Glucose-modified BSA/procyanidin C1 NPs penetrate the blood-brain barrier and alleviate neuroinflammation in Alzheimer's disease models

Linyan Duan,Zhizhong Hao,Rong Ji,Xingfan Li,Hao Wang,Yujing Su,Fangxia Guan,Shanshan Ma

Alzheimer's disease (AD) is a chronic neurodegenerative disease with high prevalence, long duration and poor prognosis. The blood-brain barrier (BBB) is a physiologic barrier in the central nervous system, which hinders the entry of most drugs into the brain from the blood, thus affecting the efficacy of drugs for AD. Natural products are recognized as one of the promising and unique therapeutic approaches to treat AD. To improve the efficiency and therapeutic effect of the drug across the BBB, a natural polyphenolic compound, procyanidin C-1 (C1) was encapsulated in glucose-functionalized bovine serum albumin (BSA) nanoparticles to construct Glu-BSA/C1 NPs in our study. Glu-BSA/C1 NPs exhibited good stability, slow release, biocompatibility and antioxidant properties. In addition, Glu-BSA/C1 NPs penetrated the BBB, accumulated in the brain by targeting Glut1, and maintained the BBB integrity both in vitro and in vivo . Moreover, Glu-BSA/C1 NPs alleviated memory impairment of 5 × FAD mice by reducing Aβ deposition and Tau phosphorylation and promoting neurogenesis. Mechanistically, Glu-BSA/C1 NPs significantly activated the PI3K/AKT pathway and inhibited the NLRP3/Caspase-1/IL-1β pathway thereby suppressing neuroinflammation. Taken together, Glu-BSA/C1 NPs could penetrate the BBB and mitigate neuroinflammation in AD, which provides a new therapeutic approach targeting AD.

Preparation of novel cinnamaldehyde derivative–BSA nanoparticles with high stability, good cell penetrating ability, and promising anticancer activity

Shuhua Chang,Dawei Qin,Lizhen Wang,Mengli Zhang,Rongjun Yan,Cuiyun Zhao

The purpose of our research is to find a new nanoparticle to conjugate BSA with a low water-soluble antitumor compound, cinnamaldehyde (CA). In this study, a cinnamaldehyde derivative (CAH) was synthesized by reacting CA with anthraniloyl hydrazine (AH) to obtain a pharmaceutical carrier with medicinal activity, and the yield can be 95%. BSA–CAH nanoparticles (NPs) were prepared by a desolvation method. Following modification, the isoelectric point of the NPs increased to 5.5–6.0, with negative changes in tumor micro environment. The microscopy analyses revealed that the NPs were homogeneously dispersed and exhibited a circular-granular structure, with 172 nm size. The modified BSA was characterized for the structure by various spectral analysis. The results showed that the content of β-turns and the antiparallel β-sheet increased, the fluorescence intensity at 370 nm was also significantly enhanced and red-shifted, so that the degree of conjugation increased after modification. Gel permeation chromatography (GPC) revealed that the molecular weight of the NPs was 83.629 kDa, confirming that 56 CAH molecules were conjugated to one molecule of BSA. By stability analysis, in solution, the NPs were stable for 30 days at room temperature , comparing with free CA. Moreover, the cell uptake experiment revealed that BSA–CAH NPs entered cancer cells as carriers and inhibited the majority of the studied cancer cell lines by FITC label. Notably, the laryngeal cancer inhibition rate was >80%. BSA–CAH NPs prepared herein exhibited promising anticancer activity.

BSA Nanoparticles Modified with N-Acetylcysteine for Improving the Stability and Mucoadhesion of Curcumin in the Gastrointestinal Tract

Shanshan Zhang,Sajid Asghar,Feng Yu,Zhipeng Chen,Ziyi Hu,Qineng Ping,Feng Shao,Yanyu Xiao

A major obstacle to the clinical use of curcumin (CUR) is its reduced bioavailability because of the drug’s hydrophobic nature, low intestinal absorption, and rapid metabolism. In this study, a novel oral drug delivery system was constructed for improving the stability and enhancing mucoadhesion of CUR in the gastrointestinal (GI) tract. First, CUR was encapsulated in the bovine serum albumin nanoparticles (CUR-BSA-NPs). Then, N-acetyl cysteine (NAC)-modified CUR-BSA-NPs (CUR-NBSA-NPs) were obtained. The average particle size and zeta potential of CUR-NBSA-NPs were 251.6 nm and −30.66 mV, respectively; encapsulation efficiency and drug loading were 85.79 and 10.9%, respectively. CUR-NBSA-NPs exhibited a sustained release property and prominently enhanced stability in simulated GI conditions. Additionally, enhanced mucoadhesion of CUR-NBSA-NPs was also observed. An MTT study showed that the CUR-NBSA-NPs were safe for oral administration. Overall, NAC-modified BSA-NPs may potentially serve as an oral vehicle for improving CUR stability in the GI tract and enhancing mucoadhesion.

Characterizing the binding interaction between antimalarial artemether (AMT) and bovine serum albumin (BSA): Spectroscopic and molecular docking methods

Jie-hua Shi,Dong-qi Pan,Xiou-xiou Wang,Ting-Ting Liu,Min Jiang,Qi Wang

Artemether (AMT), a peroxide sesquiterpenoides, has been widely used as an antimalarial for the treatment of multiple drug-resistant strains of plasmodium falciparum malaria . In this work, the binding interaction of AMT with bovine serum albumin (BSA) under the imitated physiological conditions (pH   7.4) was investigated by UV spectroscopy , fluorescence emission spectroscopy , synchronous fluorescence spectroscopy, Fourier transform infrared spectroscopy (FT-IR), circular dichroism (CD), three-dimensional fluorescence spectroscopy and molecular docking methods. The experimental results indicated that there was a change in UV absorption of BSA along with a slight red shift of absorption wavelength, indicating that the interaction of AMT with BSA occurred. The intrinsic fluorescence of BSA was quenched by AMT due to the formation of AMT–BSA complex. The number of binding sites ( n ) and binding constant of AMT–BSA complex were about 1 and 2.63   ×   10 3   M −   1 at 298   K, respectively, suggesting that there was stronger binding interaction of AMT with BSA. Based on the analysis of the signs and magnitudes of the free energy change (Δ G 0 ), enthalpic change (Δ H 0 ) and entropic change (Δ S 0 ) in the binding process, it can be concluded that the binding of AMT with BSA was enthalpy-driven process due to |   Δ H °|   >   |   T Δ S °|. The results of experiment and molecular docking confirmed the main interaction forces between AMT and BSA were van der Waals force . And, there was a slight change in the BSA conformation after binding AMT but BSA still retains its secondary structure α-helicity. However, it had been confirmed that AMT binds on the interface between sub-domain IIA and IIB of BSA.

Preparation and Properties of Sodium Alginate/Gelatin Hydrogel-Microspheres

Wen-jie Tang,Jin-xin Zhang,Liang Liu,Lu-yao Zhao,Liu Wang,Chang Shi,Yang Liu,Mei-ling Wen

Calcium alginate gel microspheres exhibiting pH response release characteristics were prepared, and utilizing Bovine serum albumin (BSA) to evaluate drug release performance. Hydrogels composed of gelatin (GL) and sodium alginate oxide (NALO), with varying ratios (7:3; 4:1; 3:2) were prepared by controlling their composition. Scanning electron microscopy (SEM) and a microplate reader (ELIASA) were used to characterize the microspheres’ microstructure and in vitro BSA release behavior, respectively. The results demonstrated that, under acidic conditions, the BSA release was slower than in neutral conditions. The hydrogels’ physical and chemical properties were analyzed using optical microscopy, Fourier transform infrared spectroscopy (FTIR), swelling experiments and mechanical testing of the hydrogels and microsphere loaded hydrogels. The results showed that a GL to NALO ratio of 7:3 increased the swelling degree, while a 4:1 ratio optimized the mechanical characteristics. The study concluded that the microsphere-loaded hydrogels exhibited significant potential for human drug delivery applications.

Polyferric-titanium composite coagulants with hydrogen bond domain expansion effect for superior coagulation performance

Wenxing Jin,Jun Nan,Hua Chai,Meng Chen,Zonghua Wang,Jinjin Jia,Langrun Song,Fangmin Wu,Bohan Liu

This study developed novel polyferric titanium chlorides (PFTC) with varying Fe/Ti ratios and basicity (B) to achieve safe and effective surface water treatment. The formation of Fe-O-Ti was confirmed through the characterization of PFTC and its impact on physicochemical properties and coagulation performance was thoroughly investigated. Additionally, the molecular-level coagulation mechanism was elucidated. The results demonstrated that PFTC exhibited remarkable stability, with a distinct pH-stable phase during alkaline titration. PFTC showed excellent bridging adsorption capacity, achieving bovine serum albumin (BSA) removal efficiency of 78.33%. The flocculation rate increased to more than 222%, producing larger, denser flocs with superior settling performance, while posing no environmental risks to treated water. XPS and FTIR analyses revealed that hydrogen bond was the primary mechanism driving bridging adsorption. Molecular docking analysis indicated that PFTC hydrolysates formed stronger hydrogen bonds with higher binding energy at key BSA residues compared to polyferric chloride (PFC). According to XDLVO theory, the dominant hydrogen bond range between PFTC and BSA was more than 2.75 times greater than that of PFC and BSA. The hydrogen bond domain expansion effect, coupled with robust hydrogen bond, overcame distance barriers, enabling PFTC to establish strong bridging adsorption interactions with a broader range of BSA molecules, thereby enhancing coagulation efficiency. This study provides valuable insights into the interaction mechanisms between PFTC and BSA, supporting the practical application of polymeric metal composite coagulants.

Preparation and properties of PVDF/SiO2@GO nanohybrid membranes via thermally induced phase separation method

Zhong-Kun Li,Wan-Zhong Lang,Wei Miao,Xi Yan,Ya-Jun Guo

Graphene oxide (GO), one of the most promising filter materials, is intensively employed to prepare the membranes via non-solvent induced phase separation(NIPS) method, but it has not been found to modify polymeric membranes via thermally induced phase separation (TIPS) method. In this work, the derivative of GO, SiO 2 @GO nanohybrid , is fabricated and employed to synthesize PVDF/SiO 2 @GO nanohybrid membranes via TIPS method for the first time. The results indicate that the PVDF/SiO 2 @GO nanohybrid membranes experience liquid-liquid phase separation mechanism, and exhibit bi-continual and asymmetric structure. The included SiO 2 @GO nanohybrid is uniformly dispersed in the membrane matrix. With the addition of SiO 2 @GO, the top surface becomes denser and the pore size decreases; but overhigh SiO 2 @GO addition for membrane M-5 triggers in the adverse trend. This is caused by the combined actions of nucleation and growth of PVDF and viscosity increase of cast solution due to the addition SiO 2 @GO nanohybrid. The BSA rejection of membrane gradually increases with the addition of PVDF/SiO 2 @GO nanohybrid accompanied with the decline of pure water permeation flux. As the SiO 2 @GO content increases to 0.9   wt%, the PVDF/SiO 2 @GO nanohybrid membrane M-4 presents the highest BSA rejection of 91.7% and the lowest permeation flux of 182.6   L   m −2   h −1   bar −1 . However, the overhigh SiO 2 @GO addition (1.2   wt%) leads to the outstanding pure water permeation flux of 679.1   L   m −2   h −1   bar −1 and a much lower BSA rejection. The addition of PVDF/SiO 2 @GO nanohybrid evidently improves the surface hydrophilicity and antifouling ability of resultant membranes. The XRD patterns and FTIR spectra of membranes verify the exclusive α-phase of PVDF, and the melting temperature( T m ) and crystallinity( x c ) evidently increase with the addition of SiO 2 @GO nanohybrid up to 0.9   wt% in the dopes. Overhigh SiO 2 @GO addition (1.2   wt%) leads to small decline for these parameters.

Enhanced Chiral Electrochemical Recognition of Tryptophan Enantiomers Using a Novel Triple-Layered GO/BSA/CS Modified Glassy Carbon Electrode

Zou Jiao,Huang Lele,Jiang Xinyu,Jiao Feipeng,Yu Jingang

A novel chiral electrochemical sensor was fabricated and used for recognition of tryptophan (Trp) enantiomers. Graphene oxide (GO) was prepared by the modified Hummers' method. Moreover, the suspensions/solutions of GO, bovine serum albumin (BSA) and chitosan (CS) in deionized-distilled water were coated onto the surface of GCE by a layer-by-layer drop method, and the novel triple layered GO/BSA/CS composite modified glassy carbon electrode (GO/BSA/CS/GCE) was fabricated. The electrochemical recognition and quantification of the Trp enantiomers on the GO/BSA/CS/GCE were performed using differential pulse voltammetry (DPV). The Trp monomer concentrations exhibited a good linear relationship with the peak current values. The GO/BSA/CS/GCE exhibited good coefficients of determination (R 2 D-Trp = 0.98917 and R 2 L-Trp = 0.94824) for D- and L-Trp enantiomers in the concentration range of 1 to 8 mM, respectively. The GO/BSA/CS/GCE fabricated in this study exhibited superior recognition performance, good oxidation potential and current response signals for D- and L-Trp. The novel GO/BSA/CS/GCE fabricated in this study exhibited potential to be used as a convenient, fast and efficient chiral electrochemical sensor for qualitative and quantitative determination of Trp enantiomers.

BSA-seq Integrated with Transcriptomics and Metabolomics Revealing the Candidate Genes Associated with Safflower Colors and Flavonoid Glycosides Biosynthesis

Wang Haotian,He Beixuan,Qi Shuyi,Gao Yue,Dong Xin,Guo Meili

Safflower is featured with time-honored medical and economic values and developing into diverse phenotypic and genetic variations. In this study, to explore the critical genes associated with color phenotypes and flavonoid derivatives biosynthesis of safflower, BSA-seq, conflated with transcriptomic and metabolic methods were performed in two extreme colors (yellow and white) in the population of “ZHH0119” and “XHH007”. After crossing two parent plants reciprocally, the F4 generation of two accessions were used to construct near-isogenic gene pools for the two extreme traits of yellow and white safflower. BSA-seq results located five QTLs regions on chromosomes 2, 8, 9, 10 and 12 including six CtPALs, three CtC4Hs, two Ct4CLs, one CtCHS, thirty-two CtUGTs and seventy CtCYPs which tied to the yellow color phenotype of safflower. Through transcriptome analysis of two accessions and at different flowering stages, 1 CtPAL, 5 CtC4Hs, 4 CtCHSs, 3 CtCHIs, 3 CtFLSs, 48 CtUGTs, 51 CtCYPs and 75 transcription factors were revealed as significantly upregulated in the yellow accession compared to the white. Integrated analysis identified eight CtUGTs (CtUGT50-57) which exhibited significant positive correlations with chalcone glycosides of yellow safflower. Based on functional characterization, CtUGT52 was found to boost Hydroxysafflor yellow A (HSYA) content in yellow safflower which possessing substrate promiscuity (chalones, flavonols and flavonoids) and catalytic promiscuity (flavonols and flavonoids), revealing its vital role in the HSYA biosynthesis through transgenic overexpression. Combining catalytic mechanism verification of CtUGT52 towards phloretin, kaempferol and luteolin, our study to some extent, elucidated the modification function of CtUGTs for flavonoid aglycones in the flavonoid biosynthesis pathway of safflower.

Construction of BSA-CuNCs@UiO-66 nanoprobe based on MOF confinement effect and its ultrasensitive fluorescence sensing for creatinine

Lian-Lian Duan,Wen-Jun Liu,Rui Zhai,Zhen-Guang Wang,Hong-Yuan Yan,Yun-Kai Lv

This study employed a pore-confined synthesis strategy to achieve the in situ growth of bovine serum albumin-capped copper nanoclusters (BSA-CuNCs) within the UiO-66 framework (BSA-CuNCs@UiO-66). This nanocomposite enables highly sensitive and specific detection of creatinine (CR). Results demonstrated that the spatial confinement imposed by UiO-66 induced aggregation of the BSA-CuNCs and suppressed non-radiative transitions, leading to an approximately 10-fold enhancement in fluorescence intensity and a 11-fold increase in quantum yield. Leveraging the specific adsorption and enrichment capability of the UiO-66 framework toward CR, the BSA-CuNCs@UiO-66 fluorescence probe exhibited significant fluorescence quenching upon exposure to CR, achieving a detection range of 50–1000 nM and a detection limit of 30.81 nM. This work presented a novel confinement engineering strategy utilizing metal-organic frameworks (MOFs), establishing a new design paradigm for high-performance fluorescence probes with significant potential in bioanalytical applications.

Red Monascus pigments attenuate fructose-driven bovine serum albumin (BSA) glycation via non-covalent and covalent interactions with proteins

Shufen Wu,Chengru Zhao,Xinxin Ma,Jingjie Cai,Changyan Dong,Shan Zhao,Zhongli Yan,Benxu Yang,Chao Li,Mianhua Chen,Changlu Wang

Two red Monascus pigments (R 1 and R 2 ) demonstrate inhibitory activity against AGEs. • R 1 and R 2 protect BSA from glycation through covalent modification. • Their anti-glycation effect is also mediated by noncovalent interactions with BSA. • Monascus pigments synergize with curcumin against glycation in a simulated system. • Monascus pigments and curcumin also act synergistically in grilled chicken wings. Two red Monascus pigments (rubropunctamine, R 1 ; monascorubramine, R 2 ) were obtained through the derivatization of orange Monascus pigments, demonstrating great promise for preventing protein glycation. R 1 and R 2 (1.4 mM) showed 43.72 ± 1.21% and 64.48 ± 1.81% inhibition of AGE formation, respectively, versus 50% for the aminoguanidine control. They also lowered fructosamine levels, and protected proteins from oxidation, cross-linking, and structural alterations. LC-MS/MS analysis revealed that R 1 and R 2 altered both the number and distribution of glycation sites on BSA and formed covalent adducts with the protein. Moreover, R 1 and R 2 were also found to interact with BSA non-covalently, thereby inducing structural modifications in the protein that ultimately resulted in further glycation suppression. Based on their IC 50 values, Monascus pigments and curcumin were combined at a fixed ratio, yielding a synergistic inhibition of AGE formation. Furthermore, this synergy was confirmed in a grilled chicken wing model, where the combination significantly reduced AGE levels. BSA@Cu3(PO4)2 hybrid nanoflower as bioinspired tyrosinase-mimicking nanozyme for production of amino acid surfactant from caffeate

Jiadong Chen,Xiao Song,Yinna Wu,Junwei Zhu,Wenbin Liu

BSA@Cu 3 (PO 4 ) 2 hybrid nanoflower as nanozyme with tyrosinase mimic activity. • Salt hydrate Na 2 CO 3 ·10H 2 O as essential cofactor for catalysis in organic solvent. • High catalytic performance for oxidation diphenol to o-quinone. • N Cu coordination as nucleation sites for HNF and active sites tyrosinase mimetics. • Production of melanin-like amino acid surfactants from ethyl caffeate. Amino acid surfactants are widely used in daily life and food industry. Melanin-like amino acid surfactant (MLAAS) is a novel type surfactant. Herein, a robust nanozyme with tyrosinase mimetic activity was first developed by fabricating BSA@Cu 3 (PO 4 ) 2 organic-inorganic hybrid nanoflower (HNF) using bovine serum albumin and copper ions as an organic and inorganic components, respectively. HNF nanozyme biomimetically oxidized ethyl caffeate to o-quinone, achieving yield of 59.57% in organic solvents with Na 2 CO 3 ·10H 2 O as essential cofactor. Michaelis constant was 15.05 mM and turnover number was 6.32 s −1 . It exhibited broad substrate scope toward diphenols and outstanding stability in various organic solvents. o-Quinone was conjugated with leucine to produce MLAAS, which possessed excellent emulsifying and foaming capacities with a critical micelle concentration of 8.12 mM. It presents a novel strategy by combination of HNF and salt hydrate for production of amino acid surfactant with promising emulsification and foaming potential in food processing.
