Impact of weakly charged insoluble karaya gum on zein nanoparticle and mechanism for stabilizing Pickering emulsions

Baoguang Wu,Shipei Zhang,Xiaofen Jiang,Pengfei Hou,Yuelin Xin,Lei Zhang,Jinsong Zhang,Deyi Zhou

The effect of weakly charged insoluble karaya gum (KG) on zein colloidal nanoparticles (ZKGPs) for stabilizing Pickering emulsions was investigated. Due to weak surface charge, KG could cover the surface of zein particles by hydrogen bonds and weak electrostatic interactions. With the increase in coverage, the zeta potential of ZKGPs changed from positive to negative values close to zero and the average particle size tended to become larger. The closest neutral wettability (89.85°) was achieved when the zein/KG mass ratio was 1:1. The samples prepared with high oil volume fraction (φ = 0.5–0.75) and high particle concentration (1.0–1.3 %, w / v ) formed emulsion gels easily and showed higher storage stability. CLSM images also confirmed that ZKGPs could be distributed in the continuous phase to enhance the emulsion network structure. Consequently, weakly charged ZKGPs reduced the emulsification energy barrier and increased the coverage and steric hindrance of particles at the oil/water interface. These findings provide new ideas for the development of stable Pickering emulsions for application in food textural modification as well as encapsulation and delivery of bioactive substances.

Pengquan Zhao,Jiandong Wang,Xinyu Yan,Zihui Cai,Lina Fu,Qi Gu,Lin Liu,Haojie Jin,Yujie Fu

In this study, the bioactive extract of Rosa roxburghii Tratt (RRT) leaves was incorporated into a chitosan/zein matrix to produce a food packaging film with higher antioxidant and antimicrobial activities. The natural deep eutectic solvent (NADES) was taken as the extraction solvent and green plasticizer to prepare the film matrix. The physical and structural properties of chitosan/zein films with different concentrations of RRT leaves extracts were characterized by infrared spectroscopy analysis , thermogravimetric analysis , X-ray diffraction and scanning electron microscope. The incorporation of RRT leaves extract significantly improved the mechanical properties , light barrier properties and water vapor permeability of the chitosan/zein films. Moreover, the films containing RRT leaves extract had better antioxidant activities and antibacterial activities, with the highest DPPH and ABTS radical scavenging activities of 88.17 % and 86.84 %, respectively, and also effectively inhibited the growth of microorganisms and extended the shelf life of blueberries and cherry tomatoes .

Preparation and evaluation of lecithin/zein hybrid nanoparticles for the oral delivery of Panax notoginseng saponins

Wen Fu,Yiping Liang,Zhonghui Xie,Hangyi Wu,Zhenhai Zhang,Huixia Lv

Panax Notoginseng Saponins (PNS) has been widely used in the prevention and treatment of cardiovascular and cerebrovascular diseases such as myocardial infarction, heart failure and cerebral infarction . However, oral administration of PNS showed low bioavailability because of its instability and poor membrane permeability in the gastrointestinal tract. Here, lipoprotein-inspired hybrid nanoparticles of PNS-Lecithin-Zein (PLZ-NPs) were prepared by using a simple phase separation method, which possessed a core-shell structure, where zein was used as protein part to replace the animal origin protein to increase the resistance to acid and enzymes while lecithin was used as the lipid composition to improve the oral absorption of PNS as well as to increase the drug loading capacity of PNS into the nanocarriers . The results of stability test showed that PLZ-NPs had robust enzymolysis resistance ability for acid and digestive enzymes of gastrointestinal environments. The fluorescent resonance energy transfer (FRET) assay confirmed the ability of LZ-NPs to be intactly absorbed by Caco-2 cell monolayer. Cell transport studies demonstrated that the permeability of PLZ-NPs in Caco-2/HT29-MTX co-culture cell model was 1.5-fold that of PNS. Meanwhile, the single-pass intestinal perfusion assay proved the absorption parameter Peff of PLZ-NPs was 1.75 and 1.80 times higher than that of PNS in the ileum and jejunum , respectively. Finally, the in vivo pharmacokinetic experiment showed that the relative oral bioavailability of PLZ-NPs was 1.71-fold that of free PNS in SD rat . In summary, the employment of the Lecithin/Zein hybrid nanoparticles could be considered as a promising approach for PNS analogues.

Epoxide Cross-Linked and Lysine-Blocked Zein Ultrafine Fibrous Scaffolds with Prominent Wet Stability and Cytocompatibility

Lu Liu,Jiancai Jiang,Xinyang Jin,Ran Li,Liqian Huang,Ni Wang,Narendra Reddy,Wanshuang Liu,Qiuran Jiang

The healing of chronic wounds, which bring profound problems, can be effectively promoted by skin tissue engineering using scaffolds with features of extracellular matrix (ECM) for supporting native cell growth. Protein ultrafine fibrous scaffolds, although with similar morphology and chemical composition to ECMs, show poor wet stability which leads to substantial deformation, low mechanical properties, and fast degradation. This research provides a two-step dry state treatment including a cross-linking process by ethylene glycol diglycidyl ether (EGDE) and a blocking process by lysine for the modification of zein ultrafine fibrous scaffold model. This distinctive two-step dry state treatment could effectively avoid fiber deformation before fully cross-linking and more importantly the concern of cytotoxicity. The modified zein/EGDE scaffolds displayed remarkable reduced shrinkage of merely 1.25%, enhanced thermal stability, improved mechanical properties around 3–4 fold, retardant degradation to above 60 days, and promoted cytocompatibility about three fold. This work revealed the possibility to develop strong, wet stable, and cytocompatible ultrafine fibers from various proteins for a wide variety of applications in the fields, such as, tissue engineering scaffolds, biosensors, and drug carriers.

Changes in microstructure and rheological properties of konjac glucomannan/zein blend film-forming solution during drying

Chong Li,Fei Xiang,Kao Wu,Fatang Jiang,Xuewen Ni

During film formation at 60 °C, the microstructure and rheological properties of konjac glucomannan (KGM) film-forming solution and KGM/zein blend film-forming solution were investigated. The drying process of film-forming solutions was divided into two stages according to the drying curves. Scanning electron microscopy showed that KGM chains in the blend solution aggregated into thicker chains and formed a molecular network with larger pores. Zein particles grew larger but were homogeneously distributed during drying as observed by confocal laser scanning microscopy. The addition of zein improved the thermal stability of the film-forming solution. As the drying proceeded (up to 8 h), KGM solution exhibited a typical concentrated solution behavior due to molecular entanglement; whereas the blend solution gradually formed a weak gel after 2 h. Complex viscosity data for the film-forming solutions were well-fitted by the power-law model. The information obtained from the study is important for understanding the film-forming mechanism.

Facile preparation of succinylated-zein-ZIF-8 hybrid for enhanced stability and pH-responsive drug delivery

Wei Zhu,Wenquan Huang,Liping Ye,Yuehua Deng,Qiuling Xie,Yanbin Jiang

Zein nanoparticles have good prospects as a pharmaceutical carrier but limited in further application by its poor stability and re-dispersibility. To obtain high stability and pH-responsive drug delivery, succinylated-zein (Suc-zein) was synthesized by nucleophilic substitution reaction, the material obtained was used to prepare Suc-zein nanoparticles (SZs). The SZs prepared not only had good dispersibility but showed better stability than zein particles on pH, storage and salinity. After loading indomethacin (Indo) into zeolitic imidazolate framework-8 (ZIF-8), the core-shelled Indo-Suc-zein-ZIF-8 (Indo-SZZs) were fabricated by coating Suc-zein onto the surface of ZIF-8 with better stability and high drug loading. Indo-SZZs were pH-responsive and sustained a drug release profile in a simulated environment. Also SZZs presented as being non-toxic, having good biocompatibility and good cellular up-take performance, and the cytotoxicity of Indo-SZZs was significantly higher than that of Indo. This provides a new insight into the preparation of a zein-based oral drug delivery system.

Electrospun Hydrophobic Nanofiber Films from Biodegradable Zein and Curcumin with Improved Tensile Strength for Air Filtration

Xie Shiyu,Xu Bo,Yuan Li,Zhao Yaxin,Ma Na,Wang Yaomin,Liu Dagang,Xiang Aimin,Ouyang Yuge,Tian Huafeng

Zein as a natural protein had been widely used in various fields due to its biodegradability and biocompatibility. However, its sensitivity to humidity and poor mechanical performance limited its application in practice. In this study, zein-based composite nanofibers loaded with curcumin were prepared by electrospinning assisted with polyvinyl alcohol (PVA). The filtration efficiency of the modified nanofibers to the particles with diameters larger than 0.5 μm was all above 98%. The loaded curcumin interacted with protein molecular chains to form a network structure within tightly connected nanofibers, which exhibited excellent moisture resistance and good adhesion to cellulose paper towels used as air filter substrates. Meanwhile, its tensile strength had also been enhanced vastly to 0.72 MPa compared with the initial tensile strength of 0.21 MPa. This study provides a new electrospinning strategy for the preparation of zein-based composite nanofibers that can be widely utilized in air filtration with high moisture resistance.

Interfacial properties and structure of pickering emulsions co-stabilized by different charge emulsifiers and zein nanoparticles

Song Cui,David Julian McClements,Xiaoyang He,Xingfeng Xu,Fenghua Tan,Dongdi Yang,Qingjie Sun,Lei Dai

The stability of Pickering emulsions can be improved by the synergistic action of colloidal particles and small molecule emulsifiers. In addition, the interaction between particles and different charge emulsifiers at the interface may result in different interfacial film structures, thus affecting the stability of Pickering emulsions. However, little is currently known about the impact of emulsifier charge on the fabrication and characteristics of these co-stabilized Pickering emulsions. Herein, we investigated the influence of Pickering emulsions co-stabilized by zein nanoparticles and anionic (quillaja saponin, QS), non-ionic (Tween 80), or cationic (lauroyl arginate ethyl hydrochloride) emulsifiers on the interfacial properties. The adsorption kinetics and rheological behavior of the molecular emulsifiers at oil/water interfaces were studied. Changes in interfacial structure and adsorption behavior were studied by confocal laser scanning microscopy (CLSM), and quartz crystal microbalance-dissipation (QCM-D) analysis. The dynamic interface characteristics showed that QS has good emulsification performance. The CLSM images indicated that the emulsifier charge impacted the interfacial structure of the Pickering emulsions. The combination of QS and zein nanoparticles formed a coating at the oil/water interface that was denser than for the other two emulsifiers, which improved the stability of the Pickering emulsions. QCM-D revealed that the interfacial film formed by anionic emulsifiers and zein nanoparticles was the thickest (about 6.6 nm). Moreover, anionic emulsifiers and zein nanoparticles stabilized Pickering emulsions provided the best protection against curcumin. These results may be important for optimizing the performance of Pickering emulsions in the food and pharmaceutical industries.

Zein nanospheres assisting inorganic and organic drug combination to overcome stent implantation-induced thrombosis and infection

Hua-Jie Wang,Meng-Fei Hao,Guan Wang,Hao Peng,Fazli Wahid,Yan Yang,Lei Liang,Shan-Qin Liu,Ren-Long Li,Shu-Ying Feng

The complication of stent implantation is the biggest obstacle to the success of its clinical application. In this study, we developed a combination way of 3D printing and the coating technique for preparation of functional polyurethane stents against stent implantation-induced thrombosis and postoperative infection. SEM, XPS, static water contact angle, and XRD demonstrated that the functional polyurethane stent had a 37 μm-thickness membrane composed of zein nanospheres (250–350 nm). Meanwhile, ZnO nanoparticles were encapsulated in zein nanospheres while heparin was adsorbed on the surface, causing 97.1 ± 6.4 % release of heparin in 120 min (first-order kinetic model) and 62.7 ± 5.6 % release of Zn 2+ in 9 days (Korsmeyer-Peppas model). The mechanical analysis revealed that the functional polyurethane stents had about 8.61 MPa and 2.5 MPa tensile strength and bending strength, respectively. The in vitro biological analysis showed that the functional polyurethane stents had good EA.hy926 cells compatibility (97.9 ± 3.8 %), anti-coagulation response (comparable plasma protein, platelet adhesion and suppressed clotting) and sustained antibacterial activities by comparison with the bare polyurethane stent. The preliminary evaluation by rabbit ex vivo carotid artery intervention experiment demonstrated that the functional polyurethane stents could maintain blood circulation under the continuous stresses of blood flow. Meanwhile, the detailed data from the simulated implant infection experiment in vivo showed the functional polyurethane stents could effectively reduce microbial infection by 3–6 times lower and improve fibrosis and macrophage infiltration.

Secondary bond interface assembly of polyethyleneimine on zein microparticles for rapid adsorption of Reactive Black 5

Zaifei Zhuang,Xin Cheng,Liyan Cao,Guiqiang He,Jian Zhou,Yanxia Wei

Textile dye wastewater has the characteristics of high concentration, complex composition and changeable color degree and pH, which is difficult to be effectively and completely treated, and easy to cause environmental pollution. Here, a strategy of secondary bond interface assembly of polyethyleneimine on zein microparticles (PEI) ( [email protected] ) was constructed to achieve rapid and efficient removal of Reactive Black 5 (RB5), which is one of the most widely used reactive dyes in the textile industry. Structural analysis indicated that the as-prepared PEI layer immobilized on zein microparticles was constructed based on the interface assembly dominated by hydrophobic interactions and electrostatic attraction between PEI molecules and zein chains. The novel interface showed excellent absorption performance for RB5 with an absorption capacity of 631.0 mg·g -1 , rapid adsorption in 2 min, wide pH range of 4–10. Mechanism analysis suggested the effective adsorption of RB5 by [email protected] microparticles was mainly attributed to secondary bond interface such as electrostatic interaction and hydrogen bond between RB5 and PEI immobilized on the surface of zein microparticles. Moreover, due to the presence of secondary bond interface, RB5 adsorbed on microparticles can be easily desorbed by using 0.01 M NaOH. Therefore, the strategy of secondary bond interface assembly with polyethyleneimine on zein microparticles has high potential for practical application in the treatment of dye-containing wastewater.

Bimodal structure in electrospun Zein/PLA-chitosan films with enhanced filtration efficiency and low pressure drop

Mingfang Sun,Jun Wang,Jun Kang,Yaxin Zhao,Huafeng Tian,Yujuan Jin,Rakesh Kumar,Junfeng Wang

This study utilizes electrospinning to develop bio-based fibrous films composed of zein and polylactic acid (PLA), with chitosan added in varying concentrations. The resulting films exhibit remarkable filtration efficiency, capturing over 95 % of airborne pollutants, while maintaining a low pressure drop of less than 4 mbar. A key innovation in this work is the integration of chitosan. Chitosan, a naturally derived polysaccharide, significantly increases the conductivity and viscosity of the spinning solution, enhancing interactions among PLA, chitosan, and protein molecules. This leads to a robust network structure with improved hydrophobicity and a bimodal structure, both essential for maintaining high filtration efficiency without reducing breathability. The addition of chitosan effectively addresses the common issue of high pressure drop in PLA/zein composite films, improving the product's efficiency and practicality for air filtration applications. Furthermore, the inherent mechanical strength and ductility of chitosan contribute to the fibrous films' overall durability and functionality. These properties make chitosan-modified zein/PLA fibers highly suitable for air pollution control, especially in personal protective equipment. This research underscores the potential of chitosan-enhanced bio-based materials as sustainable and effective solutions for advanced air filtration technologies.
