivity and selectivity. The detection limit was as low as 3.4 fg·mLl −1 , equivalent to, or better than, those of the reported ECL immunosensors for PSA.

Sustained release of valproic acid loaded on chitosan nanoparticles within hybrid of alginate/chitosan hydrogel with/without stem cells in regeneration of spinal cord injury

Jafarimanesh Mohammad Amin,Ai Jafar,Shojaei Shahrokh,Khonakdar Hossein Ali,Darbemamieh Goldis,Shirian Sadegh

Hydrogels have been increasingly applied in tissue regeneration and drug delivery systems (DDS). In this study, the capacity of valproic acid (Val) encapsulated within hybrid of alginate (Alg)-chitosan (Cs) (Alg-Cs) hydrogel containing Cs nanoparticle (Npch) with/without human endometrial stem cells (hEnSC) was initially examined for regeneration of spinal cord injury (SCI). To evaluate the stability of the synthesized hydrogels zeta potential necessary measurements were made. Physicochemically, the developed hydrogels were evaluated using Fourier-transform infrared (FTIR) spectroscopy. The physical properties including degradation rate, swelling ability, and tunability of the synthesized hydrogels were studied. To evaluate the nerve regeneration ability of the synthesized hydrogels, 35 Sprague–Dawley rats were undergone SCI. The spinal cords were exposed using laminectomy in T9-T10 area and the hemi-section SCI model was made. The rats were then randomly divided into 5 groups ( n  = 7) including, Alg-Cs/Npch, Alg-Cs/Npch/hEnSCs, Alg-Cs/Npch/Val, and Alg-Cs/Npch/hEnScs/Val, and the control groups without any intervention. The FTIR spectra showed band frequencies and assignments of Val, Alg-Cs, and alginate. Nanoparticles were formulated with a mean diameter of 187 and 210 nm, for Val/Alg-Cs and Alg-Cs, respectively. The loading of Val into Alg-Cs led to its reduced size by about 40 nm. The Cs-Npch/Val hydrogels degraded faster than the Alg-Cs-/Npch/Val hydrogel specifically in extended time of incubation. A higher swelling capacity of Alg-Cs/Npch hydrogel, compared to Cs/Npch/Val and Alg-Cs/Npch/Val hydrogels, was found. The Cs-Npch/Val hydrogels degraded faster than Alg-Cs-/Npch/Val hydrogel. The Alg-Cs/Npch/hEnSCs/Val could regenerate the damaged nerve fibers and histologically prevent the SCI-induced vacuolization spaces. The prepared Alg-Cs/Npch/Val could be a suitable polymeric carrier for taurine drugs as bioactive substrate in nerve tissue engineering (NTE) and DDS.

The chitosan/carboxymethyl cellulose/montmorillonite scaffolds incorporated with epigallocatechin-3-gallate-loaded chitosan microspheres for promoting osteogenesis of human umbilical cord-derived mesenchymal stem cell

Wang Jin,He Wubo,Tan Wen-Song,Cai Haibo

Epigallocatechin-3-gallate (EGCG) is a plant-derived flavonoid compound with the ability to promote the differentiation of human bone marrow-derived mesenchymal stem cells (MSCs) into osteoblasts. However, the effect of EGCG on the osteogenic differentiation of the human umbilical cord-derived mesenchymal stem cells (HUMSCs) is rarely studied. Therefore, in this study, the osteogenic effects of EGCG are studied in the HUMSCs by detecting cell proliferation, alkaline phosphatase (ALP) activity, calcium deposition and the expression of relevant osteogenic markers. The results showed that EGCG can promote the proliferation and osteogenic differentiation of the HUMSCs in vitro at a concentration of 2.5–5.0 μM. Unfortunately, the EGCG is easily metabolized by cells during cell culture, which reduces its bioavailability. Therefore, in this paper, EGCG-loaded microspheres (ECM) were prepared and embedded in chitosan/carboxymethyl cellulose/montmorillonite (CS/CMC/MMT) scaffolds to form CS/CMC/MMT-ECM scaffolds for improving the bioavailability of EGCG. The HUMSCs were cultured on CS/CMC/MMT-ECM scaffolds to induce osteogenic differentiation. The results showed that the CS/CMC/MMT-ECM scaffold continuously released EGCG for up to 22 days. In addition, CS/CMC/MMT-ECM scaffolds can promote osteoblast differentiation. Taken together, the present study suggested that entrainment of ECM into CS/CMC/MMT scaffolds was a prospective scheme for promotion osteogenic differentiation of the HUMSCs. Graphical Abstract

Characterization and antibacterial effect of quaternized chitosan anchored cellulose beads

Yinfeng Tan,Hao Wu,Tianzhi Xie,Lingling Chen,Shuqian Hu,Huafeng Tian,Yutang Wang,Jianguo Wang

Some thermoduric food spoilage bacteria pose great threat to beverage industry . To tackle the challenge, hydroxypropyl trimethyl ammonium chloride chitosan (HTCC) grafted magnetic cellulose beads have been prepared via a dropping technology. Sodium periodate oxidation process was carried out to form dialdehyde functional groups on the regenerated cellulose beads mixed with maghemite nanoparticles . HTCC was anchored on the beads through Schiff base reaction. The structure and properties of HTCC anchored beads were evaluated by Fourier transform infrared (FTIR) spectra, field emission scanning electron microscope (FESEM), energy dispersive X-ray spectroscopy (EDS), X-ray diffraction (XRD) and vibrating sample magnetometer (VSM). Thermal stability of the beads was estimated by thermogravimetric analysis (TGA) and differential thermal gravity (DTG), and the decomposition temperature of the beads were around 200–300 °C. A long-term antibacterial activities of the beads against Alicyclobacillus acidoterrestris were confirmed caused by the covalent bond between HTCC and the beads. The biodegradable HTCC grafted cellulose beads may provide a novel approach for food safety management .

New Family of Benzimidazole-Based Chitosan Derivatives against Penicillium expansum

Yan Lv,Jing Liu,Yujun Zhang,Yiyu Zhou,Jianying Huang,Wenjie Wang,Xiancheng Ye

Penicillium expansum is the major fungus that causes blue mold and produces patulin, threatening human health. Due to health and environmental pollution concerns, chitosan (CS) has attracted more and more attention as a safer alternative to synthetic fungicides for the control of blue mold. In the present study, four different benzimidazole groups were introduced onto CS by the acylation reaction to obtain benzimidazole-based chitosan derivatives (R1b–R4b). After being well-characterized with Fourier transform infrared spectroscopy (FTIR), ultraviolet–visible spectroscopy (UV–vis spectra), and nuclear magnetic resonance (NMR), their antifungal activities against P. expansum were screened. Results showed that the inhibitory effects of chitosan derivatives against the pathogen were significantly correlated with chitosan derivatives’ concentration and their structures. R4b was shown as optimum with good solubility and antifungal activity with a minimum inhibitory concentration (MIC) value of 0.5 mg/mL and a minimum fungicidal concentration (MFC) value of 2.0 mg/mL. The remarkable antifungal efficiency of R4b against P. expansum was further demonstrated in terms of spore germination, mycelial growth, patulin production, and the preliminary antifungal mechanism. R4b exhibited significant inhibition of patulin production, while its antifungal mechanism was revealed by destroying cell membrane integrity and inducing membrane depolarization. Furthermore, R4b treatment could significantly reduce the incidence of blue mold rot in apple fruit, and the MTT assay showed the nontoxicity of R4b against Raw 264.7, HBZY-1, and Caco-2 cells. Altogether, these results indicate that it is promising to be used as a fruit preservative in the future.

The Preparation of Capsaicin-Chitosan Microspheres (CCMS) Enteric Coated Tablets

Jian Chen,Gui-Dong Huang,Si-Rong Tan,Jiao Guo,Zheng-Quan Su

This study aimed to research the preparation and content determination of capsaicin-chitosan microspheres (CCMS) enteric coated tablets. The core tablets were prepared with the method of wet granulation. Nine formulae were designed to determine the optimal formula of the core tablet. Eudragit L100 was used to prepare the CCMS enteric-coated tablets. The effect of enteric coated formulation variables such as content of talc (10%, 25% and 40%), plasticisers (TEC and DBS), dosage of plasticiser (10%, 20% and 30%) and coating weight (2%, 3% and 5%) were evaluated for drug release characteristics. Thein vitrorelease was studied using 0.1 N HCl and pH 6.8 phosphate buffer. Enteric coated tablets without ruptures or swelling behaviour over 2 h in 0.1 N HCl indicated that these tablets showed acid resistance. The accumulated release rate in phosphate buffer (pH 6.8) revealed that the prepared tablets were able to sustain drug release into the intestine and a first-order release was obtained for capsaicin. This research is the first report of the preparation and content determination of CCMS enteric coated tablets. The sustained release behavior of enteric coated formulations in pH 6.8 phosphate buffer demonstrated that it would be a potential drug delivery platform for sustained delivery of gastric irritant drugs.

Amidoxime functionalized magnetic MXene/chitosan composites for efficient uranium extraction

Hao Liang,Nana Wang,Wei Tian,Hongping Zhang,Cuicui Li,Ruibin Guo,Zunli Mo,Nijuan Liu

Uranium extraction from unconventional uranium resources is among the major constraints to the development of the nuclear industry at present. In this dissertation, we here propose to effectively graft amidoxime groups on magnetic MXene and chitosan (CS) composites to extract uranium from low-enriched uranium solutions (MCF-AO). The data of adsorption experiments displayed that MCF-AO possessed fast adsorption kinetics (within 120 min), outstanding selectivity and salient adsorption capacity (1305.7 mg/g at pH=4, T=298.15 K) for uranium and could be easily separated with a magnet. Adsorption studies revealed a behavior consistent with the pseudo-second-order kinetic model and the Langmuir adsorption isotherm model, which is suitable for the spontaneous heat-absorption chemical monolayer adsorption process . Its sorption remains at more than 55 % of the original after 5 adsorption-desorption tests, and has great tolerance to interfering ions. Mechanistic research pointed out that the selective adsorption of uranium by MCF-AO was significantly enhanced by the presence of hydroxyl, amino and amidoxime groups. Meanwhile, MCF-AO exhibited superb resistance to two common bacteria (E. coli and B. subtilis ) and achieved more than 95 % uranium extraction capacity in simulated seawater , even more 4.12 mg/g of uranium extraction in Salt Lake water. Consequently, MCF-AO has a broad application prospect in the recovery and treatment of complex uranium systems.
