Integrated design of epigallocatechin gallate nanoparticles and tannic acid to reinforce the porous structure of κ-carrageenan/pullulan absorbent pads for extending beef shelf life

Xingnan Wang,Yike Han,Wenzhi Lei,Hongcai Li,Shiqi Li,Zhenpeng Gao

This study aimed to develop sustainable bioactive absorbent pads as a green alternative for meat packaging. Gelatin (Gel) self-assembled nanocarriers were constructed to encapsulate epigallocatechin gallate (EGCG), yielding Gel@EGCG nanoparticles with an encapsulation efficiency of 76.18% and a loading capacity of 10.65%. Subsequently, these nanocomposites were incorporated together with natural crosslinker tannic acid (TA) into a κ-carrageenan/pullulan (Car/Pul) matrix to fabricate biopolymer absorbent pads (CPTGE). The resulting three-dimensional porous structure (stabilized by hydrogen bonding and electrostatic interactions) endowed CPTGE pads oil–water superabsorbent characteristics, while reinforcing compressive resistance. Moreover, CPTGE2 (Car/Pul containing TA and 0.2%wt Gel@EGCG) effectively scavenged 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radicals, with inhibition rates of 86.72% against Escherichia coli and 92.07% against Staphylococcus aureus , conferring active antioxidant and antibacterial activities. Notably, CPTGE2 significantly delayed quality deterioration by suppressing myoglobin oxidation (reducing metmyoglobin content by 50.47%) and lipid peroxidation (lowering thiobarbituric acid reactive substances [TBARS] values by 51.10%), ultimately extending beef shelf life. This provides new insights for designing active packaging that integrates efficient liquid absorption with antimicrobial and antioxidant functions.

Chickpea protein isolate-pullulan polysaccharide hydrogel enhances gel performance and saltiness perception in low-salt shrimp surimi gels via controlled sodium release and structural optimization

Kexin Zhang,Yan Guo,Maolin Tu,Zhongyuan Liu,Dayong Zhou,Deyang Li

This study investigates the novel application of a chickpea protein isolate (CPI)- pullulan polysaccharide (PUL) hydrogel to enhance the gel properties and saltiness perception of reduced-salt (1.5 %) shrimp surimi gels, with conventional salt addition (3 %) serving as a benchmark. The incorporation of 15 % CPI-PUL hydrogel significantly improved the functionality of reduced-salt gels: Water holding capacity increased by 38 %, hardness increased by 1.49 times, and microstructural analysis revealed a markedly denser and more stable gel network compared to reduced-salt controls without hydrogels. Crucially, electronic tongue analysis and inductively coupled plasma optical emission spectrometer confirmed that the CPI-PUL hydrogel effectively enhanced the perceived saltiness of reduced-salt gels. Elemental mapping indicated a heterogeneous Na + distribution within the gel matrix. By employing in vitro oral mastication and digestion simulations, the mechanism underlying saltiness enhancement was elucidated. Analysis of digestive fluid conductivity demonstrated rapid Na + release into simulated saliva within the first 30 s of mastication. Notably, during the subsequent chewing phase (30–60 s), the CPI-PUL hydrogel modulated Na + release kinetics, delaying its liberation and thereby prolonging the duration of salty taste perception. This targeted temporal control of sodium release presents an innovative strategy for effective salt reduction in surimi-based products without compromising sensory quality.

Construction of Pickering Emulsion of Amomum tsaoko Essential Oil Based on Cellulose Nanocrystals to Enhance Pullulan Film: Structure, Antibacterial Property, and Preservation Effect of Mango

Lin Zhu,Jiameng Liu,Zhikai Zhuang,Shaokai Zhang,Lijing Lin

Mango is prone to postharvest microbial infection, which leads to deterioration. Although essential oils show great potential for antibacterial and antioxidant applications, their effectiveness is limited by poor stability. In this study, cellulose nanocrystal (CNC)-stabilizedAmomum tsaokoessential oil emulsion (AEO–CNC) was incorporated into a pullulan (Pul) matrix through hydrogen bonding to prepare antibacterial films. A stable AEO–CNC emulsion was obtained by precisely adjusting the AEO concentration. The effects of emulsion incorporation on microstructure evolution, molecular interactions, physiochemical properties, and biological functionalities of Pul-based composite films were systematically investigated, and their preservation effect on mango was evaluated. With the increasing incorporation of AEO Pickering emulsion, the ultraviolet-blocking property (transmittance <30%), antioxidant capacity (46.83%), and antibacterial activity of PP-CNC-AEO films were significantly improved. In addition, these films exhibited excellent mechanical properties (tensile strength of 30.62 MPa) and thermal stability. The PP-CNC-AEO films had a more uniform and compact surface structure, with no bubbles and fewer internal pores, which indicates the formation of an effective cross-linked network. When PP-CNC-50%AEO-F was used to preserve mango, it reduced weight loss, delayed the increase in soluble solids, and postponed the post-ripening process, exhibiting strong potential for extending the shelf life of fruit.

Pullulan/PVA based film incorporated with cinnamon essential oil submicron Pickering emulsion stabilized by carboxymethyl chitosan modified with phenolic acids: Preparation and application on mango preservation

Chuang Zhou,Li Zhang,Zhengang Zhao,Yunhao Liu,Ziming Yang,Puwang Li

Phenolic acids grafting enhanced the DPPH/ABTS + radical scavenging activity of CMCS. • CEO submicron emulsions stabilized by CMCS-g-GA showed 60-day storage stability. • Films loaded with CEO submicron emulsions extended shelf life of mango by 5–10 days. Food packaging films with antibacterial and antioxidant properties play a crucial role in the postharvest preservation of fruits. In this study, pullulan/polyvinyl alcohol (PUL/PVA) composite films incorporated with cinnamon essential oil (CEO) Pickering emulsions with submicron droplets stabilized by carboxymethyl chitosan modified with gallic acid (CMCS-g-GA) were fabricated for mango preservation. Antioxidant assays revealed that CMCS-g-GA at 50.0 μg/mL exhibited DPPH and ABTS + radical scavenging rates of 91.06% and 93.40%, respectively, representing 8.46-fold and 10.37-fold increases compared to those of unmodified CMCS. The prepared CEO emulsion, consisting of submicron droplets with an average size of approximately 704 nm, exhibited excellent storage and centrifugal stability, showing no phase separation after 60 days of storage or centrifugation at 2000 rpm for 2.5 h. Incorporation of the CEO emulsion enhanced the antibacterial and barrier properties of composite films (PP/CG-CEO), with the PP/CG-CEO20 film exhibiting the lowest water vapor permeability (3.61 × 10 −10  g/m·s·Pa) and oxygen permeability (3.53 × 10 −10  g/m∙s∙Pa). Room-temperature storage experiments demonstrated that the PP/CG-CEO coating films exerted an effective preservative effect on mangoes, maintaining the postharvest quality, enhancing antioxidant enzyme activity, delaying yellowing, and retarding the respiratory peak. These results demonstrated that the multifunctional cinnamon essential oil Pickering emulsion films have practical potential as eco-friendly packaging for postharvest fruit preservation. The favorable role of oxidized pullulan as a multipurpose crosslinker in polyvinyl alcohol (PVA)/chitosan/collagen films for promoting human skin fibroblast viability, antibacterial activity and healing of methicillin-resistant Staphylococcus aureus (MRSA) infected wounds in mice

Mingzhen Kang,Mojtaba Koosha,Tianduo Li,Xiwen Geng

In this study, pullulan was oxidized by NaIO 4 into low, medium and high oxidation degrees (OPL1,OPL2,OPL3) and used as a crosslinker to enhance physicochemical and biological properties of the wound dressings based on polyvinyl alcohol(PVA)/chitosan/collagen (FPL,FOPL1,FOPL2,FOPL3). Physicochemical, mechanical and biological properties of OPL samples as well as crosslinked films were characterized. The results showed that OPL not only increased the cell viability of human skin fibroblasts but also promoted antibacterial activity. Fourier transform infrared spectroscopy, rheological and gel content results proved the crosslinking effect of OPL. Mechanical properties of FOPL2 and FOPL3 films was >2 times higher than FPL film. The viability of human skin fibroblast cells was higher than the control for all films, and antibacterial activity of the film FOPL3 was significantly higher compared to other films. Wound healing effect of FOPL3 film was evaluated in a methicillin-resistant staphylococcus aureus (MRSA) infected full-thickness skin defect model in mice. The results showed a significantly lower wound area after 7,10,14 days of treatment compared to the control group treated by normal saline. The findings of this research enlighten wound healing potentials of oxidized pullulan to be used as a multifunctional ingredient for the development of future skin care biomaterials.
