The Lysine Acetylation Modification in the Porin Aha1 of Aeromonas hydrophila Regulates the Uptake of Multidrug Antibiotics

Lishan Zhang,Zujie Yao,Huamei Tang,Qingli Song,Huanhuan Song,Jindong Yao,Zhen Li,Xiaofang Xie,Yuexu Lin,Xiangmin Lin

Protein lysine acetylation (Kac) modification plays important roles in diverse physiological functions. However, there is little evidence on the role of Kac modification in bacterial antibiotic resistance. Here, we compared the differential expressions of whole-cell proteins and Kac peptides in oxytetracycline sensitive and oxytetracycline resistance (OXYR) strains of Aeromonas hydrophila using quantitative proteomics technologies. We observed a porin family protein Aha1 downregulated in the OXYR strain, which may have an important role in the OXY resistance. Interestingly, seven of eight Kac peptides of Aha1 decreased abundance in OXYR as well. Microbiologic assays showed that the K57R, K187R, and K197R Aha1 mutants significantly increased antibiotic resistance to OXY and reduced the intracellular OXY accumulation in OXY stress. Moreover, these Aha1 mutants displayed multidrug resistance features to tetracyclines and β-lactam antibiotics. The 3D model prediction showed that the Kac states of K57, K187, and K197 sites located at the extracellular pore vestibule of Aha1 may be involved in the uptake of specific types of antibiotics. Overall, our results indicate a novel antibiotic resistance mechanism mediated by Kac modification, which may provide a clue for the development of antibiotic therapy strategies.

Bioinspired design of mannose-decorated globular lysine dendrimers promotes diabetic wound healing by orchestrating appropriate macrophage polarization

Yuhang Jiang,Wentao Zhao,Shuangshuang Xu,Jingjing Wei,Fernando López Lasaosa,Yiyan He,Hongli Mao,Rosa María Bolea Bailo,Deling Kong,Zhongwei Gu

A large number of cytokines or growth factors have been used in the treatment of inflammation. However, they are highly dependent on an optimal delivery system with sufficient loading efficiency and protection of growth factors from proteolytic degradation. To develop the immunotherapy capacity of peptide dendrimers themselves, inspired by the structure and immunoregulatory functions of mannose-capped lipoarabinomannan (ManLAM), we thus propose a hypothesis that mannose-decorated globular lysine dendrimers (MGLDs) with precise molecular design can elicit anti-inflammatory activity through targeting and reprogramming macrophages to M2 phenotype. To achieve this, a series of mannose-decorated globular lysine dendrimers (MGLDs) was developed. Size-controlled MGLDs obtained were spherical with positive surface charges. The mean size ranged from 50-200 nm in varying generations and modification degrees. The initial screening study revealed that MGLDs have superior biocompatibility. When cocultured with MGLDs, mouse bone marrow-derived macrophages (BMDMs) acquired an anti-inflammatory M2 phenotype characterized by significant mannose receptor (MR) clustering on the cell surface and the elongated shape, an increased production of transforming growth factor (TGF)-β1, interleukin (IL)-4 and IL-10, a downregulated secretory of IL-1β, IL-6, and tumor necrosis factor (TNF)-α, and increased ability to induce fibroblast proliferation . Then in vivo studies further demonstrated that topical administration of optimized MGLDs accelerates wound repair of full-thickness cutaneous defects in type 2 diabetic mice via M2 macrophage polarization. Mechanistically, MGLDs treatment showed an enhanced closure rate, collagen deposition, and angiogenesis , along with mitigated inflammation modulated by a suppressed secretory of pro-inflammation cytokines, and increased production of TGF-β1. These findings provide the first evidence that the bioinspired design of MGLDs can direct M2 macrophage polarization, which may be beneficial in the therapy of injuries and inflammation.

Synergistic recovery and enhancement of gelling properties of oxidatively damaged myofibrillar protein by l-lysine and transglutaminase

Yungang Cao,Baoling Li,Xin Fan,Jiankang Wang,Zhenbao Zhu,Junrong Huang,Youling L. Xiong

The influence of combined Lysine (Lys) and transglutaminase (TG) on the conformation and gelling properties of oxidatively damaged myofibrillar protein (MP) was investigated. The addition of Lys (5 mM) significantly increased the α-helix content (by 47.8%) and decreased the particle size of oxidatively damaged MP, and improved the cooking yield (by 16.8%) and the breaking strength of MP gels (by 65.5%). The treatment with TG (E:S = 1:500) led to a slightly reduced α-helix content but improved breaking strength (by 41.8%) and cooking loss (by 13.3%) of the gels. Their combination (Lys + TG) showed the greatest and synergistic overall improvement, with the set gel displaying a fine, smooth and compact network structure. Notably, the gelling ability of oxidatively damaged MP upon Lys + TG treatment was significantly stronger than that of non-oxidized MP far exceeding its recovery. Therefore, significantly enhanced gelling properties of oxidatively damaged MP can be attained through the combination Lys and TG.

Development of Nonfouling Zwitterionic Copolymerized Peptides Based on Glutamic Acid and Lysine Dimers for Adjustable Enzymatic Degradation

Yumeng Guo,Liangbo Xu,Weifeng Lin,Shengfu Chen

Nonspecific protein adsorption-resistant materials, the so-called nonfouling materials, are crucial biomaterials in biomedical applications. Up-to-date, little attention was paid to the biodegradability of these materials. In this work, nonfouling zwitterionic copolymerized peptides composed of the N-l-glumatyl-l-lysine dimer (EK) and δ-l-lysinyl-l-glutamic acid dimer (E-K, glutamic acid with the lysine side chain) at various ratios were synthesized to investigate the enzymatic degradation rate. Two types of proteases (trypsin and alkaline protease), which represent a site-specific and less site-specific cleavage protease, respectively, were used to demonstrate the adjustable degradability by tracking the molecular weight (Mw) at different digestion times. Results showed that higher compositions of the E-K dimer lead to slower degradation rates by both proteases and larger fragments after 120 min digestion. With the composition of the E-K dimer over 50%, the degradation of copolymerized peptides by both proteases becomes very slow. This indicated that the bulky lysinyl side chain on E-K can alter the enzymolysis process for adjusting the enzymatic degradability of the newly synthesized zwitterionic copolymerized peptides, which could be promising candidates for biomedical applications in vivo.

Use of encapsulated L-lysine-HCl and DL-methionine improves postprandial amino acid balance in laying hens

Sun Mingfa,Zhao Jingpeng,Wang Xiaojuan,Jiao Hongchao,Lin Hai

The supplementation of dietary limiting amino acids (AA) with crystalline AA makes the use of low-protein diets an option in poultry production. The differing absorption rates of crystalline and protein-bound AA may lead to temporally imbalanced AA in the postabsorptive period. In this study, two experiments were conducted to evaluate the effect of encapsulated L-lysine-HCl (L-Lys-HCl) and DL-methionine (DL-Met) on the laying performance of hens. In exp. 1, a total of 135 forty-seven-wk-old Hy-Line Brown hens were subjected to three dietary treatments for 8 wk: basal diet supplemented with 0.14% L-Lys-HCl and 0.17% DL-Met to satisfy the NRC (1994) total Lys and Met recommendation (control) and basal diet supplemented with encapsulated L-Lys-HCl and DL-Met at the levels of 60% (60CLM, 0.084% L-Lys-HCl and 0.102% DL-Met) or 80% of control (80CLM, 0.112% L-Lys-HCl and 0.136% DL-Met), respectively. In exp. 2, 24 fifty-five-wk-old Hy-Line Brown hens were individually reared in cages and subjected to the same treatments as in exp. 1. The plasma concentrations of free AA and nitrogen metabolites were measured 2, 4, and 6 h after fed. The results showed that dietary AA treatment had no significant influence on body weight (BW), feed intake, laying rate, egg weight, egg mass, or feed efficiency. The expression levels of AA transporters CAT-1, y+LAT1, b0,+AT, B0AT, rBAT, EAAT3, and PepT1 in the duodenum, jejunum, and ileum were not influenced (P > 0.05) by dietary treatment. There was an interaction of dietary AA treatment and time (P < 0.05) and the 80CLM hens exhibited higher concentrations of Lys (P < 0.05) than the controls at 2-h time point. In contrast, plasma Met concentration was not influenced (P > 0.05), while Cys was reduced in the 60CLM hens at every time point. The 80CLM hens had higher taurine concentrations than those receiving the control diet at every postprandial time point. In conclusion, these findings demonstrate that by using encapsulated form, the supplemental levels of synthetic L-Lys-HCl and DL-Met can be effectively reduced by approximately 20% with no negative effect on laying performance. The result suggests that encapsulated Lys and Met may ameliorate the postabsorptive AA balance and contribute to the reduced dietary AA supplemental levels.

Hydrothermal synthesis of nitrogen-doped ordered mesoporous carbon via lysine-assisted self-assembly for efficient CO2 capture

Xia Wan,Yuchen Li,Huining Xiao,Yuanfeng Pan,Jie Liu

Nitrogen-doped ordered mesoporous carbons (NOMCs) were synthesized by single-step hydrothermal self-assembly using F127 as a soft template, hexamine as a formaldehyde source, L-lysine as a polymerization catalyst, and 3-aminophenol as both carbon and nitrogen sources. The microstructure of the NOMCs was characterized by XRD, N2 adsorption/desorption, TEM, FTIR, and XPS. The results indicated that the obtained NOMCs exhibited a large specific surface area, uniform pore size distribution and highly ordered 2-D hexagonal mesostructure (P6mm). Besides, the nitrogen was uniformly doped into the carbon framework in the form of various nitrogen species. The adsorption isotherms of CO2 and N2 were also determined and could be well fitted by a DSL model. The capture capacity of CO2 was affected by both the nitrogen content and mesostructure of the adsorbents. Overall, NOMC-L-0.5 displayed excellent CO2 capture capacity (0 °C, 3.32 mmol g−1; 25 °C, 2.50 mmol g−1), and still demonstrated great regenerability with only 2% loss after several CO2 adsorption/desorption cycles. Moreover, the CO2/N2 selectivity calculated by IAST was as high as 43.2 at 25 °C in a typical composition of flue gas (binary mixtures with 15% CO2). The superior adsorption performance enables NOMCs to be a promising CO2 adsorbent in practical applications.

Carboxymethyl cellulose-based nanocomposites reinforced with montmorillonite and ε-poly-l-lysine for antimicrobial active food packaging

Yunqing He,Xiang Fei,Hui Li

Traditional packaging materials with single function or weak activity cannot meet the requirements of active packaging. In this study, antimicrobial carboxymethyl cellulose (CMC)-based nanocomposites containing montmorillonite (MMT) and ε-poly-( l -lysine) (ε-PL) were prepared by solution casting method. The morphological, optical, mechanical, barrier, thermal, and antimicrobial properties of the resulted ternary nanocomposites were investigated. The surface morphology of the films was dependent on the contents of the ε-PL used. The incorporation of MMT and ε-PL into CMC matrix improved the UV barrier property, tensile strength, hydrophobicity, and water vapor barrier property. When incorporation of 7.5 wt % ε-PL, the tensile strength increased more than threefold and the water vapor permeability decreased by about 35% compared with pure CMC film, the water contact angle reached up to the highest value of 65.38°. The ε-PL incorporated CMC/MMT films exhibited good antimicrobial activities against Gram-positive ( Staphylococus aureus ) and Gram-negative ( Escherichia coli ) bacteria and fungi ( Botrytis cinerea and Rhizopus oligosporus ). Microbial growth inhibition rate of above 90% was obtained when the content of ε-PL got up to 7.5 wt %. In addition, CMC/MMT/ε-PL7.5 wt % film-forming solution having the appropriate properties was used as coating for the strawberries preservation, the shelf life of strawberries was extended for 2 days. © 2019 Wiley Periodicals, Inc. Interfacial superassembly of flower-like NiMn-LDH@poly-l-lysine composites for selective electrochemical sensing of tryptophan

Jing Qian,Juan Yang,Yuanyuan Zhang,Ting Zeng,Qijin Wan,Nianjun Yang

Development of sensitive, selective, and facile electrochemical analytical approaches to monitor tryptophan in different samples is of high significance. For such approaches, efficient electrodes play the key role. Herein, a NiMn-layered double hydroxide (LDH)@poly- l -lysine (PLL) composite has been prepared through in-situ electro-polymerization and further utilized to monitor tryptophan in medicament and biological systems. The as-synthesized NiMn-LDH@PLL composite is highly stable and features a flower-like morphology. It is found that the electron transfer rate of NiMn-LDH@PLL is greatly improved due to the introduction of PLL. The existing Lewis acid—base interaction between LDH and PLL contributes to the increase of unsaturated metal sites and endows this heterojunction composite with high electrocatalytic activity . Especially, the NiMn-LDH (2:1)@PLL composite displays much improved electrochemical response toward tryptophan sensing. This is due to reasonable balance between the outstanding electrocatalytic ability of nickel and the conductivity improvement caused by manganese species. On the other hand, the coordination promotion between NiMn-LDH and PLL is also conducive to improve the sensitivity of electrochemical response. The fabricated sensor exhibits wide linear response in the concentration ranges of 0.1–40 μM and 40–130 μM for tryptophan detection, along with a low detection limit of 52.7 nM. It further displays a high anti-interference ability for tryptophan monitoring even in the presence of other coexistent amino acids and small-sized biological molecules. This study provides a new way to employ LDHs as brilliant sensing platform to ensemble electrochemical sensors for the monitoring of biomolecules.

Hyperbranched Poly-L-Lysine Based Water-Insoluble Complexes as Antibacterial Agents with Efficient Antibacterial Activity And Cytocompatibility

Qi Chen,Yuanjing Xu,Jingyi Feng,Xingshuang Lv,Xiaohui Fu,Shuaishuai Yuan,Zhibo Li

Despite the advances in technology, bacterial infection associated with biomedical devices is still one of the most challenging issues in clinical practice. Incorporation of antimicrobial agents is regarded as an efficient way to combat medical device associated infectious. However, most of antimicrobial agents have high toxicity to host cells. Thus, fabrication of novel antimicrobial agents that simultaneously fulfil the requirements of antibacterial activity as well as biocompatibility is urgently needed. Herein, a series of water-insoluble antibacterial complexes based on hyperbranched poly-L-lysine (HBPL) and four different surfactants through non-covalent interactions were developed. Such kinds of surfactants had great effects on the antibacterial property of poly(ɛ-caprolactone) (PCL) films that incorporated with the HBPL-based complexes. The results revealed that the PCL films that doped with HBPL/phosphate ester surfactant complexes showed the highest bacterial killing efficiency. Moreover, the cytocompatibility of the composite films was also investigated. Hemolysis experiments indicated that all the films had low hemolytic activities. Considering the excellent antimicrobial and cytocompatibility properties, we believe that the optimized complexes have great potential to be used as antimicrobial agents in biomedical field. Tannic acid-Fe3+ dual catalysis induced rapid polymerization of injectable poly(lysine) hydrogel for infected wound healing

Wenxia Guo,Yang Li,Chenhui Zhu,Zhiguang Duan,Rongzhan Fu,Daidi Fan

Infected wounds are difficult to heal because they are vulnerable to bacterial attacks, inflammatory responses, and oxidative stress . To promote the healing of infected wounds, we developed an injectable dual-network hydrogel TFAEP (TA-Fe, APS, EPL-GMA, PVA) based on ε-poly- l -lysine- graft -glycidyl methacrylate (EPL-GMA), polyvinyl alcohol (PVA), and tannic acid-iron (TA-Fe). TA-Fe formed a stable redox pair, which acted as a dual-autocatalytic system to activate ammonium persulfate , generate free radicals, and subsequently induce EPL-GMA polymerization. Then PVA formed hydrogen bonds with TA molecules. Here, TA-Fe not only simulated peroxidase to convert H 2 O 2 into hydroxyl radicals ( OH), but also exhibited good near-infrared photothermal conversion efficiency, which all endowed the hydrogel with excellent antibacterial ability. In addition, the hydrogel could remove excessive reactive oxygen species and reactive nitrogen species, alleviating oxidative stress and reducing inflammation response due to the presence of TA molecules. Moreover, the hydrogel showed good injectability and tissue adhesion, ensuring the close adhesion of the hydrogel to the wound and achieving the maximum function. In vivo experiments demonstrated that the hydrogel promoted infected wound healing by accelerating epidermal regeneration, promoting angiogenesis and collagen deposition, and facilitating the expression of anti-inflammatory factors.

HPLC-MS/MS method for the simultaneous determination of lysine-derived markers for protein carbonylation in meat

Shouyin Wang,Shengjie Li,Wenhui Wang,Xiuxia Guo,Jinfeng Pan,Xiuping Dong

This paper aimed to develop a quantitative method based on high-performance liquid chromatography (HPLC) coupled with triple quadrupole tandem mass spectrometry (MS/MS) for the simultaneous determination of two protein carbonylation products derived from lysine residues in meat proteins, namely, α-aminoadipic semialdehyde (AAS) and lysinonorleucine (LNL). Mass spectrometry conditions and derivatization parameters were optimized and the linearity, repeatability, bias, the limit of detection (LOD), the lower limit of quantification (LLOQ), and recovery were validated. The developed method showed satisfactory linearity, repeatability, and sensitivity for both AAS and LNL. This method was applied to analyze the contents of these compounds in different meat products, and the formation of AAS was observed to be closely related to the severity of the manufacturing conditions. Moreover, AAS was demonstrated to be a reliable but more sensitive marker for protein carbonylation compared to the DNPH method, which could also provide information regarding the specific pathway of protein carbonylation.
