Although exopolysaccharide (EPS) has been applied to various fields, EPS for UVR-mediated oxidative stress repair still needs further exploration. In this study, a novel EPS was isolated from the fermentation medium of Bacillus sp. QDR3-1 and its yield was 4.8 g/L (pH 8.0, 12 % glucose, 30 °C and 6 % NaCl). The pure fraction (named EPS-M1) was purified by DEAE-cellulose and Sephadex G-100 column. EPS-M1 was a heteropolysaccharide composed of Man, Glc, Gal, and Fuc with a molecular weight of 33.8 kDa. Scanning electron microscopy (SEM) observed a rough surface and reticular structure of EPS-M1, and EPS-M1 formed spherical aggregates in aqueous solution observed in atomic force microscopy (AFM). Thermal analysis revealed that the degradation temperature of EPS-M1 was 306 °C. Moreover, methylation and NMR analysis determined that EPS-M1 was consisted of →3)-Man p -(1→, →2,6)-Man p -(1→, →4,6)-Glc p -(1→, →3)-Glc p -(1→, →4)-Gal p -(1→, →4)-Fuc p -(1→, and T-Man p -(1→. Furthermore, the cytotoxicity and the repair ability of UVR-mediated cell damage of EPS-M1 were studied with L929 cells. The results showed that EPS-M1 had good biocompatibility and it could mitigate UVR-mediated cell damage by regulating the levels of cellular reactive oxygen species (ROS), depolarization of mitochondrial membrane potential (MMP) and Caspase-3/7 activity. Overall, the structure analysis and the protective effects of EPS against L929 cells exposed to UVR provided an experimental basis for EPS in practical applications.

Sensitive, simple, and rapid pathogen separation and detection methods are essential for the early diagnosis and immediate medical treatment of bacterial infections. In this study, a novel strategy based on vancomycin (Van)-modified ovalbumin (OVA) mediated magnetic nanoprobes (MBs-OVA-Van) combined with multiplex polymerase chain reaction (mPCR) was developed for efficient isolation and detection of Staphylococcus aureus ( S. aureus ) and Bacillus cereus ( B. cereus ). Van conjugated to ovalbumin coated magnetic microcarrier via strong covalent interaction. In addition, Van binds through hydrogen bonds with d -alanyl- d -alanine moieties on the surface of gram-positive bacteria cell wall. Under optimum conditions, the MBs-OVA-Van showed that the capture efficiency (CE) for S. aureus and B. cereus in PBS and juice samples were around 91%, 92% and 89%, 85%, respectively. In addition, with a mixture containing both target bacteria in PBS and juice samples, the CE for S. aureus and B. cereus were recorded at about 90%, 91% and 80%, 84%, respectively. The MBs-OVA-Van plus mPCR method applied in PBS and juice samples for both bacteria showed limits of detection at 10 2 CFU mL -1 in a < 3 h total assay time. The results indicated that the method holds promise for isolation and enrichment of gram-positive bacteria with S. aureus and B. cereus as models.

The present study focuses on the inhibitory effect of volatile metabolites released byBacillus velezensisCT32 onVerticillium dahliaeandFusarium oxysporum, the causal agents of strawberry vascular wilt. The CT32 strain was isolated from maize straw compost tea and identified asB. velezensisbased on 16S rRNA gene sequence analysis. Bioassays conducted in sealed plates revealed that the volatile organic compounds (VOCs) produced by the strain CT32 possessed broad-spectrum antifungal activity against eight phytopathogenic fungi. The volatile profile of strain CT32 was obtained by headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography-mass spectrometry (GC-MS). A total of 30 volatile compounds were identified, six of which have not previously been detected in bacteria or fungi: (Z)-5-undecene, decyl formate, 2,4-dimethyl-6-tert-butylphenol, dodecanenitrile, 2-methylpentadecane and 2,2’,5,5’-tetramethyl-1,1’-biphenyl. Pure compounds were tested in vitro for their inhibitory effect on the mycelial growth ofV. dahliaeandF. oxysporum. Decanal, benzothiazole, 3-undecanone, 2-undecanone, 2-undecanol, undecanal and 2,4-dimethyl-6-tert-butylphenol showed high antifungal activity, with benzothiazole and 2,4-dimethyl-6-tert-butylphenol being the most potent compounds. These results indicate that the VOCs produced byB. velezensisCT32 have the potential to be used as a biofumigant for management of vascular wilt pathogens. Keywords:biocontrol;Bacillus velezensis;volatile organic compounds;vascular wilt pathogens

Nanofiber membranes have recently been considered as promising supports for the immobilization of microorganisms due to the simplicity and cost-effectiveness of electrostatic spinning technology and the ability to control fiber morphology, such as obtaining higher surface area and porosity. In this study, electrospun polyvinyl alcohol/sodium alginate/attapulgite (PVA/SA/ATP) nanofiber membrane was prepared as support for immobilized Bacillus cereus LY-1 for diesel degradation in an aqueous medium and a significant improvement in diesel removal efficiency was realized. The effect of modified ATP concentration on diesel removal was investigated. The results showed that the nanofiber membranes complexed with cetyl trimethyl ammonium bromide (CTAB) and 1% ATP (w/w) had the best capacity for diesel removing. When the initial diesel concentration was 2 g L−1, about 87.8% of diesel was removed by the immobilized LY-1 cells after 72 h. Immobilization of bacteria improves the ability of bacteria to survive in adverse environments. Immobilized LY-1 cells maintain the nature to remove diesel at high salinity or pH range of 6–9. Furthermore, the reusability of the LY-1 cells-immobilized PVA/SA/CTAB–ATP nanofiber membrane was tested. A diesel removal rate of 64.9% could be achieved after 4 times of use. PVA/SA/CTAB–ATP nanofibrous membranes with immobilized LY-1 cells are feasible, economical and environmentally friendly for remediation of diesel contamination in the aqueous medium, and have potential applications in the future.

This study explored the possible application of collagen hydrolysate (CH) in the fermentation process using Bacillus pumilus as probe bacterial strain. When CH acted as an only source of carbon and nitrogen in the culture medium, B. pumilus was observed to show normal characterizations during its growing process. Meanwhile, the utilization degree of CH by B. pumilus could be enhanced when wheat bran was added (used as carbon source) in the fermentation process. It was found that the highest alkaline protease activity was 325 U/mL when only CH was used, while it was increased to 620 U/mL with the addition of wheat bran. Besides, with different combination of soybean meal and CH, it was found that the ability using CH to produce alkaline protease is comparable to that of soybean meal in the presence of wheat bran. Moreover, the growth behaviors of the strain were not affected by inoculation concentrations, but the time of producing alkaline protease was greatly saved as reducing loading volume. Present investigation suggested that CH can be used as carbon and nitrogen sources in the fermentation process.

Abstrcact Highly sensitive and selective detection of specific DNA sequences is of great importance in clinical diagnosis, environmental and food monitoring, but it still remains challenges to develop a facile method for real sample detection in aqueous solution. Here, a simple and recyclable surface enhanced Raman scattering (SERS) sensor was constructed for Bacillus thuringiensis (Bt) special gene fragment detection by Fe 3 O 4 magnetic beads (MBs) and Au–Ag core-shell nanorods (Au@Ag NRs). A hairpin DNA with sulfhydryl and biotin was attached to Au@Ag NRs as indicator, and MBs with streptavidin (SA) were acted as the capture probe. On the basis of the biotin–SA specific interaction, target sequences were first hybridized with the hairpin DNA and exposed the biotin. Subsequently, the Au@Ag NRs were captured by the streptavidin modified MBs, which reduced the suspended NRs and led to the change of Raman intensity . Under the optimal conditions, the SERS intensity revealed a good linearity with Bt transgene fragment ranging from 0.1 pM to 1 nM with a detection limit of 0.14 pM (S/N=3). To demonstrate the specificity of the strategy, the single-base mismatch in DNA was discussed in the SERS assay. The results showed that the sensitivity and accuracy of the proposed method was acceptable in DNA detection, revealing a great potential in special gene detection.

The silicon coated Magnetic nanoparticles (MNps) were prepared and applied to the genetically modified detection for Cry1Ac protein. TEM observations showed the bare MNps were in the size of 3–5 nm and the silicon coated core/shell MNps were in the range of 30–60 nm 1 M HCl acidic solution was used to test the chemical stability of the resultant MNps and results showed the coated MNps were very stable in the acid environment even after 12 h. Cy3 labelled IgG tests showed that the probe on the MNps was kept the bioactivity in the immunoassays. MNps based detection results showed that this analytical strategy had similar linear relation which R = 0.995 with the commercial ELISA kit. The MNps based detections were reliable for the Cry1 Ac protein in the linear range from 0.1 pg/L to 1.0 mg/L, and showed better performance in the low concentration of analyte. This study provides a promising technique to detect the Cry1Ac protein with high sensitivity in the examination of the GM products which is of importance in food safety.

A sensitive and precise HPLC-DAD method with pre-column PMP derivatization was established and validated, for analyzing the polysaccharides in Bacillus Calmette-Guérin polysaccharide and nucleic acid (BCG-PSN) isolates, after acid hydrolysis. And the HPLC fingerprint profiling was used to analyze its monosaccharide composition. The monosaccharide concentration-peak area calibration curve was of good linearity (R 2 > 0.99), over the range of 0.016–0.08 mg/mL for mannose or 0.24–1.20 mg/mL for glucose, with high recovery of 93–105 % for quality control samples. The intra-day RSD values of mannose and glucose concentration were less than 2.5 % and 2.1 %, respectively, and their inter-day RSD values were less than 4.3 % and 2.2 %, respectively, and remained stable for up to 14 days. This method also remained durable against changes in chromatographic parameters, but it's susceptible to the flow rate of mobile phase. Additionally, the method was applied to analyze the content of mannose and glucose in 22 batches BCG-PSN powder and 17 batches BCG-PSN injection. The results showed that the HPLC-DAD fingerprint spectra of all the BCG-PSN powder and BCG-PSN injection samples had a high degree of similarity, with the similar indexes up to 0.999 and 0.998, respectively. The HPLC-DAD method with pre-column PMP derivatization is highly rapid, effective, visual, and accurate for determination of monosaccharide contents. The validated method was successfully applied to the analysis of polysaccharide in both BCG-PSN powder and injection.

Films with excellent mechanical properties and antimicrobial activity have potential applications in food preservation. The antimicrobial film was prepared by blending probiotic Bacillus velezensis 906 metabolites (906), potassium sorbate (PS), and polyvinyl alcohol (PVA). The film was characterized using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and thermal stability analysis. The mechanical, barrier, and antimicrobial properties of the film were tested. The results of the physicochemical analysis indicated that, compared to the pure PVA film, the film's mechanical properties decreased, while its barrier properties and hydrophobicity increased with the addition of PS and 906, respectively. The results of the antimicrobial tests revealed that the blend film exhibited significant antimicrobial activity against Vibrio parahaemolyticus, Psychrobacter maritimus , Oceanobacillus kimchii, Pseudomonas aeruginosa , and Listeria monocytogenes . SEM demonstrated good fusion between the components of the PVA/PS/906 film, resulting in a dense and crack-free structure. FTIR suggested that PS and 906 enhance molecular interactions within the film matrix. XRD revealed that the film was crystallized, with 906 increasing the crystallinity of the bonds between the film components. Thermal analysis indicated the thermal stability of the films was enhanced. Consequently, the PVA/PS/906 blend film shows significant potential for application in the preservation of aquatic products.

Understanding the role of common microorganisms in the spoilage of cooked sausages is essential for developing effective preservation strategies. In the framework of this study, six potential spoilage microorganisms were isolated from spoiled cooked sausages, identified through morphological, physiological, and genetic analyses as Bacillus cereus (two strains), Bacillus safensis , Bacillus altitudinis , Bacillus megaterium , and Bacillus infantis . All isolates exhibited significant protein or lipid degradation activities, except for B. infantis . The spoilage potential of Bacillus was systematically evaluated using a cooked sausage inoculation model in situ based on physical, chemical, and biological changes. Results indicated that Bacillus rapidly colonized cooked sausages, accelerating protein and lipid degradation and thereby deteriorating appearance and texture. Additionally, 74 volatile organic compounds (VOCs) were identified in both Bacillus -inoculated and control samples, revealing that Bacillus transformed aroma compounds such as terpenes into off-flavor compounds like alcohols, aldehydes, and acids ( e.g. , 1-octen-3-ol, nonanal, and phenylacetic acid). Correlation analysis confirmed the association of Bacillus with spoilage characteristics and VOCs evolution. This study elucidates the spoilage potential and mechanisms of Bacillus in cooked sausages, providing a foundation for developing an effective quality control strategy.

Peanut meal, as an important feed and protein source, is highly susceptible to aflatoxin contamination. Probiotics with the advantages of aiding digestion and improving intestinal function are regarded as potential ways to detoxify aflatoxins. In this study, six probiotic strains screened from peanut meal were identified and used for aflatoxin detoxification. The strain of Bacillus velezensis NWPZ-8 noted as A43 showed the best ability to detoxify aflatoxins. The optimized aflatoxin B1 detoxification conditions by A43 in peanut meal were proposed. At these conditions, the strain not only detoxified 75.29% aflatoxin B1 but also increased the content of protein and amino acids in peanut meal. It was found that the aflatoxins detoxification activity by A43 was mainly attributed to the extracellular proteins. Therefore, this study provides a potential probiotics strain that can be used for aflatoxin detoxification in peanut meal.

Intravesical Bacillus Calmette–Guérin (BCG) immunotherapy is a standard treatment for intermediate-risk and high-risk nonmuscle-invasive bladder cancer. The prolonged duration of BCG in the bladder can enhance the immunotherapeutic efficacy. However, intravesical BCG is rapidly excreted with urine by urination at 2 h postinstillation, compromising therapeutic outcomes. Here, this study develops a floating drug delivery system based on air microbubbles to achieve long-term retention of BCG in the bladder. By encapsulating BCG within floating air microbubbles, we successfully develop BCG-loaded microbubbles (BCG-MBs) that are capable of prolonged release of BCG in the bladder. Compared with conventional intravesical BCG immunotherapy, intravesical instillation of BCG-MBs induces more durable immune responses and demonstrates enhanced therapeutic efficacy in SD rats with orthotopic bladder cancer. Furthermore, the floating drug delivery system based on air-microbubbles not only represents a potential intravesical drug carrier for bladder cancer, but also a versatile platform that can encapsulate diverse therapeutic agents, thereby showing great potential applications in immunotherapy, targeted drug delivery, and combination therapy.
