d-tryptophan potentiates dielectric barrier discharge cold plasma inactivation of Shewanella baltica through selective singlet oxygen generation

Jian Chen,Shiya Guo,Huan Li,Shigenobu Koseki,Jianhao Zhang,Xiaowei Sheng,Yanbo Wang

Dielectric barrier discharge (DBD) cold plasma combined with d -tryptophan ( d -Trp) exhibited a pronounced synergistic inactivation effect against Shewanella baltica , a dominant spoilage bacterium in aquatic products, and the underlying reactive oxygen species (ROS)-mediated mechanism was elucidated. 40 s combined treatment (DBD + d -Trp) achieved a 7.31 log CFU/mL reduction, markedly exceeding the 1.3 log reduction obtained with DBD treatment alone. Physicochemical analysis showed elevated hydrogen peroxide and nitrite levels accompanied by decreased pH in the synergistic system. Electron paramagnetic resonance and radical scavenging assays revealed that d -Trp selectively enhanced singlet oxygen ( 1 O 2 ) generation while suppressing hydroxyl radicals and superoxide anions, thereby reshaping the ROS profile compared to DBD treatment alone. Severe cellular damage was observed by scanning electron microscopy, and increased alkaline phosphatase release, potassium leakage, and membrane depolarization confirmed disruption of the cell envelope integrity. Elevated intracellular ROS accumulation and dysregulation of antioxidant enzyme activity indicated aggravated oxidative stress leading to bacterial death. The synergistic inactivation effect was further validated in fish juice, demonstrating superior efficacy compared to individual treatments. These findings demonstrate that d -Trp potentiates plasma inactivation through selective modulation of ROS composition and provide a mechanistic foundation for developing targeted control strategies against specific spoilage bacteria in aquatic products.

Cd migration in water-level fluctuation zones of Three Gorges reservoir: Interactions of periphytic biofilms and tryptophan

Ting Liu,Mengting Xiao,Rongshan Du,Meng Li,Xinyi Li,Ying Xi,Yuanfei Lv,Honglin Liu,Yingping Huang,Tao Xu

Periphytic biofilms (PBs) and dissolved organic matter (DOM) are key factors affecting the migration of Cd at the “water-sediment” interface. However, the specific effects of PBs and protein-like components of DOM on Cd migration within the “water-biofilm-sediment” system remain poorly understood. This study simulates the dissolution and re-immobilization of Cd at the “water-biofilm-sediment” interface in the water-level fluctuation zone (WLFZ) of the Three Gorges Reservoir (TGR), aiming to clarify the role of PBs in Cd stability. The results indicate that the concentration of dissolved Cd increased by 0.96-fold (R 2  = 0.992) with each doubling of L-tryptophan (L-Trp) concentration, as the functional groups of L-Trp ( COOH and NH) formed complexes with Fe-Mn oxide-bound Cd in the sediment. In the presence of PBs and at an L-Trp concentration of 150 mg·L −1 , PBs utilized L-Trp, resulting in a 16.7 % reduction in dissolved organic carbon (DOC) ( p  < 0.05) and an increase in protein content. Additionally, PBs contributed to a 35.12 % reduction in the peak concentration of dissolved Cd, thereby stabilizing the final Cd levels. Cd enrichment within PBs, facilitated by functional groups such as >C C<, OH, >C O, and C O , led to a significant increase in Cd content (0.43 mg·kg −1 , p  < 0.05). These findings suggest that L-Trp degradation and Cd enrichment by PBs act synergistically to promote the re-immobilization of dissolved Cd. This study offers a novel perspective on Cd migration in WLFZs and provides insights that may be applicable to other aquatic environments where PBs are present.

Design, Synthesis, and Evaluation of New 2-Arylpropanoic Acid-l-Tryptophan Derivatives for Mitigating Cisplatin-Induced Nephrotoxicity

Ming Yuan,Huai Wang,Mingjun Yu,Sen Yao,Risheng Yao

Cisplatin (CIS) is a widely used chemotherapeutic agent that is highly effective against various cancers. However, its clinical application is frequently limited by its substantial nephrotoxic side effects. The gastrin-releasing peptide receptor (GRPR), a critical regulator in inflammatory diseases, has been identified as a promising therapeutic target. Our previous studies have demonstrated that the GRPR antagonists PD176252 and RH-1402 can mitigate CIS-induced nephrotoxicity through anti-inflammatory mechanisms. Based on these findings, we designed and synthesized a series of 2-arylpropanoic acid-L-tryptophan derivatives to enhance the therapeutic effects. Among these compounds,3mexhibited superior renal protection by significantly improving mouse renal tubular epithelial cell (mRTEC) viability from 50.2 ± 2.6% to 80.5 ± 3.9%, surpassing PD176252 (70.8 ± 1.4%) and RH-1402 (73.9 ± 3.7%). Moreover, compound3mmarkedly reduced the expression of kidney injury molecule-1 (KIM-1) and inflammatory cytokines [Tumor Necrosis Factor-α (TNF-α), Interleukin-6 (IL-6), Monocyte Chemoattractant Protein-1 (MCP-1)]. Finally, molecular docking results revealed that3mexhibited a high binding affinity for GRPR. Computational predictions using SwissADME further indicated that3mpossesses favorable drug-like properties, thereby supporting its potential as a promising candidate for mitigating CIS-induced nephrotoxicity.

A Eu-MOF-Based Fluorescent Sensing Probe for the Detection of Tryptophan and Cu2+ in Aqueous Solutions

Gong Yafei,Fu Yan,Lou Dawei

Abnormal tryptophan (Trp) metabolism can be used as an important indicator of chronic hepatitis, paranoia, Parkinson’s disease and other diseases. Deficiency or excessive accumulation of Cu 2+ can cause diseases such as Wilson’s disease and Alzheimer’s disease. Eu-based metal-organic framework (Eu-MOF) was successfully prepared for fluorescence sensing of Trp and Cu 2+ in an aqueous solution (pH = 7.4). Eu-MOF showed high selectivity and sensitivity for Trp and Cu 2+ with detection limits of 0.22 µM and 0.09 µM and K sv of 6.17 × 103 M − 1 and 2.37 × 10 4  M − 1 respectively. Trp and Cu2+ had overlapped UV absorption spectra with that of Eu-MOF and competed for the excitation light source. Trp also attenuated the antennae effect of organic ligands on Eu-MOF, thus quenching the red fluorescence of Eu-MOF. This study provides insights into the application of MOFs in bioanalysis and diagnostics.

Tryptophan-Restricted Intermittent Diet Alleviates Estrogen Deficiency-Induced Osteoporosis via Regulating Coupling Effects of “Gut-Bone” Axis

Tingwen Xiang,Shiyu Xiao,Chuan Yang,Dongyang Zhang,Yanghao You,Langlang Xie,Zhiguo Ling,Xiaohua Wang,Chengmin Zhang,Gang Huang,Dong Sun,Yueqi Chen,Fei Luo

Osteoporosis (OP) is a chronic and severe skeletal metabolic disorder resulting from excessive bone erosion activity and compromised bone formation. Emerging evidence highlights that the “gut-bone” axis plays a critical role in maintaining bone homeostasis via modulating gut microbiota and relevant metabolites. Individually, diet-derived tryptophan acts as one of the primary environmental factors that modulate the microbiota-bone crosstalk. Nevertheless, the promising modulatory mechanism of the “diet-microbiome-bone” axis remains unknown during OP progression. In this regard, the study focused on the dominant role of tryptophan-restricted intermittent diet during estrogen deficiency-induced OP. Upon these findings, micro-computed tomography (μCT) evaluation and histomorphometric analysis have confirmed that treatment with tryptophan-restricted intermittent diet effectively mitigated bone loss and improved bone microarchitecture. To unravel the underlying mechanism, we performed 16S rDNA gene sequencing and untargeted metabolomics to confirm the alteration of microbial community composition and metabolite profiles. Additionally, biotin was further identified as a significant microbiota-derived metabolite involved in M1 macrophage polarization and mature osteoclast apoptosis when administered with tryptophan-restricted intermittent diet. Thus, we summarized that treatment with tryptophan-restricted intermittent diet could perform a protective function during OP via modulating the coupling effects of gut microbiota and bone homeostasis, which may provide a potential therapeutic strategy for OP.

Simultaneous determination of tryptophan, kynurenine, kynurenic acid and two monoamines in rat plasma by HPLC-ECD/DAD

Ting-ting Du,Ting Cui,Hong-mei Qiu,Nian-ru Wang,Dan Huang,Xin-hui Jiang

A high-performance liquid chromatography method with a diode array and an electrochemical detection (HPLC-ECD/DAD) was developed to determine the levels of tryptophan (TRP), kynurenine (KYN), kynurenic acid (KYA), 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) in rat plasma. The prepared samples were separated on a BDS column (4.6 mm × 250 mm, 5 mm) with column oven temperature of 25 °C. The mobile phase consisted of 5% acetonitrile and a buffer solution, which contained 25 mmol/L sodium acetate and 0.01 mmol/L EDTA, adjusting pH to 4.5 with acetic acid, and it was pumped at a flow-rate of 1.0 mL/min. KYN and KYA were measured by a variable wavelength detector at wavelengths 360 nm and 333 nm respectively, TRP and vanillic acid (as IS) both were measured at 280 nm. Determination of 5-HT and 5-HIAA was accomplished at the electrochemical working potential of 700 mV. Total run time was 14 min. Several parameters of the developed method were validated including linearity, accuracy precision, and stability. The results showed the established method had good LOD and separation for all of the five compounds and IS in the biological matrix. The method is simple, fast, economical and accurate. The analytical method and the results could provide a reference for the clinical and scientific research of depression.

Determination of tryptophan and tyrosine by chemiluminescence based on a luminol–N-bromosuccinimide–ZnS quantum dots system

Shifeng Li,Min Xing,Haiyan Wang,Li Zhang,Yiman Zhong,Lei Chen

In this work, water-soluble ZnS quantum dots (QDs) modified with 3-mercaptopropionic acid (MPA), L-cysteine (L-Cys) and thioglycolic acid (TGA) were synthesized. The nanostructure and optical properties were characterized by X-ray powder diffraction (XRD), Transmission electron microscopy (TEM), UV-Vis absorption spectrum and fluorescence spectrum. Through the study of the ZnS QDs effects on the chemiluminescence (CL) system of luminol, we found that ZnS QDs could obviously enhance the CL of the luminol–N-bromosuccinimide (NBS) system in an alkaline medium. At the optimal experimental conditions, we researched the impact of 17 kinds of amino acids on this system. It was found that L-tryptophan (Trp) and L-tyrosine (Tyr) had significantly inhibitory effects on luminol–NBS–ZnS QDs CL. Based on the inhibitory effects, a novel CL method with a wider linear range and lower detection limit for determination of Trp and Tyr was developed. The detection limits were 1.5 × 10−11 g mL−1 and 2.0 × 10−10 g mL−1, respectively. The possible CL reaction mechanism was also discussed briefly.

Free-standing molecularly imprinted photonic hydrogels based on β-cyclodextrin for the visual detection of L-tryptophan

Zhaokun Yang,Dongjian Shi,Mingqing Chen,Shirong Liu

Inverse opals of molecularly imprinted photonic hydrogels (CD-MIPHs) based on β-cyclodextrin were first elaborated by using a colloidal crystal template method. The film can be exfoliated from the substrate in H2O to get a free-standing photonic crystal film. L-Tryptophan imprinted polyacrylamide photonic hydrogels (PAM-MIPHs) and CD-MIPHs were fabricated in this paper. The PAM-MIPHs exhibited a good sensing property to L-Trp and the redshift of the Bragg diffraction peak was 83 nm. Furthermore, improved recognition of L-Trp can be achieved by using CD-MIPHs. The Bragg diffraction peak of CD-MIPHs redshifted by 126 nm when the CD-MIPHs were immersed in a series of concentrations of L-Trp solution, and a visually perceptible color change was clear. Moreover, enantioselective recognition of Trp enantiomers was accomplished by CD-MIPHs instead of PAM-MIPHs. In short, the resultant CD-MIPHs showed a rapid response, high selectivity, high sensitivity and specificity towards imprinted molecules in an aqueous environment.

Constructing electrochemical sensor using molecular-imprinted polysaccharide for rapid identification and determination of l-tryptophan in diet

Yan-Jun Li,Li-Li Yang,Li Ni,Jia-Min Xiong,Jia-Yuan He,Lian-Di Zhou,Ling Luo,Qi-Hui Zhang,Chun-Su Yuan

An imbalance of l -tryptophan ( l -Trp), a basic component of a healthy diet, is harmful to human health. Traditional methods for detecting l -Trp have many limitations. To correct a deficiency or excess of l -Trp in human diets, it is necessary to develop a novel method that is rapid, low-cost, and high-sensitivity. Herein, a molecularly imprinted polysaccharide electrochemical sensor termed MIP/CS/MWCNTs/GCE (molecularly imprinted polymer/chitosan/multiwalled carbon nanotubes/glassy carbon electrode) targeting l -Trp was first constructed on a glassy carbon electrode, which was modified with multiwalled carbon nanotubes and chitosan using bifunctional monomers. The MIP/CS/MWCNTs/GCE obtained a wide linear range (1–300 μM) for detecting l -Trp and accurately detected the proportion of l -Trp in mixtures of Trp enantiomers. In milk samples, the spiked recoveries of l -Trp were 86.50 to 99.65%. The MIP/CS/MWCNTs/GCE electrochemical sensor possessed good recognition and detection performance for l -Trp and has promising potential for practical application.

Chiral recognition of tryptophan enantiomer based on the electrode modified by polyaniline adsorption bovine serum albumin complex

Wenyan Yao,Sha Li,Licheng Xie,Yan Jiang

A chiral sensing platform was constructed via adsorptive functionalization of ammonium persulfate doped polyaniline (APS-DPANI) with bovine serum albumin (BSA). The novelty of this work is the construction of such chiral interface with adsorption principle. The material has been characterized by scanning electron microscope, Fourier transform infrared and X-ray photoelectron spectroscopy, and thermogravimetric and water contact angle analyses. It displayed considerable stability in multi-run cyclic voltammetric scanning. Moreover, the superior conductivity of APS-DPANI and the decent binding ability of BSA endowed this sensing platform with an excellent recognition effect for tryptophan (Trp) enantiomers in the differential pulse voltammetry (DPV) test. The recognition was highly reproducible, and the detection limits for L- and D-isomer were 0.071 and 0.0478 mM, respectively.

Sinomenine ameliorates rheumatoid arthritis by modulating tryptophan metabolism and activating aryl hydrocarbon receptor via gut microbiota regulation

Zheng-Meng Jiang,Su-Ling Zeng,Tian-Qing Huang,Yang Lin,Fang-Fang Wang,Xing-Jiao Gao,Jing Li,Ping Li,E-Hu Liu

Gut microbiota dysbiosis is associated with the development of rheumatoid arthritis (RA). Sinomenine (SIN) is an effective immunosuppressive and anti-inflammatory drug used for treating RA, but how SIN regulates gut microbiota to alleviate RA remains underexplored. To identify the critical gut microbial species and microbial metabolites associated with the RA-protective effects of SIN, the microbiota-dependent anti-RA effects of SIN were assessed by 16S rRNA gene sequencing, antibiotic treatment, and fecal microbiota transplantation. Metabolomics analysis, transcriptional analysis, and targeted bacteria/metabolites gavage were conducted to explore how SIN regulates gut microbiota to reduce the severity of RA. SIN could restore intestinal microbial balance by mainly modulating the abundance of Lactobacillus , and significantly relieve collagen-induced arthritis (CIA) symptoms in a gut microbiota-dependent manner. SIN significantly elevated microbial tryptophan metabolites indole-3-acrylic acid (IA), indole-3-propionic acid (IPA), and indole-3-acetic acid (IAA). Tryptophan metabolites supplementation could activate aryl hydrocarbon receptor (AhR) and regulate Th17/Treg balance in CIA rats. Intriguingly, SIN relieved the arthritis symptoms involving the enrichment of two beneficial anti-CIA Lactobacillus species, L. paracasei and L. casei by mono-colonization. The promising therapeutic function of SIN was mostly attributed to the activation of AhR by explicitly targeting the Lactobacillus and microbial tryptophan metabolites. The intestinal bacterium L. paracasei and L. casei may be used to reduce the severity of CIA.

Interfacial superassembly of flower-like NiMn-LDH@poly-l-lysine composites for selective electrochemical sensing of tryptophan

Jing Qian,Juan Yang,Yuanyuan Zhang,Ting Zeng,Qijin Wan,Nianjun Yang

Development of sensitive, selective, and facile electrochemical analytical approaches to monitor tryptophan in different samples is of high significance. For such approaches, efficient electrodes play the key role. Herein, a NiMn-layered double hydroxide (LDH)@poly- l -lysine (PLL) composite has been prepared through in-situ electro-polymerization and further utilized to monitor tryptophan in medicament and biological systems. The as-synthesized NiMn-LDH@PLL composite is highly stable and features a flower-like morphology. It is found that the electron transfer rate of NiMn-LDH@PLL is greatly improved due to the introduction of PLL. The existing Lewis acid—base interaction between LDH and PLL contributes to the increase of unsaturated metal sites and endows this heterojunction composite with high electrocatalytic activity . Especially, the NiMn-LDH (2:1)@PLL composite displays much improved electrochemical response toward tryptophan sensing. This is due to reasonable balance between the outstanding electrocatalytic ability of nickel and the conductivity improvement caused by manganese species. On the other hand, the coordination promotion between NiMn-LDH and PLL is also conducive to improve the sensitivity of electrochemical response. The fabricated sensor exhibits wide linear response in the concentration ranges of 0.1–40 μM and 40–130 μM for tryptophan detection, along with a low detection limit of 52.7 nM. It further displays a high anti-interference ability for tryptophan monitoring even in the presence of other coexistent amino acids and small-sized biological molecules. This study provides a new way to employ LDHs as brilliant sensing platform to ensemble electrochemical sensors for the monitoring of biomolecules.
