Ethylene promotes ABA biosynthesis by repressing the expression of miR161 in postharvest strawberry fruit

Renchi Chen,Yingyin Wu,Xiaobo Wei,Zhihai Huang,Linchun Mao

Both ethylene and abscisic acid (ABA) are identified to be crucial phytohormones that positively regulate strawberry fruit ( Fragaria × ananassa Duch.) ripening. In this work, the interplay between ethylene and ABA involved in the ripening of strawberry fruit was explored. Results suggested that ethylene promoted ABA biosynthesis via repressing miR161 expression. Meanwhile, miR161 silencing (STTM161) verified that miR161 negatively regulated ABA biosynthesis via suppressing FaNCED1 expression. In addition, dual-luciferase reporter demonstrated the suppression of miR161 on the promoter of FaNCED1 . These results illuminated a potential molecular mechanism of the collaboration between ethylene and ABA in postharvest strawberry fruit through miR161.

Potato Starch-Based Film Incorporated with Tea Polyphenols and Its Application in Fruit Packaging

Nan Chen,Hao-Xiang Gao,Qiang He,Wei-Cai Zeng

Effects of tea polyphenols (TP) on the physical properties, barrier properties and functionality of potato starch-based film were determined, while the interaction mechanism between TP and starch in film and the application of this film in fruit packaging were further evaluated. TP exhibited different effects on the physical properties of potato starch-based film, including thickness (0.083 to 0.087 mm), moisture content (9.27% to 9.68%), color (ΔEvalue: 5.41 to 10.55), light transmittance (51% to 62%), tensile properties and thermal properties, and improved its barrier properties, including water vapor permeability (9.68 to 11.84 × 10−11g m−1s−1Pa−1),oxygen permeability (1.25 to 2.78 × 10−16g m−1s−1Pa−1) and antioxidant activity. According to the determination of wide-angle X-ray diffraction, Fourier transform infrared and scanning electron microscope, TP could interact with starch chains via hydrogen bonds to form non-crystal complexes, thus affecting the cross-linking among starch chains and further changing the microstructure of film. Furthermore, film incorporated with TP could improve the storage quality (including weight and texture) of blueberries, and inhibit the enzymatic browning of fresh-cut bananas during storage. All present results suggested that tea polyphenols had potential to enhance the properties and function of potato starch-based film, and the film exhibited the application prospect in fruit packaging and preservation. Keywords:tea polyphenols;potato starch;composite film;antioxidant activity;fruit packaging

Effective fractionation and valorization of Camellia oleifera fruit shells under the concept of lignocellulosic biorefinery

Di Meng,Qiulu Chu,Shuiping Ouyang,Jing Wang,Shufang Wu

Camellia oleifera fruit shells (CFS), a by-product of Camellia oleifera oil production industry, are rich in hemicellulose and lignin components. In this work, efficient fractionation and valorization of CFS were investigated. Malic acid (MA) pretreatment (6 g/L MA, 5 g/L 2-naphthol (2 N), 160 ℃, 30 min) resulted in a maximum yield of XOSs of 73.3 %. Besides, alkaline hydrogen peroxide/ethanol (AHP/E) biphasic delignification at mild conditions extracted over 90 % lignin from the MA2N pretreated solid, which was fabricated as lignin nanoparticles (around 170 nm) for drug delivery. Finally, the MA2N-AHP/E combinational pretreatment provided a cellulose-enriched residue, with cellulose retention rate of 93.1 %, which was enzymatically converted to glucose with nearly 100 % hydrolysis efficiency. Results revealed that the combinational pretreatment was an ideal first step of lignocellulosic biorefinery using CFS as feedstock for co-production of XOSs, fermentable sugars and lignin nanocarriers, which potentially alleviated environmental problems caused by discarding, burning and composting of CFS.

Dragon fruit peel pigment extraction residue based intelligent and biodegradable film for indicating milk freshness

Liangting Deng,Qun Liu,Wuyin Weng,Yucang Zhang

pH-responsive intelligent films for food freshness monitoring have attracted a great deal of attention in recent years. In this study, dragon fruit peel was used to develop a new type of active and functional film. The film was prepared by using residues extracted from dragon fruit peel (RPP) as the matrix, Curcumin (Cur) as the active ingredient and ZnO-NPs as the antibacterial material. The effect of the incorporation of Cur and ZnO-NPs on the properties of the films was studied. The addition of Cur increased the oxidation resistance of the films (more than 90 %), and ZnO-NPs increased the bacteriostasis and hydrophobicity of the films (water contact angle (WCA):122.00°). The colorimetry response of the RPP/Cur/ZnO-NPs film was performed, and the color of the film changed from brown to tawny to yellow. These properties highlight the potential of the RPP/Cur/ZnO-NPs film for practical applications, indicating its potential applications as smart packaging materials for the food industry. Therefore, the RPP/Cur/ZnO-NPs composite film based on dragon fruit peel can be used to monitor the freshness of milk, and to a certain extent contribute to the sustainable value of dragon fruit by-products in the food industry.

A validated HPLC method for kallikrein activity assessment and its use in evaluating fruit extracts

Hanying Ren,Lin Xie,Can Zhao,Qiaojia Ling,Shuang Gu,Xiangyang Wang

The conventional ultraviolet-visible (UV–vis) spectrophotometric assay for kallikrein (KLK) activity, which monitors the hydrolysis of N-benzoyl-L-arginine ethyl ester (BAEE) at 253 nm, is prone to significant error due to spectral interference, especially in complex matrices. This study aimed to establish a robust high-performance liquid chromatography (HPLC) method to overcome this limitation. The method achieved complete separation of BAEE and its product, N-benzoyl-L-arginine (BA), with retention times of approximately 15.5 and 5.5 min, respectively. It demonstrated excellent linearity within 1–5 min of reaction time, along with high precision, repeatability and accuracy (mean recovery: 100.9%). The limit of detection (LOD) for BA was 0.25 mg/L. When applied to screen aqueous, ethanolic, and pepsin-hydrolyzed extracts from eighteen fruits, the validated method revealed distinct modulatory effects. Ethanolic extracts tended to promote KLK activity, whereas aqueous extracts were predominantly inhibitory. Notably, pepsin hydrolysates exhibited the most potent and divergent effects, ranging from strong inhibition (e.g., kiwifruit) to marked activation (e.g., mango). These findings confirmed that the established HPLC method was a reliable tool for assessing KLK activity in complex samples and successfully identified specific fruit extracts as sources of novel KLK modulators.

Preparation of PAN@TiO2 Nanofibers for Fruit Packaging Materials with Efficient Photocatalytic Degradation of Ethylene

Zhu Zhu,Ye Zhang,Yibo Zhang,Yanli Shang,Xueji Zhang,Yongqiang Wen

Ethylene causes faster deterioration of perishable crops during postharvest transportation and storage. The present study aimed to develop TiO2-coated nanofibers with efficient photocatalytic activities to enhance the degradation of fruit-emitted ethylene. The consecutive electrospinning of polyacrylonitrile (PAN) and TiO2deposition was successfully performed to produce PAN@TiO2nanofibers. The scanning electron microscopy results indicate the uniform distribution of TiO2nanoparticles on the surface of the PAN nanofiber. The PAN@TiO2composite nanofibers exhibited enhanced photocatalytic activity for ethylene degradation under low-intensity UV light irradiation. Furthermore, a tomato fruit-ripening test confirmed the effectiveness of the PAN@TiO2nanofibers. The PAN@TiO2nanofibers exhibited effective ethylene degradation and slowed the color shift and softening of the tomatoes during storage. The results suggest great potential for use of the PAN@TiO2composite nanofibers as ethylene scavenging packaging material for fresh fruits and vegetables.

The Medicine-Food Homology Fruit Gouqi (Lycium barbarum L.) Mitigates Kidney Stone via Multiple Bioactivities

Wen Zhang,Yunuo Ren,Xiaoxi Liu,Xingmiao Liu,Weiwei Tang,Yingjie Ju,Wei Yu,Zhimou Guo,Huanhuan Qiao,Xiaoli Yu,Suxia Ren,Tianxiang Li,Yuqing Dong,Zengyi Huang,Youxin Li,Zhiguang Yuchi,Dong Li,Yiwen Wang

Background/ObjectivesLycium barbarum L. (Gouqi, GQ) is a prominent component of traditional Chinese medicine with well-established medicine-food homology properties. Owing to its high nutritional profile, GQ is widely recognized for its diverse pharmacological benefits, including protective effects on the kidney. The primary objective of this study was to evaluate the anti-urolithiatic activity of GQ aqueous extract and to elucidate its underlying therapeutic mechanisms in the context of calcium oxalate (CaOx) nephrolithiasis (Kidney stone).MethodsThe anti-kidney stone activity of the GQ extract was assessed in a Drosophila melanogaster (fruit fly) model of CaOx kidney stones induced by dietary sodium oxalate (NaOx). Oxidative stress levels were quantified via dihydroethidium (DHE) staining. The expression profiles of key enzymes involved in endogenous oxalate synthesis—namely, agxt (the ortholog of human alanine-glyoxylate transaminase, AGT) and daao1/daao2 (encoding two D-amino acid oxidases, DAOs)—were analyzed using RT-qPCR technology. Excretory function was evaluated by counting fly fecal deposits. In vitro crystallization assays were performed to examine evolution of CaOx crystal morphology and explore the regulatory activity of the GQ extract during crystal formation.ResultsOur results demonstrated that the GQ extract effectively attenuated CaOx kidney stone formation through multiple interconnected mechanisms. Specifically, GQ extract mitigated NaOx-induced oxidative stress, improved urinary excretion, reduced endogenous oxalate biosynthesis, and directly altered the morphological dynamics of CaOx crystallization.ConclusionsThese findings substantiate the significant stone-preventive potential of GQ, whose therapeutic effects are mediated via a pleiotropic mode of action. Collectively, our data suggest that GQ may serve as a valuable dietary supplement or potential therapeutic agent for the prevention and management of CaOx kidney stones.

Multifunctional Chitosan Composite Films Reinforced With Rhodiola rosea Extract/ZnO/Palygorskite Nanocomposites for Fruit Preservation

Wenting Gao,Bin Mu,Fangfang Yang,Yalong Li,Yu Tang,Zhihe Luo,Aiqin Wang

The potential of chitosan (CS) films for the preservation has been explored, and it is recognized as one of the promising approaches to enhancing the comprehensive capability of CS films for food control after incorporation of functional nanofillers. Herein, Rhodiola rosea extract/ZnO/palygorskite (RRE/ZnO/Pal) nanocomposites were facilely prepared and incorporated into CS matrix to obtain RRE/ZnO/Pal/CS (RZPC) composite films, in which ZnO/Pal nanocomposites were synthesized by a green phyto-mediated method followed by loading of R. rosea extract via a semi-dry grinding process to form organic/inorganic bioactive nanocomposites. The composite films exhibited significantly enhanced antioxidant, antibacterial, mechanical and UV-shielding properties compared with the pure CS film, as well as the maximum scavenging activities on hydroxyl radicals and 2,2-diphenyl-1-picrylhydrazine radicals of composite films were 92.85% ± 1.59% and 56.89% ± 0.56%, respectively. The antibacterial ratios of composite films containing 6.25% of RRE/ZnO/Pal nanocomposites were 94.92% ± 2.67% and 95.48% ± 2.08% for E. coli and S. aureus , respectively. In addition, the RZPC films effectively delayed browning and deterioration of bananas. This study provides a green method for construction of bioactive nanocomposites and new strategy to design multifunctional CS films for food preservation based on the synergistic effect of organic and inorganic components.

Developing high-strength, multifunctional films from tamarind seed polysaccharides and propolis for sustainable fruit preservation

Feifan Yang,Jiayu Zhang,Ruiying Ji,Lebing Wang,Yuetian Yang,Binxin Cai,Juan Sun,Yang Zhou,Jiantao Zhang

Fiber-reinforced tamarind seed polysaccharide-starch film has high tensile strength. • Propolis enables UV-barrier, antioxidant and antibacterial functions (>97%). • Extends fruit shelf life, exhibits soil biodegradability and clean combustion. Developing sustainable food packaging that integrates structural integrity with active functionality remains a central challenge for next-generation biopolymer materials. This study presents a hierarchical design strategy to fabricate a high-performance tamarind seed polysaccharide (TSP) based composite film, wherein natural seed fibers provide macroscopic reinforcement and propolis extract (PE) serves as a molecular-scale active modulator. Despite the inclusion of starch, the fiber-containing tamarind gum/pre-gelatinized starch/propolis extract composite films (TPP-0%) maintained a high tensile strength of 27.51 ± 0.97 MPa, comparable to that of pure TSP film (27.31 ± 1.19 MPa). Multiscale structural analysis reveals that PE induces a densified amorphous matrix and a dynamically reconfigured hydrogen-bond network, which collectively balance mechanical robustness and flexibility. The films demonstrate superior barrier properties against water vapor and UV radiation, alongside potent antioxidant and broad-spectrum antibacterial activity (>97% against E. coli and S. aureus ). Practical preservation tests confirm their efficacy in extending the shelf life of fruits by mitigating oxidative browning and microbial proliferation. Coupled with favorable soil biodegradability and clean combustion behavior, this work establishes a scalable and eco-friendly paradigm for structurally integrated, multifunctional food packaging.

The Phytochemical Characterization of a Cili (Rosa roxburghii) Fruit Low-Temperature Extract with Hepatoprotective Effects

Rifeng He,Ziling Lian,Zhongjun Cheng,Yang Liu,Xiaoyan Peng,Yong Wang,Hang Ma,Xue Zhou,Fahuan Ge

Cili (Rosa roxburghiiTratt) fruit is a nutrient-rich edible plant known for its antioxidant and hepatoprotective properties. However, conventional extraction methods often lead to the degradation of its bioactive compounds. In this study, we developed a low-temperature homogenate-assisted high-pressure disruption extraction (HHPD) method to obtain a phytochemically enriched cili fruit extract (HHPD-CFE). The chemical characterization of the HHPD-CFE showed that it contained higher levels of polyphenols, polysaccharides, and superoxide dismutase (SOD) than those in conventional squeeze extraction. The hepatoprotective effects of the HHPD-CFE were evaluated in oxidative stress-induced liver injury and hepatic fibrosis models. The HHPD-CFE mitigated oxidative damage by reducing malondialdehyde while enhancing SOD and glutathione activity. Additionally, the HHPD-CFE inhibited the activation of hepatic stellate cells (HSC-T6) and reduced collagen deposition, suggesting a protective role against liver fibrosis. These findings support that the HHPD-CFE is a promising botanical extract with enriched bioactive compounds and liver-protective properties. This study supports the potential application of optimized extraction techniques to preserve thermosensitive compounds and improve the efficacy of functional foods for liver health.

Ethnopharmacological and scientific validation of Caryota mitis Lour fruit peel for osteoarthritis and gout management

Pham Thi Nhat Trinh,Tong Thanh Danh,Do Chiem Tai,Phung Van Trung,Luu Hong Truong,Dang Thi Le Hang,Truong Minh Dung,Le Tien Dung

Ethnopharmacological relevance Traditional medicine in Vietnam has long utilized Caryota mitis Lour fruit peel to relieve joint pain, back pain, and inflammation. Despite its widespread use, scientific validation of its therapeutic effects remains limited. The application of the fruit peel in disease treatment is largely based on anecdotal evidence, highlighting the need for rigorous scientific research to substantiate its efficacy and elucidate its mechanisms of action. Aim of the study This study aimed to investigate the immunomodulatory effects, including anti-xanthine oxidase activity, reactive oxygen species (ROS) inhibition, and anti-inflammatory properties, alongside cartilage-protective effects such as in vitro chondrocyte proliferation, chondrogenic activity, and in vivo osteoarthritis treatment. Furthermore, phytochemical profiling was conducted to identify bioactive compounds in the fruit peel extract. Methods Active compounds were extracted using various chromatographic techniques. Cytotoxicity and chondrogenic assays were performed on human chondrocytes to assess cellular responses. A model of LPS-induced RAW264.7 macrophages was used to evaluate nitric oxide (NO), interleukin-6 (IL-6), and interleukin-10 (IL-10) secretion, as well as ROS inhibition. Xanthine oxidase inhibition assays were conducted to assess the impact on uric acid production. In vivo efficacy was examined using a CFA-induced osteoarthritis mouse model, with histological analysis and biomarker quantification to assess cartilage regeneration and anti-inflammatory effects. The phytochemical profile was analysed using LC-MS/MS and molecular networking, while the chemical structures of isolated compounds were confirmed through spectral analysis. Results All extracts—including crude ethanol (CME), hexane (CMH), chloroform (CMC), ethyl acetate (CMEA), and methanol (CMM)—significantly enhanced chondrocyte proliferation at concentrations below 1 μg/mL. At 0.1 μg/mL, CME, CMC, and CMEA strongly promoted cartilage formation. Additionally, CME, CMH, and CMEA exhibited potent immunomodulatory effects by reducing intracellular ROS, inhibiting xanthine oxidase activity, and suppressing inflammatory mediators such as NO and IL-6. In vivo, osteoarthritis-induced mice treated with CME exhibited significant cartilage regeneration, as evidenced by Safranin O staining and histopathological evaluation. The extract reduced ankle joint swelling, suppressed inflammatory cytokines, and improved cartilage integrity, demonstrating comparable efficacy to diclofenac treatment. Phytochemical analysis identified 18 bioactive compounds, primarily flavonoids and anthraquinones, in the fruit peel. Among them, emodin and esculetin, isolated from the CMEA fraction, demonstrated significant anti-inflammatory and xanthine oxidase inhibitory activities. Conclusion This study provides the first pharmacological validation of Caryota mitis fruit peel in joint disease management. The extracts exhibited immunomodulatory effects, cartilage-protective properties, and xanthine oxidase inhibition, supporting their potential role as a natural alternative to NSAIDs for osteoarthritis and gout. These findings underscore the need for further clinical research to facilitate the integration of Caryota mitis into modern phytotherapy.
