Okara, a soybean by-product, holds significant industrial potential but is currently limited by its unfavorable physicochemical properties. This study innovatively explores the synergistic effects of dry planetary ball milling and Lactobacillus plantarum UFG10 fermentation on enhancing the nutritional and functional properties of okara. As the ball milling time increased (0–10 h), the particle size of okara significantly decreased. UFG10 fermentation significantly reduced the particle size of okara that was un-milled (UO), milled for 2 h (M2), and milled for 5 h (M5), with the particle sizes decreasing from 899 µm, 588 µm, and 251 µm to 367 µm (UF), 360 µm (F2), and 177 µm (F5), respectively. Fermentation had no significant effect on the zeta potential of okara but significantly improved its rheological properties. Furthermore, this dual treatment significantly improved the functional properties of okara such as water holding capacity, water swelling capacity, oil holding capacity, foaming ability and cation exchange capacity. Fourier transform infrared (FT-IR) spectroscopy and X-ray diffraction (XRD) analyses confirmed structural changes in okara, highlighting the effectiveness of the combined treatment in enhancing its physicochemical properties. The fermentation treatment significantly increased the total phenolic content (with an average increase of over 10-fold) and antioxidant activity of okara, which were positively correlated with the ball milling time. Our findings demonstrate that integrating dry planetary ball milling with fermentation represents a novel and effective strategy to transform okara into a high-quality functional food ingredient, offering new insights for its valorization in the food industry.

Atherosclerosis and non-alcoholic fatty liver disease (NAFLD) have been increasing at an alarming rate worldwide. Many clinical studies have underlined the link between NAFLD and atherosclerosis. Our previous experiments have discovered that Lactobacillus (L.) plantarum ATCC14917 supplementation could decrease the progression of atherosclerotic lesion formation. In this study, we aimed to investigate the role of supplementation of L. plantarum ATCC14917 mitigates liver injury in rats fed with a high-fat diet (HFD, 45% kcal from fat). A total of 32 rats were randomly divided into four groups, including two intervention groups, who fed with HFD and administering either 1 × 107 or 1 × 109 colony forming units (CFU) of L. plantarum ATCC14917, the normal control group, and the HFD control group. The results showed that supplementation with low-dose and high-dose of L. plantarum ATCC14917 for 8 weeks could alleviate the body weight gain (p < 0.05), hepatic steatosis, and serum lipid metabolism (p < 0.05) in HFD-fed rats. Moreover, supplementation of L. plantarum ATCC 14917 decreased total cholesterol (TC), triglyceride (TG), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) levels (p < 0.05) in serum, and improved HFD-associated inflammation (p < 0.05). Furthermore, cecal contents were analyzed by high-throughput 16S ribosomal RNA sequencing. The results indicated that supplementation of L. plantarum ATCC 14917 could ameliorate HFD-induced gut dysbiosis. In summary, our findings suggest that supplementation of L. plantarum ATCC 14917 could mitigate NAFLD in rats, suggesting it may be considered as a probiotic agent for preventing HFD-induced obesity.

Background/Aim Probiotics have been suggested as prophylactic measure in colon carcinogenesis . This study aimed at determining the potential prophylactic activity of Lactobacillus rhamnosus GG CGMCC 1.2134 (LGG) strain on colorectal carcinogenesis via measuring its effect on Nuclear factor kappa B (NFκB) inflammatory pathway and apoptosis . Materials and methods 64 Sprague Dawley rats were grouped into four as follows; Group 1 (Healthy control), Group 2 (LGG), Group 3 (cancer control Dimethyl hydrazine (DMH)) and Group 4 (LGG + DMH). LGG was administered orally to LGG and LGG + DMH groups. Colon carcinogenesis was chemically induced in LGG + DMH and DMH groups by weekly injection of 40 mg/kg DMH. Animals were sacrificed after 25 weeks of experiment and tumor characteristics assessed. The change in expression of NFκB-p65, COX-2, TNFα, Bcl-2, Bax, iNOS, VEGFα, β-catenin, Casp3 and p53 were evaluated by western blotting and qRT-PCR. Results LGG treatment significantly reduced tumor incidence, multiplicity and volume in LGG + DMH treatment group compared to DMH cancer control group. Also, LGG treatment reduced the expression of β-catenin and the inflammatory proteins NFκB-p65, COX-2 and TNFα; the anti-apoptotic protein Bcl-2, but increased the expression of the pro-apoptotic proteins Bax , casp3 and p53 compared with DMH group. Conclusion LGG have a potential protection effect against colon carcinogenesis; inducing apoptosis and ameliorating inflammation, and may hold a promise as bio-therapeutic dietary agent.

The functional properties of foods are closely related with its matrix. This study examined the impact of curcumin-fortified Lactobacillus helveticus MB2-1 fermented milk gel on the intestinal lipid digestion and bioaccessibility of curcumin through dynamic digestion method. The gel properties, microstructure, oil droplets stability, free fatty acid (FFA) release and curcumin bioaccessibility during digestion of the curcumin-fortified milk gels fermented by L. helveticus MB2-1 or tradition commercial starters ( Streptococcus thermophilus and Lactobacillus bulgaricus ) were investigated and compared. The curcumin-fortified L. helveticus MB2-1 fermented milk gel was found to present higher apparent viscosity, more thicker properties, and firmer texture, which affect the gastrointestinal digesta status together with the release of oil droplets and solubilized curcumin, promote lipid digestion as well as enhance the bioaccessibility of curcumin during digestion. These results may provide valuable references for the design of therapeutic dairy products beneficial to health in terms of food matrix.

Encapsulation technology is a crucial method for enhancing probiotic survival through gastrointestinal digestion. This study utilized ethanol-modified pomelo pith matrix (PP) passed through 40-, 100-, and 200-mesh sieves (PP-40, PP-60, and PP-100) and sodium alginate and/or fructooligosaccharide (FOS) to create gel beads for encapsulating Lactobacillus plantarum (LP). The effects of these gel beads on probiotic loading, protection, release behavior, and gut microbiota regulation were investigated. The 100-mesh pomelo pith matrix increased the specific surface area and pore volume, enabling uniform probiotic distribution within gel beads and supporting controlled swelling behavior. Pomelo pith matrix gel beads, particularly PP-LP-100, exhibited superior acid tolerance, controlled release, higher loading capacity, improved survival during simulated digestion, and greater protection during pasteurization and storage for Lactobacillus plantarum . Incorporating FOS into pomelo pith matrix gel beads boosted microbial fermentation and diversity, leading to increased gas production, total SCFA levels, and butyric acid content. The gel beads containing FOS increased the abundance of Bifidobacterium , Blautia , Faecalibacterium , and Ruminococcus , while PP-LP gel beads enriched Bacteroides and Lactobacillus . This study provides a foundation for developing advanced probiotic formulations with improved stability and controlled release.

L-Arabinose isomerase (L-AI) has been commonly used as an efficient biocatalyst to produce D-tagatose via the isomerization of D-galactose. However, it remains a significant challenge to efficiently synthesize D-tagatose using the native (wild type) L-AI at an industrial scale. Hence, it is extremely urgent to redesign L-AI to improve its catalytic efficiency towards D-galactose, and herein a structure-based molecular modification ofLactobacillus plantarumCY6 L-AI (LpAI) was performed. Among the engineered LpAI, both F118M and F279I mutants showed an increased D-galactose isomerization activity. Particularly, the specific activity of double mutant F118M/F279I towards D-galactose was increased by 210.1% compared to that of the wild type LpAI (WT). Besides the catalytic activity, the substrate preference of F118M/F279I was also largely changed from L-arabinose to D-galactose. In the enzymatic production of D-tagatose, the yield and conversion ratio of F118M/F279I were increased by 81.2% and 79.6%, respectively, compared to that of WT. Furthermore, the D-tagatose production of whole cells expressing F118M/F279I displayed about 2-fold higher than that of WT cell. These results revealed that the designed site-directed mutagenesis is useful for improving the catalytic efficiency of LpAI towards D-galactose.

Lipoteichoic acid (LTA), a component of the Gram-positive bacterial cell wall, exhibits potent immunomodulatory properties. However, it is highly unstable and susceptible to degradation under gastrointestinal conditions, which limits its in vivo functional application. In this study, LTA from theLactobacillus johnsonii was encapsulated into chitosan-tripolyphosphate (CS-TPP) nanoparticles for the first time, establishing a robust delivery system. The resulting LTA@CS-TPP nanoparticles displayed uniform nano size, favorable stability, high encapsulation efficiency, and sustained release behavior. Simulated digestion assays demonstrated that nanoencapsulation effectively protected LTA from gastric degradation and enabled controlled release in the intestinal phase. Cellular experiments showed that LTA@CS-TPP enhanced antioxidative stress defenses, reduced intracellular ROS levels, and alleviated LPS-induced inflammatory injury. Immunofluorescence analysis further revealed that LTA@CS-TPP significantly suppressed NF-κB activation and nuclear translocation, suggesting interference with the TLR4/MyD88/NF-κB signaling cascade. Compared with free LTA, nanoencapsulation not only preserved but also amplified its bioactivity, whereas blank CS-TPP nanoparticles exerted minimal effects. Collectively, these findings provide the first evidence of chitosan-based nanoencapsulation of LTA and highlight its potential as a bioactive delivery platform for inflammation modulation, laying the groundwork for future oral administration studies with promising applications in functional foods and therapeutic strategies.

Loganic acid (LA), a major iridoid glycoside from Gentiana macrophylla Pall., possesses anti-inflammatory and immunomodulatory properties, but its therapeutic potential in rheumatoid arthritis (RA) and its interaction with the gut microbiota remain poorly defined. Here, we show that LA markedly and dose-dependently attenuates disease severity in collagen-induced arthritis mice, as evidenced by reduced clinical arthritis scores, systemic inflammatory cytokines, and synovial tissue damage. LA reshaped the gut microbiota, and fecal microbiota transplantation from LA-treated donors partially reproduced its protective effects, indicating a causal contribution of the altered microbial community. In particular, LA selectively enriched OTUs annotated as Lactobacillus murinus ( L. murinus ) and increased levels of the microbial tryptophan metabolite indole-3-acetic acid (IAA). Both LA and IAA activated aryl hydrocarbon receptor (AhR) signaling, suppressed tumor necrosis factor-α-induced inflammatory responses in human synoviocytes, and, in vivo, enhanced IAA–AhR–interleukin-10 (IL-10) signaling in arthritic joints. Pharmacological inhibition of AhR with CH223191 substantially reversed LA's therapeutic effects, demonstrating that AhR signaling plays a critical role in its anti-arthritic activity. Collectively, these findings demonstrate that LA protects against RA through a dual mechanism: direct activation of AhR in joint tissues and indirect reinforcement of AhR signaling through an L. murinus -associated microbial IAA axis. This study highlights a gut microbiota–metabolite–AhR pathway as a key mediator of LA's anti-arthritic effects and supports LA as a promising microbiota-informed therapeutic candidate for RA.

Longan polysaccharides (LP) have been demonstrated to possess various health benefits related to gut microbial modulation. Herein, the effects of superfine grinding and enzymolysis pretreatments on the colonic fermentation characteristics of hydrothermal-extracted LP were investigated. LP extracted with combined superfine grinding and enzymolysis pretreatment (LP-SE) exhibited a greater degree of degradation (85.82%) than the untreated polysaccharides (LP-H) during fermentation. The enhanced degradation of LP-SE relative to LP-H was primarily attributed to a shift in molecular weight distribution toward a lower range (2.38-667.8 kDa) and a simpler glycosidic linkage composition, rather than to changes in the proportions of the major monosaccharides, including arabinose and galacturonic acid. The enrichment of Bifidobacterium , Desulfovibrio , and Lactobacillus during LP fermentation highlighted their involvement in polysaccharide metabolism and synthesis of short-chain fatty acids. Notably, LP fermentation products significantly promoted mucin-adhesion properties of representative Lactobacillus strains by upregulating mucBP expressions, particularly for LP-SE. These findings indicate that combined superfine grinding and enzymolysis serves as a potential strategy for enhancing polysaccharide bioactivities. Oral administration of probiotic bacteria represents a promising strategy for the prevention and alleviation of colitis; however, its translational potential is frequently constrained by low bacterial viability under gastrointestinal conditions, inadequate mucosal adhesion, oxidative stress, and suboptimal host–microbe interactions at inflamed mucosal sites. To address these limitations, we developed a bilayer nanoencapsulation system for riboflavin-producing L. plantarum B2, consisting of an inner gallic acid–Ca²⁺ metal–polyphenol network and an outer coating of either hyaluronic acid (HA) or bovine serum albumin (BSA). Successful formation of uniform, conformal nanocoatings on the bacterial surface was confirmed by transmission electron microscopy (TEM), scanning electron microscopy (SEM), atomic force microscopy (AFM), zeta potential and particle size analyses, and laser scanning confocal microscopy (LSCM). Among the coated formulations, HA-coated L. plantarum B2 (H-G- L. plantarum B2) exhibited markedly improved storage stability, enhanced resistance to simulated gastrointestinal fluids, and greater tolerance to hydrogen peroxide-induced oxidative stress, while largely preserving bacterial growth kinetics, acidification capacity, riboflavin production, and isoflavone biotransformation activity during soymilk fermentation. Furthermore, HA-based nanoencapsulation significantly enhanced bacterial adhesion to Caco-2 intestinal epithelial monolayers in vitro and prolonged gastrointestinal retention in mice. In a dextran sulfate sodium (DSS)-induced colitis model, oral administration of nanoencapsulated L. plantarum B2 attenuated body weight loss, colon shortening, histopathological injury, and colonic oxidative stress. Mechanistically, the treatment suppressed the levels of TNF-α, IL-1β, calprotectin, MPO, and MDA; elevated IL-10, CAT, and reduced GSH; restored the expression of tight junction proteins ZO-1 and occludin; and modulated gut microbiota composition. These findings establish bilayer nanoencapsulation as an effective and biocompatible strategy to enhance probiotic oral delivery and highlight the therapeutic potential of riboflavin-producing L. plantarum B2 as a microbiome-targeted intervention for colitis management.

Lactobacillus plantarum R is the predominant bacterium widely used in food fermentation. However, it has a low tolerance to ethanol during fermentation, which restricts its broader application in the food industry. Although biofilm formation enhances bacterial resistance to various stresses, regulating biofilm formation in L. plantarum R remains challenging. In this study, we established a pathway for producing autoinducer 2 (AI-2), a quorum-sensing (QS) signaling molecule, by heterologously expressing pfs and LuxS in L. plantarum R. This approach aimed to modulate cell membrane structure and improve bacterial ethanol resistance. The findings showed that exogenous AI-2 significantly enhanced the growth of L. plantarum R under high-ethanol concentration and improved its biofilm formation at different ethanol concentrations. Furthermore, a multi-omics analysis revealed potential mechanisms by which AI-2 improved the tolerance of L. plantarum R to ethanol. Notably, the upregulation of the crr gene, a component of the phosphotransferase system (PTS), enhanced biofilm formation in response to ethanol stress. In addition, differential metabolites enriched in cysteine and methionine metabolic pathways further supported the role of QS in enhancing the tolerance of L. plantarum R to ethanol. These findings lay the groundwork for enhancing the fermentation performance of strains applied in the food fermentation industry, presenting a novel method for expanding the utilization of L. plantarum R in high-ethanol fermentation.

Simple SummaryPb2+and Cd2+contamination in water threatens ecosystems and human health. To develop efficient, eco-friendly adsorbents and valorize industrial by-products (red mud, distiller’s grains), this study synthesized a novel ternary composite biochar (RM) by immobilizing red mud andLactobacillus plantarumon distiller’s grain-derived biochar. Characterized via SEM-EDS, XRD, and FTIR, RM exhibited a superior porous structure. Batch adsorption experiments indicated that the adsorption on RM followed pseudo-second-order kinetics, with Cd2+adsorption fitting the Langmuir model and Pb2+fitting the Freundlich model. Key mechanisms included surface complexation, ion exchange, and coprecipitation. RM integrates the synergistic advantages of minerals, microorganisms, and biochar, offering a promising, sustainable solution for heavy metal-contaminated water remediation.Pb2+and Cd2+represent common heavy metal contaminants in aquatic environments, posing significant risks to ecosystem stability and human health. To develop efficient adsorbents for removing Cd2+and Pb2+while achieving resource utilization of industrial by-products (red mud and distiller’s grains), this study synthesized a novel composite biochar—red mud–Lactobacillus plantarumcomposite biochar (RM)—by immobilizing red mud andLactobacillus plantarumonto biochar derived from distiller’s grains. The structural and chemical properties of RM were characterized using SEM-EDS, XRD, and FTIR. Batch adsorption experiments were conducted to evaluate the effects of various experimental factors on Cd2+and Pb2+adsorption. The adsorption process was further elucidated through kinetic and isothermal models, revealing that it follows the pseudo-second-order kinetic model. Equilibrium data were best described by the Langmuir model for Cd2+and the Freundlich model for Pb2+. The maximum adsorption capacities reached 12.13 mg/g for Cd2+and 130.10 mg/g for Pb2+. The primary mechanisms involved in Cd2+and Pb2+adsorption by RM include surface complexation, cation–π interactions, ion exchange, and coprecipitation. These findings demonstrate that RM represents a promising and effective adsorbent for the remediation of heavy metal-contaminated water.
