Zinc is a common redox element and plays a vital role in immune response, catalysis, structure, and signal regulation. Tracing intracellular Zn2+ dynamics is, therefore, crucial for understanding physiological processes. Since a glass nanopipette is a good sensing platform for single-cell detection with little damage to the cell membrane, in this study, we developed DNAzyme-functionalized glass nanopipettes (D-GNs) for single-cell Zn2+ detection based on the catalytic activity of Zn2+-dependent DNAzymes. In the presence of Zn2+, DNAzyme-modified gold nanoparticles (D-AuNPs) were released from the inner wall of the nanopipette, leading to a change in ionic current rectification. D-GNs exhibited good sensing performance in vitro and can be applied to effectively detect Zn2+ in HeLa cells under electrical stimulation. This functional nanopipette-based platform offers a new strategy for probing intracellular ion variations and evaluating the physiological state of individual cells.

Dual-mode sensor uses CeO 2 NR@Pt and Mg 2+ -dependent DNAzyme for AFB 1 detection with broad linear range and low LODs. • Signal amplification via CHA and DNAzyme cycling boosts sensitivity and specificity. • CeO 2 NR@Pt show strong peroxidase activity, PEI/rGO@AuNRs enhance electrode performance. • Practical tests in peanuts and corn confirm high recovery rates and stability for real-world use. This study developed a dual-mode colorimetric/electrochemical aptasensor for detecting aflatoxin B1 (AFB 1 ) in food, utilizing cascade amplification of bipedal Mg 2+ - dependent DNAzyme and catalytic hairpin assembly (CHA). The system employed CeO 2 nanorods (CeO 2 NRs) as blocking probes for toluidine blue (TB), with in-situ Pt modification enhancing peroxidase-like activity. Gold nanorods (AuNRs) and polyethyleneimine-modified reduced graphene oxide (PEI/rGO) served as substrate materials to optimize the sensing platform. After amplification, electrode-captured CeO 2 NRs@Pt blocked TB charge transfer while enabling colorimetric detection via peroxidase activity. The sensor achieved wide linear ranges (1 × 10 −4 -1 × 10 2 ng/mL electrochemical; 1 × 10 −1 -5 × 10 2 ng/mL colorimetric) with low detection limits (3.7 × 10 −5 ng/mL and 5.6 × 10 −2 ng/mL, respectively). The method demonstrated excellent specificity, stability, and recovery rates in real-sample analysis, offering a robust approach for food safety monitoring. Sensitive and reliable quantification of Ochratoxin A (OTA) in food is essential for ensuring food safety and protecting public health. In this study, a dual-mode colorimetric and Surface-Enhanced Raman Spectroscopy (SERS) biosensor was developed by integrating DNAzyme-responsive hydrogel and Au@Pt@4-MBN nanozyme for signal amplification. Upon introduction of OTA, a specific cascade reaction was triggered, activating the generated DNAzymes and leading to the disintegration of the hydrogel network and the subsequent release of encapsulated nanozymes. The extent of hydrogel disintegration exhibited a positive correlation with OTA concentration, enabling straightforward quantitative detection. The biosensor demonstrated excellent sensitivity, with detection limits of 0.0668 ng mL⁻¹ (colorimetric) and 0.0342 ng mL⁻¹(SERS). When applied to spiked corn samples, it achieved high recoveries of 98.70%-100.93% (colorimetric) and 95.6%-104.27% (SERS), confirming its reliability. The method also demonstrated high practical applicability for authentic non-spiked samples, with a positive detection rate of 10% that was consistent with the results of the national standard method. This robust platform offers significant potential for sensitive mycotoxin monitoring in food and environmental safety. Graphical The alternative text for this image may have been generated using AI.

Manipulating the expression of cellular genes through efficient CRISPR/Cas9 delivery is rapidly evolving into a desirable tumor therapeutics. The exposure of CRISPR/Cas9 to a complex external environment poses challenges for conventional delivery carriers in achieving responsive and accurate release. Here, we report a Trojan horse-like nanocapsule for the on-demand delivery of CRISPR/Cas9 in a microRNA-responsive manner, enabling precise tumor therapy. The nanocapsule comprises a nanoassembled, engineered DNAzyme shell encasing a Cas9/sgRNA complex core. The DNAzyme, functioning as a catalytic unit, undergoes a conformational change in the presence of tumor-associated microRNA, followed by activating a positive feedback-driven autonomous catabolic cycle of the nanocapsule shell. This catabolic cycle is accomplished through chain reactions of DNAzyme “cleavage–hybridization–cleavage”, which ensures sensitivity in microRNA recognition and effective release of Cas9/sgRNA. Utilizing this Trojan horse-like nanocapsule, as low as 1.7 pM microRNA-21 can trigger the on-demand release of Cas9/sgRNA, enabling the specific editing of the protumorigenic microRNA coding gene. The resulting upregulation of tumor suppressor genes induces apoptosis in tumor cells, leading to significant inhibition of tumor growth by up to 75.94%. The Trojan horse-like nanocapsule, with superior programmability and biocompatibility, is anticipated to serve as a promising carrier for tailoring responsive gene editing systems, achieving enhanced antitumor specificity and efficacy.

DNA methylation serves as a critical epigenetic biomarker for early disease screening and clinical diagnostics, exemplified by the FDA-approved detection of methylated Septin9 gene in peripheral blood for colorectal cancer (CRC) screening. Conventional CRISPR/Cas12a-based detection systems rely on fluorophore-quencher-labeled ssDNA probes to transduce trans-cleavage activity into fluorescence signal, which yet suffers from limitations including complex probe synthesis, background noise induced by incomplete quenching and high cost. In this study, we developed a cascaded and label-free CRISPR/Cas12a system for Septin9 methylation detection using DNAzyme and split DAP-10. The method employed methylation-sensitive Aci I digestion to selectively hydrolyze non-methylated DNA, leaving methylated Septin9 DNA fragment intact to activate CRISPR/Cas12a system. Activated Cas12a triggered trans-cleavage of DNAzyme, a bifunctional molecule that acted as both trans-cleavage substrate of Cas12a and hydrolysis regulator of linker DNA that facilitated split DAP-10 reassembly to light-up Auramine O (AO) fluorescence proportionally to methylation levels. The assay achieved a linear range from 2 to 200 nM with a detection limit of 1.74 nM and robust performance in accuracy, precision, and selectivity. The proposed method was successfully applied in methylated Septin9 detection in cell lines, clinical tissues and blood samples, which indicated application potential for CRC screening.

Cadmium ion (Cd 2+ ) is a highly toxic heavy metal ion that poses serious risks to environmental safety and human health. Its accumulation through food chains and drinking water can result in renal dysfunction, neurological damage, and carcinogenic effects. Therefore, the development of simple, sensitive, and field-deployable detection methods for Cd 2+ is of great importance. In this study, a novel approach was employed using graphene oxide–systematic evolution of ligands by exponential enrichment (GO-SELEX) to screen aptamer specific to Cd 2+ . The method reduced the screening time by 30%–50% compared with conventional SELEX methods. The selected aptamer was subsequently integrated with catalytic hairpin assembly (CHA) and a G-quadruplex DNAzyme to construct a colorimetric biosensor for Cd 2+ detection. Signal transduction was achieved via the peroxidase-like catalytic oxidation of ABTS in the presence of hydrogen peroxide, enabling visual and spectrophotometric readout without the need for complex instrumentation or sample pretreatment. Under optimized conditions, the biosensor exhibited a linear detection range from 0.1 to 20 μM with a detection limit of 57 nM. The assay demonstrated excellent selectivity toward Cd 2+ over other common metal ions and satisfactory performance in spiked environmental water samples. The findings of the study demonstrate that the proposed biosensor provided a cost-effective and practical platform for on-site monitoring of Cd 2+ . It holds strong potential for real-world applications, such as assessments of water quality at riverbanks or evaluations of industrial effluents, offering a seamless transition from laboratory research to field deployment.

Herein, we propose an all-DNA self-assembled G-quadruplex/hemin DNAzyme nanosphere (GHN) and apply it to construct an electrochemical biosensor for sensitive detection of kanamycin (KAN). Unlike conventional metal-core-dependent biosensors, the GHN eliminates inorganic components, offering superior biocompatibility, simplified synthesis, and reduced environmental hazards. High loading of G-rich sequences was achieved by performing multiple branches of the hybridization chain reaction on self-assembled DNA spherical core. In addition to exhibiting excellent catalytic performance, the GHN inherently provides a strong current response, facilitating highly sensitive detection of target analytes. Through a one-step displacement reaction triggered by the target, the GHN is captured on the electrode surface, generating a measurable current signal. Under optimal experimental conditions, the electrochemical signal of the proposed sensor showed a good linear relationship with the logarithm of the KAN concentration over the range of 0.1 pM to 1 μM, with a detection limit of 71 fM (S/N = 3). For practical applications, the sensor achieved 93.98% to 102.0% recovery in spiked milk samples, demonstrating its reliability in complex matrices. The target-independent synthesis of GHN allows facile reconfiguration by altering aptamer sequences, enabling universal detection of environmental pollutants and biomarkers. This platform offers a new approach for early monitoring of antibiotic residues, which is expected to provide valuable references for the formulation of relevant environmental protection policies and the safeguarding of human health.

The authors describe an ultrasensitive amperometric enzymatic assay for uranyl ion. It is based on the use of mesoporous silica nanoparticles (mesoSiNPs) loaded with Methylene Blue (MB) and functionalized with an UO 2 (II)-dependent DNAzyme. The electroactive label MB was sealed in the inner pores of the mesoSiNPs along with double stranded DNA (containing the DNAzyme and the substrate strand). In the presence of UO 2 (II), the DNAzyme is actived to cleave the substrate strands. This leads to the cleavage of the caps and the release of MB from the mesoSiNPs. The amount of released MB depends on the concentration of UO 2 (II) and can be determined amperometrically, best at a working voltage of −0.25 V (vs SCE), by using a chitosan coated carbon paste electrode. Response is linear in the 20 pM to 0.1 nM UO 2 (II) concentration range, and the detection limit is as low as 0.15 pM. Recoveries from spiked samples varied from 91.3 to 99.4%. The assay is highly specific, selective, and not interfered by other metal ions.

Due to similar charges, atomic radii, and chemical properties of most metal ions , the selective sensing of these metal ions and imaging in vivo is still a challenge. A DNAzyme assemblied and near-infrared (NIR) light excited nanosensor was developed to detect and image Pb 2+ . In this nanosensor, NaYF 4 :Yb, Er upconversion nanoparticles (UCNPs) introduced as NIR-to-Vis transducer were the donor of luminescence resonance energy transfer (LRET), and DNAzyme-functionalized black hole quencher 1 (BHQ1) acted as energy transfer acceptor. This proposed nanosensor was applied to detecting Pb 2+ in solution with high sensitivity and selectivity . Furthermore, we have successfully demonstrated the imaging ability of this nanosensor towords Pb 2+ in living cells and early-stage zebrafish with negligible autofluorescence and good photostability. The UCNP-DNAzyme nanosensor would enrich the method of imaging Pb 2+ in vivo, and might serve as a potential tool for understanding the metabolic pathways of Pb 2+ and the mechanism of lead poisoning in biological system.

A short G-rich sequence with three G-tracts can form a G-triplex (G3), a recently identified noncanonical DNA structure . Until now, very limited functional study and application of G3 is reported. Herein, we integrated G3 with isothermal exponential amplification reaction (EXPAR) for achieving simple and sensitive biosensing strategy. In this strategy, the cascade EXPAR cycles produces larger numbers of short G-rich sequences, which self-assemble into G3 structure and then bind hemin to form G3/hemin DNAzyme. This G3/hemin DNAzyme–based EXPAR strategy could detect as low as 4.7 fM target DNA with colorimetric detection, and the sensitivity of G3/hemin DNAzyme–based EXPAR strategy was much higher than that of the conventional G-quadruplex/hemin DNAzyme–based EXPAR strategy. We explored the reason for higher sensitivity of G3/hemin DNAzyme–based EXPAR strategy. The experimental results demonstrated that G3/hemin DNAzyme is an ideal signal generator for EXPAR-based biosensing platform. This work opens a new avenue to develop effective signal amplification strategy for ultrasensitive biosensing. This work is also helpful for a deeper understanding of G3 structure and the future application of G3.

Ultrasensitive detection for low concentration and abnormal expression of microRNA-155 (miR-155) in serum (fM level) at the early stage of cancer still exist great challenges. Therefore, a dual-model photoelectrochemical biosensor was developed based on label-free trimetallic nanoprobe and Mg 2+ dependent DNAzyme driven DNA walker to generate ratiometric dual-signal output for sensitive biosensing of miR-155 from breast cancer cells (MCF-7) and cervical cancer cells (HeLa). In signal amplification strategy, PtCo@Prussian Blue nanozyme was selected as a signal nanoprobe with high catalytic activity and large specific surface area, while two dimensional MXene/AuNPs as surface modification material was applied to improve the active sites of the electrode. With the presence of the target, the DNAzyme assisted DNA walker process was initiated and abundant signal nanoprobe was introduced to the electrode surface , producing high current response and weak ultraviolet-visible spectrophotometry (UV–vis) response. Hence, the biosensor for miR-155 shows a low limit of detection of 1.2 fM from the linear range of 5 fM to 5 nM, exhibiting excellent stability and reproducibility. Moreover, the proposed biosensor demonstrates superior performance for miR-155 in complex human serum samples and tumor cell lysates, presenting great application potential for early-stage clinical diagnosis.

Based on the different affinities of graphene oxide (GO) toward ssDNA and dsDNA, a fluorescence assay using split G-rich probes and magnetic GO (Fe3O4/GO) was developed for authentication of Pseudostellaria heterophylla based on the ITS sequences. In this assay, the probes mixed with Fe3O4/GO nanoparticles, hybridize with T-DNA for a double-stranded structure and then their G-rich sequences can fold into a G-quadruplex which combines hemin for DNAzyme. After magnetic separation, the DNAzyme supernatant can catalyze 2′,7′-dichlorodihydrofluorescein diacetate (H2DCFDA) oxidation with H2O2 and release a fluorescence signal. Herein, the Fe3O4/GO nanoparticles play an important role in decreasing the background signal by absorbing the excess probes and hemin. This assay allows the analysis of T-DNA in the range of 1.0 × 10−14 to 1.0 × 10−7 mol L−1 with a low detection limit of 1.87 × 10−15 mol L−1. In addition, this newly designed assay allows specific authentication of Pseudostellaria heterophylla from different provenances.
