A novel electrochemiluminescence (ECL) biosensor based on CdSe@CdS quantum dots (QDs) functionalized MoS2 modified electrode was developed for sensitive detection of Pb(II) using HRP-mimicking DNAzyme biocatalytic precipitation (BCP) for ECL signal quenching. Polycationic poly(diallyldimethylammonium chloride) (PDDA) and negatively charged QDs were adsorbed on the surface of MoS2, in sequence, to form the MoS2–QDs composites, which were employed as a matrix for the immobilization of the G-rich DNA strand, i.e., T30695. The MoS2–QDs were characterized by UV-vis absorption spectroscopy, fluorescence spectroscopy, energy dispersive spectrometry, transmission electron microscopy, and field emission scanning electron microscopy. CdSe@CdS QDs can produce an ECL signal using 0.1 M K2S2O8 as a co-reactant. T30695 can combine with Pb(II) ions to form a stable parallel G-quadruplex. The G-quadruplex can further combine with hemin to form DNAzyme, which can catalyze H2O2 and oxidize 4-chloro-1-naphthol (4-CN) to produce an insoluble precipitation of benzo-4-chlorohexadienone on the surface of the sensor. When the concentration of Pb(II) in the solution was increased, more of DNAzyme was generated and subsequently more 4-CN molecules were oxidized catalytically, leading to a decrease of the ECL signal. Using this quenching strategy, the ECL signal of the biosensor was linearly dependent on the logarithm of the Pb(II) concentration ranging from 1.0 × 10−15 to 1.0 × 10−11 M with a detection limit of 0.98 f M. The stability and selectivity of the sensor were also investigated. The sensor was applied in the determination of Pb(II) in the real water samples and satisfactory results were obtained.

A flexible electrochemiluminescence (ECL) hydrogel sensor exhibiting good self-healing was constructed. A transparent self-healing oxidized sodium alginate/hydrazide polyethylene glycol (OSA/PEG-DH) hydrogel was prepared by crosslinking dynamic covalent acylhydrazone bond. The introduction of 4-amino-DL-phenylalanine, a catalyst with good biocompatibility, allows rapid gelation and self-healing of hydrogel under mild conditions. Using the hydrogel as the sensing substrate, the ionic liquid (IL) 2-hydroxy- N , N , N -trimethylethanaminium chloride and the luminescent reagent N -(aminobutyl)- N -(ethylisoluminol) (ABEI) were simultaneously immobilized in the OSA/PEG-DH hydrogel to obtain the ABEI/IL/OSA/PEG-DH hydrogel. The ABEI/IL/OSA/PEG-DH hydrogel can be directly used as a semi-solid electrolyte for constructing a flexible ECL hydrogel sensor for the detection of H 2 O 2 , which acted as a coreactant of ABEI. The prepared flexible ECL sensor showed good self-healing performance, can restore ECL signal intensity within 20 min after physical damage, and showed high accuracy in the analysis of complex serum samples. This research shed new light on the development of flexible ECL sensor for bioanalytical applications. Graphical abstract

Stroke, or cerebral apoplexy, is a paroxysmal disease that seriously threatens the neurological functions of middle-aged and older men, resulting in high rates of disability and mortality. The blood level of Corin can accurately predict the risk and subtype of stroke. Herein, an immunosensor was developed utilizing a nanocomposite of Au nanoparticles and Co 3 O 4 nanocubic cages (AuNPs/Co 3 O 4 NcC) as the sensing substrate, with luminol serving as the sensing probe for Corin detection. Through the application of 3-aminopropyltrimethoxysilane, AuNPs/Co 3 O 4 NcC was affixed to the surface of indium tin oxide (ITO)-coated glass, considerably enhancing the electrochemiluminescence (ECL) performance of luminol due to its peroxidase-like activity. To confer biological specificity by immobilizing the Corin antibody on this substrate and to block residual active sites with bovine serum albumin, an immunosensor was developed. The capture of Corin protein by the antibody through an immune reaction, leading to its binding on the sensor, resulted in a decreased ECL signal. Under optimized conditions, the proposed ECL immunosensor demonstrated a broad linear response range from 1 pg/mL to 4800 pg/mL and a low detection limit of 0.26 pg/mL. This ECL immunosensor is characterized by its specificity, stability, and reproducibility and can detect Corin in real blood samples.

It is the first time to develop electrochemiluminescence resonance energy transfer (ECRET) strategy using luminol-doped silica nanoparticles (luminol-SiNPs) for both “turn-off” detection of 2,4,6-trinitrophenol (TNP) and “turn-on” detection of anesthetic drug lidocaine hydrochloride (LDC). The spectral overlap of ECL emission of luminol-SiNPs with the absorption of TNP triggers ECRET from luminol-SiNPs to TNP and results in the decreased ECL signals. Besides, the specific interaction between TNP and LDC consumes TNP, accordingly prevents ECRET from luminol-SiNPs to TNP and achieves increasing ECL signals. The proposed ECRET strategy using luminol-SiNPs exhibits fast response (within 10 min), high sensitivity with LOD of 22.4 nM for TNP and 92.5 nM for LDC, respectively. It is also successfully applied in ECRET detection of TNP in lake water, LDC in human serum and commercial drug sample detections with satisfactory recoveries. More importantly, ECL of luminol-SiNPs is measured via linear sweep voltammetry by electrochemical reduction of dissolved oxygen at negative potential (– 1 V) and in no need of adding hydrogen peroxide (H 2 O 2 ). Reactive oxygen species be generated in situ and make luminol-SiNPs system emit ECL, which makes ECL of the whole luminol-SiNPs system more stable than luminol-H 2 O 2 system.

Cu 2+ serves as a significant biomarker in various physiological processes. This study presents the development of a novel Ru-UiO-TPEA functionalized metal-organic framework (MOFs) probe for Cu 2+ detection, employing both electrochemiluminescence (ECL) and electrochemical (EC) methods. Employing a dual ligand-assisted energy transfer approach, we synthesized Ru-UiO with optimal ECL and EC properties by adjusting the synthesis ratio of biphenyldicarboxylic acid to Ru(dcbpy) 3 2+ . This was followed by its conjugation with N-(2-aminoethyl)-N, N′, N′-tris(pyridin-2-ylmethyl) ethane-1,2-diamine (AE-TPEA) to form the highly conjugated Ru-UiO-TPEA probe for Cu 2+ detection. As the Cu 2+ concentration increased, the probe exhibited a diminished ECL signal and an enhanced differential pulse voltammetry (DPV) signal. The dual-mode detection method enabled cross-validation of the ECL and EC signals, enhancing accuracy and effectively minimizing errors. The ECL and EC methods had detection limits of 0.26 nM (range: 1 nM to 10 μM) and 0.65 nM (range: 2 nM to 80 μM), respectively. Meanwhile, these two methods can effectively detect Cu 2+ in rat brain microdialysis fluid. Notably, the measured concentrations correlated well with those determined by Inductively Coupled Plasma Atomic Emission Spectroscopy (ICP-AES), highlighting its potential in biological and clinical bioelectronic sensing applications.

An ultrasensitive disposable immunosensor based on a gold nanoparticles (AuNPs)/indium tin oxide (ITO) substrate has been constructed to detect ketamine (KET) conveniently. In this study, we obtained a AuNPs/ITO basic electrode by only a two-step ecofriendly drip-coating procedure. The self-assembled monolayers of AuNPs were used as a hetero-bifunctional cross-linker and activator, respectively. Ketamine antibody was used as a biorecognition determinant and immobilized onto the AuNPs/ITO through electrostatic interaction or Au–S or Au–N bonds. The architectures of AuNPs/ITO were characterized by transmission electron microscopy, scanning electron microscopy, cyclic voltammetry, and electrochemical impedance spectroscopy mainly. Electrochemiluminescence (ECL) was used to monitor the specific immune response between the antigen and antibody by measuring changes of intensity in the process of ECL. When the immune complex was incubated on the electrode, it blocked the electron communication and mass transfer, and the ECL signal was quenched. According to the quenched ECL signal, the quantification of KET can be realized. Under optimal conditions, the immunoassay showed a linear range from 10 to 100 pg/g. The low detection limit is 5.73 pg/g. The sensor has been applied to detect ketamine in human hair successfully. In addition, the immunosensor has excellent selectivity, outstanding reproducibility, and acceptable stability. What’s more, the immunosensor exhibits low-cost, simplicity, small sample consumption, and easy miniaturization, which allows it to be integrated into portable platforms to be utilized on the site of crime easily.
