Dual pH- and magnetic-responsive Fe3O4@ZIF-8 core–shell nanoparticles for ultra-fast adsorption and desorption of histidine-rich proteins

Ma Kuanjie,Li Nana,Lu Guochao,Zhang Lianwei,Hu Zhiwei,Liu Wenjing,Cai Yurong

To address the need for rapid desorption in protein adsorption applications, we developed and characterized magnetic core–shell nanoparticles composed of a zeolite imidazolate framework-8 (ZIF-8) coating on magnetite (Fe 3 O 4 ) cores (Fe 3 O 4 @ZIF-8). This nanostructure integrates the pH-sensitive properties of ZIF-8 with the magnetic responsiveness of Fe 3 O 4 , enabling rapid separation within one minute via an external magnetic field. In the synthesis of Fe 3 O 4 @ZIF-8, the particle size of the ZIF-8 shell was found to increase concurrently with the molar ratio of zinc nitrate hexahydrate to 2-methylimidazole. The resulting shells were uniformly distributed on the surface of the Fe 3 O 4 cores. Furthermore, a positive correlation was observed between the ZIF-8 particle size and the adsorption capacity for histidine-rich proteins. In adsorption experiments using bovine hemoglobin (BHB) as a model protein, Fe 3 O 4 @ZIF-8 achieved an adsorption capacity of 1013.5 mg/g within 3 min, representing 87% of its maximum adsorption potential. Saturation adsorption, with a capacity of 1164.3 mg/g, was reached within 8 min. Importantly, the pH-responsive nature of the ZIF-8 component facilitated highly efficient desorption. A high release efficiency of 90% was achieved in just 1 min using a phosphate buffer at pH 5.2. This dual pH- and magnetic-responsive system demonstrates significant potential for applications requiring fast and controllable protein adsorption and desorption, positioning it as an ideal tool for targeted protein recovery in various biochemical and industrial processes.

Effect of l-histidine on the heat-induced aggregation of bighead carp (Aristichthys nobilis) myosin in low/high ionic strength solution

Ruichang Gao,Yanmin Wang,Jianlou Mu,Tong Shi,Li Yuan

In this study, the effects of l -histidine (L-His) on the aggregation of bighead carp ( Aristichthys nobilis ) myosin induced by heating in low (0.1) or high (0.5) ionic strength solutions were studied. The solubility of natural myosin was significantly enhanced by L-His both in low and high ionic strength solutions. More small particles are suspended in the heated solution containing L-His, inducing a higher solubility than that solution without L-His. The surface hydrophobicity of myosin in a low ionic strength solution containing L-His is higher than that of myosin without L-His. The results of morphology show that L-His induced myosin to form finer aggregates and a honeycomb-like network in a low/high ionic strength solution during heat treatment. These results demonstrate that L-His might disrupt the electrostatic property of unfolded myosin heat-induced through electrostatic binding with the exposed negatively charged amino acid residues, which might disarrange hydrophobic surfaces, resulting in weakening hydrophobic interactions, which ultimately suppresses the fierce aggregation of carp myosin.

Carboxymethyl-β-cyclodextrin and histidine-zeolitic imidazolate framework-8 used for enantioseparation of three basic drugs in open-tubular capillary electrochromatography

Pandeng Miao,Jie Gan,Jian Zhang,Mingxuan Ma,Xiaoqi Li,Yingxiang Du,Zijie Feng,Liu Zhang

Metal organic frameworks (MOFs) have drawn broad attention as a novel stationary phase due to their highly porous structure, modifiable pores, large specific surface areas, and satisfactory stability. In this paper, histidine-zeolitic imidazolate framework-8 (His-ZIF-8) synthesized at room temperature was physically coated to the internal surface of the capillary column and the carboxymethyl-β-cyclodextrin (CM-β-CD) as the chiral selector was chemically bonded to the His-ZIF-8@capillary column. The prepared CM-β-CD@His-ZIF-8@capillary column was used for the enantioseparation of amlodipine, propranolol, and atenolol in capillary electrochromatography. In contrast to the CM-β-CD@capillary column without His-ZIF-8, the CM-β-CD@His-ZIF-8@capillary column reveals significantly improved enantiodiscrimination performance for amlodipine ( R s : 0 → 2.29), propranolol ( R s : 0 → 1.69), and atenolol ( R s : 0 → 0.79). His-ZIF-8 concentration, buffer pH, buffer concentration, and the proportion of organic modifier were evaluated in detail with enantiomerically separating chiral molecules. The repeatability of intraday, day-to-day, and column-to-column have been discussed; the result was preferable, and the relative standard deviation (RSD) of separation parameters was <6.7%.

pH-Responsive Hexa-Histidine Metal Assembly (HmA) with Enhanced Biocatalytic Cascades as the Vehicle for Glucose-Mediated Long-Acting Insulin Delivery

Sijie Zhou,Ruhui Yang,Xiaoling Xie,Liwen Wang,Shengwu Zheng,Na Li,Sicheng Tang,Xingjie Zan

Diabetes has been listed as one of the three major diseases that endanger human health. Accurately injecting insulin (Ins) depending on the level of blood glucose (LBG) is the standard treatment, especially controlling LBG in the long-term by a single injection. Herein, the pH-responsive hexa-histidine metal assembly (HmA) encapsulated with enzymes (GOx and CAT) and Ins (HmA@GCI) is engineered as the vehicle for glucose-mediated insulin delivery. HmA not only shows high proteins loading efficiency, but also well retained proteins activity and protect proteins from protease damage. Within HmA, the biocatalytic activities of enzymes and the efficiency of the cascade reaction between GOx and CAT are enhanced, leading to a super response to the change of LBG with insulin release and efficient clearance of harmful byproducts of GOx (H 2 O 2 ). In the treatment of diabetic mice, HmA@GCI reduces LBG to normal in half an hour and maintains for more than 5 days by a single subcutaneous injection, and nearly 24 days with four consecutive injections. During the test period, no symptoms of hypoglycemia and toxicity to tissues and organs are observed. These results indicate that HmA@GCI is a safe and long-acting hypoglycemic agent with prospective clinical application.

Self-assembly of MOFs-like materials by Zn2+ and histidine functionalized graphene quantum dots for the efficient removal of VOCs simulants

Youliang Cheng,Yue Zhang,Jiaxian Li,Changqing Fang,Jing Chen,Jiarui Zhao,Tong Zhao,Jia Zhu

Volatile organic compounds (VOCs) in the field of printing and packaging, have caused great harm to the environment. Therefore, the removal of volatile organic compounds (VOCs) is a hot topic, and metal-organic frameworks (MOFs) and graphene quantum dots (GQDs) have been applied in this field. However, improving the removal efficiency of different VOCs by using the two kinds of materials is still a great challenge. Herein, ZIF-8 like materials of Zn-His-GQDs were prepared by assembling Zn 2+ and histidine functionalized GQDs (His-GQDs) in this work. The morphologies, structure and adsorption performances of Zn-His-GQDs were investigated. These results showed that mesoporous Zn-His-GQDs with a specific surface area of 63.3 m 2 ·g −1 had similar structure to ZIF-8. When the content of Zn-His-GQDs was 200 mg, the equilibrium adsorption capacity for toluene solution of 500 mg·L −1 and ethyl acetate solution of 1600 mg·L −1 was 350 mg·L −1 and 669.44 mg·L −1 , respectively. Due to channel filling, cation-π interaction, π-π interaction, hydrogen bond and high surface energy, Zn-His-GQDs exhibited a relative high removal efficiency for high concentration of toluene and ethyl acetate. The removal rate of Zn-His-GQDs on toluene and ethyl acetate after five cycles remained 59.80 % and 36.75 %, respectively. Moreover, this kind of MOFs like materials maintained stable structure during the adsorption in different pH solutions. Therefore, as-prepared Zn-His-GQDs with excellent adsorption performances for typical VOCs simulants in this work have potential application in the treatment of VOCs in the field of printing and packaging.

Synthesis of micron-sized magnetic agarose beads chelated with nickel ions towards the affinity-based separation of histidine-tagged/rich proteins

Ya-Qi Zhao,Shi-Song Yu,Meng-Ying Chen,Yuan Wang,Yu-Jun Shi,Xin-Yu Wang,Jia-Meng Zhao,Lin-Yi Dong,Zhen-Yu Zhao,Xian-Hua Wang

Developing high-performance magnetic particles for the effective separation and purification of target proteins has become an important topic in the area of biomedical research. In this work, a simple and novel strategy was proposed for fabricating magnetic Fe3O4@agarose-iminodiacetic acid-Ni microspheres (MAIN), which can efficiently and selectively isolate histidine-tagged/rich proteins (His-proteins). Based on the thermoreversible sol-gel transition of agarose , basic magnetic agarose microspheres were prepared through the inverse emulsion method, in which the emulsion contained agarose and amine-modified Fe 3 O 4 nanoparticles. The size of the emulsion was controlled by the emulsification of a high-speed shear machine, which improved the specific surface area of MAIN. Subsequently, the amine-modified Fe 3 O 4 nanoparticles were covalently crosslinked with agarose through epichlorohydrin, which could avoid leakage of the magnetic source during use and increase the stability of MAIN. The microsized MAIN exhibited a clearly visible spherical core-shell structure with a diameter range from 3.4 μm to 9.8 μm, and excellent suspension ability in aqueous solution. The maximum adsorption capacity of MAIN for histidine-rich bovine hemoglobin was 1069.2 mg g −1 at 35 °C, which was higher than those of commercialized and most reported magnetic agarose microspheres/nanoparticles. The MAIN showed excellent adsorption ability and selectivity toward His-proteins in a mixture of histidine-rich bovine serum albumin (BSA) and histidine-poor lysozyme (LYZ). When the amount of LYZ was 5-fold higher than that of BSA, the recovery of BSA reached 75.0%. To prove its practicability, MAIN was successfully employed for the enrichment of histidine-tagged RSV-F0 from the cell culture medium supernatant. According to the optimized conditions, MAIN could enrich approximately 0.1 mg of RSV-F0 from 1 mL of complex biological sample. Therefore, we believe that the novel MAIN could be applicable for efficient separation and purification of His-proteins from complex biological systems.

Rapid in situ analysis of l-histidine and α-lactose in dietary supplements by fingerprint peaks using terahertz frequency-domain spectroscopy

Yongmei Wang,Zongshan Zhao,Jianyuan Qin,Huan Liu,Aifeng Liu,Mengmeng Xu

A simple, green and nondestructive method based on terahertz fingerprint peaks has been developed for rapid in situ analysis of l -histidine and α-lactose in dietary supplements. Fingerprint absorption peaks of l -histidine and α-lactose located at 0.77 and 0.53 THz could be directly used for identification and quantitation of these analytes in commercial dietary supplements. Compared with the partial least squares regression model (PLSR), the linear least squares regression (LLSR) method based on peak areas presented better performance, with the linear correlation coefficients of 0.9899 and 0.9910 for l -histidine and α-lactose, respectively. Furthermore, analysis time per sample can be shortened to less than 1 min due to the narrower spectral acquisition region. The accuracies were 94.8–110% and 98.9–110%, comparable to those of ion chromatography for l -histidine and high-performance liquid chromatography for α-lactose. The results presented great potential of the developed method for rapid in situ analysis of nutrients in dietary supplements.

Separation of dithiothreitol and dithioerythritol by an open tubular capillary electrochromatographic column with a MOF modified by histidine as stationary phase

Mo Chen,lidi Gao,Huiying Liu,Jiawen Yu,Fuquan Zhao,Ming Li Bai,Hongtao Chu,Ming Zhao,Shili Qin

A separation and determination method of dithiothreitol (DTT), as an important small molecule reducing agent, and dithioerythritol (DTE), as a C3-epimer of DTT, was developed by capillary electrochromatography. A functional MOF (His-ZIF-93) was first prepared by using ZIF-93 (synthesized via coordination between 4-methylimidazole-5-carbaldehyde and Zn2+) as the framework and D-histidine (D-His) as a modifier, and then was bonded to the inner wall of the capillary column to obtain His-ZIF-93 column by Schiff base reaction. The results showed that the baseline separation of DTT and DTE was easily obtained under a very wide experimental conditions with a extremely short analysis time with less than 3 min. Separation mechanism was proved by the binding constant experiments. There was a good linearity between concentration (0.10-5.0 mg/mL) and peak area of DTT and DTE (R2 ≥ 0.996), and the relative standard deviations for the peak area and the retention time were less than 0.93%. The average impurity content of DTE in a 5.0 mg/mL DTT were 0.79%. Thus, the method was able to realize for the simple and fast separation and precise quantification of DTT and DTE.

Mechanistic insights into PS80 oxidative degradation in glass vials: Roles of critical factors (iron, histidine, aluminum, zinc) and their synergistic interactions

Zhuan Cheng,Pengzhen Wang,Xu Gao,Luting Liu,Quanmin Chen,Jeremy Guo

Polysorbate 80 (PS80), a vital stabilizer in biotherapeutic formulations, faces persistent oxidative degradation challenges that threaten drug product stability during storage. While PS80 instability has been studied for decades, the oxidation mechanisms in specific formulation-packaging systems remain poorly understood. This work resolves this knowledge gap by demonstrating that PS80 oxidative degradation occurs exclusively in histidine-buffered formulations stored in glass vials. This phenomenon was found to be driven by four factors: iron (leaching from vials promoted by histidine), histidine, zinc (histidine-introduced) and aluminum (glass-derived). These four interdependent factors exhibited a unique functional synergy: 1) iron as the radical chain initiator; 2) histidine as a pro-oxidative cofactor lowering iron’s catalytic threshold via formation of an iron-histidine complex; 3) zinc and aluminum accelerate Fe 2+ /Fe 3+ redox cycling through distinct potential-specific mechanisms, enhancing the overall catalytic efficiency. This study for the first time provides the systematic evidence of the histidine-glass system as an essential prerequisite for PS80 oxidation and reveals novel mechanistic insights on the interplay between formulation composition and packaging-derived contaminants. The findings establish a predictive framework to mitigate PS80 oxidative degradation, ultimately enhancing the stability and safety of protein therapeutics.

H-Shaped copolymer ((His)PHEMA)2-b-PEG-b-(PHEMA(His))2 with histidine modified: Synthesis, characterization and its drug release study

Sa Zongpeng,Li Yapeng,Zhao Haochen,Hu Qi,Wei Jia,Wang Jingyuan

Through H-shaped macroinitiator glycol-chlorine((Cl) 2 -PEG-(Cl) 2 ) to initiate the ATRP reaction of 2-Hydroxyethyl Methacrylate (HEMA) we synthesized H-shaped copolymer (PHEMA) 2 - b -PEG- b -(PHEMA) 2 . After we modified the copolymer with histidine (His), the self-assembling and drug release behavior was studied. The morphology of the copolymer micelles and drug loaded micelles were examined by transmission electron microscope (TEM). Spherical micelles and core-shell structure with a diameter of 165 nm were obtained (at pH = 7.0). Due to the presence of imidazole-ring in histidine the copolymer show commendable pH-sensitivity, which made the copolymer potential application as drug carrier exhibiting good control-released property.

Fabrication and adsorption performance evaluation of flower-like Fe3O4@NiO core-shell nanospheres for efficient separation and purification of histidine-tagged proteins

Mochou Gao,Haoxuan Mi,Haocheng Xu,Haowen Ren,Congkai Xu,Zhihao Yang,Chenglin Xu,Haotian Li,Lu Sun,Yanbin Yin,Yanbao Zhao,Wensheng Yang,Xueyan Zou

Hierarchical Fe 3 O 4 @NiO core-shell nanospheres are synthesized. • High adsorption capacity of 65.83 mg/g for His-tagged proteins is obtained. • Fe 3 O 4 @NiO NSs exhibit rapid magnetic separation and good reusability (>80% after 5 cycles). • The adsorption follows pseudo-second-order kinetics and Langmuir isotherm model. Magnetic nanomaterials are highly efficient for the separation and purification of histidine-tagged (denoted as His-tagged) proteins. This article reports the facile synthesis of flower-like magnetic Fe 3 O 4 @NiO core-shell nanospheres (NSs) for that purpose. The synthesis involved the initial solvothermal preparation of Fe 3 O 4 NSs, followed by their controlled reaction with nickel chloride under alkaline condition to in-situ deposit a hierarchical NiO nanosphere shell. The unique hierarchical flower-like architecture could provide abundant accessible Ni 2+ coordination sites for protein binding. When isolating the target protein from cell lysate, Fe 3 O 4 @NiO NSs exhibited a high adsorption capacity of 65.83 mg/g toward His-tagged green fluorescent protein with an equilibrium time of 60 min, together with the merit of fast magnetic separation. Systematic adsorption studies revealed that the adsorption process of the target proteins by Fe 3 O 4 @NiO NSs followed pseudo-second-order kinetics and fitted the Langmuir isotherm model, referring to a chemisorption-dominated monolayer adsorption mechanism. Furthermore, the Fe 3 O 4 @NiO nano-adsorbent exhibited excellent stability and reusability, retaining over 80% of initial separation efficiency after five consecutive adsorption-desorption cycles. Therefore, the hierarchical Fe 3 O 4 @NiO NSs, as a kind of high-performance magnetically recoverable nano-adsorbent, could have significant potential for downstream bioseparation and protein purification applications.

A versatile logic-gated fluorescence turn-on nitrogen-doped carbon dots: STM32 microcontroller intelligent platform for assay of L-Histidine and Al3+

Yong-qing Liu,Bo-wen An,Tao-Zhu,Rui-yong Jiang,Ye-Wang,Chan Huang,Su-qian Cai,Bi-xiao Zhang,Yan-li Leng,Jing-zhen Pan,Le Liang,Xiao-hua Cai

Rapid and intelligent detection of bioactive molecules and metal ions is vital for health and environmental safety. Herein, a dual-functional nitrogen-doped carbon dots (N-CDs) for L -Histidine ( L -His) and Al 3+ detection was developed employing chitosan (CTS) and umbelliferone (UMB) as the substrates. The N-CDs exhibited a distinct "turn-on" fluorescence response to L -His and a subsequent signal recovery towards Al 3+ with low detection limits (LODs) of 0.950 μM and 4.393 μM. The underlying sensing mechanisms were rigorously elucidated via density functional theory (DFT) calculations and independent gradient model based on Hirshfeld partition (IGMH) analyses, highlighting the pivotal role of surface functional groups. The practical applicants of N-CDs were demonstrated through the fabrication of fluorescent hydrogels, solid-state films for light-emitting diodes (LEDs), and anti-counterfeiting patterns. Notably, a portable and intelligent sensing platform driven by STM32 microcontroller was engineered to capture and analyze RGB signals to realize smart and on-site quantification of L -His, which provided a robust and cost-effective strategy for real-time monitoring of analytes in complex matrices.
