Flurbiprofen axetil (FA) is a prodrug of flurbiprofen (FP), and it is hydrolyzed to the active FP by carboxylesterase in plasma after intravenous injection. The pharmacological action of FP is closely related to its chirality, and S-FP shows better analgesic effects than R-FP. Therefore, it is of great significance to compare the in vivo pharmacokinetic behaviors of R-FP and S-FP. In this study, we designed a sensitive high performance liquid chromatography–tandem mass spectrometry method and used CHIRALPAK-IG3 column for chiral separation to quantify the concentrations of R-FP and S-FP in rat plasma. The results show that this method can accurately and effectively analyze the contents of R-FP and S-FP in plasma. In addition, the systemic exposure was approximately 3.09-folds for the S-FP compared with the R-FP following intravenous administration of the FA to rats at a single dose of 4.5 mg/kg. More importantly, the clearance rate of S-FP is significantly smaller than that of R-FP. Therefore, the development of S-FA injectable emulsion for clinical treatment of postoperative pain is very necessary.

The dimension is a leading parameter in customization of unique optoelectronic properties of halide perovskite, while it is still challenging to achieve dimension control beyond the well-developed composition regulation-based strategy considering their intrinsic feature of ionic crystal. In this paper, ligands-directed confinement effect is emphasized in rendering the reduced-dimensional engineering of halide perovskite, and quasi-2D (OAm) 2 CsPb 2 Br 7 ( n = 2) nanosheets are obtained by introducing a large amount of metal ions. As a representation, with Mg 2+ participation, the initial adsorption of organic ligands (i.e., OA and OAm) to inorganic components (i.e., [PbBr 6 ] 4− ) is promoted, conferring a ligands-directed surface reconstruction to form the lamellar structure composed of alternate organic and inorganic layers. Namely, the inorganic–organic lamellar ensemble will play as a soft template, which effectively restricts the growth of [PbBr 6 ] 4− in the c -axis direction even after the Cs-OA injection, thus producing the anisotropic layered perovskite of (OAm) 2 CsPb 2 Br 7 from CsPbBr 3 scenario. Notably, such reduced-dimensional engineering enables a remarkable luminescence tailoring, of which the deep blue emission centered at 439 nm is achieved. In addition, benefiting from such universal strategy, the luminescence is also dependent on the species of introduced metal ions (e.g., Ca 2+ , Co 2+ , Sr 2+ , and Mn 2+ ).

The composite particles (St/Fe3O4) of starch and Fe3O4 were prepared by a co-precipitation method. Then AA and HEMA were grafted and cross-linked on St/Fe3O4 by a one-step method to obtain St/Fe3O4-g-p(AA-r-HEMA). The effects of the adsorbent dosage, pH value, initial Cu(II) concentration, time and temperature on adsorption were studied. Experiments showed that St/Fe3O4-g-p(AA-r-HEMA) showed a good adsorption performance for Cu(II), with a unit adsorption capacity of 75.5 mg g−1 and a maximum removal rate of 95.0%. The adsorption process followed the Langmuir model and the pseudo-second-order kinetic model, was dominated by chemical adsorption, and belonged to single-layer adsorption. Renewable performance studies showed that after 8 adsorption cycles, 91.2% of the first adsorption effect was still retained.

In this study, The solubility of (R,S)-mandelic acid in different solvents, including mono-solvent (H 2 O, methanol, ethanol, n-propanol, isopropanol) and binary solvents (methanol + H 2 O, ethanol + H 2 O, n-propanol + H 2 O, isopropanol + H 2 O, n-butanol + H 2 O), using analytical stirred-flask method and weight analysis method to determine the compositions of the saturated solutions at the atmospheric pressure. We found that the experimental solubility values increased with the rising of temperature accordingly. The experimental solubilities were correlated by the Ideal model, Apelblat model, λh (Buchowski) model, Wilson model, NRTL model. The results have shown that the Wilson model is the best model, which correlates the solubility of (R, S)-mandelic acid in the mono-solvent and binary solvents and fits well with experimental data. In addition, the thermodynamic properties of dissolution were calculated according to Van’t Hoff equation and experimental solubility, including Gibbs energy, entropy and enthalpy. The result confirms that the dissolution process in all solvents is always endothermic and entropy driven.

(R)-1-phenyl-1,2-ethanediol is an important synthon for the preparation of β-adrenergic blocking agents. This study identified a (2R,3R)-butanediol dehydrogenase (KgBDH) from Kurthia gibsonii SC0312, which showed high enantioselectivity for production of (R)-1-phenyl-1,2-ethanediol by reduction of 2-hydroxyacetophenone. KgBDH was expressed in a recombinant engineered strain, purified, and characterized. It showed good catalytic activity at pH 6–8 and better stability in alkaline (pH 7.5–8) than an acidic environment (pH 6.0–7.0), providing approximately 73 and 88% of residual activity after 96 h at pH 7.5 and 8.0, respectively. The maximum catalytic activity was obtained at 45°C; nevertheless, poor thermal stability was observed at >30°C. Additionally, the examined metal ions did not activate the catalytic activity of KgBDH. A recombinant Escherichia coli strain coexpressing KgBDH and glucose dehydrogenase (GHD) was constructed and immobilized via entrapment with a mixture of activated carbon and calcium alginate via entrapment. The immobilized cells had 1.8-fold higher catalytic activity than that of cells immobilized by calcium alginate alone. The maximum catalytic activity of the immobilized cells was achieved at pH 7.5, and favorable pH stability was observed at pH 6.0–9.0. Moreover, the immobilized cells showed favorable thermal stability at 25–30°C and better operational stability than free cells, retaining approximately 55% of the initial catalytic activity after four cycles. Finally, 81% yields (195 mM product) and >99% enantiomeric excess (ee) of (R)-1-phenyl-1,2-ethanediol were produced within 12 h through a fed-batch strategy with the immobilized cells (25 mg/ml wet cells) at 35°C and 180 rpm, with a productivity of approximately 54 g/L per day.

The neuroprotective role of schizandrin (SA) in cerebral ischemia-reperfusion (I/R) was recently highlighted. However, whether SA plays a regulatory role on autophagy in cerebral I/R injury is still unclear. This study aimed to explore whether the neuroprotective mechanisms of SA were linked to its regulation of AMP-activated protein kinase (AMPK)/mammalian target of rapamycin (mTOR)/autophagy pathway in vivo and in vitro. The present study confirmed that SA significantly improved oxygen-glucose deprivation/re-oxygenation (OGD/R)-induced PC12 cells injury. The results of immunoblotting and confocal microscope showed that SA decreased autophagy in OGD/R-injured PC12 cells, which was reflected by the decreased Beclin-1 and LC3-II expression, autophagy flux level, and LC3 puncta formation. In addition, the autophagy inducer rapamycin partially prevented the effects of SA on cell viability and autophagy after OGD/R, whereas the autophagy inhibitor 3-methyladenine (3-MA) exerted the opposite effect. The results of Western blotting showed that SA markedly decreased the phosphorylation of AMPK (p-AMPK), whereas the phosphor-mTOR (p-mTOR) levels increased in the presence of OGD/R insult. Furthermore, pretreatment with the AMPK inducer AICAR partially reversed the protective effects and autophagy inhibition of SA. However, AMPK inhibitor Compound C pretreatment further promoted the inhibition of SA on autophagy induction and cell damage induced by OGD/R. Taken together, these findings demonstrate that SA protects against OGD/R insult by inhibiting autophagy through the regulation of the AMPK-mTOR pathway and that SA may have therapeutic value for protecting neurons from cerebral ischemia.Keywords:OGD/R;autophagy;AMPK/mTOR;schizandrin;ischemic stroke

Limonene dioxide (LDO) is essential for manufacturing bio-based polycarbonate and non-isocyanate polyurethanes. Herein, we report a strategy for the chemoenzymatic epoxidation of (R)-(+)-limonene to LDO with high selectivity using Rhizopus oryzae whole cells. The presence of sufficient excess acid in the system is essential, in addition to overcoming the hydrolysis of the intermediate product, 1,2-limonene oxide, to accomplish the double epoxidation of limonene. When using a high concentration of H 2 O 2 and trisodium citrate, the conditions were effectively established, even when Novozym 435 was substituted, which had previously been reported to be ineffective in achieving high-yield double epoxidation of limonene. The transfer of H 2 O 2 and peracids between the cell and solvent slowed production. The total amount of peracids generated remained constant, but their concentrations remained moderate, favoring the double epoxidation of limonene. An LDO yield of 93 % and no intermediate product, limonene oxide, were obtained under optimized conditions within 20 h.

Simplifying the anchoring process of Ru nanoparticles (NPs) and activating their bifunctional activity are challenges in the construction of Ru-based catalysts. In this work, FeCoP nanosheets anchored by Ru NPs (r-Ru/FeCoP) were innovatively synthesized using an oxygen defect-assisted-gas-phase phosphorization strategy. Surprisingly, the η100 values of r-Ru/FeCoP in the alkaline hydrogen evolution reaction (HER) and oxygen evolution reaction (OER) processes were 0.8 and 0.58 times those of Pt/C and RuO2, respectively. Notably, the rich Ru NPs as HER active sites on r-Ru/FeCoP enhanced the conductivity of the catalyst and promoted the reaction kinetics for HER, while the metal atoms on the FeCoP nanosheets served as OER active sites to accelerate the desorption of O2. The synergy between the two promoted the improvement of overall water splitting efficiency. Overall, this work provides a new approach for the development of low-cost bifunctional catalysts with high Ru utilization.

Despite the potential of sonodynamic therapy (SDT) in treating malignant tumors, the lack of effective sonosensitizers has limited its clinical implementation. In this study, we explored the relationship between the heteroatom doping concentration in metal–organic frameworks and interface formation after pyrolysis by regulating the addition of manganese sources and successfully derived Z-scheme heterojunctions MnxOy/(A/R)TiO2 (MTO) in situ from MIL-125-NH2 (Ti/Mn). The electron transfer pathway introduced by interfacial contact promoted carrier separation and greatly preserved the effective redox components, significantly influencing the performance of reactive oxygen species generation. Upon reaching the tumor sites, MTO effectively depleted glutathione to alleviate the suppressive tumor environment, and the heterojunctions and MnxOy in MTO facilitated SDT and synergistic chemodynamic therapy (CDT), respectively, leading to enhanced immunogenic cell death (ICD). Furthermore, Mn2+ uptake by dendritic cells (DCs) and the tumor-associated antigens released due to ICD activated the stimulator of interferon genes pathway, which elicited a robust tumor-specific immune response by driving the maturation of DCs and the activation of T cells. In addition, the activated T cells secreted high levels of interferon-γ to enhance Mn3+/Mn2+-mediated ferroptosis in metastatic tumor cells. The combination of MTO-mediated synergistic therapy and PD-L1 checkpoint blockade exhibited vaccine-like functions, inducing stronger systemic immunity and durable immune memory to inhibit tumor progression, metastasis, and recurrence. To summarize, we synthesized a self-enhancing nanoplatform for synergistic SDT/CDT/immunotherapy using multifunctional MOF-derived Z-scheme heterojunctions. This study provides an experimental basis for amplifying the potential of sonosensitizers while optimizing SDT-mediated systemic immunity while avoiding interference caused by additional adjuvants.

Lactobacillus plantarum R is the predominant bacterium widely used in food fermentation. However, it has a low tolerance to ethanol during fermentation, which restricts its broader application in the food industry. Although biofilm formation enhances bacterial resistance to various stresses, regulating biofilm formation in L. plantarum R remains challenging. In this study, we established a pathway for producing autoinducer 2 (AI-2), a quorum-sensing (QS) signaling molecule, by heterologously expressing pfs and LuxS in L. plantarum R. This approach aimed to modulate cell membrane structure and improve bacterial ethanol resistance. The findings showed that exogenous AI-2 significantly enhanced the growth of L. plantarum R under high-ethanol concentration and improved its biofilm formation at different ethanol concentrations. Furthermore, a multi-omics analysis revealed potential mechanisms by which AI-2 improved the tolerance of L. plantarum R to ethanol. Notably, the upregulation of the crr gene, a component of the phosphotransferase system (PTS), enhanced biofilm formation in response to ethanol stress. In addition, differential metabolites enriched in cysteine and methionine metabolic pathways further supported the role of QS in enhancing the tolerance of L. plantarum R to ethanol. These findings lay the groundwork for enhancing the fermentation performance of strains applied in the food fermentation industry, presenting a novel method for expanding the utilization of L. plantarum R in high-ethanol fermentation.

Hepatic ischemia-reperfusion (I/R) injury is a major clinical complication, and the precise monitoring of key inflammatory mediators like neutrophil elastase (NE) in vivo remains a significant challenge. To address this, we developed HDS-NE, a highly specific and activatable molecular probe based on a sulfur-substituted hemicyanine scaffold, engineered for the simultaneous detection of NE via fluorescence and photoacoustic imaging modalities. The probe demonstrated exceptional sensitivity and selectivity toward NE, enabling the real-time visualization of elevated NE activity in a murine model of hepatic I/R injury. Both in vivo and ex vivo imaging results confirmed a markedly enhanced signal in injured livers compared to controls, which was further validated by serum assays and histology. Crucially, this study represents the successful achievement of non-invasive, in vivo imaging of NE activity during hepatic I/R processes, offering a powerful tool for diagnosing and studying inflammatory liver diseases.

Rare-earth ions doped Ca 0.9 R 0.1 CeNbMoO 8 ( R = Y, Sm, Nd, La) ceramics have been successfully prepared by solid-state method, and their modifications to the microstructure and electrical properties are also investigated. The rare-earth ions doped ceramics exhibit the scheelite structure. With the increase in the radius of rare-earth ions, the lattice distortion and bond interaction will be enhanced, and the consistency of grain size will be reduced. The ceramics exhibit negative temperature coefficient (NTC) thermistor characteristics in the temperature range of 473 K-1273 K, and the activation energy decreases with the increase of the radius of rare-earth ions. Rare-earth ions doping can increase the content of Ce 3+ ions and promote the conductivity of ceramics. Except for Sm 3+ -doped ceramics, the high-temperature aging rate of other ceramics is less than 2%. The existence of some metastable Sm 2+ ions in Sm 3+ -doped ceramics not only increases the activation energy, but also reduces the high-temperature stability of the ceramics.
