Background Precise and specific miRNA detection plays a vital role in exploring development mechanisms of cancer disease, thereby it can significantly improve relevant prevention and treatment strategies. Results In this work, a surface-enhanced Raman spectroscopy (SERS)-based microfluidic chip has been devised with a microcone array SERS substrate (MCASS) for the miR-141 detection. This substrate excels in unique SERS activity and large surface area for DNA oligonucleotide modification. As the presence of miR-141, the DNAzyme walker induced cleavage reaction took place on the finely designed and prepared dual DNA conjugated SERS nanoprobes. The SERS nanoprobes can anchor on MCASS by the DNA hybridization that achieved an impressive detection limit in the femtomolar level. Significance With this integrated SERS-based microfluidic chip, we provided a miRNA detection strategy using DNAzyme walker amplification technology. It is believed that this strategy could be a powerful tool for miRNA detection and related cancer screening test .

N 6 -methyladenosine (m 6 A) is a crucial post-transcriptional epigenetic modification in RNAs which associates with numerous cancers development. Here, a cost-effective, and easy-to-operate fluorescence strategy for m 6 A analysis was developed based on the different cleavage efficiency of 8–17 DNAzyme for methylation and unmethylation sites. By arranging 8–17 DNAzyme to recognize and cleave 5'-rAG-3' and 5'-rm 6 AG-3' motifs in the target sequence respectively, various short ssDNA oligoes were produced, leaving different amounts of target sequence. Benefiting from the varying degrees of retardation of CHA amplification reaction, the tiny discrepancy between m 6 A and unmethylated adenine (A) can be distinguished. Moreover, we found that 8–17 DNAzyme was more sensitive to m 6 A than to A. Importantly, the strategy we proposed not only avoids sophisticated chemical labelling and expensive proteins, but also can be easily performed on the ordinary heating apparatus of accessible laboratory. The DCHA strategy demonstrated good analytical performance in complex biological matrices, and was successfully applied to hepatocellular carcinoma-related m 6 A analysis in CTNNB1, MGAT5 and SOCS-2 mRNAs with satisfactory results, showing its potential in clinical analysis.

Orthopedic implants have a high failure rate due to insufficient interfacial osseointegration, especially under osteoporotic conditions. Type H vessels are CD31+EMCN+ capillaries with crucial roles in mediating new bone formation, but their abundance in osteoporotic fracture site is highly limited. Herein, we report a nanoengineered composite coating to improve the in situ osseointegration of a Ti implant for osteoporotic fracture repair, which is realized through inhibiting the stimulator of interferon genes (STING) in endothelial cells (ECs) to stimulate type H vessel formation. Autonomously catalytic DNAzyme-ZnO nanoflowers (DNFzns) were prepared through rolling circle amplification (RCA) of STING mRNA-degrading DNAzymes, which were then integrated on the Ti surface and further sequentially complexed with thioketal-bridged polydopamine and naringenin (Ti/DNFzn/PDA-Nar). ECs and mesenchymal stem cells (MSCs) can be recruited to the implant surface by galvanotaxis, accounting for the negative charges of DNFzn/PDA-Nar, subsequently released Nar under reactive oxygen species (ROS) stimulation to upregulate endothelial nitric oxide synthase (eNOS) in recruited ECs, leading to enhanced local angiogenesis. Meanwhile, the coordinately released DNFzns would abolish STING expression in ECs to transform the newly formed vessels into Type H vessels, thus substantially promoting the osseointegration of Ti implants. This study provides application prospects for improving implant osteointegration for osteoporotic fracture treatment.

Ultrasensitive detection for low concentration and abnormal expression of microRNA-155 (miR-155) in serum (fM level) at the early stage of cancer still exist great challenges. Therefore, a dual-model photoelectrochemical biosensor was developed based on label-free trimetallic nanoprobe and Mg 2+ dependent DNAzyme driven DNA walker to generate ratiometric dual-signal output for sensitive biosensing of miR-155 from breast cancer cells (MCF-7) and cervical cancer cells (HeLa). In signal amplification strategy, PtCo@Prussian Blue nanozyme was selected as a signal nanoprobe with high catalytic activity and large specific surface area, while two dimensional MXene/AuNPs as surface modification material was applied to improve the active sites of the electrode. With the presence of the target, the DNAzyme assisted DNA walker process was initiated and abundant signal nanoprobe was introduced to the electrode surface , producing high current response and weak ultraviolet-visible spectrophotometry (UV–vis) response. Hence, the biosensor for miR-155 shows a low limit of detection of 1.2 fM from the linear range of 5 fM to 5 nM, exhibiting excellent stability and reproducibility. Moreover, the proposed biosensor demonstrates superior performance for miR-155 in complex human serum samples and tumor cell lysates, presenting great application potential for early-stage clinical diagnosis.

In this paper, we developed an amplified fluorescence biosensor for acetylcholinesterase (AChE) activity detection by taking advantage of the mercury ion-mediated Mgzyme (Mg2+-dependent DNAzyme) activity. The catalytic activity of Mgzyme can be inhibited by the formation of T–Hg2+–T base pairs between the Mgzyme and mercury ions. Therefore, the Mgzyme–Hg2+ complex has no activity on a molecular beacon (MB) substrate, which afforded a very weak fluorescence background for this biosensor. After the addition of acetylcholinesterase (AChE), the substrate acetylthiocholine could be hydrolyzed to thiocholine, which has a stronger binding power with mercury ions than T–Hg2+–T base pairs. Therefore, the Mgzyme activity was recovered. The activated Mgzyme could hybridize with the MB substrate and undergo many cleavage cycles, resulting in a significant increase of fluorescence intensity. This biosensor displayed high sensitivity with the detection limit as low as 0.01 mU mL−1. Moreover, this design did not require complex composition and sequence design; thus it is simple and convenient. This biosensor was also applied for the determination of AChE in human blood and showed satisfactory results.

The regulation of the catalytic activities of DNAzymes is of great significance for their biosensing and biomedical applications. Herein, thermoregulation of the catalytic activity of DNAzyme is achieved through the immobilization of DNAzymes into the crosslinked polymeric matrix of poly- N -isopropylacrylamide (pNIPAM) microgels prepared with a microfluidic device. At a temperature lower than the lower critical solution temperature (LCST) of pNIPAM, the immobilized DNAzymes exhibit lower catalytic performance due to the restricted entry of DNA substrates into the polymeric networks; while at a temperature higher than the LCST, the hydrophilic DNAzyme structures are exposed to exhibit high catalytic activity due to the shrinking of the pNIPAM networks. Results show a more than 60% of increase in catalytic activity by raising the temperature from 29 to 34 ° C and reversible regulation of the DNAzyme catalytic activity is achieved by cyclic raising and lowering of the solution temperature. Through a simple heating-centrifugation process, the DNAzyme/pNIPAM microgels can be recycled for further applications. Moreover, by introducing photothermal MXene nanosheets, the remote control of the catalytic activity of the DNAzyme by near-infrared light irradiation is achieved. These DNAzyme/pNIPAM microgels as low-cost, stable, reversible, and recyclable biocatalyst may have potential in biosensing and biomedical applications.

The investigation on invertase (INV) and glucose oxidase (GOx)-dominated biological process offers a new opportunity for the development of clinical diagnosis and prognostic treatment. Herein, a ZnO nanoflowers (ZnONFs)-assisted DNAzyme-based electrochemical platform for INV- and GOx-dominated biosensing is proposed by the change of pH in microenvironment. In this strategy, INV can usually catalyze the dissolution of sucrose to generate glucose, and glucose is then consumed by GOx to produce H 2 O 2 and gluconic acid , in which ZnONFs can be effectively etched into free Zn 2+ ions. Subsequently, the released Zn 2+ ions have a shearing action for Zn 2+ -specific DNAzyme, thus triggering hybridization chain reaction along with the imbedding of methylene blue . The excellent electrochemical signals illustrate the method can be employed well for testing sucrose, INV and GOx with a low detection limit (0.019 μM, 0.047 mU/mL and 0.012 mU/mL, respectively). Finally, a series of basic and advanced logic gates (YES, AND, INHIBIT, and AND-AND-INHIBIT) in the biological process are constructed with different logic inputs, providing a valuable platform for the establishment of advanced molecular devices for bioanalysis and clinical diagnostics.

Zeolitic imidazolate frameworks (ZIFs), a famous subfamily of metal-organic frameworks (MOFs), are considered promising electrocatalysts. Herein, ZIF-67 was selected as an electrocatalyst for designing electrochemical sensors due to having the best electrocatalytic activity in ZIFs. To overcome the insufficient electrocatalytic activity of ZIFs, ZIF-67 derivatives (QZIF-67-X, where X represents calcination time) were obtained by calcining at 250 °C for a certain time. The porous structure of the precursor in QZIF-67-X is maintained, exposing more active centers. QZIF-67-X could accelerate electron transfer and lead to improve the electrocatalytic performance. Moreover, QZIF-67-2 was chosen as an Au nanoparticle-supported nanocarrier to further bind G-quadruplex/hemin DNAzymes with strong catalytic activity due to the best supporting activity of QZIF-67-2 among QZIF-67-X. The synergistic catalysis of QZIF-67-2 and G-quadruplex/hemin DNAzymes effectively amplified the reduction current signal of H 2 O 2 . The linear range of the prepared electrochemical sensor was 2 μM-65 mM, and the detection limit was 1.2 μM. Moreover, the real-time detection of H 2 O 2 from HepG2 cells was achieved by the sensor, providing a novel technique for efficient anticancer drug evaluation. These results suggested that QZIF-67 can be utilized as an efficient electrocatalyst for improving the sensitivity of sensors.

Calcium is one of the most abundant and indispensable elements in biology, as it is a vital component of nerves, bones, and muscles and maintains the excitability of normal neuromuscular muscles. However, it may be harmful to the human body and even damage the organs if the calcium content exceeds the standard value by several times. To evaluate the level of calcium ions (Ca 2+ ), an electrochemical biosensor (FET/SWNTs/Cazyme) was developed using a nonspecific DNAzyme with high stability, which combined the unique advantage of field-effect transistors and single-walled carbon nanotubes, while being easy-to-use and having excellent sensitivity. The incubation time and voltage after optimization were 15 min and +0.02 V. The nonspecific DNAzyme-based biosensor was sensitive to Ca 2+ , but it was also interfered with by Pb 2+ , which affected the detection accuracy. To solve this shortcoming, an electrochemical device was proposed, in which FET/SWNTs/Cazyme combined with other specific biosensors for Pb 2+ , and then established some data processing models were established through support vector machine regression (SVMR) and artificial neural network fitting (ANNF). For the optimal SVMR, the electrochemical device can determine the Ca 2+ concentration in the range of 7.5–1000 μM with a detection limit of 5.48 μM. Finally, the prepared electrochemical device was employed to detect the Ca 2+ in different milk and water samples.

Due to similar charges, atomic radii, and chemical properties of most metal ions , the selective sensing of these metal ions and imaging in vivo is still a challenge. A DNAzyme assemblied and near-infrared (NIR) light excited nanosensor was developed to detect and image Pb 2+ . In this nanosensor, NaYF 4 :Yb, Er upconversion nanoparticles (UCNPs) introduced as NIR-to-Vis transducer were the donor of luminescence resonance energy transfer (LRET), and DNAzyme-functionalized black hole quencher 1 (BHQ1) acted as energy transfer acceptor. This proposed nanosensor was applied to detecting Pb 2+ in solution with high sensitivity and selectivity . Furthermore, we have successfully demonstrated the imaging ability of this nanosensor towords Pb 2+ in living cells and early-stage zebrafish with negligible autofluorescence and good photostability. The UCNP-DNAzyme nanosensor would enrich the method of imaging Pb 2+ in vivo, and might serve as a potential tool for understanding the metabolic pathways of Pb 2+ and the mechanism of lead poisoning in biological system.

T4 polynucleotide kinase (T4 PNK), a bifunctional repair enzyme, catalyzes the transfer of γ-phosphate residues from adenosine triphosphate (ATP) to 5′- hydroxyl termini in nucleic acids, which plays an essential role in regulating DNA phosphorylation modes during the repair of damaged DNA. Herein, a sensitive DNAzyme-driven DNA walker biosensor was developed for amplified electrochemical detection of T4 PNK activity and inhibition based on T4 PNK-triggered phosphorylation reaction and λ exonuclease (λ exo) cleavage. A novel functional hairpin-shaped swing arm DNA (SA) containing a DNA sequence of the Pb 2+ -dependent DNAzyme is designed to specifically bind to and hydrolyze the substrate strand (rSS) sequence that can drive DNA walker. Upon the reaction of SA, T4 PNK and ATP, λ exo removes the mononucleotides from the stem of the phosphorylated SA, unfolding the hairpin structure and initiating DNAzyme-driven DNA walker. Subsequently, the DNA walker can repeatedly bind and cleave rSS with the aid of Pb 2+ , releasing the methylene blue (MB) - labeled fragment of rSS. The T4 PNK activity can be facilely monitored by recording the electrochemical signal change of MB. The proposed biosensor exhibited a broad linear range from 0.01 U mL −1 to 15 U mL −1 with a low detection limit of 0.001 U mL −1 . The proposed biosensor has also been utilized to screen the T4 PNK inhibitors and exhibited good performance in real sample analysis, making such a strategy hold potential promise in biological fields and clinical diagnosis.

DNA logic-gate technology has significantly advanced cancer diagnostics by enhancing detection specificity through programmable molecular recognition. However, conventional implementations face critical limitations including inadequate sensitivity from dependence on nucleic acid strand displacement reactions without intrinsic amplification, alongside operational complexity imposed by enzymatic dependencies and diverse reagent requirements. To overcome these constraints, we developed an enzyme-free Logic-gated Cascade with Multi-DNAzyme Activation (LCMDA). This system employs an AND-gated architecture where target miRNAs such as miRNA-141 and miRNA-375 convert into specific DNAzymes. These DNAzymes then cleave substrates to release initiators for hybridization chain reaction, whose products self-assemble into catalytically active multi-component deoxyribozymes that drive secondary signal amplification. The platform delivers three pivotal advantages through its enzyme-free AND-gated architecture. First, cascaded signal amplification enables ultrahigh sensitivity, achieving detection limits of 4.11 fM for miRNA-141 and 7.97 fM for miRNA-375. Second, complete elimination of proteolytic enzymes reduces costs while enhancing operational stability. Third, room-temperature operation facilitates point-of-care deployment without compromising performance. Validated in prostate cancer biomarker analysis, LCMDA achieves single-base mismatch discrimination and demonstrates near-quantitative recovery rates between 102.23 % and 95.04 % in human serum samples. Its modular architecture permits straightforward adaptation to diverse miRNA targets through simple reconfiguration of binding domains, establishing a versatile tool for precision oncology research and clinical diagnostics.
