Highly sensitive electrochemical microfluidic thread-based nanobiosensor for lung cancer biomarker miRNA-21

Xuan Weng,Guanbo Wang,Longyan Chen,Hai Jiang

This study presents the development of a microfluidic thread-based electrochemical nanobiosensor for the ultrasensitive detection of the lung cancer biomarker microRNA-21 (miR-21). The biosensor was constructed on nylon thread electrodes modified with poly(3,4-ethylenedioxythiophene):polystyrene sulfonate (PEDOT:PSS)–MXene–gold nanoparticles (AuNPs) (PMA) nanocomposites. Capture probes (CPs) were immobilized on the thread electrode surface via Au-S bonds. A robust signal amplification was achieved through horseradish peroxidase (HRP) catalysis, activated upon the formation of a sandwich hybridization structure between the target miR-21 and biotin-labeled signal probes (SPs). The electrochemical properties of the nanobiosensors were characterized using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Under optimized conditions, the biosensor exhibited a wide linear detection range from 1 fM to 1 nM, with an ultralow detection limit of 0.15 fM. It also demonstrated high specificity against mismatched sequences, good reproducibility, and acceptable stability. When applied to spiked human serum samples, the recovery rates ranged from 96.4% to 104.3%, confirming its potential for clinical sample analysis. The thread-based three-electrode system offers advantages over conventional screen-printed and glassy carbon electrodes, including adjustable electrode spacing and layout flexibility, low cost and a simple fabrication process. The proposed method provides a promising technological platform for developing sensitive, stable, and point-of-care (POC) diagnostic tools for miR-21 and other disease-related biomarkers.

Nano-delivery of miRNA inhibiting CENPF combined with cisplatin for bladder cancer treatment

Ruixiang Song,Xin Chen,Zhensheng Zhang,Huiqing Wang,Guanghua Chen,Jinshan Xu,Shuxiong Zeng,Wentao Zhang,Xudong Yao

Bladder cancer (BCa) presents a substantial global health burden, with high rates of recurrence and metastasis that limit the effectiveness of current therapies. New therapeutic strategies are urgently needed. This study introduces a novel nanotherapeutic approach utilizing polydopamine (PDA) nanoparticles to co-deliver cisplatin and miR-205-5p for BCa treatment. Combination therapy reduces the dose-dependent toxicity of cisplatin while enhancing tumor cell cytotoxicity. miR-205-5p targets centromere protein F (CENPF), a key regulator of cancer progression. Overexpression of CENPF in BCa correlates with poor prognosis, and miR-205-5p-mediated suppression of CENPF expression inhibits tumor growth. The PDA-based system combines the DNA-damaging effects of cisplatin with the gene-silencing properties of miR-205-5p, resulting in synergistic antitumor activity. This multimodal strategy enhances therapeutic precision and efficacy, providing a promising solution for BCa treatment with significant clinical potential. Bladder cancer (BCa) presents a substantial global health burden, with high rates of recurrence and metastasis that limit the effectiveness of current therapies.

Hesperidin Alleviates Lipopolysaccharide-Induced Neuroinflammation in Mice by Promoting the miRNA-132 Pathway

Li Min,Shao Huanzhang,Zhang Xia,Qin Bingyu

Previous studies have demonstrated that hesperidin, a flavanone glycoside from citrus fruits, produces antidepressant-like effects in both mice and rats. However, whether these effects are mediated by pro-inflammatory cytokines remains unknown. In the present study, we attempted to investigate the effects of hesperidin on the depressive-like behavior; the serum corticosterone concentrations; and the interleukin (IL)-1β, IL-6, and tumor necrosis factor alpha (TNF-α) levels in lipopolysaccharide (LPS)-induced depression-like mice. In particular, we evaluated the miRNA-132 expression after LPS and hesperidin treatment. We found that LPS injection not only decreased the sucrose preference and increased the serum corticosterone levels but also elevated IL-1β, IL-6, and TNF-α in the prefrontal cortex. More importantly, LPS down-regulated the expression of miRNA-132. Pre-treatment with hesperidin (25, 50, 100 mg/kg) for 7 days prevented these abnormalities induced by LPS injection. In contrast, this effect of hesperidin was abolished by a miRNA-132 antagomir. Taken together, these results suggest that the antidepressant-like mechanisms of hesperidin are at least partially related to decreased pro-inflammatory cytokine levels via the miRNA-132 pathway in the brain.

Droplet-based PCR in a 3D-printed microfluidic chip for miRNA-21 detection

Zhiqiang Jiao,Lei Zhao,Chu Tang,Hongyan Shi,Fu Wang,Bo Hu

Recently, 3D printing technology has been widely applied in microfluidics due to its capacity of three-dimensional construction, rapid and automatic fabrication, and less time consumption. However, a droplet-based PCR in 3D-printed microfluidic chips is rarely reported. In this study, a 3D-printed microfluidic chip was designed and fabricated for droplet-based PCR detection of miRNA-21. In this chip, the capacity of monodisperse droplet generation was demonstrated. Moreover, the temperature of the chip was uniform and sufficiently stable, meeting the requirement for the PCR process. In addition, the working conditions of the droplet-based PCR process in the 3D-printed chip were explored. Finally, the droplet-based PCR detection of miRNA-21 levels in the MCF-10A cells and MDA-MB-231 cells was successfully achieved using the 3D-printed chip. We expect that the enormous potential of 3D printing technology in the research area of microfluidics will be explored in the future.

Efficient miRNA Inhibitor with GO-PEI Nanosheets for Osteosarcoma Suppression by Targeting PTEN

Lingling Ou,Haiyingjie Lin,Yuwei Song,Guoqiang Tan,Xiujuan Gui,Jinyuan Li,Xiaoting Chen,Zhendong Deng,Shaoqiang Lin

Background Gene therapy is considered a novel way to treat osteosarcoma, and microRNAs are potential therapeutic targets for osteosarcoma. miR-214 has been found to promote osteosarcoma aggression and metastasis. Graphene oxide (GO) is widely used for gene delivery for the distinct physiochemical properties and minimal cytotoxicity. Methods Polyethyleneimine (PEI)-functionalized GO complex was well-prepared and loaded with miR-214 inhibitor at different concentrations. The load efficacy was tested by gel retardation assay and the cy3-labeled fluorescence of cellular uptake. The experiments of wound healing, immunofluorescence staining, Western blot, qRT-PCR and immunohistochemical staining were performed to measure the inhibitory effect of the miR-214 inhibitor systematically released from the complexes against MG63, U2OS cells and xenograft tumors. Results The systematic mechanistic elucidation of the efficient delivery of the miR-214 inhibitor by GO-PEI indicated that the inhibition of cellular miR-214 caused a decrease in osteosarcoma cell invasion and migration and an increase in apoptosis by targeting phosphatase and tensin homolog (PTEN). The synergistic combination of the GO-PEI-miR-214 inhibitor and CDDP chemotherapy showed significant cell death. In a xenograft mouse model, the GO-PEI-miR-214 inhibitor significantly inhibited tumor volume growth. Conclusion This study indicates the potential of functionalized GO-PEI as a vehicle for miRNA inhibitor delivery to treat osteosarcoma with low toxicity and miR-214 can be a good target for osteosarcoma therapy.

Electrochemical detection of miRNA-21 coupling catalytic hairpin assembly with photo-ATRP amplification

Yunna Li,Zhidan Tian,Liang Zhou,Jinming Kong,Xueji Zhang

Target-triggered CHA generates azide-bearing duplexes on the GCE surface. • CuAAC links CHA-derived DNA assemblies with ATRP initiator sites. • Py-mediated photo-ATRP forms electroactive poly(FMMA) for signal amplification. • The platform shows a 100 aM–100 pM range and a blank-based LOD of 60 aM. • Matrix-matched calibration supports preliminary analysis in 10% diluted NHS. The electrochemical conversion of nucleic acid recognition events into amplified current signals remains important for developing sensitive miRNA biosensors. Herein, we propose a target-programmed and light-triggered interfacial signal-amplification strategy for miRNA-21 detection by coupling catalytic hairpin assembly (CHA), CuAAC click chemistry, and pyrene-mediated photo-induced atom transfer radical polymerization (photo-ATRP). In this design, miRNA-21 first triggers CHA on the H1-modified glassy carbon electrode surface, generating surface-bound H1–H2 duplexes carrying terminal azide groups. These azide-bearing duplexes are subsequently coupled with the alkyne-containing ATRP initiator PBIB through CuAAC, thereby converting the target-triggered DNA assembly product into interfacial polymerization-related sites. Under UV irradiation, pyrene activates the photo-ATRP of ferrocenylmethyl methacrylate (FMMA), leading to the in situ formation of electroactive poly(FMMA) and an enhanced square-wave voltammetry response. The stepwise sensing mechanism was supported by agarose gel electrophoresis, FTIR, component-deletion SWV experiments, EIS/CV characterization, and interfacial morphology analysis. Under optimized conditions, the biosensor showed a linear response toward miRNA-21 from 100 aM to 100 pM, with a blank-based limit of detection of 60 aM. The method also exhibited acceptable selectivity, reproducibility, short-term storage stability, and preliminary recovery performance in 10% diluted normal human serum spiked samples with matrix-matched calibration. This work demonstrates a proof-of-concept electrochemical amplification strategy that links target-catalytic DNA assembly with light-controlled interfacial polymer growth for sensitive miRNA detection. Immunomodulation of MiRNA-223-based nanoplatform for targeted therapy in retinopathy of prematurity

Keke Huang,Zhiqing Lin,Yuanyuan Ge,Xuhao Chen,Yining Pan,Ziru Lv,Xiaoting Sun,Hao Yu,Jiangfan Chen,Qingqing Yao

Retinopathy of prematurity (ROP) is characterized by pathological angiogenesis and associated inflammation in the retina and is the leading cause of childhood blindness. MiRNA-223 (miR-223) drives microglial polarization toward the anti-inflammatory phenotype and offers a therapeutic approach to suppress inflammation and consequently pathological neovascularization . However, miRNA-based therapy is hindered by the low stability and non-specific cell-targeting ability of delivery systems. In the present study, we developed folic acid–chitosan (FA–CS)-modified mesoporous silica nanoparticles (PMSN) loaded with miR-223 to regulate retinal microglial polarization. The FA–CS/PMSN/miR-223 nanoparticles exhibited high stability and loading efficiency, achieved targeted delivery, and successfully escaped from lysosomes. In cultured microglial cells, treatment with FA–CS/PMSN/miR-223 nanoparticles upregulated the anti-inflammatory gene YM1/2 and IL-4RA, and downregulated the proinflammatory genes iNOS , IL-1β , and IL-6 . Notably, in a mouse oxygen-induced retinopathy model of ROP, intravitreally injected FA–CS/PMSN/miR-223 nanoparticles (1 μg) decreased the retinal neovascular area by 52.6%. This protective effect was associated with the reduced and increased levels of pro-inflammatory (M1) and anti-inflammatory (M2) cytokines, respectively. Collectively, these findings demonstrate that FA–CS/PMSN/miR-223 nanoparticles provide an effective therapeutic strategy for the treatment of ROP by modulating the miR-223-mediated microglial polarization to the M2 phenotype.

Cu nanoclusters/nano-vesicle-based confinement-induced electrochemiluminescence strategy for miRNA-145 detection

Fengwen Cui,Peilin Wang,Kun Liu,Yuchen Guo,Qiang Ma,Yuquan He

A novel confinement-induced ECL enhancement (CIECL) sensing system has been constructed based on copper clusters (Cu NCs) and nano-vesicles in this work. The CIECL strategy resolved the shortcoming of poor stability and low luminous intensity of Cu NCs. Firstly, nano-vesicles were prepared through liquid-liquid self-assembly with amyl gemini quaternary ammonium salt and anionic surfactant. Subsequently, Cu NCs were confined in the nano-vesicles by electrostatic interaction. Due to the restriction on the intramolecular vibration and rotation of the ligands of Cu NCs, the Cu NC/nano-vesicle-based CIECL system showed the significant luminescence enhancement, good stability and high biocompatibility. Additionally, the Au nanoparticles-array with significant surface plasmon resonance effect has been prepared to further amplify the ECL intensity. Finally, the CIECL-based biosensor has been successfully used to detect miRNA-145 in clinic samples in the linear range of 100 fM to 100   nM, which displayed the potential value in the vascular calcification analysis.

Ratiometric Electrochemical Detection of MicroRNA Based on Construction of A Hierarchical C@SnS2 Nanoflower Sensing Interface

Qiu-Mei FENG,Li QIN,Peng ZHANG,Dan LI,Ming-Kai LIU,Po WANG

Building a precise, sensitive, and selective sensing platform is highly significant for microRNA (miRNA) detection. Herein, an innovative ratiometric electrochemical biosensor sensitized with carbon nanoparticles-functionalized SnS 2 nanoflowers ( [email protected] 2 NFs) and enzyme-assisted target recycling was explored for analysis of miRNA-21. In this strategy, the application of hierarchical [email protected] 2 NFs structure as electrode material not only enhanced the surface loading capability for subsequent reactions but also improved the interfacial electron transfer. After introducing enzyme-assisted target recycling, single-target miRNA-21 could initiate the cleavage of multiple capture DNA strands by duplex-specific nuclease (DSN), leading to numerous rest single-strand DNAs on the electrode. Two probes, methylene blue-labeled probe 1 (probe 1-MB) and ferrocene-labeled probe 2 (probe 2-Fc), could selectively and competitively hybridize with the capture DNA and the rest single-strand DNA, depending on the completeness of DNA hybridization. By measuring the peak current intensity ratio of Fc and MB, miRNA-21 was quantified in a wide concentration range from 1 fmol/L to 10 pmol/L, with a detection limit of 300 amol/L. Moreover, this sensing system showed good analytical performance in the presence of interfering sequences and complex matrices, illuminating excellent selectivity and practical application potential of the hierarchical [email protected] 2 NF sensing interface.

Quantitative SERS detection of multiple breast cancer miRNAs based on duplex specific nuclease-mediated signal amplification

Wei Xu,Yu Zhang,Dianhai Hou,Jianjun Shen,Jinhua Dong,Zhiqin Gao,Honglin Liu

Simultaneous and ultrasensitive detection of multiple microRNA (miRNA) biomarkers is an essential precondition for early cancer diagnosis and treatment. Here we developed a sandwich surface-enhanced Raman scattering (SERS) sensor based on Au@Ag core–shell nanorods combined with duplex specific nuclease-mediated signal amplification (DSNSA) for quantitative detection of multiple breast cancer miRNA biomarkers. The DSNSA strategy enables quantitative detection of target miRNA through rehybridizing the capture probe DNA-SERSnanotag conjugates to trigger signal amplification. The Au@Ag core–shell nanorods coated with an Ag shell exhibit excellent SERS performance, implying that molecules can be concentrated by the Ag shell at the hot spots. By monitoring the Raman signal attenuation of hot spots in the presence of target miRNAs, three breast cancer associated miRNAs (miR-21, miR-155, and let 7b) were simultaneously determined using the sandwich SERS sensor, and their detection limits (LODs) were 0.05 fM, 0.063 fM and 0.037 fM, respectively. These results indicated that our sandwich SERS sensor combined with the DSNSA strategy holds remarkable promise for multiplex detection of cancer biomarkers and contributes to early diagnosis of cancer.

Electrochemical-Enhanced Charge State Modulation of Nitrogen-Vacancy Centers for Ultrasensitive Biodetection of MicroRNA-155

Xiaojian Fan,Youqiang Xing,Ze Wu,Bingjue Li,Peng Huang,Lei Liu

Sensitive and accurate miRNA detection is important in cancer diagnosis but remains challenging owing to the essential features of miRNAs, such as their small size, high homology, and low abundance. This work proposes a novel electrochemical (EC)-enhanced quantum sensor achieving quantitative detection of miRNA-155 with simultaneous EC sensing. Specifically, fluorescent nanodiamonds/MXene nanocomposites were synthesized and modified with dual-mode signal labels, enabling miRNA-155 concentration measurement via T1 relaxation time of nitrogen-vacancy (NV) centers and EC signals. Quantum sensing was enhanced via external voltage during the EC process, which modulated the negatively charged state of the NV centers, thereby improving the sensitivity and accuracy of miRNA-155 detection. EC sensing improved the accuracy and reliability of miRNA-155 detection while enhancing quantum sensing. The limit of detection (LOD) of the EC-enhanced quantum biosensor reached 10.0 aM, nearly 106 and 10 times lower than the reported LODs of a quantum sensor using bulk diamond and fluorescent sensors, respectively. The LOD of EC sensing was 2.6 aM, aligning with previous reports. The findings of the study indicated that quantum sensing combined with EC sensing can achieve ultrasensitive miRNA-155 detection with high accuracy and reliability, providing an advanced approach for early cancer diagnosis.

Nanostructuring Synergetic Base-Stacking Effect: An Enhanced Versatile Sandwich Sensor Enables Ultrasensitive Detection of MicroRNAs in Blood

Xin-Xin Peng,Tongtong Guo,Hao Lu,Linlin Yue,You Li,Dan Jin,Guo-Jun Zhang,Fan Yang

MicroRNA (MiRNA)-based noninvasive diagnostics are hampered by the challenge in the quantification of circulating miRNAs using a general strategy. Here, we present a base-stacking effect-mediated ultrasensitive electrochemical miRNA sensor (BSee-miR) with a universal sandwich configuration. In the BSee-miR, a short DNA probe (10 nucleotides) self-assembled on a gold electrode surface could effectively capture the target miRNA synergizing with another sequence based on coaxial sandwich base-stacking, which rivals the fully complementary strength. Importantly, such a sandwich structure is flexible to incorporate signal amplification strategies (e.g., biotin–avidin) that are usually difficult to achieve in short sequence detection. Using this design, the BSee-miR achieves a broad dynamic range with a detection limit down to 7.5 fM. Furthermore, we found a high-curvature nanostructuring synergetic base-stacking effect that could improve the sensitivity of the BSee-miR by two orders of magnitude (79.3 aM). Our BSee-miR also has a single-base resolution to discriminate the highly homologous miRNAs. More importantly, this approach is universal and has been used to probe target miRNAs varying in sequences and secondary structures. Our ultrasensitive sensor could detect miRNA in cell lysates and human blood and distinguish cancer patients from normal individuals, promising a versatile tool to measure clinically relevant miRNAs for tumor diagnostics.
