The complex of oat β-glucan (OBG) and flavonoids hampered the digestion of starch-based food and retarded the blood glucose response; however, its effect on gastric emptying and its relative mechanism have not been thoroughly investigated. By using Fourier transform infrared (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM), antioxidant ability, and enzymic inhibitory tests for the characterization and in vitro semi-dynamic digestion of complexes of OBG (high and low molecular weights) and sea buckthorn flavonoids, we found that the higher molecular weight complex (FU) exhibited stronger ABTS and DPPH radical scavenging abilities and higher α-glucosidase and α-amylase inhibition rates. Mice fed with rice flour with FU addition exhibited the slowest gastric emptying and intestinal propulsion rates and blood glucose rise and had the lowest activity of digestive enzymes and levels of insulin, ghrelin, motilin (MTL), and relevant gene (ghrelin and GHSR mRNA) expression than those in the control and low-molecular-weight groups. This study provided scientific data for the development of foods with delayed gastric rate and hypoglycemic index for specific populations.

Strawberries ( Fragaria × ananassa ) are prized for bioactive flavonoids and anthocyanins, contributing to antioxidant capacity and health benefits. However, decades of breeding for yield and shelf-life have eroded their phytochemical diversity. To explore this trade-off, we compared wild strawberries ( Fragaria nilgerrensis , HM) with three cultivated varieties (pink-FY, red-RF, white-DX) using integrated LC-MS/MS metabolomics and transcriptomics. Wild HM strawberries contained twice the vitamin C (134.2 mg/100g), 2.5× more total flavonoids (138.7 mg/100g), and higher antioxidant activity (367 μg Trolox/g) than commercial varieties. In contrast, red-fruited RF showed abundant anthocyanins (pelargonidin-3-glucoside 6.6% abundance), while the wild HM genotype accumulated higher levels of flavonoids, with neohesperidin, lonicerin, hyperin and ternatumoside II collectively representing 20.9% of total flavonoids - concentrations 3-169 times greater than those observed in the three commercial varieties (FY, RF, DX). Transcriptomic analysis (RNA-seq, Illumina HiSeq 2500) revealed distinct MYB transcription factor associations: FxaYL_611g0111200 (MYB66) showed strong positive correlation with anthocyanin biosynthesis in RF (r=0.92, **p<0.005 ), while FxaYL_242g0472670 (MYB44-like) was significantly associated with HM's unique antioxidant flavonoid profile (r=0.96, ***p<0.0001 ). These regulatory relationships were corroborated by hierarchical clustering demonstrating varietal-specific expression patterns and principal component analysis (82.6% variance explained), collectively providing molecular targets for enhancing nutritional quality in strawberry breeding programs.

Background Rhododendron anthopogonoides Maxim, a kind of traditional Tibetan medicine, has been used to remove body heat, body detoxification, cough, asthma, stomachic and swelling, eliminate abundant phlegm and inflammatory for a long time. In the present study, the total phenols and total flavonoid contents as well as antioxidative properties of the crude extract and solvent fractions of R. anthopogonoides were determined using seven antioxidant assays. Additionally, the protective effect of the extracts on hypoxia-induced injury in PC12 cells was also investigated. Methods The content of total flavonoid and total phenolic was determined by the aluminum colorimetric method and Folin-Ciocalteu assay, respectively. In vitro antioxidant study, the effect of the crude extract and solvent fractions on total antioxidant activity, reducing power, DPPH radical scavenging, ABTS radical scavenging, superoxide radical scavenging, hydroxyl radical scavenging and nitric oxide radical scavenging were examined. The correlation between the phenolic and flavonoid content of the extracts and their antioxidant properties also analyzed. Furthermore, the protective effect of extracts on hypoxia-induced damage on PC12 cells was investigated by cell viability, lactate dehydrogenase (LDH) release, malondialdehyde (MDA) production and the activities of antioxidant enzymes. Results Our results showed that ethyl acetate and n-butanol fractions had higher content of phenolics and flavonoid compounds than other fractions. Except ABTS radical assay, n-butanol fraction exhibited the strongest antioxidant activity. While the hexane fraction showed the lowest antioxidant activity. Ethyl acetate also presented excellent antioxidant activity, which was just lower than n-butanol fraction. Significant correlation between the phenolic, flavonoid content of the extract and fractions with antioxidant assay excluding ABTS, OH scavenging assay was observed. Moreover, ethyl acetate and n-butanol fractions showed protective effect in PC12 cell under hypoxia condition, while crude extract and water fraction had no protective effect. In contrast, hexane fraction exhibited strong cytoprotective effect. Further study indicated that pretreatment of PC12 cells with ethyl acetate and n-butanol fractions, prior to hypoxia exposure, significantly increased the survival of cells and the activities of SOD, CAT, GSH-Px and T-AOC, as well as reduced the level of LDH and MDA. The gathered data demonstrated that ethyl acetate and n-butanol fractions were able to protect PC12 cells against hypoxia induced injury through direct free radical scavenging and modulation of endogenous antioxidant enzymes. Conclusion These findings suggested that ethyl acetate and n-butanol fractions of R. anthopogonoides had significant antioxidant activity and could prevent PC12 cells against hypoxia-induced injury. So it might be regarded as an excellent source of antioxidants and had great potential to explore as therapeutic agent for preventing hypoxia related sickness in future.

The flavonoid-rich extract fromPaulownia fortuneiflowers (EPF) has been reported to prevent obesity and other lipid metabolism disease. However, the mechanism of its protective effects is not yet clear. The objective of this study was to investigate molecular factors involved in the hypoglycemic and hypolipidemic effects of EPF in obese mice fed a high-fat diet (HFD). Male h ICR (Institute of Cancer Research) mice were fed a HFD containing or not containing the EPF (50 or 100 mg/kg) for eight weeks. EPF reduced body weight gain, lipid accumulation in livers and levels of lipid, glucose and insulin in plasma as well as reduced insulin resistance as compared with the HFD group. EPF significantly decreased serum aminotransferase activity of the HFD group. We observed that EPF administration significantly increased the level of AMP-activated kinase (AMPK) phosphorylation and prevented fat deposits in livers and HepG2 cells, but these effects were blocked by compound C (an AMPK inhibitor). The protective effects of EPF were probably associated with the decrease in HMGCR, SREBP-1c and FAS expressions and the increase in CPT1 and phosphor-IRS-1 expressions. Our results suggest that EPF might be a potential natural candidate for the treatment and/or prevention of overweight and hepatic and metabolic-related alterations induced by HFD.Keywords:Paulownia fortuneiflower;hyperlipidemia;hyperglycemia;obesity;hepatic fat accumulation;lipid metabolism;AMPK

Constructing a three-channel MnO 2 nanozyme array with multi-mechanism synergy. • Revealing structure/coordination-mediated radical scavenging and electron transfer. • Discriminating flavonoid isomers sensitively down to 1 μg/mL. • Identifying adulterated samples accurately from 0 to 100%. • Demonstrating proof-of-concept Citrus differentiation on a paper-based platform. The rapid differentiation of closely related Citrus plants remains a challenge due to their highly similar physicochemical profiles. This study developed a three-channel colorimetric sensor array based on morphologically distinct MnO 2 -I/II/III nanozymes with diverse oxidase-like activity. Synergistic free radical scavenging and electron transfer during catalysis induce specific absorbance changes of TMB/oxTMB (green, yellow, and blue) at 370, 450, and 652 nm, which may be synergistically mediated by flavonoid structures and coordination interactions. The multi-mechanism three-channel design provides a highly cross-reactive dataset on a paper-based platform, enabling HCA, PCA, and 3D LDA to: (i) discriminate flavonoid isomers (1–500 μg/mL), (ii) identify adulterated samples (0–100%), and (iii) achieve 100% accuracy on an independent test set for multi-batch Citrus samples. This work presents a mechanism-distinctive MnO 2 nanozyme array, which, coupled with chemometric analysis, serves as a promising proof-of-concept platform for point-of-care screening of structurally similar flavonoids and closely related plant samples. Simultaneous separation of structurally similar flavonoid glycosides remains challenging in functional food analysis. This study investigated the migration behavior of eight flavonoid glycosides by capillary zone electrophoresis with UV detection (CZE-UV). Phenolic deprotonation governed effective charge and electrophoretic mobility. However, strong pH dependence reduced robustness, prompting the exploration of less charge-dependent strategies. Deep eutectic solvents (DESs) provided limited improvement, whereas methyl-β-cyclodextrin (Me-β-CD) enabled stereoselective host-guest recognition and separated all eight analytes within 12 min under optimized conditions (5.0 mM Me-β-CD). The method showed good linearity and an LOD of approximately 0.3 μg/mL, supporting its use in quality control of purified moringa leaf extracts and related functional food materials. It quantified isoquercitrin and hyperoside as major flavonoid glycosides and detected two trace peaks tentatively assigned to spiraeoside and isovitexin with LC-MS/MS support. Chloromethane (dichloromethane, trichloromethane, and carbon tetrachloride), as a widely used organic solvent, has potential environmental hazards and health risks that cannot be ignored. Therefore, it is urgent to strictly monitor and take preventive measures for chloromethane. In this paper, we introduced triphenylamine groups onto the skeleton of flavonols and constructed a novel flavonoid skeleton fluorescent probe (FAC) that can be used to distinguish chloromethane. The fluorescence emission spectra of FAC in different proportions of chloromethane can exhibit a regular shift from yellow to blue, thereby achieving adjustability of fluorescence and distinguishing three chloromethane compounds by visual inspection. The detection mechanism was verified by single-crystal X-ray diffractometer calculation analysis. The test results show that FAC has the highest quantum yield in CH 2 Cl 2 , reaching 99.50 %. The quantum yields in CHCl 3 and CCl 4 were 86.87 % and 75.09 %, respectively. Moreover, based on the fluorescence performance of FAC, fluorescent wood (FAC-DBW) with detection capability was prepared. Covering FAC-DBW flakes on UV-LED chips, yellow fluorescent (540 nm), green fluorescent (508 nm), and blue fluorescent (420 nm) LEDs can be prepared when exposed to CH 2 Cl 2 , CHCl 3 , and CCl 4 vapors. In addition, FAC-DBW is biodegradable, and its design conforms to the concept of sustainable development.

Flavonoids are well known for their extensive health benefits. However, few studies compared the differences between flavonoid O-glycoside and C-glycoside. In this work, flavonoid O-glycoside (isoquercitrin), C-glycoside (orientin), and their aglycones (quercetin and luteolin) were chosen to compare their differences on antioxidant activities and metabolism during in vitro digestion and in vivo. In vitro digestion, the initial antioxidant activity of the two aglycones was very high; however, they both decreased more sharply than their glycosides in the intestinal phase. The glycosidic bond of flavonoid O-glycoside was broken in the gastric and intestinal stage, while the C-glycoside remained unchanged. In vivo, flavonoid O-glycoside in plasma was more elevated than C-glycoside on the antioxidant activity; however, flavonoid C-glycoside in urine was higher than O-glycoside. These results indicate that differences of flavonoid glycosides and their aglycones on antioxidant activity are closely related to their structural characteristics and metabolism in different samples. Aglycones possessed higher activity but unstable structures. On the contrary, the sugar substituents reduced the activity of flavonoids while improving their stability and helping to maintain antioxidant activities after digestion. Especially the C-glycoside was more stable because the stability of the C–C bond is higher than that of the C–O bond, which contributes to the difference between flavonoid O-glycoside and C-glycoside on the absorption and metabolism in vivo. This study provided a new perspective for comparing flavonoid O-glycoside, flavonoid C-glycoside, and their aglycones on their structure–activity relationship and metabolism.Keywords:flavonoid;O-glycoside;C-glycoside;aglycones;in vitro digestion;antioxidant activity

Triple functions: Cu 2+ coordination, CMCD inclusion and polarity switchable. • Selective for 4‑carbonyl-5-hydroxy flavonoids; baicalin yield 160 mg/g. • Higher extraction yield than conventional methods/organic solvents. • Mechanism clarified by DFT/docking/IGMH. To address the inherent limitations of conventional deep eutectic solvents, such as poor selectivity and tedious separation procedures, this study prepared a novel Cu 2+ -functionalized metal-supramolecular deep eutectic solvent (MSDES) for the efficient and highly selective extraction of flavonoids from Scutellaria baicalensis Georgi. The MSDES integrates three synergistic mechanisms: Cu 2+ coordination for specific molecular recognition, carboxymethyl-β-cyclodextrin host-guest inclusion for enhanced compatibility, and the combination of decanoic acid with N , N -dimethylcyclohexylamine for polarity-switchable phase separation. This design not only improves extraction yield but also simplifies recovery, eliminating the need for distillation. Under the optimized conditions, the extraction yield of baicalin reached 160 mg/g, which was significantly higher than those obtained with conventional DESs and organic solvents. The system was characterized by UV–Vis, 1 H NMR and FT-IR spectroscopies, and the extraction mechanism was elucidated by DFT calculations and molecular docking. This study addresses the critical issue of insufficient molecular recognition precision in conventional DESs, providing an efficient novel strategy for the extraction of bioactive components from traditional Chinese medicinal materials. Increasing demand for detecting environmental and biological analytes in aqueous solutions based on fluorescent probes has raised the bar for the fluorophores used. Especially, the twisted intramolecular charge transfer (TICT) state of fluorophore can trigger the formation of a charge-separated non-emissive specie, leading to a low fluorescence brightness. In this work, we performed a simple donor engineering on the flavonoid-based fluorophores to regulate the TICT formation. A systematic study on relationship between donor structure and photophysical properties has been carried out. Among a series of flavonoids, the methylpiperazine-substituted one exhibited the strongest fluorescence brightness in water since the methylpiperazine could rigidify the rotation of donor moiety to suppress TICT state. Moreover, a novel fluorescent probe based on this flavonoid successfully realized the cysteine detection both in water and in living cells. This work confirmed the validity of design strategy by inhibiting TICT to enhance the fluorescence brightness, and also provided a practical flavonoid-based fluorophore for further bioimaging applications.

There are both soluble and insoluble-bound forms of phenolics in tea-leaf products. In order to increase total soluble phenolics contents, guava leaves tea (GLT) was first fermented with Monascus anka and Saccharomyces cerevisiae , and then hydrolyzed with complex enzymes . The changes in phenolics profiles, antioxidant activities and inhibitory effect on α -glucosidase in processed GLT were investigated. Compared with the un-fermented GLT, fermentation and complex enzymatic processing (FE) significantly increased the total phenolics, total flavonoids, quercetin and kaempferol contents by 2.1, 2.0, 13.0 and 6.8 times, respectively. After the FE, a major proportion of phenolics existed in the soluble form. Quercetin was released in the highest amount among different phenolics. In addition, soluble phenolic extracts from GLT following FE exhibited a highest antioxidant activity and inhibitory effect on α -glucosidase. The paper suggested an improved method for processing GLT into high-value products rich in phenolics and flavonoids aglycones with enhanced health benefits.

Context: Flavonoid-enriched extract from Hippophae rhamnoides L. (Elaeagnaceae) seed (FSH) has shown beneficial effects in anti-hypertension and lowering cholesterol level. However, evidence for its efficacy in treating obesity is limited. Objective: We sought to determine if FSH can reduce body weight and regulate lipid metabolism disorder in high fat diet (HFD)-induced obese mouse model, and to investigate potential molecular targets involved.Materials and methods: C57BL/6 mice were fed with HFD for 8 weeks to induce obesity. The modeled mice were divided into four groups and treated with vehicle, rosiglitazone (2 mg/kg), low (100 mg/kg) and high (300 mg/kg) dose of FSH, respectively. Normal control was also used. The treatments were administered orally for 9 weeks. We measured the effect of FSH on regulating body weight, various liver and serum parameters, and molecular targets that are key to lipid metabolism. Results: FSH administration at 100 and 300 mg/kg significantly reduced body weight gain by 33.06 and 43.51%, respectively. Additionally, triglyceride concentration in serum and liver were decreased by 15.67 and 49.56%, individually, after FSH (300 mg/kg) treatment. Upon FSH (100 and 300 mg/kg) treatment, PPARα mRNA expression was upregulated in liver (1.24- and 1.42-fold) and in adipose tissue (1.66- and 1.72-fold). Furthermore, FSH downregulated PPARγ protein level both in liver and adipose tissue. Moreover, FSH inhibited macrophage infiltration into adipose tissues, and downregulated TNFα mRNA expression in adipose tissue (38.01–47.70%).Conclusion: This effect was mediated via regulation of PPARγ and PPARα gene expression, and suppression of adipose tissue inflammation.
