Bacterial identification is challenging due to several problems, such as low sensitivity, poor selectivity, and the lack of quantitative methods. To address these obstacles, a gold nanoparticles (AuNPs)@Polyaniline (PANI)/Phage electrode was developed. The bacteriophage was immobilized on the AuNPs@PANI modified electrode, serving as an electrochemical biosensor of Escherichia coli (E. coli) DH5 $\alpha$ . In application, this biosensor can detect of E. coli DH5 $\alpha$ within a dynamic range of 10–107 CFUmL-1 with the detection limit of 1.0 CFUmL-1. Modified AuNPs@ PANI/Phage electrode exhibits a signal/noise ratio equal to 3.0, but only AuNPs@ PANI displayed 1.025 in [Fe(CN)6]3−/4− This biosensor selectively detected of E. coli DH5 $\alpha$ bacteria in the presence of comparable concentrations of different bacterial strains. In addition, the modified electrode exhibited supercapacitive behavior during electrochemical characterization and showed a high specific capacitance of 782.61 Fg-1 at 1.0 Ag-1 and 94.7% retention over 10 000 GCD cycles. Further analysis was conducted using galvanostatic charge-discharge to measure capacitance and self-discharge to assess capacity retention over time, and the Dunn method to analyze the charge storage mechanism within the electrode material. In conclusion, the developed nanostructure displayed dual electrochemical behavior as a supercapacitor and biosensor. This is the first reported PANI functionalized capacitive-controlled electrodes for bacterial detection. The fabrication and detection process for a single glass carbon electrode (GCE)/AuNPs@PANI pernigraniline salt (PANI-PS)/Phage/E. coli electrode requires ~90 min, with an estimated material cost of U.S. ${\$}$ 5.28 per electrode. Furthermore, smartphone-based testing confirmed portable, realtime detectionwith <1.5% deviation, indicating promise for multichannel clinical biosensing.

The preparation of specific antibodies is of critical importance in the immunoassay of small-molecule contaminants in food. However, due to the lack of immunogenicity, these small molecules must be conjugated to a carrier to form complete antigen in order to induce the generation of antibodies, and adjuvants are typically required during the immunization when traditional proteins were used as carriers. Therefore, identifying a substance that possesses both the characteristics of carrier and adjuvant is of significant research importance for antibody preparation. Outer membrane vesicles (OMVs) are nanoscale particles with a bilayer membrane and spherical structure produced by Gram-negative bacteria through natural budding. They contain various pathogen-associated molecular patterns (PAMPs), which could be recognized by pattern recognition receptors (PRRs) on the surface of immune cells, thereby triggering a robust immune response. In this study, OMVs derived from Δ lpxl E. coli and chloramphenicol sodium succinate (CSS) were used as the carrier and hapten to synthesize the complete chloramphenicol (CAP) antigen (CSS-Δ lpxl -OMV). BALB/c mice were subsequently immunized with this antigen to generate antibodies, with the aim of evaluating the adjuvant effects of Δ lpxl -OMV and its potential as a hapten carrier. The results indicated that Δ lpxl -OMV not only served as a carrier for binding the hapten to prepare the complete antigen but also exhibited significant adjuvant properties. The synthesized complete antigen possessed significant immunogenicity and could induce the mice to produce antibodies against the target without additional adjuvants.

Robotic grasping faces a fundamental trade-off between strong adhesion and easy, rapid release, particularly when handling fragile or heavy objects. While thermoresponsive tunable adhesives have been developed to decouple this conflict by enabling tunable adhesion via temperature changes, the lack of real-time tactile feedback limits adaptive control during dynamic manipulation. Inspired by the adhesive–tactile synergy observed in tree frog toe pads, we present a multifunctional electronic skin (e-skin) that integrates a temperature-regulated adhesive layer with a tactile perception layer for closed-loop manipulation. The adhesive layer, made of a PDMA-co-LMA (Poly(N,N-dimethylacrylamide-co-lauryl methacrylate)) copolymer, provides strong adhesion (143.46 kPa at 25 °C) and rapid release (5.41 kPa at 85 °C, a 96.23% reduction). The tactile layer, based on a clay-reinforced PDMA/ILs (ionic liquids) ionogel, exhibits a wide linear range (0–300 kPa, R2 = 0.998) and high sensitivity (13.129 kPa–1). Coupled with a real-time slip detection algorithm, the system achieves over 95% success in preventing slip and object damage. By merging tunable adhesion with tactile perception, this e-skin enables robust, adaptive, and nondestructive grasping in dynamic environments and provides a promising strategy for intelligent robotic grasping.

Dextromethorphan (DXM) is a widely used cough suppressant,
and
its escalating abuse is a pressing global health concern that necessitates
rapid screening tools. However, rapid methods for detecting DXM in
complex matrices remain highly limited. Here, we report the first
isolation and engineering of DNA aptamers specifically targeting DXM.
Using library-immobilized SELEX, we identified high-affinity aptamers
( K D ∼ 1–2 μM) with
exceptional specificity against various interferents. The lead candidate
was rationally truncated into a minimal 27-nucleotide core (DM1–27)
that retains full function. Systematic mutagenesis and molecular modeling
further delineated the structure–activity relationship governing
target recognition. Utilizing this engineered aptamer, we developed
two distinct fluorescent platforms: a label-free thioflavin T displacement
assay and a fluorophore-quencher labeled hybrid beacon. Both sensors
enabled robust DXM quantification within 10 min in human saliva and
e-cigarette juice (EJ), achieving limits of detection at low ng/mL-level.
Notably, the hybrid beacon significantly improved matrix tolerance
by bypassing short-wavelength background interference. This work provides
a versatile molecular recognition tool and establishes practical,
field-deployable sensing strategies for DXM monitoring.

Functional e-textiles have garnered increasing attention in both academic and commercial domains. However, the development of e-textiles that simultaneously exhibit efficient electrothermal physiotherapy, effective electromagnetic interference (EMI) shielding, and reliable sensing capabilities remains a significant challenge. In this study, a robust superhydrophobic e-textile is developed through the fabrication of a three-dimensional interlocked conductive coating comprising MWNTs, MXene, and AgNWs, along with a low-wettability surface interface. The low-wettability interface imparts pronounced superhydrophobicity, for which the water contact angle is 155.2°, effectively protecting the conductive components from exfoliation and chemical damage. The integration of the MWNTs/MXene/AgNWs conductive coating with the prestretching process endows the superhydrophobic e-textile with exceptional strain-sensing performance, characterized by high sensitivity (414.8), rapid response time (40 ms), and a low detection threshold (0.1%). Furthermore, the formation of the interlocked conductive coating confers excellent electrical conductivity of up to 1826 S·m–1, enabling remarkable EMI shielding effectiveness (45.4 dB) and electrothermal performance (102.3 °C at 4.0 V). These results indicate that the developed superhydrophobic e-textile holds significant potential for utilization in fields such as EMI shielding, electrothermal physiotherapy, and motion tracking.

Objective This study aimed to enhance the solubility and dispersion of poorly water-soluble dexamethasone (DEX) in oral films by developing a novel pullulan-vitamin E succinate (PUL-VES) coupling compound (designated PUVS3) as the film matrix. Methods The amphiphilic coupling compound PUVS3 was synthesized, characterized and evaluated. Using PUVS3 as a solubilizing/suspension agent, and PUL as the film-forming material, a dexamethasone-fast dissolving oral film (DEX-FDOF) was prepared by solvent casting method. The film was evaluated for appearance, microstructure, mechanical properties, weight variation, content uniformity, disintegration time and in vitro dissolution. Results The DEX-FDOF formulation exhibited a smooth surface with no observable drug precipitation and demonstrated excellent transparency. Both weight variation and content uniformity complied with the pharmacopoeial standards. The film displayed rapid disintegration (<15s) and favorable mechanical properties with an elastic modulus of 35–45 MPa and elongation at break of 17%–20%. Notably, in vitro dissolution reached approximately 47 % within 10 min, representing a significant improvement over the pure drug. This enhancement was achieved through the use of a self-synthesized PUL-VES coupling compound as the film matrix. This innovative approach enabled homogeneous dispersion of the poorly water-soluble DEX within the water-soluble oral film while simultaneously improving its solubility characteristics. Conclusion The DEX-FDOF, fabricated using the self-synthesized PUL-VES coupling compound as the matrix, exhibited excellent uniformity, mechanical strength, rapid disintegration (<15 s), and notably enhanced dissolution (47 % in 10 min), demonstrating its potential as an effective delivery system for poorly soluble drugs such as DEX.

The aim of the present study was to evaluate the effects of two well-known natural antioxidants vitamin C (VC) and vitamin E (VE) on the antifungal activity of honokiol against Candida albicans . The broth microdilution method was employed to test the antifungal activities of honokiol with or without antioxidants in the medium against C . albicans strain. Intracellular reactive oxygen species (ROS) and lipid peroxidation were determined by fluorescence staining assay. Mitochondrial dysfunction was assessed by detecting the mitochondrial DNA and the mitochondrial membrane potential. We observed that VC could significantly potentiate the antifungal activities of honokiol while VE reduced the effectiveness of honokiol against C . albicans . In addition, VC accelerated honokiol-induced mitochondrial dysfunction and inhibited glycolysis leading to a decrease in cellular ATP. However, VE could protect against mitochondrial membrane lipid peroxidation and rescue mitochondrial function after honokiol treatment. Our research provides new insight into the understanding of the action mechanism of honokiol and VC combination against C . albicans .

Candida albicansis the causal agent of invasive candidiasis, which might be lethal in immunocompromised patients. Biofilm formation is considered a key virulence factor ofC. albicansand is associated with its elevated resistance to antifungals.C. albicansand bacteria likeE. coliare frequently found to form mixed biofilms on biotic or abiotic surfaces, rendering them more refractory to existing antifungals. To investigate howE. coliendogenous indole interplaying with exogenous IAA exerts modulatory effects on dual-species biofilm withC. albicans, anE. colistrain deficient in the indole biosynthetic genetnaAwas constructed, and the enzyme TnaA inhibitor was administered to block the indole production inE. colimonoculture and/orE. coli–C. albicansdual culture. Phenotypic assay revealed that indole deficiency attenuatedE. colimono-species biofilm by 12% (tnaA∆ versus WTE. coli), and the lack of indole in theE. colicell-free culture filtrate abolished the ability to promoteC. albicansbiofilms, evidenced by the fact that the treatment with WTE. coliculture supernatants exhibited a 1.7-fold promotive effect, while treatment withtnaA∆ displayed no significant difference from the broth control towardsC. albicansbiofilms. Furthermore, impairedE. coliindole production might disruptE. coli–C. albicansbiofilm, as examined by confocal laser scanning microscopy (CLSM). Moreover, indole-3-acetic acid (IAA) was found to exhibit more potent biofilm-modulatory activity than indole by CLSM imaging with dual biofilms of WTE. coli–C. albicans, in contrast to those ofE. coli tnaA∆–C. albicanspost-supplemented with exogenous IAA. This study provides evidence for indole as a signaling molecule mediating bacterial–fungal communication during mixed-biofilm formation. Indole and its derivatives, particularly in combination with existing antifungals, have potential in the development of anti-biofilm strategies to eradicate refractory fungal infections.

Food safety is a major public health concern all over the world. Therefore, the prevention of food contamination is becoming extremely crucial. In this study, an antimicrobial nanoemulsion composed of water-soluble nisin and fat-soluble octanoic acid was successfully prepared. The results showed that the average particle size and the polymer dispersity index of the nisin−octanoic acid (NOA) nanoemulsion were around 52.21 nm and 0.253, respectively. The NOA nanoemulsion required less amounts of nisin and octanoic acid to achieve the effective antimicrobial effect against Escherichia coli and Staphylococcus aureus . In addition, the growth curves of E. coli and S. aureus were determined. The OD 600 of NOA nanoemulsion was significantly lower than free nisin after being incubated for 24 h ( p < 0.001), indicating that the antimicrobial effect of NOA nanoemulsion was outstanding. Meanwhile, the synergistic antimicrobial property of NOA nanoemulsion against E. coli and S. aureus was significantly better than free nisin under nonacid conditions ( p < 0.05). Overall, the results of this study may provide guidance for the further application of nisin in more forms.

The epidemic of avian coccidiosis seriously threatens the animals’ welfare and the economic gains of the poultry industry. Widespread in avian coccidiosis, Eimeria mitis ( E. mitis ) could obviously impair the production performance of the infected chickens. So far, few effective vaccines targeting E. mitis have been reported, and the nanovaccines composed of nanospheres captured our particular attention. At the present study, we construct two kinds of nanospheres carrying the recombinant E. mitis actin depolymerizing factor (rEmADF), then the characterization was then analyzed. After safety evaluation, the protective efficacy of rEmADF along with its nanospheres were investigated in chickens. The promoted secretions of antibodies and cytokines, as well as the enhanced percentages of CD4 + and CD8 + T cells were evaluated by the ELISA and flow cytometry assay. In addition, the absolute quantitative real-time PCR (qPCR) assay implied that vaccinations with rEmADF-entrapped nanospheres could significantly reduce the replications of E. mitis in feces. Compared with the rEmADF-loaded chitosan (EmADF-CS) nanospheres, the PLGA nanospheres carrying rEmADF (EmADF-PLGA nanosphers) were more effective in up-regulating weight efficiency of animals and generated equally ability in controlling E. mitis burdens in feces, suggesting the PLGA and CS nanospheres loaded with rEmADF were the satisfactory nanovaccines for E. mitis defense. Collectively, nanomaterials may be an effective antigen delivery system that could help recombinant E. mitis actin depolymerizing factor to enhance immunoprotections in chicken against the infections of E. mitis .

The current photocatalytic bactericidal materials in the field of food pathogen control are usually consisted of metals that always suffering from poor stability and possible secondary pollution. Besides, the requirement for high energy excitation also inspires the enthusiasm on exploring non-metallic catalysts. Herein, the non-metallic composite of rice shell biochar loaded with red phosphorus (B@RP) was developed for photocatalysis and photothermal removal of bacteria. The B@RP showed effective photocatalysis performance to stimulate the generation of OH and O 2 − free radicals for the elimination of Escherichia coli ( E. coli ). At the same time, the photothermal effect of B@RP can also increase the permeability of cell membrane, which is conducive to free radicals entering the cell interior. Therefore, the non-metallic composite could achieve complete removal of E. coli within 2 h under illumination. Meanwhile, B@RP had excellent stability and the sterilization efficiency maintained 100% after 9 cycles. Hence, B@RP is expected to be a harmless and efficient bactericidal material for food industry.
