A synergistic nanoprobe enables precise, safe, non-radiative skeletal imaging. • Multi-targeting enables precise enrichment throughout the skeleton. • AA@TFN provides a versatile platform for bone disease diagnosis and monitoring. Early diagnosis of bone-related diseases remains a critical clinical challenge, attributable to persistent limitations in imaging accuracy and safety. While near-infrared Region II fluorescence (NIR-II FL) imaging confers substantial advantages for deep-tissue visualization, its clinical translation is hampered by the inadequate targeting specificity of conventional contrast agents. Here, we developed Au-Ag@Apo-Tf/-N 3 (AA@TFN), a synergistic targeting gold–silver nanocluster probe engineered with glutathione and azide functionalities, which achieved bone-specific enrichment via a hierarchical multi-targeting mechanism. AA@TFN first anchored to hydroxyapatite in the bone matrix through glutathione carboxyl-mediated chelation, then targeted transferrin receptors highly expressed in bone marrow, and finally endured macrophage-mediated uptake facilitated by azide-enhanced bone affinity. A synergistic targeting strategy enabled real-time, high-resolution NIR-II FL imaging across the complete skeleton, with enhanced probe enrichment at lesion sites driven by microenvironmental changes. AA@TFN enabled visualization of synovial inflammation in rheumatoid arthritis models and millimeter‑scale microlesions in multiple myeloma models. Furthermore, the probe exhibited excellent biocompatibility and favorable safety profiles, underscoring its strong translational potential. A versatile platform was established for the diagnosis of bone diseases and non-invasive monitoring of therapeutic efficacy. Fat embolism syndrome (FES) is a serious complication following bone fracture, primarily resulting from the release of fat microparticles into circulation. Its diagnosis remains challenging due to the absence of specific imaging techniques and laboratory tests. Near-infrared II (NIR-II, 1000–1700 nm) fluorescence imaging (FI) has gained significant attention owing to its high tissue penetration and low background signal. While FI has been widely employed in basic research and clinical practice, its application in FES has not been explored. In this study, a NIR-II organic fluorescent small molecule, FD-1080-C6, which exhibits fat affinity and viscosity responsiveness, is designed to enable FES diagnosis, for the first time. In the high-viscosity microenvironment of FES, the lipophilic molecule FD-1080-C6, which possesses a donor–acceptor–donor (D–A–D) structure, undergoes restricted single-bond rotation, promoting the formation of a rigid planar conformation and resulting in enhanced fluorescence intensity. In vitro evaluation confirms its responsiveness to lipid droplets, affinity for adipose tissue, and viscosity-dependent fluorescence changes. Furthermore, its specificity for NIR-II FI of FES is validated in the mouse. Additionally, FD-1080-C6 exhibits stable photothermal properties. FD-1080-C6 enables early diagnosis and longitudinal monitoring of disease progression in FES mice, offering a promising strategy for the management of FES.

Near-infrared-II (NIR-II) photons hold great potential in assisting non-invasive imaging and optical signal sensing. However, the integration of broadband NIR-II light sources with photodetectors to improve the imaging sensitivity and spatial resolution is highly demanded. Herein, we have developed MgO-Li 2 SrGeO 4 :Cr 4+ composite ceramics, which demonstrate a broad NIR-II emission band centered at 1160 nm originating from spin-allowed 3 T 2 → 3 A 2 ( 3 F) transition of Cr 4+ . The incorporated nano-MgO particles serve as light-guiding channels while filling the pores and prevents the reduction of Cr 4+ , enhancing the external quantum efficiency and thermal conductivity of the composite ceramics to 53% and 17.8 W·m –1 ·K –1 , respectively. The dynamic wafer-scale luminescent ceramic wheel delivers a record broadband NIR-II emission output power of 1.9 W under 20 W·mm -2 red laser excitation, which facilitates imaging sensing of commercial PbS photodetectors by enhancing their photocurrents and signal-to-noise ratios. These results establish the feasibility of high-power broadband NIR-II light sources integrated with photodetectors, and further demonstrate the combination of intelligent algorithms toward high-resolution optical imaging.

Temporal compressive imaging enables the reconstruction of multi-frame dynamic scenes from a single exposure by multiplexing spatially encoded temporal slices. This approach mitigates integration time loss and facilitates high-speed imaging via low-frame-rate cameras. Herein, a second near-infrared (NIR-II) temporal compressive wide-field microscope (TCWM) is established, utilizing the EfficientSCI algorithm for frame reconstruction. Simulations initially verify the transferability of EfficientSCI to biological microscopic imaging. Furthermore, mask acquisition with a spectrally matched NIR backlight proves effective for the high-fidelity reconstruction of fluorescent targets. Utilizing a conventional NIR camera of 25 frames per second (FPS), NIR-II TCWM achieves a maximum in vitro reconstruction rate of 333.3 FPS, surpassing the native frame rate by over 13-fold. Additionally, its robust anti-scattering capability for dynamic imaging at significant depths is substantiated using Intralipid. Finally, NIR-II TCWM is applied to in vivo cerebrovascular imaging at depths up to 300 µm. With a 100 FPS reconstruction frame rate, NIR-II TCWM successfully demonstrates simultaneous, high-speed tracking of multiple fluorescent particles across multiple vessels, highlighting the potential for comprehensive deep-tissue hemodynamic research in living organisms.

Near-infrared-II carbon dots offer exceptional deep-tissue penetration for biomedical imaging, but challenges remain in their synthesis and photoluminescence mechanisms. Here, we report three carbon dots (CDs-1, CDs-2, CDs-3) with tunable emission from the visible to the Near-infrared-II (480–1265 nm), synthesized by constructing extended aniline-based frameworks from p -phenylenediamine. Combined structural and density functional theory analyses reveal that the Near-infrared-II redshift arises from the enhanced molecular dipole moments and electron-acceptor ability of the precursor, as well as the accumulation of graphene domains and pyrrolic nitrogen doping during carbonization polymerization, which collectively drive the narrowing of the energy gap. CDs-3 shows 15 mm penetration depth in gallbladder Near-infrared-II imaging (vs. clinically used indocyanine green 2 mm). With 1.44 signal-to-noise ratio and 334.5 μm resolution, it enables precise monitoring of biliary strictures/leakage. Selenium-doping-derived functionalized composite nanomaterials (CDs-3@pPB) exhibit potent reactive oxygen species scavenging and theranostic efficacy in liver fibrosis. This work elucidates the mechanism underlying the redshift of carbon dots emission into the Near-infrared-II and establishes a nanoplatform for hepatobiliary theranostics, demonstrating substantial clinical potential.

Photothermal therapy (PTT) is widely recognized for the treatment of infected wounds. However, the wound hyperpathia caused by infection and heat from PTT is often overlooked, which might affect wound healing significantly. In this study, we systematically investigate the management of pain in bacteria-infected wounds using PTT, and develop a photothermal antibacterial hydrogel capable of sustained and controlled analgesia system (STB-Lid Gel), aiming to simultaneously achieve efficient anti-infection and precise pain management. STB-Lid Gel is constructed from a silk fibroin (SF) hydrogel matrix co-loaded with lidocaine (Lid) as an analgesic and supramolecular stable radicals (TPT-B 12 ) as a novel second near-infrared-window (NIR-II) photothermal agent. During the treatment phase, the sustained release of Lid stored in STB-Lid Gel extends localized analgesic effects beyond 30 h. Under NIR-II laser irradiation, STB-Lid Gel mediates PTT for bacterial eradicating and enhancement of Lid release. These dual effects of anti-infection and pain management, effectively eradicates bacteria, alleviates PTT-induced acute pain, reduces local inflammation, and ultimately accelerates wound healing. This innovative approach highlights the importance of pain relief from PTT-associated acute pain in healing processes, presenting a viable strategy for combining antibacterial and analgesic treatment in infected wounds.

A RE-doped Bi 4 Ti 3 O 12 perovskite-based luminescence material is developed. • Dual NIR-II emissions of Ho 3+ /Er 3+ are achieved under 980 nm excitation. • High performance (Sr=1.3% K-1) phonon-assisted ratiometric thermometer is constructed. High-performance optical thermometers operating in the second near-infrared (NIR-II) window hold great promise for biomedical applications due to their tissue penetration depth and minimal autofluorescence. However, developing reliable NIR-II thermometers with high sensitivity and robust performance across a wide temperature range remains a challenge. Herein, the luminescence properties and NIR-II ratiometric thermometric performance of Er 3+ , Ho 3+ , Yb 3+ co-doped Bi 4 Ti 3 O 12 (BTO) perovskite oxide were investigated. Upon 980 nm excitation, NIR-II emission from Er 3+ at 1560 nm ( 4 I 13/2 → 4 I 15/2 ) and Ho 3+ at 1200 nm ( 5 I 6 → 5 I 8 ) was achieved. A ratiometric thermometer was constructed using the temperature-dependent intensity ratio, I Er /I Ho . Relying on the thermal dependence of this ratio, the BTO system exhibits a relative sensitivity (S r ) of 1.3% at 333 K. Furthermore, the results suggest that phonon characteristics of the BTO host influence energy-transfer processes and NIR-II thermometric performance, highlighting importance of host properties in designing rare-earth-doped NIR-II thermometric materials. Hollow manganese-dioxide-based nanoplatforms, which combine
hollow
structures with the intrinsic theranostic properties of Mn 2+ , have shown great potential for precise tumor diagnosis and treatment.
However, single imaging or therapeutic modalities are still insufficient
for high requirements of precise cancer theranostics. Herein, a hollow
manganese-based theranostic nanoplatform, MnO 2 –IR-820-Q/DOX@TMTP1
nanocomposite (Mn-NCs), was designed and synthesized. With the assistance
of the Mn-NCs, near-infrared-II (NIR-II, 900–1700 nm) fluorescence
imaging with high spatial resolution and low autofluorescence was
combined with magnetic resonance imaging (MRI) for highly efficient
and precise multimodal imaging of cervical tumors. The organic cyanine
dye IR-820 and PbS@CdS quantum dots (QDs) were integrated to form
a hybrid nanocomposite (IR-820-Q) with strong NIR-II fluorescence
emission, which was subsequently assembled onto a MnO 2 nanocarrier
to enable MRI and protect IR-820-Q from degradation. The hollow MnO 2 nanoshells served as a nanoscale structural framework for
IR-820-Q/DOX loading, tumor microenvironment-responsive degradation,
and Mn 2+ -mediated imaging/therapeutic functions. The cervical
tumor-targeting peptide TMTP1 was introduced to improve the tumor-targeting
potential of the nanocomposite, enabling NIR-II fluorescence visualization
of primary tumors and lymph node metastatic lesions. Therefore, Mn-NCs
are well suited for tumor-targeted imaging-guided intervention of
cervical cancer and lymphatic metastasis. By constructing the theranostic
nanoplatform, the photothermal therapy (PTT) as well as photodynamic
therapy (PDT) effect from the IR-820 was enhanced, and with the hydroxyl
radicals (via Fenton-like reaction) of Mn 2+ in the tumor
microenvironment, effective chemodynamic therapy (CDT) was also achieved.
The loading of doxorubicin (DOX) further endowed the nanoplatform
with a chemotherapeutic capability. Thus, the developed theranostic
nanoplatform showed strong NIR-II fluorescence/MRI-guided synergistic
PTT/PDT/CDT and chemotherapy, which significantly improved the therapeutic
outcomes. This study demonstrates a promising strategy for precise
cervical cancer diagnosis and treatment, highlighting the potential
of organic–inorganic hybrid nanomaterials for clinical translation.

Breast cancer remains a significant global health concern owing to the limitations of conventional therapies, such as side effects, drug resistance, and high costs. Nanocatalytic therapy has emerged as a promising alternative due to its tumor specificity, spatiotemporal controllability, and noninvasiveness. However, its effectiveness is limited by endogenous antioxidants like glutathione (GSH) and the low catalytic activity of nanocatalysts. Herein, an Cu/Au-doped polypyrrole nanocatalyst, Cu-AuPP, with multiple catalytic activities is developed by sequentially polymerizing pyrrole monomers using CuCl2 and HAuCl4, followed by PEGylation. The obtained Cu-AuPP catalyzes the production of hydroxyl radicals (·OH) and facilitates the oxidation of GSH to GSSG via redox reactions mediated by multivalent Cu ions, leading to oxidative damage, mitochondrial dysfunction, and tumor cell apoptosis. Upon 1064 nm laser irradiation, these catalytic activities were enhanced by elevated temperature and electron–hole separation mediated by Au nanoclusters, resulting in more intense oxidative damage to tumor cells. Collectively, the developed Cu-AuPP nanocatalytic medicine capable of simultaneously catalyzing GSH depletion and ·OH production via several improved catalytic mechanisms, has significant promise for the treatment of malignant tumors.

Phototheranostics, which allows simultaneous diagnosis and therapy, offers notable advantages in terms of noninvasiveness, controllability and negligible drug resistance, presenting a promising approach for disease treatment. By integrating second near-infrared (NIR-II, 1,000–1,700 nm) phototheranostic agents characterized by aggregation-induced emission (AIE) and cell membranes with specific targeting capacity, we have developed a versatile type of biomimetic nanoparticle (NP) for precise phototheranostics of pulmonary tuberculosis (TB). Coating the phototheranostic agents with preactivated macrophage membranes results in the formation of biomimetic NPs, which exhibit specific binding to TB through a lesion–pathogen dual-targeting strategy, allowing the accurate detection of Mycobacterium tuberculosis via NIR-II fluorescence imaging and precise photothermal therapy using the irradiation of a 1,064 nm laser. In comparison with traditional treatments, small individual granulomas (0.2 mm in diameter) in TB-infected mice are visualized, and improved antibacterial effects are achieved upon NP administration. Here we present a standardized workflow for the synthesis of the NIR-II AIE agents, their use for the fabrication of the biomimetic NPs and their in vitro and in vivo applications as phototheranostics against M. tuberculosis . The preparation and characterization of the NIR-II AIE agents requires ~8 d, the synthesis and characterization of the phototheranostic NPs requires ~8 d, the validation of in vitro targeting capacity and photothermal eradication requires ~26 d, and the in vivo NIR-II fluorescence imaging and imaging-guided photothermal therapy requires ~74 d. All procedures are straightforward and suitable for clinicians or researchers with prior training in organic synthesis and biomedical engineering.

Liver fibrosis presents a significant global health challenge due to its potential progression to cirrhosis, liver failure, or hepatocellular carcinoma, highlighting the necessity for precise imaging of fibrosis progression. Here, a straightforward synthesis method is proposed to design bioresponsive nanoprobes capable of achieving high-resolution bioimaging of liver fibrosis. Janus MnO 2 -coated Ag/Ag 2 S nanoparticles modified by polyethylene glycol (denoted as jMAP) can inherit both the activatable properties from Ag/Ag 2 S and MnO 2 components in the pathological microenvironment. Multiple lines of evidence supported that overexpressed levels of reactive oxygen species (ROS) in liver fibrosis efficiently transformed jMAP nanoprobes to Ag 2 S products, activating bright fluorescence in the second near-infrared window (NIR-II, 1000–1700 nm). In addition, the catalase-like and superoxide dismutase-like activities of jMAP probes reduce hypoxia and oxidative stress with a clearance rate of about 73.3% toward ROS, thereby downregulating hypoxia-inducible factor (HIF-1 α ) and NADPH oxidase-4 (NOX-4) by 49.5% and 47.9%, respectively. Overall, the developed jMAP probes demonstrate impressive diagnostic accuracy, offering transformative prospects for liver fibrosis diagnosis.

Radiation-induced enteritis (RIE) remains a critical clinical challenge, demanding advanced strategies for diagnostic and therapeutic monitoring. Conventional assessment methods are often time-consuming and primarily detect anatomic changes at moderate or late stages, limiting timely intervention. Here, DSNP@786s@EM, an innovative biomimetic nanoprobe based on NaYbF 4 :2%Er,2%Ce@NaYF 4 :30%Nd downshifting nanoparticles (DSNP) is presented for real-time, in situ monitoring of RIE via ratiometric near-infrared II (NIR-II) fluorescence imaging. This system integrates downshifting core-shell nanoparticles with strong NIR-II emission under dual-wavelength excitation (808 and 980 nm), and a reactive oxygen species (ROS)-responsive IR786s dye, enabling absorption competition-induced emission (ACIE)-based dynamic ratiometric sensing. Additionally, the probiotic Escherichia coli Nissle 1917 (EcN) membrane (EM) camouflage of the nanoprobe enhances its intestinal retention and colon-specific targeting, leading to superior diagnostic performance. Comprehensive in vitro and in vivo studies validate DSNP@786s@EM as a highly sensitive and reliable tool for real-time, quantitative ROS monitoring in an RIE mouse model. Furthermore, ratiometric NIR-II fluorescence imaging effectively tracks therapeutic responses to dexamethasone, demonstrating its potential for monitoring treatment efficacy. These findings highlight the translational value of DSNP@786s@EM in advancing RIE diagnosis and management, with broader applicability to oxidative stress-related disorders and precision medicine.
