A monoclonal antibody against native grass carp IgM showed cross-reactivity with multiple cyprinid species. • Bacterial and viral infections induced expansion and local aggregation of splenic IgM + B cells in two cyprinid species. • Bacterial and viral infections promoted the proliferation of splenic IgM + B cells in two cyprinid species. Humoral immunity is a central component of host defense in teleost fish, yet the dynamics of IgM + B cells during infection remain poorly understood. In this study, we generated and characterized a monoclonal antibody (mAb) against native IgM protein purified from grass carp ( Ctenopharyngodon idella ) sera. This mAb specifically recognized the recombinant grass carp IgM heavy chain expressed in the HEK293 cells and showed cross-reactivity with IgM molecules from multiple cyprinid species, including rare minnow, zebrafish, black carp, and common carp, but not with IgM from largemouth bass. Flow cytometric analysis showed that IgM + B cells were abundant in the spleen and head kidney of grass carp, accounting for 35.40 ± 3.15% and 27.63 ± 3.97% of total leukocytes, respectively. Using this antibody, we further investigated IgM + B-cell responses in grass carp and rare minnow after infection with Aeromonas hydrophila or grass carp reovirus genotype II. Both bacterial and viral infections significantly enhanced the proliferation and local aggregation of splenic IgM + B cells. Moreover, EdU + proliferating cells were preferentially enriched within localized IgM + B-cell clusters in the spleens of infected fish. Taken together, our work provides a useful antibody tool for studying B-cell responses and reveals that bacterial and viral infections induce proliferation and localized aggregation of splenic IgM + B cells in teleost fish, highlighting infection-induced remodeling of splenic IgM + B-cell responses.

Seafood, especially marine fish, is highly prone to spoilage during processing, transportation, and storage. It releases pungent trimethylamine (TMA) gas, which severely affects food quality and safety. Metal–oxide–semiconductor (MOS) gas sensors for TMA detection offer a rapid, convenient, and accurate method for assessing fish freshness. Indium oxide (In2O3) has shown potential as an effective sensing material for the detection of TMA. In this work, one-dimensional In2O3nanowires with different grain sizes and levels of crystallinity were synthetized using the electrospinning technique and underwent different thermal calcination processes. Gas-sensing tests showed that the In2O3–3 °C/min–500 °C gas sensor exhibited an outstanding performance, including a high response (Ra/Rg= 47.0) to 100 ppm TMA, a short response time (6 s), a low limit of detection (LOD, 0.0392 ppm), and an excellent long-term stability. Furthermore, the sensor showed promising experimental results in monitoring the freshness ofLarimichthys crocea(L. crocea). By analyzing the relationship between the grain size and crystallinity of the In2O3samples, a mechanism for the enhanced gas-sensing performance was proposed. This work provides a novel strategy for designing and fabricating gas sensors for TMA detection and highlights their potential for broad applications in real-time fish freshness monitoring.

A colorimetric method for fish freshness monitoring was developed. After coating magnetic Fe 3 O 4 nanoparticle with zeolitic imidazolate framework-8 (ZIF-8), the horseradish peroxidase (HRP) was adsorbed onto it with high efficiency due to the large surface area of ZIF-8, the mesoporous SiO 2 was further decorated onto the Fe 3 O 4 @ZIF-8@HRP to form Fe 3 O 4 @ZIF-8@HRP@SiO 2 nanostructure, in order to minimize the release of HRP, and enhance the stability and catalytic activity of HRP. The morphology, elemental composition, surface functional groups of Fe 3 O 4 @ZIF-8@HRP@SiO 2 were characterized by transmission electron microscopy, X-ray diffraction, energy dispersive spectroscopy, Fourier transform infrared spectroscopy. Hypoxanthine (HX), which formed due to the degradation of adenosine triphosphate in fish, can be oxidized by xanthine oxidase and generated H 2 O 2 intermediate. Thereafter, in the presence of H 2 O 2 , HRP within Fe 3 O 4 @ZIF-8@HRP@SiO 2 nanostructure would catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to a blue product of TMBox, allowing an indirect colorimetric sensing of HX content. Under optimal conditions, HX was selectively and sensitively detected in the range of 0–100 μM, with a detection limit of 0.133 μM. Finally, the proposed method was applied to determine the HX content in grass carp continuously stored at 4 °C and 30 °C for 96 h.

Rice-fish co-culture systems, as compared to fish monoculture, offer a sustainable agriculture model that balances productivity with environmental conservation. While the important role of gut microbiome in fish growth and health is increasingly recognized, the impact of different farming practices on fish quality and their gut microbiome remains insufficiently understood. To address this gap, we conducted a comprehensive survey of 292 carps ( Cyprinus carpio ) raised in four rice-fish co-culture systems (RF) across China, comparing them with site-matched pond systems (PF). Through a combination of amplicon sequencing, metabolomic analysis, and bioinformatics, we investigated changes in carp quality, as well as alterations in gut microbiota and metabolites, and explored the potential links between them. Our results reveal that RF system significantly improve carp quality and alter gut metabolite profiles. Further analysis of gut microbial subgroup showed that RF co-culture induced homogenization in the community structure of rare microbial taxa. Ecological network analysis indicated that RF enhances the complexity and stability of microbial networks, particularly among rare taxa, which play a previously underestimated role in gut ecosystems. Community assembly analysis suggested that deterministic processes more strongly shape the microbial communities in carps from RF system, promoting habitat-dependent selection. By developing a bi-network model to explore host-microbe interactions, we identified key microbial taxa, such as Cetobacterium , that are closely associated with beneficial traits in carp quality. Overall, this study highlights the potential of Rice-fish co-culture to foster beneficial host-microbe interactions, providing a microbial ecological perspective on understanding aquaculture safety and sustainability.

Ofloxacin (OFL), a third-generation quinolone antibiotic, has been reported to cause adverse multisystem reactions in humans after residual ingestion. The aim of this study was to develop a gold core-platinum shell nanozymes (Au@Pt NPs) via a layer-by-layer seed growth method for the rapid detection of OFL. The synthesized Au@Pt NPs, with a 25 nm gold core and 6 platinum shells, exhibited robust peroxidase-like activity. Kinetic analysis revealed a Michaelis constant (Km) of 0.41 ± 0.04, a catalytic constant (Kcat) of 1.41 × 10 5 , and a catalytic efficiency (Kcat/Km) of 3.40 × 10 5 . An indirect competitive Enzyme-linked immunosorbent assay (ic-ELISA) and an immunochromatographic strip were developed based on these nanozymes. The ic-ELISA demonstrated a linear range of 0.2–7.35 ng/mL, an IC 50 of 0.96 ng/mL, and a limit of detection (LOD, IC 10 ) of 0.03 ng/mL. The immunochromatographic strip exhibited a detection range of 0.060–4.512 ng/mL, an IC 50 of 0.521 ng/mL, and an LOD of 0.017 ng/mL, with a detection time of 5–7 min. Applied to real samples, the method achieved recoveries of 86.48 %-116.96 % in fish and 73.90 %-123.37 % in chicken, with relative standard deviations (RSDs) below 20 %. Therefore, this Au@Pt NP-based immunoassay demonstrated a rapid, sensitive, and specific method for screening antibiotic residues in food.

NH 3 is widely exist in food spoilage processes, and its concentration is closely related to environmental safety and human health. PANI is diffusely used in gas sensing due to its good sensing performance to NH 3 , but pure PANI sensors have problems such as low response to NH 3 and baseline drift. In order to improve the performance, Ti 3 C 2 T x , characterized by a large specific surface area and outstanding electron transport properties, was incorporated into polyaniline (PANI) to construct a Schottky junction-based composite sensor for ammonia detection. The incorporation of Ti 3 C 2 T x effectively enhances the protonation degree of PANI and introduces surface functional groups (−OH and − O) with strong affinity for NH₃ molecules. These synergistic effects result in a markedly improved gas-sensing performance compared to that of pristine PANI. The composite material demonstrates a significantly enhanced gas response of 267 % to 50 ppm NH₃ at room temperature, along with a lowered detection limit of 1 ppm. In addition, it exhibits excellent repeatability and selectivity, making it particularly suitable for monitoring ammonia emissions associated with food spoilage. In addition, the sensor still maintains stable operation under the condition of relative humidity of 70 %, showing excellent humidity resistance and broad practical application prospects.

High carbohydrate (HC) diets have become a conventional approach in modern aquaculture. However, this feeding strategy frequently compromises the health of high-value economic fish species, which might ultimately impair the farmers’ final profits. Our prior research indicated that Saccharina japonica efficiently reduces harm from HC diets, likely due to its sodium alginate (SA) content. In this study, Monopterus albus were randomly grouped into treatments: a normal diet (20 % carbohydrate, NC), a high carbohydrate diet (32 % carbohydrate, HC), and high carbohydrate diets supplemented with 0.5 % SA (LA). Then our outcomes evinced that LA improves various health-related parameters, including reduction of hepatosomatic index (HSI), serum glucose, triglycerides, and aminotransferase levels, as well as the decrease in hepatic lipid droplets, glycogen, and collagen fiber content. Our transcriptomic analysis on liver tissue showed that detoxification-related differentially expressed genes (DEGs) were less active in HC group than in NC group. In contrast, DEGs linked to fatty acid synthesis increased in LA group versus HC group. A combined analysis of metagenomics and the short-chain fatty acids (SCFAs) profile in the posterior intestinal digesta indicated that LA diets greatly increased concentration of acetic acid. This effect was accompanied by a concurrent reduction in the abundance of the species Marinobacter guineae relative to HC diets. Overall, sodium alginate has demonstrated a substantial enhancement in Monopterus albus’ health by mitigating liver injury and ameliorating intestinal microbiota dysbiosis.

Antimicrobial peptides are important components of the host innate immune system, forming the first line of defense against infectious microorganisms. Among them, liver-expressed antimicrobial peptides (LEAPs) are a family of antimicrobial peptides that widely exist in vertebrates. LEAPs include two types, named LEAP-1 and LEAP-2, and many teleost fish have two or more LEAP-2s. In this study, LEAP-2C from rainbow trout and grass carp were discovered, both of which are composed of 3 exons and 2 introns. The antibacterial functions of the multiple LEAPs were systematically compared in rainbow trout and grass carp. The gene expression pattern revealed that rainbow trout and grass carp LEAP-1, LEAP-2A, LEAP-2B and/or LEAP-2C were differentially expressed in various tissues/organs, mainly in liver. After bacterial infection, the expression levels of LEAP-1, LEAP-2A, LEAP-2B and/or LEAP-2C in the liver and gut of rainbow trout and grass carp increased to varying degrees. Moreover, the antibacterial assay and bacterial membrane permeability assay showed that rainbow trout and grass carp LEAP-1, LEAP-2A, LEAP-2B and LEAP-2C all have antibacterial activities against a variety of Gram-positive and Gram-negative bacteria with varying levels through membrane rupture. Furthermore, cell transfection assay showed that only rainbow trout LEAP-1, but not LEAP-2, can lead to the internalization of ferroportin, the only iron exporter on cell surface, indicating that only LEAP-1 possess iron metabolism regulation activity in teleost fish. Taken together, this study systematically compared the antibacterial function of LEAPs in teleost fish and the results suggest that multiple LEAPs can enhance the immunity of teleost fish through different expression patterns and different antibacterial activities to various bacteria.

The complement system is composed of a complex protein network and is pivotal to innate immunity. Complement 3 (C3) is a critical protein in the complement cascade and participates in complement activation and immune defense. In this study, C3 from Nile tilapia (Oreochromis niloticus) was cloned and its function in resisting pathogen infection was characterized. The full length of OnC3 open reading frame is 4974 bp, encoding 1657 aa, and the predicted protein mass weight is 185.93 kDa. The OnC3 amino acid sequence contains macroglobulin domains. The expression pattern of OnC3 mRNA in the tissues of healthy fish was detected, with the highest in the liver and the lowest in the muscle. After challenged withStreptococcus agalactiaeandAeromonas hydrophila, the expression of OnC3 mRNA was significantly up-regulated in the liver, spleen, and head kidney. Further, the recombinant OnC3 protein alleviated the inflammatory response and pathological damage of tissues after infected withS. agalactiae. Moreover, the OnC3 promoted the phagocytosis of monocytes/macrophages toS. agalactiae. The data obtained in this study provide a theoretical reference for in-depth understanding of C3 in host defense against bacterial infection and the immunomodulatory roles in teleost fish. Keywords:Oreochromis niloticus;complement 3 (C3);Streptococcus agalactiae;inflammatory response;phagocytosis

Bacterial contamination is a significant concern in food safety. Traditional methods, though being a gold standard for bacterial detection, are time-consuming. In this work, we managed to establish a simple and versatile magnetic-assisted microfluidic method for rapid bacterial detection of fish muscle products, by manipulating anti-human IgG functionalized magnetic beads in a zig-zag shaped microfluidic channel, increasing the probability for bacteria capture. The captured bacteria were characterized by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS). This method is capable of isolating Escherichia coli , Staphylococcus aureus and Klebsiella pneumoniae from 5 μL of sablefish sarcoplasmic protein sample, and detecting Escherichia coli in the range of 6.0 to 6.0×10 4 CFU/mL with a detection limit of 6 CFU/mL. Bacterial growth on salmon sashimi during its period of storage was successfully monitored. The current protocol holds great potential for pathogen detection and microbial control in the food industry.

In the present study, E. coli was taken as a model bacterium, anti- E. coli functionalized magnetic beads were constructed and used to capture E. coli from aqueous extracts of fish sarcoplasmic protein (FSP) and fish muscle protein of sablefish. The excellency of the reproducibility of the present protocol was demonstrated by capturing E. coli from sablefish FSP extracts. The presence of 10 CFU/mL E. coli is still detectable. A microbial safety test on the surface of fish muscle was successfully performed. The bacterial identification accuracy from samples with different matrices was found to be excellent with RSD = 3%. High specific detection of target bacteria in complex biological samples was testified by spiking Staphylococcus aureus and Klebsiella pneumoniae in samples as interference. Ten biomarker ions were discovered for E. coli ’s recognition. It is promising to apply the present protocol in bacterial analysis in muscle food samples to ensure their safety.

Cellulose/fish oil oleogels were formed via capillary suspension. • Oleogel/hydrogel ratio regulated phase inversion and network coupling. • BG50 showed compact network with optimal cohesiveness and deformation resistance. • Oleogel/hydrogel ratio tuned IDDSI swallowing performance from level 6 to level 5. Dysphagia-friendly foods require tunable texture, structural stability, and controlled oral deformation. In this study, cellulose/fish oil oleogels were constructed through capillary suspension structuring, and further combined with fish gelatin hydrogels to produce bigels with adjustable swallowing properties. Oleogel formation was jointly regulated by cellulose and water contents. The oleogel containing 25 wt% cellulose and 5 wt% water formed a stable bridged network with improved oil-holding capacity, reaching 73.32 ± 0.45%, and its structural recovery increased from 11.85% to 66.00% with increasing cellulose content. The oleogel/hydrogel ratio markedly affected bigel phase organization, rheology, texture, stability, and IDDSI performance. Increasing oleogel fraction induced a structure transition from hydrogel-continuous phase structure to semi-bicontinuous transitional structure and finally to an oleogel-rich structure. BG50 exhibited favorable structural and mechanical performance, with compact network, enhanced gel strength, and stable deformation resistance. IDDSI tests showed that bigels containing 10∼50% oleogel corresponded to Level 6, whereas those containing 70∼90% oleogel shifted to Level 5. Additionally, bigels with an oleogel content below 50% exhibited higher centrifugal stability, supporting their potential for processing, storage, and transport. Overall, this study provides new insights and methods for the design of dysphagia-friendly foods. Download: Download high-res image (323KB) Download: Download full-size image
