Multimodal Actuation and Precise Control in Liquid Crystal Elastomer Optical Fiber Artificial Muscles

Haojun Liu,Yongcheng He,Jiajia Luo,Nuo Li,Tanxi Wang,Enhai Song,Qianyi Guo,Zhijun Ma,Zhongqiang Yang,Jiulin Gan,Zhongmin Yang

Artificial muscles mimicking the fibrous structure and functionalities of natural skeletal muscles have garnered substantial interest for applications in actuators, soft robotics, and biomedical devices. However, achieving multidirectional actuation and delicate manipulation in confined environments remains challenging. Inspired by the neuromuscular system, a novel liquid crystal elastomer optical fiber (LCEOF) is introduced as artificial muscle with multimodal actuation and precise control. Fabricated through a two-step process, the LCEOFs possess sufficient orientation order (0.65) and low optical transmission loss (0.37 dB cm −1 ), enabling over 40% contraction strain with minimal ambient interference. Bundling multiple LCEOFs yields artificial arms capable of complicated and controllable deformations, including long-distance contraction (≥5 cm), weightlifting (>4000 times their own weight), wide-range torsion (0–180°), and omnidirectional bending (0–360°). Multimodal actuation is precisely and independently regulated via terminal-coupled laser inputs for each LCEOF in bundled arrays, enabling coordinated, crosstalk-free motions. These optical fiber artificial muscles allow precise and controllable operation of an artificial hand for grasping and manipulating objects, without reliance on free-space lateral illumination. Additionally, robotics systems incorporating bundles of LCEOFs have been designed and demonstrated for tasks such as laser writing and object transfer in confined environments, thereby offering new possibilities for the advancement of smart actuators.

Spatially heterogeneous microgel-annealed PRP-derived fibrin hydrogel promotes coordinated vasculogenesis and angiogenesis after myocardial infarction

Yini Huangfu,Yiqun Zhang,Yufeng Zhang,Shuwei Wang,Sheng Ding,Rui Gao,Hang Li,Wenhao Ju,Chuangnian Zhang,Pingsheng Huang,Weiwei Wang,Anjie Dong,Deling Kong,Zujian Feng

Blood vessel reconstruction is key for ischemic disease treatment by restoring microvascular perfusion and mitigating pathological tissue stiffening. During neovascularization, the mechanical and biochemical cues presented by the cellular-scale spatial heterogeneity of extracellular matrix (ECM) facilitate the endothelial cells (ECs) spreading and mechanotransduction, thereby driving angiogenesis. Therefore, developing ECM-mimetic proangiogenic biomaterials with compartmentalized spatial heterogeneity is highly desirable but challenging. Here, inspired by the spatially heterogeneous mechanical properties of natural tissues, we designed a series of microgel-annealed hydrogels (GMP) with compartmentalized stress heterogeneity. By integrating microfluidic-synthesized rigid microgels, the soft PRP-derived fibrin matrix was annealed with spatially mechanical domains that direct ECs-guided vascular morphogenesis through mechanotransduction, where rigid microdomains provide anchorage sites for cells adhesion, while soft interstitial matrix permits stalk cells migration and morphogenesis. In vitro, heterogeneous GMP hydrogel augmented ECs mechanotransduction via activating integrin β1-p-FAK-p-MLC signaling pathway and stabilizing VE-CAD/β-cat junctions, promoting 3D vasculogenesis and angiogenesis. In a rat myocardial infarction (MI) model, treatment with the heterogeneous hydrogel enhanced myocardial neovascularization, attenuated ventricular dilation and enhanced cardiac function. These findings not only provide valuable guidance for engineering proangiogenic biomaterials via ECM biomechanical mimicry, but also highlight the promise of spatially heterogeneous hydrogel with engineered mechanical cues essential for de novo blood vessel formation in the treatment of ischemic disease. Statement of significance This work develops a proangiogenic hydrogel that mimics the spatially heterogeneous mechanical properties of the native extracellular matrix (ECM). By annealing rigid microgels into a soft fibrin matrix, we create a biomaterial with compartmentalized stress domains that guide endothelial cell behavior. This engineered heterogeneity promotes 3D vascular network formation by enhancing key mechanotransduction pathways and stabilizing cell-cell junctions. Significantly, in a rat model of myocardial infarction, the hydrogel enhances neovascularization, attenuates tissue damage, and improves cardiac function. Our findings highlight that recapitulating ECM biomechanical heterogeneity is a transformative strategy for engineering biomaterials to drive functional blood vessel regeneration, offering a promising therapeutic approach for treating ischemic diseases.

ROS-Responsive Antioxidant Hydrogel with Immunomodulatory Activity for Promoting Diabetic Wound Healing

Yurao Liu,Qianqian Yang,Ling Zhang,Qiyue Lu,Yuhang Li,Haisong Zhang,Chunfang Zhang,Pingli Wu

The accumulation of reactive oxygen species (ROS) in the microenvironment of diabetic wounds can trigger oxidative stress and hinder wound healing. This study developed a novel ROS-responsive antioxidant hydrogel relying on boronated ester bond. GMOP/TA hydrogel is constructed using methacrylic acid modified gelatin (GelMA) and phenylboronic acid modified oxidized hyaluronic acid (OHA-PBA) as the crosslinking framework, with tannic acid (TA) loaded as the active antioxidant component. GelMA and OHA-PBA crosslink through covalent bond and Schiff base, and the reversible properties of their imine and boronate ester groups enables responsive TA release under high ROS conditions. GMOP/TA hydrogel exhibits suitale mechanical properties, excellent biocompatibity and ROS-responsive antioxidant capabilities. In vivo evaluation results show that this hydrogel can effectively alleviates oxidative stress, accelerating cell migration, proliferation, and angiogenesis. Transcriptome sequencing analysis further revealed that its pro-healing effects is mechanistically associated with key signaling pathways, especially through PI3K-AKT, PPAR signaling pathway to facilitate cell proliferation and suppress inflammation. Overall, the hydrogel provides a straightforward but effective platform for chronic diabetic wound healing.

Engineered 3D Printing Bilayer Scaffold with Spatiotemporal Release of Drug for Synergistic Osteochondral Regeneration

Xueling Yin,Wanting Xia,Meng Zhang,Xiaoyu Yang,Wei Yu,Huimin Fan,Xiaona Wu,Ruikai Zhu,Ye Ren,Zhihong Zhu

Osteoarthritis (OA) is characterized by the disruption of chondro-bone homeostasis, where progressive cartilage degeneration and aberrant bone remodeling exacerbate joint dysfunction. Restoring this imbalance remains a critical challenge in OA treatment. Here, a 3D-printed bilayer scaffold with spatiotemporal release is presented, which integrates an alendronate sodium-triphenylphosphine (ALN-TPP)-loaded polyacrylic acid/methacrylated flaxseed gum (PAA/FGMA) cartilage layer and an ALN-TPP-loaded PAA/vinyl-modified montmorillonite (vinyl-MMT) bone layer. Specifically, FGMA establishes a biomimetic microenvironment that supports the migration and proliferation of chondrocytes. vinyl-MMT improves the mechanical strength of the scaffold, providing structural support for bone regeneration. Crucially, the differential degradation kinetics of the scaffolds orchestrate a temporal release of the ALN-TPP. The faster-degrading upper layer ensures rapid initial release of ALN-TPP, which specifically targets dysfunctional mitochondrial metabolism characteristic of OA, reduces oxidative stress, and inhibits the expression of matrix-degrading enzymes during the early critical phase. Simultaneously, the slower-degrading lower layer provides a sustained release profile. ALN-TPP promotes osteogenesis by stimulating osteoblast differentiation, which effectively reverses abnormal bone remodeling in osteoarthritis. In a rabbit model with osteochondral defects, the bilayer scaffold facilitates effective formation of hyaline-like cartilage and achieves subchondral bone reconstruction. This work establishes a multifunctional therapeutic platform that can advance the repair of osteochondral defects.

3D-Printed PRP-Infused Double-Network Hydrogels Orchestrate Inflammation Resolution and Vascular Regeneration in Infected Wounds

Danlei Xing,Zhiyuan Hu,Yue Tao,Hang Xia,Juanjuan Li,Guoqing Xia,Le Qiu,Jie Shan,Xiaojun Zha,Xu-Lin Chen

Bacterial infection and excessive inflammation critically impede wound healing, necessitating the development of multifunctional bioactive materials. Herein, we report a biodegradable composite hydrogel (PPG) fabricated via projection-based 3D biophotocuring, comprising an interpenetrating double-network of gelatin methacryloyl (GelMA) and poly-L-lysine methacryloyl (PLMA), embedded with platelet-rich plasma (PRP). The resulting hydrogel exhibits potent antibacterial activity and robust inhibition of biofilm formation, alongside integrated anti-inflammatory and pro-angiogenic functions. In vitro, incorporating PRP promotes the migratory and proliferative activity of human skin fibroblasts and facilitated macrophage polarization toward the M2 phenotype. In vivo , PPG accelerates wound closure in bacteria-infected models by facilitating re-epithelialization, collagen remodeling, and neovascularization, as evidenced by sustained α-SMA and elevated CD31 expression. Mechanistically, PPG activates the PI3K/AKT/eNOS signaling axis, further supporting angiogenesis and extracellular matrix deposition. RNA-seq validation showed that PPG upregulated DEFB4 and SPRR2F, promoting wound healing and modulating inflammation. Collectively, this work presents a 3D-printed, bioactive hydrogel platform with synchronized antibacterial, immunomodulatory, and pro-regenerative properties, offering strong translational potential for treating infected wounds.

Janus silk-based patch with temporary adhesion for inflammatory mediators removal in corneal alkali burn treatment

Yilin Zhang,Dongyan Li,Hui Xiao,Xiuyuan Luo,Yi Xie,Sen Hou,Lizhen Wang,Yubo Fan,Jing Ji,Linhao Li

Corneal alkali burns cause persistent inflammation, leading to corneal vascularization and fibrosis, which severely impair vision. Here, we developed a temporary adhesive and detachable Janus silk-based patch to capture and remove inflammatory mediators from the ocular surface. The lower silk layer of the Janus patch incorporates of polyamidoamine and heparin, offering adsorption capacity for inflammatory mediators on the ocular surface. The upper hyaluronan layer imparts lubrication, alleviating foreign-body sensation and reducing shear stress from blinking. The integration of the silk and hyaluronan layers is achieved through interfacial diffusion, liquid–liquid phase separation, and photopolymerization, resulting in a stable interpenetrating network interface. After water annealing, the Janus patch exhibited excellent transparency, mechanical strength, and swelling resistance, remaining attached to the rat ocular surface for 3–5 days. Adsorption tests confirmed that the patch effectively captured small-molecule dyes, proteins, and free DNA. In the rat corneal alkali burn model, imaging and histological evaluations showed significant reductions in vascularization and fibrosis after 3 days of treatment, along with improved corneal transparency. RNA sequencing revealed that patch treatment effectively inhibited the PI3K–AKT inflammatory pathway. This inflammation-removing patch represents an innovative treatment for corneal alkali burns with significant clinical potential.

A dual-active biological scaffold with in situ loaded Abaloparatide and bone marrow-derived mesenchymal stem cells synergistically regulatory T cell to promote bone regeneration

Mingyu Jia,Zhihong Chen,Huajian Zhou,Haoran Jiang,Yukang Zhang,Yangyang Liu,Min Wu

Bone defect repair remains a significant challenge faced in clinical practice. Bone tissue engineering has emerged as an effective approach for bone defect repair due to its ample donor source and remarkable repair efficacy. In this study, we combined Abaloparatide (ABL) with bone marrow-derived mesenchymal stem cells (BMSCs) and utilized methacrylated gelatin (GelMA) as a matrix to successfully develop a bio-scaffold with dual activity (ABL@GelMA/BMSCs). BMSCs regulate the bone microenvironment by secreting cytokines, which promote bone repair through a paracrine mechanism. The addition of ABL not only facilitated the osteogenic differentiation and paracrine function of BMSCs but also significantly enhanced the bone induction capability of the scaffold, as evidenced by the marked upregulation of osteogenic markers (ALP, RUNX2, OCN). Furthermore, this bio-scaffold was able to upregulate the Foxp3 gene, inducing Foxp3+ regulatory T cells, thereby modulating the local immune microenvironment to promote bone formation. After implantation of this bio-scaffold into a critical-sized cranial defect model, histological results demonstrated a higher bone mineral density (BMD), and CT reconstruction indicated good healing of the defect. In summary, this bio-scaffold exhibits a powerful capacity for bone defect repair while improving the bone microenvironment through the synergistic modulation of regulatory T cells, providing a new strategy for bone defect repair.

A Nanovesicle-Sericin-Based Hydrogel Scaffold for RGC Survival in Glaucoma via Regulation of Microglial Polarization

Wenxiang Zhu,Jiawei Chen,Qiuling Huang,Zhimin Liao,Xiaobo Wang,Xiaoyu Zhou,Wei Huang,Dengming Zhou,Xuanchu Duan

Chronic inflammation is closely linked to retinal ganglion cell (RGC) damage in glaucoma. However, naturally derived biomaterials often lack sufficient anti-inflammatory activity and fail to effectively support RGC survival. In this study, we developed a stem cell-loaded, nanovesicle-integrated sericin-based hydrogel, SerMA-PC@PNVs, designed to enhance the survival of RGCs in glaucomatous conditions. Nanovesicles (NVs) derived from periodontal ligament stem cells (PDLSCs) were obtained via ultracentrifugation and liposome extrusion. These NVs were loaded with procyanidins (PC) and functionalized with acrylate-polyethylene glycol-N-hydroxysuccinimide (AC-PEG-NHS). The modified NVs (PC@PNVs) were then covalently grafted onto methacrylated sericin (SerMA) through a photo-cross-linking reaction to form the SerMA-PC@PNVs hydrogel scaffold. In vitro studies demonstrated that the SerMA-PC@PNV scaffold enhanced the secretion of neurotrophic factors by encapsulated PDLSCs. Both in vitro and in vivo results confirmed that the SerMA-PC@PNVs/PDLSCs hydrogel effectively reprogrammed microglial polarization from the pro-inflammatory M1 phenotype to the anti-inflammatory M2 phenotype, thereby creating a neuroprotective microenvironment favorable for RGC survival. Mechanistically, RNA sequencing of retinal tissues revealed significant regulation of pathways associated with the “inflammatory response” and “apoptotic process”. Single-cell RNA sequencing further demonstrated an increase in RGC populations and a reduction in neutrophils and microglia following treatment. Microglial subclustering analysis validated the hydrogel’s regulatory effects on “inflammatory response” signaling. In conclusion, the SerMA-PC@PNVs/PDLSCs can promote the survival of glaucoma-damaged RGCs by regulating the inflammatory response.

Engineered microbial hydrogels with confined architecture and binary microbes for efficient hydrogen production

Li Xiaoyi,Long Qing,Jiang Minwen,Tan Weiwei,Ding Ning,Sun Han,Liu Xuanqi,Hu Xia,Liu Hai,Li Xiaojie,Liu Jin,Zhou Jiajing,Du Xiaosheng

Renewable energy sources, particularly hydrogen, offer a promising solution to address global energy crisis and carbon emissions. Microalgae-driven hydrogen production has attracted immense interest in both scientific and industrial fields. However, challenges such as high oxygen sensitivity, substantial water demand, and low hydrogen production efficiency limit their potential. Here, we develop a core–shell symbiotic hydrogel system for enhanced hydrogen production via leveraging coaxial 3D bioprinting to spatially separate microalgae (i.e., core component) and bacteria (i.e., shell component). These networks optimize light and nutrient utilization while providing a localized anaerobic microenvironment to facilitate hydrogen production from microalgal photosynthesis. The symbiotic system enables a high hydrogen yield (1763 ± 98 mL L – 1 ). The system not only provides a highly efficient, liquid-free strategy for biohydrogen generation, but also advances the understanding of symbiotic relationships and microorganism-material interactions for creating advanced living material systems.

Biomimetic interfacial water-regulated hybrid dual-layer hydrogel for wound healing, anti-adhesion and hemostasis post partial hepatectomy

Jun Li,Xiang Wu,Yanhua Wan,Rongsen Wang,Hanjie Shao,Ximing Wang,Huigang Li,Xinhua Zhou,Yuhang Li,Shengjun Xu,Cheng Zhu,Peiru Zhang,Lili Yan,Rongbin Qi,Hao She,Zheyuan Cao,Haili Li,Hong Li,Di Lu,Xiao Xu

Biomimetic hydrogels, with their excellent biocompatibility and tissue adaptability, have garnered significant attention in the field of wound repair. Dual-layer hydrogels can simultaneously achieve hemostasis and anti-adhesion, demonstrating notable potential for application in liver resection surgery. However, existing systems suffer from insufficient mechanical properties and wet adhesion capabilities, and their efficacy in promoting liver wound healing is limited, severely hindering their clinical translation. To address the complex microenvironment of post-liver resection wounds, this study developed a dual-layer functional hydrogel (DAR@Glu) comprising an anti-adhesion layer (AAL) and a repair layer (RL). Through the synergistic interaction of interfacial water regulation and a multi-hydrogen-bond network, this material combines stable wet adhesion with anti-adhesion properties: the repair layer achieves strong wet adhesion via dynamic covalent bonds and hydrogen bonds, while the introduction of glutamic acid further enhances liver tissue repair activity; the anti-adhesion layer forms a dense hydrated layer that effectively prevents tissue adhesion. In vitro experiments confirmed that DAR@Glu can stably adhere to a 1.5 kg weight underwater, demonstrating excellent mechanical properties and sustained wet adhesion. In a rat partial liver resection model, the introduction of glutamic acid significantly enhanced liver tissue repair efficacy. Postoperative day 7 regional quantitative immunohistochemical analysis of liver tissues showed that in the focal regenerative region at the hydrogel-adjacent liver wound edge, the positive staining levels of Cyclin D1, Ki67 and PCNA in the DAR@Glu group were all significantly increased compared with the control group. The CD68 positivity rate was the lowest, while the hydrogel also demonstrated good hemostatic effects; in a cecal injury model, this hydrogel effectively inhibited intra-abdominal adhesions. Therefore, DAR@Glu can efficiently promote wound healing, hemostasis, and anti-adhesion after liver resection, providing an innovative strategy for the clinical repair of liver injuries.

Electrospun Scaffold-Enabled Bioreactor for Scalable Manufacturing of Mechanically Programmed Macrophage-Derived Extracellular Vesicles Targeting Aged Bone Defects

Xiao Lingfei,Huang Huayi,Ding Yunxiang,Lei Ming,Lei Qingjian,Yan Feifei,Shi Chaoran,Xu Jianbin,Cai Lin,Li Wanli

Extracellular vesicles (EVs) represent promising cell-free therapeutics for the regeneration of aged bone defects. Mechanical stimulation is critical in bone regeneration and significantly influences both EV secretion and cargo composition. Despite this, translating mechanically programmed EVs to clinically relevant doses remains a significant challenge. Most current scalable bioreactor systems has achieved high yields through high-density cell culture and fluid flow, yet they do not provide programmable or spatially consistent mechanical stimulation. In this study, an electrospun scaffold-enabled bioreactor was developed to integrate high-density three-dimensional macrophage culture with programmable cyclic stretching, enabling simultaneous EV scale-up and cargo programming. The interconnected fiber junction network transmits boundary-applied strain with enhanced spatial consistency, while the highly porous scaffold supports dense cell seeding and efficient EV release and collection. This approach increases EV yield by 13.8-fold compared to conventional two-dimensional culture. Fiber-mediated mechanical conditioning activates Piezo1-dependent Ca 2+ signaling and integrin-associated mechanotransduction, promotes Yes-associated protein (YAP) nuclear translocation, and remodels EV cargo composition. The resulting mechanically stimulated EVs (ms-EVs) demonstrate concurrent pro-regenerative activities, including cellular rejuvenation, vascular repair, and osteogenic enhancement, and perform favorably compared to canonical interleukin-4 (IL-4)-polarized M2 macrophage-derived extracellular vesicles (M2-EVs). In aged rat femoral defects, ms-EV-functionalized scaffolds increase bone volume fraction by approximately 1.7-fold at 8 weeks, reduce p16 + senescent cells, and enhance CD31 + neovascularization. These findings establish electrospun scaffolds as scalable mechanobiological manufacturing substrates for the production of functionally programmed EV therapeutics. Graphical

Sodium hyaluronate-based hydrogels with enhanced mechanical properties and anti-oxidant and anti-inflammatory characteristics for wound healing

Lanlan Dong,Reyihanguli Alimu,Zhongwang Li,Xin Zheng,Patiguli Aihemaiti,Wurikaixi Aiyiti,Cijun Shuai,Houfeng Jiang

Hyaluronic acid-xanthan gum-NAGA composite hydrogels were made using polyphenols. • Tea polyphenols optimizes mechanical, pH-response, and antioxidant properties. • NHXTP significantly upregulates IL-10 and suppresses IL-1β and TNF-α expression. • NHXTP accelerates wound closure in a rat full-thickness skin defect model. • NHXTP promotes organized tissue regeneration in full-thickness skin defect rats. Functional hydrogel dressings that combine excellent mechanical properties, biological activity, and environmental responsiveness are urgently needed in wound management. Polyphenols have antioxidant, anti-inflammatory, and wound-healing properties. However, their impact on structural fidelity and long-term stability in 3D-printed hydrogel systems remains poorly understood. Additionally, previous studies lack systematic investigation into swelling behavior within the dynamic pH environment of wound healing. This study aims to develop a multifunctional wound dressing capable of adapting to dynamic pH changes within wounds through systematic modification with polyphenols. Functionalized hyaluronic acid (HA)-xanthan gum (XG)-N-acryloylglycine (NAGA) composite hydrogels (NHX) were prepared using a polyphenol (tea polyphenols (TP), tannic acid (TA), and gallic acid (GA) immersion modification strategy. The effects of polyphenol type, concentration, and immersion time on the material properties were systematically investigated. Incorporating TPs synergistically optimized mechanical properties (337% tensile strain, 0.45 MPa stress), pH-responsive swelling behavior (up to 160% swelling at pH 9), and antioxidant activity (ABTS scavenging >99%) of the hydrogel by forming dynamic hydrogen bonds within the polymer network. In vitro anti-inflammatory experiments confirmed that the NHXTP hydrogel significantly upregulates IL-10, and suppresses IL-1β and TNF-α expression. In a rat full-thickness skin defect model, NHXTP dressings markedly accelerated wound closure (98.1% healing rate at 14 days) and promoted organized tissue regeneration. These findings contribute to the development of next-generation high-performance wound dressings.
