Direct Gating of the TRPM2 Channel by cADPR via Specific Interactions with the ADPR Binding Pocket

Peilin Yu,Zhenming Liu,Xiafei Yu,Peiwu Ye,Huan Liu,Xiwen Xue,Lixin Yang,Zhongtang Li,Yang Wu,Cheng Fang,Yong Juan Zhao,Fan Yang,Jian Hong Luo,Lin-Hua Jiang,Liangren Zhang,Lihe Zhang,Wei Yang

Summary cADPR is a well-recognized signaling molecule by modulating the RyRs, but considerable debate exists regarding whether cADPR can bind to and gate the TRPM2 channel, which mediates oxidative stress signaling in diverse physiological and pathological processes. Here, we show that purified cADPR evoked TRPM2 channel currents in both whole-cell and cell-free single-channel recordings and specific binding of cADPR to the purified NUDT9-H domain of TRPM2 by surface plasmon resonance. Furthermore, by combining computational modeling with electrophysiological recordings, we show that the TRPM2 channels carrying point mutations at H1346, T1347, L1379, S1391, E1409, and L1484 possess distinct sensitivity profiles for ADPR and cADPR. These results clearly indicate cADPR is a bona fide activator at the TRPM2 channel and clearly delineate the structural basis for cADPR binding, which not only lead to a better understanding in the gating mechanism of TRPM2 channel but also shed light on a cADPR-induced RyRs-independent Ca 2+ signaling mechanism.

A Comparison of Phenolic Monomers Produced from Different Types of Lignin by Phosphotungstic Acid Catalysts

Boyu Du,Bingyang Liu,Xing Wang,Jinghui Zhou

Graphical Lignin explained : Phosphotungstic acid polyoxometalate catalyst showed high activity in the selective depolymerization of three types of lignin samples to phenolic monomers, and the impact of the structure differences on the distribution of phenolic monomers were well studied. Emphasis was placed on changing the types of lignin samples to control the secondary depolymerization of methoxy groups and greatly improving the selectivity of phenolic monomers. Herein we studied the chemical structure of different types of lignin samples and the potential to prepare phenolic monomers was illustrated by phosphotungstic acid catalysts. Different types of H/G/S lignin components had different structures. The lignin extracted from poplar had the highest molecular weight and β-O-4 aryl ether contents, followed by pine and straw lignin samples. After depolymerization by PTA catalyst, the yields of phenolic monomers detected was 8.06 wt % (poplar), 5.44 wt % (pine) and 4.52 wt % (straw), respectively. Further, the ratios of H/G/S in the phenol monomers were also different, indicating that the S, G and H types structural units were continuously transformed with each other during the reaction. In our study, the change in the types of lignin samples resulted into an improvement of the distribution of phenolic products, and also the selectivity of phenolic monomers significantly.

Exploring the Size Effect of Pt Nanoparticles on the Photocatalytic Nonoxidative Coupling of Methane

Jiayu Ma,Xianjun Tan,Qingqing Zhang,Yan Wang,Jinlong Zhang,Lingzhi Wang

The high inertness of the C–H bond makes the photocatalytic methane conversion a significant challenge. The platinum nanoparticle is a promising cocatalyst for CH4 activation, while the study of its structure characteristics and functionality remains in its infancy. Herein, the size effect of Pt on the photocatalytic nonoxidative methane conversion efficiency was systematically investigated over x-Pt/Ga2O3 with the particle size (x) ranging from 1.5 to 2.7 nm, where a volcano-shaped relation was observed. The smaller size is beneficial to the formation of Ptδ+ species, which is mainly distributed on the terrace sites according to the DFT calculation. The corner Pt atom is the geometric active site for the CH4 polarization, and the terrace Ptδ+ helps promote C–H activation since the activity is decreased on reduced x-Pt/Ga2O3 with a lower Ptδ+ content. Meanwhile, Ptδ+ species favors the oxidation of adsorbed −CH3 group to ·CH3. The volcano-shaped size effect on the NOCM activity was finally rationalized by the balance between C–H activation and C2H6 desorption from the corner sites on different sized Pt.

Efficient and controllable alcoholysis of Kraft lignin catalyzed by porous zeolite-supported nickel-copper catalyst

Liping Kong,Chunze Liu,Ji Gao,Yuanyuan Wang,Liyi Dai

The alcoholysis of Kraft lignin was catalyzed by bimetallic Ni-Cu supported on H-Beta, HZSM-5, MAS-7, MCM-41 and SAPO-11 zeolite materials in isopropanol solvent. Results showed that a higher bio-oil yield of 98.80 wt% and monomer yield of 50.83 wt% without obvious char were achieved at 330 °C for 3 h over Ni-Cu/H-Beta catalyst. Isopropanol was found to be more effective in H 2 generation and facilitated to the hydrodeoxygenation of lignin-derived phenolic compounds. Moreover, the composition of liquid products was also influenced by the acidity and pore structure of catalyst. The superior cycloalkanes yield was produced over Ni-Cu/H-Beta with larger pore size and more acidity. In contrast, a large number of cyclic ketones/alcohols and alkanes were obtained over other zeolites supported catalysts with smaller pore size and less acid content. Besides, the temperature, time and solvent effect on the lignin depolymerization were also researched.

Graphical Ready layer one : A newly synthesized catalyst with layered cobalt oxide on the surface of titania (layer-CoO x /TiO 2 ) is highly selective towards the aerobic oxidation of C−H bonds in a series of hydrocarbons. In-depth structural and functional characterization reveal that the layer-CoO x /TiO 2 readily reacts with O 2 for the adsorption and activation of C−H bonds. Construction of efficient interfaces to improve the performance of supported metal catalysts is a challenging but effective technique. A newly synthesized catalyst with layered cobalt oxide on the surface of titania (layer-CoO x /TiO 2 ) is highly selective towards the aerobic oxidation of C−H bonds in a series of hydrocarbons under sustainable conditions. The layer-CoO x /TiO 2 easily outperforms the state-of-the-art noble metal catalysts and homogeneous cobalt salts used in industry. In-depth structural and functional characterization reveal that the layer-CoO x /TiO 2 readily reacts with O 2 for the adsorption and activation of C−H bonds. The layered structure of CoO x can maximize the interfacial effect of CoO x /TiO 2 leading to a good performance for the oxidation of C−H bonds.

Unlocking a self-catalytic cycle in a copper-catalyzed aerobic oxidative coupling/cyclization reaction

Jianming Liu,Xiaopei Wang,Zhiyue Wang,Yan Yang,Qinghu Tang,Hongchi Liu,Hanmin Huang

Summary Presented here is a copper-catalyzed, aerobic oxidative C-H/C-H cyclization reaction, which occurs by cleaving the C-H and N-H bonds of 3-phenylindoles. A broad range of 3-phenylindoles can be well tolerated to produce the indole-containing polycyclic aromatic hydrocarbons (PAH) in good to excellent yields. An evaluation of the reaction mechanism is enabled by the isolation of the di- and tri-indole intermediates, highlighting the role of the substrate for this catalytic reaction. The results of these controlled experiments and kinetic studies provide solid experimental support for a self-catalysis reaction, which has rarely been observed in oxidative C-H activation reactions. Additional mechanistic studies indicate that the substrate for this reaction accelerates by the following mechanism: The substrate combines with the Cu catalyst to transform the less active di-indole intermediate into a tri-indole intermediate. This intermediate is quickly converted into the desired product along with regeneration of the substrate copper complex.

Mitigating Amine Oxidative Degradation via H-Abstraction Control in Structurally Robust Composite Amines

Zhengang Zhou,Kai Feng,Xiuqi Hu,Chengjin Pan,Junjie Zheng,Lingyu Shao,Funing Wang,Xiao Zhang,Yang Zhang,Xiaojiang Li,Shihan Zhang,Chenghang Zheng,Xiang Gao

Amine-solvent degradation stands as a key bottleneck hindering the large-scale application of post-combustion CO2 capture, motivating the development of more chemically stable solvent chemistries. High-resolution mass spectrometry and density functional theory were combined to identify degradation pathways and quantify hydrogen-abstraction barriers. Sterically hindered and cyclic amines suppress hydrogen abstraction and downstream oxidation. Solvent degradation was evaluated in high-pressure autoclave reactors under simulated industrial flue-gas conditions representative of coal-fired power plants, with CO2 concentrations of about 10−15 vol %, O2 at several volume percent, and temperatures relevant to absorber−stripper operation and solvent circulation. Compared with conventional monoethanolamine (MEA), 2-amino-2-methyl-1-propanol (AMP) reduced degradation rates by 25−60%, attributable to steric hindrance that suppresses carbon-chain hydrogen abstraction. Under all tested operating conditions, a mixed solvent system comprising N-aminoethylpiperazine (AEP) and AMP demonstrated markedly stronger antidegradation capability than MEA. Antioxidants mitigated oxidative degradation, whereas Fe2+ accelerated it; chelating agents suppressed Fe2+-catalyzed oxidation by limiting peroxyradical formation. Structure−stability relationships and targeted additive design guided multifactor optimization of the absorbent formulation, improving stability by 71.6% under accelerated degradation tests and supporting long-term, oxygen-tolerant operation.

Flying Droplet Sensor Based on H-GQDs/CNF Network Structure and Its Applications

Lu Jiaqi,Chen Xiangdong,Ding Xing,Liu Yanchang,Zhong Jialun,Liu Fang,Li Han,Li Guoping

Respiratory infectious diseases continue to pose a serious threat to public health. Their transmission relies on respiratory droplets that are small in size and evaporate rapidly, making real-time monitoring difficult. In this study, we propose a flying droplet sensor based on hydroxylated graphene quantum dots (H-GQDs) deposited on a cellulose nanofiber (CNF) film. Owing to the network water-locking structure of the film, the sensor is capable of detecting coughing droplets transmitting over long distances (≥2.4 m). By placing multiple such sensors at equal intervals along the cough airflow direction, the spatiotemporal transmission features of coughing droplets were obtained, including arrival time and signal intensity at different distances. On this basis, we investigated the transmission behavior of cough droplets from multiple individuals at various distances. The results show that regions close to the volunteer’s mouth exhibit a broader droplet size distribution, whereas at longer distances, the remaining droplets are mainly composed of small-size and low-intensity components. We further found that coughing droplets typically take 4−5 s to travel more than 2 m, indicating that a time window for timely protection still exists at this distance. In addition, the relationships between cough sound level and droplet signal intensity as well as transmission time were evaluated. No clear correlation was observed, suggesting that acoustic indicators alone cannot directly reflect the infection risk associated with cough droplet transmission. This study provides important insights for the real-time monitoring and protection against respiratory droplet transmission.

Targeting MCT1 suppresses humoral response and autoimmunity via the NAD(H) redox state

Shu-Li Wei,Ying Yang,Cong-Cong Wang,Xue-Min Li,Yuan-Wei Li,Xiao-Li Li,Yang Zhou,Tong Du,Ru-Tao Liu,Ying Liu,Bing Yang,Rui-Sheng Duan,Peng Zhang,Chun-Lin Yang

MCT1 is highly expressed by activated and germinal center B cells. • Targeting MCT1 suppressed humoral response and autoimmunity. • Targeting MCT1 inhibited B cell survival by modulating NAD(H) redox state. Immunometabolism has garnered significant attention due to its essential roles in regulating B cell activation, proliferation and differentiation. Monocarboxylate transporters (MCTs), a family of solute carrier transporters responsible for lactate transport across biological membranes, have been shown to play a pivotal role in immune responses across various immune cell types. However, the expression and functional significance of MCTs in B cells remain poorly understood. In this study, we demonstrated that germinal center B cells and activated B cells exhibit elevated levels of MCT1 expression. Pharmacologically blocking MCT1 with AZD3965 inhibited both T-cell dependent and independent humoral response and ameliorated autoimmunity in the experimental autoimmune myasthenia gravis. Mechanically, MCT1 blockade led to the accumulation of intracellular lactate, which in turn suppressed NAD + regeneration, thereby inhibiting B cell activation and survival. Taken together, these findings suggest that MCT1 might be a potential therapeutical target in antibody-mediate autoimmune disease. Contact-electro-catalyzed direct hydroxylation of aromatic C–H bonds under ambient conditions

Zhao Jiawei,Xu Mengqian,Wang Yanjie,Liang Zihan,Ye Haobin,Luo Jin,Zhao Qianwei,Yang Li,Deng Wei-Qiao,Fan Feng Ru

The selective activation of inert aromatic C–H bonds for the synthesis of high-value phenols is of considerable scientific and industrial significance. Conventional free radical-based hydroxylation methods typically require ultraviolet irradiation or large amounts of sacrificial agents, which significantly limit their applicability. Here, we present a contact-electro-catalysis (CEC) approach that enables aromatic hydroxylation through hydroxyl radicals (·OH) and strong interfacial electric fields generated at the polytetrafluoroethylene (PTFE)–H 2 O interface via contact electrification. Using benzene as a model substrate, phenol was obtained via ·OH-mediated oxidation, and a variety of substrates with diverse electronic properties underwent efficient hydroxylation. For example, the transformation of 4-hydroxybenzoic acid yielded hydroquinone and 3,4-dihydroxybenzoic acid with maximum yields of 72.45 and 210.90 μmol·g cat −1 h −1 , respectively. Electron paramagnetic resonance (EPR) confirmed the presence of ·OH, whereas nuclear magnetic resonance (NMR) and liquid chromatography-mass spectrometry (LC-MS) analyses identified the products, thereby supporting mechanistic investigations. This work extends the application of contact-electro-catalysis to organic synthesis, providing a sustainable alternative despite existing challenges in selectivity and scalability.

Steering acidic CO2 electroreduction to ethanol via interfacial H-bond-mediated proton transport

Jiapeng Ji,Zhibo Zhang,Zhixian Mao,Huajie Yin,Hui-Ming Cheng,Xiaolong Zhang

Novel Cu/CNTs-OH interface decouples gas/ion transport for acidic CO 2 conversion. • Interfacial H-bond network via -OH groups enhances proton mobility. • Shifts selectivity from ethylene to ethanol by optimizing *H coverage and proton supply. • In situ and theoretical evidences link H-bond network to key intermediates in ethanol pathway. Electrocatalytic CO 2 reduction (ECR) offers a route to convert CO 2 into valuable fuels such as ethanol, which is favored for its high energy density and ease of storage. ECR in acidic media could suppress carbonate formation but intensify competition from the hydrogen evolution reaction, limiting the selectivity of C 2+ products. Here we introduce an interfacial H-bond network strategy by coupling Cu nanoparticles with hydroxylated carbon nanotubes (CNTs-OH) to construct proton supply and steer reaction pathways for acidic CO 2 -to-ethanol electroconversion. Molecular dynamics and distribution of relaxation times analysis show that CNTs-OH enhance CO 2 diffusion while surface -OH groups form a dynamic H-bond network, facilitating proton transport via the Grotthuss mechanism. Density functional theory (DFT) calculations, isotope labeling, and rotating disk electrode results confirm that CNTs-OH promote water dissociation, supplying protons to Cu sites. In situ spectroscopy links the H-bond network to key ethanol pathway intermediates. Consequently, the introduced H-bond network at the electrode interface yields a 2.5-fold enhancement in ethanol selectivity and a dramatic shift in the ethanol/ethylene ratio from 0.4 to 1.6. This work provides an effective materials design strategy and mechanistic insight for selective CO 2 -to-ethanol conversion in acid media.

Tuning the H-bond donicity boosts carboxylic acid efficiency in ring-opening polymerization

Songquan Xu,Hui Zhu,Zhenjiang Li,Fulan Wei,Yu Gao,Jiaxi Xu,Haixin Wang,Jingjing Liu,Tianfo Guo,Kai Guo

H-bond donor (HBD) and base co-catalysis achieved great success in ring-opening polymerization (ROP). Parallel H-bond donor and Brønsted acid (HBD–BA) cocatalysis has been scarcely explored in ROPs. In developing the HBD–BA catalysis in to versatile toolbox for ring-opening polymerizations of wider scope and better control, tunable HBDs including boronate pinacol ester urea (BU), urea (U), and thiourea (TU) were evaluated in cooperation with trifluoroacetic acid (TFA) in HBD–BA catalytic mode. Each of the HBD–TFA co-catalyst promoted ROPs of δ-valerolactone (VL) and ε-caprolactone (CL) successfully. The catalytic performances of the HBD–TFA on the HBD part was in an order of BU > U > TU to their corresponding ROPs of VL and CL. The turnover frequencies of the catalyzed ROPs of VL and CL by BU–TFA was twice as that of TFA. The ROPs afforded homopolymers with predicted molecular weights (PVL, M n,NMR  = 2.51–9.92 kg mol −1 ; PCL, M n,NMR  = 2.51–11.1 kg mol −1 ) and narrow dispersities (PVL, Đ ≤ 1.16; PCL, Đ ≤ 1.18) by ratios of monomer to initiator from 20 to 100. The obtained polymers were analyzed by 1 H NMR, 13 C NMR, and MALDI-ToF MS, the results indicated the high chain-end fidelity. Chain extension experiments were conducted by a first ROP of VL/CL and second ROP of CL/VL with precise molecular weights (from 2.98 kg mol −1 to 6.15 kg mol −1 ) and narrow dispersities below 1.18. The kinetics plots showed a distinct first order relationship between ln([VL] 0 /[VL]) versus the reaction time, it indicated the controlled nature of the catalytic ROP. NMR titrations supported the proposed mechanism that HBD associated with TFA and promoted the cationic polymerization.
