In this study, trans-ferulic acid (FA) was degraded by water falling film dielectric barrier discharge (DBD) plasma coupled with CoOOH catalyst. The results reveal that CoOOH improved the degradation of FA by DBD plasma through an apparent catalytic effect. 98.2% of FA was degraded in 10 min by DBD + CoOOH (0.5 g/L dosage) system at 11.2 kV applied voltage, which was 23.8% higher than that in a sole DBD system; the corresponding energy yield ( G 50 ) was increased by 186% and the synergistic intensity for FA degradation reached up to 2.76. The evolution of reactive species reveals that the synergistic effect of DBD plasma and CoOOH catalyst can be attributed to the catalytic conversion of O 3 and H 2 O 2 into ·OH on the active sites of CoOOH surface. The results of Lewis acid sites, X-ray photoelectron spectroscopy and Raman spectroscopy suggest that the abundant oxygen vacancies and Lewis acid sites, which are offered by the lattice OH groups in CoOOH, serve as the catalytic active sites of CoOOH. According to the detected byproducts by liquid chromatograph/mass spectrometer, a possible FA degradation pathway was also tentatively proposed.

A facile approach was developed for preparing the multilayer hybrid films of mixed (phthalocyaninato) (porphyrinato) europium(III) triple-decker compound (Pc)Eu(Pc)Eu[trans-T(COOCH3)2PP] (1) and graphene oxide (GO) using the solution-processing QLS method. The combination of the nature of relatively high conductivity and great surface area for GO with the electroactive and semiconductive triple-decker compound in ITO electrode renders the hybrid film excellent sensing property for H2O2, due to the optimized triple-decker molecular packing in the uniform-sized nanoparticles (ca. 70 nm) formed on the GO surface. The amperometric responses are linearly proportional to the concentration of H2O2 in the range of 0.05–1800 μM with a fast response time of 0.03 s μM–1, a low detection limit of 0.017 μM, and good sensitivity of 7.4 μA mM–1. The present work represents the best result of tetrapyrrole-based nonenzymatic electrochemical sensor for H2O2. Nevertheless, the triple-decker/GO/ITO also shows excellent stability, reproducibility, and selectivity, indicating the great potential of electroactive tetrapyrrole rare earth sandwich compounds in combination with GO in the field of nonenzymatic electrochemical sensors.

Dalbergia odoriferais widely used to treat cardiovascular diseases. Our research group found thatDalbergia odoriferavolatile oil has a good anti-myocardial ischemic effect, and its main pharmacodynamic components are trans-nerolol and its oxides. However, the exact mechanisms underlying this effect have not yet been elucidated. This study aimed to explore the potential myocardial protective effects of trans-nerolol and its underlying molecular mechanisms. Molecular docking was used to predict and visualize the possible mechanism of the anti-apoptotic myocardial protection by trans-nerolol. The myocardial protective effect of trans-nerolol was evaluated by observing pathological injury, myocardial enzyme levels, oxidation, antioxidant levels, and the expression of related proteins. Molecular docking results showed that trans-nerolol binds closely to cytochrome C (Cytc) and apoptosis-related proteins, suggesting that it may play a role in interacting with these target proteins. The results showed that pre-treatment with dose-dependent trans-nerolol significantly mitigated the myocardial histological damage; decreased lactate dehydrogenase (LDH), creatinine kinase (CK), alanine transaminase (ALT), and aspartate transaminase (AST) levels; reduced nitric oxide (NO) production, hydrogen peroxide (H2O2), and lipid peroxide (LPO); and increased the total antioxidant content (T-AOC), glutathione (GSH), catalase (CAT), and superoxide dismutase (SOD) activities compared with the model group. In addition, dose-dependent trans-nerolol significantly increased the Na+-K+-ATPase and Ca2+-Mg2+-ATPase levels. Moreover, trans-nerolol markedly reduced the endogenous and external apoptotic pathways; downregulated the protein expression of Cytc, apoptotic protease activating factor-1 (Apaf1), Fibroblast-associated (Fas), Cysteine-aspartate protease 3 (Caspase3), Cysteine-aspartate protease 8 (Caspase8), and Cysteine-aspartate protease 9 (Caspase9); and upregulated the expression of Heat shock protein 70 (Hsp70) and B-cell lymphoma-2 (Bcl-2). These data indicate that trans-nerolol exerts protective effects against myocardial ischemia (MI), and its mechanism is associated with the suppression of the Cytc- and caspase-signaling pathways. Trans-nerolol has a therapeutic effect on MI, and its mechanism of action is related to its anti-apoptotic effect. These results suggest thatDalbergia odoriferahas a potential role to be developed as an MI-promoting therapeutic agent.

Aging entails the progressive decline in the body’s self-regulation and functionality over time. Notably, obesity and aging exhibit parallel phenotypes, with obesity further accelerating the aging process across multiple dimensions and diminishing lifespan. In this study, we explored the impact of trans fatty acid (TFA) consumption on the overall health and lifespan of male Drosophila melanogaster under an isocaloric high-sugar and high-fat diet. Our results indicate that TFA intake results in a shortened lifespan, elevated body weight, and increased triglyceride levels in flies fed a high-sugar and high-fat diet with equivalent caloric intake. Additionally, TFA exposure induces oxidative stress, locomotor deficits, and damage to the intestinal barrier in flies. Collectively, chronic TFA consumption expedites the aging process and reduces the lifespan of male Drosophila melanogaster . These results contribute supplementary evidence regarding the adverse health effects associated with TFAs. Graphical Abstract Trans fatty acid intake results in a shortened lifespan, elevated body weight, and increased triglyceride levels in male Drosophila melanogaster fed a high-sugar and high-fat diet with equivalent caloric intake. In recent years, fatty acids containing conjugated C Cs have attracted extensive research attention due to their biological activities against human diseases. However, their differentiation is challenging. This study developed a comprehensive analytical solution to accurately differentiate cis/trans- fatty acid isomers using ion mobility mass spectrometry (IM-MS) and theoretical calculations. Cis/trans -fatty acids were mobility-differentiated via simple complexation with 1,5,9-triazacyclododecane (9C3N) or 1,4,8,11-tetraazacyclotetradecane (10C4N) and metal ions , obtaining baseline separation with a peak-to-peak resolution of 0.35–0.92. Moreover, the conformation of the complexes was optimized theoretically, revealing different binding modes between the cis / trans- fatty acid-9C3N/10C4N–metal ion systems, yielding in-depth structural data on the complexes and elucidating the principles of mobility separation. Furthermore, the proposed method was assessed in terms of quantification, accuracy, and precision repeatability. Finally, the method was applied to analyze oil samples. Given its simplicity, speed, and lack of chemical derivatization or chromatographic separation, this technique has potential applications in food analysis .

( Z )-1,3-pentadiene (Zp) exhibits a rigid planar molecular structure and is known for the typical 1,5-sigmatropic hydrogen shift, while it is commonly considered an inert monomer during polymerization. Herein, by utilizing the steric effect of the terminal methyl in Zp, the high trans -1,4-polypentadiene (HTPD) with highly ordered microstructure and well-defined sequence are successfully synthesized using barium dithymol/ tert- butyllithium/ tri -ethylaluminum trimetallic initiators based on anionic polymerization. The significant dependence of polymerization rate and trans -1,4-selectivity on ratio of Ba/Al/Li is demonstrated. The modification in microstructure ( trans -1,4 content ranging from 75 % to 92 %) and stereo-regularity (isotactic triad mm % > 65 %) are confirmed by nuclear magnetic resonance (NMR). Compared with the poly(( Z )-1,3-pentadiene) (PZp) prepared by n -BuLi containing σ -ligand modifiers, the strategy using a large steric initiator to achieve regular insertion of Zp into polymer chains is proved to be effective. Kinetics studies demonstrated that Ba–Al–Li system is characterized by retarded anionic polymerization (RAP), yet it shows a quasi-living process and enabling the preparation of stereometric block polymers such as polyisoprene- b -HTPD (PI- b -HTPD). Thermal performance analysis indicates that the obtained highly ordered polymers show superior thermoplastic properties ( T m = 77.4 °C) with crystallinity as an elastomer. Furthermore, reasonable interpretations for the structure formation, sequence, and rate reduction effected by the Ba/Al/Li trimetallic initiator are proposed.

The CRISPR/Cas13 nucleases have been widely documented for nucleic acid detection. Understanding the intricacies of CRISPR/Cas13’s reaction components is pivotal for harnessing its full potential for biosensing applications. Herein, we report on the influence of CRISPR/Cas13a reaction components on its trans-cleavage activity and the development of an on-chip total internal reflection fluorescence microscopy (TIRFM)-powered RNA sensing system. We used SARS-CoV-2 synthetic RNA and pseudovirus as a model system. Our results show that optimizing Mg 2+ concentration, reporter length, and crRNA combination significantly improves the detection sensitivity. Under optimized conditions, we detected 100 fM unamplified SARS-CoV-2 synthetic RNA using a microtiter plate reader. To further improve sensitivity and provide a new amplification-free RNA sensing toolbox, we developed a TIRFM-based amplification-free RNA sensing system. We were able to detect RNA down to 100 aM. Furthermore, the TIRM-based detection system developed in this study is 1000-fold more sensitive than the off-coverslip assay. The possible clinical applicability of the system was demonstrated by detecting SARS-CoV-2 pseudovirus RNA. Our proposed sensing system has the potential to detect any target RNA with slight modifications to the existing setup, providing a universal RNA detection platform. Graphical Abstract

BACKGROUND Trans -2, 3-dihydro-3-hydroxyanthranilic acid (DHHA) is a valuable metabolic intermediate for the biosynthesis of wide-ranging benzoic acid derivatives with enormous biological or pharmaceutical activities. Pseudomonas chlororaphis GP72 is a non-pathogenic biocontrol strain that displays unique capability to produce phenazines. Nevertheless, DHHA production is quite low by the wild type strains, which necessitates yield improvement by constructing engineered strains for large-scale biotechnological applications. RESULTS In this study, two negative regulatory genes namely rsmE and lon were successively deactivated in the DA4 strain. The resulting engineered DA6 strain produced a significantly high titer of DHHA (20.6% higher than that of DA4). The influence of varying pH from 6.2 to 8.2 on DHHA production was studied in a 6-L benchtop fermenter. A controlled broth pH of 7.2 stimulated maximum DHHA production by the engineered DA6 strain. A DO-stat based fed-batch system maintaining a constant DO level (about 20%) accompanied by a glycerol feeding strategy was applied. Highest DHHA production using this approach was recorded to be 10.06 g L -1 , which was 31% higher than with traditional batch cultivation. CONCLUSION The production of DHHA in metabolically engineered GP72 was considerably improved by inactivating two negative regulatory genes in a fed-batch mode. It is concluded that fed-batch fermentation with intermittent glycerol feeding and pH-control was an effective approach to improve DHHA production yield. © 2017 Society of Chemical Industry

Space-limited living species provide new insight into the regulation of the microstructure of isoprene units in alternating copolymers composed of isoprene (Ip) and 1,1-bis(4-dimethylsilylphenyl)ethylene (DPE-2SiH) using living anionic polymerization (LAP) modified with additives. It was found that suitable modifiers, such as sodium alkoxide (RONa), led to nearly 80% cis 1,4-addition of the isoprene units, while addition of potassium alkoxide (ROK) resulted in nearly 70% trans 1,4-addition. Through analysis, we know that the additives influence the angles between two subterminal benzene rings, and promote the regulatory effect of the space-limited living species on the cis and trans isoprene units. The larger the angle between two subterminal benzene rings, the more favourable are the conditions for addition polymerization in this form of microstructure. The copolymerization mechanism was investigated with the assistance of density functional theory (DFT) with Gaussian 09 and the corresponding characteristics of copolymers were determined with SEC, 1H NMR, 13C NMR and MALDI-TOF MS techniques. Due to the living features of LAP, this method can be employed as a key control of the copolymer chain sequence and microstructure at the same time.

Drug resistance is one of the major reasons for chemotherapy failure in breast cancer clinical therapy. In this study, a novel liposomal two-drug co-delivery delivery system was assembled from all-trans retinoic acid (ATRA) dimer phospholipid conjugate (diATRA-PC) and developed as a
doxorubicin (DOX) carrier to overcome drug resistance in breast cancer. The DOX encapsulated diATRA-PC liposomes (diATRA-PC/DOX liposomes) were prepared by classic method, with a uniform size distribution between 50–100 nm. The free ATRA and DOX were released from liposomes simultaneously
under simulated tumor intracellular environment. Moreover, the diATRA-PC liposomes exhibited enhanced cellular uptake, cytotoxicity and migration, inhibiting capacity against DOX-resistant human breast cancer cell line (MCF-7/ADR) when compared with controls. Reverse transcription-polymerase
chain reaction (RT-PCR) analysis demonstrated that the diATRA-PC/DOX liposomes had an effective promotion of apoptosis in DOX-resistant cells, with up-regulation of Caspase-3 and down-regulation of Bcl-2 mRNA expression. One could speculate that the reversed drug resistance in MCF-7/ADR cells
was attributed to the enhanced cellular uptake of DOX by cancer cells in overcoming the drug efflux. In addition, pharmacokinetics studies showed that diATRAPC/DOX liposomes had a longer circulating time in comparison with free drug. In summary, the diATRA-PC liposomes can be regarded as a
potential multi-drug combination therapeutic system for reversing drug resistance in breast cancer.

The underlying mechanisms of complement activation in Stargardt disease type 1 (STGD1) and age-related macular degeneration (AMD) are not fully understood. Overaccumulation of all- trans -retinal (atRAL) has been proposed as the pathogenic factor in both diseases. By incubating retinal pigment epithelium (RPE) cells with atRAL, we showed that C5b-9 membrane attack complexes (MACs) were generated mainly through complement alternative pathway. An increase in complement factor B (CFB) expression as well as downregulation of complement regulatory proteins CD46, CD55, CD59, and CFH were observed in RPE cells after atRAL treatment. Furthermore, interleukin-1β production was provoked in both atRAL-treated RPE cells and microglia/macrophages. Coincubation of RPE cells with interleukin-1 receptor antagonist (IL1Ra) and atRAL ameliorated complement activation and downregulated CFB expression by attenuating both p38 and c-Jun N-terminal kinase (JNK) signaling pathways. Our findings demonstrate that atRAL induces an autocrine/paracrine IL-1/IL-1R signaling to promote complement alternative pathway activation in RPE cells and provide a novel perspective on the pathomechanism of macular degeneration.

Accurate and rapid determination of aflatoxin M1 (AFM1) in food is crucial for ensuring the safety of the human diet. In view of this, we proposed a proximity-inducible Cas13a activity and hydrogel-assisted paper-based distance sensor for the detection of aflatoxin M1. The single-stranded DNA (ssDNA) and AFM1 aptamer were split into two parts and linked to the terminus of two separate DNA activator strands, respectively. By introducing AFM1, it stimulated these two DNA activator strands to assemble with CRISPR RNA (crRNA) through spatial proximity mechanism between the target and these two DNA activators to activate the Cas13a trans-cleavage towards the RNA-containing site within hairpin probe to release a DNA initiator. The released DNA initiator triggered the catalytic hairpin assembly (CHA) between magnetic beads (MBs)-DNA capture probe and trypsin-DNA detection probe. After hydrolysis of gelatin hydrogel by trypsin, water released from the hydrogel flowed along the pH indicator strip and exhibited a distinct mark. Depending on this portable method, the limit of detection (LOD) was achieved at 23.6 ng L -1 . Furthermore, our developed strategy can be applied to measure AFM1 in ultra-high temperature (UHT) milk with satisfactory effects, and its excellent specificity and reproducibility also demonstrate that our platform has great potential for application in environmental monitoring.
