Identification of an efficient phenanthrene-degrading Pseudarthrobacter sp. L1SW and characterization of its metabolites and catabolic pathway

Junlan Li,Wanli Peng,Xianqi Yin,Xiaozheng Wang,Zhixiang Liu,Qinchen Liu,Zixin Deng,Shuangjun Lin,Rubing Liang

Phenanthrene, a typical chemical of polycyclic aromatic hydrocarbons (PAHs) pollutants, severely threatens health of wild life and human being. Microbial degradation is effective and environment-friendly for PAH removal, while the phenanthrene-degrading mechanism in Gram-positive bacteria is unclear. In this work, one Gram-positive strain of plant growth-promoting rhizobacteria (PGPR), Pseudarthrobacter sp. L1SW, was isolated and identified with high phenanthrene-degrading efficiency and great stress tolerance. It degraded 96.3% of 500   mg/L phenanthrene in 72   h and kept stable degradation performance with heavy metals (65   mg/L of Zn 2+ , 5.56   mg/L of Ni 2+ , and 5.20   mg/L of Cr 3+ ) and surfactant (10 CMC of Tween 80). Strain L1SW degraded phenanthrene mainly through phthalic acid pathway, generating intermediate metabolites including cis -3,4-dihydrophenanthrene-3,4-diol, 1-hydroxy-2-naphthoic acid, and phthalic acid. A novel metabolite ( m/z 419.0939) was successfully separated and identified as an end-product of phenanthrene, suggesting a unique metabolic pathway. With the whole genome sequence alignment and comparative genomic analysis, 19 putative genes associated with phenanthrene metabolism in strain L1SW were identified to be distributed in three gene clusters and induced by phenanthrene and its metabolites. These findings advance the phenanthrene-degrading study in Gram-positive bacteria and promote the practical use of PGPR strains in the bioremediation of PAH-contaminated environments.

Responses of a polycyclic aromatic hydrocarbon-degrading bacterium, Paraburkholderia fungorum JT-M8, to Cd (II) under P-limited oligotrophic conditions

Jinghua Li,Yiwen Ou,Lijuan Wang,Yue Zheng,Weiyun Xu,Jianbiao Peng,Xin Zhang,Zhiguo Cao,Junpei Ye

For the bioremediation of mixed-contamination sites, studies on polycyclic aromatic hydrocarbon (PAH) degradation or Cd (II) tolerance in bacteria are commonly implemented in nutrient-rich media. In contrast, in the field, inocula usually encounter harsh oligotrophic habitats. In this study, the environmental strain Paraburkholderia fungorum JT-M8 was used to explore the overlooked Cd (II) defense mechanism during PAH dissipation under P-limited oligotrophic condition. The results showed that the growth and PAH degradation ability of JT-M8 under Cd (II) stress were correlated with phosphate contents and exhibited self-regulating properties. Phosphates mainly affected the Cd (II) content in solution, while the cellular distribution of Cd (II) depended on Cd (II) levels; Cd (II) was mainly located in the cytoplasm when exposed to less Cd (II), and vice versa. The unique Cd (II) detoxification pathways could be classified into three aspects: (i) Cd (II) ionic equilibrium and dose–response effects regulated by environmental matrices (phosphate contents); (ii) bacterial physiological self-regulation, e.g., cell surface-binding, protein secretion and active transport systems; and (iii) specific adaptive responses (flagellum aggregation). This study emphasizes the importance of considering culture conditions when assessing the metal tolerance and provides new insight into the bacterial detoxification process of complex PAH-Cd (II) pollutants.

Ratiometric room-temperature phosphorescence platform for visual and spectral analysis of naphthalene

Shishi Shao,Jiaqiang Zhao,Wei Wang,Hanbing Nie,Yan Jie Li,Xuepeng Zhang,Cheng Zhi Huang,Peng Fei Gao

Host-guest doping room-temperature phosphorescence (RTP) materials have showed excellent potential for ultra-sensitive detection due to its bright luminescence in presence of trace guests. However, the inherent signal fluctuation of solid-state sensors restricts their widespread applications. Herein we constructed a simple internal standard method to overcome the signal fluctuation problem in RTP spectral analysis. Ultralong organic RTP can be obtained by using 1,4-dichlorobenzene (DCB) as a host and trace gaseous NL as a guest through the host-guest doping strategy. In this study, a dual-emission ratiometric phosphorescent probe pyrene (Py)/pentachloropyridine (PCP)-DCB was constructed by using the Py/PCP system fabricated via melt-casting as the reference afterglow and using the DCB as the stimuli-responsive unit of gaseous NL. This probe can be used for visual identification and content detection of NL in commercial mothballs. The ratio of phosphorescence intensity (I485/I595) was linearly related to the mass of added NL (mNL) within the range of 0.05–1.45 mg in this ratiometric RTP platform, and had a low detection limit of 0.012 mg. Furthermore, the NL content of five commercially available mothballs with different NL content was detected by this Py/PCP-DCB platform, and it had good performance compared to the gas chromatography (GC) analysis results.

A novel eco-friendly strategy for removing phenanthrene from groundwater: Synergism of nanobubbles and rhamnolipid

Wang Kai Tong,Chaomeng Dai,Jiajun Hu,Jixiang Li,Min-tian Gao,Xueji You,Xin Ru Feng,Zhi Li,Lang Zhou,Yalei Zhang,Xiaoying Lai,Long Kahon,Rongbing Fu

Nanobubbles (NBs), given their unique properties, could theoretically be paired with rhamnolipids (RL) to tackle polycyclic aromatic hydrocarbon contamination in groundwater. This approach may overcome the limitations of traditional surfactants, such as high toxicity and low efficiency. In this study, the remediation efficiency of RL, with or without NBs, was assessed through soil column experiments (soil contaminated with phenanthrene). Through the analysis of the two-site non-equilibrium diffusion model, there was a synergistic effect between NBs and RL. The introduction of NBs led to a reduction of up to 24.3 % in the total removal time of phenanthrene . The direct reason for this was that with NBs, the retardation factor of RL was reduced by 1.9 % to 15.4 %, which accelerated the solute replacement of RL. The reasons for this synergy were multifaceted. Detailed analysis reveals that NBs improve RL's colloidal stability, increase its absolute zeta potential , and reduce its soil adsorption capacity by 13.3 %–19.9 %. Furthermore, NBs and their interaction with RL substantially diminish the surface tension, contact angle, and dynamic viscosity of the leaching solution. These changes in surface thermodynamic and rheological properties significantly enhance the migration efficiency of the eluent. The research outcomes facilitate a thorough comprehension of NBs' attributes and their relevant applications, and propose an eco-friendly method to improve the efficiency of surfactant remediation.

Nanoleaf-derived carbon materials as a sensitivity coating for solid‑phase microextraction of polycyclic aromatic hydrocarbons

Hu Xingru,Pang Long,Wu Mingkai,Wang Chaohai,Li Jiansheng

Metal–organic framework-derived carbon materials have shown extensive application in the sensitive extraction of polycyclic aromatic hydrocarbons (PAHs), but more active sites for its adsorption were still a tireless pursuit. In this study, ZIF-nanoleaf-derived carbon (NLCs) was synthesized and developed as a solid-phase microextraction (SPME) fiber (NLCs-F). The extraction performance was compared with ZIF-dodecahedron-derived carbon (DHCs) coated fiber (DHCs-F), which was prepared by only changing the ratio of the reactants. The unique morphology of NLCs provided abundant adsorption active sites for the selected PAHs, while the large average aperture facilitated selective extraction of high molecular weight analytes. Additionally, the high carbon content enhanced the strong enrichment capability for hydrophobic PAHs. Hence, the prepared NLCs-F coupled with GC–MS showed a good correlation coefficient (0.9975) in a wide linear range, low limits of detection (0.3–1.8 ng L −1 ), satisfactory repeatability, and reproducibility, which made it apply in the enrichment of PAHs in actual tea and coffee samples. Green preparation of magnetic pyrene-based hyper-cross-linked polymer using dual-purpose ferric chloride reagent for extraction of polycyclic aromatic hydrocarbons from natural water bodies

Fang Song,Li Ma,Li Gao,Yiwen Han,Shuai Zong,Lijun He,Shusheng Zhang,Wenjie Zhao

A magnetic hyper-cross-linked polymer Fe 3 O 4 /HCP PYR was prepared using pyrene as the monomer and formaldehyde dimethyl acetal (FDA) as the cross-linking agent. The objective of green chemistry was achieved by employing FeCl 3 during the synthesis, as it played a dual role of a catalyst for the Friedel-Crafts reaction and an iron source for the synthesis of magnetic Fe 3 O 4 , thus maximizing efficiency and minimizing waste. Fe 3 O 4 /HCP PYR was applied as a sorbent for magnetic solid-phase extraction (MSPE) to extract fifteen polycyclic aromatic hydrocarbons (PAHs) from water. The effects of different parameters such as the quantity of adsorbent, the extraction time, the desorption conditions, the pH value and the effect of the salt concentration on the extraction efficiency were optimized. A simple and efficient method in combination with gas chromatography-mass spectrometry (GC–MS) (Fe 3 O 4 /HCP PYR −MSPE/GC–MS) was developed and successfully applied for the detection of PAHs in environmental water samples The analytical method showed LODs in the range of 0.004−0.06 µg L −1 , which proved to be adequate for the detection all 15 PAHs at trace concentration. Spiked recoveries of PAHs in actual water samples ranged from 85.2 % to 118.5 % with relative standard deviations (RSDs) below 10.2%. These results indicate that the method has a good potential for reusability and possesses excellent sensitivity. This study showcased the potential of Fe 3 O 4 /HCP PYR composites in effectively removing organic pollutants from the aqueous environments, demonstrating their ability for water treatment applications.

Hierarchical metal-dielectric-metal nanostructures as SERS sensors for sensitive detection of polycyclic aromatic hydrocarbons

Shiqiang Wang,Huiyun Jiang,Junjie Feng,Yan Jin,Fei An,Liang Zhu,Anshan Xiao

Due to the wide variety, inaccessible self-degradation, high mutagenicity and carcinogenicity of polycyclic aromatic hydrocarbons (PAHs), it still remains challenging to efficiently identify them, especially in real environment. Herein, we develop a bottom-up self-assembly strategy to construct a sandwich-like hierarchical metal-dielectric-metal system composed of mercaptan-modified Au nanospheres-graphene oxide (GO)-Ag nanowires as surface-enhanced Raman spectroscopy (SERS) sensors for sensitive detection of PAHs. The synergistic integration of metallic layers and the organic mercaptan and GO layer not only provides localized enhanced electromagnetic field region which is induced by the nanogap between the top Au nanospheres and the bottom Ag nanowires, but also effectively traps PAHs in the middle layer through hydrophobic interaction of mercaptan and π-π stacking interaction of GO. By rationally optimizing the SERS substrates, the lowest detection concentrations of pyrene, anthracene, phenanthrene and benzo[a]pyrene could be reached, respectively, at 10 −8  M, 10 −8  M, 10 −8  M and 10 −7  M. The hierarchical system shows excellent uniformity, reproducibility and is able to identify various types of PAHs in multi-component mixtures as well as in real water environment. This strategy can provide a general platform to identify hazardous pollutants that are hydrophobic or contain benzene rings, such as pesticides, dyes and polychlorinated biphenyls.

Biochar inoculated with Pseudomonas putida alleviates its inhibitory effect on biodegradation pathways in phenanthrene-contaminated soil

Jinfeng Lu,Yuexian Liu,Ruili Zhang,Zhengyi Hu,Kai Xue,Biya Dong

Controversial results are reported whereby biodegradation of polycyclic aromatic hydrocarbons (PAHs) can be promoted or inhibited by biochar amendment of soil. Metabolomics was applied to analyze the metabolic profiles of amendment with biochar (BB) and biochar inoculated with functional bacteria ( Pseudomonas putida ) (BP) involved in phenanthrene (PHE) degradation. Additionally, metagenomic analysis was utilized to assess the impact of different treatments on PHE degradation by soil microorganisms. Results indicated that BB treatment decreased the PHE biodegradation of the soil indigenous bacterial consortium, but BP treatment alleviated this inhibitory effect. Metabolomics revealed the differential metabolite 9-phenanthrol was absent in the BB treatment, but was found in the control group (CK), and in the treatment inoculated with the Pseudomonas putida (Ps) and the BP treatment. Metagenomic analysis showed that biochar decreased the abundance of the cytochrome P450 monooxygenase (CYP116), which was detected in the Pseudomonas putida , thus alleviating the inhibitory effect of biochar on PHE degradation. Moreover, a noticeable delayed increase of functional gene abundance and enzymes abundance in the BB treatment was observed in the PHE degradation pathway. Our findings elucidate the mechanism of inhibition with biochar amendment and the alleviating effect of biochar inoculated with degrading bacteria.

Enhancing phenanthrene hydrogenation via controllable phosphate deposition over Ni2P/Al2O3 catalysts

Jieying Jing,Zhongyuan Guo,Ze Li,Yu Chen,Hao Li,Wen-Ying Li

Ni 2 P/Al 2 O 3 catalysts with phosphate deposition were synthesized by a hypophosphite decomposition method. Benefiting from its spherical structure and special electronic properties derived from the deposited phosphate on the catalyst surface, the Ni-2.5P/Al 2 O 3 catalyst offered remarkable activity and stability towards phenanthrene (PHE) hydrogenation, featuring a high turnover frequency of 44.64 × 10 -3 s −1 . Extensive characterizations revealed that the control of P/Ni molar ratio could change the phosphorus migration pathway and affect the amount of the generated phosphate deposition and then the exposed active sites of Ni 2 P. Density functional theory computations indicated that the deposited phosphate could induce electron transfer from Ni to phosphate, and a certain amount of phosphate deposition can endow Ni with an appropriate electron-deficient state, which can improve the PHE adsorption and result in a facile complete PHE hydrogenation.

Chemometrics-assisted excitation-emission matrix fluorescence spectroscopy for real-time migration monitoring of multiple polycyclic aromatic hydrocarbons from plastic products to food simulants

Kun Huang,Hai-Long Wu,Tong Wang,Ming-Yue Dong,Xiao-Qin Yan,Ru-Qin Yu

Polycyclic aromatic hydrocarbons (PAHs), as a class of organic pollutants that have attracted much attention, are likely to be formed with the production and processing of plastic products, and they may migrate from contaminated plastic products to food, causing the risk of poisoning or cancer. In this study, migration tests were carried out on disposable plastic products for food contact, and a novel strategy that combines excitation-emission matrix (EEM) fluorescence spectroscopy with the advanced second-order calibration method based on the three-direction resection alternating trilinear decomposition (TDR-ATLD) algorithm was used to monitor the migration of three PAHs anthracene (ANT), pyrene (PYR), and phenanthrene (PHE) from the plastic products to food simulants in real-time. With the “second-order advantage”, even if the fluorescence spectra of the target analytes overlapped seriously, and other unknown substances migrated from the plastic products to food simulants, accurate qualitative and quantitative results were still obtained by the proposed method. In the static system, the coefficient of determination ( R 2 ) of the three PAHs within the calibration range were all greater than 0.99, and the average spiked recoveries were 99.5-107.1%, with the standard deviation lower than 8.9%. The figures of merit (FOMs) and intra- or inter-day precision also showed good feasibility and reliability of the method. In the simulation study of the migration kinetic process, three PAHs can be quantified in real-time in complex matrix, then the related migration equations were established. The results indicate that the proposed method can be used for real-time migration quantitative monitoring of PAHs, providing a potential and available method for the study of the migration kinetics of hazardous substances from food contact materials to food or food simulants.

Removal of microplastics from water by coagulation of cationic-modified starch: An environmentally friendly solution

Wei Gao,Aoyun Mo,Jie Jiang,Yuqing Liang,Xiaomu Cao,Defu He

Microplastics (MPs) pose a potential risk to aquatic ecosystems , and there is a growing demand to alleviate the contamination of MPs. Here, we introduce cationic-modified starch (CS) as an eco-friendly bio-coagulant for removing MPs from water. CS with varying degrees of substitution was synthesized and characterized, and its performance in removing MPs was evaluated under different MP sizes, types, and aging, as well as various water conditions. The results indicated that CS efficiently removed MPs, achieving an average removal rate of 65.33 % for polystyrene particles, with higher removal rates for larger, high-density, and aged MPs. The efficiency of CS remained consistent across a wide range of water pH values, but was significantly reduced in the presence of kaolin clay or/and humic acid . The removal efficiency of CS for MPs was enhanced by the non-ionic surfactant, Tween 20, but inhibited by the anionic surfactant, cetyltrimethylammonium bromide. In addition, CS could concurrently remove both MPs and phenanthrene , as a typical water contaminant. Moreover, the applicability of CS was demonstrated in natural water samples from the Ecological Demonstration Zone of the Yangtze River Delta, China, with an average removal rate of 60.13 ± 3.15 %. Taken together, this study offers an environmentally friendly and cost-effective approach for the removal of MPs from water, demonstrating CS has significant application potential as a sustainable solution to mitigate microplastic pollution.

Application of temperature-sensitive hyperbranched polysiloxanes for the determination of polycyclic aromatic hydrocarbons in foodstuffs

Xiaomei Chen,Liulin Wei,Qian Liu,Xueyun Gao,Jingchan Zhao

In the present study, a novel temperature-sensitive hyperbranched polysiloxane dispersive liquid–liquid microextraction combined with high-performance liquid chromatography method (TS-HBPSi-DLLME-HPLC-UV) has been developed for the determination of five polycyclic aromatic hydrocarbons (PAHs) in instant noodles, in which temperature-sensitive hyperbranched polysiloxane (p(Ph-HBPSi-co-NIPAM)) was used as the extraction solvent. The method adsorbs the target analytes based on π-π and hydrophobic interaction between p(Ph-HBPSi-co-NIPAM) and PAHs, and then separates the polymer with the adsorbed target analytes from the solution by a phase transition. Microextraction parameters of the method including polymer ratio, type and concentration of salt, pH, extraction time and volume of acetonitrile were examined in detail and optimized. At the optimum conditions, the established method showed good linearity (R 2  ≥ 0.9971) with satisfactory limits of detection (0.089–0.16 ng mL −1 ) and limits of quantitation (0.27–0.48 ng mL −1 ). The values for intra-day and inter-day precisions were lower than 3.2% and 4.4%, respectively. Enrichment factors (EFs) and extraction recoveries (ERs) ranged from 62 to 70 and 75 to 85%, respectively. Finally, the proposed microextraction technique was validated by analyte addition and recovery studies, and applied to the determination of five PAHs in instant noodles, demonstrating that temperature-sensitive hyperbranched polymers have great potential for application in sample pretreatment techniques.
