Bisphenol A (BPA), a component of polycarbonate and epoxy resins, has been reported to induce learning and memory deficits. However, the mechanisms have not been fully elucidated. Growing evidence has suggested that N-methyl-d-aspartate receptors (NMDARs) are involved in cognitive impairments. In this study, BPA was administered to female Sprague–Dawley rats (six per dose group) at concentrations of 0 (control), 4, 40, and 400 μg/kg·body weight/day from gestation day 1 through lactation day 21. Spatial learning was evaluated using the Morris water maze on postnatal day 22. Expression levels of NMDARs were determined using real-time polymerase chain reaction and Western blot. The results showed that male offspring exposed to BPA exhibited increased latency in reaching the platform and reduced time in the target quadrant, and the number of crossing the platform was less, as compared with the control group. The mRNA and protein expression levels of NMDARs in the hippocampus were significantly downregulated when compared with the control group of male offspring. The data showed that maternal exposure to BPA at low dosage can cause cognitive deficits in male rat offspring, probably due to a decrease in NMDARs in the hippocampus.

It is critical for fabricating flexible biosensors with both high sensitivity and good selectivity to realize real-time monitoring superoxide anion (O 2 •− ), a specific reactive oxygen species that plays critical roles in various biological processes. This work delicately designs a Mn 3 (PO 4 ) 2 /MXene heterostructured biomimetic enzyme by assembling two-dimensional (2-D) Mn 3 (PO 4 ) 2 nanosheets with biomimetic activity and 2-D MXene nanosheets with high conductivity and abundant functional groups. The 2-D nature of the two components with strong interfacial interaction synergistically enables the heterostructure an excellent flexibility with retained 100% of the response when to reach a bending angle up to 180°, and 96% of the response after 100 bending/relaxing cycles. It is found that the surface charge state of the heterostructure promotes the adsorption of O 2 •− , while the high-energy active site improves electrochemical oxidation of O 2 •− . The Mn 3 (PO 4 ) 2 /MXene as a sensing platform towards O 2 •− achieves a high sensitivity of 64.93 µA·µM −1 ·cm −2 , a wide detection range of 5.75 nM to 25.93 µM, and a low detection limit of 1.63 nM. Finally, the flexible heterostructured sensing platform realizes real-time monitoring of O 2 •− in live cell assays, offering a promising flexible biosensor towards exploring various biological processes.

The nicotinamide adenine dinucleotide (NAD+) level shows a temporal decrease during the aging process, which has been deemed as an aging hallmark. Nicotinamide mononucleotide (NMN), a key NAD+ precursor, shows the potential to retard the age-associated functional decline in organs. In the current study, to explore whether NMN has an impact on the intestine during the aging process, the effects of NMN supplementation on the intestinal morphology, microbiota, and NAD+ content, as well as its anti-inflammatory, anti-oxidative and barrier functions were investigated in aging mice and D-galactose (D-gal) induced senescent IPEC-J2 cells. The results showed that 4 months of NMN administration had little impact on the colonic microbiota and NAD+ content in aging mice, while it significantly increased the jejunal NAD+ content and improved the jejunal structure including increasing the villus length and shortening the crypt. Moreover, NMN supplementation significantly up-regulated the mRNA expression of SIRT3, SIRT6, nuclear factor E2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), the catalytic subunit of glutamate-cysteine ligase (GCLC), superoxide dismutase 2 (SOD2), occludin, and claudin-1, but down-regulated the mRNA expression of tumor necrosis factor alpha (TNF-α). Specifically, in the D-gal induced senescent IPEC-J2 cells, 500 μM NMN restored the increased mRNA expression of interleukin 6 (IL6ST), IL-1A, nuclear factor (NF-κB1), and claudin-1 to normal levels to some extent. Furthermore, NMN treatment significantly affected the mRNA expression of antioxidant enzymes including NQO1, GCLC, SOD 2 and 3, and GSH-PX1, 3 and 4. In addition, 200 μM NMN enhanced the cell viability and total antioxidant capacity and lowered the reactive oxygen species level of senescent IPEC-J2 cells. Notably, NMN restored the down-regulated protein expression of occludin and claudin-1 induced by D-gal. The above data demonstrated the potential of NMN in ameliorating the structural and functional decline in the intestine during aging.

The hypoglycemic effects and underlying molecular mechanisms of combining two dietary compounds, quercetin (Que) and d-chiro-inositol (DCI) on hepatic insulin resistance in C57BL/6J mice that were fed a high fat diet and saturated palmitic acid (PA)-treated hepatocytes were studied. Catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx) are important antioxidant enzymes . Insufficient secretion of these enzymes will inhibit the normal phosphorylation of insulin receptors in the liver, thereby limiting the transmission of insulin signals and producing insulin resistance. The results showed that QD (both Que and DCI) led to the recovery of the activity of CAT, SOD, GPx, suggesting that QD was helpful in preventing mice hepatic insulin resistance. In PA-induced human hepatoma cells (HepG2), QD improved the antioxidant level in cells. The fluoro-immunoassay showed that QD effectively reduced the translocation of protein kinase Cε to cell membranes in the HepG2. Western blotting results suggested that QD increased the signal transduction in the IRS-2/PI3K/Akt/FoxO1 pathway. Overall, these results showed that the combination of 5 μmol Que and 5 μmol DCI ameliorated high fat diet-induced hepatic insulin resistance.

DAAO is applied as a potential catalyst in the biosynthesis of L-PPT. However, its low solubility expression constrains its broader industrial application. Herein, a novel DAAO derived from Cladophialophora carrionii ( Cc DAAO) was identified, which demonstrated superior catalytic performance toward D-Ala (specific activity: 106.38 ± 1.21 U/mg, K m : 1.56 ± 0.06 mM), along with remarkable thermostability and broad substrate spectrum. Under optimal culture conditions, the soluble expression level of Cc DAAO was enhanced through a co-expression strategy with molecular chaperones, and the enzyme activity increased by 36.3% compared with the initial level. Subsequently, Cc DAAO was constructed as a fusion protein (CGD) with catalase from Geobacillus sp. CHB1 ( Gb CAT) and applied in a D-amino acid aminotransferase (DAAT)-mediated cascade system. In a 2 L reaction system, this cascade system achieved complete conversion (> 99%) of 1 M D,L-PPT within 8 h, exhibiting a yield of 11.26 g/L/h for PPO, which represents a significant improvement over existing reports. This study presents a promising practical approach for the industrial production of optically pure L-PPT.

Piceatannol , a natural derivative of resveratrol , has been shown to exert similar anti-oxidant and anti-inflammatory effects as resveratrol. However, it remains unknown whether piceatannol has hepatoprotective effect against acute liver injury. In this study, we investigated the in vivo effect of piceatannol on D-GalN/LPS-induced fulminant hepatic failure (FHF), and its in vitro effect on ER stress-inducing drug thapsigargin (TG)-induced proinflammatory cytokines production and ROS release. Our results indicated that piceatannol markedly decreased the mortality rate, reduced the serum levels of alanine transaminase and aspartic aminotransferase, ameliorated the liver damage induced by D-GalN/LPS in mice. In addition, piceatannol reduced the expression of proinflammatory cytokines, including TNF-α, IL-1β and IL-6, the expression of ER stress markers CHOP and phosphorylated-IRE1α, and the generation of oxidative stress in D-GalN/LPS-treated mouse liver. In vitro results were consistent with in vivo observations, demonstrating that piceatannol suppressed the secretion of proinflammatory cytokines, inflammasome activation and the production of ROS induced by TG with or without LPS priming in J774A.1 macrophages. Our study proposes piceatannol as a promising medication for preventing acute liver failure and the mechanisms may be related to its inhibitory effects on ER stress, inflammation and oxidative stress.

In conjunction with the pressing global issue of climate change and the associated concern over global warming, increasing interest has emerged in the exploration of carbon dioxide (CO2) as a resource for the generation of a diverse array of products intended to serve societal needs. This study presents the development of an ATP-free and NAD-balanced in vitro synthetic enzymatic biosystem (ivSEB), which comprises only five cascade thermophilic enzymes, designed for the synthesis of L-malate through CO2 fixation powered by the utilization of D-glucose as a substrate. This designed ivSEB yields two moles of L-malate from one mole of D-glucose and two moles of CO2. Through meticulous refinement of reaction conditions and enzyme loading amounts, this ivSEB has demonstrated its capability to produce 6.85 mM of L-malate via CO2 fixation from an initial 5 mM of D-glucose with a molar product yield of 68.5%, and 2.45 mM of L-lactate as a byproduct. In the pursuit of assessing the industrial feasibility of this ivSEB, the study further subjected the system to the utilization of a high concentration (45.70 mM) of D-glucose. Although this endeavor necessitates additional optimization for enhanced efficiency, the present findings herald the emergence of an alternative avenue for the sustainable production of L-malate through CO2 fixation, thus bearing substantial promise for addressing ecological and industrial imperatives.

Introduction: Sheep placenta extract (SPE) is a representative traditional medicinal substance that exhibits multiple experimentally validated physiological properties, including anti-aging effects, wound healing acceleration, antioxidant activity, and anti-inflammatory mechanisms. However, the mechanism by which SPE influences the delay of aging is still not yet clear. Methods: Exploring the effects of sheep placenta extract on D-gal induced senescence in a mouse model of aging by macrogenomics and metabolomics. Results: In the serum of aging mice treated with SPE, the levels of antioxidant function such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT) were notably higher compared to those in the blank group, whereas malondialdehyde (MDA) levels decreased. We revealed that SPE alleviated the changes in gut microbiota caused by aging in mice, with a significant decrease in the Firmicutes/Bacteroidetes (F/B) ratio in the gut. Furthermore, Akkermansia muciniphila (A. muciniphila), which is known for its regulating immune response and potential anti-aging effects, showed a significant increase of 1177.94%. The analysis of UHPLC-QE-MS combined with orthogonal partial least squares discriminant analysis (OPLS-DA) screening of differential metabolites in mouse serum metabolic profiles revealed a significant upregulation of cis-5,8,11,14,17-eicosapentaenoic acid (EPA) and triptolide in serum metabolites, following SPE treatment, which are commonly believed to have immunosuppressive, anti-inflammatory, anti-proliferative, and anti-tumor effects.Discussion: The role of SPE in ameliorating aging may be associated with the increased abundance of A. muciniphila in the gut microbiota and the accumulation of two metabolites, EPA and triptolide, in the serum.

D-(−)-Pantolactone (D-PL) is the key chiral intermediate used to synthesize calcium D-pantothenate. In a previous study, a conjugated polyketone reductase (CduCPR) was discovered to efficiently catalyse ketopantolactone (KPL) to D-PL with a high stereoselectivity. Nevertheless, this method was limited due to the expensive coenzyme, nicotinamide adenine dinucleotide phosphate hydrate (NADPH), and the poor stability of substrate KPL in pure aqueous buffer. Here, to overcome these problems, we developed a whole-cell biotransformation process to produce D-PL in a biphasic reaction system. Recombinant CduCPR and glucose dehydrogenase (BsuGDH) were co-expressed in Escherichia coli to simultaneously achieve the synthesis of D-PL and the regeneration of NADPH. The biphasic reaction system contained 15% dichloromethane (v/v) which significantly inhibited the undesirable hydrolysis of KPL. Approximately, 2.8% of 100 mM KPL was spontaneously hydrolysed to ketopantoic acid in the biphasic system, which was lower than 54% of pure aqueous buffer. The biphasic reaction system was optimized and scaled up to prepare D-PL. In a fed-batch biphasic reaction, the substrate KPL was continuously fed into the reactor after a batch reaction. The D-PL concentration reached 0.77 mol L−1 in the reaction mixture at 7 h, and its enantiomeric excess was 99%. This study presented an alternative method to asymmetrically synthesize enantiopure D-PL for further industrial application.

Modulating the microenvironment of single-metal active sites holds excellent promises for developing highly efficiently oxygen electrocatalysts. Herein, by combining theoretical predictions and experiments, we reported a general strategy to engineer the electronic properties of iron-nitrogen-carbon (FeN 4 /C) catalysts via the incorporation of the boron (B) atom for achieving improved catalytic activity in oxygen electrocatalysis. Our theoretical results revealed that B modulation effectively tunes the d-band center of the iron (Fe) active site to optimize its adsorption strength with oxygenated species, greatly enhancing oxygen reduction reaction (ORR) and oxygen evolution reactions (OER) activity. Our experimental measurements then confirmed the above theoretical predictions: the as-synthesized B-doped FeN 4 /C (Fe-N 4 -B) material can perform as an eligible bifunctional catalyst for ORR and OER in alkaline media, and its catalytic activity even outperforms the commercial noble metal benchmarks. The present findings provide a promising strategy to further design the advanced catalysts for a wide range of electrochemical applications.

A Fe/V-CN nanozyme with synergistically enhanced POD-like activity was rationally designed. • A sensitive cascade colorimetric platform for detecting gastric cancer-related D-amino acids was successfully constructed. • The assay supports reliable quantification of D-amino acids in complex human saliva matrices. The development of a practical and reliable cascade colorimetric method for detecting D-amino acids (DAAs) provides a promising tool for the prevention and preliminary diagnosis of gastric cancer. Herein, a bimetallic Fe V supported on graphitic carbon nitride (Fe/V-CN) nanozyme was rationally designed and synthesized via a simple annealing strategy. The atomic dispersion of Fe and V species, along with their coexisting multivalent states (Fe 2+ / 3+ , V 4+ / 5+ ) synergistically boosted the intrinsic peroxidase-like (POD) activity to Fe/V-CN, which were characterized by a high substrate affinity ( K m of 0.078 and 0.083 mM for TMB and H 2 O 2 ) and an accelerated catalytic turnover ( V max of 4.94 × 10 −7 and 7.76 × 10 −7 M·s −1 , respectively). Leveraging this improved activity, a cascade colorimetric sensing system was developed by integrating Fe/V-CN with D-amino acid oxidase (DAAO) for the specific detection of DAAs. The platform exhibited favorable analytical performance for D-proline (D-Pro) and D-alanine (D-Ala), offering wide linear ranges of 0–200 μM and 0–160 μM and low detection limits of 0.93 μM and 1.45 μM, respectively, combined with acceptable selectivity against common interferents. For real-sample analysis, the platform was successfully implemented in determine DAAs in human saliva, delivering satisfactory recoveries (95.8–105.7%) and good precision (RSD < 3.5%). This work presented a synergistic Fe/V bimetallic nanozyme and a practical sensing strategy, demonstrating the feasibility of DAAs detection in complex biological matrices. Chirality is one of the ubiquitous phenomena in biological systems. The left handed (L-) amino acids and right handed (D-) sugars are normally found in proteins and in RNAs and DNAs, respectively. The effect of chiral surfaces at the nanoscale on cellular uptake has, however, not been explored. This study reveals for the first time the molecular chirality on gold nanoparticles (AuNPs) functions as a direct regulator for cellular uptake. Monolayers of 2-mercaptoacetyl-L(D)-valine (L(D)-MAV) and poly(acryloyl-L(D)-valine (L(D)-PAV) chiral molecules were formed on AuNPs surface, respectively. The internalized amount of PAV-AuNPs was several times larger than that of MAV-AuNPs by A549 and HepG2 cells, regardless of the chirality difference. However, the D-PAV-AuNPs were internalized with significantly larger amount than the L-PAV-AuNPs. This chirality-dependent uptake effect is likely attributed to the preferable interaction between the L-phospholipid-based cell membrane and the D-enantiomers.
