Objective Subarachnoid hemorrhage (SAH) is associated with high mortality and poor outcomes, which are closely related to white matter injury (WMI). (-)-Epigallocatechin-3-gallate (EGCG) exerts neuroprotective effects by inhibiting oxidative stress-related ferroptosis in astrocytes and improving neurological function. However, the role and mechanism of EGCG in regulating reactive astrocytes (RAs) to alleviate WMI after SAH remain unclear. Methods A mouse model of SAH was used to evaluate the effects of EGCG-loaded nanoparticles (EGCG-NPs). Western blot, qPCR, and immunofluorescence were performed for biochemical analysis. Neurological function was assessed using neurological deficit scores and the Morris water maze test. Results Following SAH, HO-1-mediated iron accumulation and ROS production promoted A1/A2 reactive astrocyte polarization, resulting in myelination damage and aggravated WMI. Knockdown of S100A10 inhibited iron-dependent oxidative stress in RAs and attenuated WMI. EGCG-NPs significantly suppressed HO-1/S100A10-mediated iron overload and oxidative stress in both A1 and A2 RAs, thereby alleviating WMI after SAH.Conclusion EGCG-NPs attenuate SAH-induced WMI by inhibiting the iron overload-activated HO-1/S100A10 axis in RAs, representing a promising therapeutic strategy.

Protein–polysaccharide–polyphenol ternary conjugates are promising emulsifiers for improving the oxidative stability of food emulsions. However, the relationship between antioxidant loading and interfacial effectiveness remains unclear. In this study, citrus pectins with different degrees of esterification (DE; 25%, 55%, and 77%) were used to construct whey protein isolate (WPI)–pectin–epigallocatechin gallate (EGCG) conjugates, and their structure–function relationships were systematically investigated. Molecular docking and covalent grafting assays revealed that high-DE pectin (77CP) exhibited the strongest affinity for WPI (−2.12 kcal/mol) via hydrophobic interactions, leading to the highest EGCG loading. Paradoxically, in oil-in-water emulsions, conjugates prepared with low-DE pectin (25CP) demonstrated superior oxidative stability, effectively suppressing the formation of hydroperoxides and thiobarbituric acid-reactive substances during storage. Multiscale structural analysis indicated that although 77CP enabled high antioxidant loading, its dense and thick interfacial layer induced steric masking, limiting the accessibility of EGCG to lipid radicals. In contrast, 25CP formed a thinner and more ordered interface that maximized antioxidant exposure. Furthermore, 25CP provided a synergistic defense mechanism combining potent metal chelation (via abundant carboxyl groups) with strong electrostatic repulsion. These results indicate that the performance of antioxidant emulsifiers depends on antioxidant loading, as well as interfacial architecture and antioxidant accessibility. This study provides a useful framework for designing protein–polysaccharide conjugates for oxidation-resistant emulsion systems.

Poor diabetic wound healing represents a significant threat to public health. Key obstacles include heightened oxidative stress resulting from the hyperglycemic microenvironment and increased susceptibility to bacterial infections. These factors synergistically exacerbate one another, creating a self-perpetuating cycle that hampers healing. Despite advancements in wound care, developing effective strategies to simultaneously mitigate these interconnected issues and disrupt the detrimental loop remains a critical challenge. Herein, we developed a multifunctional hydrogel dressing (PACN@CG) with glucose-depleting, reactive oxygen species (ROS)-scavenging and antibacterial properties, consisting of a double-network hydrogel, copper-based nanoenzyme and glucose oxidase (GOx), forming a combination therapy system for diabetic wound treatment. The integration of covalent and non-covalent bonds within the hydrogel endows it with a range of exceptional properties, including injectability, mechanical robustness, self-healing capability, strong biological adhesion, and biodegradability. The synergistic cascade enzyme system formed by the nanoenzyme and GOx enables self-regulated glucose depletion and ROS scavenging, thereby modulating the diabetic microenvironment while enhancing antibacterial efficacy. The efficacy of the PACN@CG hydrogel in enhancing diabetic wound healing was demonstrated using full-thickness skin wound models in diabetic mice. Consequently, this hydrogel dressing successfully reestablishes tissue redox homeostasis and promotes wound healing, presenting a highly promising approach for the treatment of diabetic wounds.

The intravenous delivery of mesenchymal stromal cells (MSCs) is often limited by pulmonary entrapment and poor survival under oxidative stress in inflammatory environments. To overcome these challenges, a multifunctional cell-surface engineering strategy driven by the synergistic coordination of polyethylene glycol (PEG), epigallocatechin gallate (EGCG), and magnesium (Mg 2+ ) was adopted in this study. PEG and EGCG were first covalently coupled to form the polymer, PEG-EGCG, in which PEG provides “stealth” shielding, and EGCG facilitates mild membrane insertion and Mg 2+ coordination. The resulting PEG-EGCG–Mg complex self-assembled on MSCs, forming a stealth layer and a metal–phenolic network. This P-E-Mg@MSCs design significantly reduced cell adhesion to endothelium and collagen, thereby diminishing pulmonary trapping and leading to increased bone marrow accumulation in a murine model of immune-mediated aplastic anemia (AA). Furthermore, the coating conferred potent antioxidant and anti-inflammatory properties, thereby improving MSC survival under oxidative stress. In AA mice, treatment with P-E-Mg@MSCs restored peripheral blood counts, reduced bone marrow adiposity, and modulated immune imbalance by upregulating regulatory T cells and downregulating cytotoxic CD8 + T cells, thereby outperforming unmodified MSCs. This work presents a versatile coating platform that integrates adhesion inhibition, microenvironment modulation, and metal-ion coordination to enhance the systemic delivery and therapeutic efficacy of MSCs for regenerative and immunomodulatory applications.

Fluoride is an essential trace element, but its beneficial range is narrow, and excess fluoride may have negative health effects. The objective of this study was to investigate the potential cytoprotective effects of epigallocatechin-3-gallate (EGCG) in cultured neuro-2a neuroblastoma cells exposed to sodium fluoride (NaF)-induced oxidative stress. Isolated Neuro-2a cells were exposed to increasing concentrations of NaF (0, 1, 2, 4, 6, and 8 mM) for 24 hours to induce oxidative stress. Moreover, to determine the concentration of EGCG necessary for protective effects, we exposed isolated Neuro-2a cells to increasing concentrations of EGCG (0, 0.5, 1, 5, 10, 20, 40, 60, 80, and 100 μg/mL) for 24 and 48 hours. Pretreatment with EGCG at various doses (0, 0.5, 1, 5, 10, 20, and 40 μg/mL) was evaluated in Neuro-2a cells for 24 hours, followed by an NaF (4 mM per culture well) challenge for 24 hours. As shown in this study, EGCG can protect Neuro-2a cells from NaF-induced apoptosis. This effect may be due to the reactive oxygen species scavenging activity of EGCC.

With recent advances in nanotechnology, debranched starch nanoparticle (DBS-NP) materials have attracted considerable interest from the fields of functional food, biomedicine, and material science, thanks to their small size, biodegradability, biocompatibility, sustainability, and non-hazardous effects on health and the environment. In this study, DBS-NP was fabricated using an eco-friendly method involving ultrasonication combined with recrystallization. The effects of ultrasonication and recrystallization times on the morphology, particle size, and crystal structure of the DBS-NPs were systematically investigated. Compared with the DBS-NPs prepared using ultrasonication treatment only, the DBS-NPs formed using ultrasonication combined with recrystallization were uniform in size and well distributed in aqueous solution. Moreover, the maximum encapsulation efficiency and loading capacity of the epigallocatechin gallate (EGCG) in the DBS-NPs with ultrasonication treatment reached 88.35% and 22.75%, respectively. The particle sizes of the EGCG@DBS-NP were more stable at a neutral pH (7.4) than at an acidic pH (2.1). The EGCG in the EGCG@DBS-NP displayed excellent radical scavenging activity and antibacterial effects, and cell assays demonstrated that the EGCG@DBS-NP was non-toxic and highly biocompatible.

Fresh litchi fruit is highly susceptible to pericarp browning after harvest. The efficacy of combined epigallocatechin-3-gallate (EGCG) and fochlorfenuron (CPPU) treatment attributes in alleviating pericarp browning in litchi fruit were investigated. The results showed that the combined treatments effectively suppressed pericarp browning and mitigated the decline in chromaticity, chlorophyll, and phenolic content. The combined treatment inhibited the accumulation of malondialdehyde and hydrogen peroxide. Following the treatments, enhanced antioxidant activity was observed, as evidenced by increased DPPH radical scavenging capacity and improved reducing power. Enzyme activity assays revealed elevated levels of phenylalanine ammonia lyase (PAL) and catalase (CAT), but decreased activities of polyphenol oxidase (PPO), peroxidase (POD), and laccase (LAC). Furthermore, the expression of key genes related to chlorophyll degradation ( LcCLH2 , LcPPH , LcNYC1 , LcPPH1/2 , LcPAO , LcRCCR ) and enzymatic browning ( LcPPO , LcPOD , and LcLAC ) was effectively regulated. The combined treatment of EGCG and CPPU might provide a practical strategy by preserving chlorophyll and phenolic compounds, and enhancing antioxidant capacity to extend the storage life of green mature litchi varieties.

Spore-forming microorganisms present a significant challenge to food safety and spoilage prevention. This study investigated the effects of dielectric barrier discharge cold plasma (CP) and epigallocatechin gallate (EGCG) on the inactivation of Clostridium perfringens spores and quality changes in salt-baked chicken during a 36-day storage. CP treatment achieved spore and total viable count reductions of 0.01 to 0.90 log CFU g −1 and 0.28 to 1.91 log CFU g −1 , respectively, depending on voltage and time. Spores treated with CP significantly lost their heat resistance under sublethal thermal conditions of 80 °C for 10 min. After 90 kV/600 s CP treatment, a minimum spore heat resistance ratio was attained 0.5% in salt-baked chicken matrix. Pyridine-2,6-dicarboxylic acid release and confocal laser scanning microscopy indicated that the CP-induced heat sensitization was associated with spore germination and structural damage. CP, EGCG, and EGCG + CP treatments extended the microbial shelf life, and suppressed the increase in total volatile basic-nitrogen values of salt-baked chicken during storage. Compared to the CP or EGCG treatments, the combined EGCG + CP application exhibited a better effect in delaying lipid oxidation. These outcomes provide scientific support for the application of CP as alternative non-thermal strategies for controlling C. perfringens spores and maintaining food quality in cooked meat products.

Antimicrobial hydrogels with NIR and enhanced mechanics benefit diabetic wound healing. • Au/MXene modifies gel network intermolecular interactions, regulating hydrogel mechanics. • NIR-triggered hyperthermia promotes wound healing via ROS/EGCG release. Biological hydrogels are ideal wound dressings due to their moisturizing, biocompatible, and drug delivery properties. Polyvinyl alcohol (PVA) and sodium alginate (Alg)-based injectable hydrogels show great potential in wound repair; however, simple hydrogels lack antimicrobial properties and cannot effectively prevent and control infections. Herein, Au@Ti 3 C 2 -E composites were obtained by anchoring gold nanoclusters on the surface of ultrathin Ti 3 C 2 nanosheets (1.5 nm) and modifying them by grafting epi gallocatechin gallate (EGCG). Under 808 nm laser irradiation, Au@Ti 3 C 2 -E@H exhibited good photothermal effects. Interestingly, elevated temperature can act as a “switch” that significantly enhances the Fenton-like catalytic activity of gold nanoclusters (3–5 nm) and promotes the release of EGCG grafted on the surface. The inhibition results showed that Au@Ti 3 C 2 -E@H achieved 94.5% and 93.0% kill rates of Methicillin-Resistant Staphylococcus Aureus (MRSA) and Ampr Escherichia coli ( E.coli ) through higher Reactive Oxygen Species (ROS) generation efficiency and disruption of bacterial biofilms. Molecular biology results showed that Au@Ti 3 C 2 -E@H significantly upregulated Vascular Endothelial Growth Factor (VEGF), Basic Fibroblast Growth Factor (bFGF), Epidermal Growth Factor (EGF) and Platelet Endothelial Cell Adhesion Molecule-1 (CD31) nearly threefold and downregulated Tumor Necrosis Factor-α (TNF-α), Interleukin-6 (IL-6), and Matrix Metalloproteinase-9 (MMP-9) nearly fourfold, which could promote angiogenesis and epithelial repair. After 21 days of the treatment cycle, the diabetic rats achieved a wound healing rate of 99.2%. Au@Ti 3 C 2 -E enhances the mechanical properties and antibacterial effects of hydrogels, synergizing photothermal and nanozyme activities to provide a promising solution for the repair of diabetic wounds infected with drug-resistant bacteria. Bisphenol A (BPA) is a typical endocrine disrupting chemical widely distributed in the environment and in food systems. The adverse effects of BPA on reproductive health have posed major concerns worldwide. The aim of this study was to determine the potential of epigallocatechin gallate (EGCG) to protect against BPA-induced male reproductive toxicity and investigate the underlying protective mechanism. The protective effects of EGCG on BPA-induced reproductive toxicity were investigated using male zebrafish model. The results showed that EGCG alleviated BPA-induced developmental toxicity of F1 generation of zebrafish, reversed testis disorder and combated the blood-testis barrier (BTB) damage caused by BPA in zebrafish. In order to unravel the underlying mechanism, TM4 cell - constructed BTB model was used for further study. The results indicated that EGCG counteracted the damage induced by BPA on the integrity of BTB model and reduced the autophagy caused by BPA. Furthermore, EGCG effectively inhibited the BPA-induced upregulation of the expression level of AMPK (p-AMPKα/AMPKα), significantly restored the BPA-mediated downregulation of the expression levels o。f p-mTOR/mTOR, p-AKT/AKT and Raptor. The results suggested that EGCG mitigated the BPA-induced male reproductive toxicity through maintaining the integrity of BTB by inhibiting the autophagy mediated by AMPK/AKT/mTOR signaling pathway. This study provided a strategy for combating the BPA-induced reproductive toxicity using a well-known bioactive component as a potential therapeutic approach.

Atopic dermatitis (AD) is an inflammatory skin disease characterized by a complex pathogenesis. Current clinical treatments exhibit several limitations, including low efficacy and considerable side effects, prompting a growing interest in novel therapeutic strategies. In this study, we developed a microneedle-assisted dual-nano drug delivery strategy that targets both oxidative stress and Janus kinase 1 (JAK1) expression, aiming for enhanced therapy of AD. Self-assembling nanoparticles (NPs) incorporating epigallocatechin gallate (EGCG) and lactoferrin were developed to effectively modulate oxidative stress. Concurrently, CRISPR-CasRx technology was employed to silence JAK1, and nanoparticles were constructed by encapsulating CasRx and siRNA within poly (β-amino ester) (PBAE). Both types of NPs were successfully delivered to skin lesions in AD mice through microneedles, either individually or combined, resulting in significant alleviation of symptoms. Treatment reduced dermatitis severity scores and splenomegaly, decreased epidermal thickness and mast cell infiltration, and increased collagen fiber content. Additionally, levels of inflammatory cytokines (IL-1β, IL-4, IL-13) and oxidative DNA damage marker (8-OHdG) in dorsal skin tissues were lowered, accompanied by decreased serum levels of thymic stromal lymphopoietin (TSLP) and IgE. The therapy also suppressed JAK1 expression and activated the Nuclear factor erythroid 2-related factor 2 (Nrf2)/Heme oxygenase 1 (HO-1) antioxidant pathway, except for PBAE-plasmid NP-loaded microneedles, which did not significantly increase HO-1 expression. Notably, microneedles containing both NPs exhibited superior efficacy in reducing IL-1β and JAK1 levels and upregulating Nrf2 expression compared to formulations with only one NP type. This innovative combination strategy demonstrates promise as a novel therapeutic approach for AD.

Diabetic wounds are marked by persistent infection, hypoxia, excessive reactive oxygen species (ROS), and impaired angiogenesis, which present significant challenges for clinical management. In this study, a pH/ROS dual-responsive multifunctional hydrogel (QOF-EGCG/Hb@PDA) was constructed by grafting 3-carboxyl-4-ffuorophenylboronic acid (FPBA) onto Quaternary ammonium chitosan (QCS) to form Schiff base bonds with Oxidized gellan gum (OGG) (pH response), and by reacting FPBA with Epigallocatechin gallate (EGCG) to form borate bonds (ROS response). Hb@PDA nanoparticles were then loaded into the hydrogel. Under acidic conditions, the cleavage of Schiff base bonds triggers the release of Hb@PDA, providing sustained oxygen delivery to alleviate hypoxia. Additionally, in ROS-rich environments, the cleavage of borate ester bonds promotes EGCG release, exerting antioxidant and anti-inflammatory effects. QOF-EGCG/Hb@PDA displays excellent self-healing capacity and tissue adhesion, as well as remarkable photothermal activity. Furthermore, it demonstrates excellent antibacterial, antioxidant, anti-inflammatory, pro-angiogenic, and hemostatic properties. In vivo studies confirmed that this hydrogel markedly accelerated diabetic wound healing by enhancing epithelial regeneration, promoting collagen deposition, regulating inflammatory responses, and facilitating angiogenesis. Transcriptomic analysis revealed that the hydrogel promotes angiogenesis by activating the HIF-1α/VEGF signaling pathway, which plays a pivotal role in vascular formation and tissue regeneration, thereby accelerating the healing of diabetic wounds. Collectively, QOF-EGCG/Hb@PDA provides a promising and comprehensive strategy with great potential for application in diabetic wound treatment.
