Effects of calcium ions on the particle performance of luteolin-loaded zein-gum arabic-tea polyphenols ternary complex nanoparticles

Renjie Li,Jin Chen,Huihui Li,Zhuangwei Zhang,Hongjin Tang

Zein-gum arabic-tea polyphenols ternary complex nanoparticles have been developed for the encapsulation of luteolin, but there are still shortages existed in their particle properties such as encapsulation efficiency, stability, and release ability of luteolin. To improve their performance, calcium ions (Ca 2+ ) were employed as a cross-linking stabilizer to investigate their effects on particle properties and functional performance. Particle parameters, physicochemical stability, antioxidant activity, and in vitro simulated gastrointestinal digestion were evaluated based on zetasizer measurement and multi-spectral methods, which were analyzed using Graphpad Prism v6.0.2. As a result, low concentrations of Ca 2+ induced the formation of smaller and more compact nanoparticles with improved luteolin encapsulation efficiency, while higher concentrations had the opposite effect. Hydrophobic, electrostatic, and hydrogen bonding interactions were found to be responsible for nanoparticle assembly and stability with Ca 2+ incorporation. Appropriate amounts of Ca 2+ promoted the physicochemical stability of the nanoparticles under different conditions such as pH, salt ion concentration, temperature, and storage time. Additionally, low levels of Ca 2+ strengthened the antioxidant activity of the nanoparticles and improved sustained release of luteolin under in vitro gastrointestinal conditions. These findings suggest that zein-gum arabic-tea polyphenols nanoparticles assembled with Ca 2+ have potential applications as a delivery system for bioactive substances.

Adding genistein or luteolin decreased the yield of citrinin and without reducing pigments in yam solid-fermentation by Monascus

Haiyun Yang,Zhiqing Peng,Liuming Xie,Jianhua Xie,Zhibing Huang

BACKGROUND Chinese yam fermented by Monascus , namely red mold dioscorea (RMD), has the potential of treating diseases. However, the production of citrinin limits the application of RMD. In the present study, the fermentation process of Monascus was optimized by adding genistein or luteolin to reduce citrinin yield. RESULTS The results showed that citrinin in 25 g of Huai Shan yam was reduced by 48% and 72% without affecting the pigment yield by adding 0.2 g of luteolin or genistein, respectively, to a 250-mL conical flask after fermentation for 18 days at 28 °C, whereas the addition of luteolin increased the content of yellow pigment by 1.3-fold. Under optimal conditions, citrinin in 20 g of iron bar yam decreased by 55% and 74% after adding 0.2 g of luteolin or genistein. Luteolin also increased yellow pigment content by 1.2-fold. Ultra HPLC coupled to quadrupole time-of-flight mass spectrometry was used for the preliminary analysis of Monascus fermentation products. It was found that the amino acid types in RMD are similar to those in yams, but there are fewer polysaccharides and fatty acids. CONCLUSION The results obtained in the present study showed that the addition of genistein or luteolin could reduce citrinin on the premise of increasing pigment yield, which laid a foundation for the better use of yams in Monascus fermentation. © 2023 Society of Chemical Industry.

Sodium hyaluronate-stabilized luteolin-loaded microemulsion gel: Interface-driven nanocarrier for enhanced skin delivery and therapeutic efficacy on atopic dermatitis

Cancan Huang,Liyang Guan,Yifan Wang,Ansar Javeed,Lei Jin,Bingnan Han

Luteolin (LUT), a promising phytochemical for atopic dermatitis (AD) therapy, is limited by poor solubility and skin permeability in topical application. To address this, a novel LUT-loaded microemulsion-based (LUT-ME) gel was developed to facilitate drug delivery. The optimal LUT-ME formulation, obtained by a pseudo-ternary phase diagram and D -optimal mixture design, comprised 7.23 % glycerol triacetate, 34.56 % Smix, and 58.21 % water. DLS analysis showed a mean droplet size of 28.77 ± 0.31 nm and a PDI of 0.31 ± 0.04, while the zeta potential was −2.95 ± 0.68 mV and the drug loading reached 7.28 ± 0.05 mg/mL. Characterization by TEM, XRD, and FTIR demonstrated uniform droplet morphology, amorphous LUT dispersion, and good drug-excipient chemical compatibility. Notably, LUT-ME more significantly suppressed reactive oxygen species (ROS) and pro-inflammatory mediators in LPS-stimulated RAW264.7 cells compared to free LUT. Incorporation of sodium hyaluronate (SH) yielded a shear-thinning gel with favorable spreadability, good stability, and negligible skin irritation. Moreover, enhanced skin permeation of LUT-ME gel (2.09-fold vs LUT gel) in ex vivo studies contributed to its superior anti-inflammatory and histopathological effects in an AD mouse model. These results indicate that LUT-ME gel markedly improves the topical therapeutic efficacy of LUT against AD.

β-Cyclodextrin metal-organic framework as a green carrier to improve the dissolution, bioavailability, and liver protective effect of luteolin

Dan Yang,Min Zhao,Yihe Huang,Liwen Chen,Jiqin Fang,Jiaonan Liu,Miao Wang,Chunjie Zhao

The incidence of acetaminophen-induced liver injury has increased, but effective prevention methods are limited. Although luteolin has hepatoprotective activity, its low solubility and bioavailability limit its applications. Cyclodextrin metal-organic frameworks (CD-MOFs) possess 3D-network structures and large inner cavities, which make them excellent carriers of poorly soluble drugs. In this study, we used CD-MOFs as carriers to improve the dissolution of luteolin and assessed their antioxidant activity, bioavailability, and hepatoprotective effects. Luteolin was loaded into β-CD-MOF, γ-CD-MOF, β-CD, and γ-CD, and characterized by powder X-ray diffractometry (PXRD) and thermogravimetric analysis (TGA). Our results showed that luteolin-β-CD-MOF was the most stable. The main driving forces were hydrogen bonds and van der Waals forces, as determined by molecular simulation. The loading capacity of luteolin-β-CD-MOF was 14.67 wt%. Compared to raw luteolin, luteolin-β-CD-MOF exhibited a 4.50-fold increase in dissolution and increased antioxidant activity in vitro . Luteolin-β-CD-MOF increased the bioavailability of luteolin by approximately 4.04- and 11.07-fold in healthy rats and liver injured rats induced by acetaminophen in vivo , respectively. As determined by biochemical analysis, luteolin-β-CD-MOF exhibited a better hepatoprotective effect than raw luteolin in rats with acetaminophen-induced liver injury. This study provides a new approach for preventing acetaminophen-mediated liver damage.

Electrochemical biosensor based on functional nanomaterials and horseradish peroxidase for the determination of luteolin in peanut shell, honeysuckle and perilla

Yu-Jie Chen,Yu-Ying Li,Bao-Lin Xiao,Lin-Lin Ma,Ke-Xin Xu,Sanad Abdalbage Mohammed Abdalsadeg,Tao Hong,Ali Akbar Moosavi-Movahedi,Reza Yousefi,Yan-Na Ning,Jun Hong

Constructing a biosensor to detect luteolin content accurately is essential, especially considering its specific health benefits at certain concentrations. In this work, the reaction of HRP catalyzed luteolin could be successfully applied in electrocatalytic processes, the oxidation process of electron loss and dehydrogenation occurring on the electrode replaced the hydrogen receptor role of H 2 O 2 in the HRP biocatalytic process. This oxidation reaction had an apparent current response, thus achieving accurate measurement of luteolin. On this biosensor, CTAB was used to disperse MWCNTs, and BSA was used to improve the hydrophobicity of MWCNTs, which was conducive to the subsequent AuNPs fixation of HRP. Three detection methods (LSV, DPV and SWV) for the detection of luteolin were compared and showed that SWV method had a wider linear range (1 × 10 −8 –2 × 10 −5  M) and lower detection limit (8 × 10 −10  M). The determination of luteolin in Traditional Chinese Medicine (TCM) by high performance liquid chromatography (HPLC) and biosensor was almost identical. Therefore, this biosensor could successfully replace HPLC in detecting luteolin in TCM.

Porous starch microspheres loaded with luteolin exhibit hypoglycemic activities and alter gut microbial communities in type 2 diabetes mellitus mice

Xiaodong Ge,Tingting Liu,Yaolin Wang,Huanhuan Wen,Zirui Huang,Ligen Chen,Jianda Xu,Hongcheng Zhou,Qin Wu,Chao Zhao,Rong Shao,Wei Xu

Luteolin (LUT), a natural flavonoid known for its hypoglycemic properties, is primarily sourced from vegetables such as celery and broccoli. However, its poor stability and low bioavailability in the upper digestive tract hinder its application in the functional food industry. To address these challenges, this study employed porous starch (PS) as a carrier to develop PS microspheres loaded with luteolin (PSLUT), simulating its release in vitro. The research assessed the hypoglycemic effects of LUT in type 2 diabetes mellitus (T2DM) mice both before and after PS treatment. In vitro findings demonstrated that PS improved LUT's stability in simulated gastric fluids and enhanced its in vivo bioavailability, aligning with experimental outcomes. PSLUT administration significantly improved body weight, fasting blood glucose (FBG), oral glucose tolerance test (OGTT), pancreatic islet function, and other relevant indicators in T2DM mice. Moreover, PSLUT alleviated abnormal liver biochemical indicators and liver tissue injury caused by T2DM. The underlying hypoglycemic mechanism of PSLUT is thought to involve the regulation of protein kinase B (AKT-1) and glucose transporter 2 (GLUT-2). After four weeks of intervention, various PSLUT doses significantly reduced the Firmicutes to Bacteroidetes ratio at the phylum level and decreased the relative abundance of harmful bacteria at the genus level, including Acetatifactor, Candidatus-Arthromitus, and Turicibacter. This microbial shift was associated with improvements in hyperglycemia-related indicators such as FBG, the area under the curve (AUC) of OGTT, and homeostasis model assessment of insulin resistance (HOMA-IR), which are closely linked to these bacterial genera. Additionally, Lachnoclostridium, Parasutterella, Turicibacter, and Papillibacter were identified as key intestinal marker genera involved in T2DM progression through Spearman correlation analysis. In conclusion, PS enhanced LUT's hypoglycemic efficacy by modulating the transcription and protein expression levels of AKT-1 and GLUT-2, as well as the relative abundance of potential gut pathogens in T2DM mice. These results provide a theoretical foundation for advancing luteolin's application in the functional food industry and further investigating its hypoglycemic potential.

Double affinity based molecularly imprinted polymers for selective extraction of luteolin: A combination of synergistic metal chelating and boronate affinity

Bailin Guo,Yukui Tong,Baoyue Zhang,Miaomiao Tian

This study addresses opportunities for large-scale extraction of health-beneficial compounds from agricultural waste peanut shell samples. We focus on preparing a novel metal-chelating and boronate-affinity double affinity molecular imprinted polymers (MB-DMIPs) adsorbent and developing a new method to resolve current problems in enrichment of luteolin (LTL) from peanut shell samples. Boronate-affinity technique can produce a synergistic effect with metal oxide to effectively improve the adsorption capacity for LTL. Compared to the single affinity molecular imprinted polymers (M-SMIPs and B-SMIPs), the MB-DMIPs possessed stronger specificity, higher sensitivity, and better efficiency. The enrichment capacities and selectivities for LTL with the MB-DMIPs were studied in detail, after enriched, LTL was detected by HPLC. Under the optimal conditions, the maximum adsorption capacity reaches 12.42 mg g −1 , and the imprinted factor is 2.89. The detection limit is 0.021 mg L −1 , quantitative limit is 0.067 mg L −1 , and the recoveries are between 85.5 and 103.1% (the relative standard deviations are less than 3.2%, n  = 3). This work will provide a new research scheme for extraction and purification of bioactive components from agricultural wastes.

Construction of an electrochemical sensor with graphene aerogel doped with ZrO2 nanoparticles and chitosan for the selective detection of luteolin

Hou Xiudan,Wu Wei,Zhao Fangyuan,Xie Wancui,Yang Qingli

A simple, fast and sensitive method for the detection of luteolin is proposed based on the chitosan/reduced graphene oxide aerogel with dispersed ZrO 2 nanoparticles modified glassy carbon electrode (ZrO 2 /CS/rGOA-GCE) as an electrochemical sensor. The ZrO 2 /CS/rGOA composite was prepared by one pot synthesis from a mixture of GO, CS and zirconyl chloride octahydrate, and subsequently be freeze-dried. Scanning electron microscope images showed a typical thin, wrinkled and fluctuant morphology of graphene nanosheets and the polymerized CS and ZrO 2 nanoparticles deposited on the surface of rGOA. Cyclic voltammetry and differential pulse voltammetry were used to measure the electrochemical response of ZrO 2 /CS/rGOA composite-based biosensor towards luteolin at the working potential window (−0.8–0.8 V). The improved performance of this biosensor was attributed to efficient electron transfer and large surface area of 3D rGOA, and high specific activity of Zr towards adjacent hydroxyl groups. Under optimized conditions, the analytical performance of this method towards luteolin was investigated with a detection limit of 1 nM and a linear range from 5 nM to 1000 nM.. Finally, the ZrO 2 /CS/rGOA-GCE electrochemical method coupled with solid phase extraction was used for the detection of luteolin in real samples. Recoveries of  spiked samples with different concentrations were in the range 78.6–103.3% with a relative RSD lower than 12.0%. Schematic representation of the preparation of the ZrO 2 nanoparticles and chitosan doped graphene aerogel modified electrode. The electrode was employed for the detection of luteolin coupled with the solid-phase extraction technique.

Reparative Effects of Ethanol-Induced Intestinal Barrier Injury by Flavonoid Luteolin via MAPK/NF-κB/MLCK and Nrf2 Signaling Pathways

Jinwen Yuan,Siyan Che,Li Zhang,Zheng Ruan

Luteolin, a dietary flavonoid, has gained increasing interest as an intestinal protectant. This study aimed to evaluate the reparative effect of luteolin against ethanol-induced intestinal barrier damage in a Caco-2 cell monolayer model and the potential mechanisms. Luteolin attenuated ethanol-induced intestinal barrier injury, by increasing transepithelial monolayer resistance (TEER, 27.75 ± 14.75% of the ethanol group, p < 0.01), reducing Lucifer yellow flux (13.21 ± 1.23% of ethanol group, p < 0.01), and upregulating the expression of tight junction (TJ) proteins zonulin occludin-1 (ZO-1), occludin, and claudin-1 (37.963 ± 8.62%, 17.69 ± 7.35%, and 29.40 ± 8.08% of the ethanol group, respectively, p < 0.01). Further mechanistic studies showed that luteolin suppressed myosin light chain 2 (MLC) phosphorylation, myosin light chain kinase (MLCK) activation, nuclear factor kappa-B (NF-κB) nuclear translocation, and mitogen-activated-protein-kinase (MAPK) phosphorylation. Moreover, luteolin also acted as antioxidants indirectly by upregulating antioxidant-responsive-element (ARE) and nuclear factor (erythroid-derived 2)-like 2 (Nrf2) nuclear translocation to relieve ethanol-induced oxidative damage and TJ dysfunction. The results of the study indicate that luteolin may play an effective role in relieving intestinal barrier damage, and this effect is at least partially due to its indirect antioxidant capacity.

N-doped luteolin-based carbon dots as a novel matrix for the analysis of small molecules by MALDI-TOF MS

Yanan Zhao,Ying-ai Piao,Long-yue Meng,Biao Jin

Biomass-derived carbon dots were synthesized by a hydrothermal method using the flavonoid luteolin, 3′,4′,5,7-tetrahydroxyflavone, as a carbon source. The reaction conditions were optimized with regard to reaction temperature, reaction time, and amount of luteolin. N-doped luteolin-based carbon dots (N-CDs) were obtained through the addition of ethylenediamine and used as MALDI-TOF matrix for the analysis of amino acids and other low molecular-weight molecules. Compared with N-free CDs, the matrix application of N-CDs displayed less background interferences, high sensitivity, superior efficiency, and good repeatability. Pyridinic N atoms were identified as the major component of the nitrogen dopants in N-CDs. The lone electron pair of this N species, which is located in an sp 2 -orbital perpendicular to the aromatic system, and the π-conjugated structure contributed to an enhanced desorption/ionization efficiency. Furthermore, the qualitative detection of amino acids in serum samples was accomplished by MALDI-TOF MS using the N-CD matrix. This new type of MALDI-TOF matrix demonstrated its potential in the analysis of small molecules present in chemical, environmental, or biological samples.

pH-responsive magnetic metal-organic framework nanocomposite: A smart porous adsorbent for highly specific enrichment of cis-diol containing luteolin

Shucheng Liu,Yue Ma,Lin Gao,Jianming Pan

A simple yet efficient strategy is urgently desired to prepare smart metal-organic framework nanocomposite for highly selective capture and separation of natural flavone from complex extraction samples. In this study, novel hydrophilic magnetic borate-functionalized metal-organic framework nanocomposites (Fe 3 O 4 @PDA@BA-MOFs) were designed and synthesized for selective enrichment of cis -diol containing luteolin (LTL). The well-known solvothermal reaction was adopted to prepare the magnetic core Fe 3 O 4 , followed by self-polymerization of dopamine, to create the polydopamine (PDA) onto Fe 3 O 4 . Thanks to the hydroxyl and amino groups of PDA, Zn 2 +  was easily adhered to the surface, taking advantage of magnetic response to fabricate the Fe 3 O 4 @PDA@BA-MOFs by layer-by-layer assembly strategy with borate ligand (5-boronobenzene-1,3-dicarboxylic acid). The ultrahigh surface area, uniform morphology, high saturation magnetization, excellent hydrophilicity and strong magnetic responsiveness of the as-prerared smart porous adsorbent (i.e. Fe 3 O 4 @PDA@BA-MOFs) were highly confirmed by the characterizations. Based on the batch mode experiments, Fe 3 O 4 @PDA@BA-MOFs showed high selectivity, obvious binding kinetics, large adsorption capacity, and excellent reusability (above 91.23% at least six repeated adsorption-desorption cycles). Moreover, pH-responsive capture and release mechanism based on the boronic acid affinity was proved, and Fe 3 O 4 @PDA@BA-MOFs would be good candidates for the enrichment and purification of the LTL.

Selective recognition of luteolin and quercetin based on the specific interaction of ortho-dihydroxy substituents with a zinc(II) complex

Bowen Xiao,Huijuan Wang,Xijuan Zhao,Yuanfang Li

Selective recognition of ortho-dihydroxy-substituted flavonoids has been developed based on their specific interaction with the fluorescent complex, 4-(methylphenyl)-2,2′:6′,2′′-terpyridine–Zn(II) (mptpy–Zn(II)). Six related flavonoids, including quercetin, naringenin, genistein, luteolin, daidzein, and morin, which have hydroxyl substituents in different positions, have been investigated. It has been observed that the ortho-3′,4′-dihydroxy substituents in luteolin and quercetin play an important role in the fluorescence quenching of mptpy–Zn(II) as ternary complexes are formed between luteolin/quercetin and mptpy–Zn(II). The quenching constants and fluorescence lifetime measurements indicated that the quenching was static. As the easy oxidation of 3-hydroxy and 4′-hydroxy in quercetin reduced the number of ortho-dihydroxy groups, luteolin had a greater quenching efficiency than quercetin under the same conditions. Furthermore, this fluorimetric method was successfully applied in the determination of the luteolin content in Lamiophlomis capsules from two factories, with satisfactory results compared with an HPLC method.
