Protocatechualdehyde (PA) is a phenolic acid present in many plants and has many biological activities. Herein, the antagonistic effects and the action mechanism of PA against methicillin-resistant Staphylococcus aureus (MRSA) were studied. The results showed that PA had both significant antibacterial and anti-biofilm activities against MRSA. Additionally, PA had synergy with ampicillin against MRSA. It was elucidated that PA was prominent in destroying cell membranes, increasing cell membrane permeability and intracellular ROS production, thus leading to bacterial cell damage. Transcriptome analysis showed that PA disrupts many physiological pathways, including increasing cell membrane permeability, inhibiting biofilm formation, decreasing resistance to antimicrobial agents, and impairing DNA replication. Finally, the antimicrobial preservation test showed that PA could inhibit the growth of MRSA and prevent the corruption of beef. In summary, PA is an effective natural antibacterial substance and has a good application potential in food preservation, even in tackling antibiotic resistance problems.

Taking the endophytic fungus UJ3-2, isolated from Urtica fissa , as the experimental material, this study aimed to explore the composition of its metabolites and the underlying mechanisms by which it inhibits Staphylococcus aureus . Initially, the MIC, MBC, inhibitory curves, biofilm growth, and extracellular nucleic acids and proteins of S. aureus in response to the metabolites were measured. Secondly, PI staining and SEM were used to evaluate the impact of the metabolites on the integrity of the cell wall and overall morphology of S. aureus . Additionally, UPLC-MS was employed to analyze the composition of the secondary metabolites. The UJ3-2 strain was identified as Xylaria grammica based on ITS sequencing and designated as Xylaria grammica UJ3-2. Our results revealed that the metabolites of UJ3-2 exhibited excellent in vitro antibacterial activity against S. aureus , with both MIC and MBC values of 3.125 mg/mL. The inhibitory curve confirmed that 1 MIC of UJ3-2 metabolites could completely inhibit the growth of S. aureus within 24 h. With increasing concentrations of UJ3-2 metabolites, the growth of S. aureus biofilms was significantly suppressed, and obvious leakage of nucleic acids and proteins was observed. PI fluorescence staining indicated that various concentrations of UJ3-2 metabolites disrupted the integrity of the S. aureus cell membrane. SEM observation revealed that the treated S. aureus surfaces became rough, and the bacteria shrank and adhered to each other, showing a dose-dependent effect. UPLC-MS analysis suggested that the main components of the fermented metabolites were 6-oxocineole (17.92%), (S)-2-acetolactate (9.91%), 3-methyl-cis,cis-muconate (4.36%), and 8-oxogeranial (3.17%). This study demonstrates that the endophytic fungus UJ3-2 exhibits remarkable in vitro antibacterial effects against S. aureus , primarily by enhancing the permeability of the S. aureus cell membrane, causing the leakage of its intracellular contents, and altering the bacterial surface morphology to inhibit the pathogen. The endophytic fungus UJ3-2 has a good antibacterial effect on S. aureus , which gives it certain application prospects in the screening and industrial production of new and efficient natural antibacterial active substances.

Photodynamic therapy (PDT) has emerged as an effective way to deal with drug-resistant bacterial infections. Especially, blue light (BL) mediated PDT (BL-PDT) presents unique advantages in the treatments of skin infection due to the strong light absorption of superficial skin, weak penetration of BL and little damage to deep tissues. However, the photosensitizers used for BL-PDT are very limited, and the ongoing development of novel BL photosensitizers is indispensable. Porphyrins are good sources for developing efficient photosensitizers. Herein, for developing more effective BL photosensitizers, five porphyrin derivatives that can be excited by BL [5,10,15,20-tetraphenylporphyrin (TPP), 5,10,15,20-tetrakis(4-bromophenyl)porphyrin (TBPP), 5,10,15,20-tetrakis(4-chlorophenyl)porphyrin (TCPP), 5,10,15,20-tetrakis(4-fluorophenyl)porphyrin (TFPP), 5,10,15,20-tetrakis(4-iodophenyl)porphyrin (TIPP)] are subjected to the investigation of PDT against MRSA (methicillin resistant Staphylococcus aureus ). The results reveal that TBPP-mediated BL-PDT shows outstanding bactericidal effects. Mechanism studies show that TBPP + BL can induce reactive oxygen species (ROS) up-regulated in MRSA, rupture cell membrane, inhibit ATP (adenosine triphosphate) production and virulence factor expression. Furthermore, TBPP + BL effectively eliminates MRSA form biofilms, inhibits biofilm formation and disintegrates mature biofilms. More importantly, TBPP-PDT significantly accelerate mouse skin wound healing in a biofilm infection model. Our work offers new insights into the development of novel BL photosensitizers. Photodynamic therapy mediated by 5,10,15,20-tetrakis(4-bromophenyl)porphyrin (TBPP) effectively eliminates drug-resistant Staphylococcus aureus , inhibits biofilm formation and accelerates wound healing in a mouse model.

Although bacteriophages (phages) are ideal biorecognition elements employed for Staphylococcus aureus detection, existing single-phage-based approaches are limited by the specific affinity of phages and false-negative results. To develop an improved phage-based colourimetric detection system and investigate its detection mechanism, we prepared a phage cocktail composed of three S. aureus phages (SapYZU10, SapYZU11, and SapYZUM13), immobilised it on Co-Mn nanozymes, and applied it to S. aureus for evaluation. The phage cocktail@Co-Mn demonstrated a sensitive, specific, and fast colourimetric evaluation of live S. aureus counts in approximately 17 min, with a detection limit of 92 CFU/mL. Moreover, the phage cocktail@Co-Mn was successfully used to detect S. aureus in food samples, with recovery rates ranging from 92.21 % to 105.56 %. The phage cocktail@Co-Mn also exhibited resilience to changes in pH (3–9), five food additives, and NaCl concentration (2 %–10 %). The detection mechanism of the phage cocktail@Co-Mn involves blockage of the active sites of the nanozyme producing O 2 •– . Notably, owing to the synergistic recognition spectra of the three phages in the cocktail, the recognition rate for the strains was a remarkable 100 %, compared with the much lower recognition rates of individual phages (between 74.7 % and 76.9 %). The recognition mechanism involves adsorption between phage tail fibres and S. aureus hosts. This pioneering study revealed that the phage cocktail is an effective biorecognition element suitable for the rapid, sensitive, specific, reliable, and colourimetric detection of viable S. aureus strains in the food industry, providing innovative directions for the phage-based detection methods.

The misuse of antibiotics in animal protein production has driven the emergence of a range of drug-resistant pathogens, which threaten existing public health security. Consequently, there is an urgent need to develop novel antimicrobials and new infection treatment options to address the challenges posed by the dramatic spread of antibiotic resistance. Piscidins, a class of fish-specific antimicrobial peptides (AMPs), are regarded as promising therapies for biomedical applications. Progress towards potential analogs from the piscidin family has been hampered by unenforceable structural optimization strategies. Here, we leverage a strategy of bioinformatics analysis combined with molecular dynamics (MD) simulation to identify specific functional hotspots in piscidins and rationally design related analogues. As expected, this approach yields a potent and non-toxic PIS-A-1 that can be used as an antibiotic adjuvant to reverse methicillin-resistant Staphylococcus aureus (MRSA) pathogens. Remarkably, the structural optimization scheme and application strategy proposed here will contribute richer therapeutic options for the safe production of animal protein.

Antibiotic-free antimicrobial strategies are urgently needed to address the rapid evolution of antimicrobial resistance and transmission of multidrug-resistance bacterial infections. Herein, we fabricated polydopamine-coated porous magnetic nanoparticles (pMNPs@PDA) for effective separation and photothermal killing of methicillin-resistant Staphylococcus aureus (MRSA). Taking advantage of the excellent bacteria-affinitive property of polydopamine, the nanoparticles were anchored on the surface of bacteria, permitting rapid and efficient MRSA capture and separation with over 99% removal via the application of a magnetic field in 30 min. It was found, for the first time, that polydopamine-coated magnetic nanoparticles displayed a selective capture of Gram-positive bacteria when compared with Gram-negative bacteria. The selectivity was attributed to the preferable binding capability of pMNPs@PDA to peptidoglycan (PGN) of Gram-positive bacteria, compared to the lipopolysaccharide (LPS) of Gram-negative bacteria. With the magnetic separation and photothermal properties, pMNPs@PDA exhibited efficient killing of the captured MRSA under the irradiation of near-infrared (NIR) light. Cell cytotoxicity testing demonstrated good biocompatibility of the nanoparticles. These antibiotic-free nanoparticles capable of fast capture, separation, and inactivation of MRSA may be potentially used for water disinfection, blood purification, and treatment of bacterial infections.

To synthesize three different grafting ratios of gallic acid (GA)-chitosan (CS) copolymer by a free radical mediated grafting method, which is further applied to the field of antibacterial materials, crosslinking structures of the compound GA-CS copolymer were characterized, fully indicating that gallic acid is resoundingly grafted onto chitosan. The grafting ratios of three copolymers GA-CS are 45.71% (I), 36.12% (II), and 18.96% (III) were determined by UV-Vis spectrophotometer. The minimum inhibitory concentrations of three GA-CS copolymers are 30 µg/mL against Escherichia coli and are ranged from 250 to 550 µg/mL against Staphylococcus aureus. By counting viable bacterial colonies, it can be found that antibacterial property is preferable by increasing the grafting ratio of GA-CS copolymers. Findings of investigation on aforementioned bacteria experiment indicate that the CFU/mL values of GA-CS (I, II, III) are 2.04×10 6 , 7.56×10 6 , 1.48×10 7 to Staphylococcus aureus , and 2.96×10 6 , 1.01×10 7 , 2.14×10 7 to Escherichia coli after 12 h treatment. In addition, the interaction process between GA-CS copolymer and bacteria can be observed through a transmission electron microscope. The specific manifestation is that the bacterial cell membranes are ruptured after being treated with the copolymer, which causes the cell contents to flow out, and the cell morphology is shrunk and out of shape.

This study explored the antibiofilm efficacy of 3,3'-diindolylmethane (DIM) on Staphylococcus aureus and its disinfection on common food-contact surfaces. The minimum biofilm inhibitory concentration (MBIC) of DIM on S. aureus was 62.5 μmol/L, while it did not impede the bacterial growth evaluated by growth curve and XTT reduction assay. DIM in the concentration range of 31.2−62.5 μmol/L demonstrated a dose-dependent antibiofilm activity to S. aureus , as confirmed by light microscopic (LM), confocal laser scanning microscopic (CLSM), and scanning electron microscopic (SEM) analyses. At DIM of 62.5 μmol/L, the biomass of S. aureus biofilm was significantly reduced by 97 % and its average thickness by 58 % ( P < 0.05). DIM of 62.5 μmol/L inhibited the bacterial initial adhesion and proliferation, as well as cell motility; the release of extracellular DNA (eDNA) and extracellular polysaccharide (EPS) were reduced by 75 % and 69 %, respectively. DIM exhibited a strong inhibition to S. aureus biofilm formation on common food-contact surfaces, including 304 stainless steel, glass, and polyvinyl chloride (PVC) but not disperse the mature biofilm. Overall, our investigation identified DIM as a promising antibiofilm agent and its suitability to prevent the biofilm formation of S. aureus on common food-contact surfaces utilized during food processing.

Methicillin-resistant Staphylococcus aureus (MRSA), one of the most important zoonotic bacterial pathogens, necessitates innovative antibacterial strategies. Rosmarinic acid (RA) possesses various biological functions, including antibacterial and anti-inflammatory effects. This study aimed to explore the synergistic effects and mechanism of RA in conjunction with ceftiofur (CF) against MRSA. Results showed that the combination of RA and CF demonstrated synergistic anti-MRSA activity, conferring protective effects in an MRSA bacteremia mouse model. Mechanistic investigations further established that RA interacts with sortase A (SrtA) through hydrogen bonds, electrostatic, and van der Waals forces at critical sites, leading to the reversible inhibition of SrtA activity. Consequently, this inhibition impeded the ability of MRSA to invade cells and form biofilms. Conclusively, RA directly inhibits SrtA activity, thus enhancing the synergistic anti-MRSA effect of CF. These findings provide a theoretical foundation for the use of RA in combination with CF as a therapeutic strategy for MRSA infections.

An innovative light scattering immunoassay was developed using an AuNPs etching strategy. Three types of anisotropic gold nanoparticles, including gold nanocubes, nanorods, and nanoflowers with distinct morphologies, were utilized to investigate how these morphological differences affect the sensitivity of light scattering signal transduction. Based on theoretical insights into light scattering and electromagnetic fields, gold nanocubes were identified as the optimal probes for enhancing light scattering signal transduction and were employed to construct an immunoassay for detecting staphylococcal enterotoxin A (SEA). The developed immunoassay achieved ultrahigh sensitivity for SEA detection in milk samples, with a detection limit of 10.39 pg mL −1 , which is 190 times lower than that of conventional ELISA. The proposed immunoassay was validated across ten food samples, demonstrating high accuracy and robustness. Given these promising results, we believe this method has significant potential for screening trace levels of SEA in food products.

Staphylococcus aureus (S. aureus) is a major pathogenic bacterium responsible for bacterial foodborne diseases, making its rapid, specific, and accurate detection crucial. In this study, we develop a ratiometric biosensor based on the recombinase polymerase amplification-clustered regularly interspaced short palindromic repeats/CRISPR associated protein 12a (RPA-CRISPR/Cas12a) system and Eu-metal–organic framework (Eu-MOF) fluorescent nanomaterials for the high-sensitivity detection of S. aureus, combining with RPA for efficient isothermal amplification, this sensor enhances specificity and sensitivity by utilizing the target activation of CRISPR/Cas12a. The Eu-MOF serves a dual function, providing stable red fluorescence as a reference signal and adsorbing FAM-labeled probes for fluorescence quenching, forming a dual-signal system that significantly reduces background interference. This ratiometric design enables accurate and quantitative detection over a wide range (7.9 × 100 to 7.9 × 108 CFU/mL) with a low detection limit of 3 CFU/mL. Overall, with these merits of simplicity, rapid response, high sensitivity, and specificity, this dual-signal biosensor offers a promising method for accurately evaluating S. aureus contamination in food under complex substrate conditions.

Staphylococcus aureus(S. aureus) is a prominent foodborne pathogen that poses a continuous threat to global public health and food safety due to its possession of a variety of toxins and its multidrug resistance. Medium-chain fatty acids (MCFAs), notably lauric acid (LA), exhibit strong antimicrobial properties, but their application is heavily constrained by poor water solubility. In this study, optimized LA emulsions stabilized by chitosan (CS) and polyvinyl alcohol (PVA) were evaluated for their antibacterial activity and detailed mode of action againstS. aureusATCC 6538. The antibacterial activities were evaluated by the maximum inhibition zone, with the 20 CS-PVA/DLTA-LA formulation exhibiting stable dispersion and potent antibacterial activity at 1%. The underlying antibacterial mechanisms againstS. aureuswere specifically focused on cell membranes and peptidoglycan. Therein, the binding of emulsion droplets to the anionic bacterial surface was driven by electrostatic attraction. Membrane degradation was also observed with membrane dysfunctions involving membrane depolarization, increased permeability, and fluidity reduction triggered by their subsequent insertion into the lipid bilayer, which may cause cell dysmetabolism, disintegration, and eventual cell death. Overall, these findings substantiate that LA emulsions disruptS. aureusby operating potentially multi-targeted effects involving cell membrane disruption and peptidoglycan interference, offering a promising alternative approach warranting further investigation for foodborne pathogen control.
