The COVID-19 pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has significantly impacted public health and social stability, highlighting the urgent need for rapid diagnostic methods to ensure timely clinical intervention. Herein, we develop a sensitive surface-enhanced Raman scattering (SERS) immunosensor based on a silver nanotripod (AgNT) array sandwich structure for simulating early SARS-CoV-2 detection. AgNT arrays modified with SARS-CoV-2 antibody serve as a capture substrate; antibody-modified gold nanoparticles (AuNPs) functionalized with dye molecules act as detection probes. Upon binding to analyte, these components form an AuNPs@AgNT sandwich immunosensor generating enhanced SERS signals for detection. The detection limit of AuNPs@AgNT for SARS-CoV-2 spike protein in phosphate-buffered saline (PBS) samples and saliva samples could reach 0.1 and 0.77 pg mL −1 , respectively. This sandwich immunosensor demonstrates excellent reproducibility with a relative standard deviation lower than 8% and maintains satisfactory detection specificity even in various saliva samples and other proteins. Furthermore, by integrating machine learning technology, the sandwich immunosensor enables accurate discrimination of multi-target samples. These results illustrate the promise of the SERS sandwich immunosensor for the early SARS-CoV-2 spike protein detection, which has a remarkable significance for the virus detection in the future. Graphical The front cover shows a surface-enhanced Raman scattering (SERS) immunosensor based on Ag nanotripod (AgNT) arrays. Au nanoparticles (AuNPs) are loaded with reporters (4-MBA, DTNB, TFMBA) and antibodies (Anti). The AgNT substrate is modified with 3-MPA and Anti. The immunosensor combined with machine learning enables detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein and distinction of tumor necrosis factor-α (TNF-α) and interleukin-1 (IL-1) in mixed samples.

An integrated, reagent-free sensing platform was constructed based on acupuncture needle and molecular imprinting technology. • 4-mercaptophenylboronic acid, poly(methylene blue), and polydopamine were employed as core functional components for targeted recognition and signal transduction. • High selectivity and sensitive detection of the SARS-CoV-2 spike protein were achieved. • The sensor exhibits excellent reproducibility, stability, and operational simplicity. • Reliability and practicality of the sensor were validated using real human serum samples, laying a foundation for clinical application. Accurate detection of the SARS-CoV-2 spike protein (S-protein) is important for the early-stage investigation and monitoring of COVID-19. We report an integrated electrochemical sensing platform with a reagent-free detection step. A modified acupuncture needle was utilized as the substrate for Au S self-assembly of 4-mercaptophenylboronic acid (4-MPBA), which enabled affinity-based capture and controllable elution of the S-protein template via reversible boronate ester interactions. Poly(methylene blue) (PMB) was electropolymerized as a built-in redox mediator, while a molecularly imprinted polymer (MIP) layer was concurrently fabricated by in situ electrophoretic deposition of polydopamine (PDA). The fabricated biosensor exhibited a linear relationship between the signal response and the logarithm of the S-protein concentration over the range of 0.01 to 10 4 ng/mL, with a detection limit of 0.0033 ng/mL (S/N = 3). It also demonstrated good selectivity, reproducibility, and stability. This work provides a proof-of-concept strategy for developing portable and low-cost electrochemical sensing platforms for protein detection. A highly sensitive fluorescent lateral flow immunoassay (LFIA) was developed for SARS‑CoV‑2 IgG detection. Polystyrene (PS) microspheres were synthesized via emulsion polymerization, doped with dipyridamole (Dip), and coated with a protective silica (SiO 2 ) shell through a sol–gel process. After amine functionalization, the SARS‑CoV‑2 nucleocapsid (N) protein was covalently immobilized onto the microsphere surface using EDC/NHS chemistry, yielding PS@Dip@SiO 2 –N fluorescent probes. The LFIA strip enabled one‑step detection within 15 min. Under optimized conditions (N protein conjugation concentration of 40 µg/mL), the strip showed excellent specificity with no cross‑reactivity against other human coronaviruses (NL63, OC43, 229E). Quantitative determination based on the T/C grayscale ratio fitted the Hill equation (R² = 0.9782). The detection limit for SARS‑CoV‑2 IgG was 10 ng/mL, approximately two orders of magnitude lower than that of a commercial colloidal gold‑based LFIA strip. This platform provides a rapid, sensitive, and low‑cost solution for point‑of‑care serological screening of COVID‑19. Graphical abstract

The outbreak of COVID-19 caused by SARS-CoV-2 urges the development of rapidly and accurately diagnostic methods. Here, one high-sensitivity and point-of-care detection method based on magnetic SERS biosensor composed of Fe 3 O 4 -Au nanocomposite and Au nanoneedles array was developed to detect SARS-CoV-2 directly. Among, the magnetic Fe 3 O 4 -Au nanocomposite is applied to capture and separate virus from nasal and throat swabs and enhance the Raman signals of SARS-CoV-2. The magnetic SERS biosensor possessed high sensitivity by optimizing the Fe 3 O 4 -Au nanocomposite. More significantly, the on-site detection of inactivated SARS-CoV-2 virus was achieved based on the magnetic SERS biosensor with ultra-low limit of detection of 100 copies/mL during 15 mins. Furthermore, the contaminated nasal and throat swabs samples were identified by support vector machine, and the diagnostic accuracy of 100% was obtained. The magnetic SERS biosensor combined with support vector machine provides giant potential as the point-of-care detection tool for SARS-CoV-2.

The ongoing outbreak of the novel coronavirus disease 2019 (COVID-19) draws worldwide concerns due to its long incubation period and strong infectivity. Although RT-PCR-based methods are being widely applied for clinical diagnosis, timely and accurate diagnosis towards COVID-19 causing virus, the SARS-CoV-2, is still limited due to labor-intensive and time-consuming operations. Herein, we report a new viral RNA extraction method based on poly-(amino ester) with carboxyl group (PC)-coated magnetic nanoparticles (pcMNPs) for the sensitive detection of SARS-CoV-2. This method combines the lysis and binding steps into one step, and refines multiple washing steps into one step, giving a turnaround time of less than 9 min. Furthermore, the extracted pcMNP-RNA complexes can be directly introduced into subsequent RT-PCR reactions without elution. This simplified viral RNA method could be well adapted in fast manual and automated high-throughput nucleic acids extraction protocols suitable for different scenarios. A high sensitivity down to 100 copies/mL and a linear correlation between 100 and 10 6 copies/mL of SARS-CoV-2 pseudovirus particles are achieved in both protocols. Benefitting from the simplicity and excellent performances, this new method can dramatically improve the efficiency and reduce operational requirements for the early clinical diagnosis and large-scale SARS-CoV-2 nucleic acid screening.

False detection of SARS-CoV-2 is detrimental to epidemic prevention and control. The scalar nature of the detected signal and the imperfect target recognition property of developed methods are the root causes of generating false signals. Here, we reported a collaborative system of CRISPR-Cas13a coupling with the stabilized graphene field-effect transistor, providing high-intensity vector signals for detecting SARS-CoV-2. In this collaborative system, SARS-CoV-2 RNA generates a “big subtraction” signal with a right-shifted feature, whereas any untargets cause the left-shifted characteristic signal. Thus, the false detection of SARS-CoV-2 is eliminated. High sensitivity with 0.15 copies/μL was obtained. In addition, the wide concerned instability of the graphene field-effect transistor for biosensing in solution environment was solved by the hydrophobic treatment to its substrate, which should be a milestone in advancing it's engineering application. This collaborative system characterized by the high-intensity vector signal and amazing stability significantly advances the accurate SARS-CoV-2 detection from the aspect of signal nature.

The origins of preexisting SARS-CoV-2 cross-reactive antibodies and their potential impacts on vaccine efficacy have not been fully clarified. In this study, we demonstrated that S2 was the prevailing target of the preexisting S protein cross-reactive antibodies in both healthy human and SPF mice. A dominant antibody epitope was identified on the connector domain of S2 (1147-SFKEELDKYFKNHT-1160, P144), which could be recognized by preexisting antibodies in both human and mouse. Through metagenomic sequencing and fecal bacteria transplant, we demonstrated that the generation of S2 cross-reactive antibodies was associated with commensal gut bacteria. Furthermore, six P144 reactive monoclonal antibodies were isolated from naïve SPF mice and were proven to cross-react with commensal gut bacteria collected from both human and mouse. A variety of cross-reactive microbial proteins were identified using LC-MS, of which E. coli derived HSP60 and HSP70 proteins were confirmed to be able to bind to one of the isolated monoclonal antibodies. Mice with high levels of preexisting S2 cross-reactive antibodies mounted higher S protein specific binding antibodies, especially against S2, after being immunized with a SARS-CoV-2 S DNA vaccine. Similarly, we found that levels of preexisting S2 and P144-specific antibodies correlated positively with RBD binding antibody titers after two doses of inactivated SARS-CoV-2 vaccination in human. Collectively, our study revealed an alternative origin of preexisting S2-targeted antibodies and disclosed a previously neglected aspect of the impact of gut microbiota on host anti-SARS-CoV-2 immunity.

Rapid and accurate discrimination of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is an available approach to implement a rapid diagnosis of the coronavirus disease 2019 (COVID-19). Here we fully exploited the cleavage properties of exonuclease III (Exo III) and hairpin DNA-assisted target cycling technology to generate bulk single-stranded DNA (ssDNA) that was employed to facilitate the constitution of a three-way junction structure on polymetallic particle (Ag-Au NPs) and Ti 3 C 2 (Ti 3 C 2 @Ag-Au) complexes. Ag-Au NPs presented favorable stability without adding extra stabilizers, demonstrating the potential value of Ag-Au NPs as an alternative to Au NPs in the field of bioanalysis. Uppon the three-way junction structure, the dumbbell hybridization chain amplification (DHCA) was occurred which generated DNA nanostructure with tight conformation. Target cycling and DHCA reactions improved the electrochemiluminescence (ECL) signal, which dramatically advanced the assay sensitivity of SARS-CoV-2 (0.59 fM). Moreover, our strategy remained to demonstrate favorable specificity and repeatability in environmental conditions and real human serum samples.

The spike protein of SARS-CoV-2 interacts with angiotensin-converting enzyme 2 (ACE2) of human respiratory epithelial cells, which leads to infection. Furthermore, low-dose radiation has been found to reduce inflammation and aid the curing of COVID-19. The receptor binding domain (RBD), a recombinant spike protein with a His tag at the C-terminus, binds to ACE2 in human body. We thus constructed a radioiodinated RBD as a molecule-targeted probe to non-invasively explore ACE2 expression in vivo, and to investigate radiotherapy pathway for inhibiting ACE2. RBD was labeled with [ 124 I]NaI using an N-bromosuccinimide (NBS)-mediated method, and 124 I-RBD was obtained after purification with a specific activity of 28.9 GBq/nmol. Its radiochemical purity was (RCP) over 90% in saline for 5 days. The dissociation constant of 124 I-RBD binding to hACE2 was 75.7 nM. The uptake of 124 I-RBD by HeLa ACE+ cells at 2 h was 2.96% ± 0.35%, which could be substantially blocked by an excessive amount of RBD, and drop to 1.71% ± 0.23%. In BALB/c mice, the biodistribution of 124 I-RBD after intravenous injection showed a moderate metabolism rate, and its 24 h-post injection (p.i.) organ distribution was similar to the expression profile in body. Micro-PET imaging of mice after intrapulmonary injection showed high uptake of lung at 1, 4, 24 h p.i.. In conclusion, the experimental results demonstrate the potential of 124 I-RBD as a novel targeted molecular probe for COVID-19. This probe may be used for non-invasive ACE2 mapping in mammals.

The rapid and sensitive diagnosis of the highly contagious severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is one of the crucial issues at the outbreak of the ongoing global pandemic that has no valid cure. Here, we propose a SARS-CoV-2 antibody conjugated magnetic graphene quantum dots (GQDs)-based magnetic relaxation switch (MRSw) that specifically recognizes the SARS-CoV-2. The probe of MRSw can be directly mixed with the test sample in a fully sealed vial without sample pretreatment, which largely reduces the testers’ risk of infection during the operation. The closed-tube one-step strategy to detect SARS-CoV-2 is developed with home-made ultra-low field nuclear magnetic resonance (ULF NMR) relaxometry working at 118 μT. The magnetic GQDs-based probe shows ultra-high sensitivity in the detection of SARS-CoV-2 due to its high magnetic relaxivity, and the limit of detection is optimized to 248 Particles mL ‒1 . Meanwhile, the detection time in ULF NMR system is only 2 min, which can significantly improve the efficiency of detection. In short, the magnetic GQDs-based MRSw coupled with ULF NMR can realize a rapid, safe, and sensitive detection of SARS-CoV-2.

The harm and impact of the COVID-19 pandemic have highlighted the importance of fast, sensitive, and cost-effective virus detection methods. In this study, we developed a DNA aptamer sensor using nanoparticle-enhanced surface plasmon resonance imaging (SPRi) technology to achieve efficient labeling-free detection of SARS-CoV-2 S protein. We used the same DNA aptamer to modify the surface of the SPRi sensor chip and gold nanoparticles (AuNPs), respectively, for capturing target analytes and amplifying signals, achieving ideal results while greatly reducing costs and simplifying the preparation process. The SPRi sensing method exhibits a good linear relationship ( R 2 = 0.9926) in the concentration range of 1–20 nM before adding AuNPs to amplify the signal, with a limit of detection (LOD) of 0.32 nM. After amplifying the signal, there is a good linear relationship ( R 2 = 0.9829) between the concentration range of 25–1000 pM, with a LOD of 5.99 pM. The simulation results also verified the effectiveness of AuNPs in improving SPRi signal response. The SPRi sensor has the advantage of short detection time and can complete the detection within 10 min. In addition, the specificity and repeatability of this method can achieve excellent results. This is the first study to simultaneously capture a viral marker protein and amplify the signal using polyadenylic acid (polyA)–modified DNA aptamers on the SPR platform. This scheme can be used as a fast and inexpensive detection method for diagnosis at the point of care (POC) to combat current and future epidemics caused by the virus.
